Interfacial solar vapor generation (SVG) has emerged as a promising strategy for water purification and desalination. Among SVG technologies, hydrogel-based evaporators stand out for their high energy efficiency and effective evaporation, but their performance often collapses under high irradiance due to insufficient water transport. Here we develop an ultralow-density rigid-network (ULR) hydrogel engineered for ultrafast water transport and evaporation. The ULR network maximizes water content and establishes steep osmotic-pressure gradients for rapid water supply, while the rigid, anti-shrinkage framework preserves hierarchical pores, sustaining capillary-driven flow and maintaining continuous vapor production under extreme irradiance. ULR evaporators surpass traditional hydrogels’ theoretical maximum water transport rate, achieving an evaporation rate of 25.57 kg m⁻² h⁻¹ at 10 suns for 100 h. In practical trials, a low-cost module produced 138 L m⁻² day⁻¹, yielding 12.42 L day⁻¹ of potable water. This design advances hydrogel-based SVG toward robust, affordable solutions for real-world water scarcity.

A wearable epifluidic patch featuring a hierarchical dual-lock mechanism is introduced. Tetrahedral DNA nanostructures co-localize enzymes at defined spacing to promote substrate channeling and accelerate kinetics, while a gelatin hydrogel stabilizes the enzymatic microenvironment during hydration cycles, ensuring uniform reaction conditions for homogeneous signal formation. This design produced a visible glucose-responsive signal within 30 s, with a linear range of 50–400 µM and a detection limit of 9.98 µM. Integrated with passive microfluidics and smartphone imaging, the patch enables sequential sampling in human trials conducted during physical exercise. The hierarchical immobilization method, device design, and system integration outlined demonstrate a platform for intuitive, electronics-free colorimetric sensing in dynamic environments. Diabetes aggravates postoperative tumor recurrence and impairs wound healing due to divergent metabolic adaptations to hyperglycemia in tumor versus normal cells. Current therapeutics fail due to the incapabilities in adaptively modulating the metabolic bifurcation across the contradictory pathological contexts. Here, we address this challenge by developing a platinum (Pt) single-atom nanocatalyst (PtSNC) with microenvironments-selective multienzyme-mimicking activities to pioneer a bi-polar bioenergetic intervention strategy. In acidic and reduced nicotinamide adenine dinucleotide (NADH)-overexpressed tumor niches, it exhibits NADH oxidase (NOX)-, oxidase-, and peroxidase-like activities, depleting NADH reserves and generating highly reactive Pt = O species to disrupt energy metabolism and induce both apoptosis/ferroptosis. While it shows NOX-, catalase-, and superoxide dismutase-like activities in neutral diabetic wounds, rectifying hyperglycemia-induced cellular NAD + /NADH abnormity and bioenergetic disorder to revitalize the cells and tissues, with promoting angiogenesis and mitigating local inflammation to accelerate regeneration. Murine diabetic melanoma resection models demonstrate its extensive capacity in effectively eradicating residual tumor tissues and suppressing the recurrence, and promoting diabetic wound healing concurrently without systemic toxicity, making PtSNC a safe and potent nanotherapeutic for diabetic tumor postoperative therapy. This study holds promise for the application of single-atom catalytic medicines in precision therapy for intractable diseases featuring pathological metabolic bifurcation. Electrocatalytic water splitting is a clean and sustainable method for hydrogen production. Cobalt diselenide (CoSe 2 ) is an electrocatalyst with great application prospects. However, due to the inherent low electrical conductivity and aggregation tendency of transition metal selenides, their practical applications are limited. In this study, under the constraint of three-dimensional porous nitrogen-doped reduced graphene oxide/nickel foam (N-rGO/NF) substrates, small-sized nitrogen-doped cobalt diselenide (CoSe 2-x N x ) nanoneedles were prepared by a one-step hydrothermal method, and excellent electrocatalysts were obtained. The optimized catalyst (N-rGO/CoSe 2-x N x /NF-100) exhibits excellent OER performance in an alkaline environment at current densities of 10, 50 and 100 mA cm −2 . N-rGO/NF not only provides good electrical conductivity for the electrocatalyst, but also limits the size of CoSe 2 . The three-dimensional porous structure constructed by small-sized CoSe 2 nanoneedles increases the specific surface area of the electrocatalyst, which is conducive to the OER process. The doping of nitrogen in CoSe 2 forms Co-Nx electron transfer channels, accelerating charge transfer. Density functional theory (DFT) calculations have confirmed that structural modification methods such as limiting the size of the electrocatalyst to avoid agglomeration, as well as electronic control methods such as nitrogen doping, can accelerate the surface structure reconstruction process of the catalyst, thereby improving the adsorption energy of the electrocatalyst for OER intermediates and reducing the overpotential.

Developing highly efficient noble-metal-free catalysts for oxygen reduction reaction (ORR) is critical for the widespread commercialization of metal-air batteries. Facet engineering provides an effective strategy to optimize the catalysts’ activity, while the controlling facet orientation in non-precious metals remains challenging. Here we show a substrate-induced growth strategy to synthesize copper nanosheets with specific (200) and (111) facet orientations over nitrogen/oxygen co-doped graphene (N/O-C) via chemical vapor deposition (Cu(200)@N/O-C and Cu(111)@N/O-C), revealing a distinct facet-dependent activity for ORR. It clarifies the substrate oxygen coordination promotes preferential Cu(200) growth under moderate deposition, whereas excess vapor drives thermodynamically favored Cu(111) formation. Theoretical and experimental analyses demonstrate Cu(200) facet in N/O-C optimizes the adsorption energies of ORR intermediates and lowers the reaction barrier for the rate-determining step to outperform the Cu(111), which also exhibits better activity compared to commercial Pt/C. When integrated into zinc-air batteries, Cu(200)@N/O-C delivers a power density of 222.5 mW·cm -2 and stable operation for over 1000 hours, highlighting the potential of facet-engineering on earth-abundant Cu for next-generation energy storage.

The peptide-centric local stability assay (PELSA) was developed to identify the protein targets and binding regions of diverse ligands at a proteome-wide scale. In this approach, the ligand is added to a cell lysate, and the proteins are digested for a short, defined time period using 0.5 mg ml −1 trypsin. Protein regions involved in ligand binding show less digestion, making it possible to identify binding sites and even quantify binding activity by dose-dependent analysis. In contrast to modification-based techniques that require chemical modification of ligands, the ligand modification-free property of the PELSA offers a key advantage by avoiding derivatization, thereby enabling proteome-wide identification of target proteins across diverse ligand types and biological contexts. The PELSA is broadly applicable for identifying protein targets of diverse ligands such as drugs and antibodies as well as endogenous metabolites and metal ions. However, performing PELSA experiments and subsequent data analysis to extract protein–ligand interaction details, including the binding protein, binding region and binding affinity, is not a trivial task. To address this, we present a detailed protocol for the experimental and computational PELSA workflow, including raw data processing and result visualization with the PELSA-Decipher software ( https://zenodo.org/records/18266507 ). Key points, such as selection of ligand concentrations, timing of trypsinization and quality control between different replicates, are discussed. Finally, we demonstrate the application of the protocol for identifying targets of staurosporine and 5-methyltetrahydrofolate, as well as for the dose-dependent PELSA analysis of inhibitors targeting the HSP90 family. The full PELSA protocol can be finished in 2 days, 1 day for sample preparation and 1 day for liquid chromatography–tandem mass spectrometry analysis and data processing. PELSA-Decipher can be downloaded from GitHub ( https://github.com/DICP-1809/ ).

The healing of the tendon-bone interface remains a significant challenge due to the lack of suitable bioactive materials for promoting interface regeneration. To address this, we developed a photosensitive hydrogel system designed to mimic the extracellular matrix. This system is based on light-induced imine crosslinking between type I collagen (Col I) and o-nitrobenzyl alcohol (NB)-modified hyaluronic acid (HANB). Experimental results demonstrate that the hydrogel (Col I-HANB) exhibits excellent controllability, low swelling rate, biodegradability, and tissue-adhesive properties. Using a fluorescent dye (8-hydroxypyrene-1,3,6-trisulfonic acid trisodium salt, HPTS) as a model molecule, we confirmed that the hydrogel enables localized and sustained release, high-lighting its potential as a drug-delivery platform for tissue engineering. To further enhance tendon-bone interface repair, the hydrogel was functionalized with two key components: (I) The E7 peptide, known for its stem cell-homing properties, was covalently immobilized into the hydrogel network via conjugation to bovine serum albumin (BSA) and followed by imine-ligation between UV-triggered aldehyde generation from HANB and BSA's amino groups; and (II) Kartogenin (KGN)-loaded nanoparticles (KGN-NPs) were incorporated to potentially promote chondrogenic differentiation, with release assays confirming sustained KGN delivery over 21 days. The composite hydrogel demonstrated excellent biocompatibility in cytotoxicity and live/dead assays. Importantly, in a rabbit rotator cuff tear model, it effectively promoted fibrocartilage regeneration and significantly improved tendon-bone healing, creating a favorable microenvironment for repair. The results above indicate that the hydrogel system provides a good microenvironment for rotator cuff injury repair and is a promising scaffold applied in treatment of rotator cuff injury.

For the first time, a series of N-doped graphitic carbon-modified g-C 3 N 4 samples were synthesized via thermal polycondensation by incorporating trace amounts of chitosan into melamine precursors. The photocatalytic performance of the samples was evaluated through Rhodamine B (RhB) degradation under visible-light irradiation. The photocatalyst prepared with 50 mg chitosan achieved complete degradation of 10 mg/L RhB within 30 min, outperforming pristine g-C 3 N 4 . After five consecutive cycles, the photocatalyst maintained 91% degradation efficiency within 30 min while preserving its original crystalline structure, demonstrating outstanding stability and reusability. The large specific surface area, enhanced electron transfer efficiency, extended visible light absorption and low photogenerated electron–hole recombination rate were considered to contribute to the extraordinary visible-light-driven (VLD) photocatalytic activity of N-doped graphitic carbon-modified g-C 3 N 4 . Furthermore, a scavenger study was conducted to identify the dominant reactive oxygen species (ROS) involved in the photocatalytic degradation of RhB, providing deeper insights into the underlying reaction mechanisms.

Radioactive organic wastewater management concerning metal source recovery and organic matter degradation conforms well to the notion of circular economy and environmental protection, but the concurrent conversion in an integrated process is challenging and has been rarely studied. Here, we introduce a high-performance photoelectrochemical system for uranyl extraction and the simultaneous degradation of co-existing urea to manage the treatment of uranyl-containing radioactive organic wastewater generated from the production of nuclear fuel for fourth-generation high-temperature gas-cooled reactor. Cost-efficient self-standing Ni-modified TiO 2 nanotube arrays (Ni/TiO 2 ) and β-cyclodextrin polymer (P-CDP@CC) were used as photoanode and cathode, respectively. Concurrent photoanodic urea degradation and cathodic uranyl reduction towards UO 2 from low-level radioactive organic wastewater demonstrated a uranyl extraction efficiency of 99.08% and the urea degradation efficiency of nearly 100% under a current density of 4.5 mA/cm 2 within 8 hours in a single-compartment electrolysis cell. This work provides an effective, environmentally benign approach to upcycle uranium resources and purify organic pollutants from complicated wastewater matrix.

Pseudomonas aeruginosa (PA) biofilm-associated chronic lung infections pose a significant therapeutic challenge due to the limited penetration and efficacy of conventional antibiotics. In this study, we developed an innovative dry powder inhaler (DPI) formulation co-loading ciprofloxacin-encapsulated liposomes (CIP-Lips) and S-nitrosoglutathione (GSNO) using an ultrasonic spray-freeze-drying (USFD) technique. GSNO, a nitric oxide (NO) donor, promotes biofilm dispersion and induces bacterial phenotypic switching, thereby enhancing the accessibility of CIP-Lips to biofilm-embedded bacteria. Subsequently, CIP released from the liposomes exerts direct antibacterial activity. The combination of CIP-Lips and GSNO produces a synergistic “biofilm-disrupting and bactericidal” therapeutic effect, leading to effective biofilm eradication. The optimized DPI formulation exhibited satisfactory in vitro aerosol performance, with a fine particle fraction of 36.51 ± 0.49%, ensuring efficient lung deposition. In vitro evaluations further confirmed pronounced biofilm disruption and bactericidal efficacy. Furthermore, in a murine model of chronic pulmonary infection, the CIP-Lips/GSNO DPI significantly reduced the bacterial burden without observable systemic toxicity. This inhalable system represents a promising strategy for the treatment of PA biofilm-associated lung infections.

Oxidative stress is a major driver of granulosa cell (GC) dysfunction and disrupted steroidogenesis. However, the mechanisms underlying oxidative stress–induced progesterone synthesis impairment and potential intervention strategies remain inadequately defined. In this study, RNA-seq re-analysis revealed that hydrogen peroxide (H₂O₂) markedly altered gene expression in mouse granulosa cells (mGCs), particularly affecting pathways related to progesterone biosynthesis. In vitro experiments confirmed that H₂O₂ suppresses progesterone production via downregulation of Sirtuin 1 (SIRT1). Conversely, β-nicotinamide mononucleotide (NMN) supplementation increased intracellular NAD⁺ levels, and upregulated SIRT1 and Nicotinamide phosphoribosyl transferase (NAMPT) expression. This regulation enhanced cell proliferation, stimulated the expression of progesterone biosynthetic enzymes, and promoted NAMPT transcriptional activity. Notably, the beneficial effects of NMN were diminished upon SIRT1 inhibition. Similarly, inhibition of NAMPT attenuated the positive regulatory effect of NMN, leading to a marked reduction in SIRT1 levels and a consequent suppression of progesterone secretion. Thus, SIRT1 activation induces NAMPT expression, which in turn feeds back to enhance SIRT1 activity and support progesterone biosynthesis. Together, these findings suggest that NMN alleviates H₂O₂-induced progesterone synthesis disruption in mGCs through activation of the NAD⁺/SIRT1/NAMPT axis, providing a potential therapeutic strategy for oxidative stress–related reproductive dysfunction.

Paeoniflorin (PF) is a bioactive monoterpene glycoside with well-established anti-inflammatory properties; however, its therapeutic potential and molecular mechanism in nephrolithiasis remain insufficiently defined. In this study, network pharmacology was employed to predict potential targets of PF. In vitro, HK-2 cells exposed to calcium oxalate (CaOx) crystals were treated with PF to evaluate inflammatory responses, crystal adhesion, and NF-κB activation. A glyoxylic acid-induced mouse model was used to assess renal crystal deposition, renal function, and expression of key regulatory targets, supported by transcriptomic profiling. Molecular docking and molecular dynamics simulations were performed to analyze the interaction between PF and Serpine1, and functional relevance was further examined using exogenous Serpine1 rescue experiments. Integrated analyses indicated that PF exerts anti-nephrolithic effects predominantly through modulation of inflammation-associated signaling. PF dose-dependently suppressed CaOx-induced NF-κB phosphorylation and nuclear translocation, reduced the release of pro-inflammatory cytokines (TNF-α, IL-6, and IL-1β), and decreased crystal adhesion in HK-2 cells. In vivo, PF markedly attenuated renal crystal deposition, improved renal functional indices, and downregulated renal Serpine1 and IL-1β expression. Computational simulations supported a stable interaction between PF and Serpine1 (binding energy − 6.22 kcal/mol). Consistently, supplementation with exogenous Serpine1 partially reversed PF-mediated inhibition of NF-κB activation and cytoprotective effects. Collectively, these findings suggest that PF mitigates CaOx-associated renal injury by suppressing inflammation-centered signaling programs involving the Serpine1/NF-κB axis, thereby providing mechanistic insight into PF as a potential multi-target therapeutic candidate for nephrolithiasis.
