NIR II light-driven nanomotor synergistically enhances immunogenic cell death through photothermal and chemodynamic therapy for melanoma immunotherapy

Xuan Hu,Ziyan Shen,Huahua Hao,Ke Ma,Aihua Zhen,Yibo Yang,Kai Liang,Zelong Chen,Jiaxuan Li,Yinghua Lv,Shuang Chao,Yuxin Pei,Zhican Qu,Zhichao Pei

Melanoma is a highly invasive and metastatic malignant skin tumor. Recently, immunogenic cell death (ICD) has attracted great attention as a promising approach to immunotherapy. However, efficiently and comprehensively activating ICD throughout the dense tumor tissue is a key challenge. Herein, we designed a NIR II light-driven asymmetric nanomotor drug delivery system (Sor@CS-ZIF-8@MO1) to achieve deep penetration into the tumor tissue. By combining photothermal therapy (PTT) and chemodynamic therapy (CDT) to synergistically induce ICD, the immunotherapeutic efficacy against melanoma is enhanced. The research results showed that Sor@CS-ZIF-8@MO1 exhibited good photothermal performance and motor-driven performance, and was able to effectively penetrate 3D tumor cell spheroids deeply. Sor@CS-ZIF-8@MO1 targeted tumor tissues through mannose and controllably released sorafenib under the low pH conditions in tumor tissues and photothermal stimulation, thereby promoting tumor tissue angiogenesis to improve its hypoxic microenvironment and effectively enhancing the CDT effect induced by Cu +/2+ . This could synergistically enhance the ICD of tumor cells with the PTT. Meanwhile, the tumor-associated antigens released by ICD, together with ovalbumin and mannose, stimulated immune response, reshaped the tumor immune microenvironment, enhanced tumor immunity, and ultimately effectively inhibited the growth and metastasis of melanoma tumors. In this work, a nanomotor delivery system that integrates multiple modalities and is capable of deeply penetrating tumor tissues to efficiently and comprehensively induce immunogenic cell death (ICD) has been designed, providing a new strategy to address the problem of insufficient induction of ICD in melanoma immunotherapy.

n-Hexyl Side Chain Engineering: Crafting NIR-II AIEgens with Balanced Fluorescence-Photothermal Performance for Precision Tumor Ablation

Binbin Wu,Qin Zhang,Min Chen,Hongchen Zhang,Lulu Zheng,Xingyu Peng,Shihua Wang,Yun-Yun Quan,Lingtian Wang,Zu-Sheng Huang,Xiaoxia Ye

Photothermal therapy (PTT) has gained significant attention as a promising cancer treatment due to its low systemic toxicity, minimal invasiveness, and other advantages. Aggregation-induced emission luminogens (AIEgens) with second near-infrared (NIR-II) fluorescence have emerged as ideal photothermal agents (PTAs), offering key benefits such as high biosafety, excellent biocompatibility, and precise tumor theranostics. Nevertheless, achieving an optimal balance between radiation-mediated NIR-II fluorescence imaging and non-radiation NIR-PTT poses a considerable challenge in designing efficient NIR-II AIEgens. In this work, we employed side chain engineering by strategically grafting n -hexyl onto the π-bridge of a conjugated D-π-A-π-D structure, leading to the development of four new AIEgens ( BQ1 ∼ BQ4 ). These compounds feature 6,7-diphenyl- [1,2,5] thiadiazolo [3,4- g ] quinoxaline as the acceptor, thiophene as the π-bridge, and phenothiazine as the donor. This molecular design enables precise regulation of backbone distortion, effectively balancing fluorescence emission and photothermal conversion. Among them, BQ1 , BQ3 , and BQ4 exhibited strong fluorescence, longer emission wavelengths, and superior photothermal conversion efficiencies (PCEs) (59.2%, 57.8%, 52% respectively), whereas BQ2 , with greater molecular distortion, showed a lower PCE of 33.4%. Notably, BQ1 nanoparticles demonstrated outstanding photothermal conversion, prolonged tumor retention, strong resistance to photobleaching in vitro and in vivo , and NIR-II imaging capability at 1055 nm. This study introduces a novel strategy for optimizing NIR-II/AIE PTAs through precise conformational control via strategic n -hexyl positioning, which remains unreported to date.

Nir-driven plasmon-enhanced composite upconversion photocatalyst β-NaLuF4@NaLuF4:Yb3+, Tm3+@Ag@BiOBr for photocatalytic pollutant degradation

Chen Zhang,Chuan Ding,Mingyue Li,Jinyang Zhou,Tao Yuan,Zhemin Shen

The infrared energy of the sun’s rays accounts for more than 40 %. Heterostructures were constructed to enable broad-spectrum response of photocatalysts, and enhance the utilization of sunlight, especially that of infrared lights. A robust heterostructured upconversion composite material decorated with Ag nanoparticles (AgNPs), β-NaLuF 4 @ NaLuF 4 :Yb 3+ ,Tm 3+ @ AgNPs@BiOBr (NLF@NLFT@Ag@Bi), was built through a multi-step hydrothermal method. NLF@NLFT@Ag@Bi demonstrates enhanced photocatalytic activity under UV–Vis-NIR, Vis-NIR, and NIR irradiation compared to pure BiOBr and NLF@NLFT@Ag/BiOBr mixture, achieving 99 % Rhodamine B (RhB) degradation in 0.5 h (UV–Vis-NIR) and 96 % in 3 h (NIR), with degradation over 80 % for Bisphenol A (BPA), Norfloxacin (Nor), and Methyl Orange (MO) in 3 h (UV–Vis-NIR). The upconversion luminescence (UCL) of NLF@NLFT@Ag is tuned by its core–shell structure and AgNPs’ plasmonic effect, while NLF@NLFT@Ag@Bi’s enhanced photocatalysis arises from boosted UCL and AgNPs-facilitated Förster resonance energy transfer (FRET) between NLFT and BiOBr. The material’s efficacy in infrared-driven photocatalysis offers a prospective approach for extending catalyst response ranges. It greatly enhances degradation effects of organic pollutants under sun’s rays.

A Photosensitizer-Engineered Nanoplatform Exposed to NIR Laser for Mitochondria Targeting and Chemo/Photodynamic Synergistic Therapy against Glioma

Fengtian Zhang,Weihong Luo,Zhongjie Min,Darong Lu,Weiliang Chen,Lijuan Wen

Targeted mitochondrial-based therapeutic strategies for tumors have emerged as a significant research focus in the field of cancer treatment in recent years. Mitochondria, often referred to as the “energy factory” of cells, play a crucial role in the metabolic reprogramming of tumor cells, as well as in oxidative stress and immune evasion. In this study, we utilized a mitochondria-targeted photosensitizer, IR780, to engineer Celastrol (Cela)-loaded lipid nanoparticles, designed as Cela@Lip-IRH, for the synergistic therapy of chemotherapy and photodynamic therapy (PDT) against glioma. Cela@Lip-IRH could respond to the weakly alkaline environment of tumor cell mitochondria for rapid Cela release. Leveraging the mitochondrial targeting capacity and PDT property of IR780, Cela@Lip-IRH nanoparticles showed enhanced cellular uptake and mitochondria targeting distribution in vitro, as well as obvious tumor accumulation in vivo. Cela@Lip-IRH nanoparticles exhibited prominent intracellular reactive oxygen species (ROS) generation upon NIR laser irradiation, which was accompanied by pronounced photocytotoxicity and cell apoptosis, finally producing remarkable tumor growth suppression efficacy in vivo. We further verified that the potential antitumor molecular mechanism was triggered by the release of Cytochrome C, leading to an activated mitochondrial apoptosis pathway, alongside an immune response characterized by enhanced recruitment of CD4+ T cells and CD8+ T cells in tumor tissues. Currently, this mitochondria-targeted nanoplatform presents an ideal method to enhance the synergistic performance of chemotherapy and PDT in glioma, and it is anticipated that such a strategy will provide a promising alternative to optimize the tumor-targeted therapies.

Fabrication and characterization of BiOBr:Yb3+,Er3+/g-C3N4 p-n junction photocatalysts with enhanced visible-NIR-light-driven photoactivities

Shuhui Liang,Manman He,Jingjing Guo,Jie Yue,Xipeng Pu,Bo Ge,Wenzhi Li

The BiOBr:Yb 3+ ,Er 3+ /g-C 3 N 4 (BYE/CN) p-n junction photocatalysts were successfully fabricated by a stepwise method. Experimental results show that Er 3+ /Yb 3+ were successfully doped into BiOBr lattice. As-obtained BYE/CN exhibited strong NIR light absorption and upconversion emission from NIR to UV–visible due to the upconversion transition of Er 3+ . The p-n junction was constructed due to the appropriate alignment band structure of BYE and g-C 3 N 4 , which distinctly suppresses the recombination rate of electron-hole pairs. Compared with pure BYE and CN, as-synthesized BYE/CN samples exhibit distinctly improved photoactivities under the irradiation of visible or NIR light, which can be attributed to the upconversion effect of Er 3+ and the construction of p-n junction. This result may provides a promising system for efficient harvest of the NIR light, which will contribute to the utilization of solar energy in the future.

In Situ Formation of Leaf-Like Ag2S/CdS Heterojunction Photocatalyst Harnessing Vis–NIR Light for Photodegradation of Organic Pollutants

Chang Libin,Zhu Gangqiang,Lu Huan,Hojamberdiev Mirabbos,Zhang Zengping,Gao Jianzhi,Wang Chenghui,Yang Woochul

Abstract The Ag 2 S/CdS composite photocatalyst with a leaf-like structure was synthesized by loading the Ag 2 S nanoparticles on the surface of the hydrothermally synthesized CdS particles. The UV–Vis–NIR absorption spectroscopy results reveal that the Ag 2 S/CdS composite photocatalyst exhibits a wide-spectrum response from ultraviolet (UV) to near-infrared (NIR), demonstrating its potential for effective utilization of solar energy. The scanning and transmission electron microscopic observations show that Ag 2 S nanoparticles with the size of about 20 nm are successfully embedded on the surface of leaf-like structure of CdS. Among the samples loaded with different molar percentages of Ag 2 S nanoparticles, the 5%-Ag 2 S/CdS composite photocatalyst exhibits a higher photocatalytic efficiency than the individually synthesized CdS and Ag 2 S for the degradation of Rhodamine B (RhB) and methyl orange (MO) under LED visible light irradiation. Furthermore, the 5%-Ag 2 S/CdS composite photocatalyst also exhibits a photocatalytic activity for the degradation of RhB under 850 and 940 nm monochromatic light irradiation. The results of the photocatalytic activity tests performed with various scavengers confirm that ·O 2 − plays a key role in the photodegradation of RhB. An enhanced photocatalytic performance of the Ag 2 S/CdS composite photocatalyst is directly attributed to the formation of a heterojunction between Ag 2 S and CdS. The developed composite photocatalyst can harness a greater share of the solar spectrum from visible to near infrared region and generate the photoinduced charge carriers more efficiently. Three-dimensional Ag2O/Bi5O7I p–n heterojunction photocatalyst harnessing UV–vis–NIR broad spectrum for photodegradation of organic pollutants

Yannan Chen,Gangqiang Zhu,Mirabbos Hojamberdiev,Jianzhi Gao,Runliang Zhu,Chenghui Wang,Xiumei Wei,Peng Liu

Ag 2 O nanoparticles-loaded Bi 5 O 7 I microspheres forming a three dimensional Ag 2 O/Bi 5 O 7 I p – n heterojunction photocatalyst with wide-spectrum response were synthesized in this study. The results of transmission electron microscopy observations revealed that the Ag 2 O nanoparticles with the diameter of ca. 10–20   nm were distributed on the surfaces of Bi 5 O 7 I nanosheets. The as-synthesized Ag 2 O/Bi 5 O 7 I exhibited an excellent wide-spectrum response to wavelengths ranging from ultraviolet (UV) to near-infrared (NIR), indicating its potential for effective utilization of solar energy. Compared with pure Bi 5 O 7 I, the Ag 2 O/Bi 5 O 7 I composite also demonstrated excellent photocatalytic activity for the degradation of Bisphenol A and phenol in aqueous solution under visible LED light irradiation. Among samples, the 20% Ag 2 O/Bi 5 O 7 I composite photocatalyst showed the highest photocatalytic activity for the degradation of Bisphenol A and phenol in aqueous solution. In addition, the 20% Ag 2 O/Bi 5 O 7 I composite also exhibited a photocatalytic activity for the degradation of Bisphenol A under NIR light irradiation. The improved photocatalytic activity is attributed to the formation of a p - n heterojunction between Ag 2 O and Bi 5 O 7 I, allowing the efficient utilization of solar energy (from UV to NIR) and high separation efficiency of photogenerated electron-hole pairs. The present work is desirable to explore a possible avenue for the full utilization of solar energy.

A novel TMTP1-modified theranostic nanoplatform for targeted in vivo NIR-II fluorescence imaging-guided chemotherapy for cervical cancer

Nuernisha Alifu,Rong Ma,Lijun Zhu,Zhong Du,Shuang Chen,Ting Yan,Gulinigaer Alimu,Linxue Zhang,Xueliang Zhang

Near-infrared II (NIR-II, 900–1700 nm) fluorescence bioimaging with advantages of good biosafety, excellent spatial resolution, high sensitivity, and contrast has attracted great attention in biomedical research fields. However, most of the nanoprobes used for NIR-II fluorescence imaging have poor tumor-targeting ability and therapeutic efficiency. To overcome these limitations, a novel NIR-II-emissive theranostic nanoplatform for fluorescence imaging and treatment of cervical cancer was designed and prepared. The NIR-II-emissive dye IR-783 and chemotherapy drug doxorubicin (DOX) were encapsulated into liposomes, and the tumor-targeting peptide TMTP1 (a polypeptide with a sequence of cyclic ASN Val Val Arg Gln Cys) was conjugated to the surface of the liposomes to form IR-783-DOX-TMTP1 nanoparticles (NPs) via self-assembly methods. The IR-783-DOX-TMTP1 NPs showed strong NIR-II emission, excellent biocompatibility and a long lifetime in vivo. Furthermore, high-definition NIR-II fluorescence microscopy images of ear blood vessels and intratumoral blood vessels were obtained from IR-783-DOX-TMTP1 NP-stained mice with high spatial resolution under 808 nm laser excitation. Moreover, IR-783-DOX-TMTP1 NPs showed strong tumor-targeting ability and highly efficient chemotherapeutic characteristics towards cervical tumors. The novel targeting and NIR-II-emissive IR-783-DOX-TMTP1 NPs have great potential in diagnosis and therapy for cervical cancer.

Facile one-step synthesis of NIR-Responsive siRNA-Inorganic hybrid nanoplatform for imaging-guided photothermal and gene synergistic therapy

Xiuna Jia,Mengmeng Lv,Yunwei Fei,Qing Dong,Hao Wang,Qiong Liu,Dan Li,Jin Wang,Erkang Wang

Diagnosis-guided synergistic treatment based on innovative nanomaterials is of great significance for the development of anti-cancer therapies. However, the low delivery efficiency of therapeutic gene and the inability to trigger release on demand are still major obstacles impeding its wide application. Herein, we report an ultra-fast one-step method within 2 min to prepare a smart carrier, liposome-coated Prussian blue @ gold nano-flower, which is named LPAR after linking with tumor-targeting peptide. The versatile LPAR not only can respond to near-infrared (NIR) light, achieve the selective delivery and the controlled release of siRNA targeting the mutant gene of Kras at its codon-12 from Glycine (G) to Aspartic acid (D) (named as G12D mutant gene) in the malignant pancreatic tumors, but also efficiently convert the absorbed NIR light into the heat to realize gene-photothermal synergistic therapy both in vitro and in vivo . Theoretical simulation results reveal that the outstanding photothermal conversion efficiency of LPAR is mainly due to its higher electric field intensity and power density distributions . Furthermore, the LPAR possesses the capabilities for triple-modal imaging. Therefore, the developed NIR light-responsive LPAR has the potential to be served as a tumor-targeted nano-delivery system for imaging-guided synergistic therapy of cancers.

Colorimetric and NIR Fluorescence Probe with Multiple Binding Sites for Distinguishing Detection of Cys/Hcy and GSH in Vivo

Kangming Xiong,Fangjun Huo,Jianbin Chao,Yongbin Zhang,Caixia Yin

Although some progress has been made in distinguishing the detection of biothiols, NIR biothiol fluorescent probes for simultaneously distinguishing detection of Cys, Hcy, and GSH in vivo have not been reported. The design of these probes involves the introduction of NIR fluorophores and multiple binding sites; thus, the integrated design of probes remains a challenge. Although Cys, Hcy, and GSH have common functional groups, a sulfydryl group and an amino group, due to their differences in spatial structure, they may react with multiple binding site probes to produce different reaction products in different bonding mechanisms, resulting in different colors and fluorescent signal changes of the system. Therefore, multiple binding site fluorescent probes can realize their discrimination detection. For an NIR fluorescent probe, it is easier to realize in vivo imaging to promote the research of biothiols in clinical diagnosis. In our work, not only were multiple binding sites constructed in the compound but also NIR fluorophores were introduced. This enables the probe to not only efficiently distinguish detection of Cys/Hcy and GSH but also achieve fluorescence imaging in vivo. We believe this result is a milestone in the discrimination detection of biothiols.

Spectral broadening and enhanced ultra-broadband emission in Ca3TiTaGa3O12:Cr3+, Yb3+ co-doped phosphor for NIR pc-LED applications

Yichen Yang,Xiaojiao Kang,Qiwen Pan,Zhennan Zhu,Wei Lü

Broadband near-infrared (NIR) phosphor-converted light-emitting diodes (pc-LEDs) are gaining significant interest due to their potential in spectroscopic applications. NIR phosphors play a critical role in pc-LED devices by converting visible light into NIR emission with high stability and tunability. In this study, an efficient broadband NIR phosphor, Ca 3 TiTaGa 3 O 12 :Cr 3+ , is developed and systematically investigated. Ca 3 TiTaGa 3 O 12 :Cr 3+ exhibits a broad NIR emission band centered at 790 nm with a full width at half maximum (FWHM) of 202 nm upon 450 nm blue light excitation. The emission bandwidth is further expanded to 303 nm by introducing an optimal amount of Yb 3+ co-doping, which produces a merged ultra-broadband Cr 3+ –Yb 3+ emission. Luminescence and decay kinetic analysis reveal that the resultant ultra-broadband emission originates from effective energy transfer Cr 3+ to Yb 3+ . Moreover, the Yb 3+ co-doping enhances the internal quantum efficiency from 30.5% to 41.3%, and improves thermal stability with emission retention increasing from 42.6% to 57.4% at 150 °C. The Ca 3 TiTaGa 3 O 12 :Cr 3+ and Ca 3 TiTaGa 3 O 12 :Cr 3+ , Yb 3+ phosphors are successfully integrated with a 450 nm blue LED chip, demonstrating their potential for practical NIR pc-LED device applications in night vision and bioimaging.

Metabolizable van der Waals enhanced Raman scattering probes for CXCR4-targeted imaging guided cancer therapy in NIR-II window

Xinru Chen,Xiaoyan Wang,Jin Li

Raman spectroscopy in the second near-infrared (NIR-II, 1000–1700 nm) window has received ever-increasing attention in intraoperative tumor imaging due to its high specificity and penetration depth. However, the overwhelming majority of existing NIR-II Raman probes are fabricated by SERS with substrate enhancement, which raises serious biosafety concerns. Herein, we present van der Waals (vdW) enhanced Raman scattering (vdW-ERS) effect based on intermolecular charge transfer of dithiolene to develop ultrabright metal-free NIR-II Raman probes. The intermolecular charge transfer within dithiolene through vdW stacking is well illustrated by independent gradient model and ultrafast spectroscopy, which significantly boosts the intrinsic Raman properties with ultrahigh scattering cross-section (1.59 × 10 −19  cm 2 ) as well as the photothermal conversion efficacy (60.4%) through an additional femtosecond-scale non-radiative decay channel. Moreover, the fluorescence-free property of dithiolene greatly inhibits the spectral backgrounds and improves the photostability. In the tumor models of triple-negative breast cancer, vdW-ERS probes enable intraoperative CXCR4-targeted Raman-guided surgical resection and postoperative hyperthermia ablation of the residual microscopic tumors, resulting in approximately 100% survival rate. Importantly, vdW-ERS probes are hepatically excreted (ca. 92% excretion within two months), in stark contrast to accumulation in the liver for typical gold substrate-enhanced counterparts. Free from fluorescence background and photobleaching, intermolecular vdW-ERS effect presents a new mechanism for fabricating charge transfer Raman probes with the potential for clinical translation.
