Nowadays, label-free atomic spectrometric bioassays are attracting great research interest because of their advantages of low cost, simple design and operation, etc. Herein, a novel and simple chemical vapor generation-atomic fluorescence spectrometry (CVG-AFS)/inductively coupled plasma-mass spectrometry (ICP-MS) label-free detection method is presented for highly sensitive and selective assay of DNA and proteins. This work mainly combined a phenomenon that CdTe quantum dots (QDs) can be used to selectively differentiate free Hg2+ and the T–Hg2+–T complex, with the use of simple membrane filtration separation to improve the performance of the label-free bioassay methods. Upon hybridization with the DNA/protein (carcinoembryonic antigen, CEA) target, the T–Hg2+–T hairpin structure was opened and Hg2+ was released; this initiated the cation exchange reaction between Hg2+ and CdTe QDs which released Cd2+ simultaneously. Subsequently, the free Cd2+ was separated by the filtration membrane without separating the CdTe QDs, which could then be separated from the sample matrices for the CVG-AFS/ICP-MS assay. Under the optimal conditions, this method possessed high sensitivity for DNA and CEA determination with limits of detection (LODs) of 0.2 nM and 0.2 ng mL−1, and linear dynamic ranges of 1–160 nM and 0.5–20 ng mL−1, respectively, and exhibited excellent DNA sequence specificity and protein selectivity. This method preserves the advantages of the label-free atomic spectrometric bioassay, and combined with the selective cation exchange reaction and simple filtration separation to improve the performance.

We report on a giant magnetoresistance (GMR) based immunosensor for the carcinoembryonic antigen (CEA) labeled with superparamagnetic microparticles (Dynabeads). The GMR sensor contains 200 GMR strips and was fabricated by micro-electromechanical system (MEMS) technology. This system can detect Dynabeads in concentrations down to 10 ng⋅mL −1 . Sandwich immunoassays were employed on the surface of a gold film modified with a self-assembled monolayer and using biotinylated secondary antibody against CEA and streptavidinylated Dynabeads. With DC magnetic fields in the range of 40 to 90 Oe, CEA can be detected with a detection limit as low as 10 pg⋅mL −1 . Samples spiked with different concentrations of CEA can be distinguished clearly, and the method is deemed to be well suited for clinical use.
Make use of antigen-antibody immune reaction, Dynabeads that conjugated with polyclonal CEA antibodies were captured onto the surface of Au film which immobilize of monoclonal CEA antibodies. Under the dc magnetic field magnetizing, the superparamagnetism Dynabeads induced a weak magnetic field that can be detected by the nearby GMR sensor, which reveals the presence of CEA. GMR: giant magneto-resistance CEA: carcinoembryonic antigens.

A photothermal immunoassay using a thermometer as readout based on polyaniline@Au organic-inorganic nanohybrids was built. Temperature output is acquired due to the photothermal effect of the photothermal nanomaterial. Polyaniline@Au organic-inorganic nanohybrids were synthesized by interfacial reactions with high photothermal conversion efficiency. A sandwich structure of the immunocomplex was prepared on a microplate for determination of carcinoembryonic antigen (CEA) by polyaniline@Au organic-inorganic nanohybrids as nanolabel. The released heat based on light-to-heat conversion from the photothermal nanolabel under NIR irradiation is detectable using the thermometer. The increased temperature is directly proportional to CEA concentration. The linear range of the photothermal immunoassay is 0.20 to 25 ng mL −1 with determination limit of 0.17 ng mL −1 . Graphical abstract
Polyaniline@Au organic-inorganic nanohybrids with high photothermal conversion efficiency was synthesized as labels to construct photothermal immunosensor. The sandwich-type immunoassay was built on 96 hole plate based on specific binding of antigen and antibody. Carcinoembryonic antigen in sample was detected quantitatively by thermometer readout.

A highly sensitive and selective photoelectrochemical (PEC) aptasensor was constructed for carcinoembryonic antigen (CEA) detection based on ZnO flower-rods (ZnO FRs) modified with g-C 3 N 4 -Au nanoparticle (AuNP) nanohybrids. The nanohybrids of g-C 3 N 4 -AuNPs can improve the visible light absorbance of ZnO FRs and enhance the PEC property. We designed a sandwichlike structure formed with DNA hybridization of NH 2 -probe1, CEA aptamer, and CuS-NH 2 -probe2 to detect CEA. The p -type semiconductor CuS nanocrystals (NCs) at the terminational part of sandwichlike structure work as electron traps to capture photogenerated electrons and consequently lead to a magnified photocurrent change. The results indicate that the photocurrent is increased when CEA antigen (Ag) is introduced. Since the sandwichlike structure is destroyed, CuS NCs are restricted to capture photogenerated electron. The PEC aptasensor for CEA determination is ranged from 0.01 ng·mL −1 to 2.5 ng·mL −1 with a detection of 1.9 pg·mL −1 . The proposed aptasensor exhibits satisfactory PEC performances with rapid detection, great sensitivity and specificity. Specially, this PEC aptasensor shows a reliable result for the determination of CEA in invalid human serum compared with the ELISA method. The designed aptasensor may provide a new idea for a versatile PEC platform to determine various molecules.

In this work, a synergetic fluorescence enhancement strategy based on micro/nanostructure optimization of ZnO nanorod arrays and in situ ZIF-8 coating was proposed for the highly sensitive microfluidic detection of carcinoembryonic antigen (CEA). The glass capillary was chosen as the microfluidic channel, and controllable construction of ZnO nanorod arrays on the inner wall of the microchannel was conducted via the intermittent reaction method. Additionally, the application of the optimal ZnO nanorod array to glass capillary-based microfluidic fluorescence detection was demonstrated. The fluorescence enhancement characteristic of the zeolitic imidazolate framework-8 (ZIF-8) towards organic fluorescence labels were investigated and successfully applied to protein marker detection. After formation of sandwich immunoassay on the ZnO/PAA nanorod arrays, the in situ ZIF-8 coating via a moderate chemical method could significantly improve the fluorescence intensity of fluorescent labels via direct contact. In CEA detection, the limit of detection (LOD) reached as low as 0.01 pg mL −1 , and the dynamic range was 0.01 pg mL −1 to 100 pg mL −1 . Hence, the microfluidic fluorescence detection strategy based on the ZnO nanorod arrays/PAA layers/ZIF-8 coating with a synergetic fluorescence enhancement effect provides a superior, promising, and highly sensitive approach for the detection of protein markers, including CEA.

A novel electrochemical immunosensor for sensitive detection of a tumor marker is reported. This protocol involves the surface-initiated atom transfer radical polymerization (SI-ATRP) of poly(ethylene glycol) monomethacrylate (PEGMA) on an indium tin oxide (ITO) electrode. Using carcinoembryonic antigen (CEA) as a model analyte, capture anti-CEA (Ab1) was conjugated to poly(poly(ethylene glycol) monomethacrylate) (P(PEGMA)) brushes, via activation of hydroxyl groups by succinic anhydride and N-(3-dimethylaminopropyl)-N′-ethylcarbodiimide hydrochloride, and N-hydroxysulfosuccinimide sodium salt (NHS/EDC). After the sandwich immunoreaction, the horseradish peroxidase-labeled signal anti-CEA antibodies were used to conjugate gold nanoparticles (HRP-Ab2–AuNPs bioconjugates). Under optimized conditions, the proposed immunosensor showed a high sensitivity and a linear range from 5 × 10−2 to 20 ng mL−1 with a low detection limit of 1.09 × 10−3 ng mL−1. The assay results of clinical serum samples were in acceptable agreement with the reference values. The designed immunoassay system with ultrahigh sensitivity and being efficient and economical, might have potentially broad applications in protein diagnostics and bioassay.

Carcinoembryonic antigen (CEA) is usually utilised as a reliable diagnostic and predictive marker of gastric cancer (GC). Herein, a target-responsive, label-free aptamer ‘signal-on’ fluorescent biosensing system was developed based on the synthesis of DNA-templated quantum dots via one-pot route and interactions with graphene oxide. CEA triggered aptamer-DNA hairpins hybridisation to release DNA to hybridise with DNA-CdTe QDs on GO and led to the fluorescence recovery of system with a ‘signal-on’ status. The fluorescence intensity was proportionate to CEA concentration, and highly sensitive sensing platform for detecting CEA was achieved with a wide linear range (5–250 ng/mL) and a low detection limit (1.17 ng/mL). This sensing strategy, the targeted detection of aptamer for CEA, provided a new direction in the detection of protein and construction of fluorescence biosensors.

We designed an ingenious plasmonic core-shell-satellite nanostructure (HPGNPs@mSiO 2 -AuNR) including a hollow porous gold nanoshell (HPGNP) core, a protective silica shell with mesoporous (mSiO 2 ) interlayer, and a high level of gold nanoring (AuNR) satellite coverage by in situ etching. The HPGNP core modified with 4-mercaptobenzoic acid (4-MBA) possessed relatively strong local-field enhancement. The mSiO 2 coating acted as a continuous interior nanogap and protective shell, and the variable external satellites in the morphology had tunable optical responses and designed “hotspot” regions. Specifically, AuNR satellites exhibited superior plasmonic properties, intense core-satellite plasmon coupling with the HPGNP core, and favorable biocompatibility. Meanwhile, the nanoassemblies had larger surface-to-volume ratios for molecular modification and were penetrable for analyte. With their abundant built-in hotspots and less hindrance for analyte penetration into the hollow interior, the labeled nanoassemblies were successfully used in SERS/fluorescence sensing system for carcinoembryonic antigen (CEA) detection. These immunoprobes with high SERS activity could be used in SERS-based immunoassay for CEA detection with a linear response ranging from 1 pg/mL to 1000 pg/mL. CEA detection in the fluorescence sensing system showed that the fluorescence quenching efficiency gradually decreased as the CEA level increased, with a linear response of 0.5–400 pg/mL. This SERS/fluorescence-based immunoassay had the same CEA detection limit of 0.1 pg/mL. The fine-engineered penetrable nanoassemblies with Au ring satellites can provide an insight into the rational and optimal design of multifunctional nanostructures. They can also broaden the field of vision for the design of multifunctional sensing probes in biochemical detection.

A facile MALDI-TOF mass spectrometric platform for quantitative analysis of protein biomarkers was developed based on magnetic ZnFe 2 O 4 nanoparticles and mass tagging signal amplification. In this platform, magnetic ZnFe 2 O 4 nanoparticles functionalized with an aptamer of the biomarker of interest was used to magnetically separate silica nanoparticles modified with another aptamer of the target biomarker and a barcoding peptide from solution phase in the presence of the biomarker of interest. After the silica nanoparticles were dissolved by KHF 2 , the released barcoding peptide was detected by MALDI-TOF mass spectrometry with magnetic ZnFe 2 O 4 nanoparticles used as assisting matrix of laser desorption ionization. Since the mass spectral intensity of the barcoding peptide is directly related to the concentration of the target biomarker, the proposed platform can be applied to the quantification of the target biomarker in complex biological samples. The effectiveness of the proposed platform was tested on the detection of carcinoembryonic antigen (CEA) in serum. Experimental results revealed that the proposed platform could achieve quite reliable quantitative results for CEA in human serum samples with accuracy comparable to a commercial CEA ELISA Kit. Its limit of detection and limit of quantification for CEA were estimated to be 0.6 × 10 −3 and 1.8 × 10 −3 ng/mL, respectively, considerably lower than the corresponding values reported in literature. Due to its features of simplicity in design, extremely low background signal, high sensitivity and selectivity , the proposed method can be further developed to be a competitive alternative for the quantification of CEA and other protein biomarkers as well.

DNA aptamers against carcinoembryonic antigen (CEA) have been identified through the systematic evolution of ligands by exponential enrichment (SELEX) technique, but their affinity needs to be improved. In this study, an in silico approach was firstly used to screen the mutation sequences of a reported DNA aptamer (the parent aptamer, denoted as P) against CEA. The affinities of several high-score DNA mutants were determined by the biolayer interferometry technique. Finally, the newly obtained aptamers were verified in an aptasensor application. For the in silico approach, Mfold and RNA Composer were combined to generate the 3D RNA structures of the DNA mutants. The RNA structures were then modified to 3D DNA structures with the Write program. The docking model and binding ability of the 3D DNA structures with CEA were simulated and predicted with the ZDOCK program. Two mutation sequences (P-ATG and GAC-P) exhibited significantly higher ZDOCK scores than P. The dissociation constant of P-ATG and GAC-P to CEA was determined to be 4.62 and 3.93 nM respectively, obviously superior to that of P (6.95 nM). The detection limit of the P-ATG and GAC-P based aptasensors was 1.5 and 1.2 ng mL−1, respectively, markedly better than that based on P (3.4 ng mL−1). The consistency between the in silico and the experimental results indicates that the developed in silico post-SELEX screening approach is feasible for improving DNA aptamers. The P-ATG and GAC-P aptamers found in this study could be used for future CEA aptasensor design and fabrication, promisingly applicable for highly sensitive CEA detection and early cancer diagnosis.

Tumor markers are usually over-expressed in human body fluids during the development of cancers. Monitoring tumor markers' level is thus important for early diagnosis and screening of cancers. One way to achieve this is based on the surface enhanced Raman scattering (SERS) technique that can drastically amplify Raman signals of analytes on a plasmonic metal (e.g., Au, Ag, and Cu) surface. However, this promising method suffers from aggregation of plasmonic nanoparticles. Here we report a stable, reproducible, and facile SERS-based readout method to detect an important tumor marker, carcinoembryonic antigen (CEA). This route utilizes Au butterfly wings with natural three dimensional (3D) hierarchical sub-micrometer structures rather than relying on the aggregates of metal nanoparticles. The Au butterfly wings show excellent SERS property and are temperature (80 °C) and time (6 months) stable on a sub-micrometer scale. Thus, the detecting antibodies and enzyme-linked secondary antibodies that are usually applied in conventional enzyme-linked immunosorbent assay (ELISA) can be replaced by chemically synthesized CEA aptamers, significantly simplifying the whole detection process. We demonstrate the feasibility of this method via quantitative detection of clinical CEA level in human body fluids. This work thus demonstrates a promising tumor marker detection technique based on a hierarchical sub-micrometer SERS structure, which could be useful for the mass screening of early stage cancers.

A flexible free-standing electrochemical biosensor to detect carcinoembryonic antigen (CEA) is described based on a conducting polypyrrole (PPy) nanocomposite film electrode. The conducting PPy composite was constructed by the sandwiched structure formed by PPy doped with pentaerythritol ethoxylate (PEE) and 2-naphthalene sulfonate (2-NS-PPy) separately via electropolymerization. Gold nanoparticles (AuNPs) were fixed on the PPy composite film by electrodeposition and then connected to CEA aptamer through self-assembly to construct a free-standing electrochemical biosensor breaking away from additional soft substrates and current collector. This PPy composite film-based electrochemical biosensor exhibits satisfying sensing performance for CEA detection, with a linear range from 10 −10 g/mL to 10 −6 g/mL and a detection limit of 0.033 ng/mL, good specificity and long-term sensing stability (96.8% of the original signal after 15 days). The biosensor also presents acceptable reproducibility with 1.7% relative standard deviation. Moreover, this electrochemical biosensor owns the deformation stability that could bear various deformations (twisting, folding, and knotting) without affecting device’s sensing performance. It can even maintain 99.4% of the original signal under 25% strain deformation. Due to the superior sensing performance, high stability (mechanical deformation and long-term storage), and flexibility, this free-standing electrochemical biosensor proves huge potential in application of flexible and wearable electronics.
