Bioinspired, surfactant-free, dual-layer asymmetric structures based on polysaccharides, gelatin, and tannic acid for potential applications in biomedicine

Huimin Li,Shen Shen,Hongbo Wang,Jiajia Fu

The formation of dual-layer asymmetric porous structures in surfactant-based systems is significantly influenced by emulsions. Surfactants self-assemble to alter the conformational arrangement of polysaccharides, while gravity disrupts the initial uniformity of the established equilibrium droplet concentration gradient in the emulsion, thus achieving delamination. Specifically, high-speed rotation and non-instantaneous freezing allow the gelatin solution to form two different states of foam layers. The integrated dual-layer asymmetric porous structure, composed of polysaccharides and tannic acid, is constructed with gelatin as a skeleton and surfactant. This innovative approach eliminates the need to consider the toxicity of chemically synthesized surfactants and expands the concept of gelatin utilization. This intriguing structure exhibits a variety of desirable characteristics within 30 days ( e.g. , tailorable performance, ultrarapid antioxidant activity, efficient antibacterial activity, low differential blood clotting index, and good hemocompatibility and cytocompatibility), suggesting its potential as a valuable reference for applying hierarchical porous structures, thereby offering more formulation flexibility for biomaterials with adjustable properties.

Biomimetic Cerium-Assisted Supra-Carbon Dots Assembly for Reactive Oxygen Species-Activated Atherosclerosis Theranostic

Qiao Chen,Xu Yang,Yao Yu,Xinmei Duan,Rongrong Ni,Guojing Song,Li Zhu,Yuan Zhong,Kai Qu,Xian Qin,Kun Zhang,Yang Luo,Wei Wu

Theranostic applications in atherosclerosis plaque microenvironment-triggered nanoplatforms are significantly compromised by the complex synthesis procedure, non-specific distribution, and limited therapeutic function. Therefore, development of a facile and feasible method to construct a pathology-based stimuli-responsive nanoplatform with satisfactory theranostic performance remains a demanding and highly anticipated goal. Herein, a novel class of multifunctional supra-carbon dots (CDs), denoted as MM@Ce-CDs NPs, by a simple nanoassembly and a subsequent coating with macrophage membrane (MM), is developed for the targeted reactive oxygen species-trigged theranostic and positive regulation of the pathological plaque microenvironment in AS. The harvested MM@Ce-CDs NPs exhibit activatable fluorescence properties, photoacoustic characteristics, and cascade enzyme performances, which can be effectively activated under ROS stimulation in the plaque pathological microenvironment, enabling precise control over theranostic functions, while markedly enhancing diagnostic accuracy and therapeutic efficacy for AS management. Besides, MM@Ce-CDs NPs can effectively manipulate the plaque microenvironment by reducing ROS levels and inflammation, alleviating M1 macrophage infiltration, and inhibiting foam cell formation, all together suppressing the pathological plaque development through the synergistic mechanisms. In addition, MM@Ce-CDs NPs inherit the biomimetic biological functions from MM, facilitating a highly specific target delivery to AS.

A tough, antibacterial and antioxidant hydrogel dressing accelerates wound healing and suppresses hypertrophic scar formation in infected wounds

Xiaoqing Liu,Yiming Sun,Jie Wang,Yongyuan Kang,Zhaolong Wang,Wangbei Cao,Juan Ye,Changyou Gao

Wound management is an important issue that places enormous pressure on the physical and mental health of patients, especially in cases of infection, where the increased inflammatory response could lead to severe hypertrophic scars (HSs). In this study, a hydrogel dressing was developed by combining the high strength and toughness, swelling resistance, antibacterial and antioxidant capabilities. The hydrogel matrix was composed of a double network of polyvinyl alcohol (PVA) and agarose with excellent mechanical properties. Hyperbranched polylysine (HBPL), a highly effective antibacterial cationic polymer, and tannic acid (TA), a strong antioxidant molecule, were added to the hydrogel as functional components. Examination of antibacterial and antioxidant properties of the hydrogel confirmed the full play of the efficacy of HBPL and TA. In the in vivo studies of methicillin-resistant Staphylococcus aureus (MRSA) infection, the hydrogel had shown obvious promotion of wound healing, and more profoundly, significant suppression of scar formation. Due to the common raw materials and simple preparation methods, this hydrogel can be mass produced and used for accelerating wound healing while preventing HSs in infected wounds.

A triple-crosslinked, self-healing, polyvinyl alcohol/nanocellulose hydrogel for versatile sensing applications

Danning Fu,Rendang Yang,Ruibin Wang,Yang Wang,Yan Li,Huiyang Bian

Flexible conductive hydrogels (FCHs) have attracted widespread interest as versatile monoliths that can be intricately integrated with various ingredients boasting multiple functionalities. The chemicophysical properties of FCHs cover a wide range, which significantly vary in their building blocks. However, achieving both favorable mechanical strength and high conductivity simultaneously through a facile approach remains a challenge. Herein, polyvinyl alcohol, dialdehyde cellulose nanofibrils, silver nanoparticles, borax, and tannic acid are readily “one-pot” incorporated into FCHs with great tensile stress (499 kPa), tensile strain (4591 %), and compressive stress (269 kPa) due to abundant hydrogen bonding, dynamic borate-diol bonding, and intermolecular acetalization. They also exhibit desired self-healing, generalized-adhesive, and antibacterial performances. Taking advantage of these, FCHs are further employed to support an epidermal sensor, on which remarkable strain sensitivity (gauge factor = 8.22), high-pressure sensitivity (≥ 0.258 kPa −1 ), and fast response (≤ 190 ms) are recorded. Its highly adaptive mechano-electric transformability and functions can be well maintained in serving as an array unit and touch screen pen. The results well addressed in this work are anticipated to pave the universal way of engineering FCHs.

Despite their potential, much remains untapped, whether in standardized extracts or isolated pure compounds. This unexplored terrain has paved the way for significant discoveries in pharmaceuticals. Notably, research has delved into the medicinal properties ofMallotus philippensis, a prominent plant in South Asia. Employing meticulous extraction techniques such as maceration, the fruit of this plant underwent initial antimicrobial screening, revealing encouraging results. Subsequent fractionation of the plant’s extracts via liquid–liquid extractions, utilizing dichloromethane and absolute ethanol, facilitated further analysis. Evaluating these fractions for antibacterial activity demonstrated efficacy against various pathogenic microorganisms, particularlyPseudomonas aeruginosaandEscherichia coli, notably by the ethanolic and dichloromethane extracts. Furthermore, a comprehensive phytochemical analysis unveiled the presence of alkaloids, flavonoids, saponins, glycosides, phenols, and tannins. An assessment of the extracts’ antioxidant potential via the 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay showcased significant activity, with a radical scavenging rate of 97%. This underscores the significance of utilizing fruit remnants, which are often rich in valuable chemical constituents yet commonly discarded, thereby adding value to both the species and the environment. Further investigation focused on the composition ofMallotus philippensisfruit, encompassing volatile and non-volatile metabolites through HPLC-MS analysis. Additionally, this study introduced the application of ionic liquid-loaded polysulfone microcapsules to enrich target constituents from crude extracts. An exploration of the key separation conditions, results, and recycling performance of these microcapsules provided insights for future research endeavors. Overall, this comprehensive study ofMallotus philippensisfruit extracts establishes a foundation for the ongoing exploration and development of this medicinal plant.

A Factor-Free Hydrogel with ROS Scavenging and Responsive Degradation for Enhanced Diabetic Bone Healing

Qin Zhang,Weikai Chen,Guangfeng Li,Zhixin Ma,Mengru Zhu,Qianmin Gao,Ke Xu,Xinru Liu,Wenyi Lu,Wencai Zhang,Yan Wu,Zhongmin Shi,Jiacan Su

In view of the increased levels of reactive oxygen species (ROS) that disturb the osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs), the repair of diabetic bone defects remains a great challenge. Herein, a factor-free hydrogel is reported with ROS scavenging and responsive degradation properties for enhanced diabetic bone healing. These hydrogels contain ROS-cleavable thioketal (TK) linkers and ultraviolet (UV)-responsive norbornene (NB) groups conjugated with 8-arm PEG macromers, which are formed via UV crosslinking-mediated gelation. Upon reacting with high levels of ROS in the bone defect microenvironment, ROS-cleavable TK linkers are destroyed, allowing the responsive degradation of hydrogels, which promotes the migration of BMSCs. Moreover, ROS levels are reduced through hydrogel-mediated ROS scavenging to reverse BMSC differentiation from adipogenic to osteogenic phenotype. As such, a favorable microenvironment is created after simultaneous ROS scavenging and hydrogel degradation, leading to the effective repair of bone defects in diabetic mouse models, even without the addition of growth factors. Thus, this study presents a responsive hydrogel platform that regulates ROS scavenging and stromal degradation in bone engineering.

Bacterial cellulose and photothermal material from biomass-based zero-waste biorefinery functionalized chitosan film for sustainable active food packaging

Li Yang,Wenyang Sheng,Yichen Yang,Ying Yang,Kaiwen He,Guangyang Jiang,Yongqiang Tian

In this study, a multifunctional food packaging film was developed using distillers' grains (DG) through a sustainable, zero-waste biorefinery approach. The objectives were to reduce foodborne contamination, lower manufacturing costs, and enhance the value of DG. Bacterial cellulose (BC) was produced by fermenting the hydrolyzed DG supernatant and then oxidized to form oxidized bacterial cellulose (OBC). OBC was combined with chitosan (CS) to create a CS/OBC composite film. Additionally, a photothermal material, FeDL, was synthesized from DG's enzymatic residue (primarily lignin) and ferrous sulfate heptahydrate. Various concentrations of FeDL were incorporated into the CS/OBC matrix. Characterization using scanning electron microscopy (SEM), X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), and thermal analysis confirmed good compatibility between FeDL and CS/OBC, with slight improvements in thermal stability and tensile strength (TS). The film containing 0.3 mg/mL FeDL achieved a 37.21% reduction in UV transmittance, a 26.81% decrease in water vapor permeability (WVP), and a 175.08% increase in antioxidant activity compared to the CS/OBC film. Under brief near-infrared light (NIR) exposure, the film enabled pasteurization, extending the shelf life of strawberries by approximately 1.75 times. The films also demonstrated biosafety and biodegradability, offering a novel zero-waste biorefining solution from waste biomass.

Bimetallic CuFe2O4 photocatalytic activates persistent free radicals and accelerates electron transfer in biochar: Efficient degradation of tetracycline

Jiahui Wen,Yating Long,Liangdi Zhu,Liang Zeng,Xu Qin,Sirong Song,Manxia Lu,Lanqing Deng

Water pollution has become an urgent issue around the world, antibiotic residues in water result in health or environmental problems worldwide. In order to address the key issues of low catalytic efficiency, poor stability, and low visible light utilization in the current photocatalytic treatment of recalcitrant organic pollutants in water, bimetallic CuFe 2 O 4 /ABC was successfully obtained from extracted Astragalus residue and used for degradation of tetracycline under visible light. First, the bimetallic active centers (Fe and Cu) enhanced the separation of charge carriers under visible light. Then, Fe and Cu active centers activated and enhanced the production of persistent free radicals (PFRs) in biochar. Fe 2+ and Cu + in the bimetallic active centers transferred e - to O 2 to promote the formation of •O 2 − , which efficiently degraded tetracycline (94.89 %) without H 2 O 2 . Notably, the persistent free radicals (PFRs) served as intrinsic electron reservoirs. By donating electrons derived from oxidized C-C bonds to Fe/Cu centers.

Amylopectin from maize-derived carbonized polymer dots as a novel high-efficiency antioxidant for periodontitis treatment

Junjun Liu,Naibo Feng,Xinyu Jiang,Xirui Xin,Yubo Hou,Zhende Jiang,Laijin Lu,Weixian Yu,Bai Yang

Periodontitis is a common chronic inflammatory disease and the local accumulation of excessive reactive oxygen species (ROS) could cause oxidative damage to periodontal tissues. Thus, antioxidative therapy is considered to be the promising strategy for treating periodontitis. However, highly efficient antioxidant activity of biomaterials with low cost, ease of synthesis, and good biocompatibility are still highly desired. Here amylopectin from maize (AFM)-derived carbonized polymer dots (AFM-CPDs-240) with excellent antioxidant activity, good water solubility and biosafty is parepared. Detailed characterizations identify that the antioxidant properties and formation mechanism of AFM-CPDs, and antioxidant activity of AFM-CPDs-240 is much higher than Trolox (a hydrophilic derivative of vitamin E) due to the large contents of conjugated C=C and C=O. Besides, in vitro and in vivo experiments suggest AFM-CPDs-240 could effectively alleviate inflammatory response, inhibit alveolar bone resorption and promote bone formation, showing excellent therapeutic effects on periodontitis. Moreover, the label-free proteomics and western blot results demonstrate AFM-CPDs-240 could accelerate the recovery of periodontitis by blocking TLR4/NF-κB (Toll-like receptor 4/ nuclear factor-kappa B) signaling pathway. This study will develop a simple approach to design highly efficient antioxidant carbon dots with good biocompatibility, and also provide a new promising nanoplatform for treating diseases associated with ROS.

An emulsion extraction system composed of hydrophobic deep eutectic solvent and water for synchronous extraction of oil and phenolic compounds from U.S. pecans [Carya illinoinensis (Wangenh.) K. Koch]

Liling Wang,Yanbin Wang,Yuchuan Qin,Liang He

U.S. pecan nuts are rich in oil and phenolic compounds with important biological functions. At present, the reports about simultaneous extraction of oil and phenolic compounds are limited, and the traditional procedure is complicated and the yield is low. In this study, a new emulsion formed by hydrophobic deep eutectic solvent (DES) and water was used as extractant for synchronous extraction of oil and phenolic components from U.S. pecans, and subsequently the two were recovered from the upper and lower phases based on the incompatibility of DES and water, respectively. The oil and phenolic compounds obtained by the optimization of extraction conditions were 615.29 mg/g and 82.11 mg/g, respectively, which were higher than those obtained by conventional methods. At the same time, the extraction mechanism is discussed through the extraction kinetics and thermodynamics. The extraction process conformed to the Fick’s second law, and it was an endothermic process. After directional recovery by macroporous resin and back-extraction, the effectiveness of the method was proved by the analysis of fatty acid composition of oil, spectral characterization of phenolic compounds and antioxidant activities.

An in situ forming cartilage matrix mimetic hydrogel scavenges ROS and ameliorates osteoarthritis after superficial cartilage injury

Zhicheng Tong,Yuanzhu Ma,Qiushi Liang,Tao Lei,Hongwei Wu,Xianzhu Zhang,Yishan Chen,Xihao Pan,Xiaozhao Wang,Huimin Li,Junxin Lin,Wei Wei,Chong Teng

Superficial cartilage defects represent the most prevalent type of cartilage injury encountered in clinical settings, posing significant treatment challenges. Here, we fabricated a cartilage extracellular matrix mimic hydrogel (GHC, consisting of Gelatin, Hyaluronic acid, and Chondroitin sulfate) to avoid the exacerbation of cartilage deterioration, which is often driven by the accumulation of reactive oxygen species (ROS) and a pro-inflammatory microenvironment. The GHC hydrogel exhibited multifunctional properties, including in situ formation, tissue adhesiveness, anti-ROS capabilities, and the promotion of chondrogenesis. The enhancement of tissue adhesion was achieved by chemically modifying hyaluronic acid and chondroitin sulfate with o-nitrobenzene, enabling a covalent connection to the cartilage surface upon light irradiation. In vitro characterization revealed that GHC hydrogel facilitated chondrocyte adhesion, migration, and differentiation into cartilage. Additionally, GHC hydrogels demonstrated the ability to scavenge ROS in vitro and inhibit the production of inflammatory factors by chondrocytes. In the animal model of superficial cartilage injury, the hydrogel effectively promoted cartilage ECM regeneration and facilitated the interface integration between the host tissue and the material. These findings suggest that the multifunctional GHC hydrogels hold considerable promise as a strategy for cartilage defect repair. Statement of Significance : Superficial cartilage defects represent the most prevalent type of cartilage injury encountered in the clinic. Previous cartilage tissue engineering materials are only suitable for full-thickness cartilage defects or osteochondral defects. Here, we developed a multifunctional GHC hydrogel composed of gelatin, hyaluronic acid, and chondroitin sulfate, which are natural cartilage extracellular matrix components. The drug-free and cell-free hydrogel not only avoids immune rejection and drug toxicity, but also shows good mechanical properties and biocompatibility. More importantly, the GHC hydrogel could adhere tightly to the superficial cartilage defects and promote cartilage regeneration while protecting against oxidation. This natural ingredients and multifunctional hydrogel is a potential material for repairing superficial cartilage defects.

An Injectable Black Phosphorus Hydrogel for Rapid Tooth Extraction Socket Healing

Fanyi Guo,Jianfeng Li,Ziyu Chen,Tianxiao Wang,Ruyu Wang,Tianyao Wang,Yifeng Bian,Yifei Du,Hua Yuan,Yongchu Pan,Jianliang Jin,Huijun Jiang,Feng Han,Jiandong Jiang,Fan Wu,Yuli Wang

The excess production of reactive oxygen species (ROS) will delay tooth extraction socket (TES) healing. In this study, we developed an injectable thermosensitive hydrogel (NBP@BP@CS) used to treat TES healing. The hydrogel formulation incorporated black phosphorus (BP) nanoflakes, recognized for their accelerated alveolar bone regeneration and ROS-scavenging properties, and dl-3-n-butylphthalide (NBP), a vasodilator aimed at enhancing angiogenesis. In vivo investigations strongly demonstrated that NBP@BP@CS improved TES healing due to antioxidation and promotion of alveolar bone regeneration by BP nanoflakes. The sustained release of NBP from the hydrogel promoted neovascularization and vascular remodeling. Our results demonstrated that the designed thermosensitive hydrogel provided great opportunity not only for ROS elimination but also for the promotion of osteogenesis and angiogenesis, reflecting the “three birds with one stone” concept, and has tremendous potential for rapid TES healing.
