MAPbI3 Quasi-Single-Crystal Films Composed of Large-Sized Grains with Deep Boundary Fusion for Sensitive Vis–NIR Photodetectors

Tao Wang,Gang Lian,Liping Huang,Fei Zhu,Deliang Cui,Qilong Wang,Qingbo Meng,Ching-Ping Wong

Perovskite single-crystal (SC) or quasi-single-crystal (QSC) films are promising candidates for excellent performance of photoelectric devices. However, it is still a great challenge to fabricate large-area continuous SC or QSC films with proper thickness. Herein, we propose a pressure-assisted high-temperature solvent-engineer (PTS) strategy to grow large-area continuous MAPbI3 QSC films with uniformly thin thickness and orientation. Dramatic grain growth (∼100 μm in the lateral dimension) and adequate boundary fusion are realized in them, vastly eliminating the grain boundaries. Thus, remarkable diminution of the trap density (ntrap: 7.43 × 1011 cm–3) determines a long carrier lifetime (τ2: 1.7 μs) and superior photoelectric performance of MAPbI3-based lateral photodetectors; for instance, an ultrahigh on/off ratio (>2.4 × 106, 2 V), great stability, fast response (283/306 μs), and high detectivity (1.41 × 1013) are achieved. The combination properties and performance of the QSC films surpass most of the reported MAPbI3. This effective approach in growing perovskite QSC films points out a novel way for perovskite-based optoelectronic devices with superior performance.

An NIR-activated polymeric nanoplatform with ROS- and temperature-sensitivity for combined photothermal therapy and chemotherapy of pancreatic cancer

Xiang Zhan,Xuan Nie,Fan Gao,Cong Zhang,Ye-Zi You,Yue Yu

Nanomedicine has attracted growing attention due to its designability and functionality, as well as its excellent pharmacokinetics with limited side effects, and recently, combined therapies have become desirable as they can obtain enhanced therapeutic efficacy by using nanomedicine. Herein, we have reported a functional drug delivery system with a dual response to temperature and reactive oxygen species to efficiently eliminate pancreatic cancer cells in a combined therapy strategy. Functional micelles with camptothecin (CPT) in the core and indocyanine green (ICG) on the surface could effectively accumulate in tumor sites through the EPR effect. The ROS in the tumor microenvironment trigger the conversion of an amino-based copolymer to a carboxy based copolymer, releasing the loaded ICG to reduce the size of the micelles with high penetration in tumor tissue. On the one hand, under 808 nm light irradiation, the micelles will produce the heat to kill tumor cells via photothermal therapy. On the other hand, the generated heat could further trigger the transition of a copolymer from a hydrophobic to a hydrophilic state, releasing the loaded CPT into the deep tumor cells to achieve chemotherapy. The in vitro and in vivo experiments revealed that this combined therapy could combat pancreatic cancer cells with an enhanced therapeutic effect.

A tumor-targeted theranostic nanomedicine with strong absorption in the NIR-II biowindow for image-guided multi-gradient therapy

Qi Chen,Ziliang Zheng,Xiaojing He,Shuo Rong,Yufei Qin,Xiaoyang Peng,Ruiping Zhang

Developing new strategies to enhance drug accumulation in the tumor and therapeutic efficacy is of great importance in the field of tumor therapy. Herein, a peanut-like multifunctional nanomedicine (CuS-PGH NMs) made of CuS nanoparticles encapsulated in poly(L-lysine)(PLL)/glucose oxidase (GOx)-hyaluronic acid (HA) shells has been constructed via layer-by-layer (LbL) assembly, and shows good biocompatibility and effective multi-gradient therapy. Because of the enhanced permeability and retention (EPR) effect, the CuS-PGH NMs could significantly enhance the cellular uptake by tumors overexpressing CD44 receptors, which respond to hyaluronidase (HAase)-triggered surface charge conversion. Once internalized by the tumor, GOx was the first to be exposed and could effectively deplete endogenous glucose for starvation therapy, and the excess H2O2 was then converted into highly toxic hydroxyl radicals (˙OH) via a Cu+-mediated Fenton-like reaction for chemodynamic therapy (CDT). Meanwhile, the as-obtained Cu+ ions accompanied the regenerated less-active Cu2+ ions. Interestingly, the high content of H2O2 could, in turn, accelerate Cu2+/Cu+ conversion to promote the Cu+–H2O2 reaction for enhanced chemodynamic therapy (CDT), thereby achieving efficient tumor growth suppression via synergistic starvation/CDT therapy. Subsequently, owing to the strong NIR-II absorption capability of CuS-PGH NMs, effective photothermal tumor ablation of the weakened tumor cells could be realized with the precise guidance of NIR-II PAI. This multi-gradient therapeutic strategy has been demonstrated to have excellent antitumor activity with minimal nonspecific damages, and offers a new avenue to precise tumor therapy.

1,6-Elimination reaction induced detection of fluoride ions in vitro and in vivo based on a NIR fluorescent probe

Aiqing Feng,Yongmei Jia,Liping Huang,Lin Wang,Guohua Zhou,Sheng Wang,Peilian Liu

Near-Infrared “turn on” type fluorescent probes are attractive and promising in the fields of chemical sensing and bioimaging. Here, a new dicyanomethylene-4H-pyran derivative (DCM-Si) NIR fluorescent probe was designed and synthesized for specific lighting up F − in living cells and bodies. Si O bond was used as F − trigger, and the release of fluorophore (DCM-NH 2 ) occurred after substituent reaction and subsequent 1,6-elimination. This NIR probe displayed high sensitivity and selectivity for the sensing of F − , and the detection limit was calculated to be as low as 157 nM. Moreover, the “off-on” fluorescent signal changes can be realized by adding F − in living cells and zebrafish embryos.

Allyl sulfide-based visible light-induced dynamically reshaped liquid crystalline elastomer/SWCNT nanocomposites capable of multimode NIR photomechanical actuations

Zhongying Wu,Pin Cheng,Weiguang Zhao,Jianglin Fang,Tianchi Xu,Dongzhong Chen

Liquid crystalline elastomers (LCEs) combine the ordering character of a liquid crystal and the elasticity of a polymer network. The development of fully light-driven LCE-based soft actuators with good reshapeability and recyclability is in high demand. In this work, we prepared SWCNT/LCE composite films through the introduction of allyl sulfide-based dynamic covalent crosslinks in the LCE matrix and the well-dispersed filler single-walled carbon nanotubes (SWCNTs) with a strong photothermal effect. The SWCNT/LCE composite films were facilely fabricated with reorientation and remouldable capabilities regulated by 405 nm visible light. The monodomain composite films demonstrated multimode actuations of contraction, bending, curling, stretching recovery, and even shape memory driven by 808 nm near-infrared (NIR) light. Moreover, the monodomain spiral ribbon or stretching flat strip and the original polydomain composite film can be readily transformed into each other, and reshaped through easily achievable fabrication conditions. Therefore, the well-prepared SWCNT/LCE composite films with exchangeable covalent crosslinks showed promise as candidates for the development of multimode NIR photomechanical soft actuators with fully light-driven controllability and good remouldability.

A dual-usage near-infrared (NIR) cell membrane targeting chimeric peptide for cancer cell membrane imaging and photothermal ablation

Chen Pei-Ling,Shi Qun-Ying,Chen Tian,Wang Ping,Liu Yun,Liu Li-Han

Due to the low background and deep tissue penetration of near-infrared light, near-infrared fluorescence (NIRF) probes have received much attention in recent years. Cell membrane targeting probes can be used in in vitro and in vivo for living cell imaging, disease diagnosis and tumor targeted therapy. However, most of the currently available cell membrane probes are blue or green emissive fluorophores, NIRF probes are relatively rare. Herein, we designed and prepared a NIRF chimeric peptide probe named C 16 -CARK, which could target the tumor cell membrane with longtime retention (more than 4 h). Moreover, the C 16 -CARK can heat and disrupt the cell membrane upon high irradiation dose, revealing its dual roles as a cell membrane imaging probe and a photothermal agent. This kind of near-infrared fluorescence probe with cell membrane targeting and retention property provides a well design paradigm for the field of cancer imaging and treatment.

AgBiS2-TPP nanocomposite for mitochondrial targeting photodynamic therapy, photothermal therapy and bio-imaging under 808 nm NIR laser irradiation

Mingdi Sun,Dan Yang,Chen Wang,Huiting Bi,Yuan Zhou,Xiangxi Wang,Jiating Xu,Fei He,Shili Gai,Piaoping Yang

808 nm near-infrared (NIR) light-induced biological theranostics is gradually becoming a popular method for cancer treatment. Meanwhile, mild synthetic methods to prepare medicines and gentle treatment conditions for cancer patients are becoming increasingly important to oncotherapy. Herein, tiny AgBiS2 nanodots were synthesized via a simple method, and for the first time, discovered to produce a photodynamic therapy (PDT) effect for cancer treatment under 808 nm laser irradiation, which was characterized by both chemical probe and intracellular reactive oxygen species (ROS) detection. Subsequently, because tumor cells have more mitochondria than normal cells to generate more energy to maintain rapid growth for population expansion, and triphenylphosphonium (TPP) can transport tiny nanoparticles into the mitochondria, the as-synthesized AgBiS2 nanodots were combined with TPP in a facile route. In our design, the AgBiS2 nanodots exhibit photothermal properties and TPP can enhance the photothermal properties of the AgBiS2 nanodots to a certain extent, which make the AgBiS2-TPP nanocomposite applicable in photothermal therapy (PTT). Furthermore, the AgBiS2-TPP nanocomposite showed a remarkable computed tomography (CT) imaging performance for tumor diagnosis. The facile synthetic strategy, satisfactory anticancer effect, CT imaging and mitochondrial targeting of the AgBiS2-TPP nanocomposite demonstrate its high potential in the anticancer field.

Flexible and Reusable Non-woven Fabric Photodetector Based on Polypyrrole/Crystal Violate Lactone for NIR Light Detection and Writing

Yang Shuangping,Macharia Daniel K.,Ahmed Sharjeel,Zhu Bo,Zhong Qiuping,Wang Haifeng,Chen Zhigang

Rapid NIR light detection and/or writing has drawn much attention, but their practical applications have been limited by obtaining such NIR photodetectors. To address this problem, we have developed a simple and versatile strategy to prepare a non-woven fabric photodetector. The blue non-woven fabric photodetector has been prepared by coating photo-thermochromic ink (including crystal violet lactone (CVL) as the thermo-sensitive dye, polypyrrole (PPy) nanospheres as the photothermal component and hydroxyethyl cellulose (HEC) as the polymer matrix) on white non-woven fabric. When the blue fabric photodetector is irradiated by NIR (808-nm as model, 0.75 W cm −2 ) laser, the discoloration occurs in 35 s, and higher laser intensity confers more rapid discoloration. This discoloration results from the photothermal effect of PPy which confers the elevation of temperature (> 50 °C) and then converts CVL to its leuco form (colorless). When the laser is turned off, the temperature drops to below the transition temperature (< 43 °C), and then CVL reverts to its initial blue color. Moreover, different figures and images can be easily printed on the fabric photodetector by 808 nm laser, and then they can be erased automatically under ambient conditions, with excellent cycling stability. Therefore, this fabric photodetector may act as a new platform for rapid NIR light detection and writing.

NIR-Triggered Theranostic Bi2S3 Light Transducer for On-Demand NO Release and Synergistic Gas/Photothermal Combination Therapy of Tumors

Yao Huang,Junqing Huang,Mingyang Jiang,Songjun Zeng

Gaseous therapy based on nitric oxide (NO) has emerged as a potential tumor therapy route. However, the development of a NO generator with precisely controlled NO releasing in the target site is still facing enormous challenges. Herein, a typical NO delivery platform by integrating the hollow structured polydopamine (PDA) coated Bi2S3 nanohybrids (denoted as H-Bi2S3@PDA) with a heat-sensitive S-nitrosothiol (RSNO) molecule was explored for near-infrared (NIR) light-responsive on-demand NO-releasing and synergetic gas/photothermal combination therapy of tumors. The designed H-Bi2S3@PDA nanocomposites hold high photothermal conversion efficiency of 46.13% under irradiation of a NIR laser, leading to NIR light-triggered controllable generation of NO gas via breaking the S–NO bonds of the heat-sensitive RSNO molecules. More importantly, synergistic gas/photothermal therapy of the tumor was also achieved, which presented remarkable inhibition growth and ablation of tumors. These findings provide the opportunity to design a NIR light-activated NO generator with a combination of photothermal materials and heat-sensitive molecules for integrative gas/photothermal therapy in deep tumors.

Bimetallic nanodots for tri-modal CT/MRI/PA imaging and hypoxia-resistant thermoradiotherapy in the NIR-II biological windows

Meng Lyu,Daoming Zhu,Yanhong Duo,Yang Li,Hong Quan

Hypoxic tumor microenvironment leads to resistance or failure of radiotherapy (RT). As a non-invasive therapy, photothermal therapy (PTT) can improve the tumor hypoxic microenvironment in addition to directly killing tumor cells. PTT combined with RT (thermoradiotherapy) becomes an emerging treatment. Multi-functional nanoparticles used for hypoxia-resistant thermoradiotherapy in the second near-infrared (NIR-II) biological windows (1000–1700 nm) are urgently needed to be developed. Here, a facil method synthesis of ultra-small cysteamine (Cys)-coated FePd bimetallic nanodots (NDs) is reported. These NDs can not only produce effective hyperthermia (35.4%) when irradiated in the NIR-II region (1064 nm) but also have an enhanced radiation effect due to i) Hypoxic improvement in tumor tissues by photothermal treatment in the NIR-II Biological Windows can greatly enhance the sensitivity of tumor cells to radiotherapy ii) The ability of NDs to deposit radiation energy in tumors has further enhanced the sensitivity of tumor cells to radiotherapy. Meanwhile, NDs was a contrast agent for tri-modal imaging including computed tomography (CT)/magnetic resonance imaging (MRI)/photoacoustic imaging (PAI) in vitro and in vivo . Both in vitro and in vivo tests demonstrated good biocompatibility and excellent stability of NDs, indicating great potential for clinical applications.

Rational Design of IR820- and Ce6-Based Versatile Micelle for Single NIR Laser–Induced Imaging and Dual-Modal Phototherapy

Xu Hu,Hailong Tian,Wei Jiang,Aixin Song,Zhonghao Li,Yuxia Luan

Phototherapy as a promising cancer diagnostic and therapeutic strategy has aroused extensive attention. However, single-wavelength near-infrared (NIR) light–triggered combinational treatment of photothermal therapy (PTT) and photodynamic therapy (PDT) is still a great challenge. Herein, a multifunctional micelle activated by a single-wavelength laser for simultaneous PTT and PDT as well as fluorescence imaging is developed. Briefly, new indocyanine green (IR820) is conjugated to d -α-tocopheryl polyethylene glycol 1000 succinate (TPGS) via the linker 6-aminocaproic acid, and then, chlorin e6 (Ce6) is encapsulated into the micelles formed by TPGS–IR820 conjugates to fabricate TPGS–IR820/Ce6 micelles. As the well-designed TPGS–IR820 conjugate shares a similar peak absorption wavelength with Ce6, this micelle can be applied with a single NIR laser (660 nm). The stable micelles exhibit excellent photothermal conversion efficiency in vitro and in vivo as well as high singlet oxygen generation capacity in tumor cells. After efficient cellular internalization, the as-prepared micelles display outstanding anticancer activity upon single NIR laser irradiation in vitro and in vivo. Furthermore, TPGS–IR820/Ce6 micelles show negligible systemic toxicity. The highly safe and effective TPGS–IR820/Ce6 micelles can offer an innovative strategy to construct single NIR light–induced PTT and PDT combined phototherapy nanoplatforms via suitable modification of organic phototherapeutic agents.

CuS@mSiO2-PEG core–shell nanoparticles as a NIR light responsive drug delivery nanoplatform for efficient chemo-photothermal therapy

Xijian Liu,Qilong Ren,Fanfan Fu,Rujia Zou,Qian Wang,Guobing Xin,Zhiyin Xiao,Xiaojuan Huang,Qian Liu,Junqing Hu

We report a facile and low-cost approach to design a difunctional nanoplatform (CuS@mSiO2-PEG) as a near-infrared (NIR) light responsive drug delivery system for efficient chemo-photothermal therapy. The nanoplatform demonstrated good biocompatibility and colloidal stability, as well as high loading capacity for the anticancer drug (26.5 wt% for doxorubicin (DOX)). The CuS nanocrystals (core) within these CuS@mSiO2-PEG core–shell nanoparticles can effectively absorb and convert NIR light to fatal heat under NIR light irradiation for photothermal therapy, and the release of DOX from the mesoporous silica (shell) can be triggered by pH and NIR light for chemotherapy. When the CuS@mSiO2-PEG/DOX nanocomposites were irradiated by 980 nm light, both chemotherapy and photothermal therapy were simultaneously driven, resulting in a synergistic effect for killing cancer cells. Importantly, compared with chemotherapy or photothermal treatment alone, the combined therapy significantly improved the therapeutic efficacy.
