Rationale: Rapid and facile detection of pathogenic bacteria is challenging due to the requirement of large-scale instruments and equipment in conventional methods. We utilize D-amino acid as molecules to selectively target bacteria because bacteria can incorporate DADA in its cell wall while mammalian cells or fungi cannot. Methods: We show a broad-spectrum bacterial detection system based on D-amino acid-capped gold nanoparticles (AuNPs). AuNPs serve as the signal output that we can monitor without relying on any complex instruments. Results: In the presence of bacteria, the AuNPs aggregate and the color of AuNPs changes from red to blue. This convenient color change can distinguish between Staphylococcus aureus ( S. aureus ) and methicillin-resistant Staphylococcus aureus (MRSA). This system can be applied for detection of ascites samples from patients. Conclusion: These D-amino acid-modified AuNPs serve as a promising platform for rapid visual identification of pathogens in the clinic.

Infections caused by methicillin-resistant Staphylococcus aureus ( MRSA ) are a significant public health challenge, necessitating the development of innovative antibacterial strategies. Nanomaterials are recognized for their ability to serve as photothermal conversion agents, transforming light energy into heat for antibacterial applications. These materials act as biological enzymes capable of generating reactive oxygen species (ROS), leveraging their catalytic activity to combat infections. While they show considerable promise, enhancing their ROS-generating efficiency and overall catalytic performance remains a critical challenge. In this work, a high catalytic bimetallic nanoplatform was developed, building on “large electronegativity difference induction/etch-assisted/diffusion termination” mechanism to improve antibacterial efficacy. The synthesized nanoparticles were found to exhibit exceptional photothermal conversion efficiency and surface defects, enabling efficient bacterial eradication. Both in vitro and in vivo experiments demonstrated that H-Pt 3 Sn nanoparticles effectively eliminated MRSA while significantly inhibiting and disrupting biofilms formed by the pathogen. The nanoparticles also facilitated tissue repair by modulating macrophage-related molecular markers. In conclusion, this study presents a promising photothermal synergistic nanosystem with high catalytic functionality for advanced anti-infective applications.

Skin and soft tissue infections (SSTIs) are among the most common bacterial infections reported in outpatients. Drug-resistant bacteria are the major cause of treatment failure and increased mortality rate in patients with SSTIs, posing significant challenges to human health. In this study, new-generation rhodium nanoplates (RhNPs) and glycol chitosan- and polydopamine-functionalized RhNPs (Rh@GCS) are developed for the treatment of drug-resistant SSTIs. RhNPs exhibited favorable antibacterial activity against methicillin-resistant Staphylococcus aureus (MRSA) and Ag-resistant MRSA. The modified Rh@GCS exhibited enhanced antibacterial activity and can directly kill various drug-resistant bacteria by increasing the permeability of cell membranes, including gram-positive MRSA and gram-negative multidrug-resistant Escherichia coli ( E . coli ) and Pseudomonas aeruginosa (PA). Moreover, Rh@GCS effectively inhibited bacterial growth and promoted the healing of skin lesions in MRSA-induced SSTI mouse models. These results suggest that Rh@GCS is a promising nonantibiotic antimicrobial agent for the treatment of drug-resistant SSTIs.

The successful treatment of osteomyelitis remains a great challenge in the field of orthopedics. The clinical method for treating refractory bone infection requires a combination therapy of long-term systemic antibiotics administration and surgical debridement. It is highly desirable to develop an antibiotic-free, noninvasive, rapid strategy to eradicate osteomyelitis. Herein, we fabricate a piezoelectric-enhanced sonosensitizer that consists of a porphyrin-based hollow metal–organic framework (HNTM), MoS2 nanosheets, and a red cell (RBC) membrane. We find that the ultrasound (US)-induced piezoelectric polarization of MoS2 can improve the charge transfer of HNTM at the heterointerface of HNTM-MoS2, increasing the production of reactive oxygen species (ROS). Besides, MoS2 increases the asymmetric shape of HNTM, leading to the strong US-propelling ability of HNTM–MoS2. The produced ROS and strong mechanical force can kill methicillin-resistant Staphylococcus aureus (MRSA) with an antibacterial efficiency of 98.5% under 15 min of US treatment, resulting in intracellular DNA damage and increased oxidative stress and disturbance of purine metabolism, tryptophan metabolism, and pantothenate and CoA biosynthesis of MRSA. Together with the toxin neutralization ability, the RBC-HNTM-MoS2 successfully eliminates the bone infection and suppresses inflammation and bone loss. This work provides another strategy for developing an efficient sonosensitizer through piezoelectric-assisted sonocatalysis and enhancing US-propelling ability.

The development of new antibiotics continues to pose challenges, particularly considering the growing threat of multidrug-resistant Staphylococcus aureus . Structurally diverse natural products provide a promising source of antibiotics. Herein, we outline a concise approach for the collective asymmetric total synthesis of polycyclic xanthene myrtucommulone D and five related congeners. The strategy involves rapid assembly of the challenging benzopyrano[2,3-a]xanthene core, highly diastereoselective establishment of three contiguous stereocenters through a retro -hemiketalization/double Michael cascade reaction, and a Mitsunobu-mediated chiral resolution approach with high optical purity and broad substrate scope. Quantum mechanical calculations provide insight into stereoselective construction mechanism of the three contiguous stereocenters. Additionally, this work leads to the discovery of an antibacterial agent against both drug-sensitive and drug-resistant S. aureus . This compound operates through a unique mechanism that promotes bacterial autolysis by activating the two-component sensory histidine kinase WalK. Our research holds potential for future antibacterial drug development.

Bacterial infection, oxidative stress, and persistent inflammation impede wound repair, and antibiotic-dependent treatment may promote the emergence of bacterial resistance. We developed an infection-adaptive, near infrared (NIR) regulated self-healing hydrogel dressing (PACH) comprising polydopamine anchored silver nanostructures (PDA@Ag) within a dynamic Schiff base network of carboxymethyl chitosan and oxidized hyaluronic acid. Because Methicillin-resistant Staphylococcus aureus (MRSA) secretes hyaluronidase during infection, the oxidized hyaluronic acid network was designed to undergo infection-adaptive degradation; NIR-induced photothermal heating further loosened and disassembled the network, thereby increasing the accessibility of PDA@Ag-related antibacterial components. PDA@Ag exhibited enhanced NIR absorption and photothermal performance (ΔT _max = 35.7 °C at 0.75 W/cm 2 ). PACH protected fibroblasts against H₂O₂ induced oxidative injury, promoted fibroblast migration, and attenuated inflammatory responses in lipopolysaccharide stimulated macrophages. In vitro, PACH exhibited potent antibacterial and antibiofilm activity against MRSA, with 808 nm irradiation reducing relative bacterial viability to 0.001%. In an MRSA infected full-thickness wound model, PACH with NIR reduced relative bacterial viability to 1.42 ± 0.59% on Day 3 and residual wound area to 2.518 ± 0.97% on Day 14, and enhanced re-epithelialization, collagen deposition, angiogenesis, and macrophage polarization toward a reparative phenotype without evident systemic toxicity. PACH is therefore a bio based hydrogel that forms in water at room temperature without organic solvents and provides antibacterial treatment without conventional antibiotics. These findings support an integrated dressing strategy for bacterial control and wound repair. Download: Download high-res image (148KB) Download: Download full-size image

Antimicrobial peptides (AMPs), which were conserved constituents of the innate immune system, exhibit antimicrobial properties similar to those of traditional antibiotics and have a low potential for the development of resistance. Thus, they were showing a great capacity for application. This study performed screening of twelve candidate antimicrobial peptides by means of pig skin enzymatic hydrolysis and activity prediction. The synthesis of these peptides was undertaken using Fmoc solid phase synthesis and purified via HPLC than 95%. The Methicillin-resistant Staphylococcus aureus (MRSA), Staphylococcus aureus (SA) and Escherichia coli (E. coli) were selected to evaluate antibacterial activity in vitro. The results indicated that peptide P5 exhibited the most significant inhibitory effect on MRSA and SA, with an inhibition rate of 91.83%. Furthermore, its antibacterial activity against MRSA and SA was found to be stronger than that against E. coli, demonstrating a dose-dependent relationship. Growth curve analysis revealed that P5 completely inhibited the growth of MRSA within 16 h at 3 mg/mL. Cytotoxicity was assessed using the CCK-8 assay and live/dead cell staining. The survival rates of both L929 and MDCK cells exceeded 87%, indicating good biocompatibility. Transcriptome sequencing (RNA-seq) and quantitative reverse transcription polymerase chain reaction (qRT-PCR) analyses demonstrated that treatment with the P5 was associated with significant transcriptional changes in MRSA. The differentially expressed genes were enriched in the ABC transporter pathway, as well as pathways related to arginine biosynthesis and metabolism, histidine metabolism, and quorum sensing. These findings suggest a potential link between P5 and these pathways, thereby providing a foundation for future mechanistic studies. In conclusion, the porcine skin-derived antimicrobial peptide P5 exhibited significant antimicrobial activity, low cytotoxicity, and a mechanism of action that was distinct from traditional antibiotics, thereby demonstrating promising potential for clinical translation in the fight against drug-resistant bacterial infections.

Infections caused by drug-resistant bacteria have become a new challenge in infection treatment, gravely endangering public health. Chloramphenicol (CL) is a well-known antibiotic which has lost its efficacy due to bacterial resistance . To address this issue, herein we report the design, synthesis and biological evaluations of novel triphenylphosphonium chloramphenicol conjugates (TPP + -CL). Study results indicated that compounds 39 and 42 possessed remarkable antibacterial effects against clinically isolated methicillin-resistant Staphylococcus aureus (MRSA) with MIC values ranging from 1 to 2 μg/mL, while CL was inactive to the tested MRSA strains. In addition, these conjugates exhibited rapid bactericidal properties and low toxicity, and did not readily induced bacterial resistance , obviously outperforming the parent drug CL. In a mouse model infected with a clinically isolated MRSA strain, compound 39 at a dose of 20 mg/kg exhibited a comparable or even better in vivo anti-MRSA efficacy than the golden standard drug vancomycin , while no toxicity was observed.

Infections caused by Methicillin-resistant Staphylococcus aureus (MRSA) have emerged as one of the most pressing global public health challenges. In concert with global rise of antimicrobial resistance at alarming rate, there is an urgent need for alternative strategies to combat MRSA. Here, the high throughput screening indicated that the Alisol A 24-acetate (AA) effectively inhibits the mevalonate (MVA) synthesis in MRSA. The mechanistic analysis revealed that AA competitively inhibits the 3-hydroxy-3-methyl glutaryl coenzyme A reductase (HMGR) protein to blockade the MVA pathway, thereby disrupting the bacterial membrane integrity and functions. Further investigations showed that this disruption consequently restores the β-lactam susceptibility in MRSA by retarding the expression of PBP2a protein and dampens the virulence of MRSA by reducing the exotoxins secretion. In addition to the effect on MRSA, AA has been found to exert host-acting activity to reduce the MRSA-induced inflammation. The promising anti-MRSA activity of AA was further confirmed in vivo . Collectively, the current study highlighted the potential of AA as a proposing drug for combating MRSA and emphasize the MVA pathway as an ideal therapeutic target for MRSA treatment.

Hydrogel-based delivery systems have now emerged as a pivotal platform for addressing chronic tissue defects, leveraging their innate capacity to suppress pathogenic infections and facilitate expedited tissue regeneration. In this work, an injectable hydrogel dressing, termed AgNPs-dermal matrix hydrogel (Ag@ADMH), has been designed to expedite the healing process of wounds afflicted with methicillin-resistant Staphylococcus aureus (MRSA), featuring sustained antibacterial efficacy. The synthesis of the hydrogel dressing entailed a self-assembly process of collagen fibers within an acellular dermal matrix to construct a three-dimensional scaffold, encapsulated with plant polyphenol-functionalized silver nanoparticles (AgNPs). The Ag@ADMH demonstrated exceptional biocompatibility, and enables a sustained release of AgNPs, ensuring prolonged antimicrobial activity. Moreover, the in vitro RT-qPCR analysis revealed that compared with ADMH, Ag@ADMH diminish the expression of iNOS while augmenting CD206 expression, thereby mitigating the inflammatory response and fostering wound healing. Especially, the Ag@ADMH facilitated a reduction in M1 macrophage polarization, as evidenced by a significant decrement in the M1 polarization trend and an enhanced M2/M1 ratio in dermal matrix hydrogels laden with AgNPs, corroborated by confocal microscopy and flow cytometry analyses of macrophage phenotypes. The in vivo assessments indicated that Ag@ADMH minimized fibrous capsule formation. In a full-thickness skin defect model of MRSA infection, the formulation significantly attenuated the inflammatory response by reducing MPO and CD68 expression levels, concurrently promoting collagen synthesis and CD34 expression, pivotal for vasculogenesis, thereby accelerating the resolution of MRSA-infected wounds. These attributes underscore the injectable extracellular matrix hydrogel as a formidable strategy for the remediation and regeneration of infected wounds. Graphical Abstract

The increasing prevalence of multidrug-resistant bacteria is a significant global health threat. In contrast to conventional antibiotic treatments, photodynamic therapy (PDT) offers a promising alternative by reducing the bacterial adaptability to antibiotics and bactericides. However, traditional photosensitizers encounter poor antimicrobial efficacy due to poor hydrophilicity of photosensitizers, short lifespan, narrow diffusion radius of reactive oxygen species (ROS), and the risk of exacerbating inflammation. In this study, we report a bacterial-targeting supramolecular nanophotosensitizer for combating multidrug resistant bacteria. The nanophotosensitizer, formed through host–guest interactions and self-assembly of tetra-cyclodextrin-modified silver porphyrin (AgTPP-CD4), adamantyl-modified phenylboronic acid (Ad-PBA), and curcumin (Cur), can effectively target and kill methicillin-resistant Staphylococcus aureus (MRSA). Moreover, it reduces inflammation and promotes wound healing in MRSA-infected wounds without inducing drug resistance. The combination of supramolecular chemistry and targeted PDT offers a promising strategy for combating multidrug-resistant bacterial infections.
