Antibacterial, antioxidant and injectable hydrogels constructed using CuS and curcumin co-loaded micelles for NIR-enhanced infected wound healing

Pengpeng Jia,Yu Zou,Jiang Jiang

Constructing antibacterial and antioxidant hydrogels is critical for treating infected full-thickness skin wounds. Herein, we report a co-encapsulation strategy to load CuS nanoparticles and hydrophobic antioxidant curcumin (cur) in aldehyde-terminated F127 micelles, which are then cross-linked with carboxymethyl chitosan through a Schiff base reaction to form a functional composite hydrogel (CF–CuS–cur). Apart from its suitable swelling and degradation behavior, good biocompatibility, and injectability for treating irregular wounds, the CF–CuS–cur hydrogel displayed excellent photothermal antibacterial ability under 1064 nm NIR laser irradiation, and antioxidant activity to protect cells from excessive oxidative stress. Using a full-thickness infected wound model, we demonstrated that the CF–CuS–cur hydrogel accelerated the wound healing process by effective sterilization and decreased inflammation, under synergistic action from CuS, curcumin and NIR irradiation. Histological and immunohistochemistry analysis further revealed the promoted skin attachments and regeneration, collagen deposition, neovascularization, and early transition to anti-inflammatory M2 macrophages, when the wounds were treated with the CF–CuS–cur hydrogel. This work demonstrates a facile strategy to construct functional hydrogels with NIR-enhanced antibacterial and antioxidant properties, which can be potentially applied as wound dressings for treating chronic wounds.

Functionalized MoS2-nanosheets with NIR-Triggered nitric oxide delivery and photothermal activities for synergistic antibacterial and regeneration-promoting therapy

Mu Zhixiang,Jin Ting,Chu Tengda,Lu Hongyang,Chen Yuanqi,Li Sisi,Zeng BaiRui,Huang Chen,Lei Kezheng,Cai Xiaojun,Deng Hui,Hu Rongdang

Bacterial infection in skin and soft tissue has emerged as a critical concern. Overreliance on antibiotic therapy has led to numerous challenges, including the emergence of multidrug-resistant bacteria and adverse drug reactions. It is imperative to develop non-antibiotic treatment strategies that not only exhibit potent antibacterial properties but also promote rapid wound healing and demonstrate biocompatibility. Herein, a novel multimodal synergistic antibacterial system (SNO-CS@MoS 2 ) was developed. This system employs easily surface-modified thin-layer MoS 2 as photothermal agents and loaded with S-nitrosothiol-modified chitosan (SNO-CS) via electrostatic interactions, thus realizing the combination of NO gas therapy and photothermal therapy (PTT). Furthermore, this surface modification renders SNO-CS@MoS 2 highly stable and capable of binding with bacteria. Through PTT’s thermal energy, SNO-CS@MoS 2 rapidly generates massive NO, collaborating with PTT to achieve antibacterial effects. This synergistic therapy can swiftly disrupt the bacterial membrane, causing protein leakage and ATP synthesis function damage, ultimately eliminating bacteria. Notably, after effectively eliminating all bacteria, the residual SNO-CS@MoS 2 can create trace NO to promote fibroblast migration, proliferation, and vascular regeneration, thereby accelerating wound healing. This study concluded that SNO-CS@MoS 2 , a novel multifunctional nanomaterial with outstanding antibacterial characteristics and potential to promote wound healing, has promising applications in infected soft tissue wound treatment.

Biosynthesis of Quercetin-Loaded Melanin Nanoparticles for Improved Antioxidant Activity, Photothermal Antimicrobial, and NIR/pH Dual-Responsive Drug Release

Di Zhang,Xianrui Chen,Nitong Bu,Liying Huang,Huanglong Lin,Lizhen Zhou,Ruojun Mu,Lin Wang,Jie Pang

Quercetin (QCT) is a promising dose-dependent nutraceutical that usually suffers from poor water solubility and low bioavailability issues. In this work, a novel QCT-loaded nanoscale delivery system was constructed based on the oxidative self-polymerization of melanin (Q@MNPs). The FT-IR, XRD, and Zeta potential analyses confirmed that QCT was successfully absorbed on the melanin nanoparticles (MNPs) via Π−Π and hydrogen bonding interactions. The encapsulation efficiency and particle size of Q@MNPs were 43.78% and 26.68 nm, respectively. Q@MNPs improved the thermal stability of QCT and the antioxidant properties in comparison to MNPs. Meanwhile, Q@MNPs presented fantastic photothermal conversion capacity and stability triggered by the NIR laser, which significantly enhanced the antibacterial capability with a sterilization rate of more than 98% againstE. coliandS. aureus. More importantly, Q@MNPs exhibited NIR/pH dual-responsive drug release behavior and good biocompatibility (at concentrations of < 100 μg/mL). Thus, Q@MNPs show promising prospects for flavonoid delivery. Keywords:melanin nanoparticles;quercetin;antioxidant activity;photothermal antimicrobial;stimuli drug release

Biotin-decorated hollow gold nanoshell for dual-modal imaging-guided NIR-II photothermal and radiosensitizing therapy toward breast cancer

Yong-Jian Chen,Wei Men,Ming Chen,Lian-Ying Zhang,Ming-Wa Chen,Xiao-Tong Chen,Jian Peng,Nai-Han Huang,Wen-Hua Zhang,Jin-Xiang Chen

Radiotherapy (RT) is dominantly used in breast cancer therapy but is facing fierce side effects because of the limited difference between tumor and normal tissues in response to ionizing radiation. Herein, we construct a core-shell nanoparticle of UiO-66-NH2@AuNS. Then the solid gold shell was etched into hollow AuNS (HAuNS) and further modified with biotin-PEG-SH (PEG-bio) to obtain HAuNS@PEG-bio. HAuNS@PEG-bio demonstrates effective near infrared II (NIR-II) region photothermal therapy (PTT) performance, and the increase of temperature at the tumor site promotes the blood circulation to alleviate the hypoxia in the tumor microenvironment (TME). Meanwhile, HAuNS exhibits strong X-ray absorption and deposition ability due to the high atomic coefficient of elemental Au (Z = 79) and hollowed-out structure. Through the dual radiosensitization of the high atomic coefficient of Au and the hypoxia alleviation from PTT of HAuNS, the breast cancer cells could undergo immunogenic cell death (ICD) to activate the immune response. The outstanding anticancer effects of HAuNS@PEG-bio by cell targeting to exert IR thermal and CT imaging-guided NIR-II photothermal, radiosensitizing therapy, and ICD in vivo. This work highlights that the constructed biotin-decorated hollow gold nanoshell has a promising potential as a diagnostic and treatment integration reagents for the breast cancer.

Selenium-integrated conjugated oligomer nanoparticles with high photothermal conversion efficiency for NIR-II imaging-guided cancer phototheranostics in vivo

Yang Lele,Gao Yijian,Wei Jinchao,Cheng Zehua,Wu Sijia,Zou Liang,Li Shengliang,Li Peng

Second near-infrared (NIR-II) fluorescence imaging in the range of 1000–1700 nm has great prospects for in vivo imaging and theranostics monitoring. At present, few NIR-II probes with theranostics properties have been developed, especially the high-performance organic theranostics material remains underexploited. Herein, we demonstrate a selenium (Se)-tailoring method to develop high-efficient NIR-II imaging-guided material for in vivo cancer phototheranostics. Via Se-tailoring strategy, conjugated oligomer TPSe-based nanoparticles (TPSe NPs) achieve bright NIR-II emission up to 1400 nm and exhibit a relatively high photothermal conversion efficiency of 60% with good stability. Moreover, the TPSe NPs demonstrate their photothermal ablation of cancer cells in vitro and tumor in vivo with the guidance of NIR-II imaging. It is worth noting that the TPSe NPs have good biocompatibility without obvious side effects. Thus, this work provides new insight into the development of NIR-II theranostics agents.

Injectable hydrogel-mediated combination of hyperthermia ablation and photo-enhanced chemotherapy in the NIR-II window for tumor eradication

Gang Wang,Na Zhang,Ziyang Cao,Zhenghai Zhang,Zhongming Zhu,Gengyun Sun,Liangjie Jin,Xianzhu Yang

Local administration of therapeutic agents with long-term retention capabilities efficiently avoids nonspecific distribution in normal organs with an increased drug concentration in pathological tissue. Herein, we developed an injectable and degradable alginate-calcium (Ca2+) hydrogel for the local administration of corn-like Au/Ag nanorods (NRs) and doxorubicin hydrochloride (DOX·HCl). The immobilized Au/Ag NRs with strong absorbance in the near-infrared II (NIR-II) window efficiently ablated the majority of tumor cells after 1064 nm laser irradiation and triggered the release of DOX to kill residual tumor cells. As a result, injectable hydrogel-mediated NIR-II photothermal therapy (PTT) and chemotherapy efficiently inhibited tumor growth, resulting in the complete eradication of tumors in most of the treated mice. Furthermore, owing to the confinement of the Au/Ag NRs and DOX·HCl within the hydrogel, such treatment exhibited excellent biocompatibility.

A Biofilm Microenvironment-Activated Single-Atom Iron Nanozyme with NIR-Controllable Nanocatalytic Activities for Synergetic Bacteria-Infected Wound Therapy

Qiqi Xu,Yusheng Hua,Yuetong Zhang,Mingzhu Lv,Huan Wang,Yang Pi,Jiani Xie,Chengyan Wang,Yuan Yong

Biofilm microenvironment (BME)-activated antimicrobial agents display great potential for improved biofilm-related infection therapy because of their superior specificities and sensitivities, effective eliminations, and minimal side effects. Herein, BME-activated Fe-doped polydiaminopyridine nanofusiform-mediated single-atom nanozyme (FePN SAzyme) is presented for photothermal/chemodynamic synergetic bacteria-infected wound therapy. The photothermal therapy (PTT) function of SAzyme can be specifically initiated by the high level of H 2 O 2 and further accelerated through mild acid within the inflammatory environment through “two-step rocket launching-like” process. Additionally, the enhanced chemodynamic therapy (CDT) for the FePN SAzyme can also be endowed by producing hydroxyl radicals through reacting with H 2 O 2 and consuming glutathione (GSH) of the BME, thereby contributing to more efficient synergistic therapeutic effect. Meanwhile, FePN SAzyme could catalyze biofilm-overexpressed H 2 O 2 decomposing into O 2 and overcome the hypoxia of biofilm, which significantly enhances the susceptibility of biofilm and increases the synergistic efficacy. Most importantly, the synergistic therapy of bacterial-induced infection diseases can be switched on by the internal and external stimuli simultaneously, resulting in minimal nonspecific damage to healthy tissue. These remarkable characteristics of FePN SAzyme not only develop an innovative strategy for the BME-activated combination therapy but also open a new avenue to explore other nanozyme-involved nanoplatforms for bacterial biofilm infections.

Noninvasive In Vivo Imaging and Monitoring of 3D-Printed Polycaprolactone Scaffolds Labeled with an NIR Region II Fluorescent Dye

Linzhi Jing,Mingtai Sun,Pingkang Xu,Kai Yao,Jiao Yang,Xiang Wang,Hang Liu,Minxuan Sun,Yao Sun,Runyan Ni,Jie Sun,Dejian Huang

Significant progress has been made in fabricating porous scaffolds with ultrafine fibers for tissue regeneration. However, the lack of noninvasive tracking methods in vivo makes it impossible to track the fate of such scaffolds in situ. The development of near-infrared region II (NIR-II, 1000–1700 nm) dyes provides the possibility of performing noninvasive visualization with deep-tissue penetration and high spatial resolution in vivo. Herein, we developed a polycaprolactone (PCL) ink containing the small organic NIR-II dye SY-1030 and the fluorescently labeled macromolecular dye SY-COO-PCL and fabricated high-resolution NIR-II active scaffolds via electrohydrodynamic jet (EHDJ) printing. All printed scaffolds subcutaneously implanted in mice were clearly imaged one week after the operation. Compared with scaffolds containing SY-1030, the fluorescence intensity emitted from scaffolds containing SY-COO-PCL can be tracked for up to three weeks. Moreover, the image quality can be optimized by adjusting the dye concentration, laser power, and exposure time. The advantage of such NIR-II active scaffolds is evidenced by the lower dye concentration, longer tracking period, and better in vivo stability. We also demonstrated the biocompatibility and biodegradability of the scaffolds containing SY-COO-PCL over a 3-month period. The developed NIR-II active scaffolds have potential applications in biopolymer implant tracking, tissue reconstruction monitoring, and target-position-based drug delivery.

Surface-Defect-Induced and Synergetic-Effect-Enhanced NIR-II Electrochemiluminescence of Au–Ag Bimetallic Nanoclusters and Its Spectral Sensing

Li Fu,Xuwen Gao,Shuangtian Dong,Hsien-Yi Hsu,Guizheng Zou

Aqueous electrochemiluminescence (ECL) in the second near-infrared biowindow (NIR-II, 900–1700 nm) was anticipated for ECL evolution and spectral multiplexing. Herein, aqueous and monochromatic ECL with a single emission peak beyond 900 nm was achieved by employing methionine (Met)-capped Au–Ag bimetallic nanoclusters (BNCs) as luminophores and triethanolamine (TEOA) as a coreactant. The Met-capped Au–Ag BNCs with surface-defect-induced PL around 756 nm were water-soluble and synthesized via doping Met-capped Au NCs with Ag in a doping-in-growth way. By extensively exploiting the red-shifting nature of surface-defect-induced ECL to PL and the synergetic-effect-enhanced ECL of BNCs, physically surface-confined Au–Ag BNCs exhibited efficient NIR-II ECL around 906 nm in aqueous medium. A spectrum-based NIR-II ECL immunoassay around 915 nm was also achieved by immobilizing the Au–Ag BNCs onto an electrode surface via forming a sandwich immunocomplex, which could selectively determine CA125 from 5 × 10–4 to 1 U/mL with a detection limit of 5 × 10–5 U/mL (S/N = 3). The combined strategy of surface-defect-induced ECL and synergetic-effect-enhanced ECL would enable promising biorelated application of NIR-II ECL.

Noble Metal-Free FeOOH/Li0.1WO3 Core–Shell Nanorods for Selective Oxidation of Methane to Methanol with Visible–NIR Light

Yi Zeng,Xin Luo,Feng Li,Anhua Huang,Hongmei Wu,Guo Qin Xu,Song Ling Wang

Hydroxyl radicals (•OH) generated in the photocatalytic process are crucial to the conversion of methane (CH4) to value-added methanol (CH3OH) at room temperature. However, utilizing noble metal-free catalysts and low-energy photons of solar light, such as visible and near-infrared light (vis–NIR), is difficult to provide more electron states to form •OH radicals. Here, we developed FeOOH/Li0.1WO3 core–shell nanorods via a two-step in/out co-modification of hexagonal tungsten oxide (h-WO3): (1) lithium ions intercalating into the hexagonal tunnels of h-WO3 to form Li0.1WO3 nanorods and (2) using FeOOH-wrapped Li0.1WO3 to obtain FeOOH/Li0.1WO3 core–shell nanorods. Introduction of lithium induces polaron transition in Li0.1WO3, enabling the absorption of vis-NIR light. Interestingly, FeOOH-based Fenton-like reaction when H2O2 is selected as an oxidant favors the generation of more •OH radicals available for CH4 oxidation to CH3OH. Meanwhile, FeOOH with FeIII as an “electron sink” highly improves the separation of photoinduced electrons and holes in Li0.1WO3. Eventually, efficient selective formation of CH4OH is achieved with remarkable generation rates up to ∼342 and ∼160 μmol g–1 at visible light (420–700 nm) and NIR light (≥800 nm), respectively. Our finding opens up new possibilities for developing noble metal-free catalysts for solar energy-driven CH4 conversion to CH3OH under ambient conditions.

A Full Solar Light Spectrum Responsive B@ZrO2–OV Photocatalyst: A Synergistic Strategy for Visible-to-NIR Photon Harvesting

Yumei Qin,Zhaoyang Ding,Wenwei Guo,Xiaolu Guo,Cheng Hou,Bang-Ping Jiang,Chun-Guang Liu,Xing-Can Shen

The utilization of diffusive solar energy through photocatalytic processes has received tremendous attention. Herein, a full-solar-spectrum photocatalyst with strong near-infrared (NIR) photoactivity, which is based on the synergy of oxygen vacancies (OVs) and doped boron that is cointegrated into zirconia (B@ZrO2–OV), is designed in this work. The doped photocatalyst is prepared via a controlled one-step NaBH4 reduction from white ZrO2. Density functional theory (DFT) calculations reveal that simultaneously introduced OVs and boron dopants on ZrO2 induced a narrow band gap (4.78 eV) and achieved charge separation (work function of 4.12 eV), thus confirming its excellent photocatalytic activity in the degradation of rhodamine B under NIR irradiation. In contrast, oxygen-deficient ZrO2 and white ZrO2 were inactive under NIR light. Therefore, this synergistic strategy provides an efficient way to use the narrow band gap of a classic semiconductor zirconia as a highly efficient catalyst for full-solar-spectrum-driven photocatalysis.

Noble metal-free MoS2-modified In2S3 for excellent visible-NIR-light-driven photocatalytic of Cr6+ removal in alkaline wastewater

Wang Linjuan,Zan Ling

MoS 2 -modified In 2 S 3 ( x wt.%MoS 2 -In 2 S 3 ) samples (0 ≤  x  ≤ 20) were fabricated through a facile one-pot hydrothermal process. All In 2 S 3 samples modified by MoS 2 deliver an excellent Cr 6+ removal ability under visible-NIR-light-driven, and 5% MoS 2 -In 2 S 3 can reduce 20 mg/L Cr 6+ solution completely within 30 min with a trace photocatalyst concentration (0.27 mg/mL) which is 3.2-hold higher than that of single In 2 S 3 . Further investigations demonstrate that the loaded MoS 2 can not only enhance the absorption of light scope but also promote the interfacial electron transfer from In 2 S 3 to MoS 2 and then to Cr 6+ , thus greatly improving the Cr 6+ removal ability. ESR shows that the modifying of MoS 2 can accelerate the transformation of electrons from the In 2 S 3 to MoS 2 which results in that the intensity of O 2 − • significantly increased. PL, photocurrent, and EIS further identified that the modification of MoS 2 can greatly improve the charge carrier separation efficiency. The fabricated MoS 2 -In 2 S 3 samples also exhibit characteristic features such as stable and reducing Cr 6+ in the weak alkaline environment for the first time, which could always be a great challenge for wastewater purification in alkaline condition.
