Cyclophosphamide (CTX) is an anticancer drug known to induce immunosuppression. This study investigated the immunomodulatory effects of Sargassum thunbergii glucuronomannan and its oligomers (Gs) in CTX-induced immunosuppressive mice. Various Gs, including G2, G4, G6, G8, and Gn, were administered to the mice. Among these, G6, G8, and Gn notably improved the spleen index, cytokine levels (IL-2, IFN-γ, NO), and antioxidant activity (SOD, GSH-Px). G6 treatment increased GSH-Px by 29.3% and promoted macrophage viability. Additionally, G6 enhanced mRNA expression of iNOS, IL-2, and TNF-α. Molecular docking analysis and dynamics analysis showed strong interactions between G6 and immune cytokines IL-2 and TNF-α, enhancing receptor-binding efficiency. These results suggested that Gs, particularly G6, played a key role in immunomodulation and might serve as potential candidates.

Electrochemical direct air CO 2 capture (eDAC) using redox carriers enables sustainable decarbonization but suffers efficiency losses from O 2 -induced side reactions such as oxygen reduction and carrier oxidation. We introduce a composite electrode BPT−GPL by integrating 2,5-bis(4-pyridyl)−1,3,4-thiadiazole (BPT) with ether oxygen (–O–) enriched gas permeation layer (GPL), forming a tunable gas transport channel that facilitates CO 2 diffusion while limiting O 2 penetration. Under atmospheric conditions (~400 ppm CO 2 , 21% O 2 ), the BPT − GPL system demonstrates high eDAC capacity of 3.3 ± 0.2 mmol g –1 BPT across 48 cycles with negligible redox carrier degradation. The thiadiazole ring-based BPT molecules increase electron delocalization, exhibiting moderate CO 2 binding strength and high O 2 tolerance. Moreover, GPL allows CO 2 diffusion and suppresses O 2 permeation creating low-O 2 microenvironment that inhibits side reactions and maintains cycling stability. Molecular dynamics (MD) simulations suggest that ether oxygen’s stronger affinity for CO 2 than O 2 increases CO 2 solubility within GPL, resulting in preferential CO 2 permeation. The BPT − GPL system favors CO 2 delivery and restricts O 2 transport, enabling efficient eDAC and highlighting the potential for practical implementation.

Pseudocapacitive materials employ rapid, non-diffusion-limited faradaic processes to store charge, demonstrating significant potential for fast-charging batteries and supercapacitors. However, the high redox potentials of existing pseudocapacitive anodes substantially lower the overall cell voltage and energy density. Here, a cation-disordered rock-salt lithium titanium oxyfluoride (DRX-Li x TiOF 2 , 0 < x < 2) is reported to reversibly accommodate approximately 1.19 mol of Li + (~ 310 mAh g −1 ) and delivers high-rate performance (exceeding 64.4 C) via pseudocapacitive Li + storage within a low potential window extending down to 0.1 V vs. Li + /Li. This pseudocapacitive behavior is characterized by several structural and electrochemical features: the absence of phase transformation during Li + intercalation, quasi-rectangular cyclic voltammetry curves, sloping charge/discharge profiles and a surface-controlled current response. We further reveal that the pseudocapacitive characteristics originate from a three-dimensional percolation network that facilitates fast Li + migration with low energy barriers, enabled by a cation/anion-disordered structure arising from the mixed occupancy of Li/Ti cations and O/F anions. Owing to its low operating potential and high-rate capability, DRX-Li x TiOF 2 allows a lithium-ion capacitor to attain 4.0 V cell voltage and achieve energy and power densities several times higher than those obtained with conventional anodes, such as battery-type Li 4 Ti 5 O 12 or pseudocapacitive materials like Nb 2 O 5 and TiO 2 .

WO 3 quantum-dot electrochromic devices have exhibited significant commercial potential, owing to their ultra-fast response speed, wide optical transmittance modulation, and low-cost preparation process. However, the application of quantum-dots in electrochromic devices has encountered bottleneck issues. Specifically, quantum-dots tend to agglomerate or be eroded by electrolyte. Herein we propose strategies to manipulate the coating material of quantum-dots at the molecular scale for these issues, achieving single-dot and multi-dot coating of quantum-dots. The single-dot coated quantum-dot device has a transmittance modulation of 54.5% and 69.1% at wavelengths of 580 and 800 nm, respectively, with colouring efficiencies of 101.7 and 163.1 cm 2 /C, and a bleaching/colouring time of 4.0/7.5 s. Meanwhile, the transmittance modulation of the multi-dot coated quantum-dot device is up to 77.4% at 580 nm and 86.0% at 800 nm with colouring efficiencies of 80.7 and 135.3 cm 2 /C and a bleaching/colouring time of 11.0/13.0 s. After 10,000 CV-tests, the multi-dot coated quantum-dot device still operates stably. These indicate advantages of the strategies.

Background The rapid expansion of the aquaculture industry has intensified the discharge of nitrogen-rich wastewater, posing significant ecological risks. Biological treatment with HN-AD bacteria is sustainable, but most conventional strains lack long-term environmental resilience. The key bottleneck is identifying novel, robustly adaptable strains, especially spore-forming Gram-positive bacteria. Moreover, the non-canonical metabolic mechanisms that distinguish such strains remain largely unexplored. This study isolated a novel HN-AD strain, Paenibacillus glycanilyticus DQ-1, from aquaculture sludge and investigated its nitrogen removal performance and genomic metabolic pathways. Methods Strain DQ-1 was isolated and identified through 16 S rRNA gene sequencing and morphological analysis. Its nitrogen removal efficiency was evaluated under aerobic conditions with different nitrogen sources, including ammonium nitrogen (NH₄ + -N), nitrate nitrogen (NO₃ − -N), total inorganic nitrogen (TIN) and hydroxylamine nitrogen (NH 2 OH-N). Whole-genome sequencing was conducted using the PacBio Sequel II platform to identify functional genes and reconstruct the nitrogen metabolic pathways. Results Strain DQ-1 achieved 93.26% NH₄⁺-N and 100% NO₃⁻-N removal within 36 h, with 95.92% TIN removal. Genomic analysis revealed a complete assimilatory nitrate reduction pathway ( nasAB and nirBD ). Notably, while the canonical hydroxylamine oxidoreductase gene ( hao ) is absent, the strain showed 74.67% NH 2 OH-N removal and harbors a putative ammonia monooxygenase gene ( amo ). These results suggest that DQ-1 employs a novel non-canonical pathway, distinct from known HN-AD models, for efficient nitrogen transformation. Conclusions P. glycanilyticus DQ-1 differs from typical HN-AD bacteria in its spore-forming ability and a novel hao -independent pathway. This study provides the first comprehensive genomic insight into the nitrogen removal capacity of P. glycanilyticus , expanding the microbial resource pool for sustainable environmental remediation.

With the acceleration of industrialization, mercury pollution in water bodies has become a severe threat to both the ecological environment and human health. The development of efficient and cost-effective technologies for mercury ion removal has become crucial. This study designed and synthesized a novel triazine-based porous organic polymer, TTIA, using cyanuric chloride and 2-aminoimidazole as monomers via a nucleophilic substitution reaction, with the aim of creating a high-performance Hg(II) adsorption material. Systematic characterization of the material’s structure and morphology confirmed that TTIA features a stable cross-linked network, a surface enriched with nitrogen-based active sites, and a mesoporous-dominated porosity (BET surface area: 34.82 ± 0.15 m 2 /g), which provides abundant binding sites and efficient mass transfer pathways for Hg(II) adsorption. Adsorption performance testing demonstrated that TTIA exhibits exceptional adsorption efficiency toward Hg(II) in aqueous solutions. Langmuir model fitting revealed a maximum adsorption capacity of 1545.2 mg/g (R 2 = 0.989), significantly surpassing most reported adsorbents. Mechanistic analysis indicates that TTIA forms coordination bonds with Hg(II) through the lone pair electrons of nitrogen atoms in both the triazine ring and imidazole unit. The nitrogen-enriched region serves as the key active center. Graphical abstract

Dextranase (DEX) is a high-value biocatalyst with substantial potential in oral hygiene and diverse industrial bioprocesses, yet its practical application is frequently constrained by the insufficient yields of wild-type strains. In this study, a high-efficiency DEX production system was established through an integrated strategy comprising Atmospheric and Room Temperature Plasma (ARTP) mutagenesis, molecular mechanism elucidation, and host-specific promoter engineering. A stable mutant, Chaetomium gracile B12, was isolated, exhibiting a 30.6-fold increase in DEX activity (16.23 U/mL) compared to the parental strain. Sequence analysis and multi-scale computational simulations, including AlphaFold2-based modeling and 100 ns molecular dynamics, generated a testable hypothesis that this mutation may contribute this enhancement. This substitution was found to augment structural rigidity and refine the spatial orientation of the substrate within the active pocket, facilitating a more "productive alignment" for catalysis. Furthermore, heterologous expression optimization uncovered a distinct host-specific promoter preference: the heterologous Pcdna1 promoter was most effective in C. gracile, whereas the endogenous inducible PgpdA proved superior in Aspergillus niger. Notably, this pioneering heterologous expression of DEX in A. niger streamlined the production cycle, slashing the fermentation period from 6 days to 48 h. Following systematic optimization of carbon/nitrogen sources and aeration intensity, the engineered A. niger strain H achieved a peak DEX activity of 36.00 U/mL. Biochemical characterization revealed that the recombinant DEX possessed a favorable near-neutral optimal pH (6.0-7.0) and a temperature optimum of 50 °C. Overall, this study reports the first heterologous expression of a dextranase in A. niger, reducing the fermentation period from 6 days to 48 h, and provides a comprehensive strategy for the overproduction of dextranase and offers molecular insights into enzymatic "fine-tuning", yielding a high-titer, neutral-pH compatible biocatalyst with significant promise for oral care and industrial applications.

Zn-based aqueous batteries have attracted widespread research attention, while the lack of nucleation theory for electrochemical interactions at the Zn-water interface constrains efforts to suppress the thermodynamically spontaneous hydrogen evolution reaction and dendrite formation, thereby stalling practical development. Elucidating Zn electrodeposition in aqueous media requires Zn-specific nucleation theory and a descriptor to regulate interfacial electrochemistry. Conventional Li-based spherical nucleation models disregard Zn’s crystallography and the interfacial resistance that governs nucleation, thereby focusing on polarization variations. In this work, we reformulate the classical spherical nucleation theory derived by Li for the hexagonal close-packed structure of Zn and establish a dimensionless descriptor ( W f ) to quantitatively rationalize interfacial electrochemistry. W f synthesizes the polarization driving force and interfacial resistance into a stability metric. Higher W f values facilitate uniform Zn deposition, as evidenced by the literature. Accordingly, we develop a high- W f electrolyte to inhibit dendrites and side reactions, achieving over 700 h at 100% depth of discharge and 7660 cycles at 10 A g −1 in a Zn||NaV 3 O 8 cell. This work provides a fundamental nucleation theory and a generally applicable quantitative metric for the rational design of Zn-based aqueous batteries.

Osteoarthritis (OA) is a common degenerative joint disease with limited disease-modifying therapies. Emerging evidence suggests that epigenetic dysregulation contributes to cartilage degeneration, but effective strategies to selectively target these pathways remain lacking. Here we show that the BRD4/Nav1.7 axis drives inflammatory and metabolic dysfunction in OA. Integrated single-cell and transcriptomic analyses identify BRD4 as a key regulator that enhances Nav1.7 transcription, promoting mitochondrial impairment and catabolic activation in chondrocytes. To therapeutically target this pathway, we develop a biomimetic hydrogel system incorporating chondrocyte membrane-coated nanoparticles for cartilage-specific delivery of a BRD4 proteolysis-targeting chimera (PROTAC), a molecule designed to induce selective protein degradation. This nanoplatform enables efficient intra-articular delivery, immune evasion and targeted retention in cartilage. Treatment suppresses inflammatory responses, restores mitochondrial function and reduces cartilage degeneration and pain behaviors in two mouse models of OA. These findings establish targeted BRD4 degradation as a disease-modifying strategy and provide a precision nanotherapeutic platform for OA.

Lactate significantly accumulates in intervertebral disc degeneration (IVDD) to promote inflammation storm and nucleus pulposus cells (NPCs) senescence. However, eliminating the lactate efficiently and inhibiting the inflammation storm and NPCs senescence stimulated by lactate remains a challenge. Here, we show a lactate metabolism reprogramming reactor, which converts lactate to the inhibitor of NPCs senescence (alanine) through orthogonal tandem catalysis (OTC) reaction, thereby guiding the lactate metabolism reprogramming. Enzymes and substrates are encapsulated to prepare OTC nanoparticles, which are embedded in hydrogel microspheres to form the reactors. The lactate metabolism reprogramming efficiently performs in vitro and sustainably conducts in vivo to down-regulate lactate to reduce NLRP3 activity and up-regulate alanine to decrease oxidative stress, significantly inhibiting the inflammatory storm and NPCs senescence. The biomechanical function of neo-generated tissues reaches 94% of that of normal tissues, showing clinical potentials in reversing the IVDD. Lactate accumulates in intervertebral disc degeneration to promote inflammation storm and nucleus pulposus cells (NPCs) senescence. Here, the authors address these issues by developing a lactate metabolism reprogramming reactor, which converts lactate to the inhibitor of NPCs senescence (alanine) through orthogonal tandem catalysis reaction, guiding the lactate metabolism reprogramming.

The adsorption of charged nanoparticles at water-oil interfaces constitutes a fundamental phenomenon, underlying pivotal technologies spanning from emulsion stabilization to the sophisticated fabrication of foams. However, the diffusional behavior of these nanoparticles remains poorly understood. Here, we use single-molecule tracking experiments to show that the diffusion of like-charged nanoparticles at the water-oil interface not only becomes anomalous but also displays a diffusional aging phenomenon at the interfacial coverage that is not associated with the glassy state. We further develop a theoretical framework that quantitatively reproduces all experimental observations. Molecular dynamics simulations reveal the necessity of the coexistence of attraction and repulsion for the emergence of aging dynamics. The interplay between attraction and repulsion leads to nanoparticle adhesion taking place over observable timescales, which is manifested as aging dynamics. The discovery of diffusional aging demonstrates that interfacial evolution persists beyond adsorption equilibrium, suggesting that this effect must be accounted for in applications involving water-oil interfaces laden with charged nanoparticles.

Gouty arthritis faces multiple challenges, such as recalcitrance to cure and recurrent flares, owing to its intricate pathophysiological microenvironment. Recent studies have focused on urate reduction, pain relief, or anti-inflammation effects alone, which often fail to provide satisfactory results. To address these challenges, an implantable manganese platinum microscale galvanic cell (Mn@Pt micro-GC) is developed. This system utilizes the urate oxidase-like activity of platinum to degrade deposited monosodium urate crystals, while the Mn 2+ and hydrogen gas work synergistically to effectively reversing the pro-inflammatory environment. In addition to decreasing the production of pain signals associated with gout, Mn@Pt micro-GC effectively inhibits pain signal transmission along the CXCL5-neuronal CXCR2-TRPA1 axis through continuous electrical stimulation. This multimodal treatment approach emphasizes the strong relationship between bone and nerve in gout, taking into account the importance of reducing uric acid, anti-inflammation, and pain relief in gout therapy, which has led to improved treatment results.
