Identification and molecular docking of xanthine oxidase and α-glucosidase inhibitors in Opuntia ficus-indica fruit

Huixian Wang,Xiaolu Zhou,Yixuan Liu,Wenxuan Xie,Derui Yang,Dongxue Huo,Quan Guo,Ruimin Wang

Opuntia ficus-indica fruit (OFI) is rich in bioactive compounds, which can promote human health. In this work, the purified OFI extract was prepared from OFI and its bioactivities were investigated. Xanthine oxidase (XOD) and α-glucosidase (α-Glu) inhibitors of the purified OFI extract were screened and identified by bio-affinity ultrafiltration combined with UPLC-QTRAP-MS/MS technology. The inhibitory effect of these inhibitors on enzymes were verified, and the potential mechanism of action and binding sites of inhibitors with enzymes were revealed based on molecular docking. The results showed that the total phenolic content of the purified OFI extract was 355.03 mg GAE/g DW, which had excellent antioxidant activity. Additionally, the extract had a certain inhibitory effect on XOD (IC 50 = 199.00 ± 0.14 µg/mL) and α-Glu (IC 50 = 159.67 ± 0.01 µg/mL). Seven XOD inhibitors and eight α-Glu inhibitors were identified. Furthermore, XOD and α-Glu inhibition experiments in vitro confirmed that inhibitors such as chlorogenic acid, taxifolin, and naringenin had significant inhibitory effects on XOD and α-Glu. The molecular docking results indicated that inhibitors could bind to the corresponding enzymes and had strong binding force. These findings demonstrate that OFI contains potential substances for the treatment of hyperuricemia and hyperglycemia.

Immunomodulatory hydrogel patches loaded with curcumin and tannic acid assembled nanoparticles for radioactive dermatitis repair and radioprotection

Ruoyuan Huang,Wenjie Sun,Wenzhao Li,Rui Hu,Rui Meng,Zheng Peng,Rongbo Yang,Tianyu Huang,Jinhui Du,Luoran Shang,Congying Xie

Radiation dermatitis, as a major side effect of radiotherapy, is a form of skin injury linked to the production of reactive oxygen species (ROS). Development of functional patches with excellent ROS scavenging ability and biosafety is crucial for radioactive dermatitis repair. Herein, we present a nanohybrid hydrogel patch composed with natural-derived substances. The patch possesses dual functionalities of chemically scavenging ROS and physically absorbing radiation, thereby alleviating radiation damage. Curcumin@tannic acid (Cur@TA) nanoparticles are synthesized through spontaneous coordination of TA and iron ions (Fe 3+ ), enabling the encapsulation of Cur. The nanoparticles are then loaded into methacrylate gelatin (GelMA) hydrogel patches, which endow the hydrogel with tissue adhesion as well as photothermal properties. Simultaneously, the patch exhibits powerful ROS scavenging capability and promotes M2 polarization of macrophages. Additionally, the high-water content of the GelMA hydrogel enables physical absorption of low-energy X-rays, providing radiation shielding. Based on these characteristics, we have demonstrated the augmented therapeutic efficacy of the hydrogel patch loaded with Cur@TA nanoparticles in the mouse model of radiation dermatitis. Our findings introduce a novel therapeutic approach for radiation protection and treatment, with potential clinical applications.

Improved antioxidant activity of pretreated lignin nanoparticles: Evaluation and self-assembly

Mingfu Li,Yingchuan Zhang,Hongli Ma,Qida Peng,Douyong Min,Pingjun Zhang,Liqun Jiang

Lignin nanoparticles (LNPs) have gained significant attention for their potential as natural antioxidants . This study investigated the effect of various pretreatment methods on the lignin structure and subsequent antioxidant activity of LNPs. Among four pretreated LNPs, hydrothermal LNPs exhibited the highest antioxidant activity , surpassing unpretreated, acid-pretreated and kraft LNPs, with an impressive efficacy of 91.6%. The relationship between LNPs' structure and antioxidant activity was revealed by 2D heteronuclear singular quantum correlation ( 1 H 13 C HSQC) and 31 P nuclear magnetic resonance (NMR). 1 H 13 C HSQC suggested the cleavage of β-O-4 ether bonds, as well as a decrease in ferulic acid and p -coumaric acid, which directly influenced the antioxidant activity of LNPs. 31 P NMR demonstrated a positive correlation between the total hydroxyl group content and the antioxidant activity . Besides, an isothermal kinetic model for scavenging free radicals was established based on Langmuir kinetic model instead of Freundlich model. Moreover, multilayer LNPs, based on layer-by-layer self-assembly, were prepared and exhibited remarkable antioxidant activity of 95.8%. More importantly, when blended with pure cosmetic cream , the multilayer LNPs maintained antioxidant activity of 86.7%. These finding may promote the practical applications of biomolecules, e.g. lignin additives in cosmetics and pharmaceuticals.

Inflammation-Targeted Nanomedicines Alleviate Oxidative Stress and Reprogram Macrophages Polarization for Myocardial Infarction Treatment

Danrong Hu,Ran Li,Yicong Li,Meng Wang,Lu Wang,Shiqi Wang,Hongxin Cheng,Qing Zhang,Chenying Fu,Zhiyong Qian,Quan Wei

Myocardial infarction (MI) is a critical global health challenge, with current treatments limited by the complex MI microenvironment, particularly the excessive oxidative stress and intense inflammatory responses that exacerbate cardiac dysfunction and MI progression. Herein, a mannan-based nanomedicine, Que@MOF/Man, is developed to target the inflammatory infarcted heart and deliver the antioxidative and anti-inflammatory agent quercetin (Que), thereby facilitating a beneficial myocardial microenvironment for cardiac repair. The presence of mannan on the nanoparticle surface enables selective internalization by macrophages rather than cardiomyocytes. Que@MOF/Man effectively neutralizes reactive oxygen species in macrophages to reduce oxidative stress and promote their differentiation into a reparative phenotype, reconciling the inflammatory response and enhancing cardiomyocyte survival through intercellular communication. Owing to the recruitment of macrophages into inflamed myocardium post-MI, in vivo, administration of Que@MOF/Man in MI rats revealed the specific distribution into the injured myocardium compared to free Que. Furthermore, Que@MOF/Man exhibited favorable results in resolving inflammation and protecting cardiomyocytes, thereby preventing further myocardial remodeling and improving cardiac function in MI rats. These findings collectively validate the rational design of an inflammation-targeted delivery strategy to mitigate oxidative stress and modulate the inflammation response in the injured heart, presenting a therapeutic avenue for MI treatment.

Insights into folic acid functionalization of self-assembled octenyl succinic anhydride starch micelles towards targeted delivery of selenium nanoparticles

Fang Xie,Nian Liu,Xiaoqing Liu,Xiaofang Feng,Zhaoxing Yang,Zhijun He,Jie Cai

Selenium nanoparticles (SeNPs) exhibit significant potential in antitumor therapy. However, challenges such as aggregation and lack of targeting capability limit their application. Herein, we developed selenium-loaded octenyl succinic anhydride starch (OSAS) micelles functionalized with folic acid (FA) for targeted tumor delivery. The FA-OSAS-SeNPs were synthesized through self-assembly, incorporating SeNPs into FA-conjugated OSAS micelles. Fourier Transform Infrared (FTIR) spectroscopy and UV–visible spectrophotometry confirmed the successful synthesis of FA-OSAS-SeNPs. The nanoparticles exhibited an average size of 131.66 ± 7.88 nm and a zeta potential of −19.54 ± 0.33 mV, with encapsulation efficiency and drug loading capacity of approximately 87.28 % and 8.96 %, respectively. FA-OSAS-SeNPs demonstrated good stability across various conditions, including different dilution ratios, temperatures, pH levels, and ionic strengths. In vitro studies showed that FA-OSAS-SeNPs exhibited significant targeted inhibitory effects on cervical cancer (HeLa) cells and markedly increased intracellular ROS levels, inducing apoptosis. This study presents a novel and effective strategy for targeted SeNPs delivery systems in tumor therapy, offering a valuable reference for future development of nanomaterials for clinical applications in cancer treatment.

Insights into the Chemical Structure and Antioxidant Activity of Lignin Extracted from Bamboo by Acidic Deep Eutectic Solvents

Kaiqin Song,Liping Yu,Shoulu Yang,Yan Cao,Lifen Li,Zhigang Wu,Hongtao Shi,Qiaorun Ma

Deep eutectic solvents (DESs) composed of choline chloride as hydrogen bond acceptors (HBAs) and six organic acids as hydrogen bond donors (HBDs) were used to extract lignin from bamboo (Phyllostachys edulis (Carrière) J. Houz.). The structures of the DES-extracted lignin samples were analyzed by Fourier-transform infrared spectroscopy (FT-IR), UV–visible spectroscopy (UV–vis), thermogravimetric analysis (TG), and gel permeation chromatography (GPC) to investigate the relationship between the chemical structure of lignin and its antioxidant activity. The results showed that DES treatment removed a large portion of the lignin (73.37–86.38%) from bamboo, and the chemical structure of lignin was changed due to the use of different types of HBDs. The extracted lignin exhibited good UV–vis light shielding properties, thermal stability, and antioxidant activity. Moreover, the total phenolic hydroxyl content of lignins was positively correlated with their antioxidant activity, while the molecular weight of lignins was negatively correlated with their antioxidant activity. Notably, lignin extracted with choline chloride-p-toluenesulfonic acid had the highest phenolic hydroxyl content and lower molecular weight, showing the strongest antioxidant activity (IC50 DPPH = 417.69 μg/mL, IC50 ABTS = 58.62 μg/mL). This study confirms the high thermal stability, excellent antioxidant activity, and UV shielding properties of lignin extracted with choline chloride-organic acid DESs, suggesting its potential application in the fields of antioxidants and material modifiers.

Isolation and Identification of Indole Alkaloids from Aspergillus amstelodami BSX001 and Optimization of Ultrasound-Assisted Extraction of Neoechinulin A

Shuyao Li,Xiaobo Liu,Qiuya Gu,Xiaobin Yu

This study aimed to investigate the alkaloid secondary metabolites ofAspergillus amstelodamiBSX001, a fungus isolated from Anhua dark tea, and to improve the extraction yield of the active ingredients by optimizing the extraction process. The structural characterization of the compounds was investigated using mass spectrometry (MS) and nuclear magnetic resonance (NMR) spectroscopy. The antioxidant activity of echinulin-related alkaloids was evaluated by determining the total reducing power and DPPH radical scavenging capacity. The extraction process of the compound with optimum activity was optimized by a single-factor test and response surface methodology (RSM) combined with Box–Behnken design (BBD). The optimized result was validated. Finally, a new alkaloid 8-hydroxyechinulin (1), and four known alkaloids, variecolorin G (2), echinulin (3), neoechinulin A (4), and eurocristatine (5), were isolated. Echinulin-related compounds1,3, and4possessed certain antioxidant activities, with IC50values of 0.587 mg/mL, 1.628 mg/mL, and 0.219 mg/mL, respectively, against DPPH radicals. Their total reducing power at a concentration of 0.5 mg/mL was 0.29 mmol/L, 0.17 mmol/L, and 4.25 mmol/L. The extraction process of neoechinulin A was optimized with the optimum extraction parameters of 72.76% methanol volume fraction, 25 mL/g solid–liquid ratio, and 50.8 °C soaking temperature. Under these conditions, the extraction yield of neoechinulin A was up to 1.500 mg/g.

Isolation of a novel polysaccharide from Tribonema minus and preparation of polysaccharide–selenium nanoparticles with enhanced antioxidant activity

Rundong Yang,Yuhao Guo,Yanchao Mao,Wenxiang Zhang,Feifei Wang,Zhenzhou Zhu

Tribonema polysaccharide (TP), a biologically active polysaccharide containing β-1,3-glycosidic bonds, has not been extensively studied as a dispersing agent for stabilising selenium nanoparticles (SeNPs), and its synergistic effects with SeNPs remain poorly understood. This study investigates the relationship between the antioxidant activity and physicochemical properties of TP–SeNPs, which were synthesised through chemical reduction at varying TP-to‑selenium mass ratios (TPSMRs). TP was isolated from Tribonema minus using ultrasonic-assisted extraction, achieving a 119.64 % increase in yield under optimal conditions (400 W, 25 min, 1:400 solid-to-liquid ratio). Structural analysis identified TP as a novel β-1,3-glucan (8.46 kDa) composed exclusively of glucose units. Moreover, TP–SeNPs prepared at varying TPSMRs exhibited significantly higher antioxidant activity than either TP or SeNPs alone, demonstrating a clear synergistic effect. Notably, TP–SeNPs prepared at a TPSMR of 2:3 exhibited the lowest half-maximal inhibitory concentrations for scavenging 2,2-diphenyl-1-picrylhydrazyl, superoxide anions, and hydroxyl radicals (0.30, 0.77 and 0.46 mg/mL, respectively). This was attributed to the increased selenium content and reduced particle size of these complexes. Additionally, TP–SeNPs displayed excellent dispersibility and stability, likely due to non-covalent interactions between TP and SeNPs. In cellular assays, TP–SeNPs exhibited low cytotoxicity and dose-dependently enhanced cell viability under H 2 O 2 -induced oxidative stress. Cells treated with 200 mg/L TP–SeNPs exhibited a 35.83 % increase in viability compared with the model group. This protection was associated with elevated antioxidant enzyme activity, increased glutathione levels and reduced malondialdehyde content, thereby mitigating oxidative damage in HepG2 cells. This study is the first to characterise a novel β-1,3-glucan from T. minus and to elucidate the structure–activity relationships of TP–SeNPs, highlighting their potential as functional additives or pharmaceutical supplements.

Impact of Co-60 γ-ray irradiation on the cross-linking and stability of fish collagen: Structural changes and digestibility

Junwei Qin,Litong Liu,Chengdu Miao,Bifeng Lan,Tao Liao,Xiaofei Tian,Zhenqiang Wu

Irradiation can promote the cross-linking of collagen to enhance its biological properties, but the molecular changes in irradiated collagen have few been investigated. This study aimed to explore the impact of irradiation treatment on the molecular structure stabilization and cross-linking degree of skin collagen. The results showed that collagen in solution was more susceptible to Co-60 gamma irradiation than that in fish skin and in sponge forms. The solubility of collagen in solution decreased by more than 80% and the carbonyl content increased by more than 4 times compared with the control group after 12 kGy irradiation. Spectroscopic analysis and sodium dodecyl sulfate - polyacrylamide gel electrophoresis (SDS-PAGE) showed that collagen in solution was cross-linked, especially at high irradiation doses. However, the triple helical structure of skin collagen and collagen sponge remained intact. Furthermore, irradiation also reduced the sensitivity of collagen to collagenase. Gastrointestinal digestion results showed that the antioxidant activity of collagen increased at 1–3 kGy doses and decreased at 9–12 kGy doses. Collagen irradiated at high doses (9–12 kGy) was more difficult to digest and had a larger average particle size. This study provides a potential new approach for the preparation of stable fishery-derived collagen with excellent antioxidant activity and resistance to digestion by irradiation treatment.

Inclusion complex of quercetin with sulfobutylether β-cyclodextrin: preparation, characterization, antioxidant and antibacterial activities and the inclusion mechanism

Kunkun Dai,Jiayi Wu,Xinyang Liu,Suilou Wang,Yihang Liu,Hehe Li,Haixiang Wang

Quercetin (QCT) has a variety of pharmacological effects, such as antioxidant, antibacterial, anticancer, anticardiovascular and antiaging effects. However, its poor water solubility, stability and bioavailability limit its applications. The special structure of cyclodextrins and their derivatives with a hydrophobic inner cavity and hydrophilic outer wall can load a variety of hydrophobic drugs of a suitable size and shape, thereby improving the stability and solubility of these molecules. In this study, an inclusion complex of quercetin and sulfobutylether-β-cyclodextrin was prepared. It was characterized via FT-IR, UV, 1H NMR, XRD, DSC, and SEM analysis, which revealed the successful formation of the inclusion complex. In vitro biological activity estimations were carried out and the results indicated that the inclusion complex displayed higher antioxidative and antibacterial properties compared with free QCT. In addition, the mechanisms of inclusion were explored using 1H NMR analysis and docking calculations, thus providing a theoretical basis for obtaining an inclusion complex.

Incorporating ε-polylysine hydrochloride, tea polyphenols, nisin, and ascorbic acid into edible coating solutions: Effect on oxidation and structure of marinated egg proteins

Jingbo Liu,Lu Han,Dongkun Cheng,Shengrao Li,Xiumei Chen,Yiding Yu,Deju Zhang,Ting Zhang

Egg processing products lose their attraction due to protein oxidation and degradation, while the edible coating incorporating antioxidant and antimicrobial agents can address this deficit. In this work, chitosan (CS) in combination with ε-polylysine hydrochloride, tea polyphenols, nisin, and vitamin C (CS–NH-TC) was applied as a novel edible coating material for preserving marinated eggs for 20 days and its effects on egg quality, and protein oxidation and structure were determined. The CS–NH–TC coating exhibited the highest antioxidant and antibacterial activities among all coatings. Compared to uncoated marinated eggs (Sulfhydryl: 12.02 μmol/g, dityrosine: 6251.8, protein hydrophobicity: 3691, turbidity: 0.13 mg/mL, particle size: 342.13 nm), CS- and CS–NH–TC-coated eggs exhibited improved protein stability, as evidenced by a higher sulfhydryl (22.20 and 30.92 μmol/g) and lower dityrosine content (4722 and 4216), protein hydrophobicity (2891.72 and 865.75), turbidity (0.06 and 0.03 mg/mL), and particle size (292.06 and 256.57 nm). Additionally, edible coatings, especially CS–NH–TC coatings, were positively correlated with smaller intermolecular force changes in marinated eggs, corresponding to suppressed protein oxidation and conformational changes. Furthermore, CS–NH–TC-coated samples showed more stable protein secondary structures and less disorderly and aggregated protein microstructure, and better color, texture, and sensory scores. Our study could provide a theoretical basis for stabilizing the protein structure of egg processing products.

In Situ Reaction-Generated Aldehyde-Scavenging Polypeptides-Curcumin Conjugate Nanoassemblies for Combined Treatment of Spinal Cord Injury

Yixuan Liu,Feng Lin,Cheng Wu,Wanguo Liu,Hao Wang,Chunsheng Xiao,Xuesi Chen

The microenvironment after traumatic spinal cord injury (SCI) involves complex pathological processes, including elevated oxidative stress, accumulated reactive aldehydes from lipid peroxidation, excessive immune cell infiltration, etc. Unfortunately, most of current neuroprotection therapies cannot cope with the intricate pathophysiology of SCI, leading to scant treatment efficacies. Here, we developed a facile in situ reaction-induced self-assembly method to prepare aldehyde-scavenging polypeptides (PAH)-curcumin conjugate nanoassemblies (named as PFCN) for combined neuroprotection in SCI. The prepared PFCN could release PAH and curcumin in response to oxidative and acidic SCI microenvironment. Subsequently, PFCN exhibited an effectively neuroprotective effect through scavenging toxic aldehydes as well as reactive nitrogen and oxygen species in neurons, modulating microglial M1/M2 polarization, and down-regulating the expression of inflammation-related cytokines to inhibit neuroinflammation. The intravenous administration of PFCN could significantly ameliorate the malignant microenvironment of injured spinal cord, protect the neurons, and promote the motor function recovery in the contusive SCI rat model.
