Lab-on-tip: Protruding-shaped all-fiber plasmonic microtip probe toward in-situ chem-bio detection

Ling-Xin Kong,Yan-Xin Zhang,Wei-Gang Zhang,Yun-Shan Zhang,Tie-Yi Yan,Peng-Cheng Geng,Biao Wang

Lab-on-tip technology opens a new door for optical fiber surface plasmon resonance (SPR) sensors. In this paper, we first demonstrate an ultra-compact reflective SPR microtip probe on the flat fiber end in experiment, which has a length of tens of microns and a diameter of several microns. Simulations and experiments show that this new fiber SPR microtip probe coated with a nano-goldfilm has excellent refractive index (RI) sensing characteristic and great potential for biochemical detection at single cell level. Based on the high RI sensitivity, a bio-probe assembled with Chitosan/Polysodium Styrene Sulfonate (Cs/PSS) nano-polyelectrolyte film and the sheep anti-human IgG-HRP is prepared to recognize human IgG specifically. Due to the high surface sensitivity, the SPR microtip probe exhibits an ultra-low detection limit for human IgG. Due to the high bulk sensitivity and high thermo-optical coefficient (TOC) of polydimethylsiloxane (PDMS), a reference temperature probe based on SPR microtip is proposed, which is used to compensate for the temperature cross-sensitivity of the bio-probe. This new SPR microtip probe can be used not only in optical fiber sensor, but also in Tip-Enhanced Near-Field Raman Microscopy and Scanning Near-Field Optical Microscope.

A portable solar light-driven biophotoelectrocatalytic system for pollutant removal powered by photovoltaic cells

Kai Yan,Jianqiao Liu,Jin Qin,Jingdong Zhang

Photoelectrocatalytic (PEC) technology has been considered as one of the most efficient advanced oxidation processes for wastewater treatment , but the necessity of external electric power supply limits the portability of PEC system for on-field applications. Herein, a portable solar-powered biophotoelectrocatalytic system driven by photovoltaic (PV) cells was explored for degradation of pollutant by coupling BiVO 4 /Fe 2 O 3 -deposited photoanode and horseradish peroxidase (HRP)-immobilized cathode. The integration of PV cells in this PEC device could not only avoid the necessary introduction of external electric power supply, but also improve the utilization of solar irradiation. The characterization of BiVO 4 /Fe 2 O 3 photoanode demonstrates that ultrathin Fe 2 O 3 layer on BiVO 4 electrode could facilitate the holes transfer to the surface efficiently and thus avoid charge recombination. After HRP was immobilized on the cathode, the removal efficiency for 2,4-dichlorophenol (2,4-DCP) was obviously promoted, attributed to the efficient HRP-catalyzed oxidation reaction by in-situ generated H 2 O 2 from PEC process. Under natural solar irradiation, the proposed portable biophotoelectrocatalytic device exhibited a satisfying removal efficiency of 92.0% for 100 ppm of 2,4-DCP after 3-h treatment. The intermediate products formed during the degradation process were identified by liquid chromatography-mass spectrometry, and possible 2,4-DCP degradation pathway was also proposed.

Functional lightweight polystyrene@polydopamine nanoparticle for high-performance ELISA

Xueyu Zhang,Mingyan Liu,Minghui Yang,Wangqing Cheng,Juan Xiang,Wu Zhu,Xiang Chen

Nanoparticles are usually used as carrier to load more antibody and enzyme to improve the sensitivity of enzyme-linked immunosorbent assay (ELISA). However, limited by high density and complicated modification procedure, the traditional nanoparticles such as Au nanoparticles (AuNPs) usually induce large background signal and poor reproducibility in ELISA. In this work, functional lightweigh nanoparticle [email protected] ( [email protected] ) was prepared and induced as the carrier of detection antibody and horseradish peroxidase (HRP) to form [email protected] @Ab 2 /HRP biojungates. The appropriate density (close to water) and good hydropilicity ensure the good dispersion of [email protected] @Ab 2 /HRP in solution, preventing the physical sedimention and decreasing the background signal even though the bioconjugate's size is close to 200 nm. The large surface area and abundant active group from PDA facilitate the loading of detection antibody and HRP, improving the loading efficiency and stability of biojungates. Based on it, taking interleukin-17A (IL-17A, a biomarker of psoriasis) as the detection target, we developed a [email protected] sandwich ELISA, achieving a sensitive dynamic range from 0.3 to 80 pg/mL and a detection limit of 0.2 pg/mL. Furthermore, the contents of IL-17A were assayed successfully in 10-fold diluted serum samples from psoriasis patients. Compared with those commercial or AuNP-based ELISA, our [email protected] ELISA method exhibits higher sensitivity, lower background interference, and higher stability, which will significantly improve the application of ELISA in the low-abundant biomolecule assays.

Pomegranate-Bionic Encapsulating Horseradish Peroxidase Using Dopamine Flexible Scaffold-Coated Multishell Porous ZIF-8 To Enhance Immunochromatographic Diagnosis

Mingxuan Song,Juanxia Xing,Huan Cai,Xin Gao,Chunyang Li,Changjin Liu,Xinmin Li,Xuhuai Fu,Shijia Ding,Wei Cheng,Rui Chen

Nanoparticle-natural enzyme complexes are receiving increasing attention as the promising signal reporters for colorimetric lateral flow immunoassay (LFIA). Nonetheless, it remains a challenge to develop the nanocomplexes with high loading efficiency, catalytic efficiency, and colorimetric signal brightness. Herein, inspired by pomegranate structure, we reported the synthesis of a colorimetric catalytic nanocomplex ((HRP@ZIF-8)3@PDA@HRP), using dopamine flexible scaffold-coated multishell porous zeolitic imidazolate framework-8 (ZIF-8) as a hierarchical scaffold to encapsulate horseradish peroxidase (HRP), and described its potential to promote an ultrasensitive colorimetric LFIA of cardiac troponin I (cTnI). (HRP@ZIF-8)3@PDA@HRP exhibited ultrahigh HRP loading efficiency and catalytic activity due to the epitaxial shell-by-shell overgrowth of porous ZIF-8 scaffold, which provided more cavities for enzyme immobilization and a diffusion path for the catalytic substrate. Furthermore, the polydopamine (PDA) layer on the (HRP@ZIF-8)3 surface both enhanced the colorimetric signal brightness and acted as a flexible scaffold to immobilize HRP, further increasing the amount of enzyme. Following integration with LFIA, the developed platform achieved an ultrasensitive colorimetric test strip assay for cTnI with pre- and postcatalytic naked-eye detection sensitivities of 0.5 ng mL–1 and 0.01 ng mL–1, respectively, which were 4/2- and 200/100-fold higher than gold nanoparticles (AuNPs)/PDA-based LFIA and comparable to chemiluminescence immunoassay. Further, the quantitative testing results of the developed colorimetric LFIA on 57 clinical serum samples agreed well with the clinical data. This work provides ideas for the design of natural enzymes-based colorimetric catalytic nanocomplex to encourage applications for the development of ultrasensitive LFIA for early diseases diagnosis.

Biosensors with vancomycin and polymetallic metal-organic frameworks for colorimetric-fluorescent dual-mode detection and sterilization of bacteria

Wei Chen,Xiayu Peng,Lichao Kang,Shengnan Dong,Jian Zhang,Yunfeng Zhao,Fengxia Sun

The development of a versatile platform for bacterial assay and elimination is urgently needed due to the danger that bacteria pose to human life. Here, we synthesized a trimetallic deposition and horseradish peroxidase (HRP)-embedded porous coordination network-224 hybrid nanozymes (PCN-224 @AuPdPt@HRP) with outstanding peroxidase activity and fluorescence quenching ability. On this basis, we designed a dual recognition strategy-driven colorimetric-fluorescence dual-mode detection platform using Listeria monocytogenes as a pattern analyte. The platform consisted of an aptamer-modified PCN-224 @AuPdPt@HRP (PCN-224 @AuPdPt@HRP@Aptamer) specifically recognizing Listeria monocytogenes and vancomycin-coated 96-well plates. In the presence of vancomycin, which has the ability to recognize and inactivate gram-positive bacteria, the significant peroxidase activity of PCN-224 @AuPdPt@HRP@Aptamer in the precipitate was able to catalyze the color change of the substrate by H 2 O 2 . Meanwhile, the residual PCN-224 @AuPdPt@HRP@Aptamer in the supernatant was able to change the fluorescence of fluorescein-labeled deoxyribonucleic acid (FAM-DNA). In summary, this paper presents a multifunctional platform capable of detecting and eliminating residual bacteria in real environments. This strategy is expected to facilitate the development of multifunctional biosensors based on metal–organic framework probes and also provide environmental health.

A novel colorimetric sensing platform for the detection of S. aureus with high sensitivity and specificity

Yun Zhang,Shuyou Shi,Jiajia Xing,Wenqing Tan,Chenguang Zhang,Lin Zhang,Huan Yuan,Miaomiao Zhang,Jinjuan Qiao

In this study, a novel colorimetric sensing platform was developed for the detection of S. aureus using dog immunoglobulin G (IgG) as the capture antibody and chicken anti-protein A immunoglobulin Y labeled with horseradish peroxidase (HRP-IgY) as the detection antibody. Dog IgG labeled with magnetic beads was used to capture S. aureus through the interaction between the Fc region of dog IgG and Staphylococcal protein A (SPA). HRP-IgY was introduced to recognize the residual SPA on the surface of S. aureus and to create a sandwich format, after which a soluble 3,3′,5,5′-tetramethylbenzidine (TMB) substrate was added. A stop solution was utilized to cease the enzymatic chromogenic reaction, and then optical density was read at 450 nm. Under optimal conditions, the proposed method displayed a low detection limit of 1.0 × 103 CFU mL−1 and a wide linear range of 3.1 × 103 to 2.0 × 105 CFU mL−1. This detection method exhibited high specificity against other foodborne bacteria. The recovery rates ranged from 95.2% to 129.2%. To our knowledge, this is the first report to employ dog IgG and chicken IgY as an antibody pair to detect S. aureus. This technique exhibits high application potential for S. aureus monitoring in various kinds of samples.

Construction of an “aptamer-target-peptide” Sandwich Electrochemical Biosensor for Ultrasensitive Assay of Amyloid-Beta Oligomers based on Bimetallic Covalent Organic Framework

Lv Yubing,Zhou Yanli,Lv Chaoran,Wang Mengyao,Sheng Nan,Lin Yuanyuan,Wang Cancan,Xu Maotian,Zhang Jing,Yan Mei

In view of the importance of quantification of beta-amyloid (Aβ), an “aptamer -target- peptide” sandwich electrochemical biosensor to assay Aβ42 was fabricated by using gold–palladium bimetallic nanoparticles covalent organic framework-carbon cloth (Au–Pd@COF TTF-Thi -CC) and prion protein (PrP C ) labeled with horseradish peroxidase (HRP) nanocomposites. The concentration of Aβ42 showed a positive correlation with the peak current resulting from the oxidation of hydroquinone (HQ) by HRP. In contrast, the reference current signal from thionin (Thi), derived from covalent organic framework (COF), showed a negative correlation. The current response of the electrochemical biosensor was proportional to the Aβ concentrations ranging from 0.01 to 2.5 nM with a detection limit (LOD) of 2.17 fM ( S / N  = 3). The sensitivity of the sandwich electrochemical biosensor could be primarily amplified based on the substrates of Au–Pd@COF TTF-Thi -CC. The AuNPs and PdNPs not only facilitated the bonding of the linker based on the Au–S bond but also improved the performance of COF TTF-Thi -CC. Secondary sensitivity amplification of the biosensor could be achieved by using PrP C -labeled HRP to catalyze hydroquinone in solution. Furthermore, the biosensor showed excellent selectivity, anti-interference performance, and stability. The Aβ concentrations in cerebrospinal fluid (CSF) were successfully evaluated using the constructed biosensor. This assay provides a potential tool for early-stage Alzheimer's disease diagnosis.

Positional assembly of multi-enzyme cascade reaction in polyelectrolyte doped microcapsule through electrospray and layer-by-layer assembly

Shiyi Che,Jie Wang,Xiaoyuan Ji,Zhiguo Su,Shaomin Wang,Songping Zhang

Polyelectrolyte-doped microcapsules (PDM) was fabricated by coaxial electrospray of a mixture of glycerol and water containing 10 mg/mL cationic polyelectrolyte poly(allylamine hydrochloride) (PAH) fed as the core phase solution, and a N,N-dimethylacetylamide solution of 10 wt% polyurethane fed as the shell phase solution. Multi-enzyme system involving Candida Antarctica lipase B (CALB), glucose oxidase (GOD), and horseradish peroxidase (HRP) for cascade reaction was assembled in the PDM at three different places, namely, surface, shell, and lumen. Placing of enzyme inside aqueous lumen of the PDM was realized by in situ encapsulation through adding the enzyme in the core-phase solution for coaxial electrospray. By ion-pairing of enzyme with cationic surfactant CTAB, an organic soluble enzyme-CTAB complex was prepared, so that in situ embedding of enzyme in the shell of the PDM was realized by adding it into the shell phase solution. Surface attachment of enzymes was achieved by layer-by-layer (LbL) technology, which is based on the ion-exchange interactions between oppositely charged enzymes and PAH that was doped in PDM. The enzyme-decorated microcapsule was then studied as a micro-bioreactor, in which 1-Oxododecyla-α-glucopyranoside was converted by CALB to glucose, which was oxidised by GOD to gluconolactone in a second step. The hydrogen peroxide produced was then used by HRP to oxidize ABTS to form coloured radical cation ABTS • + for activity analysis. The successful fabrication of the PDM and precise localization of enzymes in the PDM by different strategies were fully characterized. By varying the immobilization strategy, totally six PDM bioreactors with three enzymes precisely positional assembled in different strategies were constructed and their activities for the cascade reaction were investigated and compared. The PDM micro-bioreactor prepared by novel electrospray technologies provide a smart platform for positional assembly of multi-enzyme cascade reaction in a precise and well-controlled manner.

Synergistic Effects of DNA Structure for Ultrasensitive Detecting OTA in Grains

Yuan Wei,Lu Lixia,Lu Yichen,Xiong Xiong,Li Yi,Cui Xiaowen,Liu Yuanjian,Xiong Xiaohui

Ochratoxin A (OTA) is considered as one of the most hazardous mycotoxins commonly occurring in grains. Thus, the monitoring of OTA is extremely significant. Herein, a synergistic strategy to detect OTA in grains based on the combination of DNA tetrahedron-structured aptamer (DTA) and hybridization chain reaction (HCR) was proposed. Briefly, DTA was assembled on the gold electrode. In the presence of OTA, the trigger DNA on the top of the DTA was exposed to unfold the biotin-modified hairpin structure DNA to HCR. It could conjugate plentiful streptavidin-labeled horseradish peroxidase (SA-HRP) to form polyaniline (PANI), which was regarded as electrochemical active molecule to detect OTA. In this way, the synergistic effect of DTA and HCR not only can provide stable probe structure to improve the recognition rate of OTA, but also can supply a large number of templates to synthesize PANI under mild conditions. As a result, the biosensor has a good analytical performance for OTA in a linear range of 0.05 to 50 ng mL −1 , with a detection limit of 0.017 ng mL −1 . The accuracy of sensor is similar with high-performance liquid chromatography (HPLC) analysis.

Light-addressable electrochemical immunoassay for multiplexed detection of antigen

Jian Wang,Fangming Chen,Qiaoyu Yang,Yao Meng,Mingrui Jiang,Yaqiong Wang,De-Wen Zhang,Liping Du

Multi-channel evaluation on a single sensor interface is highly valuable but full of challenge. Herein, a universal light-addressable electrochemical immunoassay based on silicon photoelectrodes was proposed for spatially-resolved detection of prostate-specific antigen (PSA) as a model target. To stabilize and functionalize silicon substrate, a simple and robust antifouling coating was prepared by cross-linking bovine serum albumin (BSA) matrix with glutaraldehyde (GA). For the PSA detection, an electrochemical sandwich-type immunosensor was performed on the basis of [email protected] functionalized silicon, in which horse radish peroxidase (HRP) was used to catalyze the reaction of H 2 O 2 and hydroquinone (HQ), resulting in in situ generation of benzoquinone (BQ). The light-induced local reduction photocurrent of BQ was then recorded as the sensor response by irradiating the target spot of photoelectrode respectively with a moveable light pointer. The results demonstrated the feasibility of localized measurements of enzymatic reactions . Coupling with light-addressable electrochemistry (LAE) and enzyme-linked immunosorbent assay (ELISA) protocol, multiplexed PSA detection could be achieved on a single sensor chip with good sensitivity and specificity. This proof-of-concept study provides a promising platform for high-throughput and accurate immunoanalysis in diagnostic or biodefense applications.

End-labeling-based electrochemical strategy for detection of adenine methylation in nucleic acid by differential pulse voltammetry

Yang Hongmei,Wang Yafen,Tang Jing,Wang Fang,Chen Zilin

A promising electrochemical strategy for assay of N6-methyladenosine (m6A)/N6-methyladenine (6mA) in RNA/DNA is proposed. The key of this strategy is the end-labeling of nucleic acid, which makes it possible to detect methylation level in unknown sequence. Firstly, the end of m6A-RNA or 6mA-DNA was labeled with sulfhydryl group through T4 polynucleotide kinase (T4 PNK) and then directly assembled on a gold nanoparticle–modified glassy carbon electrode (AuNPs/GCE). Secondly, methylation sites in RNA/DNA were specifically recognized by anti-m6A-antibody, and then, horseradish peroxidase–labeled goat anti-rabbit IgG (HRP-IgG) was further conjugated on the antibody. Thirdly, HRP-IgG catalyzed the hydroquinone oxidation reaction to generate amplified current signal which correlates with the amount of m6A/6mA in nucleic acid. This method showed a wide linear range from 0.0001 to 10 nM for m6A-RNA, 0.001 to 100 nM for 6mA-dsDNA, and 0.0001 to 10 nM for 6mA-ssDNA. The method was successfully applied to detection of m6A/6mA in RNA/DNA from HeLa cells and E. coli cells and validation of the decrease of m6A-RNA in HeLa cells after treatment with FTO protein. Microfluidic fabrication of tunable alginate-based microfibers for the stable immobilization of enzymes

Wen Zhang,Wenbo Ye,Yajun Wang,Yunfeng Yan

Immobilized enzymes have drawn extensive attention due to their enhanced stability, easy separation from reaction mixture, and prominent recyclability. Nevertheless, it is still an ongoing challenge to develop potent immobilization techniques which are capable of stable enzyme encapsulation, minimal loss of activity, and modulability for various enzymes and applications. These microfibers were able to efficiently encapsulate bovine serum albumin (BSA), glucose oxidase (GOx), and horseradish peroxidase (HRP). But the physically adsorbed enzymes readily diffused into the catalytic reaction system. The leakage of enzymes could be substantially inhibited by conjugating to poly(acrylic acid) (PAA) and incorporating into alginate-based microfibers, enabling stable immobilization, improved recyclability, and enhanced thermostability. In addition, GOx and HRP-loaded microfibers were fabricated under the optimized conditions for the visual detection of glucose using the cascade reaction of these enzymes, showing sensitive color change to glucose with concentration range of 0–2 m m . Due to the tunability and versatility, this microfluidic-based microfiber platform may provide a valuable approach to the enzyme immobilization for the cascade catalysis and diagnoses with multiple clinical markers. Graphical and Lay Summary Development of potent techniques for the immobilization of enzymes with stable encapsulation, minimal loss of activity, and good reusability is an ongoing challenge. In this study, authors developed co-flow microfluidic devices for the fabrication of alginate-based microfibers with tunable size and crosslinking degree for the stable immobilization of glucose oxidase and horseradish peroxidase. This microfluidic platform has great potential in the construction of customizable enzyme-loaded microfibers for the cascade catalysis and diagnoses with multiple clinical markers.
