Preparation, characterization and antioxidant activity of polysaccharide from spent Lentinus edodes substrate

Hongji Zhu,Li Tian,Lei Zhang,Jingxiu Bi,Qianqian Song,Hui Yang,Jianjun Qiao

This study explored the potential of spent Lentinus edodes substrate, a by-product of mushroom industries causing environmental pollution, serving as materials to produce antioxidant polysaccharide. The extraction process of spent Lentinus edodes substrate polysaccharide (SLSP) was optimized and the effects of drying methods on chemical composition, morphological property and antioxidant activity were investigated. Results showed that freeze-dried SLSP (SLSP-F) exhibited the best quality in terms of the polysaccharide yield (13.00%) and antioxidant activity. The EC 50 values of SLSP-F on DPPH, ABTS and superoxide anion radicals was 0.051   mg/mL, 0.379   mg/mL, 0.719   mg/mL, respectively, which was significantly lower than that of freeze-dried Lentinus edodes polysaccharide (LP-F). After purification by Sephadex G-150, the purified SLSP-F (PSP) has a molecular weight of 16.77   kDa. Compared with LP-F, PSP has more reducing sugars and uronic acids in chemical composition and higher contents of xylose, glucose and galactose in monosaccharide composition. FT-IR and NMR spectroscopy analysis revealed that PSP has both α and β glycosidic bonds and massive acetyl groups, which is different from LP-F mainly composed of 1, 3 linked α-D-Man p residue with some acetyl groups. The findings provided a reliable approach for the development of antioxidant polysaccharide from spent Lentinus edodes substrate.

Preparation and properties of quercetin-incorporated high density polyethylene/low density polyethylene antioxidant multilayer film

Li-Jing Lu,Li-Xin Lu

High density polyethylene/low density polyethylene (LDPE) antioxidant multilayer films were prepared by the co-extrusion method, and quercetin was incorporated in the LDPE layers as an antioxidant. The release rates of quercetin and the antioxidant activities of films were adjusted by changing the amount of ethylene vinyl acetate (EVA) and diatomite added into the LDPE active layer. The morphologies of the films were observed by SEM, and the release property of quercetin was characterized by a high-performance liquid chromatography (HPLC) method. The mechanical properties and heat sealing performance of the films were influenced to a certain extent by the amounts of EVA and diatomite in the active layers, while the barrier properties of the films were almost unchanged. The release of quercetin from the active films to a food simulant (95% alcohol) at 37°C was measured over 55 days. When the EVA amounts were 30% and 40% to 50%, the diffusion coefficients, D, were 10 −14 and 10 −13 cm 2 /s, respectively. In addition, the antioxidant activity values of the films were enhanced as the EVA amount increased. When adding diatomite into the active layer with 50% EVA, the diffusion coefficient, D, was 10 −11 cm 2 /s, and the quercetin was almost completely released with a partition coefficient, K, of less than 1. Meanwhile, the antioxidant activity values of the films exceeded 95%. The antioxidant release rate could be adjusted within a wide range; thus, these active films could be used for food antioxidant protection.

Effect of different Agrobacterium rhizogenes strains on hairy root induction and analysis of metabolites in Physalis peruviana L.

Yijia Zhong,Zhie Zhou,Zhongping Yin,Lu Zhang,Qingfeng Zhang,Yihai Xie,Jiguang Chen

Physalis peruviana L. ( P. peruviana ) is an edible medicinal plant rich in bioactive phenolics. This study aimed to establish a hairy root (HR) culture of P. peruviana as a potential source for the synthesis of natural compounds. HRs were induced in P. peruviana using different Agrobacterium rhizogenes strains (R1601, C58C1, A4, and K599). Notably, K599 did not induce HR formation, whereas R1601, C58C1, and A4 yielded transformation frequencies of 57.78, 65.14, and 72.31%, respectively. Secondary metabolite production and antioxidant capacity were further examined in HRs induced using C58C1, R1601, and A4. It was found that A. rhizogenes R1601 induced the greatest increase (44% compared to that observed in the non-transformed culture). The methanolic extract of HRs induced by A. rhizogenes R1601 exhibited strong antioxidant capacity, with IC 50 values of 1.41 mg DE/mL and 2.33 mg DE/mL for 2,2-diphenyl-1-picrylhydrazyl and 2,2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid), respectively. The HR culture showed higher production of phenolic compounds and higher antioxidant capacity than the non-transformed cultures. Ultra-performance liquid chromatography time-of-flight tandem mass spectrometry was used to identify eight alkaloids, phenolics, and glycoside compounds. A. rhizogenes R1601 is emerging as a possible strain for the mass production of HR and bioactive phenolic compounds in P. peruviana .

Development of a multifunctional active food packaging membrane based on electrospun polyvinyl alcohol/chitosan for preservation of fruits

Ying He,Tian Zhong,Yiguo Liu,Menghui Wan,Lei Sun,Yanbao Zhao,Zhihua Wang

To mitigate environmental impacts in food preservation, the development of a multifunctional membrane for packaging is of importance. In this study, we have successfully fabricated a nanofibrous membrane using an eco-friendly electrospinning technique, comprising polyvinyl alcohol (PVA), chitosan (CS), and tannic acid (TA). The resulting nanofibrous membranes were crosslinked with glutaraldehyde (GA) and surface modified with ZnO. Our findings demonstrate that the crosslinking process enhances water resistance, reduces water vapor permeability, improves tensile strength (from 3 to 18 MPa), and enhances thermal stability (increasing decomposition temperature from 225 °C to 310 °C). Furthermore, the incorporation of TA and ZnO provides antioxidant properties to the membrane, effectively preventing food decomposition caused by UV-induced oxidation. Additionally, CS, TA, and ZnO synergistically exhibit a remarkable antibacterial effect with a bacteriostasis rate exceeding 99.9 %. The strawberry fresh-keeping experiment further confirms that our developed membrane significantly extends shelf life by up to 6 days. Moreover, cytotoxicity assays confirm the non-toxic nature of these membranes. The innovative significance of this study lies in proposing a robust GA-PVA/CS/TA@ZnO nanofibrous membrane with excellent mechanical properties, biocompatibility, and multiple functionalities including antibacterial, anti-ultraviolet, and anti-oxidation capabilities. It has tremendous potential for applications in active food packaging materials.

Development of a multifunctional nano-hydroxyapatite platform (nHEA) for advanced treatment of severely infected full-thickness skin wounds

Shixin Zhang,Tinghan He,Fengxin Zhao,Qinling Tan,Dongxiao Li,Qiguang Wang,Yumei Xiao,Xingdong Zhang

The treatment of full-thickness skin injuries complicated by severe infection is hampered by the lack of comprehensive solutions that can regulate the various stages of wound healing . Consequently, there is an urgent need for a multifunctional dressing capable of multi-level regulation. In this study, we propose a novel solution by covalently integrating ε-poly-l-lysine-grafted gallic acid (EG) and in situ bioreduced silver nanoparticles (AgNPs) onto nano-hydroxyapatite (nHAP), thereby developing a multi-layered, multifunctional nanoplatform (nHEA). Cell experiments have shown that, compared to nHAP and nHAP loaded only with EG (nHEG), the addition of AgNPs to nHEA confers excellent antibacterial properties while maintaining optimal biocompatibility . The incorporation of EG onto nHEG and nHEA imparts antioxidation, anti-inflammatory, and pro-angiogenic functions, and the release of Ca 2+ and EG further enhances fibroblast migration and collagen secretion. In a rat model of full-thickness skin injury with severe infection, nHEA demonstrates remarkable antibacterial and anti-inflammatory effects, along with promoting collagen remodeling and regeneration. Together, both cell experiments and animal studies confirm the significant potential of this innovative multifunctional nanoplatform in the treatment of full-thickness skin injuries with severe infection. Statement of significance Treating infected full-thickness skin injuries poses a longstanding challenge due to the lack of comprehensive solutions that can regulate different stages of wound healing. This study introduces a novel multifunctional nanoplatform, nHEA, developed by covalently integrating ε-poly-l-lysine grafted with gallic acid (EG) and in situ bioreduced AgNPs onto nano-hydroxyapatite (nHAP). Cell experiments reveal that the integration of AgNPs enhances nHEA's antibacterial performance while maintaining optimal biocompatibility. The inclusion of EG bestows antioxidant, inflammation-regulating, and angiogenetic properties upon nHEA, and the release of Ca 2+ and EG stimulates the migration and collagen secretion of fibroblast cells. Consequently, nHEA exhibits superior antibacterial and inflammation-regulating efficacy, and stimulates collagen remodeling and regeneration in vivo , making it a promising treatment for severely infected skin injuries.

Development of antioxidant arabinoxylan-tea polyphenol composite films for enhanced preservation of fresh grapes

Rongrong Luo,Zimeng Peng,Na Wu,Li Zhang,Bin Peng,Rong Shao,Wei Xu,Lei Yang

In this study, arabinoxylan (AX) was used as a substrate, and tea polyphenol (TP) as a functional additive to create degradable, non-polluting, and antioxidant packaging materials. The effects of different TP concentrations on light transmittance and antioxidant activity of the AX-TP composite films were analyzed. The colors of the films gradually deepened with increasing TP content. The maximum scavenging rate of 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals using the AX-TP composite film solution of 59.13 ± 2.19 % was achieved at the TP concentration of 2.0 %. AX-TP composite films with different TP concentrations were applied to the surfaces of grapes, and the sensory quality, shriveling and decay rates, titratable acid, and weight loss rate of grapes during storage were evaluated using data from different experimental groups. The AX-TP composite films coated on grapes reduced the transpiration of water in the fruit and delayed grape spoilage, demonstrating an excellent preservation effect. These results show that AX-TP composite films increase the shelf life of fresh grapes.

Development of pH response film based on potato starch/polyvinyl alcohol using Kyoho grape skin extract to monitor pork freshness

Shihui Wang,Biao Zhao,Yanling Hao,Qixiang He

In this study, the optimal ratio of starch and polyvinyl alcohol (PVA) was successfully selected by response surface method. The novel packaging films were fabricated by immobilizing Kyoho skin extract (KSE) within starch/PVA composite film by the casting method. The mechanical property, PH sensitivity, and other properties of films with different KSE content (1–5 mL) were studied. The starch/PVA film with 3 mL KSE (SPK3.0 film) showed more uniform structure than the starch/PVA film in microscope. At the same time, the film has strong mechanical properties and low water barrier capacity. Infrared analysis of the film further revealed hydrogen bond interaction between film matrix and KSE. The smart packaging film displayed distinguishable changes of color from purple to green in pH (1–13). Furthermore, the smart films possessed excellent antioxidant. Therefore, the SPK films had the potential application potential in the field of packaging. Highlights Barrier properties and antioxidant activity were enhanced significantly by the addition of KSE. The film components are connected by hydrogen bonds so that they have good compatibility. The functional film shows different colors in the range of pH 1–13.

Development of Zein–PEG400/PVA–Chitosan Bilayer Films for Intelligent Packaging

Rong Sun,Liangliang Li,Jiangjie Zhou,Yongfeng Zhang,Haiya Sun,Datong Zhang,Qi Wu

Zein exhibits excellent biodegradability, thermal stability, UV resistance, and water barrier properties, making it a promising candidate for food packaging applications. However, pure zein films suffer from brittleness and poor mechanical strength, which limit their practical use. In this study, a unique bilayer packaging film (ZP/P-C) was developed using a layer-by-layer solution casting technique, where hydrophobic zein was coated onto a polyvinyl alcohol and chitosan composite layer (P-C). Incorporating PEG400 into the zein layer improved the interfacial compatibility of the bilayer film, increasing its uniformity and toughness. The resulting bilayer films demonstrated enhanced mechanical properties, flexibility, and water vapor barrier performance. Specifically, the ZP7.5/P-C bilayer film showed an elongation at break of 68.74% and a modulus of elasticity of 187.19 MPa. It had a water vapor permeability of 6.60 × 10−11g·m·m−2·s−1·Pa−1and provided near-complete UV protection within the 200–350 nm range. Furthermore, an intelligent detection bilayer film was created by integrating anthocyanin extract into the zein layer. Adding anthocyanin improved the film’s antioxidant properties and allowed it to respond colorimetrically to total volatile basic nitrogen. The bilayer film ZPBA1.0/P-C displayed an excellent antioxidant activity (45.8%) and remarkable color change (ΔE = 20.2) in response to ammonia, effectively indicating shrimp spoilage in 48 h (ΔE > 10). This investigation spotlights the potential of zein-based bilayer films in active and intelligent food packaging, offering innovative strategies to improve food safety and extend the shelf life of perishable goods.

Chitosan-curcumin conjugate prepared by one-step free radical grafting: Characterization, and functional evaluation

Xuerong Luo,Lingyu Zhao,Imran Mahmood Khan,Lin Yue,Yin Zhang,Zhouping Wang

Curcumin (Cur) is a naturally hydrophobic polyphenol, and it has a wide range of physiological functions. But the practical application of Cur is constrained by its low water solubility and poor stability. To improve these deficiencies of Cur, a novel Cur derivative (CS-Cur) was prepared by grafting chitosan (CS) with Cur through a one-step reaction of a free radical-mediated redox system. A series of characterizations provided evidence that the grafting of CS with Cur was successful. The obtained CS-Cur showed lower crystallinity and thermal properties than CS and Cur. After grafting, the water solubility of CS-Cur was found to be 9.76 ± 2.45 g/L and greatly improved. Meanwhile, the CS-Cur showed good photothermal stability, antioxidant activity, and photodynamic antibacterial activity in an aqueous solution, and it had good in vitro biosafety. This provides an idea for the design and synthesis of novel highly water-soluble Cur derivatives and also improves the practical application of Cur in aqueous systems.

Conformational and processing properties of a high-active ingredient involving soy protein isolates bound with anthocyanins and its application in cake baking

Wu Han,Li Bin,Corradini Maria,Zhong Liang,Zhou Jianzhong,Liu Xiaoli

Soybean protein is of plant origin and is commonly appropriate for improving the processing characteristics of foods. This study aimed to explore a novel functional ingredient that contained soybean protein isolate (SPI) and blueberry anthocyanins (BANs). The spatial conformation and secondary structure of SPI-BANs complexes were analyzed using circular dichroism and fluorescence spectroscopy, the processing properties were investigated as well as the retention of antioxidant activity during thermal treatments. Results showed that the contents of free sulfhydryl and free amino groups in complexes increased to 3.50 and 1.19 folds than those of SPI, respectively, while the surface hydrophobicity decreased by 74.23%. Compared with SPI, the BANs-modified SPI had a smaller particle size of 29.12 nm and a lower zeta-potential of -8.73 mV and on the other hand, the complexes possessed higher solubility (83.08%) and foaming and emulsifying properties (115.08% and 54.03 m 2 /g). After fortification with SPI-BANs, the baking loss rate and adhesiveness of chiffon cake were reduced by 10.82% and improved to 0.24 N.mm, respectively. The high antioxidant activities of SPI-BANs under heat led to the cake’s bioactivities largely enhanced by 1.99 ~ 12.71 folds, being 345.19 µg Trolox/g for the DPPH radical scavenging activity. This study developed the functional food ingredients as antioxidants and a substitute for animal-based proteins in bakery products, which was safe and sustainable by using the dietary components from plant resources.
