A bacterial strain ( Gordonia sp. Lff) capable of efficiently degrading di-(2-ethylhexyl) phthalate (DEHP) was isolated from river sludge. The optimal pH and temperature for the degradation of DEHP by Lff were 7.0 and 35 °C, respectively. Lff could degrade high concentrations of DEHP (100–2000 mg/L) with a degradation efficiency of over 91.43%. The DEHP degradation curves fit well with first-order kinetics, with a half-life ranging from 0.598 to 0.746 d. Substrate inhibition analyses showed that the maximum specific degradation rate, half-saturation constant and inhibition constant were 0.8 d −1 , 45.8 mg/L and 462.18 mg/L, respectively. A detailed biodegradation pathway of DEHP was proposed based on GC–MS analysis. Furthermore, Lff could also efficiently degrade DEHP in soils. DEHP or DEHP plus Lff changed the bacterial community in soils, and Lff accelerated the shaping of the bacterial community. To the best of our knowledge, this study is the first to perform a detailed investigation into the biodegradation of DEHP in soil by Gordonia sp. and its effect on the soil bacterial community. These results suggest that Lff is an ideal candidate for the bioremediation of DEHP-contaminated environments.

Di (2-ethyl) hexyl phthalate (DEHP) is a common environmental endocrine disruptor that induces oxidative stress, posing a significant threat to human and animal health. Oxidative stress can activate the PTEN/PI3K/AKT pathway, which is closely related to cell apoptosis. However, it is unclear whether DEHP induces apoptosis of chicken liver cells by regulating the PTEN/PI3K/AKT pathway through oxidative stress. In this experiment, male laying hens were continuously exposed to 400 mg/kg, 800 mg/kg, and 1600 mg/kg DEHP for 14 d, 28 d, and 42 d. The results showed that liver injury was aggravated with the dose of DEHP gavage, and the ROS/MDA levels in L, M, and H DEHP exposure groups were significantly increased, while the T-AOC/T-SOD/GSH-PX levels were decreased. Meanwhile, DEHP exposure up-regulated the mRNA and protein expression levels of PTEN/Bax/Caspase-9/Caspase-3 and down-regulated the mRNA and protein expression levels of PI3K/AKT/BCL-2, indicating that DEHP may lead to hepatocyte apoptosis through ROS regulation of PTEN/PI3K/AKT axis. In order to further clarify the relationship between oxidative stress and liver injury, we treated chicken hepatocellular carcinoma cell line (LMH) with 2.5 mM N -acetylcysteine (NAC). NAC attenuated these phenomena. In summary, our study suggests that DEHP can induce apoptosis of chicken liver through ROS activation of the PTEN/PI3K/AKT axis.

Male infertility is an attracting growing concern owing to decline in sperm quality of men worldwide. Phthalates , in particular to di (2-ethylhexyl) phthalate (DEHP) or its main metabolite mono-2-ethylhexyl phthalate (MEHP), affect male reproductive development and function, which mainly accounts for reduction in male fertility. Lycopene (LYC) is a natural antioxidant agent that has been recognized as a possible therapeutic option for treating male infertility. Testosterone (T)/androgen receptor (AR) signaling pathway is involved in maintaining spermatogenesis and male fertility. How DEHP causes spermatogenesis disturbance and whether LYC could prevent DEHP-induced male reproductive toxicity have remained unclear. Using in vivo and vitro approaches, we demonstrated that DEHP caused T biosynthesis reduction in Leydig cell and secretory function disorder in Sertoli cell, and thereby resulted in spermatogenic impairment. Results also showed that MEHP caused mitochondrial damage and oxidative damage, which imposes a serious threat to the progress of spermatogenesis. However, LYC supplement reversed these changes. Mechanistically, DEHP contributed to male infertility via perturbing T/AR signaling pathway during spermatogenesis. Overall, our study reveals critical role for T/AR signal transduction in male fertility and provides promising insights into the protective role of LYC in phthalate-induced male reproductive disorders.

Di (2-ethylhexyl) phthalate (DEHP) is a persistent pollutant that harms the environment and human health. Nanomaterial modified biochar (NMBs) significantly enhances the removal efficiency of organic pollutants due to its unique properties and structures. The aim of this research is to clarify the mechanisms of DEHP biodegradation in black soil over a 60 day incubation period, especially regarding the role of NMBs (nano-hydroxyapatite modified (NHBC), nano-zero valent iron modified (NZFBC)), including examining the involved metabolic pathways, the functional microorganisms facilitating the process, the functional genes responsible and the synergies between them. Results indicated that the impact of biochar on DEHP is multilayer adsorption and chemisorption. Among the treatments, NHBC exhibited the optimal degradation performance in black soil, it increased DEHP degradation efficiency from 17.79% (unamended original soil) to 91.12% after 60 days. NMBs addition notably raised the soil pH, soil organic matters (SOM), humus (HU), fulvic acid (FA), humic acid (HA) and enzyme activity. We identified three major metabolic pathways of DEHP enhanced by NHBC treatment ((phthalic acid (PA) to protocatechuic acid (PCA) to tricarboxylic acid (TCA) cycle), (Benzoic acid (BA) to cis-1,2-dihudroxycyclohexa-3,5-diene-1-carboxylate to TCA cycle) and (BA to Benzoate-CoA to TCA cycle). Additionally, 22 bacterial species and 55 functional genes involved in DEHP degradation under the action of NMBs, and identified 10 potential DEHP-degrading bacterias and 20 high-priority genes. The results provide a scientific basis for DEHP adsorption and biodegradation.

Bis(2-ethylhexyl) phthalate (DEHP) is not only a widely used plasticizer but also a common endocrine disruptor that frequently lingers in water, posing a threat to the health of aquatic organisms . Quercetin (Que) is a common flavonol found in the plant kingdom known for its antioxidant, anti-inflammatory, and immunomodulatory effects. However, it is still unclear whether DEHP can cause pyroptosis and affect the expression of cytokines of grass carp L8824 cells and whether Que has antagonistic effect in this process. In our study, grass carp L8824 cells were treated into four groups after 24 h, namely NC group, DEHP group (1000 μM DEHP), Que group (5 μM Que), and DEHP + Que group (1000 μM DEHP + 5 μM Que). Our results indicate a significant increase in the level of ROS in L8824 cells after exposure to DEHP. DEHP upregulated oxidative stress markers (H 2 O 2 and MDA) and downregulated antioxidant markers (CAT, GSH, SOD , and T-AOC). DEHP also upregulated MAPK and NF-κB signal pathway-related proteins and mRNA expressions (p-p38, p-JNK, p-EPK, and p65). As for cell pyroptosis and its related pathways, DEHP upregulated pyroptosis-related protein and mRNA expressions (GSDMD, IL-1β, NLRP3, Caspase-1, LDH , pro-IL-18, IL-18, and ASC). Finally, DEHP can up-regulated cytokines (IL-6 and TNF-α) expression, down-regulated cytokines (IL-2 and IFN-γ) expression, and antimicrobial peptides (β-defensin, LEAP2, and HEPC). The co-treatment of L8824 cells with DEHP and Que inhibited the activation of the ROS/MAPK/NF-κB axis, alleviated pyroptosis, and restored expression of immune-related indicators. Finally, NAC was applied to reverse intervention of oxidative stress. In summary, Que inhibited DEHP-induced pyroptosis and the influence on cytokine and antimicrobial peptide expression in L8824 cells by regulating the ROS/MAPK/NF-κB pathway. Our results demonstrate the threat to fish health from DEHP exposure and confirmed the harm of DEHP to the aquatic ecological environment and the detoxification effect of Que to DEHP, which provides a theoretical basis for environmental toxicology.

The global rate of human male infertility is rising at an alarming rate owing to environmental and lifestyle changes. Phthalates are the most hazardous chemical additives in plastics and have an apparently negative impact on the function of male reproductive system. Ferroptosis is a recently described form of iron-dependent cell death and has been linked to several diseases. Transferrin receptor (TfRC), a specific ferroptosis marker, is a universal iron importer for all cells using extracellular transferrin. We aim to investigate the potential involvement of ferroptosis during male reproductive toxicity, and provide means for drawing conclusions on the effect of ferroptosis in phthalates-induced male reproductive disease. In this study, we found that di (2-ethylhexyl) phthalate (DEHP) triggered blood-testis barrier (BTB) dysfunction in the mouse testicular tissues. DEHP also induced mitochondrial morphological changes and lipid peroxidation, which are manifestations of ferroptosis. As the primary metabolite of DEHP, mono-2-ethylhexyl phthalate (MEHP) induced ferroptosis by inhibiting glutathione defense network and increasing lipid peroxidation. TfRC knockdown blocked MEHP-induced ferroptosis by decreasing mitochondrial and intracellular levels of Fe 2+ . Our findings indicate that TfRC can regulate Sertoli cell ferroptosis and therefore is a novel therapeutic molecule for reproductive disorders in male patients with infertility.

Soil microbial fuel cells (MFCs), a novel ecosystem technology, have recently been intensively studied for antibiotic-polluted soils. However, actual agricultural soils are always contaminated by mixed pollutants, especially plasticizers from extensively used agricultural plastic films. The aim of this study is to investigate the effect of di-2-ethylhexyl phthalate (DEHP, a representative plasticizer in soil) on the removal of sulfadiazine (SDZ, a frequently detected antibiotic in natural environments), antibiotic resistance genes (ARGs), and the microbial community in soil MFCs. Our results confirmed that soil MFCs maintained a good antibiotic removal ability even under the influence of residual DEHP and achieved a higher removal performance at higher DEHP concentrations due to enhanced power generation. Specifically, a higher DEHP concentration had a favorable effect on antibiotic removal in soil MFCs because the biodegradation efficiency was improved in both the upper ( i.e., to 0.268 ± 0.021 mg kg −1 from 4.867 ± 0.221 mg kg −1 ) and lower layers ( i.e. , to 0.293 ± 0.047 mg kg −1 from 4.867 ± 0.221 mg kg −1 ) of polluted soils. Moreover, a high DEHP concentration significantly promoted the abundance of bacteria associated with electricity generation compared with a lower DEHP concentration, resulting in the promotion of extracellular electron transfer, and enhancing SDZ degradation. The increased ARG abundance may have been caused by the enrichment of ARG potential hosts brought about by high DEHP concentrations, likely due to the increased conjugative transfer rates of the RP4 plasmid due to decreased membrane permeability and an increased reactive oxygen species content. The results revealed the ecological risk of residual DEHP in soil that promotes ARG transmission in soil MFCs, although it has the potential to reduce SDZ toxicity through horizontal gene transfer. We also highlight concerns regarding the management of antibiotics and plasticizers in soil. The negative effects of plasticizers on antibiotic removal should be carefully evaluated when using soil MFCs for the in situ remediation of antibiotic-contaminated soil.

As the most produced phthalate, di-(2-ethylhexyl) phthalate (DEHP) is a widely environmental pollutant primarily used as a plasticizer, which cause the harmful effects on human health. However, the impact of DEHP on spleen and its underlying mechanisms are still unclear. Pyroptosis is a novel form of cell death induced by activating NOD-like receptor family pyrin domain containing 3 (NLRP3) inflammasomes and implicated in pathogenesis of numerous inflammatory diseases. The current study aimed to explore the impact of DEHP on immune inflammatory response in mouse spleen. In this study, the male ICR mice were treated with DEHP (200 mg/kg) for 28 days. Here, DEHP exposure caused abnormal pathohistological and ultrastructural changes, accompanied by inflammatory cells infiltration in mouse spleen. DEHP exposure arouse heat shock response that involves increase of heat shock proteins 60 (HSP60) expression. DEHP also elevated the expressions of toll-like receptor 4 (TLR4) and myeloid differentiation protein 88 (MyD88) proteins, as well as the activation of NF-κB pathway. Moreover, DEHP promoted NLRP3 inflammasome activation and triggered NLRP3 inflammasome-induced pyroptosis. Mechanistically, DEHP drives splenic inflammatory response via activating HSP60/TLR4/NLRP3 signaling axis-dependent pyroptosis. Our findings reveal that targeting HSP60-mediated TLR4/NLRP3 signaling axis may be a promising strategy for inflammatory diseases treatment.

Di-(2-ethylhexyl) phthalate (DEHP) is a common environmental endocrine disrupting chemical that may induce male reproductive disorders. Exposure to DEHP at a prepubertal stage could lead to prepubertal testicular injury, but the underlying mechanisms remain unclear. In this study, we exposed Sprague-Dawley rats to 0, 250, and 500 mg DEHP per kg body weight per day at the prepuberty stage from postnatal day 22 (PND 22) to PND 35 by oral gavage. Testicular injury and oxidative stress were evaluated, and the levels of 6-methyladenosine (m6A) modification and expression of modulator genes for RNA methylation were measured in testes . Furthermore, m6A modification of the important antioxidant transcription factor Nrf2 was analyzed using methylated RNA immunoprecipitation qPCR. Our results show that DEHP worsened testicular histology, decreased testosterone concentrations, downregulated expression of spermatogenesis inducers, enhanced oxidative stress, inhibited the Nrf2-mediated antioxidant pathway, and increased apoptosis in testes. Additionally, DEHP increased global levels of m6A RNA modification and altered the expression of two important RNA methylation modulator genes, FTO and YTHDC2 . Moreover, m6A modification of Nrf2 mRNA increased upon DEHP exposure. Overall, these findings link oxidative stress imbalance with epigenetic effects of DEHP toxicity and provide insight into the testicular toxicity of DEHP from the new perspective of m6A modification.

Mono(2-ethylhexyl) phthalate (MEHP) is a metabolite of DEHP which is one of phthalic acid esters (PAEs) widely used in daily necessities. Moreover, MEHP has been proven to have stronger biological toxicity comparing to DEHP. In particular, several recent population-based studies have reported that intrauterine exposure to MEHP results in adverse pregnancy outcomes. To explore the mechanisms and metabolic biomarkers of MEHP exposure, we examined the metabolic status of HTR-8/Svneo cell lines exposed to different doses of MEHP (0, 1.25, 5.0, 20 μM). Global and dose–response metabolomics tools were used to identify metabolic perturbations and sensitive markers associated with MEHP. Only 22 metabolic features (accounted for <1 %) were significantly changed when exposed to 1.25 μM. However, when the exposure dose was increased to 5 or 20 μM, the number of significantly changed metabolic features exceeded 300 (approximately 10 %). In particular, amino acid metabolism, pyrimidine metabolism and glutathione metabolism were widely affected according to the enrich analysis of those significant altered metabolites, which has and have previously been reported to be closely related to fetal development. Moreover, 5′-UMP and N -acetylputrescine with the lowest effective concentrations (EC −10 = 0.1 μM and EC +10 = 0.11 μM, respectively) were identified as sensitive endogenous biomarkers of MEHP exposure.

Waste scrap tyre as microbial immobilization matrix enhanced degradation of phthalate esters (PAEs), di (2-ethylhexyl) phthalate (DEHP), dibutyl phthalate (DBP), and diethyl phthalate (DEP). The hybrid (physical adsorption and microbial immobilization) degradation process performance of scrap tyres was examined for the PAEs degradation. The scrap tyre was shown with competitive adsorption capacity toward PAEs, influenced by pH, temperature, dosage of adsorbent (scrap tyre), and concentration of PAE. The primary adsorption mechanism for tyres toward PAEs was considered hydrophobic. The immobilization of previously isolated Bacillus sp. MY156 on tyre surface significantly enhanced PAEs degradation as well as bacterial growth. The enzymatic activity results implied immobilization promoted dehydrogenase activity and decreased esterase activity. The cell surface response during PAEs degradation, in terms of morphological observation, FTIR and XRD analyses, and extracellular polymeric substance (EPS) release, was further assessed to better understand the interactions between microorganisms and tyre surface. Waste scrap tyres could be a promising potential candidate to be reused for sustainable environmental management, including contaminants removal.

The pervasive use of the plasticizer di(2-ethylhexyl) phthalate (DEHP) poses potential risks to global aquatic ecosystems. This study systematically evaluated the adverse effects of chronic exposure to environmentally relevant concentrations of DEHP on gill tissues of crucian carp, utilizing histological examination, metabolomic, and transcriptomic analysis. The results demonstrated that DEHP induced significant histopathological alterations in gill tissues, with significant enrichment observed in multiple pathways associated with amino acid, hormone, lipid, and xenobiotic metabolism. Metabonomics-transcriptomics analyses indicated that DEHP-induced significantly over-activation of cytochrome P450 1B1-like ( p < 0.001 ) and cytochrome P450 3A30-like ( p < 0.05 ) via the nuclear xenobiotic receptors pathway was a key factor contributing to the disruption of tryptophan metabolism and steroid hormone biosynthesis, as well as inducing circadian rhythm disruption. Moreover, circadian rhythm disruption further exacerbated the imbalance of cytochrome P450 (CYP450) enzyme system as well as linoleic acid, arachidonic acid, sphingolipid, and glycerophospholipid metabolism. Overall, the feedback regulation between the CYP450 enzyme system and circadian rhythms emerged as the primary mechanism underlying DEHP-induced metabolic and transcriptional disruptions, ultimately resulting in gill toxicity. This study not only enriched the toxic effects on aquatic organisms of chronic exposure to DEHP, but provided potential biomarkers for the environmental risk assessment of DEHP.
