Application of curcumin in clinic was limited due to its poor solubility, stability and low bioavailability. The purpose of this study is to investigate the efficacy of some common excipients including Poloxamer 188 , polyvinyl pyrrolidone K30 (PVP K30), Tween 80 , Gelucire 4414 and polyethylene glycol 400 (PEG 400) for overcoming these limitations of curcumin. Results showed that these excipients were efficient in improving the solubility and stability of curcumin and moreover significantly increased curcumin absorption in a concentration-dependent manner after their administrations to rat lungs. Studies of pulmonary membrane damage evaluated by measuring amount of protein and activity of lactate dehydrogenase (LDH) released from rat lung demonstrated that these excipients at studied concentrations did not cause obvious membrane damage to lung tissues. The bio-efficacy studies showed that combinatorial treatment of curcumin and PEG 400 (45%, v/v) resulted in a therapeutic effect on the sepsis-induced acute lung injury (ALI) of rats, characterized by obvious reduction of lung W/D weight ratio, protein content, MPO activity, and alleviated pathological symptoms of lung tissues. These findings suggested that intrapulmonary delivery of curcumin combined with these excipients, especially 45% (v/v) PEG 400, was promising for treating pulmonary diseases including ALI.

Background/Aims: This study investigated the underlying mechanisms of the antidepressant effects of curcumin and dexanabinol-loaded solid lipid nanoparticles in corticosterone-induced cell and mice depression models. Methods: Curcumin and dexanabinol-loaded solid lipid nanoparticles (Cur/SLNs-HU-211) were synthesized via an emulsifcation and low-temperature solidification method. Antidepressant activities of nanoparticles in a corticosterone-induced major depression model were investigated by MTT assay, cellular uptake by flow cytometry, behaviour by Forced Swimming Test and rotarod test, neurotransmitters by High Performance Liquid Chromatography, Western blotting, qPCR and immunofluorescence. Results: Treatment with Cur/SLNs-HU-211 induced greater dopamine (DA)/5-hydroxytryptamine (5-HT) release with reduced corticosterone-induced apoptotic cell death in PC12 cells. Additionally, in vivo Cur/SLNs-HU-211 significantly induced recovery from depressive behaviour with increased DA/5-HT levels, CB1 mRNA levels and CB1, p-MEK1 and p-ERK1/2 protein expression levels in the hippocampus and striatum. Cur/SLNs-HU-211 improved CB1 expression and inspired the proliferation of astrocytes in the hippocampus and striatum, exerted neuroprotective effects by preventing corticosterone -induced BDNF/NeuN expression reduction. Conclusion: Our study implies that Cur/SLNs-HU-211 may be a useful approach for treatment of major depression

Artificial graft serves as the primary grafts used in the clinical management of sports-related injuries . Until now, optimizing its graft-host integration remains a great challenge due to the excessive inflammatory response during the inflammatory phase, coupled with an absence of tissue-inductive capacity during the regeneration phase. Here, a multi-layered regenerated silk fibroin (RSF) coating loaded with curcumin (Cur) and Zn 2+ on the surface of the PET grafts (Cur@Zn 2+ @PET) was designed and fabricated for providing time-matched regulation specifically tailored to address issues arising at both inflammatory and regeneration phases, respectively. The release of Cur and Zn 2+ from the Cur@Zn 2+ @PET followed a time-programmed pattern in vitro. Specifically, cellular assays revealed that Cur@Zn 2+ @PET initially released Cur during the inflammatory phase, thereby markedly inhibit the expression of inflammatory cytokines TNF-a and IL-1β. Meanwhile, a significant release of Zn 2+ was major part during the regeneration phase, serving to induce the osteogenic differentiation of rBMSC. Furthermore, rat model of anterior cruciate ligament reconstruction (ACLR) showed that through time-programmed drug release, Cur@Zn 2+ @PET could suppress the formation of fibrous interface (FI) caused by inflammatory response, combined with significant new bone (NB) formation during regeneration phase. Consequently, the implementation of the Cur@Zn 2+ @PET characterized by its time-programmed release patterns hold considerable promise for improving graft-host integration for sports-related injuries.

Ultrasonic assisted acetic acid hydrolysis was applied to prepare starch nanocrystals (SNCs) from native starches with different crystalline structures (A, B, and C types). The structure properties, morphology, Pickering emulsion stability and curcumin deliver capacity of both SNCs and native starches were investigated and compared. Compared with native starches, SNCs showed smaller size and higher crystallinity. The size of SNCs varied with different crystalline types, with C-type starch exhibiting the smallest SNCs (107.4 nm), followed by A-type (113.8 nm), and B-type displaying the largest particle size (149.0 nm). SNCs-Pickering emulsion showed enhanced stability with smaller emulsion droplets, higher static stability, and denser oil/water interface. SNCs-Pickering emulsions displayed higher curcumin loading efficiency (53.53 %–61.41 %) compared with native starch-Pickering emulsions (13.93 %–19.73 %). During in vitro digestion, SNCs-Pickering emulsions proved to be more proficient in protecting and prolonging the biological activity of curcumin due to their smaller size and better interfacial properties. These findings demonstrated the potential of SNCs for application in Pickering emulsion and delivery of bioactive components.

Curcumin (Cur) is renowned for its antioxidant and anti-inflammatory properties; however, its application faces many challenges, including poor bioavailability and low solubility. Alginate (SA) gel system is a promising platform for the delivery of Cur, but its efficacy remains limited and requires further improvement. In this study, Cur/oil was encapsulated into water-in-oil nanoemulsions, which were subsequently incorporated into SA microparticles modified with chitosan (CS) or inulin (IN) to fabricate four types of microparticles: Cur/SA, Cur/SAIN, Cur/SACS, and Cur/Mix. Their characteristics were analyzed, including particle size and morphology, and their stability and release properties were assessed using in vitro models. It demonstrated that the incorporation of IN and CS slightly improved the encapsulation efficiency and stability of SA-based microparticles in simulated human upper digestive tract conditions. The in vitro results further indicated that the incorporation of IN and CS enhanced the release of Cur from SA microparticles in simulated intestinal conditions through pH- and biodegradation-dependent mechanisms. Moreover, Cur/Mix exhibited a longer residence time in the small intestine and colon compared to the other microparticles. The four microparticles exerted distinct effects on the composition of gut microbiota, with Cur/Mix enhancing microbiota richness and diversity. Furthermore, these microparticles altered microbiota metabolite profile, particularly amino acid metabolism, with Cur/Mix exhibiting a more pronounced effect on arginine biosynthesis. This study suggests that incorporating IN and CS into SA microparticles not only improves their physicochemical properties but also achieves a synergistic effect between Cur and polysaccharides.

Polylactic acid (PLA), as a biodegradable material, is considered an ideal alternative to petroleum-based plastics. The intrinsic mechanical fragility and lack of multifunctional characteristics significantly constrain its practical utilization in advanced applications. This study designed and prepared a ternary PLA composite material with uniform surface and enhanced functionality by uniformly dispersing esterified CNC (E n CNC) Curcumin (Cur) hybrid nanostructures in the polymer matrix. The synergistic interaction between the E n CNC and Cur significantly enhanced their interfacial compatibility with the PLA matrix, leading to uniform dispersion and significantly improved flexibility. The flexibility of resulting E n CNC_Cur_PLA composite films was significantly improved, with elongation at break increased to 64.42 %, representing a 628.59 % increase compared to pure PLA. The addition of EnCNC increased the bioavailability of Cur and demonstrated multifunctional effects, including controlled release of Cur in ethanol, enhanced antioxidant activity (80.6 % DPPH clearance) and antibacterial activity (over 99 % against Escherichia coli, 87.7 % against Staphylococcus aureus ). Overall, these results highlight a simple and effective strategy to fabricate multifunctional, sustainable bio-based films with improved mechanical performance and biological activity, suitable for advanced food packaging applications.

Cold plasma (CP) modification, combined with nanotechnology, has demonstrated significant potential for enhancing the performance of bio-based packaging films. In this study, zein-curcumin-quercetin-chondroitin sulfate nanoparticles (ZCQC NPs) were incorporated into a chitosan film and modified by CP (0, 10, 30, 50 kV) to enhance the physicochemical properties, bioactivity, and preservation ability of the chitosan-based film. Fourier transforms infrared spectrometer (FT-IR) and X-ray diffraction (XRD) analyses confirmed the presence of hydrogen bonding and electrostatic interactions between the NPs and chitosan, as well as their good compatibility. Thermogravimetric analysis revealed that CP treatment synergistically enhanced the thermal stability of NPs-incorporated composite films. The incorporation of CP and NPs enhanced the film's mechanical strength, hydrophobicity, and barrier performance synergistically. Among them, the CP-C-NPs 30 kV film exhibited lower water vapor permeability (1.33 ± 0.15 g/m·d) and oxygen permeability (4.16 ± 0.27 × 10 −6 g/m·d·Pa), along with excellent antioxidant and antibacterial properties. More importantly, the CP-C-NPs 30 kV film significantly reduced the total viable count (13.64 % compared to the control) and the degree of lipid oxidation (TBARS value decreased by 42.86 %) in packaged fish, extending the shelf life at 4 °C to 8 days, which was twice that of ordinary packaging film. This study not only reveals the synergistic enhancement mechanism of CP modification on the performance of active packaging materials but also provides new strategies for developing efficient food preservation materials.

Natural products showed many benefits for human health and are important resources in drug discovery and herbal medication development. However, their pharmacological activities are often constrained by their poor water solubility. To address these challenges, various drug delivery systems (DDS) have been developed to enhance the physicochemical properties of drugs, thereby improving their pharmacological effects or reducing adverse effects. In this study, we reported the construction of a novel natural products-based DDS through a self-assembly engineering approach, in which curcumin (Cur) was encapsulated by phosphorylated capsaicin (Cap-p). The physicochemical properties, drug loading and release behavior as well as therapeutic effects of Cap-p and Cap-p@Cur were investigated in detail. Results demonstrate that both Cap-p and Cap-p@Cur can self-assemble into nanovesicles and exhibit high water dispersibility. Results from biological assays suggested Cap-p exerts favorable biocompatibility toward MDA-MB-231 cells at concentration of 40 µg/mL. Compared to free Cur and Cap-p, Cap-p@Cur displays obviously enhanced cell killing capability against MDA-MB-231 cells and its cell killing ability was significantly amplified upon light irradiation. In addition, Cap-p displayed obvious bone-targeting capability, indicating its potential for eradicating bone-metastasis of triple-negative breast cancer (TNBC). The above results suggest that Cap-p@Cur holds considerable promise in treatment of TNBC via combining chemotherapy and photodynamic therapy. This work will provide some valuable insights into construction of natural product-based carrier-free DDS.

Combination therapy has been proved to be an effective strategy to inhibit metastasis, however, its efficacy is always compromised by the poor delivery efficiency of drugs. In this study, multi pH-sensitive polymer–drug conjugate mixed micelles were fabricated by the self-assembly of PEOz-PLA-ace-Cur, a conjugate of curcumin (Cur) with poly(2-ethyl-2-oxazoline)-poly(D,L-lactide) (PEOz-PLA) through the linkage of the pH-cleavable acetal bond, and PEOz-PLA-imi-DOX, a conjugate of doxorubicin (DOX) with PEOz-PLA through the linkage of the pH-cleavable benzoic imine bond. The mixed conjugate micelles (PP-Cur/PP-DOX-Mix-PMs) with accurately and conveniently controlled mass ratio of the two drugs were demonstrated to have a small particle size (40–128 nm), high drug loading capacity and pH-dependent drug release behavior. Notably, PP-Cur5/PP-DOX1-Mix-PMs exhibited slower DOX release under physiological conditions compared with PEOz-PLA-imi-DOX micelles, resulting in deeply reduced side effects in vivo. Furthermore, the mixed conjugate micelles showed synergistically enhanced inhibition of MDA-MB-231 cell growth and metastasis evidenced by the results of in vitro anti-invasion, wound healing and anti-migration assessment, and in vivo bioluminescence imaging in nude mice, and significant reduction of the side effects of DOX compared with dual drug physically loaded polymeric micelles. Mechanistic studies demonstrated that the possible inhibitory mechanism of PP-Cur5/PP-DOX1-Mix-PMs on tumor metastasis could be assigned to their inhibition of the invasion, migration, intravasation and extravasation of tumor cells. In conclusion, the multi pH-sensitive polymer–drug conjugate mixed micelles with synergistically enhanced anti-tumor and anti-metastasis activity are potential candidates for safe and effective cancer combination therapy.

This study aims to elucidate the interaction mechanisms between curcumin (Cur) and four collagen subtypes (fish type I [FCI], bovine type I [BCI], fish type II [FCII], bovine type II [BCII]), with parallel characterization of the structural and functional attributes of their derived nanoparticles. Type I Collagen/Cur nanoparticles exhibited superior solution stability compared to type II. Cur binding significantly enhanced the surface hydrophobicity, absolute ζ potential, and surface tension of collagen, while reduced dynamic interfacial tension. The binding type of Cur to collagen was static, and binding process was enthalpy-driven exothermic reaction. Molecular dynamics simulations revealed that hydrophobic interactions, hydrogen bonds, and electrostatic forces dominated the binding process. The binding affinity followed the order: FCI/Cur > BCI/Cur > FCII/Cur > BCII/Cur. The binding sites of Cur to type I collagen and type II collagen were around Ser129-Glu135 and Asn179-Ser183 residues. Collagen/Cur nanoparticle stabilized emulsions and improved oxidative stability and storage modulus.

Protein-based complexes are critical for enhancing food functionality, particularly owing to the synergistic effects of ternary covalent complexes containing proteins, polysaccharides, and polyphenols. This study investigated the formation mechanism, structural characteristics, and functional properties of ternary covalent complexes composed of β-lactoglobulin (β-Lg), hyaluronic acid (HA), and three dicaffeoylquinic acid (DCQA) isomers (3,5-DCQA, 3,4-DCQA, and 4,5-DCQA); the complexes were prepared using different addition sequences. These complexes exhibited reduced surface hydrophobicity, altered secondary structures, and enhanced thermal stability. They showed significantly better antioxidant activities and emulsifying properties in comparison with native β-Lg. The protein-polysaccharide-polyphenol sequence yielded more stable and compact structures with higher polyphenol binding capacity. Subsequently, the protein-polysaccharide-polyphenol complexes were used to encapsulate curcumin (Cur), and it yielded encapsulation efficiencies up to 76.14 % (β-Lg-HA-3,4-DCQA-Cur), which was significantly higher than the encapsulation efficiency of native β-Lg-Cur (36.35 %). These complexes also improved the physical stability and antioxidant activity of Cur under adverse conditions. These findings highlight the critical roles of the addition sequence and polyphenol structure in modulating the functionality of protein-based complexes, and offer promising strategies for enhancing the delivery of bioactive compounds in food applications.

Oxidative stress of neurons caused by a series of complex neuropathological processes will induce certain neurodegenerative disorders including epilepsy. Curcumin (Cur) is an effective natural antioxidant compound; however, the poor bioavailability obstructs its neural protective applications. In this study, Cur is encapsulated in solid lipid nanoparticles (SLNs) for better neuroprotective efficacy. In vitro study certified that Cur-SLNs functioned obviously better against neuronal apoptosis than Cur, by significantly decreasing the level of free radical and reversing mitochondrial function through the activation of the Bcl-2 family. In vivo experiments showed that SLNs transported Cur through the blood–brain barrier (BBB). The behavioral performance of epileptic mice was improved by Cur-SLNs, with more NeuN but less TUNEL positive cells observed in hippocampus. The in vivo mechanism was also explored. Cur-SLNs reduced neuronal apoptosis through Bcl2 family and P38 MAPK pathways. Overall, Cur-SLNs have better protective effects toward oxidative stress in neurons than free Cur both in vitro and in vivo, which suggests they may be a promising agent against neurodegenerative disorders including epilepsy.
