Effect of Ginkgo biloba seed exopleura extract and chitosan coating on the postharvest quality of ginkgo seed

Fang Tian,Weiliang Chen,Gongjian Fan,Tingting Li,Xiaohong Kou,Cai'E Wu,Zhihao Wu

Background The effects of Ginkgo biloba seed exopleura extract (GSEE) and chitosan (CH) coating on the preservation of ginkgo seeds were investigated. Results Results showed that CH could alleviate the decay rate, and that CH combined with GSEE (CH–GSEE) treatment further inhibited the development of seed decay due to the additional antifungal activity of GSEE. The nutrient contents, including water, starch and soluble protein, were maintained by both CH–GSEE and CH treatments. CH–GSEE treatment led to better restriction on oxidative stress by decreasing superoxide anion production rate, membrane permeability, malondialdehyde content, respiration rate and ethylene production rate. The antioxidant enzyme activities of peroxidase, superoxide dismutase and catalase in ginkgo seeds were maintained by treatment with CH–GSEE at a higher level. These results were consistent with the enhanced visual appearance, qualities and storability of the CH–GSEE-treated seeds. Principal component analysis provided a global view of the internal relations of the ginkgo seeds with different treatments. Conclusions The postharvest qualities of CH–GSEE-treated seeds were better than those of other treatment groups. Therefore, CH–GSEE is an effective and alternative way for inhibiting decay, maintaining quality and extending the postharvest life of ginkgo seeds. © 2018 Society of Chemical Industry

Functional and correlation analyses of dihydroflavonol-4-reductase genes indicate their roles in regulating anthocyanin changes in Ginkgo biloba

Jun Ni,Rujue Ruan,Lijun Wang,Zhifang Jiang,Xiajun Gu,Lisha Chen,Maojun Xu

Anthocyanins, a class of flavonoids, are synthesized in a branch of the flavonoid pathway. Dihydroflavonol-4-reductase (DFR) catalyzes the first reaction leading to anthocyanins, and it is considered a key regulatory enzyme of anthocyanin biosynthesis in plants. In this research, we isolated three new DFR genes by searching the whole-genomic sequence data of Ginkgo biloba . The genes have similar structures and are clustered in the same scaffold. Sequence analyses of GbDFR proteins revealed that these GbDFRs have a closer relationship to Arabidopsis thaliana DFRs than previously identified GbDFRs. Expression analyses revealed that GbDFR4 and GbDFR6 were preferentially expressed in leaves and fruit, respectively. Young and mature leaves shared similar gene expression patterns, while fruit harvested in May had significantly higher expression levels of GbDFR s than fruit harvested in June. Transgenic tobacco plants ectopically overexpressing GbDFR genes showed higher DFR enzymatic activity levels, higher anthocyanin contents and darker flowers than the control. Ginkgo seedlings treated with NaCl showed increased total flavonoid and anthocyanin contents, and the expression levels of GbDFR genes were also induced. In contrast, UV-B irradiation increased the total flavonoid content but decreased both the anthocyanin content and GbDFR expression levels. A salicylic acid treatment induced and inhibited the total flavonoid content with and without light, respectively. In contrast, both the GbDFR expression levels and the anthocyanin content were inhibited by the salicylic acid treatment regardless of the light conditions. This research validated the functions of GbDFRs and expands our understanding of the environmental stress-response mechanisms of ginkgo trees.

Exposure to benzaldehyde and larval feeding experiences modulate olfactory and oviposition behaviors in female Grapholita molesta (Lepidoptera: Tortricidae)

Yi Wang,Wei-Na Kong,Rui-Wei Shen,Hang Chen,Yu-Song Zhao,Jie Li,Rui-Yan Ma

BACKGROUND Grapholita molesta is an important pest of rosaceous fruit trees. Female oviposition affects larval infestation levels in orchards. This behavior, shaped by larval feeding experiences and female olfactory responses, points to an experience-modulated semiochemical strategy based on host plant secondary metabolites. To elucidate the combined effects of non-volatile and volatile components on female oviposition, we analyzed the volatile profiles of peach, pear, and apple shoots/fruits under intact, mechanically injured, and larval damage states throughout the growing season, along with female oviposition preferences, and compared olfactory responses (electroantennogram (EAG) and wind-tunnel experiments) and oviposition bioassays with benzaldehyde (amygdalin hydrolysate) between females with two larval amygdalin-feeding experiences. RESULTS Female oviposition preference and benzaldehyde emissions, both modulated by host factors, exhibited synchronous fluctuations. In EAG and wind-tunnel experiments, females not fed amygdalin during larval stages exhibited a strong attraction to benzaldehyde at concentrations of 10 and 0.1 μg/μL; however, females from larvae fed amygdalin showed a weaker long-distance orientation but greater responses to benzaldehyde during close-range approach and landing behaviors. Females fed amygdalin during larval stages were more likely to be responsive to lower concentrations of benzaldehyde (0.01, 0.1, and 1.0 μg/μL) for oviposition. In contrast, close-range attraction and oviposition behaviors were stronger for females that developed from amygdalin-fed larvae than those that were not fed amygdalin. CONCLUSION Dietary exposure to amygdalin during the larval stages increased the olfactory responses of females to benzaldehyde. Female oviposition behavior is co-regulated by larval experience derived from amygdalin ingestion and benzaldehyde as a host semiochemical signal. © 2025 Society of Chemical Industry.

N, B co-doped porous carbon materials with high heteroatom content derived from Pleurotus eryngii for high-performance symmetrical supercapacitors

Jinfeng Zheng,Hao Wu,Xiaoyang Cheng,Baopeng Ding,Jianwen Hu,Jingwei Li,Qingqing Hu,Haojie Gao,Zhipeng Zhao

N and B co-doped carbon materials were prepared using urea and H 3 BO 3 as activators and Pleurotus eryngii as the carbon source. Structural characterization shows that the heteroatom content of PENB (co-modified by urea and H 3 BO 3 ) is as high as 62 %, which is much higher than that of carbon materials modified by a single activator. The specific surface area of PENB is 2.2 times that of PEB (modified only by H 3 BO 3 ) and 127.4 times that of PEN (modified only by urea). The high specific surface area of PENB can provide more active sites. The excellent structure of PENB can be attributed to the fact that urea and H 3 BO 3 retain heteroatoms in the reaction system through neutralization reaction, which promotes the contact between heteroatoms and biomass, thereby enhancing the etching effect of heteroatoms on carbon materials. In addition, N and B atoms can induce amorphous carbon materials to produce graphene microcrystals, which can enhance the electronic conductivity of PENB. Interestingly, this method of increasing the content of heteroatoms and generating graphene is also applicable to other biomass. Electrochemical tests show that PENB exhibits the highest electrochemical performance. The symmetrical supercapacitor (SSC) assembled with PENB has an energy density of 8.8 Wh kg −1 at a power density of 400 W kg −1 . After 10,000 cycles, the capacitance retention rate is 100 %. This work provides a new universal strategy to improve the heteroatom content of carbon materials and the preparation of graphene, which can well cope with the energy crisis.

Ginkgolide B alleviates glucolipid metabolism disorders and adipose tissue inflammation by inhibiting thromboxane A2 synthesis

Yufeng Dai,Jinxiang Chen,Jialong Fang,Shuxiao Liang,Hao Zhang,Haitao Li,Wei Chen

Thromboxane A 2 (TXA 2 ) is traditionally known for its role in inducing platelet aggregation, but our previous study has discovered its crucial involvement in hepatic glucolipid metabolism and adipose tissue inflammation. Ginkgolide B (GB), a naturally occurring terpene lactone, possesses various pharmacological functions through its antagonism of the platelet activating factor (PAF) receptor. However, the metabolic regulation mechanism link between the anti-obesity efficiency of GB and TXA 2 has remained elusive. Therefore, this research aimed to explore the potential of GB in protecting against obesity by inhibiting TXA 2 synthesis. The results showed that GB inhibited TXA 2 biosynthesis which shares a similar action with eicosapentaenoic acid (EPA) and aspirin. GB treatment ameliorated hyperglycemia by decreasing gluconeogenesis. Additionally, GB supplementation relieved hepatic lipid deposition and dyslipidemia. Furthermore, GB intervention promoted macrophage M2 polarization, thus mitigating adipose tissue inflammation. In summary, our study provided evidence that GB alleviated glucolipid metabolism disorders and adipose tissue inflammation through the inhibition of the TXA 2 pathway. These findings shed light on the mechanism underlying the anti-obesity efficacy of GB and lay foundations for the future use of GB as a functional dietary supplement to alleviate high-fat diet-induced obesity.

Effect of Bacillus natto solid-state fermentation on the functional constituents and properties of Ginkgo seeds

Na Guo,Yi-Wei Jiang,Xin-Rui Song,Yan-Yan Li,Zhi-Ming Liu,Yu-Jie Fu

In the present investigation, fibrinolytic Ginkgo seeds were produced by solid-state fermentation (SSF) with Bacillus natto strains, and some parameters of the fermentation processes were investigated. Under optimal fermentation conditions, the fibrinolytic activity of Ginkgo seeds reached 3,682 ± 43 IU/g with the fermentation parameters of relative humidity 80%, initial water content 73%, fermentation temperature 38°C, inoculation volume 18%, and fermentation time 38 hr, respectively. The fermented Ginkgo seeds possessed a superior potential for the production of Nattokinase. What's more, the fermented Ginkgo seeds possessed higher total flavonoid and lower ginkgolic acids contents, which could enhance bioactivity and guarantee food safety. Sensory evaluations indicated that Ginkgo seeds produced by SSF could also be consumed as a kind of popular food. Practical applications Fermented food is popular in countries. It can not only improve the sensory properties of the products, reduce undesirable constituents, and make nutrients easily absorbable, but also improve the nutritional properties. Ginkgo biloba L is one of the oldest species that has existed on earth for more than 200 million years. However, the application of Ginkgo seeds has been limited because of the ginkgolic acids. In a previous study, immobilized Bacillus natto acted upon Ginkgo seeds to enhance the bioactivity and safety of fermented Ginkgo seeds. However, separating the fermented Ginkgo seeds from the liquid needs a large amount of energy. The solid-state fermentation of Ginkgo seeds is a good choice to produce functional Ginkgo seed products.

Excessive hydrogen sulfide-induced activation of NMDA receptors in the colon participates in anxiety- and compulsive-like behaviors in a rodent model of hemorrhagic shock and resuscitation

Rong-Xin Song,Xiao-Yi Ma,Ting-Ting Zhou,Zhi-Fang Yu,Jun Wang,Bao-Dong Li,Yu-Mo Jing,Han Wang,Yue Fu,Rui-Zhao Lv,Shi-Yan Jia,Xiao-Ming Li,Li-Min Zhang

Objective Hemorrhagic shock and resuscitation (HSR) cause inflammatory responses in the gastrointestinal tract and is associated with substantial morbidity and mortality rates. Hydrogen sulfide (H 2 S), a gasotransmitter with pleiotropic activity, exhibits anti-inflammatory benefits at physiological levels. However, deleterious effects are observed when its concentration increases. In this investigation, we employed a mouse model of HSR to examine the effects of an H 2 S scavenger on the gastrointestinal tract and brain, with emphasis on N-Methyl- d -Aspartate (NMDA) receptor function. Methods Mice were immediately administered dl -propargylglycine (PAG) intragastrically as an H 2 S scavenger after HSR exposure. The O-maze and buried beads tests were used to assess compulsive- and anxiety-like behaviors. Pathological changes in the intestine were evaluated at 24 and 30 days after HSR. Subsequently, at 30 days after HSR, we examined electrophysiological and pathological changes in the amygdala. Results Within 24 h of HSR exposure, animals treated with PAG showed significantly lower colonic injury. Additionally, compared to the HSR-treated mice 30 days after HSR, the PAG-treated mice displayed reduced buried beads, increased open-arm time, lower blood levels of Diamine Oxidase (DAO) and considerably improved ZO-1 intensity, a stronger association between the delta rhythm phase and beta activity amplitude, and lower neuroinflammatory response in the amygdala. MK-801, an NMDA receptor inhibitor, significantly reversed H 2 S-induced intestinal and cerebral injury. Conclusion This experimental data suggests that H 2 S-induced excessive activation of NMDA receptors contributes to anxiety- and compulsive-like behaviors caused by HSR.

Effect of screw pressing temperature on apricot (Prunus armeniaca L.) kernels oil quality properties and apricot kernels protein isolate functional properties

Hongyu Wu,Li Zhang,Ruiguo Cui,Chuxuan Zhang,Man Xu,Weiwei Liu,Mengshi Wang,Ruijie Liu,Long Xu,Lijun Song

This study evaluated the effects of screw pressing temperature (40 °C–200 °C) on the yields, physicochemical properties, oxidation stabilities, and antioxidant activities of apricot kernels oil (AO). Additionally, this study comparatively analyzed the functional properties of different apricot kernels protein isolates (APIs). The oil yield ranged from 30.06% to 49.07% with AO-200 showing the highest yield. Compared with cold pressing, high-temperature pressing significantly increased the trace components (such as total phenols, tocopherols, and phytosterols), oxidation stabilities, and antioxidant activities of AOs. High-temperature pressing also increased the functional properties of API. Multivariate analysis revealed that spiral pressing at 120 °C may be an optimal strategy for obtaining high-quality oil and protein isolate from apricot kernels. These findings provide a theoretical basis for the extraction and application of AO and API.

Rapid screening and evaluation of XOD inhibitors and O2•− scavenger from total flavonoids of Ginkgo biloba leaves by LC–MS and multimode microplate reader

Jing Wang,Yong Zhang,Meiqi Zhou,Meizhu Zheng,Jing Cui,Zhiqiang Liu,Chunming Liu,Shu Liu

Superoxide anion radical scavenger and xanthine oxidase inhibitor play an important role in the treatment of several relevant human diseases. In the present study, ultrafiltration liquid chromatography–mass spectrometry coupled to microplate reader was applied to screen and identify superoxide anion radical scavengers and xanthine oxidase inhibitors from total flavonoids of Ginkgo biloba leaves. As a result, four compounds (quercetin, apigenin, kaempferol and isorhamnetin) were screened as xanthine oxidase inhibitors by ultrafiltration LC–MS, and the 50% scavenging concentration values of the screened flavonoids were lower than those for allopurinol. Lineweaver–Burk plot results indicated that kaempferol was a competitive xanthine oxidase inhibitor; the other flavonoids were all anticompetitive inhibitors. Four flavonoids—rutin, quercetin, kaempferol and isorhamnetin—were screened as superoxide anion radical scavengers by LC–MS. The results demonstrate that the method for screening and evaluation of superoxide anion radical scavenger and xanthine oxidase inhibitor from a complex mixture system is feasible and efficient.

Astrocyte-mediated regulation of BLAWFS1 neurons alleviates risk-assessment deficits in DISC1-N mice

Xinyi Zhou,Qian Xiao,Yaohui Liu,Shuai Chen,Xirong Xu,Zhigang Zhang,Yuchuan Hong,Jie Shao,Yuewen Chen,Yu Chen,Liping Wang,Fan Yang,Jie Tu

Assessing and responding to threats is vital in everyday life. Unfortunately, many mental illnesses involve impaired risk assessment, affecting patients, families, and society. The brain processes behind these behaviors are not well understood. We developed a transgenic mouse model (disrupted-in-schizophrenia 1 [DISC1]-N) with a disrupted avoidance response in risky settings. Our study utilized single-nucleus RNA sequencing and path-clamp coupling with real-time RT-PCR to uncover a previously undescribed group of glutamatergic neurons in the basolateral amygdala (BLA) marked by Wolfram syndrome 1 (WFS1) expression, whose activity is modulated by adjacent astrocytes. These neurons in DISC1-N mice exhibited diminished firing ability and impaired communication with the astrocytes. Remarkably, optogenetic activation of these astrocytes reinstated neuronal excitability via D-serine acting on BLA WFS1 neurons’ NMDA receptors, leading to improved risk-assessment behavior in the DISC1-N mice. Our findings point to BLA astrocytes as a promising target for treating risk-assessment dysfunctions in mental disorders.

Ginkgo leaf derived amphiphilic and biocompatible carbon dots for sterilization and MRSA biofilm destruction

Peili Li,Gan Zhao,Long Meng,Jumen Wei,Junhua Chen,Yunhe Xu,Da Zhang,Xiang Ke,Zirong Li

Multidrug-resistant (MDR) bacterial infections, typified by methicillin-resistant Staphylococcus aureus (MRSA), have become a severe global challenge to healthcare systems. In response, antibacterial carbon dots (CDs) have attracted growing interest in the fight against MRSA. In this study, nitrogen self-doped ginkgo leaf (GL) carbon dots (GLCDs) were synthesized through a facile one-step solvothermal method combined with extraction technology. The prepared GLCDs are spherical nanoparticles with a crystalline structure and carry a positive charge of + 7.62 mV. The minimum inhibitory concentration (MIC) of GLCDs against MRSA was found to be 250 μg/mL. Notably, at a concentration of 750 μg/mL, GLCDs could completely eliminate 1 × 10 6 MRSA within 240 minutes and effectively suppress MRSA biofilm formation in a dose-dependent manner. The antimicrobial mechanisms involve electrostatic interaction enabling penetration into MRSA, disruption of the cell structure leading to intracellular component leakage, induction of oxidative stress, and inhibition of antioxidant enzymes and respiratory function. Moreover, GLCDs demonstrated excellent biocompatibility in hemolysis and in vitro cytotoxicity tests. This research presents a novel natural-plant-based antimicrobial nanomaterial, providing valuable insights into the rational utilization of natural biomass to combat MRSA infections.
