Enhancement of Colorimetric pH-Sensitive Film Incorporating Amomum tsao-ko Essential Oil as Antibacterial for Mantis Shrimp Spoilage Tracking and Fresh-Keeping

Yunxia He,Yuay Yuan,Yuanyuan Gao,Mianhong Chen,Yingying Li,Ying Zou,Liangkun Liao,Xiaotong Li,Zhuo Wang,Jihua Li,Wei Zhou

Anthocyanin-based smart packaging has been widely used for food freshness monitoring, but it cannot meet the requirements of smart films with antibacterial properties. This study aimed to enhance the antibacterial properties of intelligent films by incorporatingAmomum tsao-koessential oil (AEO) for mantis shrimp spoilage tracking and keeping the product fresh. A smart film was designed by introducing AEO and purple potato anthocyanin (PPA) to a polyvinyl alcohol/cellulose nanocrystal (PVA/CNC) polymer matrix. Our findings revealed that APP and AEO imparted the smart film with a favorable oxygen barrier, UV protection, mechanical properties, and antioxidant and pH/NH3-sensitive functions. Interestingly, the PVA/CNC-AEO-PPA film achieved 45.41% and 48.25% bactericidal efficacy againstS. putrefaciensandV. parahaemolyticus, respectively. Furthermore, a visual observation confirmed that the target film (PVA/CNC-AEO-PPA) changed color significantly during mantis shrimp spoilage: rose red—light red—pink—light gray—dark gray. Meanwhile, the PVA/CNC-AEO-PPA film retarded the quality deterioration of the mantis shrimp effectively. The PVA/CNC-AEO-PPA film shows great application potential in mantis shrimp preservation and freshness monitoring; it is expected to become a rapid sensor for detecting seafood quality non-destructively and a multifunctional film for better preservation of product quality.

Cadmium-induced oxidative stress, histopathology, and transcriptomic changes in the hepatopancreas of Fenneropenaeus merguiensis

Zhimin Ju,Charles Brighton Ndandala,Yiguo Lei,Vicent Michael Shija,Junliang Luo,Panmei Wang,Chongqing Wen,Huafang Liang

Cadmium (Cd) is a threatening heavy metal pollutant to aquatic organisms. The hepatopancreas plays a crucial role in combating environmental stress and enhancing the immune defense of aquatic species. The exact mechanism behind the damage induced by Cadmium in the hepatopancreas of Fenneropenaeus merguiensis is unclear. Thus, this study aimed to elucidate the potential mechanisms of Cadmium's impact on the hepatopancreas of F. merguiensis through physiological, histological, and transcriptomic analyses. After 72 h of F. merguiensis exposure to the 0.5 mg/L concentration of cadmium results in the reduction of superoxide dismutase and catalase activities, coupled with an increase in hydrogen peroxide and malondialdehyde levels, which leads to damage in both the gills and hepatopancreas. This causes severe histological changes in the gills and hepatopancreas of F. merguiensis , characterized by irregularities in the gill filaments and ruptures in the basal membrane of the hepatopancreas. Comparative transcriptomic analysis of F. merguiensis revealed the upregulation of several genes associated with pathways related to metabolism (ko01100), antigen processing and presentation (ko04612), and cell apoptosis (ko04215), indicating adaptive response mechanisms to Cadmium exposure. Transcriptomic analysis also showed that Cadmium exposure induced the expression of genes related to detoxification and apoptosis, such as heat shock proteins, metallothionein, and Caspase-3 , suggesting adverse effects on the normal physiological functions of F. merguiensis . Our study provides comprehensive insights into the toxicological mechanisms and impacts of Cadmium on F. merguiensis.

Biocompatible zein-gum Arabic nanoparticles for astaxanthin encapsulation and topical delivery: Preparation and characterization

Wenpeng Song,Wei Wei,Biao Zhou,Ning Lv,Heng Qin,Fei Huan,Xiaoling Zhang

Astaxanthin (AST), a potent antioxidant, holds great promise for skin health and cosmetic applications but is limited by poor water solubility and stability. This study presents the development of zein nanoparticles modified with gum arabic (GA) for the encapsulation of AST, serving as a topical nanocarrier for AST to enhance its dermal bioavailability and topical delivery. The optimized zein-GA-AST nanoparticles (Z-GA-AST) exhibited a spherical nanostructure with a mean particle size of 199.03 ± 4.63 nm (PDI <0.2) and achieved a high encapsulation efficiency of 90.34 ± 1.74 %. FT-IR spectroscopy analysis confirmed the successful encapsulation of AST within the nanoparticles. The AST-loaded nanoparticles exhibited significantly superior antioxidant activity compared to free AST. At an equivalent astaxanthin concentration of 10 μg/mL, the ABTS and DPPH radical scavenging efficiencies of the nanoparticles were at least 153 % and 128 % higher than those of free AST, respectively. The incorporation of GA enhanced the physicochemical stability and astaxanthin retention of the nanoparticles under various conditions, including different salt ion concentrations, temperatures, and storage durations, by facilitating the formation of a more compact nanostructure. The skin permeation behavior of the nanoparticles was evaluated using Franz diffusion cells and porcine skin. The results demonstrated that Z-GA2-AST1 exhibited significantly higher skin retention (48.37 %) and lower astaxanthin permeation (10.20 %) compared to free astaxanthin. This study provides valuable insights into the preparation and characterization of biocompatible nanoparticles for AST encapsulation, could become a potential formulation for topical delivery applications in cosmetics and topical drug delivery systems.

Sphingopyxis sp. YF1 and metabolite astaxanthin alleviate MC-LR-induced kidney injury

Yafang Li,Jia Wei,Xiaofang Liang,Jiangping Wang,Sisi Yan,Lemei Zhu,Zhen Ding,Chunhua Zhan,Xiaoya Ren,Fei Yang

Microcystin-LR (MC-LR) is a widely distributed cyanotoxin that causes kidney injury. Our previous work identified Sphingopyxis sp. YF1 as a novel bacterial strain capable of efficiently degrading MC-LR, but its protective role and mechanisms against MC-LR-induced kidney injury remain unexplored. In this study, the bioactive metabolite of strain YF1 was identified by mass spectrometry. The beneficial effect of strain YF1 and its bioactive metabolite against MC-LR-induced kidney injury was validated in vivo and in vitro . We found strain YF1 effectively ameliorated MC-LR induced kidney disruption and functional impairment. Mechanistically, strain YF1 significantly improved oxidative stress (SOD, CAT, GSH, and MDA) and inhibited pro-inflammatory factors expression (including IL-6 , IL-18 , IL-1β , and TNF-α ) induced by MC-LR. Notably, strain YF1 not only reduce MC-LR level in mice but also produce a bioactive metabolite astaxanthin. Consistently, both in vivo and in vitro experiments demonstrated that astaxanthin alleviated MC-LR-induced kidney injury by modulating antioxidant pathways and inflammatory cascades. This study suggested that strain YF1 could alleviate MC-LR-induced kidney injury by removing MC-LR and generating astaxanthin that mediated antioxidant and anti-inflammatory effects. These findings highlighted microbial-based antidotes for simultaneously eliminating MC-LR and mitigating its harmful effects.

Fabrication and characterization of chitosan/anthocyanin intelligent packaging film fortified by cellulose nanocrystal for shrimp preservation and visual freshness monitoring

Dan Zheng,Shumin Cao,Dengming Li,Yihan Wu,Peijun Duan,Shanshan Liu,Xing Li,Xiaoyu Zhang,Yisheng Chen

In this study, a multi-functional packaging film was fabricated, utilizing the natural polysaccharide chitosan (CS) as the base material, integrating natural blueberry anthocyanin (AN) as pH-responsive indicator, and reinforced with cellulose nanocrystals (CNCs). The implications of addition levels of CNCs on the characteristics of the films were systematically investigated, resulting in that CS-AN-CNCs 9 % film exhibited optimal performance. Specifically, the film showed a substantial enhancement in maximum tensile strength from 15 MPa to 35 MPa; On the other hand, the swelling degree properties, the oxygen permeability and water vapor permeability decreased from 159.2 % to 92.0 %, from 51.7 g/(m 2 d) to 12.2 g/(m 2 d), from 31.6 × 10 −12  g/(m·s·Pa) to 1.6 × 10 −12  g/(m·s·Pa), respectively. Moreover, the CS-AN-CNCs 9 % film exhibited antioxidant, antibacterial, coupled with a color metrically responsive to pH variations, displaying great potential in indicating the shrimp freshness and delaying spoilage. Another notable advantage of the-prepared packaging material lies in its completely biodegradability, therefore meeting the requirement of environmental protection. Therefore, the prepared CS-AN-CNCs film as an intelligent packaging solution with potential applications in food preservation and freshness monitoring applications.

Hybrid chiral nanocellulose-cyanidin composite with pH and humidity response for visual inspection and real-time tracking of shrimp quality and freshness

Yingxuan Feng,Jian Lin,Xinjie Chen,Hao Chai,Linping Tian,Jiayi Zhang,Qingfeng Sun,Yushan Yang,Yingying Li

Biobased multi-stimulation materials have received considerable attention for intelligent packaging and anti-counterfeiting applications. Cellulose nanocrystals (CNCs) and cyanidins are good material candidates for monitoring food freshness as they are eco-friendly natural substances. This work incorporated cyanidin with a CNC-hosting substrate to develop a simple, environment-friendly colorimetric device to visualize food freshness. Across the pH range of 2–13, the indicator exhibited noticeable color changes ranging from red to gray and eventually to orange. The CNC-cyanidin (CC) film exhibited a dramatic color change from blue to dark red and high sensitivity at a relative humidity of 30 %–100 %. In corresponding to the total volatile elemental nitrogen (TVB-N) level of shrimp, the indicator showed distinguishable colors at different stages of shrimp. The findings imply that the samples have substantial potential for use as an intelligent indicator for tracking shrimp freshness.

Characterization of astaxanthin-loaded Pickering emulsions stabilized by conjugates of pea protein isolate and dextran with different molecular weights

Zhi-Wei Guo,Heng-Juan Li,Ning Peng,Ying-Qiu Li,Yan Liang,Yu-Ru Zhao,Cai-Yue Wang,Zi-Yue Wang,Chenying Wang,Xidong Ren

BACKGROUND Pea protein isolate (PPI) is gaining increasing popularity in the food industry. It provides a diverse range of health benefits, such as hypoallergenic and gluten-free characteristics. However, the functional performance of PPI is hindered by its low solubility and poor stability. Therefore, in this article, PPI and dextran (DX) of different molecular weights were grafted to investigate the effects of grafting DX with different molecular weights on the interface properties and antioxidant properties of PPI. Additionally, the stability and digestive properties of the glycated PPI nanoemulsion system were explored. RESULTS The result showed that the grafting degree of PPI–DX conjugates (PPI–DC) decreased with an increase in the molecular weight of DX. Surface hydrophobicity, antioxidant activity and solubility of PPI–DC were significantly improved after grafting compared with PPI and PPI–DX mixtures (PPI–DM). Astaxanthin-loaded emulsions stabilized by grafted conjugates had smaller droplets and higher astaxanthin encapsulation rate compared to PPI emulsions. In vitro digestion demonstrated that the bioavailability of PPI–DC emulsions was higher than of PPI emulsion. Furthermore, after 24 days of storage, retention rate of astaxanthin-loaded emulsions prepared by conjugates remained above 70%, surpassing that of PPI emulsion. CONCLUSION These results indicated that DX grafting can improve the emulsion properties of PPI. In addition, the DX with a molecular weight of 5 kDa showed the most significant improvement. This study contributes to the advancement of natural emulsifiers by modifying PPI through glycation, and furnishes a valuable reference for its utilization in functional foods. © 2024 Society of Chemical Industry.

Mechanisms of breakdown of Haematococcus pluvialis cell wall by ionic liquids, hydrochloric acid and multi-enzyme treatment

Zhang Ye,Xing-He Tan,Zhi-Wei Liu,Rana Muhammad Aadil,Yi-Cheng Tan,Muhammad Inam-ur-Raheem

Summary The robust cell wall structure of Haematococcus pluvialis ( H. pluvialis ) consists of polysaccharides and tough non-hydrolysable sporopollenins, which makes it difficult to extract superpotent antioxidant, astaxanthin from these cells. Therefore, breakdown of cell wall is a key step in the overall process of astaxanthin recovery. In this study, the mechanism of three well-established chemical techniques for cell disruption of H. pluvialis cysts [ionic liquids (IL), hydrochloric acid (HCl) and multiple enzymes (multi-enzyme, ME)] on deconstruction of the cyst cell wall of H. pluvialis was explored and characterised by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), nuclear magnetic resonance (NMR) and gas chromatography–mass spectrometry (GC-MS) analyses. The results demonstrated that the three cell wall breakdown techniques exhibited high extraction efficacies for the recovery of astaxanthin from H. pluvialis [IL (86.71 ± 2.06%), HCl (80.52 ± 2.28%) and ME (71.08 ± 2.49%)]. However, their performances on disrupting the trilayered cell walls of H. pluvialis were significantly different, which were confirmed by distinct morphologies of the treated cell walls visualised by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Meanwhile, the results of FTIR confirmed that, to some extent, cellulose, hemicellulose and lignin in the cell walls were hydrolysed by HCl, IL and ME treatments. However, ME exhibited a less hydrolytic effect on lignin than HCl and IL. Moreover, XRD and NMR analyses implied that the amorphous region of cell wall was susceptible to hydrolysis/breakdown by the three techniques.

Genome mining of astaxanthin biosynthetic genes from Sphingomonas sp. ATCC 55669 for heterologous overproduction in Escherichia coli

Tian Ma,Yuanjie Zhou,Xiaowei Li,Fayin Zhu,Yongbo Cheng,Yi Liu,Zixin Deng,Tiangang Liu

As a highly valued keto-carotenoid, astaxanthin is widely used in nutritional supplements and pharmaceuticals. Therefore, the demand for biosynthetic astaxanthin and improved efficiency of astaxanthin biosynthesis has driven the investigation of metabolic engineering of native astaxanthin producers and heterologous hosts. However, microbial resources for astaxanthin are limited. In this study, we found that the α-Proteobacterium Sphingomonas sp. ATCC 55669 could produce astaxanthin naturally. We used whole-genome sequencing to identify the astaxanthin biosynthetic pathway using a combined PacBio-Illumina approach. The putative astaxanthin biosynthetic pathway in Sphingomonas sp. ATCC 55669 was predicted. For further confirmation, a high-efficiency targeted engineering carotenoid synthesis platform was constructed in E. coli for identifying the functional roles of candidate genes. All genes involved in astaxanthin biosynthesis showed discrete distributions on the chromosome. Moreover, the overexpression of exogenous E. coli idi in Sphingomonas sp. ATCC 55669 increased astaxanthin production by 5.4-fold. This study described a new astaxanthin producer and provided more biosynthesis components for bioengineering of astaxanthin in the future. Graphical Limited microbial resources for astaxanthin limit its widespread applications in nutritional supplements and pharmaceuticals. Sphingomonas sp. ATCC 55669 can produce astaxanthin naturally. Whole-genome sequencing with gene functional confirmation identifies the discrete astaxanthin biosynthetic genes, and overexpressed related genes in E. coli can increase astaxanthin production. The new astaxanthin producer and biosynthesis components provide more possibilities for bioengineering of astaxanthin in the future.

Boosting one-step degradation of shrimp shell waste to produce chitin oligosaccharides at smart nanoscale enzyme reactor with liquid-solid system

Ziteng Wang,Yijin Cai,Mingxuan Li,Xiaoru Wan,Li Mi,Wenge Yang,Yonghong Hu

Chitin oligosaccharides (CTOS) possess potential applications in food, medicine, and agriculture. However, lower mass transfer and catalytic efficiency are the main kinetic limitations for the production of CTOS from shrimp shell waste (SSW) and crystalline chitin. Chemical or physical methods are usually used for pretreatment to improve chitinase hydrolysis efficiency, but this is not eco-friendly and cost-effective. To address this challenge, a chitinase nanoreactor with the liquid-solid system ( Bc ChiA1@ZIF-8) was manufactured to boost the one-step degradation of SSW and crystalline chitin. Compared with free enzyme, the catalytic efficiency of Bc ChiA1@ZIF-8 on colloidal chitin was significantly improved to 142 %. SSW and crystalline chitin can be directly degraded by Bc ChiA1@ZIF-8 without any pretreatments. The yield of N , N′ -diacetylchitobiose [(GlcNAc) 2 ] from SSW and N -acetyl- D -glucosamine (GlcNAc) from crystalline chitin was 2 times and 3.1 times than that of free enzyme, respectively. The reason was that Bc ChiA1@ZIF-8 with a liquid-solid system enlarged the interface area, increased the collision frequency between enzyme and substrate, and improved the large-substrates binding activity of chitinase. Moreover, the biphasic system exhibited excellent stability, and the design showed universal applicability. This strategy provided novel guidance for other polysaccharide biosynthesis and the conversion of environmental waste into carbohydrates.

Bai Liufu,Qiyao Su,Kunhao Hong,Jie Wei,Yakun Wang,Zhiqiang Han,Lingyun Yu

Simple Summary17α-methyltestosterone (MT) is known to suppress ovaries and induce spermatogenesis; yet, its effects in crustaceans remain underexplored. This study investigates the impact of dietary MT on gonad development and intestinal microbiota in juvenileMacrobrachium rosenbergiithrough sex ratio determination, histological observation, the expression of reproductive development-related genes, and an analysis of the intestinal microbiota structure. The results showed that an. optimal MT concentration and treatment duration promoted gonadal differentiation and growth. However, excessive MT concentrations inhibited male gonad development and slowed growth rates. MT supplementation significantly increased microbial abundance and altered the intestinal microbial community structure. These findings clarify the effects of adding MT to feed on the reproductive development and intestinal microbiota of juvenileM. rosenbergii, providing a reference for MT application in crustaceans.17α-methyltestosterone (MT) is known for its ability to suppress ovaries and induce spermatogenesis; yet, its effects in crustaceans are underexplored. This study investigates the impact of varying MT concentrations on the gonadal development and intestinal microbiota of juvenileMacrobrachium rosenbergii. Feeds containing different MT doses were provided, and the sex ratios, histological observations, reproductive gene expression, and intestinal microbial composition were analyzed. The results revealed short-term feeding (60 days) of 1000 mg/kg MT resulted in the highest male ratio, while long-term feeding (150 days) of 500 mg/kg MT achieved the same outcome. Conversely, long-term feeding of 1500 mg/kg MT led to the lowest male ratio and retarded male germ cell development. An intestinal microbiota analysis showed that MT supplementation significantly increased microbial abundance and altered the intestinal microbial community structure. Additionally, MT suppressed the expression of female reproductive-related genes. This study provides insights into the effects of MT on reproductive development and gut microbiota in juvenile prawns, offering a valuable reference for the application of MT in crustacean aquaculture.

Sensory characteristics and key odor-active compounds of oxidized Antarctic krill oil: A comparative study with oxidized fish oil

Gui-Mei Dong,Rui-Ying Wang,Fa-Wen Yin,De-Yang Li,Lei Qin,Da-Yong Zhou

Oxidized Antarctic krill oil (AKO) has unique sensory characteristics that have not been fully elucidated. This study was aimed to explore the sensory characteristics and key odor-active compounds of oxidized AKO by comparing with oxidized fish oil (FO) through accelerating storage experiments. The sensory evaluation results exhibited that oxidized AKO had a distinct ammonia odor and oxidized FO had an obvious fishy odor. GC–MS and GC-O analysis identified 21 and 28 odor-active compounds in AKO and FO samples, respectively, and the correlation analysis showed a good correlation between sensory attributes and odor-active compounds. Trimethylamine (27.73 mg/kg) was the key odor-active compound with an ammonia odor in AKO, and not found in FO. Although AKO samples also contains odor-active compounds such as 1-penten-3-ol (4.93 mg/kg) and 1-penten-3-one (1.12 mg/kg) contributing to fishy odor in FO samples, the masking effect between the odor compounds prevented obvious fishy odor from being sensed.
