Spatiotemporally co-delivery of triple therapeutic drugs via HA-coating nanosystems for enhanced immunotherapy

Chaoqun Ma,Yichao Duan,Chaohui Wu,Erjuan Meng,Pingping Li,Zhenzhong Zhang,Chunhua Zang,Xueling Ren

There is growing empirical evidence that certain types of chemotherapy and phototherapy trigger immunogenic cell death and enhance the therapeutic anticancer efficacy of genetic immunotherapy. However, the main challenge is spatiotemporally co-delivering different drugs to maximize the therapeutic index of the combination therapy. In this study, a drug delivery system (HTCP-Au/shPD-L1/DOX) was designed with a polysaccharide-wrapped shell and a condensed DNA core. To construct the HTCP-Au vector, dodecyl side chains with a polyethylenimine (PEI) head were grafted onto hyaluronic acid, and AuNPs were grafted via Au-S bonds. During drug loading, PEI arrested shRNA plasmid DNA targeting programmed cell death ligand 1 (shPD-L1) via electrostatic interactions. It also formed a PEI-DNA core that was automatically enclosed when aliphatic hydrocarbons pulled the hyaluronic acid backbone. A hydrophobic interlayer consisting of dodecyl bridge chains between the PEI-DNA core and the hyaluronic acid shell was required to accommodate hydrophobic doxorubicin. In vitro and in vivo assays demonstrated that this core-shell drug delivery system could efficiently load and transport three different drugs and effectively target tumors. Moreover, it could activate the immune system, thereby providing promising therapeutic efficacy against tumor growth and metastasis.

Fiber optic Fabry–Perot interferometer based on HA/PVA composite film for humidity sensing

Zhaoxu Zhang,Huaping Gong,Changgui Yu,Kai Ni,Chunliu Zhao

A fiber optic humidity sensor based on Fabry-Perot (F-P) interferometer with the composite film of hyaluronic acid (HA) and polyvinyl alcohol (PVA) is proposed. The humidity characteristics of the sensors with different mixture ratios of HA/PVA composite are investigated for the first time. The experimental results show that the addition of HA to pure PVA contributes to the improvement of the humidity sensitive properties of the material. Furthermore, the sensor with the ratio of 1:2 HA/PVA composite film has the best performance for relative humidity (RH) sensing, whose humidity sensitivity is −233 pm/%RH. And the stability and response time of the sensor were tested. The sensor has the advantages of high sensitivity, low temperature cross sensitivity, fast response time and good stability.

Construction of coral rod-like MoS2@HA nanowires hybrids for highly effective green antisepsis

Zejun Wang,Bozhen Li,Jiarui Ren,Yufeng He,Pengfei Song,Rongmin Wang

Antibacterial materials have been rapidly emerging as a primary component in the mitigation of bacterial pathogens, and green functional materials play a vital role in the antibacterial field. In this study, biocompatible hydroxyapatite nanowires (HANW) was used as a carrier, a coral rod-like nanowires hybrid of MoS 2 and HANW (CR-MoS 2 @HANW) was synthesized via a facile two-step hydrothermal approach. After being characterized by scanning electron microscope (SEM), energy dispersive spectrometer (EDS), X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), fourier transform infrared spectroscopy (FT-IR), Brunauer-Emmet-Teller (BET) and thermogravimetric (TG) analysis, the antibacterial activity and environmental compatibility were assessed. It was found that MoS 2 nanosheets were in-situ assembled onto surface of HA nanowires, and the obtained nanohybrid exhibited excellent stability. CR-MoS 2 @HANW endowed a desirable long-term antibacterial activity against both gram-negative E. coli and gram-positive S. aureus. It was sufficient to inhibit the growth of bacteria within 72 h, and nanohybrids effectively promoted the growth of plants. In summary, the combination of MoS 2 and HANW created a novel eco-friendly nanohybrids that could be applied as a promising multi-functional green antisepsis. And the CR-MoS 2 @HANW possessed enormous potential for biomedical applications.

HA-coated collagen nanofibers for urethral regeneration via in situ polarization of M2 macrophages

Niu Yuqing,Stadler Florian J.,Yang Xu,Deng Fuming,Liu Guochang,Xia Huimin

In situ tissue engineering utilizes the regenerative potential of the human body to control cell function for tissue regeneration and has shown considerable prospect in urology. However, many problems are still to be understood, especially the interactions between scaffolds and host macrophages at the wound site and how these interactions direct tissue integration and regeneration. This study was designed to evaluate the efficacy of hyaluronic acid (HA) functionalized collagen nanofibers in modulating the pro-healing phenotype expression of macrophages for urethral regeneration. Tubular HA-collagen nanofibers with HA-coating were prepared by coaxial electrospinning. The formation of a thin HA-coating atop each collagen nanofiber endowed its nanofibrous mats with higher anisotropic wettability and mechanical softness. The macrophages growing on the surface of HA-collagen nanofibers showed an elongated shape, while collagen nanofibers’ surface exhibited a pancake shape. Immunofluorescence and ELISA analysis showed that elongation could promote the expression of M2 phenotype marker and reduce the secretion of inflammatory cytokines. In vivo experiments showed that tubular HA-collagen nanofibers significantly facilitate male puppy urethral regeneration after injury. In the regenerated urethra bridged by tubular HA-collagen nanofibers, anti-inflammatory M2 macrophages are recruited to the surface of the scaffold, which can promote angiogenesis and endogenous urothelial progenitor cell proliferation.

Fabrication of FA/HA-functionalized carbon dots for human breast cancer cell targeted imaging

Junting Tao,Hao Zou,Xiaokun Liao,Xinjian Lu,Jun cao,Jiaqi Pan,Chaorong Li,Yingying Zheng

Green fluorescent carbon dots (CDs) were prepared by one-step hydrothermal method and then modified into folic acid functionalized carbon dots (FA-CDs) and hyaluronic acid functionalized carbon dots (HA-CDs) with targeted function to study their application in breast cancer cells imaging. The microstructure of the CDs observed through TEM showed the CDs with a scale of 2.69 nm. FT-IR and XPS showed the changes of bonds and functional groups that confirmed the transformation of COOH and NH 2 to amide bonds. FA-CDs and HA-CDs had good water solubility and cytocompatibility, which laid a foundation for their application in human breast cancer cells imaging. At the same time, FA-CDs and HA-CDs had strong fluorescence excitation, and the optimal emission wavelength was about 450 nm. In fluorescence imaging of cells, carbon dots had bright green fluorescence in both breast cancer cells (MCF-7 cells) and normal cells (EC cells). After targeted endocytosis, FA-CDs and HA-CDs could emit bright green fluorescence in cancer cells but could not in normal cells, which proved that the synthesized FA-CDs and HA-CDs had targeting properties. FA-CDs and HA-CDs could be used to accurately identify breast cancer cells and normal cells as cancer diagnosis material, which had the potential application in early cancer diagnosis.

Mechanical Properties and In Vitro Biocompatibility of Hybrid Polymer-HA/BAG Ceramic Dental Materials

Yuanyuan Chen,Cheng Sun,Jinfang Cao,Yuanyuan Wu,Bencang Cui,Jianfeng Ma,Huining Wang

The aim of this study is to prepare hybrid polymer–ceramic dental materials for chairside computer-aided design/computer-aided manufacturing (CAD/CAM) applications. The hybrid polymer–ceramic materials were fabricated via infiltrating polymerizable monomer mixtures into sintered hydroxyapatite/bioactive glass (HA/BAG) ceramic blocks and thermo-curing. The microstructure was observed by scanning electron microscopy and an energy-dispersive spectrometer. The phase structure was analyzed by X-ray diffraction. The composition ratio was analyzed by a thermogravimetric analyzer. The hardness was measured by a Vickers hardness tester. The flexural strength, flexural modulus, and compressive strength were measured and calculated by a universal testing machine. The growth of human gingival fibroblasts was evaluated by a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) colorimetric assay and immunofluorescence staining. The results showed that the sintering temperature and BAG content affected the mechanical properties of the hybrid polymer–ceramic materials. The X-ray diffraction analysis showed that high-temperature sintering promoted the partial conversion of HA to β-tricalcium phosphate. The values of the hardness, flexural strength, flexural modulus, and compressive strength of all the hybrid polymer–ceramic materials were 0.89–3.51 GPa, 57.61–118.05 MPa, 20.26–39.77 GPa, and 60.36–390.46 MPa, respectively. The mechanical properties of the hybrid polymer–ceramic materials were similar to natural teeth. As a trade-off between flexural strength and hardness, hybrid polymer–ceramic material with 20 wt.% BAG sintered at 1000 °C was the best material. In vitro experiments confirmed the biocompatibility of the hybrid polymer–ceramic material. Therefore, the hybrid polymer–ceramic material is expected to become a new type of dental restoration material. Keywords:hybrid polymer–ceramic;hydroxyapatite;bioactive glass;mechanical properties;human gingival fibroblasts

DLP printing of BT/HA nanocomposite ceramic scaffolds using low refractive index BT crystals

Ruixuan Xue,Peimei Yuan,Bo Zhao,Fei Jing,Xuyuan Kou,Wangyang Yue,Yan wang,Dongmei Wang,Galhenage Asha Sewvandi,Dengwei Hu

Biological piezoelectric materials have significant potential for bone repair and energy harvesting owing to their excellent biocompatibility and piezoelectric effect. The BaTiO 3 /Ca 10 (PO 4 ) 6 (OH) 2 (BT/HA) composite material is an outstanding representative of biological piezoelectric materials, which has not been individually designed using digital light processing (DLP) 3D printing because of the large difference in the refractive index of its components. Therefore, in this work, double-sided-tooth plate-like BT crystals with high curvature were prepared via a hydrothermal process, and BT/HA ceramic slurries were grinded out using dispersed intermittent ball milling scheme, and BT/HA nanocomposite ceramic scaffolds were fabricated by DLP 3D printing technology. The nanostructure, dielectric properties, and piezoelectric energy harvesting performance of the BT/HA nanocomposite ceramic scaffolds were evaluated. The influences of different morphologies and contents for BT on the piezoelectric potential and stress distribution were analyzed based on a multi-physics coupling finite element simulation. The cell proliferation and adhesion abilities were investigated also. The BT/HA nanocomposite ceramic scaffolds present excellent dielectric properties, cell proliferation and adhesion abilities, and an open circuit voltage of 8 V during piezoelectric energy harvesting. The material properties and multi-physics coupling finite element analysis imply that the double-sided-tooth plate-like BT plays an important role for the fastness structure and electric field distribution in the BT/HA nanocomposite. Thus, this work provides a strategy for the application of the customized BT/HA nanocomposite ceramic scaffolds in new-generation orthopedic implants and biological energy harvesting.

Multifunctional CaCO3@Cur@QTX125@HA nanoparticles for effectively inhibiting growth of colorectal cancer cells

Hu Shengyun,Xia Kunkun,Huang Xiaobei,Zhao Ye,Zhang Qingqing,Huang Dongdong,Xu Weiyi,Chen Zhengju,Wang Chenfei,Zhang Zhiyong

Colorectal cancer (CRC) is a major cause of cancer-related deaths in humans, and effective treatments are still needed in clinical practice. Despite significant developments in anticancer drugs and inhibitors, their poor stability, water solubility, and cellular membrane permeability limit their therapeutic efficacy. To address these issues, multifunctional CaCO3 nanoparticles loaded with Curcumin (Cur) and protein deacetylase (HDAC) inhibitor QTX125, and coated with hyaluronic acid (HA) (CaCO3@Cur@QTX125@HA), were prepared through a one-step gas diffusion strategy. Dynamic light scattering (DLS), transmission electron microscopy (TEM), and scanning electron microscopy (SEM) showed that CaCO3@Cur@QTX125@HA nanoparticles have uniform spherical morphology and elemental distribution, with diameters around 450 nm and a Zeta potential of − 8.11 mV. The controlled release of Cur from the nanoparticles was observed over time periods of 48 h. Cellular uptake showed that CaCO3@Cur@QTX125@HA nanoparticles were efficiently taken up by cancer cells and significantly inhibited their growth. Importantly, CaCO3@Cur@QTX125@HA nanoparticles showed specific inhibitory effects on CRC cell growth. Encouragingly, CaCO3@Cur@QTX125@HA nanoparticles successfully internalized into CRC patient-derived organoid (PDO) models and induced apoptosis of tumor cells. The multifunctional CaCO3@Cur@QTX125@HA nanoparticles hold promise for the treatment of CRC.

A pH-sensitive hyaluronic acid prodrug modified with lactoferrin for glioma dual-targeted treatment

Yatao Yin,Chaoping Fu,Mei Li,Xiangpen Li,Mengying Wang,Lei He,Li-Ming Zhang,Ying Peng

Gliomas are the most common and lethal type of primary malignant brain tumor. But the existence of blood brain barrier (BBB) and blood-tumor barrier (BTB) hinder drug from reaching the tumor site. To address this problem, we developed a novel prodrug (Lf-HA-DOX) by conjugating hyaluronic acid with doxorubicin (HA-DOX) by an acid-labile hydrazone linkage, which is released in an acidic environment and accumulates in tumor tissues. Lactoferrin (Lf) was coupled for transporting across the BBB. In vitro, the release of DOX from Lf-HA-DOX was pH-dependent. At lower pH (5.0 and 6.0), the release of DOX was much quicker than that at pH   7.4. In the cellular uptake studies, flow cytometry analyses and confocal laser scanning microscopy results showed significantly enhanced cellular uptake of Lf-HA-DOX and HA-DOX in C6 cells compared to DOX. In BALB/C mice bearing C6 glioma, enhanced accumulation of Lf-HA-DOX in the glioma was observed by real time fluorescence image. Correspondingly, glioma-bearing mice treated with Lf-HA-DOX displayed the longest median survival time, which was 2-fold longer than that of saline group. In conclusion, Lf-HA-DOX was able to significantly increase drug delivery to the glioma, which might provide a promising strategy for antiglioma therapy.

Oral colon-targeted responsive alginate/hyaluronic acid-based hydrogel propels the application of infliximab in colitis

Manxiu Huai,Mingliang Pei,Jiaxing Pan,Yun Zhu,Yingwen Chen,Peng Du,Yanming Duan,Huixiong Xu,Wensong Ge

Currently, commercialized infliximab (IFX) has rapidly propelled the clinical treatment of IBD, however, its inherent attributes, such as off-target effects and rapid metabolism, severely limit practical applications. Moreover, high doses injection of IFX can result in IBD treatment failure, which may induce other side effects. In this study, an colon microenvironment-responsive hydrogel (AL/HA hydrogel), consisting of acid-resistant sodium alginate and colon-degraded and targeted hyaluronic acid, was constructed by simple Ca 2+ /Zn 2+ cross-linking. The ion-mediated hydrogel exhibited the protective effect of gastrointestinal tract to avoid early drug leakage, while the inflammation environments showed well-controlled drug release and significant biodegradable behaviors. Additionally, oral hydrogel exhibited long-standing enteritis areas compared with normal mice. Therefore, hydrogel-assisted enteritis treatment has great potential in IBD as an oral agent. After that, IFX was packaged in hydrogel to fabricate a facile oral antibody delivery system to treat IBD. IFX-embedded hydrogel showed remarkable therapeutic effect on IBD compared with free IFX. Surprisingly, oral hydrogel below 7 times IFX achieve the same amount of IFX-infused treatment that will further help alleviate the drawbacks of IFX. Our work elaborated on the efficacy of oral AL/ [email protected] in IBD, providing a guarantee for the future of promoted clinical transformation.

Antibacterial and antioxidant films based on HA/Gr/TA fabricated using electrospinning for wound healing

Xiang Bao,Qianying Zhu,Yunyun Chen,Huijuan Tang,Weimin Deng,Haixia Guo,Li Zeng

Prevention of bacterial contamination, maintenance of redox balance in the environment, and acceleration of wound healing are key requirements for wound dressing. In the present study, hyaluronic acid (HA)/graphene (Gr)-electrospun fibre films loaded with polyphenolic tannic acid (TA) were prepared using electrospinning. The antioxidant activity of the films was then examined to determine whether they contained optimal TA concentrations for subsequent research. Following that, the surface morphology and physicochemical properties of the films were determined and in vitro experiments were conducted to assess their biocompatibility and antibacterial activity. Finally, the in vivo effects of the electrostatically spun fibre films on infected wound healing in mouse models were observed. The HA/Gr/TA-electrospun fibre film with 0.3% w/v TA concentration displayed the best antioxidant activity and better mechanical, water-absorption, water-retention, and degradation properties than the film without TA. In addition, it displayed superior antibacterial activity and biocompatibility, as well acceleration of infected wound healing, than the film without TA. Therefore, the HA/Gr/TA-electrospun fibre film is a promising alternative option for wound dressings.

Modification of 3D printed PCL scaffolds by PVAc and HA to enhance cytocompatibility and osteogenesis

Jingqi Ma,Lili Lin,Yi Zuo,Qin Zou,Xin Ren,Jidong Li,Yubao Li

In the study, a specific material system that contains poly-(ε-caprolactone) (PCL), polyvinyl acetate (PVAc) and hydroxyapatite (HA) was used to fabricate porous scaffolds employing a 3D printing technique for bone regeneration. Four groups of 3D printing scaffolds were fabricated: PCL, PCL/PVAc, PCL/HA and PCL/PVAc/HA for comparision. The morphologies, mechanical properties and biological characteristics of these scaffolds were analyzed using SEM, a material testing machine, in vitro cell culture and in vivo animal experiments. The results showed that these 3D printed scaffolds possessed porous channel structures with a hole size of 375–475 μm and porosity of 74.1–76.1%. The compressive moduli of the scaffolds increased with the addition of HA and decreased with the addition of PVAc. The PCL/PVAc/HA scaffold exhibited higher cell proliferation and bone formation rates than other groups (p < 0.001), which could be attributed to the synergistic effect of PAVc and HA components. Two types of new bone formation patterns in the scaffold were found in this study: one is the new bone formed directly on the grid matrix, and the other is the new bone initially formed in the center of the scaffold channel and then remolded to concentric circles. The osteogenesis pattern of the latter is analogous to the osteon structure of a cortical bone. The 3D printed scaffold based on PCL/PVAc/HA tri-component system is a promising prospect for future individualized bone repair applications.
