The development of single-excitation dual-emission (SEDE) ratiometric nanoprobes has emerged as a crucial strategy for the detection of heavy metal ions. In this study, a rare earth-based SEDE nanoprobe, designated ZG-Tb, was developed for rapid and sensitive Hg 2+ detection through Tb 3+ coordination onto guanine-rich hairpin DNA (GH5)-functionalized Cr-doped zinc gallogermanate nanoparticles. Upon 260 nm excitation, ZG-Tb exhibits two distinct emission bands: an intrinsic 696 nm emission from Z nanoparticles and a Tb 3+ -mediated 545 nm emission sensitized by surface-immobilized GH5. In the presence of Hg 2+ , competitive interaction between Hg 2+ and Tb 3+ for GH5 results in the selective quenching of the 545 nm emission while leaving the 696 nm emission unaltered. The ratiometric luminescence response ( I 545 / I 696 ) enables quantitative detection of Hg 2+ within 1 min, with a linear range of 0–30 nmol/L and a detection limit of 1.45 nmol/L. The nanoprobe also shows reliable performance in drinking and tap water samples, underscoring its potential for practical applications.

The spatial organization of enzymes in natural cells enables efficient metabolic cascades through localized microenvironments and controlled intermediate diffusion. Mimicking this compartmentalized architecture in artificial systems remains a key challenge in biocatalysis. Here, we introduce a DNA-mediated self-assembly strategy to construct the biomimetic core-satellite structure of metal-organic frameworks (MOFs) that spatially organize multiple enzymes into defined compartments. In this system, DNA bridges larger “core” MOFs and smaller “satellite” MOFs, forming ordered assemblies with tunable microenvironments tailored for individual enzymes. Using glucose oxidase (GOx) and horseradish peroxidase (HRP) as a model cascade, we demonstrate that the core-satellite structure facilitates substrate diffusion, mitigates inhibition by intermediates, and enhances overall catalytic efficiency. This platform was further applied to support multi-enzyme cascades, including the two-enzyme system of N -acetyl hexosamine 1-kinase (NahK)/UDP- N -acetylgalactosamine pyrophosphorylase (GlmU) for glycosyl donor synthesis, and the three-enzyme system of β-galactosidase (β-Gal)/GOx/HRP for sequential cascade catalysis. Compared to the single MOF system, spatially compartmentalized MOFs exhibit up to 34-fold higher activity and improved thermal, solvent, and operational stability. This work provides a versatile platform for the design of cell-inspired biocatalytic systems and underscores the potential of DNA-guided MOF assembly in constructing programmable multi-compartment enzyme reactors.

Highly sensitive detection of cell secretions at the single-cell level is critical for efficient cell screening and monoclonal antibody (mAb) production. Herein, we present a highly sensitive microfluidic chemiluminescence immunoimaging (μCLII) method for rapid and quantitative screening of high-yield specific hybridoma cells (HSHCs) by detecting single-cell secreted antibodies on a rationally designed lantern-shaped microchamber array chip. This method can effectively capture and isolate single hybridoma cells to in situ detect their secreted antibody by visual CL imaging with a proximity recognition amplification strategy. The proposed strategy can be performed by antibody-triggered proximity hybridization to release a DNA sequence, which initiates a rolling circle amplification reaction to turn on abundant hemin-DNA enzymes for generating strong CL emission of the imidazole-enhanced luminol-H2O2 system. This method shows a detection limit of ∼60 antibodies per chamber, and thus enables a precise sorting of HSHCs within 70 min. Using antiproprotein convertase subtilisin/kexin type 9 mAb (PCSK9-mAb) as a model, all of the cells selected by the μCLII method show high-producing hybridomas with PCSK9-mAb yield more than 80 fg during the screening process, indicating a promising potential of the proposed method for single-cell analysis and mAbs screening.

5-methylcytosine (5mC), the most well-known form of DNA methylation, is typically regulated by two types of enzymes: DNA methyltransferases (DNMT), “writers” responsible for the deposition of methylation marks, and ten-eleven translocation dioxygenases (TET), “erasers” controlling their active removal. Therefore, these two types of enzymes play fundamental roles in regulating active turnover of 5mC modification. Given that 5mC-based approaches have been widely applied in diagnosis and treatment, and that a subtle imbalance in these enzyme activities often precedes aberrant 5mC deposition, we developed a single-molecule array–based method for sensitive detection of both enzymes, enabling activity analysis at the single-cell level. Notably, in human peripheral blood mononuclear cells (PBMCs), DNMT activity is significantly higher in elders than in young individuals, while TET activity remains unchanged. Moreover, the relative active demethylation ratio (TET activity/DNMT activity) is significantly higher in young individuals. When applied to clinical disease samples, bone marrow mononuclear cells (BMNCs) from pediatric patients with active hematologic disease showed elevated TET activity and an increased active demethylation ratio. Overall, our approach provides a valuable tool for investigating diseases associated with aberrant 5mC modifications, with promising implications for biomarker development and disease monitoring.

Despite the promise of cuproptosis in antitumor therapy, developing strategies to enhance its therapeutic efficacy within the tumor microenvironment remains a challenge. Inspired by the chronotherapy that manipulate circadian rhythms to enhance drug effectiveness, herein we report for a CRISPR-customized copper-DNA nanoplatform (Cu-RNP) that synergistically induces multimodal cell death, including potentiated cuproptosis, by manipulating circadian and metabolic pathways. Cu-RNP integrates coordination-driven self-assembly of Cu2+-DNA nanospheres with Cas13d/crRNA ribonucleoproteins targeting BMAL1. Upon cellular internalization, the acidic and reducing endo/lysosomal environment triggers Cu-RNP disassembly, releasing RNP to silence BMAL1 and disrupt circadian oscillations, leading to WEE1 downregulation and p21 upregulation, thereby inducing apoptosis. Simultaneously, liberated Cu2+ generates cytotoxic hydroxyl radicals for chemodynamic therapy (CDT) and concurrently depletes GSH, promoting mitochondrial copper overload for cuproptosis. Importantly, we demonstrate that silencing BMAL1 disrupts circadian rhythms, inhibits glycolysis, enhances mitochondrial respiration, and redirects metabolic flux to the TCA cycle, thereby amplifying the cell’s vulnerability to copper-induced cuproptosis. In vitro and in vivo results demonstrate that Cu-RNP sensitizes cancer cells to cuproptosis and elicit strong antitumor response through the synergistic combination of cuproptosis, CDT, apoptosis, and circadian-metabolic modulation. This study demonstrates a mechanistic link between BMAL1-regulated circadian rhythms and cuproptosis sensitivity, suggesting a potential treatment strategy for multimodal, cuproptosis-potentiating cancer therapies.

Photodynamic therapy (PDT)-mediated antitumor immune response depends on oxidative stress intensity and subsequent immunogenic cell death (ICD) in tumor cells, yet the inherent antioxidant system restricts reactive oxygen species (ROS)-associated oxidative damage , which is highly correlated with the upregulated nuclear factor erythroid 2-related factor 2 (Nrf2) and the downstream products, such as glutathione (GSH). Herein, to overcome this dilemma, we designed a versatile nanoadjuvant ( [email protected] ) to enhance the sensitivity of tumor cells to oxidative stress via Nrf2-specific small interfering RNA (siNrf2). The constructed [email protected] could significantly amplify photooxidative stress and achieve robust DNA oxidative damage, activating the stimulator of interferon genes (STING)-dependent immune-sensing to produce interferon-β (IFN-β). Additionally, [email protected] together with laser irradiation reinforced tumor immunogenicity by exposing or releasing damage-associated molecular patterns (DAMPs), showing the prominent adjuvant effect for promoting dendritic cell (DC) maturation and T-lymphocyte activation and even alleviating the immunosuppressive microenvironment to some extent.

The predictability of Watson–Crick base pairing provides unique structural programmability to DNA, facilitating the development and application of biomolecules in biocomputing. However, in DNA-based biocomputing, the scale of operation that can be achieved by an existing reaction system is very limited. How to expand the operation range of a logic circuit and realize the integration and extensibility of circuits is always the key problem to be solved in this field. In this work, by designing a multifunctional DNA–nanostructure-based reaction platform, which can realize an output of up to 2n scalable fluorescence signals, combined with the construction of an input “library” and a modular distribution strategy of output signals, for the first time, we successfully performed the calculation of both square roots and cube roots of consecutive integers within a decimal number of “10” and in each result of the operation, two digits after the decimal point are preserved (). We believe that the design concept presented in this work can help effectively solve the urgent problems of biological computing in terms of computational scaling, integration and scalability, and can open up new horizons for the design of new functional devices and complex computing circuits.

Ochratoxin A (OTA) with high toxicity represents a serious threat to the agriculture and food chain, consequently to human health. Herein, a simple electrochemiluminescence (ECL) biosensor was constructed for ultrasensitive detection of OTA based on mercaptopropionic acid templated Au nanoclusters (Au NCs) as intensive signal probe and a non-enzymatic 2D DNA walking machine as the effective amplification strategy. Specifically, the target related bipedal DNA walker efficiently moved along 2D DNA tracks through toehold-mediated DNA strand displacement, which triggered abundant signal probes for combining to the DNA tracks. Moreover, the Au NCs could exhibit strong ECL emission due to fast electron transfer from massive Au-S electronic pathways under the electrochemical excitation. Thus, the biosensor possessed significant ECL response for achieving ultrasensitive detection toward OTA with low detection limit of 3.19 fg/mL. Impressively, the sensing platform was also applied to detect OTA from edible oils, exhibiting great application potential in food analysis.

Nucleic acid markers have been widely used in the detection of various virus-related diseases, including hepatitis B virus (HBV), which is spreading worldwide. The trans -activated CRISPR-Cas system has shown excellent sensitivity and specificity in nucleic acid detection. However, nucleic acid testing usually requires amplification of the target nucleic acid for more accurate and specific detection; furthermore, current nucleic acid assays are time-consuming, costly, and are limited by non-specific cross-reactivity. We developed an amplification-free viral DNA biosensor-based diagnostic method that uses a clustered regularly interspaced short palindromic repeats-associated system (CRISPR/Cas)-based approach with surface enhanced Raman spectroscopy . This method can specifically identify the target site by changing the crRNA sequence. In addition, the incubation period and development of the disease can be determined by quantitative detection of viral DNA. This system could achieve rapid and highly sensitive detection of HBV DNA within 50 min and vast detection range from 0.1 pM to 1 nM. Therefore, a combined CRISPR/Cas12a-SERS-based assay would improve the sensitivity of detection in assays using multiple biomarkers. In conclusion, our CRISPR/Cas12a-based biosensor would enable rapid, simple, and sensitive detection of HBV nucleic acids.

Manganese oxide nanomaterials (MONs) are emerging as a type of highly promising nanomaterials for diseases diagnosis, and surface modification is the basis for colloidal stability and targeting delivery of the nanomaterials. Here, we report the in-situ functionalization of MnO 2 with DNA through a biomineralization process. Using adsorption-oxidation method, DNA templated Mn 2+ precursor to biomineralize into nano-cubic seed, followed by the growth of MnO 2 to form cube/nanosheet hybrid nanostructure. Among four types of DNA homopolymers , poly-thymine (poly-T) was found to stably attach on MnO 2 surface to resist various biological displacements (phosphate, serum, and complementary DNA). Capitalized on this finding, a di-block DNA was rationally designed, in which the poly-T block stably anchored on MnO 2 surface, while the AS1411 aptamer block was not only an active ligand for tumor targeting delivery, but also a carrier for photosensitizer (Ce6) loading. Upon targeting delivery into tumor cells, the MnO 2 acted as catalase-mimic nanozyme for oxygenation to sensitize photodynamic therapy, and the released Mn 2+ triggered chemodynamic therapy via Fenton-like reaction, achieving synergistic anti-tumor effect with full biocompatibility. This work provides a simple yet robust strategy to functionalize metal oxides nanomaterials for biological applications via DNA-templated biomineralization.

A branched DNA-based electrochemical biosensor was designed to sensitively detect specific nucleic acids. On this platform, novel a branched DNA with three sticky ends could be used as a biosensor to sensitively and specifically detect nucleic acids. Meanwhile, we also employed branched DNA-modified AuNPs as a signal amplifier to further improve the sensitivity. Branched DNA sensors, target DNA, and DNA-modified AuNPs formed a sandwich structure to produce an electronic signal for target DNA detection. The reaction primarily involved DNA hybridization without bulky thermal cyclers and enzymes. We proved that the hybridization reaction easily occurred under different conditions, such as the NaCl concentration, reaction time, pH, and temperature, except for a pH lower than 4. The limit of detection could go as low as 0.09 pM (S/N = 3) with excellent specificity and selectivity. There was a correlation curve relationship between the peak current and the logarithm of the target DNA concentration (0.10 pM to 10 nM). The correlation coefficient reached 0.987. The electrochemical platform enables a branched DNA nanostructure to determine nucleic acids for disease diagnosis. Keywords:amplifier;branched DNA;diagnosis;detection;nucleic acids

Cardiac troponin I (cTnI), a principal biomarker of acute myocardial infarction, possesses superior cardiac specificity and can provide a long detection window for cardiac injury. Although the potential of cTnI as a cardiac biomarker is promising, sensitive and rapid detection of cTnI remains challenging. In this work, a novel colorimetric biosensor was developed for cTnI detection based on multifunctional hybrid nanoprobes coupled with triple-aptamer-modified tetrahedral DNA nanostructures (TAPT-TDNs). First, we evaluated the affinity of four TAPT-TDNs with three vertices modified by two kinds of aptamers in different proportions. The four TAPT-TDNs exhibited much better affinity than the single-stranded aptamer for cTnI, which was attributed to the fact that TAPT-TDNs could improve the capture efficiency of cTnI with excellent interface density and constant support. Second, the selected TAPT-TDNs with highest affinity were immobilized on Fe 3 O 4 -Au magnetic nanoparticles as capture probes (named Fe 3 O 4 -Au-TAPT-TDNs). TAPT-TDNs and HRP were both labeled on NH 2 -MIL-101(Fe) to form signaling probes (named MIL-101(Fe)-HRP-TAPT-TDNs). Then, with the help of capture probes and signaling probes, the target protein was recognized and captured to form capture probe-cTnI-signaling probe sandwich-like structures, causing a decrease in signaling probes in the supernatant with magnetic separation and eventually leading to a visual color change. This colorimetric aptasensor displays excellent analytical performance toward cTnI with a detection limit of 27 pg·mL −1 . The anisotropic aptamer-mediated colorimetric sensor may serve as an alternative to conventional immunoassays for quick and accurate detection of cTnI in clinical samples.
