The increasing drug resistance of bacteria due to the inappropriate use of various antibiotics has become an ongoing public health problem around the world. In this work, a novel Cu-hemin nanosheet with multilayer structure was synthesized by a simple green method. Integrating the peroxidase properties of hemin with the excellent photothermal conversion performance of IR780, the as-prepared Cu-hemin@IR780 can not only effectively catalyze hydrogen peroxide to produce reactive oxygen species (ROS), but also has a highly efficient and stable photothermal effect for sterilization. The antibacterial efficiency against MRSA can reach up to 98.96% by using chemodynamic therapy (CDT) and photothermal therapy (PTT) for synergistic sterilization.

Bisphenol S (BPS), a common replacement for bisphenol A (BPA) in packaging and food containers, has posed a threat to human health through dietary consumption. In this study, the effects of BPS binding on the redox states and stabilities of hemeproteins were investigated. Spectroscopy and molecular docking indicated that BPS could cause conformational alterations of hemoglobin (Hb) and the conversion of ferrous Hb to ferric Hb, which subsequently led to increased liberation of free hemin. Next, free hemin significantly caused cell membrane damage, reactive oxygen species formation, lipid peroxidation, and a decline of cell viability in endothelial cells. The selective inhibitors of ferroptosis significantly suppressed hemin-induced toxicity, indicating a hemin-mediated ferroptotic cell death mechanism. Our findings illustrate the binding mechanism of BPS with the hemeprotein as well as the cytotoxicity, which have important implications for the environmental and human health impacts of BPA replacements.

A foam consisting of reduced graphene oxide was synthesized by a one-pot hydrothermal method. The foam was used to prepare a nanocomposite with hemin which is formed via π-interactions. The nanocomposite was incorporated via a Nafion film and then placed on a glassy carbon electrode (GCE). The modified GCE displays outstanding catalytic activity towards H 2 O 2 . It is assumed that this is due to (a) the redox-active center [Fe(III/II)] of hemin, and (b) the crosslinked macroporous structure of the foam. Both improve the electron transfer rate and electrochemical signals. Under the optimum experimental conditions and a working voltage of typically −0.41 mV (vs. SCE), the sensor has a 2.8 nM H 2 O 2 detection limit, and the analytically useful range extends from 5 nM to 5 mM with a sensitivity of 50.5 μA μM −1 cm −2 . The modified GCE has high sensitivity and fast response. It was utilized to quantify H 2 O 2 in spiked environmental water samples.

Mitochondrial damage mediated by reactive oxygen species (ROS) is a major contributory factor to Parkinson’s disease (PD) pathogenesis. The use of hemin-based nanomaterials to control ROS production can protect against neurological damage. Here, we report the rational design of a uniform and water-soluble artificial nanozyme based on an ultrathin graphitic carbon nitride (g-C 3 N 4 ) nanosheet with loaded hemin to reduce ROS and treat PD. The g-C 3 N 4 not only acted as a scaffold for hemin loading but also improved the stability, cytotoxicity, and catalytic action of hemin. The integration of g-C 3 N 4 increased hemin’s peroxidase activity by 31.7%, promoting effective oxidation of the substrate 3,3′,5,5′-tetramethybenzichne (TMB) in the presence of H 2 O 2 to a blue-colored solution after 10 min’ incubation at room temperature. Moreover, CN-hemin exhibited good biocompatibility and could effectively scavenge intracellular ROS and reduce cytotoxicity induced by H 2 O 2 and 6-hydroxydopamine (6-OHDA). In vivo animal studies using nematode transgenic hus111 strain and 6-OHDA pretreated C. elegan s wild-type Bristol (N2) as models further showed that CN-hemin extended their lifespan and restored dopamine-dependent behaviors such as area-restricted searching, ethanol avoidance, and the basal slowing response by reducing ROS-mediated neurotoxicity. Nanomaterials using CN-hemin enzyme mimicry have potential biomedical applications, including the treatment of PD and disorders related to ROS metabolism.

Constructing porous carbon adsorbents with SF 6 -matched ultramicropores is an effective route for recovering SF 6 from nitrogen-rich gas streams. Herein, hemin was introduced into ZIF-8 precursors to regulate pyrolysis-induced pore evolution and produce Fe,N-containing porous carbons without corrosive KOH activation. The optimized Fe,N-C-1000 displayed a large BET surface area of 1772.60 m 2 ·g −1 along with abundant ultramicropores mainly distributed at 0.5–0.65 nm, providing suitable confined spaces for adsorption of SF 6 . The Fe,N-C-1000 delivered an SF 6 adsorption capacity of 4.58 mmol·g −1 and an IAST selectivity of 222 at 298 K and 1.0 bar. Fixed-bed breakthrough tests verified the preferential retention of SF 6 over N 2 , as well as good regenerability and cycling stability. Structure-performance analysis suggests that the improved separation performance is mainly governed by hemin-directed ultramicropore formation, while the Fe/N-containing surface environment contributes additionally to SF 6 affinity. DFT calculations further support stronger SF 6 interaction with the Fe,N-C model than with the undoped carbon model. These results demonstrate that hemin-assisted pyrolysis provides a feasible KOH-free strategy for constructing MOF-derived ultramicroporous carbons for the capture and recovery of SF 6 .

A facile sequential crystallization strategy is developed to fabricate isostructural HOF-on-HOF architectures with periodic interfacial lattice contrast. • Spatially stepwise immobilization of hemin across core and shell regions extends visible-light harvesting and provides reversible Fe(III)/Fe(II) redox sites. • The optimized LPFC-1@hemin@NPFC-1@hemin catalyst achieves a hydrogen evolution rate of 2345.1 μmol·g −1 ·h −1 under visible light (λ ≥ 420 nm), 1.84-fold higher than pristine LPFC-1. • The synergistic effect of interfacial charge regulation and hemin sensitization opens a new avenue for high-performance HOF-based photocatalyst design. Hydrogen-bonded organic frameworks (HOFs) have emerged as promising photocatalysts for sustainable hydrogen production, yet their performance suffers from intrinsically poor visible-light absorption and sluggish photogenerated carrier transfer kinetics. To address these issues, we herein report a facile sequential crystallization strategy to construct isostructural HOF-on-HOF materials, wherein a periodic interfacial lattice contrast spontaneously forms at the core-shell boundary. Moreover, this kind of structure further enables stepwise immobilization of hemin molecules in both the core and shell domains via hydrogen bonding and π···π stacking interactions. Benefiting from the synergistic effects between periodic interfacial lattice contrast and hemin immobilization, the optimal LPFC-1@hemin@NPFC-1@hemin (large-sized pyrene-based hydrogen-bonded organic frameworks, denoted as LPFC-1, nano-sized pyrene-based hydrogen-bonded organic frameworks, denoted as NPFC-1) catalyst exhibits an exceptional photocatalytic hydrogen evolution rate of 2345.1 μmol·g −1 ·h −1 under visible light irradiation (≥ 420 nm), which is 1.84 times higher than that of pristine LPFC-1. This work not only establishes a robust strategy to construct continuous HOF crystalline layers on preformed HOF crystals, but also presents an efficient route for stepwise hemin immobilization toward boosted photocatalytic hydrogen evolution. A stepwise layered growth strategy constructs well-defined HOF-on-HOF materials and achieves spatial immobilization of hemin for superior photocatalytic hydrogen generation. In this paper, using a strong coordination of the multi-amino groups of polyethylenimine (PEI)/SiO 2 nanoparticles with Cu 2+ and the weaker carboxyl-amine interactions with heme chloride (hemin), novel Cu 2+ -hemin dual-functionalized PEI/SiO 2 nanoparticles (abbreviated as Cu 2+ /hemin/PEI/SiO 2 NPs) were conveniently synthesized. The Cu 2+ /hemin/PEI/SiO 2 NPs exhibit both the peroxidase-like activity of hemin and the electrochemical signaling properties of Cu 2+ . Moreover, a large number of hemin molecules were successfully loaded onto Cu 2+ /PEI/SiO 2 NPs through electrostatic interactions. This not only prevented their self-aggregation-induced inactivation in aqueous solutions, but also catalytically amplified the electrooxidation signal of Cu 2+ /PEI/SiO 2 NPs by shortening the electron transfer distance. Subsequently, these nanoparticles spontaneously assembled on the overoxidized polypyrrole-modified reduced graphene oxide (abbreviated as OPPy/rGO) electrode surface through Cu N coordination between OPPy and Cu 2+ , and π-π stacking interactions between hemin and rGO. As a novel immunosensing platform, a label-free electrochemical immunosensor for carbohydrate antigen-125 (CA125), a specific biomarker for epithelial ovarian cancer was developed by covalently immobilizing CA125 capture antibodies onto the Cu 2+ /hemin/PEI/SiO 2 NPs@OPPy/rGO hybrid nanomaterials. Elevated serum CA125 levels correlate significantly with ovarian cancer progression. With the increasing concentration of CA125 antigen, the current responses of this immunosensor were decreased due to the formation of antigen-antibody insulating immune complexes. The proposed immunosensor exhibited excellent performances within a wide linear range from 5 × 10 −5 to 200 U·mL −1 for CA125 and a low detection limit of 1.2 × 10 −5 U·mL −1 (S/N = 3). Additionally, this immunosensor demonstrated good stability, reproducibility, and selectivity, making it a promising tool in clinical screening, early diagnosis, and prognostic monitoring of ovarian cancer.

Cryptococcosis, a serious systemic fungal infection caused by Cryptococcus neoformans ( C. neoformans ) and its variants, poses a significant clinical challenge due to its poor prognosis and severe health implications. The treatment of cryptococcal infections is complicated by several unique factors, stemming from both the pathogenic characteristics of the fungi and the biological barriers they exploit. These include the fungi's protective capsule, their ability to reside within host macrophages—thereby evading pharmacological intervention—and their involvement in multi-organ infections such as the lung and brain, in particular their strategic positioning within the brain, protected by the blood-brain barrier (BBB). To overcome these obstacles, precise active targeting emerges as a pivotal strategy. Identifying common targets is imperative to enhance therapeutic efficacy while ensuring the druggability of delivery systems. However, research on the methodology for selecting such shared targets remains sparse. In our investigation, we have pioneered the use of secreted proteins as shared target to trace the pathogens and their infection pathways. We identified the mannoprotein Cig1, prominently expressed on the surfaces of infected macrophages, lungs, and brains, as a viable shared target. On this basis, we utilized Hemin, a ligand for Cig1, to design liposomes (Hemin Lip) tailored for addressing complex fungal infections. By leveraging the interaction with the secreted protein Cig1, Hemin Lip specifically identifies and binds to organs and macrophages harboring cryptococcal infections, thereby facilitating targeted and efficacious clearance of both intracellular and extracellular fungus. Moreover, we have extended this targeting mechanism to other nanomedicinal platforms, including albumin nanoparticles. This study proposes an innovative drug delivery model that targets extracellular secretory proteins within the infection microenvironment, offering a streamlined formulation with the potential for effective therapy against complex infections.

The high-residual and bioaccumulation property of organophosphorus pesticides (OPs) creates enormous risks towards the ecological environment and human health, promoting the research for smart adsorbents and detection methods. Herein, 2D hemin-bridged MOF nanozyme (2D-ZHM) was fabricated and applied to the efficient removal and ultrasensitive dual-mode aptasensing of OPs. On the one hand, the prepared 2D-ZHM contained Zr-OH groups with high affinity for phosphate groups, endowing it with selective recognition and high adsorption capacity for OPs (285.7 mg g −1 for glyphosate). On the other hand, the enhanced peroxidase-mimicking biocatalytic property of 2D-ZHM allowed rapid H 2 O 2 -directed transformation of 3,3′,5,5′-tetramethylbenzidine to oxidic product, producing detectable colorimetric or photothermal signals. Using aptamers of specific recognition capacity, the rapid quantification of two typical OPs, glyphosate and omethoate, was realized with remarkable sensitivity and selectivity. The limit of detections (LODs) of glyphosate were 0.004 nM and 0.02 nM for colorimetric and photothermal methods, respectively, and the LODs of omethoate were 0.005 nM and 0.04 nM for colorimetric and photothermal methods, respectively. The constructed dual-mode aptasensing platform exhibited outstanding performance for monitoring OPs in water and fruit samples. This work provides a novel pathway to develop MOF-based artificial peroxidase and integrated platform for pollutant removal and multi-mode aptasensing. Graphical Abstract

Herein, we propose an all-DNA self-assembled G-quadruplex/hemin DNAzyme nanosphere (GHN) and apply it to construct an electrochemical biosensor for sensitive detection of kanamycin (KAN). Unlike conventional metal-core-dependent biosensors, the GHN eliminates inorganic components, offering superior biocompatibility, simplified synthesis, and reduced environmental hazards. High loading of G-rich sequences was achieved by performing multiple branches of the hybridization chain reaction on self-assembled DNA spherical core. In addition to exhibiting excellent catalytic performance, the GHN inherently provides a strong current response, facilitating highly sensitive detection of target analytes. Through a one-step displacement reaction triggered by the target, the GHN is captured on the electrode surface, generating a measurable current signal. Under optimal experimental conditions, the electrochemical signal of the proposed sensor showed a good linear relationship with the logarithm of the KAN concentration over the range of 0.1 pM to 1 μM, with a detection limit of 71 fM (S/N = 3). For practical applications, the sensor achieved 93.98% to 102.0% recovery in spiked milk samples, demonstrating its reliability in complex matrices. The target-independent synthesis of GHN allows facile reconfiguration by altering aptamer sequences, enabling universal detection of environmental pollutants and biomarkers. This platform offers a new approach for early monitoring of antibiotic residues, which is expected to provide valuable references for the formulation of relevant environmental protection policies and the safeguarding of human health.

Kanamycin (KANA) residue in meat is particularly harmful to public health and there is an urgent need to establish a fast, accurate and low-cost method to determinate KANA in food quality control. In this paper, hemin–reduced graphene oxide-carboxylated multiwalled carbon nanotubes (hemin–rGO–cMWCNTs) were designed and prepared, and the characteristics of hemin–rGO–cMWCNTs are presented. After that, an aptamer/hemin–rGO–cMWCNTs sensor for determination of KANA was developed. The electrochemical characteristics were studied by cyclic voltammetry (CV), differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS). Under optimal conditions, the sensitive response of the aptasensor towards KANA presented a wide concentration range of 10−9 to 10−6 M and a low detection limit of 0.36 nM (S/N = 3). Meanwhile, the aptasensor showed prominent selectivity, high stability and acceptable reproducibility in the application of KANA detection. In addition, the aptasensor detection in real samples correlated well with that obtained by liquid chromatograph mass spectrometer (LCMS).

A short G-rich sequence with three G-tracts can form a G-triplex (G3), a recently identified noncanonical DNA structure . Until now, very limited functional study and application of G3 is reported. Herein, we integrated G3 with isothermal exponential amplification reaction (EXPAR) for achieving simple and sensitive biosensing strategy. In this strategy, the cascade EXPAR cycles produces larger numbers of short G-rich sequences, which self-assemble into G3 structure and then bind hemin to form G3/hemin DNAzyme. This G3/hemin DNAzyme–based EXPAR strategy could detect as low as 4.7 fM target DNA with colorimetric detection, and the sensitivity of G3/hemin DNAzyme–based EXPAR strategy was much higher than that of the conventional G-quadruplex/hemin DNAzyme–based EXPAR strategy. We explored the reason for higher sensitivity of G3/hemin DNAzyme–based EXPAR strategy. The experimental results demonstrated that G3/hemin DNAzyme is an ideal signal generator for EXPAR-based biosensing platform. This work opens a new avenue to develop effective signal amplification strategy for ultrasensitive biosensing. This work is also helpful for a deeper understanding of G3 structure and the future application of G3.
