Background Rapid industrial development has generated serious pollution, including the presence of toxic and harmful heavy metal ions. Among them, trivalent chromium ion (Cr 3+ ) is a very important element that poses a threat to life and health in our industrial wastewater pollution. Thus, it is important to develop efficient fluorescence methods for Cr 3+ detection. In this study, an upconversion luminescence biosensor for detecting Cr 3+ was constructed based on a DNAzyme, strand displacement reaction (SDR), and DNA-functionalized upconversion nanoparticles (UCNPs). Results The sulfonate-rich poly (sodium 4-styrene sulfonate) (PSS) was modified onto the surface of UCNPs, forming UCNPs@PSS. Then, NH 2 -Capture probe DNA (NH 2 -Cp) was further modified onto the UCNPs@PSS surface through sulfonylation, resulting in UCNPs@PSS@NH 2 -Cp. The DNAzyme activated by Cr 3+ triggered the release of the primer probe (Pp), which initiated the SDR system cycle, thereby releasing a tetramethylrhodamine (TAMRA)-modified signal probe (TAMRA-Sp). Finally, UCNPs@PSS@NH 2 -Cp bound to TAMRA-Sp through complementary base pairing, causing UCNPs and TAMRA to approach each other. Because of the luminescence resonance energy transfer (LRET) mechanism, the upconversion luminescence (UCL) signal of the UCNPs was quenched by TAMRA, enabling the detection of Cr 3+ by the change of I 585 /I 545 ratio. This biosensor has good stability, selectivity, and sensitivity, with a linear range of 0.5–75 nM and a detection limit of 0.135 nM for Cr 3+ . Significance and novelty Firstly, based on LRET between UCNPs and TAMRA, the quantitative analysis of Cr 3+ is achieved through the changes of ratio fluorescence. Secondly, the specificity of the biosensor is improved by utilizing the specific recognition of DNA enzymes. Thirdly, the signal amplification technology of the SDR cycle greatly improves the sensitivity of biosensor. This biosensor will be useful for future environmental safety monitoring and biopsy of biological fluids.

When using circular dichroism (CD) spectroscopy for quantitative analysis, the samples to be analyzed must be free of light-absorbing interferences. However, in real-world samples, the presence of background absorbers is practically unavoidable. The difference in the matrices between the real-world samples to be analyzed and the standard samples (on which either univariate or multivariate calibration model was built) would result in systematic errors in the quantification results of CD spectroscopy. In this contribution, a novel calibration strategy for quantitative CD spectroscopic analysis was proposed. The main idea of the proposed calibration strategy is to project the CD spectra of both the standard samples and the real-world sample to be analyzed onto a projection space orthogonal to the space spanned by the background CD spectrum of the real-world sample and then build a multivariate calibration model on the transformed CD spectra of the standard samples. The performance of the proposed calibration strategy was tested and compared with conventional univariate and multivariate calibration methods in the quantification of Pb 2+ in cosmetic samples using CD spectroscopy in combination with a G-quadruplex DNAzyme (e.g. PS2.M). Experiments results revealed that the proposed calibration strategy could mitigate the influence of the difference in the matrices between the standard samples and cosmetic samples and realized quantitative analysis of Pb 2+ in cosmetic samples, with precision and accuracy comparable to atomic absorption spectroscopy . The proposed calibration strategy has the features of simplicity and effectiveness, its combination with CD spectroscopic probes can realize accurate and precise quantification of analytes in complex samples using CD spectroscopy.

A novel signal amplification strategy based on a Pb 2+ -dependent DNAzyme is proposed for detecting Pb 2+ and radon through Pb 2+ -induced DNAzyme cleavage and the auto-assembly of a duplex-hemin/G-quadruplex with two loop-stem structures and enzyme-mimicking catalytic activity. First, the Pb 2+ -specific DNAzyme cleaves a primer sequence, which induces assembly of the hairpin probe Hp1/Hp2 into a double helix structure. Subsequently, a G-quadruplex forms after the insertion of hemin into the free G-rich sequences; this G-quadruplex possesses the catalytic activity of horseradish peroxidase and changes colorless TMB to its deeply colored oxidized state. lead-dependent DNAzymes were constantly sheared by Pb 2+ , and the free primer strands were continuously assembled into double chains by hybridization with hairpin probes, providing amplification for the detection of lead (II) and radon. Under the optimum conditions, there was a good linear relationship between ΔA and the lead concentration for Pb 2+ concentrations ranging from 2.58 to 18 nM, and the detection limit was 0.77 nM. Analysis of actual samples indicated that when the radon concentration was in the range of 5.41 × 10 3– 1.65 × 10 5 Bq·h/m 3 , the radon concentration and the absorbance were linearly correlated with a detection limit of 1.62 × 10 3 Bq·h/m 3 . During the process of radon sampling and detection, radiation damage from the radioactive gas radon can be avoided. In this study, the primer dissociated by the DNAzyme was in a free state. Compared with the fixed-state primer chain, this system will be more convenient for the biological analysis of ultratrace metal ions. Therefore, this strategy has good application prospects for biosensors and is expected to become a novel platform for the amplification and detection of metal ion signals.

In this study, an innovative electrochemical biosensor was developed for the rapid, specific, and sensitive detection of Acinetobacter baumannii without the need for sample pretreatment. The biosensor utilized an aptamer as a specific capture probe for A. baumannii and employed a self-powered DNAzyme walker cleavage cycle reaction to achieve signal amplification. Upon introduction of the target bacteria, the aptamer captured the bacteria and released the Trigger, activating the DNAzyme to cleave the substrate chain containing methylene blue (MB). This led to the release of MB-labeled DNA fragments from the electrode surface, resulting in a significant decrease in the square wave voltammetry (SWV) signal of MB on the sensing platform. The limit of detection (LOD) for A. baumannii was determined to be 30 CFU/mL, enabling discrimination of the target bacteria from other common clinical isolates. Furthermore, the biosensor’s potential for real sample analysis was demonstrated in cerebrospinal fluid (CSF), showcasing its efficacy and versatility as a biosensing tool with wide-ranging applications in disease diagnosis and bioanalysis.

Multifunctional therapeutic nanoplatforms with an activatable property have demonstrated the ability to enhance cancer therapy due to their intrinsic controllability in drug delivery. Although much effort has been focused on constructing multifunctional therapeutic nanoplatforms with an activatable property, developing logically activatable nanoplatforms to exert improved therapeutic efficiency against cancer remains an imperative need. In this study, we established a tumor microenvironment activable nanoplatform by the DNAzyme assisted coating of polyaniline (PANI) on the surface of mesoporous silica nanoparticles (MSNs). This mild synthetic strategy can avoid the destruction of the loaded biomolecules and guarantee the biosafety of the nanoplatforms. Furthermore, the activable polymer can be used to load siRNA for combined cancer treatment. When the composite nanoplatforms were enriched in the tumor, the acidic tumor microenvironment activated the nanocarrier and unloaded the drugs and siRNA upon near-infrared (NIR) irradiation to achieve controlled drug delivery for enhanced cancer therapy.

BCL-2 gene as well as its products is recognized as a promising target for the molecular targeted therapy of tumors. However, due to certain defense measures of tumor cells, the therapeutic effect based on the gene silencing of BCL-2 is greatly reduced. Here we fabricate a smart response nucleic acid therapeutic that could silence the gene effectively through a dual-targeted and cascade-enhanced strategy. In brief, nano-graphene oxide (GO), working as a nano-carrier, is loaded with a well-designed DNAzyme, which can target and silence the BCL-2 mRNA. Furthermore, upon binding with the BCL-2 mRNA, the enzymatic activity of the DNAzyme can be initiated, cutting a substrate oligonucleotide to produce an anti-nucleolin aptamer AS1411. Nucleolin, a nucleolar phosphoprotein, is known as a stabilizer of BCL-2 mRNA. Via binding and inactivating the nucleolin, AS1411 can destabilize BCL-2 mRNA. By this means of simultaneously targeting mRNA and its stabilizer in an integrated system, effective silencing of the BCL-2 gene of tumor cells is achieved at both the cellular and in vivo levels. After being dosed with this nucleic acid therapeutic and without any chemotherapeutics, apoptosis of tumor cells at the cellular level and apparent shrinkage of tumors in vivo are observed. By labeling a molecular beacon on the substrate of DNAzyme, visualization of the enzymatic activity as well as the tumor in vivo can be also achieved. Our work presents a pure bio-therapeutic strategy that has positive implications for enhancing tumor treatment and avoiding side effects of chemotherapeutics.

A DNAzyme-powered DNA walking machine was constructed to develop the fluorescence aptasensing for sensitive detection of kanamycin. The aptamer for kanamycin is partially hybridized with complementary DNA (cDNA) modified on magnetic beads (MBs). The specific interaction of target and aptamer triggered the cDNA to be free tentatively, which captured walker DNA. Then the autonomous motion of DNA walker on MBs surface was propelled via DNAzyme digestion of recognition sites. The signal probe was separated, and the amplified fluorescence signal was achieved by the accumulation of the signal probe. Kanamycin was used as a model analyte, and the developed assay achieves a detection limit of 0.00039 ng·mL −1 (S/ N = 3) within a linear detection range from 0.001 to 2000 ng·mL −1 . This aptasensing strategy can be extended for detection of other antibiotics by adapting corresponding target recognition aptamer sequence.

Cardiovascular disease is one of the main causes of death in the world, which is closely associated with dyslipidemia. Dyslipidaemia is usually manifested as a relatively higher level of low-density lipoprotein (LDL) and lower level of high-density lipoprotein (HDL). Thus, the quantitative detection of the LDL and HDL particles is of great importance to predict the risk of cardiovascular diseases. However, the traditional methods can only indirectly reflect the HDL/LDL particle concentrations by detecting the cholesterol or proteins in HDL/LDL particles and are always laborious and time-consuming. Thus, the accurate and efficient approach for the detection of intact HDL and LDL particles is still lacking so far. We developed an enzyme- and isolation-free method to measure the concentration of HDL and LDL based on DNAzyme and hybridization chain reaction (HCR)-based signal amplification. This method can be used to directly and accurately detect the concentration of “actual” HDL and LDL particles instead of the cholesterol in HDL and LDL, with limits of detection of 10 and 30 mg/dL, respectively, which also satisfied the lipoprotein analysis in clinical samples. Therefore, this HCR–DNAzyme platform has great potential in clinical applications and health management.

The detection and speciation analysis of metal-ion is very important for environmental monitoring. A novel electrochemical biosensor for Nickel(II) detection based on a DNAzyme-CdSe nanocomposite was developed. We firstly hybridized with capture probe (DNA1) and sequentially with DNA (DNA2) on the gold electrode. Then CdSe QDs were incorporated the specific recognition of DNA2 by covalent assembling. Upon addition of nickel ion into the above system, the substrate strand of the immobilized DNAzyme was catalytically cleaved by target Ni 2+ , resulting in disassociation of the shorter DNA fragments containing CdSe QDs. The remaining CdSe QDs on the electrode surface detected by differential pulse anodic stripping voltammetry (DPASV). Under optimal conditions, the as-prepared sensor exhibited high sensitivity and fast response to Ni 2+ with the linear range from 20 nM to 0.2 mM and a low detection limit of 6.67 nM. The prepared biosensor also shows good stability and good reproducibility and high selectivity toward target Ni 2+ against other metal ions because of highly specific Ni 2+ -dependent DNAzyme. Thus, our strategy has a good potential in the environment surveys.

Escherichia coli (E. coli) is a common Gram-negative bacterium that is widely distributed in nature and is susceptible to natural and random genetic factors. It is a pathogenic factor for many diseases. Sensitive and specific detection of E. coli is an important means to ensure food safety and prevent foodborne disease outbreaks. Ultrasensitive and specific detection of E. coli is still an urgent problem that needs to be solved. An intuitive and convenient colorimetric assay method with high sensitivity based on a DNAzyme-powered AuNR-DNA walker is proposed here for detection of E. coli. The strategy, dependent on the targeted activation of Mg2+-DNAzyme, starts an efficient DNA walking machine, induces the formation of countless peroxidase-mimicking enzymes, and produces a color change via a chromogenic reaction for visual detection of E. coli. This strategy features several aspects. First, this is the first time that an AuNR-based DNA walker is integrated with a colorimetric assay of E. coli. By combining the DNAzyme-powered AuNR-DNA walker and visual detection via the peroxidase-mimicking enzyme, the detection sensitivity is significantly improved compared to those of previously reported colorimetric-based methods, which provides a paradigm for the development of highly sensitive colorimetric assay methods. Second, an activity-controlled nano-DNAzyme is constructed via target-triggered formation of G-quadruplex DNAzyme to achieve low background and high signal-to-noise detection. In addition, this strategy has the advantages of simple operation, short analysis time, low cost, and easy on-site detection. Therefore, the proposed strategy has the potential for application in the detection of E. coli and related food safety analysis and clinical diagnosis.

α-synuclein oligomer is a marker of Parkinson's disease. The traditional enzyme-linked immunosorbent assay for α-synuclein oligomer detection is not conducive to large-scale application due to its time-consuming, high cost and poor stability. Recently, DNA-based biosensors have been increasingly used in the detection of disease markers due to their high sensitivity, simplicity and low cost. In this study, based on the DNAzyme-driven DNA bipedal walking method, we developed a signal-on electrochemical sensor for the detection of α-syn oligomers. Bipedal DNA walkers have a larger walking area and faster walking kinetics, providing higher amplification efficiency compared to conventional DNA walkers. The DNA walker is driven via an Mg 2+ -dependent DNAzyme, and the binding-induced DNA walker will continuously clamp the MB, resulting in the proliferation of Fc confined near the GE surface. The linear range and limit of detection were 1 fg/mL to 10 pg/mL and 0.57 fg/mL, respectively. The proposed signal-on electrochemical sensing strategy is more selective. It will play a significant role in the sensitive and precise electrochemical analysis of other proteins.

Aberrant DNA methylation, which is caused by the abnormal level of DNA methyltransferase (MTase), has been considered associated with a growing number of human diseases. Although there are various methods paying close attention to DNA methyltransferase (MTase) detection, most of them are generally complex and expensive. Here, a simple electrochemical strategy for sensitive detection of DNA methyltransferase (MTase) and inhibitor screening based on DpnI digestion triggering the formation of G-quadruplex DNAzymes has been developed. In this paper, a probe richness of guanine (G) was first self-assembled on the surface of the electrode through Au–S bond and then hybridized with the complementary DNA. Without DNA methylation, G-quadruplex DNAzymes cannot be formed due to the double helix structure and a weak electrochemical response can be observed. On the contrary, an obvious enhancement of the electrochemical response can be achieved after the cleavage of the methylated double-strand DNA by DpnI since G-quadruplex DNAzymes can be obtained, which catalyze the oxidation of hydroquinone by H 2 O 2 with the assistance of the cofactor hemin. This method is under a detection limit of 0.96 U/mL and can monitor the change of DNA methylation level selectively. Moreover, RG108 was selected as a representative inhibitor for studying the inhibition activity of DNA MTase.
