Digital microfluidics (DMF) shows a great application prospect in droplet manipulation. However, the fouling of the hydrophobic surfaces caused by biomolecules limits its development. In this study, we report a new strategy to enhance the functionality and anti-biofouling performance of the DMF chip by using a hydrophobic liquid surface (HLS) rather than a regular hydrophobic solid surface (HSS). The DMF chip with such a configuration can efficiently drive various liquids with full-function operations. Moreover, our DMF chips can directly manipulate biomolecular droplets without restrictive conditions like adding surfactants or filling with silicon oil. The liquid-liquid contact between the droplet and the hydrophobic surface ensures that the non-specifically adsorbed biomolecules move along with the droplet. Thus, no residue is left behind to ruin the hydrophobicity of the hydrophobic surface. Meanwhile, the long-term reversibility of contact angle change and stability of droplet movement demonstrate the excellent ability against biofouling. In addition, high- and low-temperature tests also show the temperature stability of the HLS. Finally, a biochemical application, plasmid extraction of Escherichia coli ( E.coli ) cells, is successfully carried out on the DMF chip with HLS as a proof of its usability. This HLS is expected to offer versatile functionalities and anti-biofouling performance for DMF chips in handling biomolecular droplets.

The effect of hydrodynamics on crystallization kinetics in confined flow systems (e.g., biological tubules) remains unclear. Herein, we developed a home-made microfluidic chip platform mimicking the curved, confined flow microenvironment of human renal tubules to investigate the growth kinetics and inhibition efficacy of calcium oxalate monohydrate (COM) crystals under controlled flow rates and constant supersaturation. Time-resolved imaging quantified COM growth rates at varied flow rates, identifying a transition from diffusion-controlled to reaction-controlled growth at 6 mL/h. COM growth is predominantly mediated by the screw-dislocation mechanism but shifts to 2D nucleation within the narrow supersaturation range of 1.8–2.1. The inhibition efficacy of polyphenols is regulated by the coupled effect of flow rate and concentration: decreasing flow rate either weakens or enhances inhibition depending on inhibitor concentration, with transition concentration thresholds varying among inhibitors and correlating with their crystal surface interaction strength. Simulations of pathological conditions showed reduced [Ca 2+ ]:[C 2 O 4 2− ] ratios diminish inhibitory efficacy via competitive adsorption between inhibitors and oxalate ions. However, ellagic acid outperforms citric and gallic acids, retaining over 95% inhibitory efficiency in high-calcium environments. This study highlights the key role of hydrodynamics and provides a theoretical basis for developing targeted, environment-adaptive strategies to prevent kidney stone formation.

Microfluidic lab-on-a-chip technology has shown great potential in various fields such as bioscience, medical diagnostics, and environmental monitoring. However, its widespread adoption has been hindered by challenges in functional integration, operational autonomy, and manufacturing scalability. To address these limitations, we present a 3D-printed self-sensing magnetically actuated microfluidic (SMAM) chip designed for autonomous bioanalysis. This innovative device utilizes stereolithography apparatus (SLA) 3D printing to rapidly prototype and integrate microchannel networks alongside with a magnetically driven functional module. The chip employs magnetic actuation for precise, wireless manipulation of fluids within the microchannels, eliminating the need for bulky external pumps. Additionally, the system features an integrated self-sensing mechanism, enabling flow monitoring and on-chip analyte detection. The SMAM chip demonstrates exceptional dual-function performance, achieving a high pumping flow rate of up to 972 μL min−1 and a good piezoresistive sensitivity of 43.1 MPa−1. We first demonstrate its system-level utility by assembling the chip into a modular, wirelessly monitored microfluidic platform with an integrated flow rectifier. Furthermore, its potential for therapeutic interventions is validated through a proof-of-concept of an untethered device for magnetically guided, on-demand drug release. This work provides a novel approach for developing intelligent analytical devices, promising to enable new paradigms in automated biological research and diagnostics.

Phosphor-converted light emitting diodes (pc-LEDs) are essential for current lighting and display areas because of their unique properties of being uncomplicated, durable and economical. In previous studies, Pr 3+ doped red-emitting phosphors have attracted enormous research interest due to their high color purity. However, their photoluminescence excitation (PLE) spectra are always hard to be combined with LED chips. This mismatch is a great obstacle to achieve Pr 3+ phosphors utilizing in LED devices. Here, to circumvent this issue, Pr 3+ -doped Ca 3 MgTiGe 3 O 12 (CMTG) red-emitting phosphor was optimized with PLE band shifting from 330 nm to 360 nm after tuning with Bi 3+ co-doping based on the structure and phosphorescence properties. The chip-matched excitation makes it a good candidate for white LEDs (wLEDs) fabrication to improve the output quality of device. Meanwhile, the high color purity of 93.02 % was obtained, which is conducive to widening the color gamut of backlight display applications. The photoluminescence (PL) spectra, fluorescence lifetime, and X-ray photoelectron spectroscopy (XPS) analysis were employed and discussed in detail to explain the role of Bi 3+ played in the system. Finally, wLEDs were fabricated with CMTG:Pr 3+ , Bi 3+ , showing a high color rendering index (Ra) of 94.4. And the color gamut covers ∼80 % of the National Television System Committee (NTSC) standard, suggesting great potential for applications of wLEDs and backlight display.

A facile approach to fabricating a surface-enhanced Raman scattering (SERS)-active chip by integrating silver dendrites with copper substrate through a one-step process was explored. The structures of dendrites were synthesized and controlled by an AgNO3/PVP aqueous system, and the fabrication parameters amount of AgNO3/PVP and reaction time were systematically investigated. The optimized silver dendrites, closely aggregated on the surface of the copper chip, exhibited high SERS activity for detecting rhodamine 6G at a concentration as low as 3.2 × 10–11 M. Meanwhile, the prepared SERS-active chip displayed a high thermal stability and good reproducibility. Moreover, the potential application for analysis of polycyclic aromatic hydrocarbons was demonstrated by detection of fluoranthene at a low concentration of 4.5 × 10–10 M. This SERS-active chip prepared by the convenient and high-yield method would be a promising means for rapid detection under field conditions.

Multicellular hepatic spheroids that recapitulate biomimetic cellular assembly and enhanced cellular interaction have become useful 3D culture models for liver tissue engineering and hepatotoxic drug screening. Despite progress, current strategies for generating heterogeneous spheroids with multiple cell types mostly depend on cell random aggregation, lacking a well-defined architecture that mimics natural liver tissue structure. Here, a microarray chip with a triangular chamber arrangement is developed to the fabrication of heterogeneous spheroids comprising hepatocytes, stellate, and endothelial cells in a multilayer organization. The triple cell co-culture spheroids formed by in sequence seeding above three types of cells, yielding structures with a hepatic plate-like core orderly coated by stellate and endothelial shells. These structured spheroids exhibit an enhanced structural integrity and liver functions such as albumin, MRP2, and the CYP3A4 and CYP1A2 enzyme expression levels compared to randomly distributed spheroids. Screening of hepatotoxic drugs by the hepatic spheroid microarray show increased sensitivity compared to 2D cultures. This approach represents a significant advancement toward control over cell spatial distribution in spheroids, and offers a valuable structured spheroid model with multilayer cellular organization for regenerative medicine and drug hepatotoxic evaluation.

In infections with high bacterial loads, such as peritonitis, antibiotic efficacy is often compromised by the inoculum effect. Antimicrobial peptides, despite their broad-spectrum activity, are similarly affected. To elucidate the mechanisms underlying the inoculum effect of antimicrobial peptides, this study employed microfluidic chips to isolate and analyze single bacterial cells. Comparison of population-level minimum inhibitory concentration obtained from 96-well assays with single-cell minimum inhibitory concentration revealed that minimum inhibitory concentration increased with inoculum density, whereas single-cell minimum inhibitory concentration remained unchanged. Further investigation revealed that intracellular components released from lysed cells and OmpT-mediated peptide degradation substantially elevated population resistance, effects that were absent under single-cell isolation. These results demonstrate that the inoculum effect of antimicrobial peptides stems from population-dependent interactions, intracellular material release, and enzymatic degradation rather than intrinsic single-cell resistance. This work provides mechanistic insight into the inoculum effect of antimicrobial peptides and informs the optimization of their clinical application.

The photocatalytic degradation for environmentally hazardous substances has been widely explored with the use of various photocatalysts and techniques. It still remains a challenge to achieve efficient degradation and removal of residual antibiotics in the water environment. Here, we report an improved photocatalytic activity chip for degradation of antibiotic tetracycline hydrochloride by perpendicular ZnS-coated ZnO nanorod arrays (ZnO@ZnS NAs). The enhanced photocatalytic activity contributed to the polycrystalline ZnS shell’s effective inhibition of the recombination of photogenerated electron–hole pairs. Meanwhile, vertically aligned ZnO@ZnS nanorod arrays can increase the light harvesting ability by enhancing scattering of light among ZnO@ZnS NAs. On the basis of these findings, an improved photocatalytic activity ZnO@ZnS NA chip has been fabricated by growth of ZnO NAs on a piece of silicon wafer and further sulfurization. More importantly, ZnO@ZnS NA chips have been utilized to construct a ladderlike device, purifying antibiotics wastewater in one step, to effectively degrade tetracycline hydrochloride with enhanced photocatalytic efficiency in flowing contaminated water. In addition, the ZnO@ZnS NA device exhibited excellent recyclability in multiple repeated cycles. The ZnO@ZnS NA chip provided a convenient and fast strategy for removal of antibiotics and pharmaceutical residues in wastewater.

Sandwich immunoassay plays a significant role in disease diagnostics and prognosis. However, this technology always requires cumbersome manual washing procedures and well trained operators. To address the issues, we made an effort to develop a self-powered microfluidic chip (SP-Chip) to accomplish multiple washing processes and antigen-antibody binding reaction automatically in one-step sliding manner. This smart microfluidic platform integrated with fluorescence microscopic counting technique for rapid and quantitative detection of human chorionic gonadotropin (hCG) and carcinoembryonic antigen (CEA) in 10 min. Moreover, the versatile strategy is not only applied to various biomarkeres detection, but also suitable for designing other sample-in-answer-out diagnostic system. Taken together, the autonomous, accurate, inexpensive, self-contained, and easy to use chip lays a solid foundation for quantitative detection of biomarkers in the fields of disease treatment and personalized diagnostics.

Conventional microchannels provide effective chip cooling with constant flow rate and this comes at the cost of high pumping power consumption. Considering the dynamic heat load in reality, a self-adaptive microchannel cooling is desired for regulating the flow rate to diminish the operational cost while preserving the inherent cooling effectiveness. Taking the advantages of easy synthesis, well tunable lower critical solution temperature (LCST) and remarkable mechanical strength, this paper reports the self-adaptive microchannel cooling with the thermal-sensitive nanocomposite (NC) P(MEO 2 MA- co -OEGMA) hydrogel. A novel route is developed to integrate the hydrogel with the chip using template-aided imprint molding and in situ polymerization. Circular hydrogel is elaborately designed and anchored firmly in the channel. The cooling performance is tested with a custom test vehicle under different operating conditions. The hydrogel acts as a smart valve and tailors the flow rate from 0.14 mL/s to 0.46 mL/s based on heat load variation. Compared with conventional channels, heat flux of 100 W/cm 2 can be extracted with similar temperature rise while the coefficient of performance (COP) is improved by an order of magnitude. The transient characteristics and repeatability are also validated. The self-adaptive scheme offers both effective and economical liquid cooling for modern electronic devices.

A nanoliter self-priming compartmentalization (SPC) microfluidic chip suited for the digital polymerase chain reaction (dPCR) analysis in point-of-care testing (POCT) has been developed. This dPCR chip is fabricated of polydimethylsiloxane (PDMS). After the dPCR chip is evacuated, there will be a negative pressure environment in the chip because of the gas solubility of PDMS. The negative pressure environment can provide a self-priming power so that the sample solutions can be sucked into each reaction chamber sequentially. The whole sampling process requires no external power and is valve-free. Channels that contain water are designed around each sample panel to prevent the solvent (water) from evaporating during dPCR process. A glass coverslip is also used as a waterproof layer, which is more convenient and more efficient than other waterproof methods seen in literature. This dPCR chip allows three samples to be amplified at the same time. Each sample is distributed into 1040 reaction chambers, and each chamber is only 2.08 nL. Human β-actin DNA solutions of known concentrations are used as the templates for the dPCR analyses to verify the sensitivity and accuracy of the method. Template DNA solutions diluted to concentrations of 300, 100 and 10 copies/μL are tested and shown that this simple, portable and self-priming dPCR chip can be used at any clinic as a real POCT technique.

A “sandwich” microfluidic surface enhanced Raman scattering (SERS) chip with Ag film@nanoAu prepared in a microchannel was proposed and fabricated in situ. The integrated composite Ag film@nanoAu was prepared in situ by way of chemical plating and self-assembly. Firstly, the preparation conditions were optimized. On the basis of that, the influence of the laser focus position along the vertical direction and the materials of the cover plates on the SERS effect were investigated and optimized in detail. The optimized SERS effect was observed when the laser was focused on the interface of the sample and the SERS substrate. The SERS intensity of Rhodamine 6G (R6G) decreased 2.5 times and 2.1 times when PDMS or glass was used as the cover plate separately. Compared to single gold and silver SERS substrates, the composite of Ag film@nanoAu integrated in the microfluidic channel resulted in the peak intensity increasing 3–5 times during the SERS detection of R6G at 1507 cm−1. The enhancing effect could be attributed to the change in the local electromagnetic field which was caused by the synergistic sensitizing effect between the Au nanoparticles and the underlying silver mirror film. It was illustrated that the detection limit for R6G was 10−8 M and the enhancement factor (EF) was 3.8 × 105 with a relative standard deviation (RSD) of approximately 10% in the as-prepared microfluidic SERS chip.
