Lipase immobilization plays a pivotal role in enhancing enzyme stability, reusability, and practical applicability in ligand fishing, a technique used to identify bioactive compounds from traditional Chinese medicine (TCM). However, conventional methods often lead to significant activity loss due to suffering from enzyme leakage, conformational distortion, and mass transfer resistance. In this study, we present a dynamic defect-engineering (DDE) strategy within zeolitic imidazolate framework-8 (ZIF-8) to overcome these limitations. The DDE process enables adaptive formation of transport channels driven by enzyme–framework interactions, facilitating the migration of lipase from surface adsorption to intracrystalline encapsulation. Brunauer–Emmett–Teller (BET) analysis confirmed successful encapsulation, showing a reduced specific surface area of 565.1 m 2 /g for lipase@ZIF-8, indicative of enzyme incorporation within the defect-engineered framework. This structural optimization significantly improved biocatalytic performance, achieving 82 % activity retention and 65 % immobilization yield, compared to 44 % activity retention in conventional in situ encapsulation. Utilizing the preserved catalytic activity, we established a lipase-based ligand fishing platform, which successfully identified 20 potential inhibitors (10 anthraquinones and 10 flavonoids) from Cassiae Semen and Citri Reticulatae Pericarpium using HPLC-Q-TOF-MS/MS analysis. Overall, this work demonstrates the effectiveness of the DDE strategy in enhancing lipase immobilization within ZIF-8, offering a robust and efficient platform for the high-throughput screening of lipase inhibitors from natural products and advancing the application of enzyme-based ligand fishing in TCM research.

Immobilized protein-based affinity chromatography offers a promising alternative for high-throughput drug screening, yet its effectiveness is often hampered by conventional immobilization techniques as they result in functional loss of the protein due to random orientation and multi-step purification. Herein, we developed a universal strategy for fabricating highly efficient chromatographic methods by integrating genetic code expansion with alkyne-azide click cycloaddition in the immobilization of dopamine D2 receptor (DRD2) and glucocorticoid receptor (GR) strain-promoted alkyne-azide cycloaddition (SPAAC). The site-specific incorporation of p -azidophenylalanine ( p AzF) into the two receptors using an engineered E. coli C321.ΔA strain enabled direct, oriented, and covalent immobilization of them onto dibenzocyclooctyne (DBCO)-functionalized silica by a copper-free click reaction without the need for prior purification. The resulting immobilized DRD2 p AzF and GR p AzF stationary phases were thoroughly characterized by XPS, SEM/EDS, and immunofluorescence, confirming successful immobilization and preserved receptor activity. These exhibited exceptional specificity, stability (>30 days), and reproducibility (RSD<1%). Applications immobilized DRD2 and GR in analyzing Wendan Decoction identified liquiritigenin, neohesperidin, and naringin as active components of the prescription. Among them, liquiritigenin, neohesperidin were characterized as dual-target ligands. Determination of their binding affinities to the receptors by frontal analysis enables calculation of drug-like efficiency indices (SEI, BEI, LLE). Cellular assays in HT22 cells further confirmed their bioactivities and target specificity to the receptors. This work is possible to advance the discovery of potential leads from natural products as it has the properties of streamlining the construction of reliable receptor chromatography methods, integrating the screening with binding affinity measurement, and preliminary drug-likeness evaluation.

Alkaline As(III)-contaminated wastewater, which is prevalent at tunnel-construction sites, poses a serious risk to the environment and human health. However, the existing research on the treatment of this alkaline wastewater is scarce. Herein, we proposed a treatment method based on a Fe(II)/CaSO 3 system enhanced by highly alkaline As(III)-contaminated wastewater (pH: ~12) itself. Mechanistic investigations revealed that alkaline conditions enhanced the process in two ways: (i) by accelerating the Fe(II)/Fe(III) cycle to sustain · OH and SO 4 · − radical generation and (ii) by promoting the reaction between M(Ca/Fe)–OH and the oxidation products of AsO 4 3− species. These dual effects promoted the formation of Ca 5 (AsO 4 ) 3 OH and FeAsO 4 precipitates that immobilized As(V). Furthermore, the Fe(II)/CaSO 3 system exhibited high resistance against interference from suspended solids and other coexisting substances. The system achieved >98 % removal of 30 mg·L −1 As(III) within 10 min at pH ~12. Notably, the final effluent pH value could be reduced to an ecofriendly value of ~8, and the residual SO 4 2− concentration was lower (140 mg·L −1 ) than that observed in common sulfur-based systems (e.g., peroxomonosulfate, peroxydisulfate, and sodium sulfite). This study presents a perspective shift in treating alkaline wastewater streams by viewing a dominant waste characteristic as a resource rather than as a barrier. Moreover, it provides an effective method for As(III) decontamination of alkaline wastewater.

In this study, mesoporous silica SBA-15 was modified by organic functional groups through silanization. Series of organosilane compounds were grafted onto the SBA-15, and the obtained functionalized carriers were then used to immobilize the lipase from Rhizomucor miehei (RML). The enzymatic properties of the obtained immobilized RML samples were evaluated, and the catalytic efficiencies in glycerolysis of triacylglycerols (TAG) reaction were studied. Compared with the parent SBA-15 immobilized RML, the organic modification gave a maximum improvement of enzymatic activity from 200.00 to 13211.11 U/g; in addition, TAG conversion and diacylglycerols (DAG) content increased from 21.28 to 84.24% and 15.45 to 59.03% respectively. The organic modification also decreased the sensitivity of immobilized RML in extreme pH values and increased their thermostability.

Phosphate materials (PMs) combine with phosphate solubilizing bacteria play an essential roles in lead (Pb) immobilization, but their resulting ability to reduce Pb bioavailability may vary depending on PMs used. In this study, Pseudomonas edaphica GAU-665 and three PMs: tricalcium phosphate, calcium phytate and nano-hydroxyapatite were respectively encapsulated into bio-beads by sodium alginate, which immobilization efficiency of Pb 2+ were 99.11%, 97.76% and 99.02% at initial Pb 2+ concentration of 200 mg L −1 , respectively. The Pb 2+ immobilization performance of bio-beads under different conditions and their organic acids secreted were examined. Most Pb 2+ was immobilized by bio-beads through combined functions of adsorption, precipitation, ion exchange and biomineralization, accompanied by the formation of more stable compounds such as Pb 3 (PO 4 ) 2 , Pb 5 (PO 4 ) 3 OH and Pb 5 (PO 4 ) 3 Cl. Meanwhile, pot experimental results indicated that the inoculation of CPhy (calcium phytate) bio-beads with PSB have highest biomass and root growth of oat ( Avena sativa L.) in Pb-stressed compared with CK, which increased the content of chlorophyll b (167.51%) in shoot. In addition, the CPhy bio-beads enhance the peroxidase, catalase activities and reduce the malondialdehyde content to alleviating lead physiological toxicity in oat, which reductions the Pb accumulation in shoot (52.06%) and root (81.04%), and increased the residual fraction of Pb by 165.80% in soil. These findings suggest the bio-beads combined with P. edaphica GAU-665 and calcium phytate is an efficient Pb immobilization material and provided feasible way to improve safety agricultural production and Pb-contaminated soil remediation.

Membrane fouling significantly limits the application of membrane separation technology in the treatment of textile dyeing wastewater. In this study, a loose nanofiltration membrane made of polybenzimidazole (PBI) with enhanced antifouling and high-efficiency dye/salt separation capabilities was developed through Ag@UiO-66-NH₂ immobilization and hydrophilic polymer brush grafting. Initially, Ag@UiO-66-NH₂ nanoparticles, possessing antibacterial properties, were incorporated into the PBI base membrane. Subsequently, a brush-like antifouling layer was constructed on the membrane surface by grafting linear polypropylene glycol (PPG) and polyethylene glycol (PEG). Experimental results revealed that the grafting modification substantially improved the membrane's performance: the rejection of the small-molecule dye Methyl Red increased from 16.8 % to 59.6 %, and the membrane surface hydrophilicity was enhanced, leading to a slight improvement in pure water flux. Additionally, the grafted layer effectively suppressed the adsorption of bovine serum albumin, with a flux recovery rate remaining above 90 % after four fouling cycles. Furthermore, the membrane exhibited rejection of <10 % for inorganic salts (Na 2 SO 4 , NaCl, MgSO 4 and MgCl₂), achieving efficient separation of dyes and salts. In tests with simulated textile dyeing wastewater, the membrane's rejection for industrial dyes remained stable at over 90 %, and the addition of surfactants and auxiliaries had no significant impact on its performance. This study shows that the combination of immobilizing MOF nanoparticles with controlled release properties of biocides and grafting hydrophilic polymer brush can effectively improve the separation efficiency and antifouling performance of the membrane, which provides a new method for the treatment of complex textile printing and dyeing wastewater.

The extraction of uranium (U) from wastewater is of vital importance for ensuring the sustainable and safe development of nuclear energy, as it addresses both environmental protection and resource recycling. Vanadium (V) and its compounds are extensively employed in iron and steel manufacturing, and their utility in pollutant remediation further promotes the circular use of V-bearing materials. However, studies on utilizing inexpensive and readily available vanadium oxides (e.g., V 2 O 5 ) for the treatment of highly acidic uranium-containing wastewater remain scarce. This study investigated the effectiveness of V 2 O 5 in removing U(VI) and immobilizing it into crystalline uranyl vanadates, providing the first confirmation of the feasibility of using V 2 O 5 for uranium elimination and capture. The results indicate that V 2 O 5 exhibits excellent extraction capacity and selectivity for uranyl ions in contaminated solutions, achieving a U(VI) extraction rate exceeding 97% even under highly acidic conditions ( e.g. , pH = 2.0). Adsorption equilibrium was attained within 250 min, and kinetic studies combined with model fitting indicated that the process was governed by both intra-particle diffusion and chemical adsorption mechanisms. It was discovered that the captured U(VI) was ultimately incorporated into a stable crystalline phase, identified as uvanite (U 2 V 6 O 21 ·15H 2 O), through a coprecipitation pathway, revealing a novel crystallization mechanism. The overall immobilization process was attributed to the synergistic effects of chemical adsorption and coprecipitation. These findings support the development of vanadium oxide-based technologies for U(VI) purification and recovery from complex wastewater.

Immobilized proteins have been conceived as promising for developing assays, including biosensors and chromatographic methods. Conventional efforts are geared toward the attachment of purified or recombinant protein, while lacking strategies for capturing endogenous protein in live cells. Herein, we developed a rapid, two-step method for the target- and site-selective immobilization of endogenous natural γ-amino butyric acid receptors (GABAAR) directly from live hippocampal neuronal cells (HT22 cells). The method employed a ligand-tethered reagent as a recognition motif and a derivative of acyl-imidazole as an alkyne donor, which selectively introduces an alkyne click handle into specific sites of GABAAR. Subsequent click reaction between the alkyne-protein and azide-modified dye or support surface enables visualization of the receptor in live cells, as well as fabrication of GABAAR-immobilized surface plasmon resonance (SPR) sensor chips or affinity chromatography stationary phases. Compared to a typical affinity-driven one-step capture method, this approach significantly shortens the immobilization time from 6 h to 15 min while preserving receptor functionality. The immobilized endogenous GABAAR was successfully applied in two analytical platforms: as an SPR sensor chip for enhancing sensitivity in revealing drug-receptor interactions, and as a stationary phase in affinity chromatography, where it markedly improved resolution, selectivity, and accuracy in analyzing drug-receptor interactions. Such unique features indicate that the two-step strategy may be a versatile tool for modifying endogenous natural proteins in live cells while minimizing the influence on their functions. It has great potential to facilitate the construction of affinity analytical assays using immobilized endogenous proteins.

This study presents a novel FeMg-layered double hydroxide (LDH) modified biochar composite (LB) designed to synergistically address phosphate recovery from wastewater and heavy metal immobilization in contaminated soil. The composite exhibits an exceptional phosphate adsorption capacity of 25.02 mg/g, representing a 169.00 % increase compared to pristine biochar. The improved performance results from precipitation and anion exchange within the LDH layers. And the competitive adsorption tests further confirm that LB better retains phosphate in simulated wastewater. When repurposed as a soil amendment, phosphate-laden LB (P-LB) demonstrates remarkable heavy metal immobilization efficiencies, achieving 41.23 % Pb, 30.52 % Cu and 25.63 % As immobilization at a 1.0 % dosage, representing an 18.70–26.42 % improvement over LB. Beyond its metal stabilization capabilities, P-LB effectively enhances soil phosphorus cycling, increasing total phosphorus by 14.41 % and bioavailable phosphorus by 9.02 %. Furthermore, the remediated soil exhibits significantly enhanced enzymatic activity, with urease and sucrase activities increasing by 21.38 % and 380.10 %, respectively. Pot experiments confirm the agronomic benefits of P-LB, demonstrating a 68.16 % increase in plant height and a 39.27 % increase in biomass accumulation in the remediated soil. This integrated approach, leveraging LDH-phosphate synergy, not only achieves phosphate recovery in sewage but also realizes metal immobilization and soil fertility remediation in contaminated soil.

A feasible and green sol-gel method is proposed to fabricate well-distributed nano-particulate Fe-Ni2P incorporated in N, P-codoped porous carbon nanosheets (Fe-Ni2P@N,P-CNSs) using biomass agarose as a carbon source, and ethylenediamine tetra (methylenephosphonic acid) (EDTMPA) as both the N and P source. The doped Fe in Ni2P is essential for a substantial increase in intrinsic catalytic activity, while the combined N,P-containing porous carbon matrix with a better degree of graphitization endows the prepared Fe-Ni2P@N,P-CNSs catalyst with a high specific surface area and improved electrical conductivity. Benefiting from the specific chemical composition and designed active site structure, the as-synthesized Fe-Ni2P@N,P-CNSs manifests a satisfying catalytic performance toward both oxygen reduction reaction (ORR) and oxygen evolution reaction (OER) in an alkaline solution, with low overpotential, small Tafel slope and long-term durability, relative to the counterparts (Fe-free Ni12P5/Ni2P2O7@N,P-CNSs and CNSs) with single components and even comparable to Pt/C and RuO2 catalysts. The present work broadens the exploration of efficient bifunctional oxygen electrocatalysts using earth abundant biomass as carbon sources based on non-noble metals for low cost renewable energy conversion/storage.

Cell wall is the major component of root apoplast which is the main reservoir for iron in roots, while nitric oxide (NO) is involved in regulating the synthesis of cell wall. However, whether such regulation could influence the reutilization of iron stored in root apoplast remains unclear. In this study, we observed that iron deficiency elevated NO level in tomato ( Solanum lycopersicum ) roots. However, application of S-nitrosoglutathione, a NO donor, significantly enhanced iron retention in root apoplast of iron-deficient plants, accompanied with a decrease of iron level in xylem sap. Consequently, S-nitrosoglutathione treatment increased iron concentration in roots, but decreased it in shoots. The opposite was true for the NO scavenging treatment with 2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (cPTIO). Interestingly, S-nitrosoglutathione treatment increased pectin methylesterase activity and decreased degree of pectin methylation in root cell wall of both iron-deficient and iron-sufficient plants, which led to an increased iron retention in pectin fraction, thus increasing the binding capacity of iron to the extracted cell wall. Altogether, these results suggested that iron-deficiency-induced elevation of NO increases iron immobilization in root apoplast by decreasing pectin methylation in cell wall.

Cascade reactions catalyzed by two or more enzymes have been widely used in industrial production and exhibited many advantages over the single-enzyme catalytic system. In this study, two components of hydroxylase monooxygenase (HpaBC) were first co-immobilized by Ni 2+ -nitrilotriacetic acid (Ni-NTA) functionalized magnetic silica nanoparticles (Ni-NTA/H 2 N-SiO 2 @Fe 3 O 4 ) for enhancing the stability and activity of biocatalysts with multi-components. These two components, HpaB and HpaC, were modified with histidine-tag and employed to construct a bi-enzyme catalytic system. After co-immobilization, the activity of the bi-enzyme system was 2.6 times of free enzymes. Meanwhile, the co-immobilized system was more stable against high temperature and alkaline condition, and maintained 76.6% of the initial activity for storage 12 days. Moreover, the co-immobilized HpaBC remained more than 60% catalytic activity after 7 cycles. These results showed that co-immobilized multi-component enzymes based on functionalized magnetic nanoparticles without purification would play a great potential role in the field of industrial biocatalysis.
