Currently, coronary heart disease (CHD) is prevalent worldwide, with its harmful impact second only to that of malignant tumors. A growing number of studies have confirmed that Choerospondias axillaris can significantly improve the therapeutic effect of CHD treatment and reduce the incidence of adverse reactions. However, its bioactive components and pharmacological mechanisms remain unclear. Based on a targeted fishing strategy, this study synthesized magnetic nanomaterials immobilized with 5-lipoxygenase (5-LOX) for screening and identifying 5-LOX inhibitors from the crude extract of Choerospondias axillaris . A total of 21 active components were identified and analyzed using ultra-high performance liquid chromatography-Q-Exactive Orbitrap tandem mass spectrometry (UHPLC-Q-Exactive Orbitrap-MS/MS). By comparing the screened active components with reported 5-LOX inhibitors, 9 novel 5-LOX inhibitors were discovered. Further molecular docking was performed on these 9 inhibitors, and the pharmacological activity of the selected ligands was verified using a 5-LOX inhibitor screening kit (fluorescence method). The compounds are ranked by activity from strongest to weakest: Lobetyolin (23.92 μM), Anhydroicaritin (29.55 μM), Narirutin(33.66 μM), Fisetin (47.28 μM) and Naringin(59.02 μM). This novel enzyme-immobilized targeted fishing technology is expected to efficiently and accurately screen bioactive components targeting different receptors from complex substrates.

Electrocatalytic 5-hydroxymethylfurfural oxidation reaction (HMFOR) to produce 2,5-furanedicarboxylic acid (FDCA) offers a promising strategy to produce high value chemicals. Herein, CuO(S-ER-1.5) with excellent catalytic performance was prepared by anodic oxidation, sulfidation and electrochemical reconstruction on the substrate of copper foam. The experimental results of catalytic performance showed that CuO(S-ER-1.5) can catalyze HMFOR with a high current density (200 mA cm −2 ) at a low potential (1.48 V vs RHE) and exhibit outstanding stability. The open-circuit potential time test displayed that more HMF molecules were adsorbed in the Helmholtz layer of CuO(S-ER-1.5), which proved that the CuO(S-ER-1.5) could improve the kinetic adsorption of HMF. The electrochemical impedance spectroscopy further showed that CuO(S-ER-1.5) had faster reaction kinetics than Cu(OH) 2 precursor. The product analysis showed that CuO(S-ER-1.5) electrocatalytic HMFOR followed the DFF path, and the catalytic mechanism was explored by the three-step rate deconvolution procedure. It was found that CuO(S-ER-1.5) electrocatalytic HMFOR was dominated by direct oxidation reaction. This work provides a valuable guidance for the synthesis of value-added chemicals FDCA by electrocatalytic HMFOR.

Retinal dehydrogenase 5 (RDH5) is an important enzyme in the visual cycle. Several studies have reported that the RDH family may play crucial roles in tumor prognosis. However, the role of RDH5 in tumor prognosis is still unclear. We examined the mRNA level of RDH5 by using q-PCR in hepatocellular carcinoma (HCC) and adjacent non-cancerous tissues. The proliferation rate of HCC cells was detected by MTS assay, and the invasive ability was examined by transwell and scratch wound assays. The YAP protein localization and expression were visualized by immunofluorescence in two different cell lines. CpG islands in the promoter region were predicted by using the methprimer database. Clinical characteristics of a patient cohort data came from The Cancer Genome Atlas database. RDH5 was significantly downregulated in hepatocellular carcinoma tissues, and low RDH5 expression was associated with metastasis and poor patient prognosis. Functional assays revealed that the RDH5 promoter is methylated in HCC cell lines. Moreover, overexpressing RDH5 can suppress metastasis by reversing the epithelial-mesenchymal transition (EMT) process, and RDH5 also inhibits cell proliferation in HCC cell lines. Furthermore, suppressing RDH5 can activate the Hippo/YAP signaling pathway and promote the nuclear translocation of YAP. Clinical data demonstrated that RDH5 is an independent prognostic factor in HCC. In our study, we provided the first evidence that RDH5 plays a crucial role in suppressing proliferation and metastasis, and the RDH5 promoter is methylated in hepatocellular carcinoma. And as an important regulator, RDH5 can suppress the Hippo/YAP signaling pathway. Taken together, it revealed that RDH5 might be a potential therapeutic target in HCC patients.

Comprehensive Summary As a key epigenetic modification, 5-formylcytosine (5fC) controls many important cell functions, such as gene expression and cell differentiation. However, 5fC exists in mammalian tissues and cells with low content, and its structure is highly similar to 5-methylcytosine (5mC), 5-hydroxymethylcytosine (5hmC) and 5-carboxycytosine (5caC), which requires the detection method for detecting 5fC with high sensitivity and specificity. In this experiment, WS 2 /Bi/Bi 2 O 2 CO 3 was used as the photoactive material, 5fC antibody was used as the target molecule recognition reagent, and rolling amplification (RCA) was employed as the signal amplification strategy to construct a novel photoelectrochemical biosensor for detecting 5-formylcytosine deoxynucleotide (5fdCTP). Firstly, WS 2 /Bi/Bi 2 O 2 CO 3 nanocomposite was prepared and characterized, and then modified on the ITO electrode. Then, polyacrylic acid (PAA) was immobilized by electrostatic adsorption, which was employed as the antibody capture reagent based on the covalent reaction of –COOH on PAA with –NH 2 on Ab. Subsequently, 5fdCTP was recognized through immunoreaction, triggering the subsequent rolling circle amplification reaction with the aid of the crosslinker of Zr 4+ . Finally, positive methylene blue (MB) was adsorbed by negative ssDNA, resulting in an increased photoelectric conversion efficiency of WS 2 /Bi/Bi 2 O 2 CO 3 and improved detection sensitivity. The linear range of the PEC biosensor for detecting 5fdCTP was 0.05—100 nmol/L, and the minimum detection concentration was 12.4 pmol/L (S/N = 3). The applicability of this method has been evaluated by studying the influence of Cd 2+ and antibiotic concentration on the 5fC content in the genomic DNA of maize seedlings.

Efficient zeolitic recycling of polyolefin waste into C2–C4 light olefins offers an attractive route toward resource recovery; however, conventional zeolite catalysts suffer from severe mass transport limitations that lead to undesired secondary reactions and coke formation. Here, we report a “structure modulation and intelligent optimization” strategy to develop hierarchical ZSM-5 (H-ZSM-5), integrating tailored mesoporosity and crystal size control via Bayesian optimization (BO). Catalytic performance was evaluated in a two-stage system combining thermal pyrolysis and subsequent catalytic cracking. BO efficiently navigates this multivariate design space, identifying an optimal architecture (∼100 nm crystals, 4.0 nm mesopores) that achieved 92.6 wt % total gas yield and 85.1 wt % light olefin selectivity at 500 °C with a low catalyst-to-feed ratio (0.2). The nanoscale H-ZSM-5 also exhibited durability over 40 consecutive cycles and broad compatibility with postconsumer plastic mixtures. Mechanistic studies revealed that the synergy between nanoscale morphology and hierarchical porosity enhances mass transport and acid site accessibility, demonstrating that complex catalyst architectures can be precisely optimized via artificial intelligence strategies, such as BO.

Amorphous ZSM-5 was facilely synthesized by simply tuning the aqua ammonia amount and used as support of Mo-V-O for aerobic oxidation desulfurization. It was found that both geometric and electronic effects help to enhance the catalytic performance. Geometrically, the amorphous ZSM-5 could better disperse the metals due to its higher specific area. Electronically, the amorphous ZSM-5 could enable V to enrich electrons availing for oxygen activation to superoxide radical thanks to its low work function. The superoxide radical combines with Mo to form peroxide and then oxidize the sulfur-containing substrates. 100% desulfurization can be realized at 80°C for model oil with sulfur content of 500 ppm over the amorphous ZSM-5 supported Mo-V-O catalyst. This study provides inspiration for designing efficient catalysts for aerobic oxidation desulfurization.

An omnipresent pathogenic bacterium, Pseudomonas aeruginosa (P. aeruginosa PAO1), is easy to contaminate environmental water or foods, causing daily food spoilage and infections. The biofilm-forming ability and bacterial resistance of P. aeruginosa PAO1 make it difficult to be eradicated by traditional bacteriostatic agents. In this work, we designed and synthesized a pyridinium-based pillar[5]arene (PP5), while trimethylammonium-based pillar[5]arene (TP5) was used as a control compound. After clear characterization, the antibacterial and antibiofilm activities as well as the microbial resistance of TP5 and PP5 against P. aeruginosa PAO1 were extensively examined. It was revealed that PP5 exhibited good inhibition activity with the minimum inhibitory concentration (MIC) of 0.051 mmol/L, while no significant antibacterial and biofilm formation activity for TP5 against P. aeruginosa PAO1 was observed. More importantly, PP5 had negligible antimicrobial resistance even after 18th passages. A transmission electron microscope (TEM) showed that PP5 could physically disrupt the cell membranes, causing the leakage of internal constituents, which is possibly ascribed to the synergistic penetrability and π–π interactions of strain, thus greatly reduced the development of bacterial resistance. Overall, the presented studies indicated that pyridinium moieties could facilitate the cationic pillar[5]arene to generate surprising antibacterial and antibiofilm formation ability against P. aeruginosa PAO1.

Based on 5-nitroisophthalic acid (5-H 2 nip) and bis(4-pyridyl)amine (dpa), the complexes {[Cd 6 (5-nip) 6 (dpa) 4 ]·EtOH} n ( 1 ) and [Co 5 (5-nip) 2 (dpa) 2 (OH) 2 (SO 4 ) 2 (H 2 O) 2 ] n ( 2 ) have been synthesized and characterized by elemental analysis, single-crystal X-ray diffraction, IR spectroscopy, thermal gravimetry analysis, fluorescence and magnetic susceptibility. The carboxylate groups of the 5-nip 2− adopt three different bridging modes [ bis (chelate-bidentate), bis (bidentate) and bis (monodentate)]. The 5-nip 2− anions and dpa play as linkers, which connect Cd(II) or Co(II) ions to form the three-dimensional structures of 1 and 2 . Moreover, 1 exhibits intense solid-state luminescence emissions centered at 402 nm at room temperature, which mainly originates from the intraligand π – π* transitions of 5-nip 2− . Magnetic susceptibility measurement indicates that 2 shows antiferromagnetic interactions between the Co(II) ions.

The wear and corrosion resistance of water-based polytetrafluoroethylene (PTFE) coating need to be further improved to cope with various harsh environmental conditions. In this paper, a mulberry-like ZSM-5 molecular sieve was prepared and then was modified to enhance the uniformity, wear resistance and hardness of PTFE coating. In addition, a ZSM-5/MoS2/PTFE coating was developed by adding MoS2 in the chemical composition of coatings, which significantly increased the critical cracking thickness of PTFE to 27 μm. The wear amount of ZSM-5/MoS2/PTFE coating is only 5.7 mg, and after 680 h of salt spray, there is no obvious corrosion on the surface. The friction coefficients under applied loads of 5 N, 10 N, and 20 N are 0.007, 0.057, and 0.055, respectively. The experiment results show that the coating possesses excellent lubrication, wear resistance, and corrosion resistance, and its performance and cost are far superior to commercial products.

The composition of microtubules involving several steps, including the polymerization and depolymerization of α-tubulin and β-tubulin heterodimers. Microtubule-targeting agents can increase or inhibit microtubule polymerization, thereby disrupting the dynamic process and stalling cells in G2/M phase. Microtubule-targeting agents are generally cytotoxic, which neurological toxicity being one of the significant adverse events associated. We recently reported a novel 5-arylalkynyl-2-benzoyl thiophene (PST-3) that exhibited broad-spectrum cellular cytotoxicity and in vivo potency with high safety. PST-3 was a substrate of p-gp, which could not cross the blood-brain barrier and lead to less neurotoxicity. The antitumor activities in vitro demonstrated that PST-3 combined with the colchicine-binding site on microtubule, induces morphological changes, disrupts microtubule networks, inhibits polymerization of tubulin, arrests breast cancer cells in the G2/M phase of the cell cycle and induces apoptosis. Evaluation of the antitumor effect in vivo demonstrated that PST-3 elicited MDA-MB-468 tumor %T/C of 11.75%, whereas elicited MCF7 tumor %T/C of 44.38% in breast cancer xenograft models. Besides, in vivo experiments of a higher dose (60 mg/kg) of PST-3 treatment for 21 days did not produce any significant neurotoxicity. These results provide evidence that PST-3 might possess the potential to be developed into a new microtubule inhibitor without neurological toxicity.

The promotional effects of phosphorous modification on ZSM-5 zeolites by phosphorus oxyacids (H 4 P 2 O 7 , H 3 PO 4 , H 3 PO 3 , and H 3 PO 2 ) with different molecular sizes were investigated with catalytic cracking of n -tetradecane as the probe reaction. It was verified that the diffusion of P precursor molecules into ZSM-5 channels was important to prevent aggregation of P species for better stabilization on tetra-coordinated framework aluminum . Meanwhile, the P–Al interaction was proposed based on results of 27 Al, 31 P, and 29 Si MAS NMR. With H 3 PO 2 as the P precursor, easy diffusion into ZSM-5 channels contributed to less formation of polyphosphates, homogeneous distribution of P species, better reserved crystallinity, microporous area and acidic sites for improving cracking performance of n -tetradecane.

5-Aminolevulinic acid (ALA)-loaded nanoethosome (ALA-ES) gels are successfully prepared to realize a transdermal delivery of ALA, and they provide a feasible approach for the photodynamic therapy (PDT) of hypertrophic scars (HS). Herein, the morphological and physicochemical features indicate that ALA-ES is stable in gel matrix. In vitro transdermal penetration studies suggest ALA-ES gels can overcome the compact dermal barrier and deliver more ALA into human HS tissue. In vivo delivery studies further reveal that ALA-ES gels can penetrate into rabbit HS tissue to facilitate ALA accumulating in hypertrophic scar fibroblast (HSF) and converting into protoporphyrin IX in the cytoplasm. Utilizing transmission electron microscopy, the visual in vivo penetration process indicates ALA-ES penetrate into HS tissue utilizing its deformable membrane, enters HSF by a pinocytotic-like mechanism, and then releases ALA in the cytoplasm. Subsequently, PDT efficacy is assessed using rabbit HS models. The morphological and histological analysis reveal that ALA-ES gels can improve HS by promoting HSF apoptosis, remodelling collagen fibers and increasing MMP3 expression. The results demonstrate that ALA-ES gels are suitable in clinical treatment of HS and make a substantial progress within the field.
