Honey has been recognized for its role in disease prevention through nutritional modulation. In this study, we comprehensively assessed the physicochemical parameters, metabolic profile, antioxidant and anti-inflammatory activities of multifloral honeys produced by Apis cerana cerana in Sansha City (SS), Hainan Province. Metabolomic analysis identified SS honey had multiple bioactive compounds known for antioxidant and anti-inflammatory properties. Compared to A. cerana cerana honey from central regions of Hainan Province, SS honey showed higher levels of TFC (113.80 mg RE/100 g) and TPC (45.86 mg GAE/100 g). Furthermore, SS honey demonstrated greater antioxidant activity, as evidenced by FRAP (1503.16 μmol TE/kg), DPPH IC 50 (36.13 mg/mL), and ABTS IC 50 (11.03 mg/mL). SS honey significantly reduced the levels of cyclooxygenase-2 (COX-2), interleukin-1β (IL-1β), interleukin-6 (IL-6), inducible nitric oxide synthase (iNOS), and tumor necrosis factor-α (TNF-α) in lipopolysaccharide (LPS)-induced RAW 264.7 cells, along with suppressing the mRNA expression of these inflammatory markers.

Metal–Phenolic Networks-Reinforced Extracellular Matrix Scaffold for Bone Regeneration via Combining Radical-Scavenging and Photo-Responsive Regulation of Microenvironment

Zhiqing Liu,Tianlong Wang,Lei Zhang,Yiping Luo,Jinhui Zhao,Yixing Chen,Yao Wang,Wentao Cao,Xinyu Zhao,Bingqiang Lu,Feng Chen,Zifei Zhou,Longpo Zheng

The limited regulation strategies of the regeneration microenvironment significantly hinder bone defect repair effectiveness. One potential solution is using biomaterials capable of releasing bioactive ions and biomolecules. However, most existing biomaterials lack real-time control features, failing to meet high regulation requirements. Herein, a new Strontium (Sr) and epigallocatechin-3-gallate (EGCG) based metal–phenolic network with polydopamine (PMPNs) modification is prepared. This material reinforces a biomimetic scaffold made of extracellular matrix (ECM) and hydroxyapatite nanowires (nHAW). The PMPNs@ECM/nHAW scaffold demonstrates exceptional scavenging of free radicals and reactive oxygen species (ROS), promoting HUVECs cell migration and angiogenesis, inducing stem cell osteogenic differentiation, and displaying high biocompatibility. Additionally, the PMPNs exhibit excellent photothermal properties, further enhancing the scaffold's bioactivities. In vivo studies confirm that PMPNs@ECM/nHAW with near-infrared (NIR) stimulation significantly promotes angiogenesis and osteogenesis, effectively regulating the microenvironment and facilitating bone tissue repair. This research not only provides a biomimetic scaffold for bone regeneration but also introduces a novel strategy for designing advanced biomaterials. The combination of real-time photothermal intervention and long-term chemical intervention, achieved through the release of bioactive molecules/ions, represents a promising direction for future biomaterial development.

Microenvironment-responsive Bletilla polysaccharide hydrogel with photothermal antibacterial and macrophage polarization-regulating properties for diabetic wound healing

Zhengbo Hu,Kai Zhao,Xin Rao,XingCan Chen,Yujing Niu,Qiantao Zhang,Mingyuan Zhou,Yuchi Chen,FangMei Zhou,Jie Yu,Zhishan Ding,Bingqi Zhu

The healing of diabetic wounds (DW) is challenging due to the complex microenvironment, manifesting severe bacterial infections, persistent inflammation, and excessive oxidative stress. Therefore, overcoming these impediments to restore the wound microenvironment to a normal state is critical points and challenges in the development of effective DW dressings. Herein, a B/TF hydrogel was developed in our study using Bletilla striata polysaccharide (BSP), borax, and the tannic acid/ferric iron (TA/Fe 3+ ) complex. The crosslinking of the hydrogel constructed by dynamic borate bonds not only impart remarkable physical properties to the hydrogel, but also exhibited responsive release of BSP and TA/Fe 3+ complex to pH and glucose levels. Incorporating the TA/Fe 3+ complex provided notable near-infrared (NIR) photothermal properties to the hydrogel, thereby demonstrating significant photothermal antibacterial activity and antioxidant capability. Additionally, the B/TF hydrogel effectively regulated the transformation of macrophages from the pro-inflammatory M1 phenotype to the anti-inflammatory M2 phenotype. Animal experiments have also effectively confirmed that the treatment with B/TF hydrogel combined with NIR radiation promoted the regeneration of skin tissue in DW. In summary, the developed multifunctional B/TF hydrogel showed advantages and met clinical requirements for treating DW.

Microenvironment-Regulated Dual-Layer Microneedle Patch for Promoting Periodontal Soft and Hard Tissue Regeneration in Diabetic Periodontitis

Yaning Qu,Huajing Zeng,Lei Wang,Zhenlin Ge,Bin Liu,Zengjie Fan

Chronic periodontitis in individuals with diabetes can exacerbate the destruction of local periodontal soft tissues and accelerate the resorption of hard tissue. Currently, effective strategies to simultaneously restore both soft and hard periodontal tissues remain insufficient. To address this challenge, a multifunctional dual-layer microneedles (d-MNs) design is proposed to regenerate both periodontal soft and hard tissues in diabetic patients. The d-MNs substrate is composed of gelatin methacryloyl (GelMA) infused with nano-hydroxyapatite (nHA), which facilitates the differentiation of osteogenic cells into osteoblasts, thereby promoting alveolar bone regeneration. The tips of the d-MNs, on the other hand, are primarily made of hyaluronic acid (HA) combined with a magnesium-based metal-organic framework (Mg-MOF) loaded with glucose oxidase (GOX). This composition creates a hypoglycemic, angiogenic, and anti-inflammatory microenvironment, which supports soft tissue repair. When implanted at the site of periodontitis, the synergistic interaction between the d-MNs substrate and tips effectively promotes the regeneration of both soft and hard tissues, as demonstrated in diabetic rat models of periodontitis. These innovative d-MNs have the potential to revolutionize traditional approaches to treating diabetic periodontitis and can see broad application in dental clinics.

Microneedles Constructed by Swellable Hydrogels Loaded with Celastrol for Efficient Treatment of Skin Infections Induced by Drug-Resistant Bacterial Strains

Jianbin Deng,Mengqi Liu,Shiqi Gao,Dongjie Lei,Zhicheng Su,Fuqing Liang,Songyun Tang,Huiyuan Yang,Yuan-Yuan Huang,Weiquan Xie,Guang-Yu Pan

The urgent need for new antimicrobial drugs arises from the limited efficacy of traditional antibiotics against emerging drug-resistant strains. Celastrol (CSL) demonstrates an exceptional antibacterial property that remains unaffected by bacterial resistance, but its poor water solubility limits its wide applications. This study uses the hydrophobic inner cavity of mono-(6-diethylenetriamine-6-deoxy)-β-cyclodextrin (mβ-CD) (a derivative of cyclodextrin) to encapsulate CSL, constructing an inclusion complex (CSL@mβ-CD) to enhance the water solubility of CSL. The obtained inclusion complex is further incorporated into a swellable hydrogel microneedle (MN) to obtain CSL@mβ-CD/MN. The fabricated CSL@mβ-CD/MN can enable the sustained release of CSL, achieving effective bacterial eradication at infected sites. In vivo experiments demonstrate that CSL@mβ-CD/MN has a remarkable efficacy in the treatment of methicillin-resistant Staphylococcus aureus-induced subcutaneous abscesses and wound infections. Specifically, CSL@mβ-CD/MN can effectively penetrate the stratum corneum of the skin to realize rapid elimination of the bacteria in wounds. Moreover, CSL@mβ-CD/MN can efficiently scavenge reactive oxygen species, promote M2 polarization of macrophages, and relieve local inflammation at the wound sites. These results reveal that CSL@mβ-CD/MN holds great promise in the clinical treatment of acute skin infections induced by drug-resistant bacteria.

Modified Chitosan-Based Coating/Packaging Composites with Enhanced Antibacterial, Antioxidant, and UV-Resistant Properties for Fresh Food Preservation

Changyuan He,Liubo Yuan,Siwei Bi,Chaomei Zhou,Qin Yang,Jun Gu,Bin Yan,Jin He

Chitosan-based biomass packaging materials are a promising material for food preservation, but their limited solubility, antioxidant capacity, UV resistance, and mechanical properties severely restrict their application. In this study, we developed a novel chitosan-based coating/packaging composite (QCTO) using quaternary ammonium salt and tannic acid (TA)-modified chitosan (QCS-TA) and oxidized chitosan (OCS). The introduction of quaternary ammonium salt and TA effectively improves the water solubility and antibacterial, antioxidant, and UV-resistant properties of chitosan. The Schiff-base bond formed between OCS and QCS-TA, along with the TA-mediated multiple interactions, conferred the prepared composite film with good mechanical properties (69.9 MPa tensile strength) and gas barrier performance to water (14.3 g·h–1·m–2) and oxygen (3.5 g·mm·m–2·h–1). Meanwhile, the prepared QCTO composites demonstrate excellent biocompatibility and safety and are applied as coatings for strawberries and bananas as well as packaging films for mushrooms. These preservation experiments demonstrated that the prepared composites are able to effectively reduce weight loss, prevent microbial growth, maintain color, and significantly prolong the shelf life of fresh products (bananas, strawberries, and mushrooms extended shelf life by 6, 5, and 6 days, respectively). Therefore, the developed QCTO coating/packaging film shows great potential for applications in the field of food preservation and packaging.

Molecularly engineered lignin to polyphenol via organocatalysis as an active sunscreen ingredient

Yuanlong Guo,Pengcheng Liu,Lei Deng,Changwei Lui,Michael North,Gang Hu,Qitian Huang,Zongbao Kent Zhao,Haibo Xie

Phenolation is one of the effective strategies to synthesize lignin-based polyphenols, improve lignin's properties, and extend its value-added applications in biological, medicinal and cosmetic fields. Herein, by taking the structural feature advantage of lignin, an effective and green strategy was developed to molecularly engineer lignin into a robust lignin-3-(2-hydroxyphenyl)propionate ester (LPPE) derivative via a transesterification reaction between 3,4-dihydrocoumarin (DHC) and the aliphatic hydroxyls in lignin under organocatalysis. The strategy is optimized and the novel derivative was systematically characterized by 1 H, 13 C and 31 P nuclear magnetic resonance (NMR) and Fourier transform infrared (FT-IR) spectroscopy. The findings indicated that the successful introduction of 3-(2-hydroxyphenyl)propionate groups using a OH groups/DHC/organic base molar ratio of 1꞉1꞉0.3 at 120 °C for 6 h increased the content of phenolic hydroxyl groups from 1.793 1 to 3.017 9 mmol/g, and the LPPE exhibited excellent ultraviolet-absorbing and antioxidant performance with up to 90% free radical scavenging activity within 20 min using 5 mg/mL of LPPE. In addition, good biocompatibility and a high Sun protection factor (SPF) value of 40.9 were achieved at 5% ( w ) dosage of LPPE in the cream, indicating its significant application potential in sunscreen.

Multifunctional Nanosystem Based on Ultrasmall Carbon Dots for the Treatment of Acute Kidney Injury

Hanchun Yao,Zhihui Zhu,Mengyu Liu,Fangfang Sun,Mengyu Du,Yilin Sun,Bin Du

Acute kidney injury (AKI) is a critical medical condition characterized by high morbidity and mortality rates. The pathogenesis of AKI potentially involves bursts of reactive oxygen species (ROS) bursts and elevated levels of inflammatory mediators. Developing nanoparticles (NPs) that downregulate ROS and inflammatory mediators is a promising approach to treat AKI. However, such NPs would be affected by the glomerular filtration barrier (GFB). Typically, NPs are too large to penetrate the glomerular system and reach the renal tubules─the primary site of AKI injury. Herein, we report the development of ultrasmall carbon dots-gallic acid (CDs-GA) NPs (∼5 nm). These NPs exhibited outstanding biocompatibility and were shown not only to efficiently eliminate ROS and alleviate oxidative stress but also to suppress the activation of the NF-κB signaling pathway, leading to a reduction in the release of inflammatory factors. Importantly, CDs-GA NPs were shown to be able to rapidly accumulate rapidly in the renal tissues without the need for intricate targeting strategies. In vivo studies demonstrated that CDs-GA NPs significantly reduced the incidence of cisplatin (CDDP)-induced AKI in mice, surpassing the efficacy of the small molecular drug, N-acetylcysteine. This research provides an innovative strategy for the treatment of AKI.

Nanocomposite hydrogel orchestrating multiple modulation of degenerative microenvironment for potential application in intervertebral disc regeneration

Huitong Luo,Zhipeng Sun,Zetao Wang,Wanqing Lun,Qi Feng,Dafu Chen,Xiaodong Cao

Effective treatment of intervertebral disc degeneration with biomaterials remains a challenge, owing to the difficulty in simultaneously overcoming oxidative stress and its associated cascades in the nucleus pulposus microenvironment, which includes cellular senescence, apoptosis, inflammation, and extracellular matrix (ECM) degradation. To address these issues, a multifunctional hydrogel (HG-QNT) loaded with transforming growth factor- β 1 (TGF- β 1) and quercetin-based nanoparticles (QUNPs) is developed through borate ester bonding and Schiff base reaction-induced crosslinking. Specifically, QUNPs fabricated via coordination and hydrophobic interactions endow the hydrogel with extraordinary antioxidative properties. Benefiting from the multi-dynamic crosslinking, the hydrogel achieves self-healing, mechanical stability, and pH-responsive release of QUNPs and TGF- β 1. The HG-QNT hydrogel is demonstrated to enhance the proliferation of encapsulated nucleus pulposus cells, thereby providing an ideal platform for cell transplantation. The cooperative antioxidation of QUNPs and the hydrogel carrier renders HG-QNT effective in mitigating oxidative stress, resulting in the suppression of cellular senescence, mitochondrial dysfunction, apoptosis, excessive inflammation, and abnormal catabolism. Afterwards, TGF- β 1 and QUNPs act in synergy with the hydrogel to restore the anabolic/catabolic balance by enhancing ECM synthesis. Overall, the strategy orchestrating multiple modulation by HG-QNT hydrogel shows great potential for application in intervertebral disc regeneration.

Nutrients, Phytochemicals, and Antioxidant Capacity of Red Raspberry Nectar Fermented with Lacticaseibacillus paracasei

Feng Shi,Yin Qin,Shuyi Qiu,You Luo

Fresh raspberries are highly perishable, but lactic acid bacteria fermentation offers a favourable method for developing healthy products. This study investigated the effects ofLacticaseibacillus paracaseifermentation on the nutrients and phytochemicals of red raspberry nectar using widely targeted metabolomics, as well as its antioxidant activity. The fermentation notably disrupted the raspberry tissue structure, reshaped its non-volatile composition, and increased its DPPH and hydroxyl free radical scavenging abilities. A total of 261 compounds showed significant differences, with 198 upregulated and 63 downregulated. Among these, certain flavonoid glucosides (e.g., pelargonid-in-3-O-rutinoside, delphinidin-3-O-rutinoside-7-O-glucoside, and kaempferol-3-O-glucoside) were significantly downregulated, while some bioactive phenolic acids (e.g., 3-(4-Hydroxyphenyl)-propionic acid and DL-3-phenyllactic acid), alkaloids (e.g., deoxymutaaspergillic acid and indole-3-lactic acid), amino acids (e.g., L-phenylalanine and L-glutamine), and B vitamins (e.g., VB6, VB7, and VB3) were substantially upregulated. Furthermore, the Kyoto Encyclopedia of Genes and Genomes (KEGG) annotation and enrichment analysis revealed that metabolic pathways and the biosynthesis of secondary metabolites contributed significantly to the new profile of fermented red raspberry nectar. These findings provide valuable insights for developing fermented raspberry products usingLacticaseibacillus paracasei, which can help minimise fresh raspberry loss and enhance their valorisation.

One stone, four birds: Deep eutectic solvents serve multiple functions in the development of flexible smart tag for monitoring the freshness of high-protein food

Yuxiao Hu,Jiayan Zhang,Jiaxin Tong,Zhijian Tan

In this study, a facile “one stone, four birds” strategy was implemented to fabricate a flexible smart food packaging tag by incorporating purple potato extract obtained using deep eutectic solvents (DESs) into a polyvinyl alcohol (PVA)/gelatin (Gel) substrate. The favorable properties of DESs enabled them to serve four different important functions in tag development: anthocyanin extraction; mitigation of anthocyanin oxidation; as a plasticizer to increase the mechanical properties of the tag; and to enhance NH 3 sensitivity. The physicochemical, microstructural, and functional properties of the tag were characterized using various methods. Moreover, density-functional theory (DFT) was used to calculate the adsorption energies for NH 3 at multiple sites on the DES. The tag was employed to monitor the freshness of shrimp and fish, whereby it underwent conspicuous color changes during 10 h of storage, which enabled classification of the products as fresh, sub-fresh, or spoiled. Overall, the developed flexible smart tag shows significant potential for application in monitoring the freshness of high-protein food.

Optimization of Black Garlic Protein Extraction Process and Exploration of Its Properties and Functions with Enzymatic Hydrolysis Products

Jian Liu,Yuanyuan Wang,Bo Wang,Wei Zhang,Xiaoyu Ren,Youchuang Zhang,Lijun Jiang,Chunming Dong,Guihong Zhao

This study optimized the process of extracting protein from black garlic using an alkaline dissolution and acid precipitation method through response surface methodology. The optimal extraction conditions were determined as a solid-to-liquid ratio of 1:50, an extraction time of 100 min, an extraction temperature of 30 °C, and an alkaline extraction pH of 9.0. Under these optimized conditions, the actual black garlic protein (BGP) extraction yield was 12.10% ± 0.21%, and the isoelectric point of the obtained BGP was 3.1. Subsequently, this study extracted black garlic protein under optimal conditions and subjected it to enzymatic hydrolysis using different enzymes (trypsin, pepsin, and their mixed enzymes). The functional characteristics, antioxidant activity, and hypoglycemic activity of black garlic protein before and after enzymatic hydrolysis were compared. Among the hydrolysates, the pepsin hydrolysate (BGPH-P) had the smallest particle size (188.57 ± 1.93 nm) and the highest Zeta potential (−29.93 ± 0.42 mV). Scanning electron microscopy showed that BGPH-P had the smallest and most dispersed particles. Fourier-transform infrared (FTIR) spectroscopy revealed that the dual enzymatic hydrolysis hydrolysate (BGPH-PT) exhibited the most stable structure. Compared to BGP, the hydrolysates demonstrated significantly improved solubility, water-holding capacity, and foaming ability ( p < 0.05), while their emulsifying activity, emulsion stability, DPPH radical scavenging capacity, and hypoglycemic activity decreased. In summary, the BGP extracted using the optimized process demonstrated good antioxidant and hypoglycemic activities, while its enzymatic hydrolysate BGPH-P exhibited excellent solubility, water-holding capacity, and emulsifying properties, providing valuable insights for the further development of black garlic protein and its hydrolysates.
