Engineering Crystalline Frameworks into Porous Liquids to Fabricate Graphene Oxide/Porous Liquid Membranes for Efficient Li+/Mg2+ Separation

Liu Lingfeng,Huang Jian’an,Wang Ying,Pang Dekang,Ding Yuling,Liu Jiamin,Zhang Xiaojing,Guo Changsheng,Jia Jianbo,Zhang Mengchen,Luan Tiangang

The quest for sustainable energy transition has intensified the demand for lithium extraction from salt-lake brines, necessitating advanced membranes for high-efficiency Li + /Mg 2+ separation. Integrating porous frameworks and two-dimensional (2D) scaffolds has shown promise for fabricating ion-permselective membranes, but is hampered by poor processability. Here we transform crystalline frameworks into porous liquids (PLs), and embed them within graphene oxide (GO) laminates to engineer an ultrathin and defect-free GO/PLs membrane. Discrete porous guests are uniformly confined within 2D nanofluidic channels mediated by a sterically hindered solvent. The GO/PLs membrane exhibits ultrahigh Li + permeance (0.75 mol m −2 h −1 ) and exceptional Li + /Mg 2+ selectivity (154.4) by regulating ion dehydration and ion-to-channel interactions. Efficient lithium extraction from simulated brine is demonstrated, reversing the Mg/Li ratio of 40 to achieve a Li/Mg ratio of 4.6. This work reports the manufacturing strategy of hybrid membranes based on porous frameworks and 2D materials for precise ion separation.

Enantiomeric covalent organic polymers-based electrodes for selective electrochemical recognition of cysteine enantiomers and detection of L-Cysteine

Nie Ying,Zhang Hui,Zhao Jianwei,Fang Zhili

In this work two novel chiral sensing platforms were constructed from enantiomeric covalent organic polymers (R-CCOP and S-CCOP) prepared via a one-step solvothermal method, for the enantioselective recognition of L- and D-cysteine (Cys). The prepared materials were characterized using scanning electron microscopy (SEM), energy-dispersive spectroscopy (EDS), nitrogen adsorption-desorption (BET), and X-ray diffraction (XRD). By integrating the chiral characteristics of R-(+)-1,1-bisnaphthyl-2,2-diamine/S-(-)-1,1-bisnaphthyl-2,2-diamine building blocks with the electrochemical behavior of large π-conjugated systems formed after polymerization, two types of electrochemical chiral sensing interfaces were constructed using glassy carbon electrodes (GCE) modified with R-CCOP and S-CCOP. These interfaces were employed for the electrochemical chiral recognition of Cys enantiomers. The results showed that R-CCOP/GCE exhibited the highest chiral recognition efficiency, with an enantioselectivity coefficient ( I L / I D ) of 2.86, while S-CCOP/GCE had a chiral recognition efficiency of 1.37. Density functional theory (DFT) calculations confirmed that both R-CCOP and S-CCOP demonstrated stronger recognition capabilities for L-Cys, with hydrogen bonding being the key factor in recognition. Additionally, the R-CCOP/GCE sensing interface exhibited a linear response within a concentration range of 5 µM to 500 µM for L-Cys, with a detection limit of 0.50 µM (S/ N  = 3) for L-Cys, along with good stability and reproducibility, making it an effective chiral sensing interface for the recognition of Cys enantiomers.

METTL16 S-glutathionylation-triggered RNA m6A modification of IGF2BP3 inhibits CFTR expression and promotes chemoresistance in colorectal cancer

Xu Lu,Ma Yan,Yao Wang,Qiu Ya-ju,Xu Chao,Xu Fang-yue,Zou Zhi-miao,Dai Wu-min,Gong Wan-gang,Ma De-ning,Chen Xiao-wan,Ye Wen-kai,Huang Yi-yun,Chen Hai-tao,Hong Lue,Mo Jia-ping,Feng Jian-guo,Yu En-yan,Chen Wei,Zhang Yu-hua,Yao Qing-hua,Li Xia

5-Fluorouracil (5-FU) resistance in colorectal cancer (CRC) involves oxidative stress mechanisms, but the role of epitranscriptomic regulation remains unclear. This study investigates how oxidative post-translational modifications of the N6-methyladenosine (m 6 A) methyltransferase METTL16 contribute to 5-FU resistance. Parental (HCT8, HCT15) and 5-FU-resistant CRC cells were compared using redox proteomics, m 6 A-seq, RNA-seq, and functional assays. METTL16 S-glutathionylation was assessed via streptavidin pulldown and mass spectrometry. IGF2BP3’s role was validated through knockdown/overexpression, patient-derived organoids (PDOs), and xenograft models. Clinical relevance was evaluated in 112 CRC patient tissues and data from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) databases. Drug synergy was tested using isoliquiritigenin (ISO) combined with 5-FU. Resistant cells exhibited elevated global protein S-glutathionylation and reduced m 6 A RNA methylation. METTL16 underwent site-specific S-glutathionylation at Cys548, leading to its degradation and subsequent m 6 A loss. Integrative omics identified IGF2BP3 as a key METTL16 target: reduced m 6 A on IGF2BP3 mRNA enhanced its nuclear export, splicing, and overexpression. IGF2BP3 knockdown sensitized resistant cells to 5-FU in vitro and in vivo, while overexpression conferred resistance. Mechanistically, IGF2BP3 destabilized CFTR mRNA (an ABC transporter), reducing 5-FU uptake. Clinically, high IGF2BP3 correlated with poor survival and 5-FU resistance in CRC patients. Targeting IGF2BP3 with ISO synergized with 5-FU, overcoming resistance in cells and xenografts. Oxidative stress-induced METTL16 S-glutathionylation drives 5-FU resistance by reducing m 6 A modification, enabling IGF2BP3 overexpression and CFTR suppression. IGF2BP3 is a biomarker of clinical resistance, and its targeting with ISO represents a promising combinatorial strategy to restore 5-FU efficacy in CRC.

Multiple miRNA Analysis Based on DNA Fiber Barcodes for Cell Typing

Fang Xiaoxing,Li Xuewei,Gao Wenqi,Li Chao,Zheng Ji

Tumor cell subtyping is clinically critical for early cancer diagnosis and prognostic assessment. As key regulators of cellular phenotype, microRNAs (miRNAs) exhibit cell-specific expression profiles that serve as pivotal tumor classification biomarkers. However, current multiplex miRNA detection technologies suffer from notable limitations. Quantitative polymerase chain reaction (qPCR) is vulnerable to primer cross-amplification interference, and sequencing and microdevice-based methods are either cost-prohibitive or operationally cumbersome. These drawbacks severely limit their clinical applicability for rapid, user-friendly diagnostics. To address these limitations, this study proposes a novel DNA fiber barcode-based multiplex miRNA detection strategy for accurate tumor cell subtyping. Programmable DNA tile self-assembly constructs functional DNA fibers with target-specific recognition units. Binding of miRNAs triggers conformational changes in the hairpin probe, generating detectable fluorescent barcode signals that enable the simultaneous identification and quantification of miRNAs. Experiments demonstrate a robust linear response within a range of 10 pM–70 nM, with detection limits between 2.4 pM and 5.0 pM for the analysis of miR-21, miR-105, and miR-155. The results align consistently with qPCR findings for cancer and normal cell lines, confirming the method’s accuracy and reliability. This strategy overcomes existing technical limitations, provides new insights for miRNA-based cellular analysis, and establishes both theoretical and technical foundations for precision tumor subtyping.

Broadly neutralizing nanobodies target a defined structural pivot site on the RSV fusion protein

Qianqian Wang,Xianliang Ke,Entao Li,Dongxiang Hong,Zekai Cheng,Hongxin Li,Jiachen Zhang,Tengchuan Jin,Rui Gong,Bo Shu,Sandra Chiu

Respiratory syncytial virus (RSV) is a leading cause of acute lower respiratory tract infections in infants and elderly individuals. Although nirsevimab represents a recent breakthrough against RSV infection, the emergence of resistant variants highlight the need for additional antiviral strategies. Here, we report two nanobodies (Nbs), 1G9 and 1D8, that target the RSV fusion (F) protein and exhibit high neutralizing activity against RSV subtypes. In vivo, the Nbs-Fc demonstrated robust prophylactic and therapeutic efficacy. Cryo-electron microscopy revealed that 1G9 and 1D8 specifically engage a conformational pivot site within antigenic site IV, crosslinking the metastable heptad repeats B (HRB) and the conformationally stable domain II of the F protein. This interaction stabilizes the prefusion conformation and prevents the structural rearrangement required for membrane fusion. Notably, the binding residues are highly conserved across RSV subtypes, accounting for the broad-spectrum neutralization observed. Together, our findings identify a structurally conserved and functionally critical epitope on RSV F and highlight 1G9 and 1D8 as promising candidates for next-generation prophylactic and therapeutic interventions against RSV. Respiratory syncytial virus (RSV) poses a significant threat to the health of infants and the elderly. Although the current use of nirsevimab for prevention represents a major breakthrough, its efficacy remains limited. There is an imperative need to develop a safe, broad-spectrum, and cost-effective next-generation RSV neutralizing antibodies (Nbs). An attention-demanding hunting paradigm engages the superior colliculus–zona incerta circuit mediating analgesia in male mice

Zhang Xue,Liu Xiao-Juan,Yin Cui,Zhou Qiangqiang,Zhang Jiaqi,Wang Liwei,Cao Jun-Li,Xiao Cheng,Zhou Chunyi

Attention distraction mitigates pain, yet its neural basis remains elusive. We established cricket hunting as an attention-demanding paradigm that alleviates acute and chronic pain in male mice. Activity tagging revealed that cricket hunting activated a subset of glutamatergic neurons in the superior colliculus (SC). Selective stimulation of these hunting-activated neurons alleviated chronic hyperalgesia, primarily mediated by their projections to the zona incerta (ZI). Notably, repeated engagement, either through the hunting paradigm or by repeatedly activating hunting-activated SC-ZI projections, induced sustained analgesia (lasting at least 6 hours), linked to potentiated glutamatergic inputs to ZI GABAergic neurons. Furthermore, SC-ZI projections originating from SC neurons expressing tachykinin precursor 1 (Tac1) selectively elevated neuropathic pain thresholds via substance P release and neurokinin 1 (NK1) receptor-dependent excitatory synaptic transmission onto ZI GABAergic neurons. Together, we show that a specific SC-ZI circuit drives attention-induced analgesia, highlighting the SC Tac1 -ZI pathway as a therapeutic target for chronic pain.

Organic-inorganic hybrid scintillating glass for high-energy particle detection

Li Hongwei,Lv Shichao,Qiu Jianrong,Zhou Shifeng

Glass scintillators are promising for large-scale radiation detection but traditionally suffer from low light yield and slow response. Here, we present highly transparent, homogeneous, and size-customizable organic-inorganic hybrid scintillating glasses, combining an inorganic glass host with organic components. The resulting hybrid glasses exhibit excellent radiation response, including rapid luminescence decay and enhanced scintillating efficiency. Notably, they achieve superior α-particle detection performance. In addition, their low density enables selective α-particle detection in mixed radiation fields and minimizes γ-ray interference, outperforming conventional glass scintillators. Furthermore, the glasses can be doped with elements such as lithium, boron, and gadolinium, offering efficient thermal neutron detection. These findings highlight the potential of organic-inorganic hybrid glasses as advanced scintillating materials for high-energy radiation field detection, with enhanced functionality for applications in nuclear medicine, oil exploration, and nuclear safety.

~100% upcycling of chlorinated/fluorinated plastic mixtures to H2 and nanotubes over FeNi/Ni/C by microwave catalysis

Zhao Jun,Yang Jianlong,Wang Duanda,Zhang Huanrong,Jia Qianqian,Zhang Lei,An Zhenguo,Xue Mianqi,Xie Haijiao,Ma Wangjing,Zhang Lu,Zhao Sui,Tang Junwang

The catalytic upcycling of plastic mixtures into H 2 and carbon nanotubes (CNTs) represents a transformative strategy for advancing plastic management and the circular economy. Herein, an in-situ constructed high-performance FeNi/Ni/C catalyst combined with prompt microwave catalysis enables efficient conversion of LDPE-PVC, LDPE-HDPE-PP-PS-PVC-PTFE and landfilled waste mixture into valuable H 2 and CNTs, resulting in near-quantitative atom economy (i.e., H 2 and carbon efficiencies are nearly 100%) together with H 2 and CNTs yields reaching >922 mmol g -1 catalyst  and >10600 mg g -1 catalyst , respectively. Furthermore, this microwave catalysis strategy presents an excellent stability (>35 cycles) via Cl/F-based regeneration and energy efficiency (80% reduction vs. traditional thermal catalysis), alongside a high turnover number of 107.6 mol carbon mol -1 NiFe . Fundamentally, the Ni species contribute to C-H breaking, and Fe is beneficial for CNT formation during plastic decomposition. Here, we show a cost-effective platform for upcycling of chlorinated/fluorinated plastic mixtures, aligning with resource circularity objectives.

Harmonized sodium coordination engineering for high-energy phosphate cathodes

Li Xinyu,Zhang Duoduo,Fan Xiangfeng,Gao Lina,Chen Weixin,Wang Liguang,Cao Ruiguo,Jiao Shuhong,Sun Yang,Guo Hao,Hu Yong-Sheng,Lu Xia,Pan Huilin

Fe-based polyanionic cathodes are promising for large-scale Na-ion batteries owing to their stability, safety and elemental abundance, however their capacity remains limited by electrochemically inactive Na sites and irreversible Na loss. Here we identify that the Na + coordination environment critically influences the Na-site accessibility and redox activity in Na 4 Fe 3 (PO 4 ) 2 P 2 O 7 -type cathodes. Combined experimental and theoretical analyses reveal that precise V 3+ substitution at the Fe2 site harmonizes Na + coordination geometry and softens the polyanionic framework, thereby activating previously inert Na sites and stabilizing high-voltage redox reactions above 4 V. The optimized Na 3.4 Fe 2.4 V 0.6 (PO 4 ) 2 P 2 O 7 achieves full Na + utilization (3.4 Na + , 150.7 mAh g −1 ) and a 52% increase in energy density (487 Wh kg −1 ), approaching the practical limit of Li-ion phosphate cathodes. It also demonstrates exceptional durability over 10,000 cycles in the 2.1-4.5 V range and stable pouch-cell performance. These findings provide a coordination-based strategy to overcome intrinsic capacity limitations in phosphate cathodes, enabling high-energy, durable Na-ion batteries.

In situ formation of adaptive electronic skin in 2 seconds enabled by metal coordination

Wang Xiaojuan,Pan Xiaosen,Jiang Junzhi,Song Wanlong,Zhou Xiaoqi,Lin Xu,Zhao Jingye,Zhang Zhengjian,Liu Zhenxing,Ma Xiaojun,Liu Hongbin,Gao Meng

Epidermal electronics, which are flexible and conformable electronic systems designed to interact seamlessly with human skin, hold great promise for healthcare monitoring and personal electronics. However, traditional fabrication methods face challenges of reliance on non-sustainable materials, intricate and time-consuming processes, and material softness-induced fragile transfer to target substrates. Inspired by the “milk skin” phenomenon, we developed a rapid dipping-dipping molecular assembly method to fabricate cellulose-based bio-skin in situ within seconds, exhibiting ultra-thin, highly conformal, shape-customizable, degradable, and low-impedance performances. This technique immerses substrates sequentially into carboxymethyl cellulose (CMC) and Cu(II) solutions, leveraging strong metal-coordination interactions. Membrane formation efficiency, influenced by the oxidation and coordination characteristics of metal ions, follows the order: Cu(II) > Fe(II) > Ca(II). CMC-Ag(I)/CMC-Cu(II) form stable membranes, whereas CMC-Fe(II) forms fragmented structures, and CMC-Mg(II)/CMC-Ca(II) remain in solution. This adaptable method can also be extended to other biomacromolecules like methylcellulose and carboxymethyl chitosan, broadening applications. The bio-skin enables real-time monitoring of electrocardiograms (ECG), electrooculograms (EOG), electroencephalograms (EEG), and electromyograms (EMG), showcasing its potential for wearable, biocompatible electronics in healthcare.

Photoactivated probiotic micro-reactor synchronizes STING/TLRs agonists to spatiotemporally synergize antitumor immunotherapy

Yuzhi Qiu,Yunting Liu,Sihan Chen,Yidi Liu,Xi Yu,Xiangliang Yang,Yan Zhang,Yanhong Zhu

Reprogramming tumor-associated macrophages (TAMs) from the pro-tumoral M2-like state to the immunostimulatory M1-like phenotype has emerged as a promising strategy for tumor therapy. However, most M2-like TAMs are preferentially located in hypoxic regions of the tumor, which are poorly accessible to many advanced drug delivery systems, posing a significant challenge to effective TAM reprogramming. Here, leveraging the tropism of facultative anaerobic bacteria to localize and propagate in the hypoxic tumor, an optogenetically engineered Escherichia coli Nissle 1917 strain conjugated with murine STING agonist (EcN flaB @UPD) was developed for cancer-specific immunotherapy. Upon near-infrared light illumination, the blue and UV emissions from upconversion nanoparticles (UCNPs) simultaneously activate the expression of Toll-like receptor (TLR) agonist, flaB, from EcN flaB , and the release of photocaged murine STING agonist, DMXAA, respectively. This spatiotemporally synchronized dual release ensures co-localized STING and TLR5 agonists inside the hypoxic niche, repolarizing TAMs from the M2 to the M1 phenotype via synergistic TLR5-MAPK1-NF-κB and STING-NF-κB signaling. The polarization of TAMs enhances their antigen-presenting capacity and, more importantly, activates the cytotoxic, stem-like and memory CD8 + T cells responses. This subsequently inhibits tumor growth, relapse, and metastasis in the murine 4T1 tumor model. Collectively, our work introduces the bacteria-based system that uses near-infrared light to dual-release immunotherapeutics for systemic anti-tumor immunity, opening new avenues for precise and effective cancer immunotherapy.
