Synthesis of a GNRs@mSiO2-ICG-DOX@Se-Se-FA Nanocomposite for Controlled Chemo-/Photothermal/Photodynamic Therapy

Na An,Huiming Lin,Fengyu Qu

Graphical GNRs@mSiO 2 core–shell nanoparticles have been synthesized as host to load ICG and DOX molecules by the layer-by-layer assembly method through electrostatic interactions. A sensitive Se–Se linker was synthesized and grafted to ensure controlled release. Under NIR irradiation, the enhanced cytotoxicity to cancer cells is derived from the synergistic effect of chemotherapy, PTT, and PDT. A novel GNRs@mSiO 2 core–shell nanocomposite has been synthesized to integrate sensitive chemotherapy with photodynamic therapy (PDT) and photothermal therapy (PTT) for enhanced antitumor treatment by using gold nanorods (GNRs) as the core and mesoporous silica (mSiO 2 ) as the shell. After modifying with -NH 2 , Indocyanine Green (ICG, negative charge) and doxorubicin hydrochloride (DOX, positive charge) molecules could be stored in the mesopores by the layer-by-layer assembly method through electrostatic interactions. To inhibit leakage from the mesopores, a sensitive Se–Se linker was synthesized and adopted as a nanovalve coated on the outside of the nanoparticles (GNRs@mSiO 2 -ICG-DOX@Se-Se). Under NIR (808 nm) irradiation, heat elevation (derived from GNR/ICG) and generation of reactive oxygen species (ROS, derived from ICG) can also be detected in vitro. Owing to the redox sensitivity of Se–Se, GNRs@mSiO 2 -ICG-DOX@Se-Se showed reduction (GSH)- as well as oxidation (ROS)-triggered DOX release. To further ensure specific targeting, folic acid (FA) was grafted to the outside of the nanoparticles, because of the high expression of FA receptors on tumor cells. Detailed cell experiments performed with the nanoparticles and HepG2 as typical cancer cells showed enhanced cytotoxicity towards the cancer cells owing to the synergistic effect of chemotherapy, PTT, and PDT. On account of the different GSH concentrations in cancer and normal cells and the switch-on/off application of NIR irradiation, the GNRs@mSiO 2 -ICG-DOX@Se-Se-FA nanoparticles show potential for applications in cancer therapy.

NIR-light-responsive chemo-photothermal hydrogel system with controlled DOX release and photothermal effect for cancer therapy

Jun Yang,Zhen Sun,Qingqing Dou,Shuhan Hui,Peng Zhang,Rui Liu,Di Wang,Shan Jiang

Here a multiplex near-infrared (NIR) light-responsive hydrogel platform (SA/ [email protected] ) has been constructed for controlled synergetic chemotherapy and photothermal cancer therapy. Ag nanoparticles doped SnS 2 nanoflowers (SA), which can convert NIR light into thermal energy with efficiency of 30.0%, were embedded in low-melting-temperature agarose hydrogel as a photothermal agent. Moreover, anticancer drug doxorubicin (DOX) is loaded into the hydrogel for tumor treatment. The heat generated from NIR irradiation of SA promotes melting of hydrogel, thus the incorporated DOX can be released into the surrounding environment. By adjusting the irradiation power and time, the drug will be released with a control. In vitro study shows that the SA/ [email protected] system is highly effective to kill cancer cells (∼ 93%) at a relatively low -laser irradiation (1.2 w cm −2 , 10 min) treatment. In the mice tumor model, SA/ [email protected] with laser irradiation is capable to suppress or even eliminate the tumor completely from the mice within two weeks. The excellent anti-cancer efficiency benefits from the combined results of photothermal effect from NIR-irradiated SA and chemotherapy from DOX controlled release by heat. Therefore, this chemo-photothermal therapeutic system exhibits a great potential for cancer treatment.

Carboxymethylcellulose based self-healing hydrogel with coupled DOX as Camptothecin loading carrier for synergetic colon cancer treatment

Liping Yin,Kaiyue Zhang,Weichen Sun,Yu Zhang,Yong Wang,Jianglei Qin

The self-healing hydrogels have important applications in biomedication as drug release carrier. In this research, the Doxorubicin (DOX) was coupled onto oxidized carboxymethylcellulose (CMC) (CMC-Ald) to fabricate self-healing hydrogel with intrinsic antitumor property and loaded with Camptothecin (CPT) for synergetic antitumor treatment. The DOX coupled CMC-Ald (CMC-AD) was reacted with poly(aspartic hydrazide) (PAH) to fabricate injectable self-healing hydrogel. The coupled DOX avoided the burst release of the drug and the 100 % CPT loaded hydrogel could take the advantages of both drugs to enhance the synergetic antitumor therapeutic effect. The in vitro and in vivo results revealed the CPT loaded CMC-AD/PAH hydrogel showed enhanced antitumor property and reduced biotoxicity of the drugs. These properties demonstrate that the CMC-AD/PAH hydrogel has great application prospects in biomedication.

Rattle-type hollow CaWO4:Tb3+@SiO2 nanocapsules as carriers for drug delivery

Xuefeng Zhai,Min Yu,Ziyong Cheng,Zhiyao Hou,Ping'an Ma,Dongmei Yang,Xiaojiao Kang,Yunlu Dai,Dong Wang,Jun Lin

Rattle-type hollow nanocapsules are among of the most promising candidates as drug carriers owing to their huge inner space and multifunctional material combination. In this paper, rattle-type hollow CaWO4:Tb3+@SiO2 nanocapsules with a diameter of 100–110 nm and a wall thickness around 10 nm were fabricated. The hollow silica nanospheres were used as nano-reactors and the luminescent core of CaWO4:Tb3+ was post-filled into the nano-reactors by a vacuum nano-casting route combined with a Pechini-type sol–gel method. Subsequently, doxorubicin hydrochloride (DOX), a model of an anti-cancer drug, is loaded into the CaWO4:Tb3+@SiO2 nanocapsules and their cell cytotoxicity, cancer cell uptake and drug release behavior are investigated in vitro. The prepared multifunctional inorganic nanocapsules show a loading capacity for DOX as high as 124 mg g−1 and sustained-release properties. The release profile of the drug from DOX-loaded nanocapsules can last over five days. Besides, the blank CaWO4:Tb3+@SiO2 shows very low cytotoxicity against cancer cell lines (HeLa cell) while the DOX-loaded nanocapsules exhibit relatively high efficiency for killing of HeLa cells. The rapid cancer cell uptake process is observed by confocal laser scanning microscopy. The results indicate that a rattle-type hollow CaWO4:Tb3+@SiO2 nanocapsule has the potential to be used as drug carrier in therapy. Moreover, it is possible to extend the synthetic strategy in this study to other rattle-type multifunctional composites to meet various demands.

DOX sensitized upconversion metal–organic frameworks for the pH responsive release and real-time detection of doxorubicin hydrochloride

Feng Tian,Chen Xu,Mingyue Xu,Haiqing Gao,Ziyi Xiao,Ling Li,Yingxi Wang

Drug resistance is a major obstacle in cancer treatment, and designing a material that monitors real-time drug release remains a top priority. In this study, metal–organic frameworks doped with lanthanum and thulium were synthesized and then coated with aminated silica to form La/Tm-MOF@d-SiO2 as a drug carrier. Doxorubicin hydrochloride (DOX) was selected as a drug model, and the drug loading and release were investigated. It was found that the release of DOX under acidic conditions reached an optimal level, indicating the pH-responsiveness of La/Tm-MOF@d-SiO2. Under acidic conditions (pH = 5.8), upconversion fluorescence was generated after loading DOX on La/Tm-MOF@d-SiO2. At pH = 5.8, the longer the drug released, the stronger the upconversion fluorescence. It was found that the upconversion fluorescence intensity is directly proportional to the amount of drug released; thus, the real-time monitoring of DOX release in tumor cells can be performed based on the upconversion fluorescence.

DOX-conjugated unimolecular micelles from benzaldehyde-functionalized star copolymer via metal-free ATRP for pH-responsive drug delivery

Wei Ma,Hongyan Yang,Xinyue Ma,Jiao Huang,Li Chen,Atsushi Takahara

The development of polymer prodrugs with well-defined architectures capable of forming stable nanomicelles is important for achieving precise and efficient drug delivery. Compared with conventional linear systems, star-shaped copolymer prodrugs offer enhanced micellar stability owing to their covalently tethered architecture. However, many existing synthetic approaches rely on transition-metal catalyzed polymerization and customized monomers, which limit their sustainability and practical applicability. Herein, we report a pH-responsive doxorubicin (DOX)-conjugated benzaldehyde-functionalized star-shaped copolymer synthesized via an environmentally benign metal-free atom transfer radical polymerization (ATRP) strategy. A hydroxyl-functionalized star-shaped copolymer scaffold was first prepared from readily available monomers and subsequently modified to introduce pendant benzaldehyde groups, enabling DOX conjugation through acid-labile imine linkages. This approach affords well-controlled polymer architectures while avoiding transition-metal residues and the need for pre-functionalized monomers. The resulting DOX-conjugated star-shaped copolymers form unimolecular micelles with high colloidal stability and exhibit pH-triggered drug release under acidic conditions. In vitro studies further demonstrate effective cellular internalization and a moderated cytotoxic profile toward human large-cell lung carcinoma (H460) cells, supporting the functional viability of this micellar system as a polymer prodrug delivery platform.

Absolutely “off–on” fluorescent CD-based nanotheranostics for tumor intracellular real-time imaging and pH-triggered DOX delivery

Yuman Dong,Pengcheng Du,Peng Liu

Carbon dots (CDs) have attracted intense attention in tumor nanotheranostics recently; however, those nanotheranostics exhibited similar fluorescence in both normal and tumor tissues, limiting their practical application. In the present work, absolutely “off–on” fluorescent CD-based nanotheranostics was designed for tumor intracellular real-time imaging and pH-triggered DOX delivery via both static quenching by the crosslinking of benzaldehyde-containing diblock copolymers and dynamic quenching because of the surrounding conjugated DOX molecules. The proposed PPEGMA42-b-PFPMA122–(CDs)–DOX nanotheranostics did not exhibit fluorescence in a normal physiological medium, while strong fluorescence recovery occurred in the tumor intracellular microenvironment due to pH-triggered disintegration, releasing the CDs and DOX. The pH-triggered DOX release and absolute “off–on” fluorescence make the proposed nanotheranostics promising for tumor-specific pH-triggered DOX delivery and imaging.

“Two in one”: Simultaneous functionalization and DOX loading for fabrication of nanodiamond-based pH responsive drug delivery system

Wei Long,Hui Ouyang,Weimin Wan,Wenfeng Yan,Chaoqun Zhou,Hongye Huang,Meiying Liu,Xiaoyong Zhang,Yulin Feng,Yen Wei

Nanodiamond (ND) has been widely studied as a new type of carbon nanomaterials that is expected to be used as a promising candidate in various fields especially in the field of biomedicine. However, its poor water dispersibility and insufficient controlled release limit its practical applications. In this paper, ND-based composites with pH-responsive hydrazone bonds were successfully prepared by a simple chemical reaction between ester groups and hydrazine hydrate, in which ester groups were conjugated on the surface of ND via thiol-ene click reaction. On the other hand, CHO-PEG and doxorubicin hydrochloride (DOX) were linked on the carriers through formation of hydrazone bonds, resulting in improving water dispersibility and high drug loading capacity. The structure, thermal stability, surface morphology and particle size of ND carriers were characterized by different equipment. Results demonstrated that we have successfully prepared these functionalized ND. The release rate of DOX in acidic environment was significantly greater than that in normal physiological environment. More importantly, cell viability and optical imaging results showed that ND-based composites possess good biocompatibility, therapeutic effect, and could successfully transport DOX to HepG2 cells. Considering the above results, we believe that our new ND carriers will become promising candidates for intracellular controlled drug delivery and cancer treatment.

Nuclear-targeted p53 and DOX co-delivery of chitosan derivatives for cancer therapy in vitro and in vivo

Jian-Tao Lin,Hua Chen,Dan Wang,Liang Xiong,Jia-Zhu Li,Gui-hong Chen,Guang-Bin Chen

The nucleus is one of the most important cellular organelles. Chitosan-grafted poly-(N-3-carbobenzyloxy-lysine) (CCL) decorated with human immunodeficiency virus-1 transactivator of transcription (TAT) can co-deliver p53 and doxorubicin into the nucleus simultaneously, such that their antitumor functions are exerted. However, TAT-CCL has been shown to have an anti-tumor effect only in vitro; the effect in vivo was unsatisfactory. Here, a unique nucleus-targeted delivery system based on amidized TAT (aTAT)-CCL with aTAT functional on the surface was designed to achieve a highly efficient nucleus-targeting gene and drug delivery system for effective cancer cell elimination in vitro and in vivo. In this delivery system, TAT is amidized to inhibit its nonspecific interactions. Confocal laser scanning microscopy observations revealed that if aTAT-CCL was incubated in pH 5.0 acetate buffer solution for 24 h before use (named aTAT-CCL-HB), more aTAT-CCL-HB entered the nucleus compared with aTAT-CCL or CCL. aTAT-CCL-HB can also achieve high gene transfection and drug delivery efficiencies and low viability in HepG2 cells. However, only aTAT-CCL achieved extensive circulation in the blood compartment and high antitumor activity in vivo. Amidization of TAT in vectors may become a promising strategy for nucleus-targeted delivery systems, especially in in vivo applications.

Tumor environment responsive degradable CuS@mSiO2@MnO2/DOX for MRI guided synergistic chemo-photothermal therapy and chemodynamic therapy

Minyi Zhang,Xijian Liu,Qian Luo,Qi Wang,Linjing Zhao,Guoying Deng,Renbin Ge,Lei Zhang,Junqing Hu,Jie Lu

Chemo-photothermal therapy and chemodynamic therapy have a great promise for enhanced tumor treatment effect for integration of Reactive Oxygen Species (ROS) and O 2 generation. Hereby, a smart tumor environment (TME) responsive theranostic agent based on CuS@mSiO 2 @MnO 2 nanocomposites was fabricated by a facile method. When the nanocomposites entered tumor, the drug release was triggered by multi-model response of NIR irradiation, weak acidity and high glutathione (GSH) in TME. The O 2 and toxic •OH were generated from H 2 O 2 in TME through catalyzing by MnO 2 , which relieved hypoxia and enhanced the effect of chemodynamic therapy (CDT). Meanwhile, the generated Mn 2+ in TME was used to monitor the nanocomposites delivery by magnetic resonance imaging (MRI). Also, the excellent photothermal therapy effect could be triggered by NIR irradiation for CuS core. More importantly, the growth of tumors was greatly inhibited by the synergetic chemo-photothermal therapy and CDT. This work may open a wide route for tumor environment responsive theranostic, including relieving hypoxia, chemo-photothermal therapy, CDT and MR imaging.

pH/Reduction Dual-Responsive Oxidized Alginate-Doxorubicin (mPEG-OAL-DOX/Cys) Prodrug Nanohydrogels: Effect of Complexation with Cyclodextrins

Tingting Zhou,Jiagen Li,Xu Jia,Xubo Zhao,Peng Liu

Novel biocompatible and biodegradable pH/reduction dual-responsive oxidized alginate-doxorubicin (mPEG-OAL-DOX/Cys) prodrug nanohydrogels were designed for tumor-specific intracellular triggered release of anticancer drug DOX by conjugating DOX via acid-labile Schiff base linkage into the PEGylated oxidized alginate (mPEG-OAL) cross-linked with bioreducible disulfide bond. The effect of the complexation with cyclodextrins (α-CD and β-CD) before or after the cross-linking of the mPEG-OAL on the DOX content and controlled release performance was investigated. It was found that the cyclodextrin inclusion complex prodrug nanohydrogels mPEG(CD)-OAL-DOX/Cys, prepared by cross-linking of the mPEG-OAL after complexation with cyclodextrins, exhibited better pH/reduction dual-responsive controlled release performance than the mPEG-OAL-DOX/Cys(CD) ones prepared by cross-linking of the mPEG-OAL before complexation with cyclodextrins, owing to the supramolecular cross-linking of the adjacent pseudopolyrotaxanes. Especially for the cyclodextrin inclusion complex prodrug nanohydrogels mPEG(α-CD)-OAL-DOX/Cys, DOX was released rapidly under lower pH media mimicking the tumor microenvironment and completely released within 48 h, while the premature leakage under the simulated physiological condition was ∼40%, without burst release in both cases. The cellular toxicity and uptake results demonstrated that the mPEG(α-CD)-OAL-DOX/Cys prodrug nanohydrogels possessed similar inhibition against cancer cell growth in comparison with the free DOX and enhanced drug intracellular accumulation.

Chemo/Photoacoustic Dual Therapy with mRNA-Triggered DOX Release and Photoinduced Shockwave Based on a DNA-Gold Nanoplatform

Yundong Zang,Yanchun Wei,Yujiao Shi,Qun Chen,Da Xing

A multifunctional nanoparticle based on gold nanorod (GNR), utilizing mRNA triggered chemo-drug release and near-infrared photoacoustic effect, is developed for a combined chemo-photoacoustic therapy. The constructed nanoparticle (GNR-DNA/FA:DOX) comprises three functional components: (i) GNR as the drug delivery platform and photoacoustic effect enhancer; (ii) toehold-possessed DNA dressed on the GNR to load doxorubicin (DOX) to implement a tumor cell specific chemotherapy; and (iii) folate acid (FA) modified on GNR to guide the nanoparticle to target tumor cells. The results show that, upon an effective and specific delivery of the nanoparticles to the tumor cells with overexpressed folate receptors, the cytotoxic DOX loaded on the GNR-DNA nanoplatform can be released through DNA displacement reaction in melanoma-associated antigen gene mRNA expressed cells. With 808 nm pulse laser irradiation, the photoacoustic effect of the GNR leads to a direct physical damage to the cells. The combined treatment of the two modalities can effectively destroy tumor cells and eradicate the tumors with two distinctively different and supplementing mechanisms. With the nanoparticle, photoacoustic imaging is successfully performed in situ to monitor the drug distribution and tumor morphology for therapeutical guidance. With further in-depth investigation, the proposed nanoparticle may provide an effective and safe alternative cancer treatment modality.
