A simple and sensitive sensor based on a molecularly imprinted polymer-modified carbon paste electrode for the determination of curcumin in foods

Q. Zhou,H. Y. Zhai,Y. F. Pan,K. Li

A highly sensitive and selective molecularly imprinted polymer-modified carbon paste electrode (MIP-CPE) was designed. The curcumin molecularly imprinted polymer (CUR-MIP) was prepared by thermally induced precipitation polymerization, using curcumin (CUR) as the template molecule, methacrylic acid (MAA) as the functional monomer, 2,2′-azodiisobutyronitrile (AIBN) as the initiator, and ethylene glycol dimethacrylate (EDGMA) as the cross-linker. The modified electrode was prepared by mixing the polymer, graphite powder, and paraffin oil in specific proportions. The electrochemical behaviour of CUR on this electrode was studied, and a method for direct CUR determination using this electrode was established. The results showed that CUR exhibited a distinct oxidation peak (Epa) at 0.434 V (vs. SCE) in the mixed phosphate buffer solution (PBS) at pH 3.06. The optimum addition ratio of the polymer in the modified electrode was 20%, and the peak current (Ipa) was 4.5-fold higher than that of the bare CPE. The Ipa showed a good linear relationship with concentration in the range 0.1–50 μM, and the detection limit was 10.1 nM. The method was applied to determine CUR contents in food, giving recovery rates of 90.77–105.7%.

Development and characterization of polyvinyl alcohol/chitosan crosslinked malic acid composite films with curcumin encapsulated in β-cyclodextrin for food packaging application

Shuting Cao,Hongzhen Liu,Ming Qin,Nannan Xu,Fuhao Liu,Yuetao Liu,Chuanhui Gao

Considering that fruits are vulnerable to damage and waste during stockpiling, transport and marketing. Given this, an innovative curcumin inclusion compound (Cur@β-CD) was devised in this study to introduce oil-soluble curcumin (Cur) into water-soluble polyvinyl alcohol (PVA) materials, thereby fabricating food packaging films endowed with excellent properties. DPPH test manifested that the oxidation resistance for PCOMC-Cur@β-CD film was 95 % above PVA material. It was ascribed to the fact that the Cur@β-CD elevated the water solubility of Cur while the increase of water solubility heightened the antioxidant effect for Cur in the film. Additionally, the chitosan (CS) was crosslinked with malic acid (MA), which elevated the barrier property of the film, reduced the amount of oxygen transmission and further retarded the oxidation reaction of the fruits for packaging. The antibacterial test demonstrated that the antibacterial rates of PCOMC-Cur@β-CD film against E. coli and S. aureus reached 92 % and 95 %, respectively, which was attributed to the slow release of Cur when Cur@β-CD was dissolved in PVA material and the Schiff base reaction between Cur and amino groups on CS. These findings indicate that the PCOMC-Cur@β-CD film developed in this work can provide certain insights into the field of food packaging.

Improving the prebiotic activity and oxidative stability of carboxymethyl curdlan − quercetin conjugates stabilized Pickering emulsions for the colonic targeting delivery of curcumin

Huan Li,Wenjiang He,Zijun Wang,Qun Zhang,Die Hu,Ke Ding,Qiutao Xie,Yanqun Xu,Yang Shan,Shenghua Ding

The carboxymethyl curdlan-quercetin conjugate (CMCD-QUE) was synthesized to stabilize curcumin (CUR) −loaded Pickering emulsions. The physicochemical properties, antioxidant activity, and prebiotic activity of CMCD-QUE were investigated. The effects of different concentrations of CMCD-QUE on CUR-loaded emulsions were also explored. CMCD-QUE exhibited excellent antioxidant activity throughout in vitro simulated gastrointestinal digestion. The particle size and electrostatic repulsion of CMCD-QUE gradually increased with rising pH, indicating that CMCD-QUE was pH-responsive. CMCD-QUE exhibited prebiotic activity and specifically promoted the enrichment of Akkermansia. The droplet size of the CMCD-QUE stabilized emulsions became smaller and more uniform as the carrier concentration increased. A concentration of 5% CMCD-QUE could be stably adsorbed at the oil–water interface of the emulsion, which exhibited a maximum encapsulated efficiency (95.85%) of CUR. CMCD-QUE showed excellent antioxidant capacity, pH responsiveness, and prebiotic activity, making it a promising multifunctional emulsifier for colon-targeted delivery of functional ingredients.

Nanocomposites Based on Layered Double Hydroxides Loaded with Curcumin and Carbon Nitride Quantum Dots for Combined Chemotherapy and Chemodynamic Therapy of Tumors

Jing Zhijie,Zhang Xiaorui,Jin Yimin,Wang Jianhua,Chai Chaofan,Bian Wei

Using multifunctional nanocomposites for multiple therapeutic approaches has emerged as an innovative cancer treatment strategy. Chemotherapy and chemodynamic therapy (CDT) have many benefits for cancer therapy, in which have great significance to develop nanocomposites that enable chemotherapy/CDT combined treatment. Herein, in situ co-precipitation method was used to load curcumin (Cur) into layered double hydroxide (LDH). Then, the Cur/LDH was assembled with carbon nitride quantum dots (CNQDs) through electrostatic adsorption. Acidic microenvironment–responsive nanocomposite (Cur/LDH/CNQDs) was constructed and used for tumor treatment. The as-synthesized Cur/LDH/CNQDs exhibits good biocompatibility and chemodynamic properties. In addition, the releases of Fe(II) ions (78%) and Cur (85%) are accelerated under acidic pH conditions that mimic the tumor microenvironment. The results show that the combination of chemotherapy and CDT can enhance its tumor-killing ability compared with single therapy. In essence, Cur/LDH/CNQDs shows a good effect in combined chemotherapy/CDT treatment of tumor cells and provide a promising alternative for combined tumor therapy.

Curcumin-loaded nanoparticles developed from sodium caseinate and OSA starch and the in vitro semi-dynamic elderly digestion behaviour of fortified yogurt

Ruijie Wang,Leiyu Deng,Weiping Jin,Qi Meng,Jianzhong Han,Weilin Liu

Summary Curcumin is a bioactive substance, while poor stability and low bioavailability limit its effective utilisation. Curcumin-loaded nanoparticles were prepared using the wall materials containing OSA starch alone (HI), sodium caseinate alone (SC), and a mixture of OSA starch and sodium caseinate (HS). Compared to HI and SC, HS had obvious network structure and smaller particle size, with the loading capacity higher than 20%. The storage stability at 4 °C were higher in HS and SC, FTIR and XRD results showed that all nanoparticles encapsulated curcumin through hydrogen bonding. When nanoparticles were added to yogurt, HI showed slower curcumin release during in vitro semi-dynamic elderly digestion, whereas SC and HS can better accelerate the digestion rate of proteins in yogurt, leading to a decrease in the apparent viscosity of digestive emptying products. This study provided a reference for curcumin delivery wall materials and pushed forward development of functional foods for elderly.

Electric-field-sensitive hydrogel based on pineapple peel oxidized hydroxyethyl cellulose/gelatin/Hericium erinaceus residues chitosan and its study in curcumin delivery

Huishuang Yin,Peiqin Song,Chunhui Zhou,Huihua Huang

This study focused on synthesis of innovative hydrogels with electric field response from modified pineapple peel cellulose and hericium erinaceus chitosan and gelatin based on Schiff base reaction. A series of hydrogels were prepared by oxidized hydroxyethyl cellulose , gelatin and chitosan at different deacetylation degree via mild Schiff base reaction. Subsequently experiments towards the characterization of oxidized hydroxyethyl cellulose/gelatin/chitosan (OHGCS) hydrogel polymers were carried out by FTIR/XRD/XPS, swelling performances and electric response properties. The prepared hydrogels exhibited stable and reversible bending behaviors under repeated on–off switching of electric fields, affected by ionic strength , electric voltage and pH changes. The swelling ratio of OHGCS hydrogels was found reduced as deacetylation degree increasing and reached the maximum ratio ∼ 2250 % for OHGCS-1. In vitro drug releasing study showed the both curcumin loading capacity and release amount of Cur-OHGCS hydrogels arrived about 90 % during 6 h. Antioxidation assessments showed that the curcumin-loaded hydrogels had good antioxidation activities, in which, 10 mg Cur-OHGCS-1 hydrogel could reach to the maximum of about 90 % DPPH scavenging ratio. These results indicate the OHGCS hydrogels have potentials in sensor and drug delivery system .

Borneol serves as an adjuvant agent to promote the cellular uptake of curcumin for enhancing its photodynamic fungicidal efficacy against Candida albicans

Zhenlong Zhou,Shengli Wang,Penghui Fan,Xiaofeng Meng,Xinyu Cai,Wen Wang,Lin Ma,Hang Ma,Jianyu Su

Candida albicans ( C. albicans ), a major opportunistic pathogenic fungus, is known to cause superficial skin infections. Unfortunately, the misuse of antibiotics has led to the emergence of drug resistance in fungi. Antimicrobial photodynamic therapy (aPDT), a non-antibiotic alternative, has shown potential in treating drug-resistant fungal infections. Curcumin is a photodynamically active phytochemical whose photodynamic fungicidal efficacy is largely dependent on its intracellular accumulation. However, curcumin faces challenges in penetrating the cytoplasm due to its poor water solubility and the fungal cell wall. Borneol, another monoterpenoid phytochemical, is known for its ability to enhance drug absorption. In this study, we showed that borneol improved the cellular uptake of curcumin, thereby enhancing its photodynamic fungicidal efficacy against C. albicans . This effect was attributed to borneol's ability to increase cell permeability. Transcriptomic analysis further confirmed that borneol disrupted the normal structure and function of the C. albicans cell wall and membrane, resulting in dysregulated mRNA expression of related genes and ultimately increased cell permeability. As a result, the excessive accumulation of curcumin in C. albicans triggered the overproduction of intracellular ROS upon exposure to blue light. These excessive intracellular ROS disrupted various cellular structures, interfered with essential cellular processes, inhibited biofilm formation and reduced virulence. Remarkably, borneol was also found to enhance curcumin uptake by C. albicans within biofilms, further enhancing the anti-biofilm efficacy of curcumin-mediated aPDT (Cur-aPDT). In conclusion, the results of this study strongly support the potential of borneol as an adjuvant agent to Cur-aPDT in treating superficial cutaneous fungal infections.

Development of sodium alginate-based active packaging films reinforced with curcumin/β-cyclodextrin stabilized high internal phase Pickering emulsions for blueberry preservation

Si Shi,Junjie Tang,Yanyan Zhou,Hongyan Huang,Jianxi Zhang,Wenhao Liu,Zhiwen Hu,Hongwei Wang,Jie Pang,Chunhua Wu

Maintaining the safety and quality of minimally processed agricultural products is a significant challenge in modern food supply chains. This study developed multifunctional sodium alginate (SA)-based films incorporating high internal phase Pickering emulsions (HIPPEs) stabilized by curcumin/β-cyclodextrin inclusion complexes (CUR/β-CD). The HIPPEs demonstrated excellent colloidal stability (>21 days) with a mean droplet diameter of 12.76 ± 0.32 μm. Through solvent casting, HIPPEs were uniformly integrated into SA matrices (0.5–2 % w/w). Based on a comprehensive characterization of the films, the optimum concentration of 1.5 % for doping with HIPPEs was determined. The SA/HIPPEs1.5 films exhibited excellent tensile strength (14.21 MPa), elongation at break (26.28 %), and adequate water vapor (6.5 × 10 −11  g/(m·s·Pa)) and oxygen permeability (1.06 × 10 −3  g/m 2 ·s). Furthermore, they demonstrated dual antimicrobial-antioxidant functionality, with inhibition zones of 19.30 ± 0.70 mm ( Staphylococcus aureus ) and 20.53 ± 0.40 mm ( Escherichia coli ), along with 70.63 % DPPH radical scavenging activity. Compared to polyethylene packaging, SA/HIPPEs1.5 films effectively preserved blueberry quality at 25 °C for 18 days, showing lower weight loss (30.6 % vs. 42.2 %) and a favorable pH (3.43 vs. 4.22). These findings support the potential of SA/HIPPEs composite films as an effective active packaging solution for extending blueberry shelf life.

Construction of a Rice Glutelin-Rhamnolipid-Pectin Nanoparticle Delivery System for Curcumin Delivery: Effect of Pectin Esterification on Characterization, Stability, and Bioaccessibility

Lei Chen,Qiongqiong Wu,Shaokang Li,Shuangli Wang,Weidong Lin,Xiaodan Zang

Riceglutelin (RG)-rhamnolipids (Rha)-high-methoxyl pectin (HMP)/medium-methoxy pectin (MMP)/low-methoxy pectin (LMP) were used to attract self-assembled nanoparticles by electrostatic attraction, and the effects of pectin esterification degree and concentration on the characteristics of nanoparticles and the bioavailability of curcumin were evaluated. The minimum particle size and the highest encapsulation efficiency of the nanoparticles were at mass ratios of RG:HMP = 2:1, RG:MMP = 4:1, and RG:LMP = 2:1. The results of Fourier transform infrared spectroscopy and circular dichroism spectroscopy show that hydrogen bonding, hydrophobic interaction, and electrostatic attraction are the main driving forces for the formation of complexes. The X-ray diffraction results verified that the curcumin in the complex was in an amorphous state. In addition, curcumin embedded with nanoparticles has good photothermal and storage stability. The results of in vitro simulated digestion showed that nanoparticles constructed from LMP had high bioaccessibility. The results of this study are expected to provide new ideas for improving the bioaccessibility of curcumin.

Novel polysaccharide/polysaccharide complex microgels stabilized HIPEs formed by carboxymethyl tamarind seed polysaccharide and chitosan for improved stability and bioaccessibility of curcumin

Fusheng Sun,Chuanxin Pan,Xinru Zhang,Yuanman Liang,Weiyu Shen,Yantao Liu,Duoxia Xu,Nan Yang

In this work, Pickering emulsions, stabilized with carboxymethyl tamarind seed polysaccharide/chitosan (CMTSP/CS) complex microgels, were formulated with oil-phase ratios of 60 % and 80 % and encapsulating curcumin. The stability and in vitro digestive characteristics were systematically investigated. High internal phase emulsions (HIPEs) with 80 % oil phase demonstrated superior storage stability and improved oxidative stability. SAOS and LAOS analysis further revealed that the HIPEs stabilized by CMTSP/CS complex microgels at 1:1 ratio exhibited the highest viscoelastic property and greatest deformation resistance. Increasing the mass ratio of CMTSP to CS from 1:5 to 1:1 formed a more stable interfacial film of emulsion droplets with superior viscoelastic properties, resulting in higher centrifugal resistance and thermal stability. The Pickering emulsion (particularly HIPEs at 1:1) showed reduced curcumin degradation and higher retention rates. Lipid digestion kinetics revealed that the HIPEs at 1:1 achieved maximal free fatty acid release and the highest curcumin bioaccessibility. The system exhibited an increased electrostatic repulsion and highest viscoelastic film properties, which effectively mitigated oil droplet aggregation. It facilitated the release of free fatty acids and promoted the formation of additional mixed micelles, thereby improving the bioaccessibility of curcumin.

Curcumin delivery system based on a chitosan–liposome encapsulated zeolitic imidazolate framework-8: a potential treatment antioxidant and antibacterial treatment after phacoemulsification

Meiting Lin,Kunyuan Tang,Wendi Zheng,Shijie Zheng,Ke Hu

Curcumin is a natural polyphenol extracted from plants that can interact with various molecular targets, including antioxidant, antibacterial, anticancer, and anti-aging activities. Due to its variety of pharmacological activities and large margin pf safety, curcumin has been used in the prevention and treatment of various diseases, such as Alzheimer's, heart, and rheumatic immune diseases. To develop curcumin eye drops that can be used as antioxidant and antibacterial agents after phacoemulsification, we have designed a nano-based drug delivery system to improve curcumin bioavailability and duration of action. We successfully prepared zeolitic imidazolate framework-8 (ZIF-8) coated with chitosan–liposome (Cur@ZIF-8/CS-Lip) for curcumin delivery. It can release curcumin for over 20 h in vitro and exhibits excellent biosafety, antioxidant, and antibacterial activities. Therefore, we hypothesized that Cur@ZIF-8/CS-Lip could reduce the incidence of oxidative stress and infection after cataract surgery.

Design of oleogel-based co-delivery systems for proanthocyanidins and curcumin: Focusing on the modulation of interfacial and oleogel structures by emulsifiers

Zhaohua Huang,Baozhong Guo,Junhui Pan,Deming Gong,Guowen Zhang

This research offers valuable understanding of the structural and functional attributes of water-in-oleogel emulsions (OGEs) containing proanthocyanidins and curcumin, highlighting the relationship between their rheological characteristics and gastrointestinal fate. By altering emulsifier types (PGPR, lecithin, zein, or none), OGEs demonstrate unique mechanical characteristics and digestion profiles. Microscopic observations confirmed emulsion formation, with polarized light microscopy revealing interfacial crystallization in OGE-zein. Interfacial tension analysis indicated synergistic interactions between carnauba wax and emulsifiers. Rheological analysis showed that PGPR and lecithin weakened the crystal network, while zein enhanced its density and stability. In vitro digestion revealed that the emulsions with PGPR and lecithin had lower bioaccessibility of proanthocyanidins and curcumin, whereas OGE-zein showed the highest free fatty acids release and bioaccessibility. Correlation analysis indicated that stronger oleogel matrices improve the bioaccessibility of co-encapsulated nutrients. These results provide insights for designing functional OGEs for low-fat foods and bioactive compound delivery systems.
