Volumetric additive manufacturing has emerged as a promising technique for the flexible production of complex structures, with diverse applications in engineering, photonics and biology 1 , 2 . However, present methods still face a trade-off between resolution and volumetric build rate, restricting efficient and flexible production of high-resolution 3D structures. Here we propose a method, called digital incoherent synthesis of holographic light fields (DISH), to generate high-resolution 3D light distributions through continuous multi-angle projections with a high-speed rotating periscope without the requirement of sample rotation. The iterative optimization of the holograms for different angles in DISH maintains 19-μm printing resolution across the 1-cm range that is far beyond the depth of field of the objective and enables high-resolution in situ 3D printing of millimetre-scale objects within only 0.6 s. Acrylate materials in a range of viscosities are used to demonstrate the general compatibility of DISH. Integrating DISH with a fluid channel, we achieved mass production of complex and diverse 3D structures within low-viscosity materials, demonstrating its potential for broad applications in diverse fields.

Prolonged exposure to visible light can disrupt tear-film stability and compromise ocular lubrication, potentially leading to eye fatigue, dryness, and the development of dry-eye disease, thereby highlighting the need for durable and long-lasting ocular lubrication. In this study, we develop a photoresponsive double-network hydrogel (PDNH) for the ocular environment that addresses these issues and achieves superlubricity. The PDNH is composed of a polyvinylpyrrolidone (PVP) hydrogel framework and a responsive supramolecular system of β-cyclodextrins/polyethylene glycol (β-CD/PEG) and competitive guest azobenzene derivative (Azo-C2). Under visible-light irradiation, the competitive host–guest interaction between the β-CD/PEG supramolecule and Azo-C2 lead to the release of the PEG chain ends from the β-CD cavities, allowing the PEG chains to diffuse toward the hydrogel surface. The strong hydrophilicity of PEG promotes water retention at the ocular surface, forming a stable lubricating layer that effectively reduces ocular friction. With continuous visible-light irradiation, the friction coefficient (COF) of the hydrogel gradually decreases and reaches its optimal state after 6 h of exposure, with the COF reduced by 25%, and its mechanical properties can remain at a high level. Its self-regulating lubrication capability minimizes dependence on frequent external lubricant administration. Moreover, its excellent biocompatibility highlights the promise as a smart ophthalmic lubricant, with the photoresponsive mechanism offering broad potential for biomedical systems and a novel route to ocular superlubricity in drug delivery.

To tackle low adsorption capacity and eco-unfriendly preparation of methylene blue adsorbents, this study fabricated three polyacrylonitrile (PAN)-based composite fiber membranes via a novel ultraviolet curing-electrospinning method, namely Type I (PAN/acrylic acid (AA)-hydroxypropyl acrylate (HPA)), Type II (PAN/AA-ethylene glycol (EG)), and Type III (PAN/(AA-polyacrylamide)-EG). After 5 cycles at an initial methylene blue concentration of 520 mg/L, the membranes retain over 80% of initial adsorption capacity, demonstrating excellent reusability. Material properties were characterized, and key adsorption-affecting parameters were studied. Results indicated the Type III membrane with 8% PAN + 9% prepolymer had the best comprehensive performance, achieving a maximum methylene blue adsorption capacity of 1420.84 mg/g, much higher than Type I (958.13 mg/g) and Type II (1159.35 mg/g). Type III membrane has continuous, uniform fibers without beaded defects and excellent thermal stability. The adsorption process is highly consistent with the pseudo-second-order kinetic model and Langmuir model, belonging to chemisorption. The mechanism is dominated by electrostatic attraction between carboxyl groups and methylene blue cation, supplemented by hydrogen bonding of amide groups, combined with a flexible cross-linked network structure. This verifies the “multi-functional group synergy-microstructure optimization” design strategy for high-performance adsorbents, providing a new scheme for the green preparation of efficient MB adsorbents.

Slow and insufficient sealing is a major problem for natural hydrogels to control incompressible hemorrhage. In this study, a natural macromer with both acrylamido and catechol groups was first designed and synthesized as a hydrogel adhesive precursor, and then the hydrogel was fabricated by photopolymerization. This design with multiple conjugated structures is observed to have the strong electron conjugation effect, i.e., n–π conjugation by UV-induced orbital coupling for electron delocalization and electronic orbital overlap between adjacent chains arising from UV photo-cross-linking as well as between uncosumed acrylamido groups. Owing to this synergistic effect, the natural polysaccharide-based hydrogel demonstrates a rapid gelation within 4.3 s and a significantly enhanced burst pressure with the value of 187.2 ± 21.4 mmHg, as well as strong tissue adhesion, thus providing a fast and stable physical barrier at the bleeding sites. Together with the accelerated clotting in vitro resulting from the promotion of erythrocyte adhesion by the hydrogel, they achieve rapid and efficient hemostasis of rat liver incision and perforation, and femoral artery perforation, which are far superior to the hydrogel with only acrylate groups, commercial gelatin sponge, and gauze. Besides, they are biocompatible and biodegradable via in vitro and in vivo evaluation. Such a molecular design and strategy are suitable for natural polymers with abundant hydroxyl groups, e.g., chitosan and hyaluronic acid, appearing to offer new ideas for the development of hemostatic materials in incompressible hemorrhage control.

The extracellular matrix (ECM) is a dynamic, hierarchically structured network essential for mechanical support and regulation of cell behavior. Mimicking its complexity in synthetic materials remains a major challenge. Here, we present a straightforward strategy to fabricate ECM-inspired, dynamically phase-separated hydrogels (DyPS-MAPgel) by methacrylation of mussel adhesive proteins (MAPs), followed by metal ion-induced phase separation and in situ photopolymerization. Leveraging the intrinsic ability of MAPs to undergo biomolecular phase separation and subsequent covalent crosslinking, the resulting hydrogels exhibit tunable mechanical properties, strong tissue adhesion, self-healing, antibacterial, and antioxidant activities. Importantly, DyPS-MAPgels support stem cell proliferation and lineage-specific differentiation both in vitro and in vivo through integrin-dependent mechanotransduction pathways. This work demonstrates the potential of phase separation–mediated assembly as a robust strategy for constructing ECM-mimetic materials and provides insights into ECM-cell interactions. Our findings offer a versatile platform for regenerative medicine applications, including bioadhesion, 3D bioprinting, and tissue engineering.

Diabetic wound healing is a global biomedical challenge. Bacterial infection and microenvironmental modulation are major factors in the refractory healing of diabetic wounds. Conventional therapeutic approaches are often hindered by limitations, such as inadequate drug penetration, the rise of antimicrobial resistance, and restricted depth of drug delivery. Nanomedicine is emerging as a prospective drug delivery and anti-infective therapeutic approach in the field of wound management. Herein, all-trans retinoic acid (ATRA) was loaded into a zeolite imidazolate framework-91 salt+ (ZIF-91 salt+). A gelatin methacryloyl (GelMA) hydrogel microneedles (MNs) patch based on this multifunctional ZIFs (denoted as GelMA-ZIF-91 salt+@ATRA MNs patch) was developed, which makes transdermal drug delivery and combinatory treatment for diabetic wound healing feasible. Significantly, ATRA-containing ZIF-91 salt+ provides nanocomposites with anti-inflammatory activity through their modulation of macrophage polarization. Crucially, the prepared ZIF-91 salt+ showed efficient antibacterial activity against Staphylococcus aureus and Escherichia coli in vitro. Therapeutic effects of the GelMA-ZIF-91 salt+@ATRA MNs patch were verified in a diabetic mouse model with full-thickness cutaneous wounds. A unique MNs patch with antimicrobial, angiogenesis-inducing, and anti-inflammatory properties was designed in accordance with the physiological characteristics of wounds. Collectively, the designed MNs patch based on this multifunctional ZIF-91 salt+ provides new insights into diabetic wound therapy.

Open abdomen (OA) is a life-saving surgical intervention commonly performed in patients with severe pancreatitis, intra-abdominal infection, or major trauma. However, abdominal exposure after OA inevitably leads to continuous fluid and heat loss, making temporary abdominal closure (TAC) indispensable in these cases. Conventional materials, such as polypropylene (PP) mesh, can prevent abdominal wall retraction. However, they may cause intestinal abrasions, which can lead to enteroatmospheric fistulas. Thus, the development of wound dressings that combine structural support and bioactivity is highly important. In this study, we developed a multifunctional composite hydrogel by combining gelatin methacryloyl (GelMA) with TEMPO-oxidized cellulose nanofibers (TC) and further crosslinking with zinc ions (Zn 2+ ) coordination to form Zn-TC-GelMA (Zn-TCG). The Zn-TCG hydrogel exhibited tunable mechanical properties, excellent injectability, and sustained Zn 2+ release. It exhibited good hemocompatibility, suppressed reactive oxygen species (ROS) accumulation, and inhibited bacterial proliferation. In vitro, Zn-TCG significantly enhanced fibroblast migration and HUVEC tube formation. In vivo, it markedly accelerated re-epithelization, collagen deposition, and angiogenesis compared to the control treated with GelMA and PP mesh. Mechanistic studies confirmed the activation of the MAPK/ERK signaling pathway and upregulation of epidermal differentiation markers (Krt10, Loricrin, and Tgm1). RNA-seq further revealed enrichment in keratinization and immune-related pathways. In conclusion, the Zn-TCG hydrogel represents a promising strategy for temporary abdominal closure and wound management.

Repairing spinal cord injury (SCI) remains hindered by two major unmet challenges: the inability to precisely reconstruct irregular lesion cavities, where tissue contraction and collapse disrupt anatomical continuity, and the lack of effective platforms for stable and localized delivery of fragile therapeutics such as exosomes, whose rapid clearance severely limits efficacy. Addressing these bottlenecks requires biomaterials that can simultaneously rebuild three-dimensional structure and orchestrate a pro-regenerative microenvironment. Here, we present a morphology-adaptive injectable microporous annealed particle scaffold (MS) for localized and long-term release of human umbilical cord MSC-derived exosomes (hucMSC-Exo). GelMA/SFMA microspheres were fabricated via solvent-free microfluidic electrospraying combined with liquid nitrogen-assisted cryo-processing, preserving exosome bioactivity. This “disassemble-and-reassemble” approach enables minimally invasive injection of microspheres into irregular lesion cavities, followed by body temperature-induced annealing and in situ photocuring to form a mechanically stable, interconnected three-dimensional scaffold, which supports cell infiltration, nutrient diffusion, and sustained exosome release. In vivo , MS@Exo suppressed inflammation and scar formation, enhanced angiogenesis, and promoted neuronal survival, axonal regeneration, and myelination, ultimately achieving superior functional recovery over MS or Exo alone, demonstrating a clear synergistic effect. Transcriptomic profiling further revealed that MS@Exo reprogrammed gene expression by downregulating extracellular matrix- and collagen-related pathways while upregulating neuronal, synaptic, and ion channel signaling networks, thereby shifting the repair process from an inflammatory-proliferative phase toward a differentiation-remodeling phase. Collectively, MS@Exo acts as an intelligent multifunctional scaffold that couples advanced fabrication with exosome-based therapy to overcome fundamental limitations in SCI repair, offering a broadly applicable strategy for reconstructing irregular tissue defects in regenerative medicine.

Efficient removal and rapid in situ monitoring of Cu(II) in wastewater are of great importance for aquatic environmental remediation, yet conventional adsorbents generally fail to simultaneously achieve high adsorption capacity and quantitative detection. In this study, a N-doped carbon dot composite hydrogel strategy integrating adsorption and fluorescence sensing was proposed. A three-dimensional hydrogel network rich in N/O-containing functional groups was constructed using gelatin methacryloyl (GelMA), acrylic acid (AAc), acrylamide (AAm), and nitrogen-doped carbon dots (NCDs) were further introduced to fabricate the bifunctional fluorescent composite hydrogel NCDs-GA 1 A 2 gel. This structure not only provided abundant coordination and electrostatic adsorption sites for Cu(II), but also endowed the material with optical responsiveness for fluorescence detection. Batch adsorption experiments showed that the Cu(II) adsorption process followed the pseudo-second-order kinetic model and the Langmuir isotherm model, indicating monolayer chemisorption, with a maximum theoretical adsorption capacity of 203.1 mg·g −1 . After ten adsorption-desorption cycles, the hydrogel retained 86.7% of its initial adsorption capacity, demonstrating favorable regeneration stability. Meanwhile, NCDs-GA 1 A 2 gel could serve as a fluorescent probe for Cu(II), showing a good linear fluorescence response to Cu(II) concentration and a low detection limit of 0.65 μM. Coexisting ion interference experiments further confirmed its excellent selectivity and anti-interference capability. This work provides a facile strategy for developing multifunctional biomass-derived fluorescent hydrogel adsorbents and demonstrates promising application potential for Cu(II) removal and in situ detection in wastewater

Background/Objectives: Renal ischemia–reperfusion injury (IRI) and infection-associated renal damage are characterized by persistent inflammation, oxidative stress, microvascular dysfunction, and impaired tissue regeneration, creating a hostile microenvironment that limits effective repair. We developed an injectable, photocrosslinkable methacrylated chitosan (CSMA) hydrogel for the localized co-delivery of cerium oxide nanoparticles (CeO2NPs) and vascular endothelial growth factor (VEGF), aiming to integrate redox modulation, antibacterial activity, and regenerative support. Methods:Gelation, microstructure, rheology, degradation, and CeO2NP/VEGF were characterized. Tubular epithelial and fibroblast migration and endothelial network formation, angiogenic gene expression, and antibacterial activity againstStaphylococcus aureusandEscherichia coliwere evaluated in vitro. Theraputic performance was assessed by renal surface application in a rat renal IRI model and catheter-mediated interavsical administration in an ascending urinary tract infection model. Systematic biocompatibility was evaluated separately in a 14-day subcutaneous implantation study. Renal response were further investigated using transcriptomic and targeted molecular analyses. Results:The CSMA/VEGF/CeO2NPs hydrogel exhibited rapid in situ gelation, interconnected porous architecture, stable viscoelasticity, gradual degradation, and sustained release of both CeO2NPs and VEGF. The formulation enhanced tubular epithelial and fibroblast migration, promoted endothelial network formation and angiogenic gene expression and effectively inhibited bothS. aureusandE. coli. In a surgically controlled rat renal IRI model, direct renal-surface application of the hydrogel reduced tubular injury, inflammatory infiltration, and fibrotic remodeling. In a separate ascending urinary tract infection model, catheter-based intravesical administration reduced the ascending renal bacterial burden and infection-associated inflammatory injury. No detectable adverse systemic effects observed under the tested conditions over the 14-day observation period in the subcutaneous implantation. Transcriptomic analyses further revealed that CSMA/VEGF/CeO2NPs treatment was associated with marked remodeling of the renal injury microenvironment, characterized by suppression of antigen presentation and immune activation pathways, alongside restoration of metabolic programs associated with amino acid, lipid, and purine metabolism. These molecular changes were accompanied by downregulation of CIITA/CD74/MHC-II signaling, recovery of metabolic regulators AGXT and ACOX1, modulation of Hippo/YAP- and ECM-associated pathways, and preservation of renal structural markers including nephrin and WT1.Conclusions:The localized CSMA-mediated co-delivery of CSMA/VEGF/CeO2NPs hydrogel promotes renal repair through resolution of maladaptive immune activation, metabolic reprogramming, angiogenic enhancement, and preservation of renal structural integrity, providing a promising biomaterial strategy for the treatment of ischemic and infection-associated renal injuries.

Repair of craniofacial bone defects remains highly challenging due to the region's complex anatomy close to vital brain marrow tissue. How to make a safe and efficient repair without disturbing surrounding tissues is critical, especially in external-stimulus-enhanced regeneration strategy. Magnetic hyperthermia therapy (MHT), due to its ability of uniform heat generation and good penetration, has gained increasing attention in bone repair. An asymmetric magnetic microspheres hybrid hydrogel was developed with magnetothermal responsiveness for targeted heating at the defect, preventing the risk of potential thermal injury to adjacent critical tissues. Guided by an external magnetic field, magnetic microspheres distributed in the upper side of the hydrogel achieved localized mild heating (40°C–42°C) under an alternating magnetic field (AMF), whereas the lower side remained within a physiologically safe temperature (< 38°C). In vitro and in vivo experiments showed that the asymmetric magnetic hydrogel under safe magnetothermal stimulation significantly enhanced the osteogenic differentiation and vascularization. Furthermore, transcriptomic analysis and histological staining suggested that this platform may convert external magnetothermal stimulation into intracellular signals by activating the PI3K-Akt pathway and upregulating key osteogenic and angiogenic markers. Overall, this asymmetric magnetothermal hydrogel offers a spatially controlled, safe, and effective strategy for bone regeneration.
