Background Cardiac hypertrophy, characterized by the thickening of the heart muscle, arises from factors such as hypertension and genetic mutations, often leading to adverse outcomes like heart failure and arrhythmias. Guanxinning tablets (GXNT), a botanical drug composed of the blood-activating herbs Salvia miltiorrhiza Bunge. and Ligusticum striatum DC., are widely used in the treatment of cardiovascular diseases. However, the active ingredients and their molecular mechanisms are yet to be fully understood. Methods We evaluated the anti-hypertrophic effects of GXNT and screened its active substances via cardiac function live-imaging on an aristolochic acid A-stimulated zebrafish cardiac hypertrophy model, and through F-actin immunostaining on a phenylephrine-induced hypertrophic NRCMs model. Additionally, the protective effects of GXNT’s active substances were analyzed in a mouse model of cardiac hypertrophy using echocardiography, histopathology analysis, and western blotting. Results The anti-hypertrophic effects of GXNT were assessed using an aristolochic acid A-stimulated zebrafish model and phenylephrine-induced hypertrophic NRCMs. GXNT demonstrated significant anti-hypertrophic effects in both models. Phenotypic screening identified Senkyunolide I (Sen I) from Ligusticum striatum as the active component in the zebrafish model, while Salvianolic acid B (Sal B) and Rosmarinic acid from Salvia miltiorrhiza emerged as the key anti-hypertrophic compound in NRCMs. In a mouse model of isoproterenol-induced cardiac hypertrophy, Sal B and Sen I showed synergistic effects, improving cardiac function, reducing oxidative stress, and suppressing inflammation. Mechanistically, transcriptomic sequencing highlighted cooperative modulation of MAP3K1 signaling by the two compounds. Notably, siRNA-mediated knockdown of MAP3K1 in cardiomyocytes attenuated the hypertrophic phenotype, supporting its essential role in the pathological process. Molecular docking and dynamic simulations further supported their binding potential to MAP3K1. Conclusion These findings underscore GXNT’s potent anti-hypertrophic effects, possibly driven by the synergistic actions of Sal B and Sen I, and offer insights into its therapeutic potential through MAP3K1 signaling regulation.

The oxidation of thallium [Tl(I)] to Tl(III) by chlorine (HOCl) is an important process changing its removal performance in water treatment. However, the role of bromide (Br–), a common constituent in natural water, in the oxidation behavior of Tl(I) during chlorination remains unknown. Our results demonstrated that Br– was cycled and acted as a catalyst to enhance the kinetics of Tl(I) oxidation by HOCl over the pH range of 5.0–9.5. Different Tl(I) species (i.e., Tl+ and TlOH(aq)) and reactive bromine species (i.e., HOBr/BrO–, BrCl, Br2O, and BrOCl) were kinetically relevant to the enhanced oxidation of Tl(I). The oxidation by free bromine species became the dominant pathway even at a low Br– level of 50 μg/L for a chlorine dose of 2 mg of Cl2/L. It was found that the reactions of Tl+/BrCl, Tl+/BrOCl, and TlOH(aq)/HOBr dominated the kinetics of Tl(I) oxidation at pH < 6.0, pH 6.0–8.0, and pH > 8.0, respectively. The species-specific rate constants for Tl+ reacting with individual bromine species were determined and decreased in the order: BrCl > Br2 > BrOCl > Br2O > HOBr. Overall, the presented results refine our knowledge regarding the species-specific reactivity of TI(I) with bromine species and will be useful for further prediction of thallium mobility in chlorinated waters containing bromide.

Cu-based catalysts attract considerable attention because of their exceptional CO 2 photo-/electro-/thermo-reduction capacities, where Cu(I) is generally treated as the active species. However, a significant problem hindering the large-scale applications of Cu-based catalysts is the inactivation of Cu(I) via irreversible redox to Cu(II)/Cu(0). This study proposes a Cu(I) valence pinning method based on hydroxyapatite (HAP). Experimental and theoretical studies demonstrate that the phonon resonance among the Cu(I) ions, their adjacent heteroatoms, and the intermediates adsorbed at the Cu(I) sites yields Cu(I) valence electrons in their lowest energy states. Thus, Cu(I) is stabilized, stable, and efficient photothermal CO 2 hydrogenation is promoted. However, because of the change of Cu(I) coordination environment during the CO 2 hydrogenation reaction, Cu(I) ions migrate into the bulk phase, leading to activity attenuation. Nevertheless, Cu(I) ions can be pulled out to the surface of HAP under the oxidative humid air condition, and the catalytic activity can be easily recovered. Thus, we propose a simple cyclic reaction/regeneration process. This enables the Ca 5 (FeCuCe) 5 -HAP catalyst to achieve the CO yield of 402.8 mmol g −1 h −1 and a CO 2 conversion rate of 27.7%, which is close to the thermodynamic equilibrium. This catalyst also displays a selectivity of approximately 100% and cycle stability of 156 h at 500 °C under a pressure of 1 atmospheric. Our study provides a viable method for the scale applications of Cu(I) based catalysts in the “negative carbon” industries.

High efficiency copper(I) halide complexes containing rigid diphosphine ligands have attracted much attention. However, rigid bidentate phosphine ligands have been rarely reported so far. Here, an electron-rich heteroaryl ring grafted with two electron donating methyl groups was first included to construct a rigid bidentate phosphine ligand, three four-coordinate dinuclear copper(I) halide complexes, [CuX(dpmt)]2 (dpmt = 3,4-bis(diphenylphosphino)-2,5-dimethylthiophene, X = I (1), Br (2) and Cl (3)), were synthesized, and their molecular structures and photophysical properties were investigated. The structural analysis reveals that two copper(I) centers are bridged by two halogen ligands to form a dinuclear structure with a four-membered Cu2X2 ring. These complexes exhibit intense bluish green to yellow green emission in the solid state at room temperature and have peak emission wavelengths at 490–543 nm with microsecond lifetimes (τave = 3.9–13.6 μs, Φ = 0.26–0.35). The emission of complex 1 mainly originates from MLCT and ligand-centered transitions. Similar emissive behaviors for complexes 2 and 3 are possibly due to ligand-centered transition. The solution-processed devices of complex 3 exhibit stable yellow emission with CIE(x, y) of (0.42, 0.52) for doped and nondoped devices.

The measurement of cardiac troponin-I (cTnI) is widely used for diagnosing acute myocardial infarction (AMI) diseases because of its myocardial specificity. Packaged microbubble resonators with thin wall are utilized for label-free and specific detection of cTnI based on whispering gallery mode (WGM). This packaged structure can provide a good protection for the biosensor, improve the anti-interference ability of the sensor and reduce the system noise. The theoretical detection limit of the biosensors for cTnI in phosphate buffer saline (PBS) is 0.4 ag mL −1 (0.02 a M ). Furthermore, we demonstrated that the biosensors can be used to detect cTnI molecules in simulated serum and the theoretical detection limit is also 0.4 ag mL −1 (0.02 a M ). These results are much far below the clinical cut-off value and show a huge application potential for the detection of cardiac biomarkers of AMI.

As a highly toxic rare metal, the removal of thallium (Tl) from wastewater has been widely investigated, and adsorption is considered one of the most promising treatment technologies for Tl-containing contaminated water because of its cost-effectiveness, convenience, and high efficacy. In this work, coal tar residue (CTR)-based porous carbon was synthesized through K 2 FeO 4 activation, and applied in adsorbing Tl(I). K 2 FeO 4 could synergistically produce porosity and load iron oxide on the produced porous carbon surface because of the catalytic cracking and oxidative etching during the activation of CTR. The adsorbent was synthesized at 800 ℃ with a mass ratio of K 2 FeO 4 /CTR being 3 (PC 3–800 ) showed optimal Tl(I) adsorption performance. The removal efficiency and distribution coefficient of PC 3–800 were above 95 % and 10 4 mL/g, respectively, in a wide pH range (4−10). Furthermore, the selection and reusability of PC 3–800 were favorable. The adsorption was a spontaneous, exothermic, and entropy increase process. The adsorption process was dominated by electrostatic attraction, surface complexation, and surface oxidation . The results suggested that removing Tl(I) from contaminated water via CTR-based porous carbon was feasible.

Myocardial infarction occurs rapidly, and thus the rapid detection of cTnI levels is the key to its diagnosis. Most current assays take 10–30 min. In this study, we developed a method for accurately measuring cardiac troponin I (cTnI) levels in human sera with amplified luminescence neighborhood homogeneous assay (AlphaLISA). The method involves coupling two cTnI antibodies targeting different epitopes to the surface of carboxylated donor and acceptor beads. The final signal values were detected by the double-antibody sandwich method, and the best reaction conditions were obtained by optimizing the experimental conditions. The sensitivity, specificity, accuracy, and precision of the method were evaluated. Results showed that the method requires only 3 min to produce the results, the detection sensitivity is 27.06 ng L−1, and the measurement range is 34.56–62 500 ng L−1. cTnI-AlphaLISA has an intra-assay precision of 2.18–4.57% (<10%) and an inter-assay precision of 5.60–6.95% (<10%). The relative recovery rates are within reasonable limits. In addition, the serum assay results of the method were compared with chemiluminescence immunoassay, and the results are in agreement with one another (ρ = 0.8803; P < 0.0001). The method is expected to be developed as a routine method, but further studies and evaluations are needed.

The utilization of bifunctional ligands, specifically pyridine carboxylic acids, endowed with dual coordination sites, has been instrumental in the assembly of polymer materials. The ambidentate characteristics of these ligands play a crucial role in shaping the structure and framework of cluster-based polymers. In this study, we have synthesized a diverse array of multidimensional copper(I) alkynyl cluster-based polymers (CACPs) by employing four distinct pyridine carboxylic acids – namely, isonicotinic acid (INA), 6-isoquinolinecarboxylic acid (IQL), 4-pyridin-4-yl-benzoic acid (4-PyBA), and 3-pyridin-4-yl-benzoic acid (3-PyBA) – as linking ligands. These pyridine carboxylic acids not only serve as protective ligands but also act as pivotal linkers in constructing the cluster-based framework materials, exerting significant influence on the overall framework structures. Furthermore, the incorporation of auxiliary ligands has been shown to markedly impact the structural integrity and framework architecture of the CACPs. This study elucidates the indispensable role of pyridine carboxylic acids in the construction and stabilization of cluster-based framework materials, thereby advancing the frontier of research in metal cluster-based framework material synthesis.

Highly emissive copper halide complexes with a bidentate phosphine ligand have been paid much attention. Here, a series of four-coordinate mononuclear copper(I) halide complexes containing bi- and mono-dentate phosphine ligands, [CuX (dpmb)(PPh 3 )] (dpmb = 1,2-bis(diphenylphosphino)–4,5-dimethylbenzene, X = I ( 1 ), Br ( 2 ) and Cl ( 3 )), were synthesized, and their molecular structures and photophysical properties were investigated. These complexes exhibit intense blue emission in the solid state at room temperature and have peak emission wavelengths at 464–479 nm with microsecond lifetimes ( τ = 3.5−4.9 μs; Φ = 0.23−0.52). The emission of the complexes 1–3 mainly originates from MLCT, XLCT and intraligand transitions. Three complexes display moderate thermal stability.

Summary By leveraging a Cu(OH)F precursor, we construct a disk-like Cu(I)/Cu(0) interface model (P-Cu x /Cu 2 OF) composed of F-stabilized Cu(I) shell and Cu(0) core by one-step pulsed potential conversion. The interfacial region is shown to facilitate ethanol formation, guaranteeing a high FE(C 2+ ) of up to 80.2%. Tuning the Cu(I)/Cu(0) ratio enhances the ethanol yield by 18.5-fold over pure P-Cu, with the corresponding partial current density (j ethanol ) reaching up to 128 mA/cm 2 . This is enabled by a 3.6-fold boost in selectivity for the ethanol product (35.4%) and an overall 7.8-fold boost in activity ( j C 2 + up to 288 mA/cm 2 ). The interactions between P-Cu and Cu 2 OF upshift the d-band center of Cu sites on the interface region, which is critical to asymmetrical C-C coupling to form OCCOH∗, concurrent with a lowered energy barrier for ethanol formation. On the other hand, the ethylene-forming symmetrical C-C coupling occurs primarily on P-Cu away from the Cu(I)/Cu(0) interface.

Heparinase I (EC 4.2.2.7), is an enzyme that cleaves heparin, showing great potential for eco-friendly production of low molecular weight heparin (LMWH). However, owing to its poor catalytic activity and thermal stability, the industrial application of heparinase I has been severely hindered. To improve the catalytic activity, we proposed to engineer both the substrate and Ca2+ binding domains of heparinase I. Several heparinases I from different organisms were selected for multiple sequence alignment and molecular docking to screen the key residues in the binding domain. Nine single-point mutations were selected to enhance the catalytic activity of heparinase I. Among them, T250D was the most highly active one, whereas mutations around Ca2+ binding domain yielded two active mutants. Mutant D152S/R244K/T250D with significantly increased catalytic activity was obtained by combined mutation. The catalytic efficiency of the mutant was 118,875.8 min−1·µM−1, which was improved 5.26 times. Molecular modeling revealed that the improved activity and stability of the mutants were probably attributed to the formation of new hydrogen bonds. The highly active mutant had great potential applications in industry and the strategy could be used to improve the performance of other enzymes.

In this study, the copper-nickel (Cu–Ni) bimetallic electrocatalysts for electrochemical CO 2 reduction reaction(CO 2 RR) are fabricated by taking the finely designed poly(ionic liquids) (PIL) containing abundant Salen and imidazolium chelating sites as the surficial layer, wherein Cu–Ni, PIL–Cu and PIL–Ni interaction can be readily regulated by different synthetic scheme. As a proof of concept, Cu@Salen-PIL@Ni(NO 3 ) 2 and Cu@Salen-PIL(Ni) hybrids differ significantly in the types and distribution of Ni species and Cu species at the surface, thereby delivering distinct Cu–Ni cooperation fashion for the CO 2 RR. Remarkably, Cu@Salen-PIL@Ni(NO 3 ) 2 provides a C2+ faradaic efficiency (FE C2+ ) of 80.9% with partial current density ( j C 2+ ) of 262.9 mA cm −2 at −0.80 V (versus reversible hydrogen electrode, RHE) in 1 m KOH in a flow cell, while Cu@Salen-PIL(Ni) delivers the optimal FE C2+ of 63.8% at j C2+ of 146.7 mA cm −2 at −0.78 V. Mechanistic studies indicates that the presence of Cu–Ni interfaces in Cu@Salen-PIL@Ni(NO 3 ) 2 accounts for the preserve of high-valence Cu(I) species under CO 2 RR conditions. It results in a high activity of both CO 2 -to-CO conversion and C–C coupling while inhibition of the competitive HER.
