In this study, we addressed the challenge of enhancing the efficacy of curcumin (Cur) delivery for cancer treatment by developing Cur-loaded electrospun sheets (ES) fabricated from PLGA (poly(lactic-co-glycolic acid)) with a focus on tailoring both the dosage and the physical dimensions of the delivery device to suit specific therapeutic applications. Two formulations containing 10 % and 50 % Cur by weight relative to PLGA were designed and produced. We demonstrated that both formulations maintained the amorphous structure of PLGA, where increasing the Cur concentration enhanced the sheets' wettability, tensile extensibility, and mucoadhesive properties. Comparing the drug release trends and the cytotoxicity of the Cur-loaded ES against HeLa cells (human cervical cancer) revealed that both the Cur dosage and the sheet dimensions influenced drug release kinetics and anticancer efficacy. Hence, we highlight the importance of considering specific size-dose relevance measurements when applying polymer-drug systems in solid form. For instance, when maintaining the same dose, a smaller segment of the high dose sheet is suitable for oral delivery, thanks to the higher drug content and better mucoadhesion properties. The low dosage sheet enjoying a larger surface area, could fit local applications requiring uniform static delivery, such as post-surgery recurrence inhibition dressing.

This study aims to elucidate the interaction mechanisms between curcumin (Cur) and four collagen subtypes (fish type I [FCI], bovine type I [BCI], fish type II [FCII], bovine type II [BCII]), with parallel characterization of the structural and functional attributes of their derived nanoparticles. Type I Collagen/Cur nanoparticles exhibited superior solution stability compared to type II. Cur binding significantly enhanced the surface hydrophobicity, absolute ζ potential, and surface tension of collagen, while reduced dynamic interfacial tension. The binding type of Cur to collagen was static, and binding process was enthalpy-driven exothermic reaction. Molecular dynamics simulations revealed that hydrophobic interactions, hydrogen bonds, and electrostatic forces dominated the binding process. The binding affinity followed the order: FCI/Cur > BCI/Cur > FCII/Cur > BCII/Cur. The binding sites of Cur to type I collagen and type II collagen were around Ser129-Glu135 and Asn179-Ser183 residues. Collagen/Cur nanoparticle stabilized emulsions and improved oxidative stability and storage modulus.

Protein-based complexes are critical for enhancing food functionality, particularly owing to the synergistic effects of ternary covalent complexes containing proteins, polysaccharides, and polyphenols. This study investigated the formation mechanism, structural characteristics, and functional properties of ternary covalent complexes composed of β-lactoglobulin (β-Lg), hyaluronic acid (HA), and three dicaffeoylquinic acid (DCQA) isomers (3,5-DCQA, 3,4-DCQA, and 4,5-DCQA); the complexes were prepared using different addition sequences. These complexes exhibited reduced surface hydrophobicity, altered secondary structures, and enhanced thermal stability. They showed significantly better antioxidant activities and emulsifying properties in comparison with native β-Lg. The protein-polysaccharide-polyphenol sequence yielded more stable and compact structures with higher polyphenol binding capacity. Subsequently, the protein-polysaccharide-polyphenol complexes were used to encapsulate curcumin (Cur), and it yielded encapsulation efficiencies up to 76.14 % (β-Lg-HA-3,4-DCQA-Cur), which was significantly higher than the encapsulation efficiency of native β-Lg-Cur (36.35 %). These complexes also improved the physical stability and antioxidant activity of Cur under adverse conditions. These findings highlight the critical roles of the addition sequence and polyphenol structure in modulating the functionality of protein-based complexes, and offer promising strategies for enhancing the delivery of bioactive compounds in food applications.

The application of orally administered nanoparticles in the circulation system is limited by the secretion and shedding of intestinal tract mucous layer. In order to enhance mucoadhesion and mucus penetration of curcumin (Cur)-loaded nanostructured lipid carrier (NLC) after oral administration, a new multifunctional conjugate, N-acetyl-L-cysteine-polyethylene glycol (100)-monostearate (NAPG), was synthesized. Functionalized nanocarriers (Cur-NAPG-NLC) modified by different amounts of NAPG (the amounts of NAPG were 20, 50, and 100 mg) were prepared and investigated for in vitro and in vivo behavior. Mean particle sizes of 89–141 nm with negative zeta potential (−15 to −11 mV) and high encapsulation efficiency (EE, >90%) possessing spherical and stable nanocarriers were observed. Sustained drug release was also observed for the NAPG-NLC. In situ intestinal perfusion studies showed that with increasing the amount of NAPG increase absorption of Cur. In vivo oral pharmacokinetic evaluation suggested that the bioavailability of Cur in rats was proportional to the degree of functionalization of NLCs with NAPG. AUC0–t of Cur-NAPG100-NLC was improved by 499.45 and 116.89 folds as compared to that of Cur solution and unmodified Cur-NLC, respectively. In conclusion, NAPG modified NLC could be a promising drug delivery system for improving oral performance of BCS class IV drugs.

In the research, the solubility of curcumin was studied by static equilibrium method combined with HPLC. The solubility data of curcumin was measured in twelve pure solvents (dimethyl sulfoxide, tetrahydrofuran, acetone, ethyl acetate, isopropanol, acetonitrile, 1-butanol, ethanol, methanol, xylenes, n-hexane and n-heptane) and three binary solvent mixtures (acetone + tetrahydrofuran, acetonitrile + tetrahydrofuran and n-hexane + tetrahydrofuran) at different temperature (T = 278.15–323.15 K) under atmospheric pressure. The solubility values demonstrate that the solubility increased with the increase of temperature in all solvents. The modified Apelblat model and the Buchowski-Ksiazczak λh model were used to fitted the solubility data in pure solvents, the combined nearly ideal binary solvent/ Redlich-Kister (CNIBS/R-K) model and the Jouyban-Acree model were employed to adopt the solubility data in binary solvent mixtures. The RAD max of the modified Apelblat model and the λh model are 2.621% and 5.831%, the RMSD max are 6.9 × 10 -4 and 1.62 × 10 -3 , respectively. The RAD max of the CNIBS/R-K model and the Jouyban-Acree model are 2.249% and 4.901%, the RMSD max are 9.33 × 10 -3 and 3.67 × 10 -3 . Meanwhile, the KAT-LSER model was used to explore the effect of the solvation of curcumin. The fitting results of all equations prove the accuracy of the experimental data, these solubility data are very important for optimizing the extraction process and recrystallization of curcumin.

The combination of microwave irradiation, ultrasonication treatment, and enzyme-assisted extraction approach was applied to study the extraction efficiency and antioxidative activity of curcumin from turmeric ( Curcuma longa L.) using Response Surface Optimization (RSM). The effects of the power and treatment time of microwave irradiation and ultrasonication treatment, the types of enzymes, the amounts, and interaction time of pectinase on the extraction efficiency were studied. RSM was used to select the optimum extraction conditions by implementing Box–Behnken design. The extraction ratio and antioxidant activity of extracted species are 2.89% and 83.95% under the optimal conditions, respectively, which are close to the predicted values, and are much higher than the single extraction approach. Our results show that a versatile approach for biological compounds extraction from agricultural and natural products was successfully developed, and the bioactivity of extracted species could be kept well by using our proposed combination extraction methods.

A novel efficient photoelectrochemical (PEC) sensor was assembled for curcumin (Cur) detection using 2-mercaptoacetic acid (TGA)-capped CdTe nanoparticles and nickel tetra-amined phthalocyanine-linked graphene oxide (TGA-CdTe@NiTAPc-Gr). The TGA-CdTe nanoparticles could be firmly immobilized on the thin film structure of NiTAPc-Gr in a hydrogen-bonded manner, thus producing the proposed PEC sensor with a high photoelectrochemical activity of TGA-CdTe nanoparticles and excellent stability of NiTAPc-Gr. The TGA-CdTe@NiTAPc-Gr thin film showed a stronger blue light absorption property than the individual precursors, and this resulted in the sensitive detection of Cur ranging from 0.25–100 μM with a detection limit of 12.50 nM. In addition, the PEC sensor exhibited a high sensitivity and selectivity and quite good reproducibility. Thus, it shows potential for Cur detection in real samples.

Vascular stent implantation is a major treatment for vascular diseases, yet postoperative infection and persistent inflammation increase the risk of in-stent restenosis. Herein, core–shell structured PCL/PEO-PVP fiber membranes co-loaded with ciprofloxacin hydrochloride (CIP) and curcumin (CUR) were fabricated via coaxial electrospinning. Orthogonal experiments were conducted to optimize critical spinning parameters through multi-index comprehensive evaluation. Characterizations confirm intact core–shell architecture and stable polymeric backbone structure. In vitro release tests reveal sequential drug-delivery behavior: a rapid initial release of hydrophilic CIP and a delayed sustained release of hydrophobic CUR were observed, contributing to early-stage antibacterial and long-term anti-inflammatory effects, respectively. The “antibacterial zone” method verifies favorable antibacterial activity againstStaphylococcus aureus(S. aureus) andEscherichia coli(E. coli). ELISA results demonstrate the enhanced anti-inflammatory capacity of the dual-drug-loaded coaxial fiber membrane. Cellular assays confirm satisfactory cytocompatibility, and endothelial cells achieve normal proliferation and exhibit typical polygonal morphology on the membrane surface. This dual-drug-loaded coaxial-fiber membrane realizes coordinated sequential antibacterial and anti-inflammatory properties, which provides a feasible strategy for developing functional coatings toward vascular stents.

Nanoparticle-reinforced composite hydrogels have been widely used for tissue engineering and biomedicine. In particular, hydrogels with multiple functionalization, such as self-healing, antibacterial, and cytocompatible properties, can facilitate their use in wound-healing and bio-adhesive applications. In this work, the multifunctional composite hydrogels were fabricated using silk fibroin (SF) and gelatin methacrylate (GelMA), incorporating silver-doped reduced graphene oxide (Ag@rGO) nanosheets and curcumin-loaded PF127 (PF127-Cur) micelles. The obtained hydrogels possessed an interconnected microstructure, good swelling properties (250–350%), and strong adhesion to the surface of different matrices, including rubber, metal, glass, plastic, wood, and finger skin. In addition, the hydrogels demonstrated self-healing, injectability, and electrical conductivity. Furthermore, the designed composite hydrogels demonstrated good antibacterial properties against the typical bacteria, Gram-positive S. aureus and Gram-negative E. coli . CCK-8 assay, fluorescent images, and cell scratch assay confirmed that the composite hydrogels exhibited excellent cytocompatibility and cell pre-migration ability. The obtained data confirmed that the designed hydrogels possessed great potential for biomedical applications, such as wound healing and bio-adhesives.

Hydrogels, due to their excellent properties, were widely noted and held great potential for application in wound healing. However, conventional wound dressings are limited by insufficient antioxidant and antibacterial properties, which restricted their effectiveness in treating infected wounds. In this study, a multifunctional hydrogel (PCS/Cur@Fe&TA) was fabricated from carboxymethyl chitosan, polyvinyl alcohol, and sodium alginate via a freeze-thaw cycle method to evaluate its potential as a wound dressing. The hydrogel incorporated nanocomplex composed of curcumin, tannic acid, and Fe 3+ , which endowed the material with enhanced antioxidant and antibacterial properties. The resulting hydrogel exhibited excellent swelling capacity (400–800%) and water holding capacity (>62%), and a good degradation rate, enabling it to maintain a moist microenvironment conducive to wound healing. Moreover, after hydrogel treatment, the cell viability remained >80%, >99% hemocompatibility (hemolysis rate < 1%), and significantly enhanced antioxidant enzyme activities ( P < 0.05). The hydrogel showed marked antibacterial activity, with inhibition zones of 1.03 cm against E. coli and 0.61 cm against S. aureus . Overall, the developed hydrogel exhibits considerable promise as a therapeutic material for accelerating wound healing. Download: Download high-res image (157KB) Download: Download full-size image

Wound healing in skin infections is frequently compromised by multi-drug-resistant bacterial colonization, persistent inflammation, and bacterial biofilm formation. To address these issues, we engineered an innovative multifunctional biomaterial (Cur@MIL-101(Cr/Ag)) which integrates potent antibacterial, anti-biofilm, and tissue-regenerative properties. This composite was constructed via a bimetallic synergistic and molecularly confined strategy, where Ag + and Cr 3+ were co-anchored within a terephthalic acid-based framework (MIL-101) through hydrothermal method, followed by the in situ encapsulation of curcumin. In vitro evaluations revealed potent broad-spectrum antibacterial activity against both Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ), along with significant inhibition of biofilm formation and minimum inhibitory concentrations (MICs) of 8 ppm and 2 ppm, respectively. In vivo, topical application of Cur@MIL-101(Cr/Ag) in a wound-infected mouse model further demonstrated complete bacterial eradication, mitigated inflammatory responses, accelerated wound closure, and promoted tissue regeneration with no observable systemic toxicity within the scope of assessment (i.e., stable body weight and non-pathological organ morphology). The therapeutic efficacy can be attributed to a synergistic triple-therapy mechanism comprising: (i) electrostatic interaction, (ii) ROS-mediated oxidative stress induction, (iii) synergistic interaction of multiple antimicrobial components (Ag⁺, Cr³⁺, and curcumin). This nanoengineered platform presents a clinically translatable strategy for precision therapy of infected wounds, combining antimicrobial robustness, biofilm penetration, and pro-regenerative functions within an integrated therapeutic system.

In the pursuit of optimal delivery of hydrophobic agents such as curcumin, the role of lipid composition in the efficacy of nanostructured lipid carriers is gaining significant attention. This study aimed to investigate the effects of cocoa butter (0–10 wt%) and medium chain triglycerides oil (0–10 wt%) mass ratios on processing stability, crystallization, polymorphism transition, supersaturation, encapsulation, and in vitro digestion of curcumin-loaded nanostructured lipid carriers (Cur-NLCs). Transmission electron microscopy (TEM) analysis showed that spherical Cur-NLCs exhibited larger average particle size with the increase in cocoa butter content. Differential scanning calorimetry (DSC) and X-ray diffraction (XRD) revealed that the crystallinity and thickness of solid shells decreased with the increase in liquid lipid content. The obtained Cur-NLCs remained stable under various conditions: thermal treatments (25, 37, 60, and 121 °C), varied ionic strengths (50–500 mM), pH changes (2.0–6.0), centrifugation (21382× g for 40 min), and 60-days storage. Furthermore, Cur-NLCs showed higher supersaturation (1863 ± 20–2328 ± 10%) than that of nanoemulsion (1489 ± 6%) and solid lipid nanoparticles (1635 ± 36%). In addition, the prepared Cur-NLCs exhibited high bioaccessibility (79.4 ± 3.1–91.1 ± 2.1%) along with a controlled release of curcumin. This study provides guidelines to prepare Cur-NLCs with tailored physicochemical, functional, and digestion properties to deliver hydrophobic bioactive substances, nutraceuticals, and pharmaceuticals.
