Dynamic 3-hydroxyflavone-modified D-alanine nanoparticles for metabolic-incorporation in anaerobic, pathogenic biofilms generating CO in infected periodontal pockets

Da-Yuan Wang,Jing Li,Xiao-Han Tian,Siyu Zhang,Yao Chen,Feihe Ma,Henk.J. Busscher,Jing Shen,Linqi Shi

Light-activatable CO-generating molecules such as 3-hydroxyflavone (3-HF) derivates, are water-insoluble, which limits their applicability for infectious biofilm control. Here, a 3-HF-modified type of D-alanine (3-HFD) was coupled with hexadecyltrimethylammonium-bromide (CTAB) to self-assemble into water-suspensible, hydrophilic nanoparticles. Self-assembled 3-HFD nanoparticles did not generate CO upon visible-light irradiation, but dynamically disassembled upon metabolic-incorporation in bacterial cell-wall peptidoglycan of Gram-positive and Gram-negative periodontal pathogens, leaving molecular 3-HFD inside the cell-wall that generated CO upon visible-light irradiation. In vitro , visible-light irradiation of aerobic Staphylococcus aureus and anaerobic Porphyromonas gingivalis biofilms in presence of 3-HFD nanoparticles yielded CO-generation with strong antibiofilm activity under atmospheric oxygen conditions and in a nitrogen atmosphere. 3-HFD nanoparticles were subsequently applied for treatment of ligature-induced periodontitis in specific-pathogen-free rats with P. gingivalis inoculated, periodontal pockets. Irrigation with nanoparticle suspensions and visible-light irradiation of P. gingivalis infected periodontal pockets was demonstrated to yield in-pocket CO-generation. CO-generation improved bone-regeneration, modulated macrophages to attenuate immune-inflammation and reduced P. gingivalis colony-forming-unit counts, while hardly affecting total colony-forming-unit counts. This points to selective elimination of CO-susceptible pathogens and restoration of a healthy periodontal microbiome, as confirmed using 16S rRNA microbiome analyses. Herewith, an effective, non-surgical and non-antibiotic based three-in-one CO-based treatment of periodontitis is proposed.

Activation of Sirtuin3 by 6,4′-Dihydroxy-7-methoxyflavanone Against Myoblasts Senescence by Attenuating D-Galactose-Induced Oxidative Stress and Inflammation

Bingsi Li,Yuxuan Gu,Libing Zhou,Rui Chen,Yiwei Liu,Zexuan Wan,Ziyi Liang,Yukang Wang,Renlei Ji,Zhian Liu

Background/Objective: Cellular senescence is increasingly recognized as a key mechanism underlying sarcopenia, an age-related muscle disorder with no effective therapeutic. 6,4′-Dihydroxy-7-methoxyflavanone (DMF), a flavonoid isolated from Dalbergia odorifera T. Chen, has shown anti-senescence potential. This study aimed to investigate the protective effects of DMF against myoblasts senescence and elucidate the underlying molecular mechanisms. Method: A cellular model of senescence was established in C2C12 myoblasts using D-galactose (D-gal). The effects of DMF pretreatment were evaluated by assessing senescence phenotypes, myogenic differentiation, and mitochondrial function. The role of Sirtuin3 (SIRT3) was confirmed using siRNA-mediated knockdown. Results: DMF Pre-treatment effectively attenuated D-gal-induced senescence, as indicated by restored proliferation, reduced senescence-associated β-galactosidase activity, decreased DNA damage, and the downregulation of p53, p21Cip1/WAF1and p16INK4a. Furthermore, DMF rescued myogenic differentiation capacity, enhancing the expression of Myoblast determination protein 1, Myogenin, Myosin heavy chain and Muscle-specific regulatory factor 4, and promoting myotube formation. Mechanistically, DMF was identified as a SIRT3 activator. It enhanced SIRT3 expression and activity, leading to the deacetylation and activation of the mitochondrial antioxidant enzyme superoxide dismutase 2. This consequently reduced mitochondrial reactive oxygen species, improved mitochondrial membrane potential and ATP production, and suppressed the NF-κB pathway by inhibiting IκBα phosphorylation and p65 acetylation/nuclear translocation. Crucially, all the beneficial effects of DMF—including oxidative stress reduction, mitochondrial functional recovery, anti-inflammatory action, and ultimately, the attenuation of senescence and improvement of myogenesis—were abolished upon SIRT3 knockdown. Conclusions: Our findings demonstrate that DMF alleviates myoblasts senescence and promotes myogenic differentiation by activating the SIRT3-SOD2 pathway, thereby reducing oxidative stress and NF-κB-driven inflammation responses. DMF emerges as a promising therapeutic candidate for sarcopenia.

Multipotent anti-aging recombinant human elastin: Counteracting zebrafish caudal fin shrinkage and D-galactose-induced intrinsic skin aging in mice

Lijun Zhu,Wenjie Huang,Jingjing Shi,Fei Li,Xiuxia Sun,Jianxi Xiao

The degradation of elastin in aged skin results in the loss of elasticity, the formation of wrinkles, and even a reduced ability for wound healing after injury. However, previous studies about elastin material have been limited by high immunogenicity of animal-derived elastin, the use of single elastin peptide sequences, and residual crosslinking agents. In this study, we designed a multipotent anti-aging recombinant human elastin (rElastin) and utilized it to counteract caudal fin shrinkage in zebrafish and D-galactose-induced intrinsic skin aging in mice. The rElastin demonstrated thermoresponsive reversible phase-transition closely resembling that of natural elastin. In vitro , cell experiments demonstrated that rElastin effectively promoted the proliferation, adhesion, and spreading of HFF-1 cells. In a D-galactose-induced intrinsic aging mouse model, rElastin significantly restored dermal density, skin hydration, and TEWL to levels approaching those of healthy skin. Histological analysis further confirmed that rElastin facilitated the deposition of collagen and regeneration of elastic fibers, thereby promoting structural and functional repair of aging skin. Additionally, rElastin obviously inhibited the tail fin shrinkage after UV radiation. rElastin effectively targets skin symptoms associated with intrinsic aging, providing fresh insights into anti-aging therapies and showcasing strong potential for applications in regenerative medicine and cosmetic dermatology.

D-Tagatose attenuates DSS-induced ulcerative colitis by inhibiting inflammation, reducing intestinal barrier damage and modulating the intestinal flora composition

Le Wang,Houzhen Lu,Haitao Gui,Zifu Ni,Zhongke Sun,Zihua Wang,Zhihua Wang,Xuanyan Liu,Qipeng Yuan

Ulcerative colitis (UC) is a prevalent inflammatory bowel disease (IBD) posing a significant health threat. This study explored the protective effects of D-tagatose against DSS-induced colitis in mice and its underlying mechanisms using H&E staining, AB-PAS staining, immunofluorescence, immunohistochemistry, ELISA, qPCR, western blotting, and other assays. D-Tagatose improved colitis by increasing body weight and colon length, with decreased DAI (disease activity index) and histopathological scores. The results showed that D-tagatose inhibited the secretion of myeloperoxidase (MPO), inflammatory enzymes (iNOS and COX-2) and pro-inflammatory cytokines (TNF-α, IL-1β and IL-6) as well as increased the content of anti-inflammatory cytokines (IL-10) in vitro. In addition, D-tagatose enhanced the expression of tight junction proteins (ZO-1 and Occludin) and mucin (MUC-2). Furthermore, D-tagatose was able to modulate the gut microbiota dysbiosis caused by DSS-induced UC and increased the content of short-chain fatty acids (SCFAs). This study indicated that D-tagatose attenuated DSS-induced UC by modulating inflammatory cytokines, restoring intestinal barrier function, maintaining gut microbiota homeostasis, and enhancing SCFA production. These findings provide D-tagatose as a safe and effective novel functional food strategy for the prevention and treatment of UC.

Design and Synthesis of D–A Structure Fluorescent Probes for Specific Detection of Human Serum Albumin Using Aggregation-Induced Emission

Yiyi Qiao,Jian Cao,Yanan Li,Chuanxiao Wang,Jiayu Gao,Linlin Wang,Xiao Zhang,Le Wang,Yuanhao Li,Zhicheng Zuo

Graphical “Donor-Acceptor” structural molecules with ICT and AIE properties were constructed. The sensor Nap-B1 was able to specifically recognize HSA by modifying with pyridinium and quinolinium. In the H2O-EtOH system or highly viscous and solid phase environment, the fluorescence of sensor was turned on in an aggregated. Elevated levels of human serum albumin (HSA) serve as an early indicator of cardiovascular and renal diseases in diabetic and hypertensive patients. This study focuses on the design and synthesis of two fluorescent probes, Nap-B1 and Nap-KL, which were developed by functionalizing the “D–A” structural framework consisting of naphthalimide derivatives (A) and triphenylamine (D) with pyridine/quinoline cations. These sensors are intended for the quantitative detection of HSA in urine, specifically recognizing HSA and inducing fluorescence “turn-on” upon binding. Both Nap-B1 and Nap-KL is demonstrated high selectivity for HSA, stability independent of environmental factors, and sensitivity for rapid detection. Changes in Zeta potential, dynamic light scattering (DLS), fluorescence lifetime, and fluorescence intensity were observed, which result from alterations in HSA conformation before and after sensor binding. Molecular docking experiments further confirmed that the probes bind to HSA via hydrophobic interactions and electrostatic adsorption, forming a probe-HSA complex and triggering the fluorescent response.

Aggregation-Enhanced Photophysical Performance of D-π-A Structured Hemicyanine for NIR-II Fluorescent and Photoacoustic Imaging-Guided Photothermal Therapy

Baoling Li,E Pang,Shaojing Zhao,Guowei Deng,Shuodong Wang,Benhua Wang,Jieyun Wu,Guangle Niu,Xiangzhi Song,Minhuan Lan

Near-infrared (NIR)-II fluorescence and photoacoustic (PA) dual-model imaging-guided photothermal therapy (PTT) can precisely diagnose and treat tumors and evaluate the therapeutic efficacy in real-time. Herein, we utilized a donor-π-acceptor (D-π-A) structured hemicyanine dye (named M1) with a large conjugated structure and strong intramolecular charge transfer effect and demonstrated that the aggregation of M1 could significantly enhance its photophysical performance by improving its photostability and photothermal conversion capability as compared with M1 in a single molecular state. Furthermore, we prepared water-dispersible NIR-II fluorescent nanoparticles (M1 NPs) by wrapping M1 with DSPE-PEG2000-NH2. The obtained M1 NPs exhibit strong NIR-I absorption and NIR-II fluorescence with their maxima at 734 and 1040 nm, respectively, with a fluorescence quantum yield of 2.84%. Moreover, M1 NPs also exhibit excellent biocompatibility, good photostability, and high photothermal conversion efficiency of 77.5%. In vitro and in vivo experiments reveal that M1 NPs can effectively image tumors through NIR-II fluorescence and PA signals, inhibit DNA replication, trigger cytoskeleton collapse, and eventually induce tumor cell apoptosis under 808 nm laser irradiation. Based on these outstanding properties, the application of M1 NPs in PA and NIR-II fluorescence imaging-guided PTT of tumors is demonstrated.

A novel D-π-A molecule as ICT type fluorescent probe for endogenous hypochlorite imaging in living cells and zebrafishes

Haixia Wu,Yuze Chen,Xunli Ling,Weiwei Yuan,Bowen Li,Zhan Zhou

In order to explore the application of D-π-A molecules in bio-imaging, two novel D-π-A and ICT type molecules ( SE and PE ) are designed and synthesized by the adjustment of the π conjugation bridge. Because of strong intramolecular charge transfer (ICT) effect between the carbazole and pyrimidine unit, SE and PE exhibit weak fluorescence emission in the aqueous solution system. Interestingly, SE shows the enhanced fluorescence detection performance for hypochlorite due to “D-π-A" structure and suitable ICT effect after the oxidative cleavage of alkene unit by hypochlorite. Importantly, the cell membrane permeable SE can be used for the exogenous and endogenous ClO − in living cells and zebrafishes without apparent toxicity before and after treating with ClO − . Our study indicates that regulating “D-π-A" structure and suitable ICT effect of the organic fluorescence probes is a powerful method to enhance their detection performance for the target analyte.

Characterization of antibiofouling behaviors of PVDF membrane modified by quaternary ammonium compound – combined use of QCM-D, FCM, and CLSM

Wen Yue,Zhang Xingran,Chen Mei,Wu Zhichao,Wang Zhiwei

In this study, we systematically evaluated the antibiofouling behavior of quaternary ammonium compound (QAC) blended polyvinylidene fluoride (PVDF) membrane using quartz crystal microbalance with dissipation monitoring (QCM-D) combined with flow cytometry (FCM) and confocal laser scanning microscopy (CLSM) measurements. QCM-D tests showed that the introduction of QAC reduced bacterial attachment due to the biocidal functions of QAC. FCM indicated that cell integrity of the bacteria in the suspension flowing along QAC-modified membrane surfaces during the QCM-D test was severely affected. CLSM confirmed the significantly lower attachment of bacteria and higher dead/live cell ratio onto the surface of modified membranes after the washing step in QCM-D tests. Both FCM and CLSM results validated the antibacterial behavior of QAC-modified membranes by a contact-killing mechanism, which is in agreement with that of QCM-D tests. In addition, the bacterial cells accumulated on modified membrane surface exhibited higher reversibility compared to the control membrane, indicating ease of membrane cleaning. The results highlight that the combined use of QCM-D, FCM, and CLSM can comprehensively characterize the antibiofouling behavior of membranes.

Oxygen vacancy-mediated d-band center regulation in NiCu0.4Fe1.6O4 via Cu-doping achieves efficient peroxymonosulfate activation for durable water purification

Donglei Fu,Wei Zhang,Zhengxiong Zhao,Zhonglin Chen,Pengwei Yan,Jinxiang Zuo,Lianpeng Sun

Element-doped catalysts with oxygen vacancies (OVs) are highly effective in water purification by activating oxidants; however, the interfacial mechanisms and specific contributions of reactive oxygen species (ROS) in metal-doped catalysts featuring OVs for peroxymonosulfate (PMS) activation remain insufficiently elucidated. In this study, Cu-doped NiFe 2 O 4 catalysts with abundant OVs were synthesized via a hydrothermal-calcination process, endowing efficient PMS activation for ROS generation. Remarkably, it mediated more sulfate radicals (SO 4 · − ) and hydroxyl radicals (·OH) generation, accounting for 50.4% and 36.5% of 2,4-dichlorophenoxyacetic acid (2,4-D) degradation, respectively. Benefiting from the formation of OVs, it significantly enhanced PMS adsorption ( E ads = -7.17   eV), markedly lower than that of the pristine NiFe 2 O 4 ( E ads = -4.11   eV). Meanwhile, inducing the regulation of the d-band center of Fe and Ni sites towards the Fermi level, thereby enhancing more electron transfer (0.84 e of NiCu x Fe 2-x O 4 compared to 0.60 e of NiFe 2 O 4 ) from OVs to surface-complexed PMS, and thus voluntarily forming SO 4 · − with lower free energy (△G = -11.7   eV). This work elucidates the OVs-mediated structural regulation of the d-band center and interfacial electron transfer pathway, highlighting a synergistic multi-metal activation mechanism in response to PMS activation, which offers a novel strategy for designing efficient catalysts to trigger interfacial ROS formation.

Laminaria japonica fucoidan ameliorates D-galactose-induced cognitive impairment via the regulation of tryptophan metabolism along the gut-brain axis

Lu Wang,Chunyan Lu,Shugang Li,Tong Wu,Xiaomeng Ren,Shuang Song,Chunqing Ai

Aging is a multifactorial biological process in which chronic inflammation and oxidative stress are central to the development of age-related disorders, including neurodegenerative decline. Fucoidan, a sulfated polysaccharide extracted from brown algae, has well-documented anti-inflammatory and antioxidant effects, and therefore has the potential to be a neuroprotective agent against cognitive impairment associated with aging. In the present study, the major fucoidan fraction (LJF-2) isolated from Laminaria japonica was examined for its neuroprotective properties in a D-galactose induced aging mouse model. Oral administration of LJF-2 for 8 weeks significantly improved spatial learning and memory and suppressed neuroinflammatory responses and oxidative stress while significantly reducing the activation of astrocytes and microglia. These neuroprotective effects were linked to the regulation of key proteins involved in neuronal protection and synaptic function, such as neprilysin and synapsin, by cAMP response element-binding protein signaling. Furthermore, LJF-2 significantly remodeled the gut microbiota through a reduction in the abundance of the Bacteroidota, Proteobacteria, and several putative pathogenic genera, which enhanced the intestinal barrier integrity and modified the microbial metabolite profiles, especially those associated with tryptophan metabolism. Fecal microbiota transplantation experiments further confirmed the role of the gut microbiota modulated by LJF-2 in mediating its neuroprotective effects through reduction of oxidative stress and inflammation. Collectively, these findings suggest that LJF-2 may be a promising therapeutic approach to address the aging-related cognitive decline by modulating the gut–brain axis.

Protective effect and mechanism of taxifolin 7-rhamnoside extracted from Hypericum japonicum against LPS/D-GalN-induced acute liver injury

Hengxing Ran,Yan Wen,Yiebi Qin,Kaiwen Kang,Zhengrong Wu,Yifan Li,Yu Wang,Rihui Wu,Dongli Li,Song Ang,Lishe Gan,Jingwei Jin

This study aims to identify the most primary hepatoprotective compound from Hypericum japonicum and elucidate its underlying mechanisms of liver protection. This study screened 15 compounds from Hypericum japonicum against D-GalN-induced AML12 cell injury, identifying TAX ( Compound 8 , taxifolin 7-rhamnoside) as the most hepatoprotective. This study indicates that TAX reduced AST, ALT, and MDA levels, while enhancing SOD and GSH levels in D-GalN-induced AML12 and LPS/D-GalN -induced liver. In addition, TAX attenuated LPS/D-GalN-induced hepatic injury by lowering inflammatory infiltration and pro-inflammatory cytokines (TNF-α, IL-6 and NO) levels. Network pharmacology analysis revealed common targets between TAX and drug-induced liver injury, forming a protein-protein interaction (PPI) network. Functional enrichment (GO/KEGG) and molecular docking indicated strong binding affinity between TAX and AKT1, confirmed by molecular dynamics simulations. Western blot analyses that TAX ameliorated inflammation and oxidative stress through down-regulating the AKT/NF-κB pathways and up-regulating Nrf2/HO-1 pathways in vivo, TAX inhibited apoptosis via up-regulating Bcl-2 and down-regulating Bax in vitro. Collectively, this study demonstrates that TAX ameliorates LPS/D-GalN-induced ALI by attenuating hepatocyte apoptosis, inflammatory responses, and oxidative stress.

Electronic modulation of PtNd alloy on S-doped g-C3N4 via f-d orbital coupling for enhanced photocatalytic hydrogen evolution

Chenxi Gao,Changcun Han,Hao Xiang,Zhuo Peng,Yinbo Tong,Chengyi Zhang,Hui Lv,Yizhong Huang

Precise control of the photogenerated charge dynamics and the electronic configuration of active sites is key to enhancing the performance of photocatalytic hydrogen-producing materials. Here, we report an efficient one-pot strategy to prepare sulfur-doped g-C 3 N 4 (SCN). Furthermore, a PtNd/SCN composite photocatalyst was successfully fabricated by loading a platinum (Pt)‑neodymium (Nd) alloy onto the SCN surface via an H 2 reduction method. Using trithiocyanuric acid as a single precursor to supply C, N, and S enables uniform S doping within the g-C 3 N 4 framework, broadening the light absorption range and optimizing the band structure. Pt serves as the primary hydrogen-evolving active site, while Nd acts as an “electronic co-catalyst” for Pt, forming strong electronic interactions through its unique 4f orbital structure. HRTEM and XPS analyses confirm the formation of a Pt Nd alloy interface and directional electron transfer from Nd to Pt, effectively modulating the Pt d-band center and optimizing the adsorption free energy of hydrogen intermediates, thereby significantly enhancing hydrogen evolution reaction (HER) kinetics. The experimental results show that the optimized PtNd/SCN composite photocatalyst exhibits a markedly enhanced photocatalytic hydrogen production rate under simulated sunlight, reaching 5468 μmol·g −1 ·h −1 —5 times higher than Pt/CN (1074 μmol·g −1 ·h −1 ) and 1.3 times that of Pt/SCN (4076 μmol·g −1 ·h −1 ). PL, TRPL, and photoelectrochemical measurements confirm that the introduction of Nd promotes the migration of photo-generated electrons from SCN to Pt, thereby significantly suppressing carrier recombination. This study reveals the unique mechanism by which the rare earth element Nd acts as an electronic regulator for the d-band center of Pt, providing new insights into the design of photocatalytic systems with low Pt loading.
