Biodegradable Two-dimensional (2D) nanomaterials have attracted tremendous interest in biomedical application. Herein, a biodegradable ferrous sulfide nanosheet with ultrathin structure and uniform size was developed. Following its development, the application of the self-enhanced photothermal nanoplatform (FMPN) in vivo by combining Mn-DOTA and dimeric PD-L1 affibody for targeted dual-mode magnetic resonance imaging (MRI) guided synergistic tumor therapy is explored. Dual-contrast enhanced subtraction imaging (DESI) was used to evaluate the excellent T1-T2 MRI ability of FMPN. FMPN displayed excellent photothermal conversion efficiency (η = 40.33 %) and biodegradability, thus had good tumor photothermal therapy effects with good biosafety. In addition, FMPN with pH-responsive Fe 2+ release can effectively induce tumor cell ferroptosis, further intensifying PTT and subsequent immunologic cell death. PD-L1 affibody could endow this nanoplatform with efficient tumor targeting, terminating T cells’ immune suppression by effectively blocking the interaction between PD-1 and PD-L1. Systemic delivery of FMPN significantly inhibited tumor progression in primary, metastatic and abscopal tumors, and showed obvious dual-mode MRI enhancement. Therefore, this nanoplatform exhibited promising potential for MRI-guided synergistic ferroptosis/photothermal/immunotherapy.

From the perspective of targeting factors, iron acetylacetonate and nickel acetylacetonate were used as major materials, and oleylamine was used as a solvent. Through thermal decomposition, the uniformly distributed nanoparticle FeNi3 was obtained. Then, the α-methyltryptophan
(AMT) was used to modify FeNi3, thereby the material (FeNi3-AMT) would be biocompatible. The prepared material was used as a targeting factor to explore whether it could target the epileptogenic focus in temporal lobe epilepsy model rats. In the experiment, the animal
models of temporal lobe epilepsy were constructed by using the lithium chloride-pirocarpine method. According to the injection of the nuclear magnetic factor, the models were divided into the FeNi3 group, the FeNi3-AMT group, and the saline group. The model rats received
intravenous injections 48 hours after the onset of epilepsy. The activity of the epileptic seizure sites before and after injection, the penetration of nanoparticles, and the toxic and side effects of the material were compared. The results showed that the rat models of temporal lobe epilepsy
were successfully constructed (the success rate was more than 85%). Meanwhile, the epilepsy lesions of the rat models added with nanoparticles were darker (P < 0.05). The staining of rat brain sections confirmed the distribution of iron particles. In the FeNi3-AMT group,
there were more iron particles distributed (P < 0.05). In addition, various abnormal neurons appeared in the hippocampus of the three groups, and there was no significant difference between the different groups (P < 0.05). There was no significant difference in the number
of positive neurons in the CA3 region of the brain between different groups (P < 0.05). However, the imaging effect based on FeNi3-AMT was more prominent (P < 0.05), which confirmed that the FeNi3-AMT group had better targeting location effects. The
study confirmed that the iron nano alloy particle material had excellent application values in the localization of temporal lobe epilepsy.

Dextran-poly(glycidyl methacrylate) (Dex-PGMA) nano-suitcases were synthesized efficiently via a graft copolymerization induced self-assembly (GISA) approach. On this basis, the Dex-PGMA nano-suitcases were modified with hydrazide, and the attachment of multiple chelated Gd(III) ions to the interior of the nano-suitcases affords nanoscale MRI contrast agents with high relaxivity values. The highly fenestrated dextran shell of the nano-suitcases assures water exchange which readily occurs between the surrounding environment and the Gd(III) ions encapsulated within the hybrid nano-suitcases. The complexation between the hydrophilic hydrazide interior of the nano-suitcases and Gd(III) ions results in an impressive Gd payload at 22.6 wt% in the hybrid nano-suitcases. The longitudinal relaxivity (r1) of the hybrid nano-suitcases is reported as 44.4 L/(mmol·s), which is 9–14 folds of that of commercial Gd-DTPA agents. In vivo MRI studies demonstrate that the hybrid nano-suitcases accumulated in the lymph node of the rat due to their nanoscale dimensions and displayed strong signals in vivo . The results indicated that the hybrid nano-suitcases provide a promising platform for the diagnosis of lymph node related diseases.

In the past decade, numerous species of nanomaterials have been developed for biomedical application, especially cancer therapy. Realizing visualized therapy is highly promising now because of the potential of accurate, localized treatment. In this work, we first synthesized metal nanorattles (MNRs), which utilized porous gold shells to carry multiple MR imaging contrast agents, superparamagnetic iron oxide nanoparticles (SPIONs), inside. A fragile wormpore-like silica layer was manipulated to encapsulate 8 nm oleylamine SPIONs and mediate the in situ growth of porous gold shell, and it was finally etched by alkaline solution to obtain the rattle-type nanostructure. As shown in the results, this nanostructure with unique morphology could absorb near-infrared light, convert to heat to kill cells, and inhibit tumor growth. As a carrier for multiple SPIONs, it also revealed good function for T2-weighted MR imaging in tumor site. Moreover, the rest of the inner space of the gold shell could also introduce potential ability as nanocarriers for other cargos such as chemotherapeutic drugs, which is still under investigation. This metal rattle-type nanocarrier may pave the way for novel platforms for cancer therapy in the future.

Although molecular imaging in nuclear medicine has made remarkable progress, the development of tumor-targeted magnetic resonance imaging (MRI) contrast agents based on tumor-cell–specific targets remains a major challenge. Conventional non-targeted gadolinium chelates such as Gd-DTPA are widely used in clinical MRI but often require high doses to achieve diagnostically meaningful enhancement, and their performance is constrained by modest relaxivity and the lack of tumor-targeting capability. Attempts to introduce targeting ligands onto small-molecule gadolinium complexes may increase molecular complexity, alter pharmacokinetics, and raise biosafety concerns. Meanwhile, superparamagnetic iron oxide nanoparticles have been explored as alternative MRI agents due to their high magnetic sensitivity, yet metal deposition, biodegradation, and long-term retention toxicity continue to hinder clinical translation. Current prostate-specific membrane antigen (PSMA)-targeted PET tracers enable highly sensitive lesion detection, yet a corresponding PSMA-targeted MRI agent capable of providing high-resolution anatomical and functional information is still lacking. Therefore, constructing a true PET–MRI dual-modality nanoplatform is expected to overcome the spatial-resolution limitations of PET alone while compensating for the insufficient molecular specificity of conventional MRI contrast agents. In this study, we developed a novel multifunctional organic nanoplatform based on natural melanin nanoparticles (MNPs). By simply integrating nanocarriers, imaging moiety and targeting ligands within one system, a PSMA-targeted MRI contrast agent was obtained. The PSMA ligand was covalently conjugated onto the surface of PEG-modified MNPs, while large numbers of paramagnetic Gd 3+ ions were chelated/adsorbed onto the melanin scaffold, yielding prostate-cancer-targeted PSMA-GMNs. The hydrodynamic diameters of MNPs and PSMA-GMNs were 3.56 ± 2.09 nm and 15.74 ± 1.63 nm, respectively. PSMA-GMNs exhibited a T 1 -enhancing effect more than five-fold stronger than that of Gd-DTPA in vitro . In vivo MRI demonstrated a high T 1 -weighted signal within LNCaP tumors at 4 h and 24 h post-intravenous administration. Furthermore, 64 Cu-labeled PSMA-GMNs achieved peak tumor uptake at 16 h (SUVmax = 4.24 ± 0.50), indicating sustained PSMA-mediated internalization and prolonged tumor retention. The successfully constructed 64 Cu-PSMA-GMN s achieve functional complementarity between the high-sensitivity molecular detection of PET and the high-resolution anatomical visualization of MRI within a single nano-platform. This work addresses the current lack of prostate-cancer-specific MRI contrast agents and the separation of PET and MRI molecular information in clinical practice.

This study aimed to synthesize and characterize L-epigallocatechin gallate (EGCG) complexed Mn2+ nanoparticle (L-EGCG-Mn), a proof-of-concept pH-sensitive manganese core nanoparticle (NP), and compare its magnetic resonance (MR) properties with those of Gd-DTPA, both in vitro and in vivo. Reverse microemulsion was used to obtain the L-EGCG-Mn NPs. The physicochemical properties of L-EGCG-Mn were characterized using dynamic light scattering, transmission electron microscopy, and near-infrared fluorescence small animal live imaging. The in vitro relaxivity of L-EGCG-Mn incubated with different pH buffer solutions (pH = 7.4, 6.8, 5.5) was evaluated. The T1-weighted MR imaging (MRI) properties were evaluated in vitro using hypoxic H22 cells as well as in H22 tumor-bearing mice. Cytotoxicity tests and histological analysis were performed to evaluate the safety of L-EGCG-Mn. L-EGCG-Mn showed good biocompatibility, stability, pH sensitivity, and tumor-targeting ability. Moreover, when the pH was decreased from 7.4 to 5.5, the r1 relaxivity of L-EGCG-Mn was shown to gradually increase from 1.79 to 6.43 mM−1·s−1. Furthermore, after incubation with L-EGCG-Mn for 4 h, the T1 relaxation time of hypoxic H22 cells was significantly lower than that of normoxic H22 cells (1788 ± 89 vs. 1982 ± 68 ms, p=.041). The in vivo analysis showed that after injection, L-EGCG-Mn exhibited a higher MRI signal compared to Gd-DTPA in H22 tumor-bearing mice (p < .05). Furthermore, L-EGCG-Mn was found to have a good safety profile via cytotoxicity tests and histological analysis. L-EGCG-Mn has a good safety profile and pH sensitivity and may thus serve as a potential MRI contrast agent.

Multifunctional nanomaterials with targeted imaging and chemotherapy have high demand with great challenge. Herein, we rationally aimed to design multifunctional drug delivery systems by RGD-modified chitosan (CH)-coated nanoneedles (NDs) of gadolinium arsenate (RGD-CH-Gd-AsNDs). These NDs have multifunctionality for imaging and targeted therapy. NDs on intravenous administration demonstrated significant accumulation of As ions/species in tumor tissues, which was monitored by the change in T1-weighted magnetic resonance (MR) imaging. Moreover, NDs were well opsonized in cells with high specificity, subsequently inducing apoptosis to the HepG2 cells. Consequent to this, the in vivo results demonstrated biosafety, enhanced tumor targeting, and tumor regression in a subcutaneously transplanted xenograft model in nude mice. These RGD-CH-Gd-AsNDs have great potential, and we anticipate that they could serve as a novel platform for real-time T1-weighted MR diagnosis and chemotherapy.

Sustained and localized delivery of chemotherapeutics in postoperative cancer treatment leads to a radical improvement in prognosis and a much decreased risk of tumor recurrence . In this work, polydopamine (PDA)-coated superparamagnetic iron oxide nanoparticle (SPION)-loaded polycaprolactone and poly(lactic-co-glycolic acid) fibers were developed as a potential implant to ensure safe and sustained release of the chemotherapeutic drug methotrexate (MTX), as well as provide local contrast for magnetic resonance imaging (MRI). Fibres were prepared by co-axial electrospinning and loaded with MTX-layered double hydroxide (LDH) nanocomposites in the core, yielding organic–inorganic hybrids ranging from 1.23 to 1.48 µm in diameter. After surface coating with PDA, SPIONs were subsequently loaded on the fibre surface and found to be evenly distributed, providing high MRI contrast. In vitro drug release studies showed the PDA coated fibres gave sustained release of MTX over 18 days, and the release profile is responsive to conditions representative of the tumor microenvironment such as slightly acidic pH values or elevated concentrations of the reducing agent glutathione (GSH). In vitro studies with Caco-2 and A549 cells showed highly effective killing with the PDA coated formulations, which was further enhanced at higher levels of GSH. The fibres hence have the potential to act as an implantable drug-eluting platform for the sustained release of cytotoxic agents within a tumor site, providing a novel treatment option for post-operative cancer patients.

19F magnetic resonance imaging (MRI)-assisted drug delivery provides the possibility to monitor and track drug transportation details in situ. A series of photo-responsive amphiphilic block copolymers consisting of hydrophilic poly(ethylene glycol) and 19F-containing hydrophobic segments, poly(2,2,2-trifluoroethyl acrylate) (PTFEA), with different chain lengths were synthesized by reversible addition–fragmentation chain-transfer polymerization. In particular, the photo-sensitive functional group of o-nitrobenzyl oxygen was introduced to control the photolysis behavior of the copolymers under ultraviolet irradiation. With the extension of the hydrophobic chain length, the drug loading capacity and photoresponsivity were both enhanced, while the chain mobility of PTFEA was suppressed, and the 19F MRI signal was attenuated. When the polymerization degree of PTFEA was about 10, the nanoparticles exhibit detectable 19F MRI signals and sufficient drug loading capacity (loading efficiency = 10%, cumulative release = 49%). These results offer a promising “smart” theranostic platform for 19F MRI.

Individual theranostic agents with dual-mode MRI responses and therapeutic efficacy have attracted extensive interest due to the real-time monitor and high effective treatment, which endow the providential treatment and avoid the repeated medication with side effects. However, it is difficult to achieve the integrated strategy of MRI and therapeutic drug due to complicated synthesis route, low efficiency and potential biosafety issues. In this study, novel self-assembled ultrasmall Fe 3 O 4 nanoclusters were developed for tumor-targeted dual-mode T 1 /T 2 -weighted magnetic resonance imaging (MRI) guided synergetic chemodynamic therapy (CDT) and chemotherapy. The self-assembled ultrasmall Fe 3 O 4 nanoclusters synthesized by facilely modifying ultrasmall Fe 3 O 4 nanoparticles with 2,3-dimercaptosuccinic acid (DMSA) molecule possess long-term stability and mass production ability. The proposed ultrasmall Fe 3 O 4 nanoclusters shows excellent dual-mode T 1 and T 2 MRI capacities as well as favorable CDT ability due to the appropriate size effect and the abundant Fe ion on the surface of ultrasmall Fe 3 O 4 nanoclusters. After conjugation with the tumor targeting ligand Arg-Gly-Asp (RGD) and chemotherapy drug doxorubicin (Dox), the functionalized Fe 3 O 4 nanoclusters achieve enhanced tumor accumulation and retention effects and synergetic CDT and chemotherapy function, which serve as a powerful integrated theranostic platform for cancer treatment.

Magnetic resonance imaging (MRI) contrast agents face persistent challenges in achieving target-specific activation and high signal-to-noise ratios (SNRs). To address these limitations, we developed LP-GZF, a bioresponsive nanoprobe leveraging magnetic resonance tuning (MRET) within tumor-targeting liposomes. The system coencapsulates Gd-DOTA (paramagnetic enhancer) and Zn0.4Fe2.6O4@DMSA (superparamagnetic quencher), engineered to switch between “OFF” and “ON” states via spatial separation. It has also been confirmed that the relaxivity of LP-GZF is only related to its state, instead of its concentration. In the native state, the proximity between components suppresses Gd3+ relaxivity (0.15 mM–1 s–1), while the release of Gd-DOTA restores relaxivity to 4.25 mM–1 s–1, a 28.3-fold enhancement. In 4T1 tumor-bearing mice, LP-GZF demonstrated delayed burst-enhancement kinetics, with SNR surging from 29.00 ± 1.13 to 48.88 ± 3.32 at 2 h postinjection, clearly lighting the boundary of tumor tissue. The outstanding performance of LP-GZF is attributed to the selective enrichment in the tumor and the subsequent spatial separation of the “enhancer” and “quencher”. The nanoprobe exhibited excellent biocompatibility and hepatorenal clearance. This work establishes a paradigm for stimuli-responsive contrast agents, combining spatiotemporal precision with analytical reliability for early cancer diagnosis.

Polypyrrole (PPy) nanoparticles have been widely studied in tumor photothermal therapy (PTT) for their significant photostability, good biocompatibility, and excellent photothermal performance. Herein, we report bovine serum albumin (BSA) stabilized PPy that were mineralized by MnO2 nanozyme on the surface (PPy@BSA-MnO2) to achieve synergistic photothermal and chemodynamic therapy (CDT) for breast cancer. In this multifunctional nanoplatform, the surface-loaded MnO2 undergoes a redox reaction with glutathione (GSH) to generate glutathione disulfide (GSSG) and Mn2+. Then, Mn2+ can convert H2O2 into a highly cytotoxic ˙OH to achieve chemodynamic therapy (CDT) and possess good magnetic resonance (MR) T1-weighted imaging capabilities to realize contrast imaging of the 4T1 tumor-bearing mouse models. In addition, PPy nanoparticles can efficiently convert near-infrared light energy into heat and achieve PTT. Most importantly, PPy@BSA-MnO2 nanoprobes have excellent in vitro 4T1 cell-killing effect and in vivo tumor-suppressive properties. The acute toxicity assessment results indicate that PPy@BSA-MnO2 nanoprobes have good biological safety. Therefore, the as-prepared multifunctional PPy@BSA-MnO2 nanoprobes possess excellent performance to promote MRI-guided PTT/CDT synergistic therapy for breast cancer treatment and have extensive clinical transformation and application prospects.
