Integration of multienzyme co-immobilization and biomimetic catalysis in magnetic metal–organic framework nanoflowers for α-amylase detection in fermentation samples

Liangqiang Chen,Mengdi Hao,Wanqiu Huang,Shaoning Yu,Hao Shen,Fan Yang,Li Wang,Huabin Tu

Multiple enzymes induce biological cascade catalysis is essential in nature and industrial production. However, the shortcomings of enzymes, including unsatisfactory stability, reusability, and sensitivity in harsh microenvironment, have restricted their broader use. Here, we report a facile method for fabricating a cascade system by combining the benefits of immobilized enzymes and biomimetic catalysis based on magnetic metal–organic framework nanoflowers (mMOFNFs). mMOFNFs prepared through the layered double hydroxide-derived strategy exhibited remarkable peroxidase-like activity and accessible amino interface, enabling it to serve not only as a reliable carrier for α-glucosidase and glucose oxidase fixation, but also as a nanozyme participating in cascade. On this basis, a colorimetric biosensor of excellent sensitivity and selectivity for α-amylase detection was constructed with a wide range (2–225 U L −1 ), low detection limit (2.48 U L −1 ), and rapid operation (30 min). This work provides a versatile strategy for establishing multi-enzyme cascade systems and rapid analysis of α-amylase.

Covalent immobilization of α-amylase on hollow metal organic framework coated magnetic phase-change microcapsules for the improvement of its thermostability

Yixin Liu,Zhiping Sun,Cunhui Wu,Xinguang Qin,Gang Liu,Xinlin Wei,Haizhi Zhang

Exploring efficient immobilized carrier for α-Amylase (α-Amy) to enhance its thermostability has significant influence in starch related industry. Here, hollow ZIF-8 (HZIF-8) with amino groups coated magnetic phase change microcapsules (PCM) was designed for covalent immobilization of α-Amy. Magnetic PCM consisting n-docosane core and SiO 2 /Fe 3 O 4 hybrid shell were firstly synthesized. Then, HZIF-8 shell with amino groups was coated and α-Amy was subsequently immobilized through covalent cross-linking strategy. The morphology, chemical structure and magnetic property of PCM@HZIF-8@α-Amy (PCMHA) were comprehensively characterized. Moreover, heat control property of PCMHA was studied with encapsulation efficiency and thermal energy-storage efficiency of 50.55 % and 50.59 %, respectively. Catalytic activity of immobilized α-Amy was fully investigated with K m and V max of 2.773 mg/mL and 1.853 μmol/mL·min, respectively. From reusability and storage stability study, immobilized α-Amy not only maintained rather high activity after 9 cycles' reuse, but also exhibited good activity under high salt ion condition after 7 days.

Designable immobilization of D-allulose 3-epimerase on bimetallic organic frameworks based on metal ion compatibility for enhanced D-allulose production

Huayang Tang,Yian Chen,Dexun Fan,Fengguang Zhao,Shuangyan Han

D-allulose, a low-calorie rare sugar catalyzed by D-allulose 3-epimerase (DAE), is highly sought after for its potential health benefits. However, poor reusability and stability of DAE limited its popularization in industrial applications. Although metal-organic frameworks (MOFs) offer a promising enzyme platform for enzyme immobilization, developing customized strategies for MOF immobilization of enzymes remains challenging. In this study, we introduce a designable strategy involving the construction of bimetal-organic frameworks (ZnCo-MOF) based on metal ions compatibility. The DAE@MOFs materials were prepared and characterized, and the immobilization of DAE and the enzymatic characteristics of the MOF-immobilized DAE were subsequently evaluated. Remarkably, DAE@ZnCo-MOF exhibited superior recyclability which could maintain 95 % relative activity after 8 consecutive cycles. The storage stability is significantly improved compared to the free form, with a relative activity of 116 % remaining after 30 days. Molecular docking was also employed to investigate the interaction between DAE and the components of MOFs synthesis. The results demonstrate that the DAE@ZnCo-MOF exhibited enhanced catalytic efficiency and increased stability. This study introduces a viable and adaptable MOF-based immobilization strategy for enzymes, which holds the potential to expand the implementation of enzyme biocatalysts in a multitude of disciplines.

A facile and effective electrochemical immunosensor based on controlled and oriented immobilization of natural antibodies for sensitive detection of aflatoxin B1

Yu-Min Zhang,Hong-Ming Yang,Chong-Mei Xu,Jia-Yi Ju,Tian-Shui Xue,Yan Jiang,Zi-Yu Jia,Xiao-Ming Gong,Xian-Zhong Zeng,Jin-Bao Tang

Aflatoxin B 1 (AFB 1 ) is one of the most common and harmful toxins in agricultural products, and the development of a sensitive, simple, and accurate detection method for AFB 1 is crucial for ensuring continuous monitoring and safeguarding food safety. Herein, we describe a facile and effective electrochemical immunosensor for ultrasensitive detection of AFB 1 . In this framework, a novel Fc-specific cysteine (Cys)-modified antibody was first fabricated by employing photoaffinity labeling technique, which enables antibodies to be directionally immobilized onto a bare gold electrode via Au-sulfhydryl interaction. The proposed immunosensor exhibited detection limits (LODs) of 0.054 and 0.019 ng mL −1 for direct competitive and sandwich-type competitive assays, respectively. Notably, these LODs were approximately 8-fold and 10-fold lower, respectively, compared to those achieved with randomly immobilized antibodies. The proposed immunosensor demonstrated excellent stability and reproducibility, along with high sensitivity, accuracy, and precision in AFB 1 detection, suggesting its significant potential for applications in food safety applications.

Humic and fulvic acid-mediated dissimilatory iron reduction coupled with phosphate solubilizing bacteria inducing secondary mineral formation for efficient cadmium immobilization

Gongting Wang,Wenqing Luo,Zedong Teng,Xin Zhao,Yuxin Cheng,Yanzhong Wang,Bin Liu,Mengnan Lu,Min Li

Cadmium (Cd) pollution in sediments has become an urgent environmental problem. Humus mediated dissimilatory iron reduction and phosphorus solubilizing bacteria (PSB) were widely used for Cd immobilization. Here, we utilized humic acid (HA) and fulvic acid (FA) to promote the efficiency of iron reduction by Geobacter metallireducens GS-15 to immobilize Cd with the involvement of PSB. The results showed that both HA and FA can facilitate the reduction of Fe(III) and the release of P in ferric phosphate minerals, and then the immobilization of Cd. The immobilization of dissolved Cd 2 + in HA, FA and BCK groups were 76.25 %, 65.25 % and 63.58 %, respectively. HA and FA promoted the growth of PSB and the secretion of acetic acid, high concentration of acetic acid provided sufficient nutrients for the growth of iron-reducing bacteria. Thereby, high concentration of Fe(II) facilitated the recrystallization, which effectively immobilized Cd 2+ . Compared to the CK group, the HA and the FA groups increased the proportion of exchanged Cd converted to combined Cd in secondary minerals by 25.32 % and 19.16 %, respectively. The findings reveal that enhanced effect of HA on microbial iron reduction coupled with PSB can facilitate the immobilization of Cd, providing a promising strategy for the remediation of Cd-contaminated sediments.

Tannic acid mediated surface functionalization of polylactic acid nanofiber films for in situ AgNPs immobilization: antioxidant, antibacterial and fresh-keeping properties

Yugang Song,Rongli Li,Shunlin Yang,Hong Yu,Yanziwen Zhang,Lihua Zhang,Haibo Xie

The application of biodegradable and environment-friendly packaging materials with antioxidant and antibacterial activities is urgent and essential for food fresh-keeping. In this work, surface-functionalized biodegradable electrospun polylactic acid (PLA) nanofiber films were fabricated via a simple two-step strategy, which integrated the bioinspired coating of tannic acid (TA, a naturally occurring plant polyphenol) and the subsequent in situ immobilization of silver nanoparticle (AgNPs). The TA coating endowed the nanofiber films with excellent antioxidant activity, while simultaneously functioning as reductant and stabilizer for the in situ synthesis of AgNPs from silver source (silver nitrate), and consequently, the reduction process did not necessitate the incorporation of extra reducing or stabilizing agents. The surface morphology and chemical composition of the functionalized PLA nanofiber films were characterized by SEM, EDX and XPS. The TA-mediated surface modification increased the nanofiber diameter compared to pristine PLA film, and the generated AgNPs made the nanofibers much rougher. The fabricated PLA-TA@AgNPs nanofiber films exhibited enhanced mechanical properties, with tensile strength increasing and ∼ 193 % compared to pristine PLA films. Furthermore, the introduction of TA coating impressively imparted the films with excellent antioxidant activity, demonstrating a potent scavenging effect on DPPH and ABTS free radicals. In vitro antibacterial assay showed excellent antibacterial ability of the PLA-TA@AgNPs nanofiber films against Escherichia coli and Staphylococcus aureus. Furthermore, the functional PLA-TA@AgNPs nanofiber films extended the shelf life of strawberries to at least 8 days at room temperature, as indicated by the appearance, weight loss and firmness loss, demonstrating great potential for use in fresh-keeping packaging applications.

Co-immobilization of rhodium catalyst and LDH on carbon electrode by covalent graft strategy for electrosynthesis L-lactate in bipolar electrochemistry

Chunhua Zhang,Bowen Li,Xiang Gao,Wei Xiao,Mingfang Chen,Qilong Bian

This work demonstrates a wireless electroenzymatic platform using carbon microsphere electrodes co-immobilized with a rhodium catalyst and lactate dehydrogenase (LDH) for integrated NADH regeneration and pyruvate-to-L-lactate conversion in a bipolar electrochemical system. The 3D "aggregate reactor" architecture, functionalized via diazonium grafting, Rh coordination, and covalent enzyme coupling, enables spatial coupling of cofactor recycling and biocatalysis, while HPLC revealed a L-lactate production rate of 0.04 mM cm-2 h⁻¹ under 12.5 V cm⁻¹ with a plenty of carbon beads attributed to enhanced mass transport. This wireless, scalable design eliminates wiring constraints and offers modular adaptability for sustainable synthesis of chiral chemicals.

Poly (2-methyl-2-oxazoline) coating by thermally induced immobilization for determination of bovine lactoferrin in infant formula with capillary electrophoresis

Ke Mao,Haiqin Du,Longchao Bai,Yalin Zhang,Haikun Zhu,Yanmei Wang

In this work, a one-step coating procedure by a simple annealing protocol of poly (2-methyl-2-oxazoline)-random-glycidyl methacrylate (PMOXA-r-GMA) copolymer was used to yield covalent and cross-linked PMOXA-based antifouling coating on a fused-silica capillary inner surface, which was used to determine the bovine lactoferrin (Lf) in infant formula by capillary electrophoresis with ultraviolet (CE-UV) detection. The X-ray photoelectron spectroscopy (XPS) confirmed that PMOXA-r-GMA could be bonded onto fused-silica capillary inner wall and stable electroosmotic flow (EOF) was obtained in the PMOXA-r-GMA coated capillary at pH 2.2–9.0. The separation of a mixture of four basic proteins indicated that the PMOXA-r-GMA coated capillary exhibited excellent separation efficiency for the basic proteins. Therefore, the PMOXA-r-GMA coated capillary was used to determine the quantity of Lf in infant formula. Under the optimal conditions, the peak area (A) and the concentration of Lf showed a good linear relationship within the range of 10–500   μg/mL with a linear correlation coefficient of 0.9995. The limit of detection (LOD) and limit of quantitation (LOQ) for Lf were 5.0   μg/mL and 16.7   μg/mL, respectively. The recoveries at spiked concentrations of 0.20 and 0.40   mg/mL Lf in infant formula were 97.1±5.5% – 97.8±5.1%. The determined values of Lf in infant formula samples with Lf were consistent with the nominal values, indicating that our CE method could be successfully applied to the quantitative analysis of Lf in commercial infant formula.

Facile synthesis and immobilization of MoS2 and SnO2 co-modified g-C3N4 ternary heterostructure for enhanced visible light photodegradation of RhB

Si Jiang,Jun Xiao,Jieyi Tan,Xiangqi Li

Constructing heterojunctions is an important strategy for enhancing the separation efficiency of charge carriers in graphitic carbon nitride (g-C 3 N 4 ). Meanwhile, immobilization enables the recycle of g-C 3 N 4 -based photocatalysts from wastewater. Herein, a visible light-responsive MoS 2 and SnO 2 co-modified g-C 3 N 4 ternary heterostructure was synthesized via one-pot calcination and subsequently immobilized in a modified bacterial cellulose (BC) aerogel through freeze-drying. The prepared ternary heterojunction exhibited enhanced visible light-harvesting capacity and charge separation efficiency due to intimate interfaces and the synergy effect of different components. It showed a degrade rate constant 3.43 times higher than that of g-C 3 N 4 , and degraded 99.3 % of RhB (30 mg/L) at pH 6.5 after 30 min of irradiation, and 99.1 % at pH 5 after 15 min. Owing to dispersion improved by sodium dodecyl benzene sulfonate (SDBS) and the strength and hydrophobicity enhanced by methoxytrimethylsilane (MTMS), the prepared aerogel degraded 98.7 % of RhB (30 mg/L) at 60 min irradiation, and maintained 94.0 % after 6 cycles. This work demonstrates a highly efficient MoS 2 -SnO 2 -g-C 3 N 4 ternary photocatalyst and a practical immobilization route for application in purification of dye wastewater.

Simple Purification and Immobilization of His-Tagged Organophosphohydrolase from Cell Culture Supernatant by Metal Organic Frameworks for Degradation of Organophosphorus Pesticides

Saiguang Xue,Jiaojiao Li,Liya Zhou,Jing Gao,Guanhua Liu,Li Ma,Ying He,Yanjun Jiang

Coordinating unsaturated metal sites (CUS) on the surface of metal–organic frameworks (MOFs) could be used to adsorb His-tagged proteins. The specific adsorption between CUS and His-tagged proteins could reduce preparation steps, shorten preparation time, and could also avoid the binding between the metal ion of metalloenzyme active center and the chelating agent to ensure the enzyme activity. In this study, MIL-88A was synthesized by hydrothermal method and used to purify and immobilize His-tagged organophosphohydrolase (OpdA) in one step for organophosphate bioremediation. Under optimized conditions, OpdA@MIL-88A had a maximal activity of 1554 U/gprotein, which was nearly 5 times higher than free OpdA. Compared with free OpdA, OpdA@MIL-88A exhibited improved organic solvent tolerance, SDS tolerance, thermal stability, and storage stability. OpdA@MIL-88A was used to degrade organophosphorus pesticides on grapes and cucumbers. After reuse 6 times, OpdA@MIL-88A retained more than 66% and 61% of the initial activity, respectively. Therefore, this proposed strategy provided a facile and effective method for degradation of organophosphorus pesticides.

Arsenic speciation, oxidation and immobilization in an unsaturated soil in the presence of green synthesized iron oxide nanoparticles and humic acid

Shurui Guo,Mingqin Jiang,Jiajiang Lin,Nasreen Islam Khan,Gary Owens,Zuliang Chen

While the availability of arsenic (As) in soil is well known to be highly correlated with the presence of iron (Fe) oxides and humic acid (HA) in the soil, the relationship between Fe oxides and HA and As species in the soil is less well understood. In this study, As speciation in an unsaturated soil in the presence of external HA and green synthesized Fe oxide nanoparticles (FeNPs) showed that As(V) was mainly distributed to the specifically-bound (F2), amorphous and poorly-crystalline hydrous oxides of Fe, Al (F3) and the well-crystallized hydrous oxides of Fe and Al (F4). While As(III). This was the major component in unsaturated soil, and was mainly distributed to F4 and the residual fraction (F5). As bound to F3 and F5 was most sensitive to the addition of HA and FeNPs, while HA/FeNPs treatment increased the F3-bound As(V); however, it decreased the F5-bound As(III). Nonetheless the effect of HA on As is completely different to the HA/FeNPs treatment. The increase of As(V) in F3 resulted from F5-bound As(III) oxidation when treated by HA/FeNPs. Cyclic voltammetry confirmed that HA and Fe 3+ /Fe 2+ redox enhanced As(III) oxidation, while FTIR revealed that HA-bound As(III) was the least available fraction in the soil. Finally, a mechanism involving a combination of HA and FeNPs was proposed for explaining the redistribution of As species in the soil.

Immobilization of β-Cyclodextrin-Conjugated Lactoferrin onto Polymer Monolith for Enrichment of Ga in Metabolic Residues of Ga-Based Anticancer Drugs

Haijiao Zheng,Tenggao Zhu,Xiqian Li,Guan Wang,Qiong Jia

Biological-material-functionalized porous monoliths were prepared with lactoferrin and β-cyclodextrin via a click reaction. With the monolith as an extraction medium, a method combined with ICP-MS was developed for the determination of total gallium originating from metabolic residues of orally bioavailable gallium complexes with tris(8-quinolinolato)gallium (GaQ3) as a representative. The method exhibited favorable adsorption behaviors for gallium with high selectivity, low detection limit (2 ng L–1), and an enrichment factor of 29-fold with the sample throughput of 30 min–1. The developed approach was validated by the analysis of gallium from GaQ3 metabolic residues in a human cell line. Additionally, the practical applicability of this method was evaluated by the determination of gallium in human blood and urine samples from cancer patients. Results illustrated that the prepared monolith had potential in Ga-based anticancer drug analysis in complex biological samples.
