A self-assembled metal-polyphenolic nanomedicine for mild photothermal-potentiated chemodynamic therapy of tumors

Huizhu Yu,Ming Ma,Kaicheng Liang,Jie Shen,Zhengyi Lan,Hangrong Chen

Engineering nanotherapeutics with simple ingredients but multiple functions at large-scale via easy methods is still a formidable challenge for cancer treatment. Herein, a novel multifunctional nanotherapeutic agent (named as FeEP-NP) was fabricated by a one-pot self-assembly method based on the coordination between Fe II and (-)-epigallocatechin gallate (EGCG) with poly(vinylpyrrolidone) serving as a stabilizer. The designed FeEP-NPs could produce the toxic hydroxyl radical (•OH) effectively via Fenton reaction for chemodynamic therapy (CDT). Intriguingly, the unique binding between EGCG and Fe II ions endows nanoparticles with strong absorption and photothermal conversion capabilities in the near-infrared (NIR) region, which could achieve mild hyperthermia-enhanced CDT under NIR laser irradiation. More impressively, the partially released EGCG could accelerate the Fe III /Fe II conversion to augment the generation of •OH to further promote CDT, and simultaneously down regulate the intracellular expression of heat shock protein 90 (HSP 90) to enhance mild photothermal therapy (PTT). Both in vitro and in vivo results demonstrate that the low-temperature PTT-potentiated CDT based on FeEP-NPs could suppress tumors more efficiently. Overall, this work presents a novel multifunctional nanoplatform with succinct design, highlighting significant prospects for future clinical application.

Eco-friendly composite nanofibrous membranes loaded with chitosan microcapsules for enhanced antibacterial and deodorant application

Hongnan Zhang,Mengting Cui,Tingting Zhang,Xiaohong Qin

Traditional textiles are conductive to breed germs that might cause infection or unpleasant odor, especially in public hygiene products. Moreover, biosafety, environmental protection and cost are crucial considerations for antimicrobial materials. Herein, we developed a biocompatible microcapsules/nanofibrous membrane using eco-friendly and facile strategy. The microcapsules loaded with the Epigallocatechin and gallate (EGCG) were synthesized through an ion-crosslinking reaction between chitosan (CS) and sodium tripolyphosphate (TPP). Additionally, the addition of octadecyl trimethyl ammonium chloride (STAC) could further enhance the antibacterial activity . Following that, the microcapsules were combined with the polycaprolactone (PCL) nanofibers . The resultant composite nanofibrous membrane exhibits a slow release effect, an enjoyable deodorant activity, and strengthened antibacterial activity through contact-killing and biocide-releasing. Besides, the membrane behaved a low degradation rate , without compromising its mechanical and hydrophilic properties after a 20-day degradation. These findings provided a biocompatible and practical approach for antimicrobial and odor-proof material for hygienic and sanitary products.

ROS/Glucose-Dissociable EGCG-Coated Oxygen-Supplying Nanocomposite Hydrogel for Diabetic Wound Therapy

Shiyu Sun,Rongxue Zhu,Junneng Du,Jin Lin,Yuying Li,Qian Jiang,Tianzong Huang,Enping Huang,Yingping Jiang,Xiaoya Zhang

Background: In diabetic wounds, a detrimental and persistent inflammatory environment characterized by impaired angiogenesis and concomitant hypoxia severely impedes wound healing. This study utilized phenylboronic acid-grafted quaternized chitosan (QCS-PBA) to encapsulate calcium peroxide (CaO2) within the QCS-PBA matrix. Epigallocatechin gallate (EGCG) was then anchored onto the surface via phenylborate ester bonds to fabricate oxygen-producing nanoparticles (QP@Ca-E). Finally, the nanoparticles were integrated into a multifunctional hydrogel (GP-GL) prepared from functionalized gelatin. This system provides a promising therapeutic strategy for irregular wound closure, hypoxia alleviation, inflammatory response modulation, and angiogenesis promotion. Methods: In this study, QP@Ca-E loaded with CaO2 and EGCG were combined with functionalized gelatin grafted with double bonds/lipoic acid (GP-GL) to fabricate a photocurable and reactive oxygen species (ROS)-responsive hydrogel delivery system (GP-GL@Ca-E). The system was systematically evaluated for its ROS/glucose-responsive release behavior, antioxidant activity, and antibacterial properties. Through in vitro experiments, the hydrogel’s biocompatibility and its effects on cellular ROS/hypoxia levels, angiogenic capacity, and macrophage polarization toward the M2 phenotype were further investigated. Additionally, a full-thickness skin defect model in diabetic rats was established to validate the hydrogel’s efficacy in promoting wound healing, restoring oxygen supply, mitigating inflammatory responses, and accelerating angiogenesis. Results: The GP-GL@Ca-E oxygen-supplying multifunctional hydrogel provides an effective physical barrier for irregular wounds. Moreover, the GP-GL@Ca-E hydrogel responds to ROS and glucose in the wound microenvironment to enable sustained EGCG release. EGCG not only exhibits excellent antibacterial, anti-inflammatory, and antioxidant properties but also promotes angiogenesis. Subsequently, the exposed CaO2 reacts with water to continuously generate oxygen. The synergistic effect of EGCG and CaO2 effectively alleviates wound hypoxia and inflammatory responses while promoting neovascularization, thereby creating a favorable microenvironment for diabetic wound repair and significantly accelerating the healing process. In vivo studies confirmed that compared with the control group, the hydrogel-treated group showed significantly accelerated wound healing. By day 14, the wound healing rate reached 99.01% in the hydrogel group, significantly higher than the 86.32% observed in the control group, fully demonstrating its great potential for diabetic wound therapy. Conclusion: The results indicate the successful preparation of a multifunctional oxygen-producing hydrogel, which exhibits remarkable antibacterial properties and ROS scavenging capability. This hydrogel can effectively suppress inflammatory responses, promote macrophage polarization toward the M2 phenotype, and enhance angiogenesis, thereby creating a favorable immune microenvironment for skin regeneration. These characteristics collectively demonstrate its promising potential for application in diabetic wound healing.

Self-Assembled EGCG Nanoparticles Achieved Long-Term ROS Scavenging to Delay Osteoarthritis Progression

Xinyu Zhang,Mingda Zhao,Jiadong Li,Zhulian Li,Lei Tong,Hailong Wang,Gongbing Liu,Chunyu Tan,Yujiang Fan,Yong Sun

Oxidative stress disrupts the synthesis–degradation balance of the extracellular matrix in osteoarthritic (OA) cartilage, resulting in the loss of type II collagen (COLII). Here, we developed self-assembled nanoparticles (PE@NPs) driven by hydrophobic interaction, π–π stacking interactions and hydrogen bonding, forming an epigallocatechin-3-gallate (EGCG) core and a polyethylene glycol (PEG) shell. Compared with free EGCG, which possesses potent but short-lived antioxidant activity, PE@NPs improved molecular stability, extending reactive oxygen species scavenging activity to 24 h. Furthermore, PE@NPs significantly suppressed interleukin-1 β-induced COLII degradation in OA chondrocytes. Transcriptomic analysis revealed that PE@NPs upregulated genes involved in antioxidant defense (Selenop), cartilage homeostasis (Cytl1 and DKK3) and subchondral bone remodeling (Omd). In vivo, PE@NPs exhibited a more significant therapeutic effect than free EGCG, notably attenuating COLII degradation and improving subchondral bone mass, thereby delaying OA progression. Overall, these findings identify PE@NPs as a safe and effective therapeutic approach for OA.

(-)-Epigallocatechin-3-Gallate (EGCG) Regulates the Formation of Zein–Ovalbumin Heteroprotein Complex Coacervates

Min Huang,Jiayi Han,Hao Li,Junhao Lin,Qingbo Jiao,Zexing Cao,Qunfang Lei,Wenjun Fang,Gerui Ren,Zhijun Song,Ying Zheng,Ruibo Wu,Hujun Xie

Growing interest in sustainable nutrition has improved the importance of understanding the interactions between (-)-Epigallocatechin-3-gallate (EGCG), a polyphenol in green tea, and proteins for functional food development. In this study, we investigated the regulatory effects of EGCG on the aggregation behavior of zein, ovalbumin, and their complexes. The addition of EGCG at 100 μg mL −1 led to significant changes in particle sizes of zein, ovalbumin, and zein-ovalbumin complexes, increasing them from 215.6, 3485.3, and 2081.0 nm to 4822.3, 7857.0, and 5895.7 nm, respectively. The zeta potentials of these samples decreased from 8.4, −2.6, and 5.0 mV to −5.3, −0.66, and 2.13 mV, respectively, indicating a reduction in net surface charges and promotion of protein aggregation. UV–vis absorption spectroscopy revealed hyperchromic effects and blue shifts upon EGCG addition, suggesting conformational changes. Molecular dynamics simulations further revealed that EGCG contributed to the structural stabilization of both individual proteins and their complexes by reinforcing hydrophobic and electrostatic contacts at the molecular interface. These findings provide molecular-level insights into the regulatory role of EGCG in protein assembly and offer guidance for the rational design of plant-animal protein complexes with tailored functional properties for food applications. Graphical EGCG plays a significant regulatory role in protein aggregation behavior. EGCG interacts with protein molecules, promoting conformational changes and aggregation, particularly in systems involving zein and ovalbumin. This study highlights the potential application of EGCG in the development of functional foods.

Green Fabrication of Lactoferrin–Dendrobium Polysaccharide–EGCG Ternary Nanoparticles for Enhanced Lutein Delivery

Xiaoke Peng,Xiaofan Deng,Jianzhong Xu,Jiayi Guo,Rui Zhao,David Julian McClements,Cuicui Ma,Fuguo Liu

Lutein’s application in functional foods is limited by its poor solubility, instability, and low bioavailability. To address these challenges, ternary nanoparticles (LF-DOP-EGCG) were constructed using lactoferrin (LF), Dendrobium officinale polysaccharide (DOP), and epigallocatechin gallate (EGCG) via a surfactant-free, organic-solvent-free aqueous process. Optimized preparation conditions (0.5% LF, pH 3, LF/DOP-EGCG mass ratio 3:1) produced uniformly sized and well-dispersed nanoparticles. Spectroscopic analyses confirmed that DOP-EGCG interactions altered the secondary structure of LF. Compared with non-covalent complexes, covalently modified nanoparticles (LF-COV) exhibited improved thermal stability and antioxidant capacity. Lutein-loaded nanoparticles showed efficient encapsulation and loading, and spectral shifts verified successful incorporation. Covalently modified lutein-loaded nanoparticles (LF-COV-LUT) demonstrated markedly enhanced stability under heat, light exposure, and 40-day storage relative to free lutein. In vitro digestion further revealed that LF-COV-LUT slowed gastric degradation and facilitated intestinal release, resulting in significantly higher bioaccessibility (54.6 ± 8.7%) compared with free lutein (6.13 ± 0.83%). These findings indicate that this natural multicomponent nanoparticle system provides a green and effective approach for improving the stability and digestive availability of carotenoids in functional food applications.

CO-loaded Hemoglobin/EGCG Nanoparticles Functional Coatings for Inflammation Modulation of Vascular Implants

Wu Sui,Dong Ruichen,Xie Yinhong,Chen Wenhao,Liu Wei,Weng Yajun

During the implantation process of cardiovascular implants, vascular damage caused by inflammation occurs, and the inflammatory process is accompanied by oxidative stress. Currently, carbon monoxide (CO) has been demonstrated to exhibit various biological effects including vasodilatation, antithrombotic, anti-inflammatory, apoptosis-inducing, and antiproliferative properties. In this study hemoglobin/epigallocatechin-3-gallate (EGCG) core-shell nanoparticle containing coating on stainless steel was prepared for CO loading and inflammation modulation. Inspired by strong coordination ability with CO, hemoglobin nanoparticle was first prepared and encapsulated into EGCG metal-phenolic networks. A polydopamine linking layer was then coated on 316 stainless steel, and the hemoglobin/EGCG nanoparticles was loaded with the subsequent polydopamine deposition. It showed that the maximum release amount of CO by the coating was 17.0 nmol/cm2 in 48 hours. In vitro evaluations conducted in a simulated inflammatory environment revealed that the coating, which released CO from hemoglobin/EGCG nanoparticles, effectively mitigated the LPS-induced inflammatory response in macrophages. Specifically, it decreased the expression of TNF-α, increased the expression of IL-10, suppressed the polarization of macrophages towards the M1 phenotype, and reduced intracellular reactive oxygen species (ROS). Furthermore, under simulated oxidative stress conditions, the coating decreased the apoptosis of endothelial cells induced by oxidative stress and downregulated intracellular ROS levels. In vivo implantation results further confirmed that the coating, with its hemoglobin/EGCG nanoparticles and CO release capabilities, reduced macrophage-mediated inflammatory responses and modulated the polarization phenotype of macrophages.

The enzyme-oriented regulation of theaflavin-3, 3′-digallate synthesis and the accurate determination of its yield

Yangping Ding,Bingcan Chen,Huayi Suo,Huarong Tong

Summary Theaflavin-3,3′-digallate (TFDG) is the main product formed by oxidative condensation between (−)-Epigallocatechin-3-gallate (EGCG) and (−)-Epicatechin-3-gallate (ECG). Some sub-products, such as homo- and hetero-oligomers of EGCG and ECG, are also produced simultaneously, which significantly decreases the yield of TFDG. In general, the optimum temperature for TFDG synthesis is 20 °C below which can reduce its generation resulting in the coexistence of a large amount of unreacted substrates (ECG and EGCG). Therefore, a higher possibility of increasing the formation of TFDG could be achieved if a suitable synthetic method can be developed at a low temperature. Our results showed that simultaneous dropwise addition of both EGCG and ECG could markedly enhance the content of TFDG at ice bath. Using such technique, the production of TFDG increased by 47.60% as compared to the group synthesised under the optimum temperature. The absorbance of TFDG at 278 nm is 2.36 times higher than that of ECG with equimolar concentration. Therefore, a rapid, simple and accurate calculation method of TFDG yield was developed via the spectral correlation. Based on the counting method, the yield of TFDG was 30.58% by using the novel synthetic method of TFDG.

Production of protein-epigallocatechin gallate conjugates using free radicals induced by ultrasound and their gelation behavior

Chen Li,Yalu Zheng,Xiaohui Xiong,Feng Xue

In this study, free radicals generated by ultrasound were used to prepare conjugates of food proteins (soybean protein isolates, sodium caseinate and gelatin) with epigallocatechin gallate (EGCG). The changes in free amino and sulfhydryl group contents were used to confirm the occurrence of conjugation. The formation of covalent interactions on surface hydrophobicity, functional groups, structures, thermal stability, and gelation behavior of three proteins were investigated. The results showed that conjugation led to decrease in free amino and sulfhydryl group contents, reduction in the intensity of amide A and fluorescence intensity, and increase in β-fold content. The conjugation also resulted in a decrease in surface hydrophobicity and thermal stability of soybean protein isolates and sodium caseinate, but an increase in the surface hydrophobicity and thermal stability of gelatin. Furthermore, the covalent bonding between proteins and EGCG improved gel strength, water holding capacity, and resulted in a denser and more compact microstructure.

Mechanism of Inhibiting Spontaneous Coal Combustion by Dopa Hydrosol

Lianquan Suo,Zhilin Xi,Tong Xia,Tao Chen,Jiawei Han

A new type of biomass environmentally friendly hydrosol (DEP) was proposed to inhibit coal spontaneous combustion (CSC). The DEP consists of two parts, the PDopa side chain solution formed by the redox reaction of Dopa and EGCG and the main chain sol solution formed by the free radical polymerization of AA under the action of APS initiator. The −COOH, −NH2, and −OH in Dopa can be grafted onto the skeleton of the main chain via double strong hydrogen bonding interactions with the −COOH group in PAA to form a stable three-dimensional cross-linked network structure. DEP can inhibit CSC through three aspects: oxygen isolation, increasing the activation energy of coal oxidation, scavenging active groups and free radicals. The calculation simulation results show that EGCG in DEP can remove various active free radicals with lower activation energy at room temperature, breaking the chain cycle reaction of coal spontaneous combustion, which proves its high efficiency. This study provides a new way to apply mussel-inspired adhesion mechanisms to the prevention of CSC.

Syntheses and anticancer activities of novel glucosylated (−)-epigallocatechin-3-gallate derivatives linked via triazole rings

Shi Bo-Ya,Wang Ze-Hao,Zhang Ning,Xie Yin-Rong,Sun Xiu-Li,Yang Hao-Nan,Wu Yi-Long,Zi Cheng-Ting,Wang Xuan-Jun,Sheng Jun

Novel glucosylated (-)-epigallocatechin-3-gallate derivatives 10 – 13 having the EGCG analogs conjugated to the d -glucosyl azide were synthesized by carrying out the copper-catalyzed azide-alkyne cycloaddition (CuAAC) reaction, and were evaluated for their cytotoxicities against a panel of five human cancer cell lines (HL-60, SMMC-7721, A-549, MCF-7, and SW480) using MTT assays. Compounds 10 and 11 showed the highest levels of cytotoxicity against the HL-60 cells with IC 50 values of 4.57 and 3.78 μM, respectively, and showed moderate selectivity toward cancer cell lines. Compound 11 was also shown to induce apoptosis in HL-60 cells. Most notably, inclusion of the perbutyrylated glucose residue in an EGCG derivative was concluded to lead to increased anticancer activity.

EGCG-Loaded Zinc Ion-Cross-Linked Injectable Sodium Alginate Hydrogel for Extraction Socket Healing

Yaru Han,Chuanwen Wang,Meitong Liu,Baojin Ma,Shaohua Ge,Yang Yu

Postextraction wound healing under inflammatory conditions remains a crucial clinical challenge due to persistent oxidative stress and compromised bone regeneration. To address this critical issue, injectable hydrogel dressings offer a promising strategy for inflammatory wound healing after tooth extraction. Herein, we developed an epigallocatechin gallate (EGCG)-loaded sodium alginate/zinc ion (SA/Zn2+) composite hydrogel (designated SZE) through ionic cross-linking and multiple physical bonding interactions. The SZE hydrogel with high cytocompatibility could effectively scavenge intracellular ROS, protect mitochondria, inhibit the expression of inflammatory factors, and modulate macrophage polarization, as well as enhance the osteogenic differentiative potential of bone marrow mesenchymal stem cells. Meanwhile, the SZE hydrogel demonstrated good bactericidal effects against Staphylococcus aureus and Escherichia coli. Based on in vivo rat extraction socket models with/without S. aureus infection, SZE accelerated socket healing by eliminating infection, alleviating oxidative stress, inhibiting excessive inflammation, and promoting bone regeneration and effectively augmented infected socket healing. Overall, our research indicates that the developed SZE hydrogel offers a promising strategy for achieving accelerated and ideal healing of extraction sockets, especially under bacteria-infected inflammatory conditions.
