A tough, antibacterial and antioxidant hydrogel dressing accelerates wound healing and suppresses hypertrophic scar formation in infected wounds

Xiaoqing Liu,Yiming Sun,Jie Wang,Yongyuan Kang,Zhaolong Wang,Wangbei Cao,Juan Ye,Changyou Gao

Wound management is an important issue that places enormous pressure on the physical and mental health of patients, especially in cases of infection, where the increased inflammatory response could lead to severe hypertrophic scars (HSs). In this study, a hydrogel dressing was developed by combining the high strength and toughness, swelling resistance, antibacterial and antioxidant capabilities. The hydrogel matrix was composed of a double network of polyvinyl alcohol (PVA) and agarose with excellent mechanical properties. Hyperbranched polylysine (HBPL), a highly effective antibacterial cationic polymer, and tannic acid (TA), a strong antioxidant molecule, were added to the hydrogel as functional components. Examination of antibacterial and antioxidant properties of the hydrogel confirmed the full play of the efficacy of HBPL and TA. In the in vivo studies of methicillin-resistant Staphylococcus aureus (MRSA) infection, the hydrogel had shown obvious promotion of wound healing, and more profoundly, significant suppression of scar formation. Due to the common raw materials and simple preparation methods, this hydrogel can be mass produced and used for accelerating wound healing while preventing HSs in infected wounds.

A triple-crosslinked, self-healing, polyvinyl alcohol/nanocellulose hydrogel for versatile sensing applications

Danning Fu,Rendang Yang,Ruibin Wang,Yang Wang,Yan Li,Huiyang Bian

Flexible conductive hydrogels (FCHs) have attracted widespread interest as versatile monoliths that can be intricately integrated with various ingredients boasting multiple functionalities. The chemicophysical properties of FCHs cover a wide range, which significantly vary in their building blocks. However, achieving both favorable mechanical strength and high conductivity simultaneously through a facile approach remains a challenge. Herein, polyvinyl alcohol, dialdehyde cellulose nanofibrils, silver nanoparticles, borax, and tannic acid are readily “one-pot” incorporated into FCHs with great tensile stress (499 kPa), tensile strain (4591 %), and compressive stress (269 kPa) due to abundant hydrogen bonding, dynamic borate-diol bonding, and intermolecular acetalization. They also exhibit desired self-healing, generalized-adhesive, and antibacterial performances. Taking advantage of these, FCHs are further employed to support an epidermal sensor, on which remarkable strain sensitivity (gauge factor = 8.22), high-pressure sensitivity (≥ 0.258 kPa −1 ), and fast response (≤ 190 ms) are recorded. Its highly adaptive mechano-electric transformability and functions can be well maintained in serving as an array unit and touch screen pen. The results well addressed in this work are anticipated to pave the universal way of engineering FCHs.

Qiang Peng,Famin Wen,Xinrui Tang,Shengjie Lu,Huilin Li,Chuanju Wang,Zihao Chen,Zidan Zhou,Shaofan Tan,Huaicheng Qin,Yongguang Bi,Zhong Wang,Fansheng Kong

Wound healing and antibiotic resistance present significant clinical challenges, underscoring the need for innovative bioactive wound dressings. This study prepared green-sustainable bioactive films by incorporating Litsea cubeba essential oil nanoemulsion (LCEO-NE) into a carboxymethyl chitosan-gelatin composite matrix, with essential oil in the films stabilized by whey protein for the first time. Fourier-transform infrared spectroscopy (FTIR) confirmed the interaction between the nanoemulsion and the matrix, resulting in altered absorption peaks. Scanning electron microscopy (SEM), X-ray diffraction (XRD), and thermogravimetric analysis (TGA) revealed that the addition of LCEO-NE increased the surface roughness, crystallinity, and thermal stability of the films. Furthermore, incorporating LCEO-NE significantly enhanced the physical properties and bioactivity of the films, with CGL-3 exhibiting optimal performance: tensile strength of 2.31 MPa, elongation at break of 72.31 %, strong antibacterial and antioxidant effects, excellent water absorption, biocompatibility, prolonged essential oil release (66 h), a water vapor permeability of 40.73 g/m 2 /h, and a contact angle of 82.23°. In vivo testing in a mouse model showed a 99 % wound healing rate by day 15 following treatment with the CGL-3 film, along with enhanced collagen deposition, accelerated epithelialization, and reduced inflammation. Overall, the CGL-3 film holds great promise for advanced wound care applications.

Caseinate-coated zein nanoparticles as potential delivery vehicles for guavinoside B from guava: Molecular interactions and encapsulation properties

Haoning Yang,Yu Mu,Dan Zheng,Tess Puopolo,Lejie Zhang,Zhuo Zhang,Sai Gao,Navindra P. Seeram,Hang Ma,Xueshi Huang,Liya Li

Guavinoside B (GUB) is a characteristic constituent from guava with strong antioxidant activity ; however, its low water solubility limits its utilization. Herein, we investigated the interaction between GUB and zein , a prolamin with self-assembling property, using multiple spectroscopic methods and fabricated GUB-zein-NaCas nanoparticles (GUB- Z -N NPs) via the antisolvent coprecipitation approach. GUB caused fluorescence quenching to zein via the static quenching mechanism. Fourier-transform infrared spectroscopy and computational analysis revealed that GUB bound to zein via van der Waals interaction , hydrogen bond , and hydrophobic forces. The GUB- Z -N NPs were in the nanometric size range (< 200 nm) and exhibited promising encapsulation efficiency and redispersibility after freeze-drying. These particles remained stable for up to 31 days at 4 °C and great resistance to salt and pH variation, and displayed superior antioxidant activity to native GUB. The current study highlights the potential of zein-based nanoparticles as delivery vehicles for GUB in the food industry.

Characterisation of the phytochemical and bioactivity profiles of raw tea, stale-aroma, and betelnut-aroma type of Liupao tea through GC/LC-MS-based metabolomics

Xuancheng Wang,Zhiyong She,Hailin Zhou,Tingting An,Jianwen Teng,Ning Xia,Pingchuan Zhu,Wenhui Liu,Huanxiao Dong,Limin Tang,Shulan You,Lu Wei,Kongying Li,Lingli Wang,Li Huang,Qisong Zhang

Liupao tea (LPT) is a Chinese dark tea known to possess a unique flavour. Microbial fermentation plays a crucial role in flavour development and enrichment. Currently, the phytochemical profiles and bioactivities of LPT with and without fermentation are not fully known. In this study, we compared the chemical composition of raw tea (SF), stale-aroma (SA), and betelnut-aroma (BA) type LPT through the application of GC/LC-MS-based metabolomics, and experimentally investigated their bioactivities via antioxidant, anti-inflammatory, hypolipidemic, and hypoglycemic assays in vitro. The results indicated that fermentation enhanced the flavour of LPT as evidenced by the sweetness-producing substances, decreased bitterness and astringency-related compounds and enriched abundance of aroma-generating compounds. Two and four volatiles were detected to be major contributors to the aroma in SA and BA, respectively. Fatty acids and phosphatidylcholines were the primary lipids, among which the lysing diacylglycerol trimethyl homoserines were found to be a new class of lipids in LPT. Notably, the fermentation resulted in the degradation of compounds, particularly glycerophospholipids and saccharolipids. SF had the highest level of bioactivity, followed by BA and SA. These findings expand the present understanding regarding the development of flavour, nutrition, and medicinal value of LPT. Moreover, they provide a theoretical basis for the identification of BA and SA and serve as a reference value for consumers in their selection of LPT products.

Characterisation of an edible active film prepared from bacterial nanocellulose/forsythia essential oil Pickering emulsions with funoran and its application in fresh meat

Ming Cheng,Ying Shu,Mengli Li,Chaoyu Li,Tieqiang Liang,Zhisheng Zhang

This study sought to develop an edible active film by integrating Pickering emulsions of forsythia essential oil and bacterial nanofibers at various concentrations into a film-forming matrix composed of funoran (F). The stability of the emulsions was evaluated through examination of the micro-morphology, particle size and distribution, 7-day emulsification index, and embedding rate of the Pickering emulsions. Subsequently, selected Pickering emulsions were incorporated into F to generate the edible active film. Scanning electron microscopy, Fourier-transform infrared spectroscopy, and Raman spectroscopy revealed that the Pickering emulsion was uniformly distributed throughout the F film, interconnected by hydrogen bonds. X-ray Diffraction spectra exhibited changes in peak intensity and shifts in position attributable to the edible active film. Pickering emulsion had a minimal impact on thermal stability. The film's tensile strength significantly increased, while elongation at break decreased. The heightened concentration of hydroxyl groups in the film led to increased thickness, reduced moisture content, and enhanced hygroscopicity. The edible active film exhibited superior antioxidant and antibacterial properties, thereby more efficiently shielding against oxygen and water vapour. In preservation tests involving chicken and lamb, the Pickering emulsion led to elevations in pH, total volatile basic nitrogen, and thiobarbituric acid reactive substance levels in the meat.

Mechanism of the wine pomace tannin in hyperpigmentation inhibition: Impact on signaling pathways, cell proliferation, and tyrosinase activity

Xinqing Shi,Jiaman Liu,Wanqin Zhang,Shinian Zeng,Fan Lin,Haining Wang,Bo Teng

After winemaking, tannins with high polymerization remain in the pomace. Utilizing these tannin fractions is a concern for the wine industry. While tannins show potential in treating hyperpigmentation, their mechanisms in vivo and at the cellular level are unclear. Herein, pomace tannin fractions (PTFs) were isolated post-winemaking. Nuclear magnetic resonance spectroscopy and mass spectrometry analysis showed PTFs were composed of ( epi )catechin gallate and (epi)catechin as terminal and extensional units, with polymerization degrees of 10, 16, and 35. In vivo studies demonstrated that PTFs removed ∼76 % of skin melanin, comparable to hydroquinone. The inhibition by PTFs is due to: (1) Inhibition of the Wnt and melanogenesis pathways, downregulating key melanin synthesis proteins (TYR, TYRP1, TYRP2); (2) Inducing cell cycle arrest at the G1/S checkpoint, disrupting DNA, decreasing mitochondrial membrane potential and integrity, and slowing melanocyte proliferation; (3) Superior tyrosinase inhibitory activity by binding to tyrosinase, chelating copper ions, and demonstrating antioxidant properties. These findings suggest that PTFs inhibit melanin synthesis by the combination of the above mentioned ways, supporting the medical use of winemaking tannins.

Maillard reaction products derived from xylose-phosphatidylethanolamine: Potential anti-oxidative substances from hot-pressed flaxseed oil

Yi-kang Zhang,Ke-yao Li,Yue Du,Lin Wang,Qian Xiong,Ning Zhang,Xin Yue,Jia-zi Chen,Martin J.T. Reaney,Yong Wang,Zi-zhe Cai

Hot-pressed flaxseed oil is favored in Northwest China as cooking oil due to its flavor and oxidative stability. The mechanism of enhancing oxidation stability of hot-pressed flaxseed oil by neoformed Maillard reaction products (MRPs) remains mysterious. Maillard model reaction between phosphatidylethanolamine (PE) and reducing sugar of flaxseed, glucose (Glu)/fructose (Fru)/xylose (Xyl), was conducted to investigate the antioxidant characteristics of products. MRPs of PE-Xyl had a superior reactivity and anti-oxidative capabilities, which could improve the oxidative stability of flaxseed oil and inhibited the formation of aldehydes in the accelerated oxidation. Phosphatidylethanolamine-linked pyrrolecarbaldehyde (PLP) was the main antioxidant component in MRPs of Xyl-PE by MS and NMR analysis and verification of antioxidant activity. Furthermore, the results of quantum chemical calculation show that the antioxidant activity center of PLP is the pyrrole group. The findings of this study provide insights into the chemical basis underlying the improved oxidative stability of hot-pressed oils.

Microenvironment-Regulated Dual-Layer Microneedle Patch for Promoting Periodontal Soft and Hard Tissue Regeneration in Diabetic Periodontitis

Yaning Qu,Huajing Zeng,Lei Wang,Zhenlin Ge,Bin Liu,Zengjie Fan

Chronic periodontitis in individuals with diabetes can exacerbate the destruction of local periodontal soft tissues and accelerate the resorption of hard tissue. Currently, effective strategies to simultaneously restore both soft and hard periodontal tissues remain insufficient. To address this challenge, a multifunctional dual-layer microneedles (d-MNs) design is proposed to regenerate both periodontal soft and hard tissues in diabetic patients. The d-MNs substrate is composed of gelatin methacryloyl (GelMA) infused with nano-hydroxyapatite (nHA), which facilitates the differentiation of osteogenic cells into osteoblasts, thereby promoting alveolar bone regeneration. The tips of the d-MNs, on the other hand, are primarily made of hyaluronic acid (HA) combined with a magnesium-based metal-organic framework (Mg-MOF) loaded with glucose oxidase (GOX). This composition creates a hypoglycemic, angiogenic, and anti-inflammatory microenvironment, which supports soft tissue repair. When implanted at the site of periodontitis, the synergistic interaction between the d-MNs substrate and tips effectively promotes the regeneration of both soft and hard tissues, as demonstrated in diabetic rat models of periodontitis. These innovative d-MNs have the potential to revolutionize traditional approaches to treating diabetic periodontitis and can see broad application in dental clinics.

Microenvironment-responsive Bletilla polysaccharide hydrogel with photothermal antibacterial and macrophage polarization-regulating properties for diabetic wound healing

Zhengbo Hu,Kai Zhao,Xin Rao,XingCan Chen,Yujing Niu,Qiantao Zhang,Mingyuan Zhou,Yuchi Chen,FangMei Zhou,Jie Yu,Zhishan Ding,Bingqi Zhu

The healing of diabetic wounds (DW) is challenging due to the complex microenvironment, manifesting severe bacterial infections, persistent inflammation, and excessive oxidative stress. Therefore, overcoming these impediments to restore the wound microenvironment to a normal state is critical points and challenges in the development of effective DW dressings. Herein, a B/TF hydrogel was developed in our study using Bletilla striata polysaccharide (BSP), borax, and the tannic acid/ferric iron (TA/Fe 3+ ) complex. The crosslinking of the hydrogel constructed by dynamic borate bonds not only impart remarkable physical properties to the hydrogel, but also exhibited responsive release of BSP and TA/Fe 3+ complex to pH and glucose levels. Incorporating the TA/Fe 3+ complex provided notable near-infrared (NIR) photothermal properties to the hydrogel, thereby demonstrating significant photothermal antibacterial activity and antioxidant capability. Additionally, the B/TF hydrogel effectively regulated the transformation of macrophages from the pro-inflammatory M1 phenotype to the anti-inflammatory M2 phenotype. Animal experiments have also effectively confirmed that the treatment with B/TF hydrogel combined with NIR radiation promoted the regeneration of skin tissue in DW. In summary, the developed multifunctional B/TF hydrogel showed advantages and met clinical requirements for treating DW.

Macrophage Membrane Spontaneously Encapsulated Cyclodextrin-Based Nanomedicines for Improving Lipid Metabolism and Inflammation in Atherosclerosis

Nianlian Mou,Xinmei Duan,Kai Qu,Qiao Chen,Zhigui He,Yu Cao,Kun Zhang,Xian Qin,Li Zhu,ZhiQiang Han,Chuanwei Li,Yuan Zhong,Wei Wu

Atherosclerosis is a persistent inflammatory condition of the blood vessels associated with abnormalities in lipid metabolism. Development of biomimetic nanoplatforms provides an effective strategy. Herein, inspired by the peptide CLIKKPF spontaneously coupling to phosphatidylserine (PS) on the inner leaflet of cell membranes specifically, MM@NPs were constructed by macrophage membrane spontaneous encapsulation of cyclodextrin-based nanoparticles modified with the peptide CLIKKPF and loaded with the hydrophobic compound resveratrol. MM@NPs could be specifically phagocytized by the activated endothelium with the overexpressed VCAM-1 for enhancing target delivery into the pathological lesion. Additionally, for the ApoE–/– mice, MM@NPs provide comprehensive treatment efficiency in reducing oxidant stress, alleviating the inherent inflammation, and decreasing cholesterol deposition, subsequently resulting in the atherosclerotic plaque regression. Therefore, MM@NPs could be one possible candidate for improving lipid metabolism and inflammation in atherosclerosis.

Maltodextrin vitamin E succinate: A novel antioxidant emulsifier for formulating functional nanoemulsions

Xiang Luo,Yuanyuan Chen,Zhe Jiang,Hongze Wu,David Julian McClements,Chang Zhang,Yanyan Zhou,Hongliang Fu,Xuguang Yin,Wenna Huang,Zhixin Wang,Lemao Yu,Xin Tang,Kangli Li,Kewu Zhu

A new multifunctional emulsifier was synthesized by coupling maltodextrin with a dextrose equivalent of 19 to vitamin E succinate. Two emulsifiers with varying degrees of vitamin E succinate substitution were prepared based on different mass ratios of vitamin E succinate to maltodextrin. The molecular structure and purity of these emulsifiers were analyzed. Nanoemulsions were prepared using octenyl succinic anhydride modified starch as a control to investigate the physical stability, antioxidant capacity, oxidative stability, and in vitro simulated digestive properties of the nanoemulsions. The emulsifying and antioxidant activity of the maltodextrin-vitamin E succinate conjugate was significantly superior to that of octenyl succinic anhydride modified starch, demonstrating good physical and oxidative stability. Additionally, they were rapidly digested under simulated small intestinal conditions. This new emulsifier shows broad application potential for the encapsulation, protection, and delivery of hydrophobic bioactive substances in the fields of medicine, food, and healthcare products.
