The mortality rate of primary liver malignant tumors is very high. Curcumin (Cur) is one of the most commonly used drugs for the treatment of liver cancer. However, due to its low bioavailability, nano-drug delivery systems are rapidly developing. Zeolitic imidazolate framework-8 (ZIF-8), is pH-sensitive and biodegradable in acidic environments, providing a very suitable platform for drug delivery. Here, the anti-tumor properties of Cur and the anti-inflammatory properties of gold nanoparticles (Au NPs) were integrated and encapsulated in ZIF-8 material to prepare a composite drug delivery system (Cur/Au@ZIF-8). The prepared materials were characterized by scanning electron microscopy and elemental analysis, transmission electron microscopy, and powder X-ray diffraction. The drug loading efficiency of Cur in this delivery system reached 47.16 %. The drug release curve showed that the release rate of Cur/Au@ZIF-8 was faster at pH 5.5, with a release rate of 72.34 %. Ultrasound-guided in situ liver cancer models were constructed in SD rats to investigate the in vivo anti-tumor effects of Cur/Au@ZIF-8 and its impact on the metabolomics of tumor-bearing rats before and after treatment. In vivo results indicated that Cur/Au@ZIF-8 showed better anti-tumor effects, with a tumor inhibition rate of 46.81 %. After treatment, 26 key differential metabolites (p < 0.05) were identified from expression patterns and metabolic pathways, which were mainly related to fatty acid metabolism. This treatment strategy provides a reference for future cancer treatment and mechanism exploration.

The recurrence following the discontinuation of medication is a formidable challenge in managing psoriasis. Changes in the microbiome accompany the onset of psoriasis relapse, highlighting a potential therapeutic modality. To evaluate the superiority of the topical administration of aptamer-functionalized curcumin mesoporous silica (Apt-GA+Cur@μmS) plus blue light (BL) in restoring dysbiosis and intervening in recurrence in a murine model, a psoriasis relapse murine model with double imiquimod induction was established. With a BL-responsive shell, Apt-GA+Cur@μmS released curcumin (Cur) to assist BL to improve the preventative and therapeutic effects in the psoriasis relapse murine model, as evidenced by the psoriasis area and severity index, histology, splenic index, and dorsal IL-17A level. We also observed a negative correlation between splenic nitric oxide (NO) levels and the splenic index, indicating a possible mechanism by which Apt-GA+Cur@μmS&BL may function in the treatment of splenomegaly. Treatment with Apt-GA+Cur@μmS&BL exhibited a higher alpha diversity than the model group, with levels similar to those of healthy mice, indicating that this combination could adjust the composition of the dorsal microbiome to a healthier state. A reduction in the combined relative abundance of Staphylococcus, Streptococcus, and Corynebacterium as well as restoration of dysbiosis was also verified through 16S rDNA gene sequencing in vivo. Collectively, BL and Apt-GA+Cur@μmS cotherapy alleviates psoriasiform lesions in a double imiquimod-induced murine model by inhibiting IL-17A and increasing splenic NO. Additionally, this cotherapy restores the eubiosis of the dorsal lesions. Thus, it is a promising and innovative therapeutic modality for psoriasis inflammation alleviation and recurrence intervention.

CUR Pickering emulsion exhibited uniformity and stability. • CUR Pickering emulsion enhanced CS-based coating bioactivity and sustained release. • PE/CS coating maintained postharvest quality for plum fruits. Curcumin is a promising natural preservative, but its poor solubility and susceptibility to photo-oxidation limit its application in active coatings. This study developed a cellulose nanofiber-stabilized curcumin Pickering emulsion incorporated into chitosan for postharvest plum preservation. The Pickering emulsion prepared at a water-to-oil ratio of 40:10 ( w / w ) exhibited uniform droplet distribution and good electrostatic stability. Its incorporation enhanced the mechanical performance and water resistance of PE/CS coating films, while FTIR and XRD analyses indicated hydrogen-bonding interactions within the composite matrix. Notably, PE/CS coating films showed sustained curcumin release in 50% ethanol, reaching 65% after 96 h. This controlled-release behavior was associated with enhanced antimicrobial and antioxidant activities, as reflected by inhibition zones above 10 mm and DPPH/ABTS + scavenging rates exceeding 98%. PE/CS coating effectively delayed flesh browning, water loss, and firmness decline in plums, providing a Pickering emulsion-based strategy for active fruit preservation. Comprehensive Summary The massive use of antibiotics in aquaculture and resulted antibacterial resistance problem urgently need antibiotic substitutions. Herein, we report a promising substitution of aquaculture antibiotics using a synergistic combination of biodegradable peptide polymers and curcumin, a natural compound from plant. The synergistic combination shows strong antibacterial activity against V. fluvialis and some other common bacteria in aquaculture. The membrane-damaging antibacterial mechanism echoes our finding that the synergistic combination will not induce bacteria to develop resistance after continuous use. The synergistic combination also displays effective cure on V. fluvialis -infected zebrafish. The biodegradability of the peptide polymer enables the combination to lose antibacterial activity and will not cause selective pressure on bacterial in the environment. Our study indicates potential application of synergistic composition, biodegradable peptide polymer and curcumin, as promising antibiotic substitution in aquaculture, which represents a promising strategy to address the global challenge of antimicrobial resistance.

After an osteosarcoma excision, recurrence and bone defects are significant challenges for clinicians. In this study, the curcumin (Cur) loaded chitosan (CS) nanoparticles (CCNP) encapsulated silk fibroin (SF)/hyaluronic acid esterified by methacrylate (HAMA) (CCNPs-SF/HAMA) hydrogel for the osteosarcoma therapy and bone regeneration was developed by photocuring and ethanol treatment. The micro or nanofibers networks were observed in the CCNPs-SF/HAMA hydrogel. The FTIR results demonstrated that alcohol vapor treatment caused an increase in β-sheets of SF, resulting in the high compression stress and Young’s modulus of CCNPs-SF/HAMA hydrogel. According to the water uptake analysis, SF caused a slight decrease in water uptake of CCNPs-SF/HAMA hydrogel while CCNPs could enhance the water uptake of it. The swelling kinetic results showed that both the CCNPs and the SF increased the swelling ratio of CCNPs-SF/HAMA hydrogel. The accumulative release profile of CCNPs-SF/HAMA hydrogel showed that the release of Cur from CCNPs-SF/HAMA hydrogel was accelerated when pH value was decreased from 7.4 to 5.5. Besides, compared with CCNPs, the CCNPs-SF/HAMA hydrogel had a more sustainable drug release, which was beneficial for the long-term treatment of osteosarcoma. In vitro assay results indicated that CCNPs-SF/HAMA hydrogel with equivalent Cur concentration of 150 μg/mL possessed both the effect of anti-cancer and promoting the proliferation of osteoblasts. These results suggest that CCNPs-SF/HAMA hydrogel with superior physical properties and the bifunctional osteosarcoma therapy and bone repair may be an excellent candidate for local cancer therapy and bone regeneration. Keywords:curcumin;hyaluronic acid;silk fibroin;chitosan nanoparticle;MG-63 cells;MC3T3-E1 cells

The devastating occurrence of drug resistance such as antimicrobial resistance has aroused global concerns for public health, which has propelled a continuous pursuit of safe and effective therapeutic agents. In this study, silver nanoparticles were decorated in mesoporous silica of SBA-15 coated with melanin-like polydopamine (PDA) as nanocarriers. Meanwhile, the constructed mesopore was loaded with phytochemical curcumin (CCM) through its noncovalent interactions with PDA coatings. The obtained CCM@SBA-15/PDA/Ag composites were characterized by physicochemical methods and exhibited desirable biocompatibility and low hemolytic activity. The dual-stimuli-responsive (pH and ROS) release of curcumin and/or silver nanoparticles from the CCM@SBA-15/PDA/Ag composites was achieved to reduce the side effects of noncontrolled drug leakage under physiological conditions. Additionally, compared with that of SBA-15/PDA/Ag and CCM@SBA-15/PDA, CCM@SBA-15/PDA/Ag combination showed a prolonged inhibitory effect on bacterial growth of G–E. coli (72 h) and G+S. aureus (24 h), attributing to the enhanced effect of the bactericide of silver nanoparticles and curcumin. Furthermore, through the utilization of the nanoformulation of curcumin, improved chemotherapeutic efficiency against human cervical cancer cells (HeLa) and Taxol-resistant nonsmall cell lung cells (A549/TAX) was identified in comparison with that of free curcumin. Thus, our study rationalized the combinational design of the natural compound and silver nanoparticles as an integrated dual-responsive nanoplatform in dealing with infectious bacteria and drug resistance in cancers for enhanced therapy.

This study aimed to design a new-type curcumin (Cur)-based network nanohydrogel (H), which was composed of whey protein isolate nanofibers (WPINs) and pectin (P). Among them, WPINs were prepared with subcritical water at different treatment times (90 min: WPINs-90, 120 min: WPINs-120, and 180 min: WPINs-180). The structural characteristics, rheology and gastrointestinal release kinetics of Cur-based nanohydrogel were investigated. The results showed that WPINs-120-P-H showed the most favorable particle size (659.00 ± 3.90 nm), potential (−30.00 ± 2.00 mV), peak temperature (110.57 °C) and heating stability. The rheological results revealed that each group of nanohydrogels formed strong elastic gels. In addition, WPINs-120-P-H exhibited the highest encapsulation efficiency (98.42 ± 5.00%) of Cur with the strongest network structure. Moreover, the release curved line of Cur showed an initial release of 19.23% in the stomach, and afterwards, a continuous release trend in the small intestine, reaching a final release amount of 84.80%. Interestingly, the release kinetics of Cur was in line with the Weibull equation model, evidencing a continuous and dynamic release process. This study provided a novel way to prepare the WPINs-P nanohydrogel as delivery for Cur, which demonstrated great potential application prospects in the functional food industry.

Curcumin-loaded DES was successfully encapsulated in hydrogel beads. • Water distribution and microstructure varied with HAMS content. • Higher-HAMS formulations showed slower intestinal-stage curcumin release. • DES-hydrogel integration provides a simple strategy for hydrophobic delivery. Curcumin is a hydrophobic bioactive compound with limited oral applicability due to its poor water solubility and low stability. In this study, high-amylose maize starch (HAMS)-containing hydrogel beads were developed for the encapsulation of curcumin-loaded natural deep eutectic solvent (CLDES). Sodium alginate (SA) formed an ionically crosslinked hydrogel matrix through Ca 2+ -induced gelation, while HAMS contributed to additional physical associations within the hydrogel system. The hydrogel beads showed HAMS-content-dependent differences in their structural properties. The average bead diameter increased from 2.41 ± 0.02 mm (H0) to 3.14 ± 0.02 mm (H25). SEM showed a progressively denser internal structure with increasing HAMS content. LF-NMR analysis revealed differences in water mobility and distribution among formulations with different HAMS contents. In addition, the maximum thermal degradation temperature increased from 248.3 °C (H0) to 313.7 °C (H35), indicating formulation-dependent differences in thermal degradation behavior at higher HAMS levels. The swelling ratio and water holding capacity decreased with increasing HAMS content, while the loading capacity gradually stabilized at higher HAMS levels. Under simplified simulated gastrointestinal conditions, all hydrogel beads exhibited limited curcumin release in simulated gastric fluid (below 3% within 2 h), followed by markedly increased release in simulated intestinal fluid. Release kinetic analysis suggested that the release behavior was associated with combined effects of network relaxation, swelling, and structural disintegration rather than simple Fickian diffusion. Overall, the HAMS-containing CLDES hydrogel beads developed in this study provide a facile strategy for the encapsulation and controlled release of hydrophobic bioactive compounds. Curcumin improved survival under chronic heat stress in Siberian sturgeon. • Curcumin alleviated heat-induced oxidative stress in the sturgeon heart. • Curcumin suppressed cardiomyocyte apoptosis and restored Hsp90 levels. • Curcumin restored mitochondrial membrane potential and cardiac ATP levels. • Curcumin moderated heat-induced changes in glycolysis and the TCA cycle. Global warming increasingly threatens cold-water aquaculture, yet nutritional strategies for protecting the fish heart from chronic heat stress remain insufficiently understood. This study evaluated the protective effects of dietary curcumin against chronic heat stress (28 °C) in the hearts of Siberian sturgeon ( Acipenser baerii ) using histological, biochemical, molecular, and metabolomic analyses. Heat stress caused cardiac injury, as indicated by inflammatory infiltration, mitochondrial vacuolization, increased serum cardiac enzymes (CK, CK-MB, and α-HBDH) and glucose, and elevated myocardial reactive oxygen species (ROS), superoxide dismutase (SOD) activity, and malondialdehyde (MDA) content. Curcumin supplementation attenuated these alterations and enhanced catalase (CAT) and glutathione peroxidase (GSH-Px) activities. Curcumin also reduced cardiomyocyte apoptosis by downregulating caspase-3 , caspase-7 , and caspase-9 and restoring heat shock protein 90 (Hsp90) levels. In addition, curcumin improved mitochondrial membrane potential (MMP), Ca 2+ /Mg 2+ -ATPase activity, and ATP content. Metabolomic profiling indicated that heat stress altered energy-related metabolites such as ATP and isocitrate, whereas curcumin partially restored the metabolic profile, most notably ATP. These findings support dietary curcumin as a practical nutritional strategy for improving cardiac protection and thermal resilience in Siberian sturgeon aquaculture. Background Abnormally regulated long non-coding RNAs (lncRNAs) functions in cancer emphasize their potential to serve as potential targets for cancer therapeutic intervention. LncRNA ASBEL has been identified as oncogene and an anti-sense transcript of tumor-suppressor gene of BTG3 in triple-negative breast cancer (TNBC). Results Herein, multicomponent self-assembled polyelectrolyte nanocomplexes (CANPs) based on the polyelectrolytes of bioactive hyaluronic acid (HA) and chitosan hydrochloride (CS) were designed and prepared for the collaborative modulation of oncogenic lncRNA ASBEL with antago3, an oligonucleotide antagonist targeting lncRNA ASBEL and hydrophobic curcumin (Cur) co-delivery for synergetic TNBC therapy. Antago3 and Cur co-incorporated CANPs were achieved via a one-step assembling strategy with the cooperation of noncovalent electrostatic interactions, hydrogen-bonding, and hydrophobic interactions. Moreover, the multicomponent assembled CANPs were ulteriorly decorated with a near-infrared fluorescence (NIRF) Cy-5.5 dye (FCANPs) for synchronous NIRF imaging and therapy monitoring performance. Resultantly, MDA-MB-231 cells proliferation, migration, and invasion were efficiently inhibited, and the highest apoptosis ratio was induced by FCANPs with coordination patterns. At the molecular level, effective regulation of lncRNA ASBEL/BTG3 and synchronous regulation of Bcl-2 and c-Met pathways could be observed. Conclusion As expected, systemic administration of FCANPs resulted in targeted and preferential accumulation of near-infrared fluorescence signal and Cur in the tumor tissue. More attractively, systemic FCANPs-mediated collaborative modulating lncRNA ASBEL/BTG3 and Cur co-delivery significantly suppressed the MDA-MB-231 xenograft tumor growth, inhibited metastasis and extended survival rate with negligible systemic toxicity. Our present study represented an effective approach to developing a promising theranostic platform for combating TNBC in a combined therapy pattern.

A pH-responsive double-layer film comprising chitosan (CS), zein (Z), curcumin-β-cyclodextrin complex (Cur- β-CD), alizarin (AL) and cinnamaldehyde (CIN) was developed to detect spoilage and prolong the shelf life of pork. The outer Z/AL is the colorimetric responsive and highly protective outer layer, while the CS/Cur- β-CD/CIN is the fluorescence responsive and functional layer. The CS/Cur- β-CD/CIN-Z/AL film demonstrated excellent barrier properties against oxygen (8.48 × 10 -13 g (m s Pa) -1 ) and water (2.42 × 10 -11 g (m s Pa) -1 ) primarily due to the polar interactions in the Z structure and its hydrophobic nature. The addition of AL and Cur provided the CS/Cur- β-CD/CIN-Z/AL film with pH colorimetric and fluorescence response capabilities, respectively. Furthermore, the inclusion of Cur- β-CD, CIN and AL significantly enhanced the film’s antioxidant and antibacterial properties, with radical scavenging rates of 79.29 % (DPPH) and 89.84 % (ABTS), as well as antibacterial efficiencies of 96.2 % ( S. aureus ) and 85.78 % ( E. coli ). To test the effectiveness of the double-layer film, a freshness monitoring and maintenance experiment was conducted on pork stored at 4 °C for 8 days. Various parameters of pork, including surface color, pH value, total volatile bases nitrogen (TVB-N), total viable counts (TVC), thiobarbituric acid reactive substance (TBARS), hardness and springiness, were measured, along with the color and fluorescence intensity (FL) of the film. The shelf life of the pork was extended by at least 2 days compared to the control group, and the pork was considered inedible if ΔE ≥ 12 or FL intensity ≤ 2160. Overall, this food packaging film shows promise in simultaneously monitoring and maintaining the freshness of pork.

Aim We aimed to design RGD-anchored liposomes encapsulating an antipyroptosis drug that could efficiently target macrophages and relieve the rate of cytokine release syndrome, providing a new strategy for sepsis treatment, especially sepsis-induced acute renal injury. Background Sepsis is a clinical syndrome of life-threatening organ dysfunction caused by host response disorders due to infection. Sepsis has a high incidence and remains one of the leading causes of death worldwide. Objective Macrophage-mediated pyroptosis plays an important role in the occurrence and development of cytokine release syndrome and organ injury caused by sepsis. Curcumin can inhibit inflammasome assembly and slow the progression of pyroptosis by scavenging intracellular reactive oxygen species, but it has poor water solubility and low bioavailability. The emergence of drug-delivery nanosystems has overcome this problem, but there is still a lack of research on how to accurately deliver antipyroptotic drugs to innate immune cells and subsequently hinder pyroptosis. Methods We constructed a curcumin-loaded RGD-modified liposome (RGD-lipo/Cur) and demonstrated that RGD-lipo/Cur could effectively target macrophages. Results In vitro , RGD-lipo/Cur reduced the upregulation of caspase-1, caspase-3, NLRP3, IL-1β and GSDMD, inhibiting pyroptosis, reducing oxidative stress, and attenuating the proinflammatory cytokine cascade. Conclusion RGD-lipo/Cur was considered to have great potential for sepsis treatment.
