Atomically precise Ag clusters for intelligent NIR-II imaging

Huizhen Ma,Ling Liu,Hua He,Si Sun,Shuyu Yang,You Huang,Yuan Li,Kexin Tan,Haile Liu,Shuhu Liu,Yonghui Li,Changlong Liu,Pengfei Liu,Hao Wang,Xiao-Dong Zhang

Ligand-protected metal clusters are essential in optics, catalysis, and biomedicine due to their unique electronic transfers and rapid renal clearance, but their stability and photostability are still highly challenged. Here, we develop atomically precise Ag 29 clusters with 900–1,200 nm emissions that exhibit ultrahigh structural stability for 2 months and distinct photostability for 140 min via Pt single-atom engineering. The density functional theory shows that Ag 29 behaves as a molecular-like semiconductor with a direct band gap of 1.3 eV. Meanwhile, the Pt single atom introduces rich 5 d states in the highest occupied molecular orbital, leading to a continuous electron supply that ensures high photostability. We acquire numerous images of blood vessels, which enables machine learning to predict high-resolution near-infrared II (NIR-II) images. The NIR-II light sheet microscopy achieves a large field of view and deep-tissue imaging without photobleaching and accurately surveils the pathological progression of intestinal injury at the depth of 2.4 μm.

Exploiting single molecular species synchronously affording powerful second near-infrared (NIR-II) fluorescence, superior photoacoustic output, prominent reactive oxygen species generation, and satisfactory photothermal conversion is supremely appealing for phototheranostics, yet remains formidably challenging. In this work, electron donor/π-bridge engineering is implemented on the basis of 6,7-di(thiophen-2-yl)-[1,2,5]thiadiazolo[3,4- g ]quinoxaline moiety. The optimal molecule, namely TPATO-TTQ, is demonstrates to exhibit those notable features requested by exceptional phototheranostics, which are systematically elucidated through the depictions of excited-state energy dissipation pathways and the influence of intramolecular motion on the photophysical properties, with assistances of quantum chemical calculation and molecular dynamic simulation. By utilizing TPATO-TTQ nanoparticles, unprecedented performance on NIR-II fluorescence-photoacoustic-photothermal trimodal imaging-navigated type I photodynamic-photothermal synergistic therapy to orthotopic breast cancer is authenticates by the precise tumor diagnosis and complete tumor ablation.

InSe Nanosheets for Efficient NIR-II-Responsive Drug Release

Chi Huang,Zhengbo Sun,Haodong Cui,Ting Pan,Shengyong Geng,Wenhua Zhou,Paul K. Chu,Xue-Feng Yu

Near-infrared-II (NIR-II) biowindow is appealing from the perspectives of larger maximum permissible exposure in comparison with the near-infrared-I biowindow, so the NIR-II-responsive drug-delivery nanoplatform is highly desirable. In this work, two-dimensional InSe nanosheets (InSe NSs) are modified with poly(ethylene glycol) and evaluated as an effective NIR-II-responsive cancer treatment nanoplatform. The InSe NSs synthesized by liquid exfoliation exhibit prominent NIR-II-responsive photothermal conversion efficiency (39.5%) and photothermal stability. Moreover, the InSe NSs have a doxorubicin (DOX) loading capacity as high as 93.6%, along with excellent NIR-II-responsive DOX release characteristic. The superior synergistic chemo/photothermal effects have also been demonstrated by the in vitro experiments in killing cancer cells. In combination with good biocompatibility, the InSe NSs have great potential in therapeutic applications.

Ligand Engineering of Gold Nanoclusters for NIR-II Imaging

Meili Guo,Gang Zhang,Ruoli Zhao,Huizhen Ma,Yuxing Yan,Shuyu Yang,Jian Meng,You Huang,Xiao-Dong Zhang,Hao Wang,Ruiping Zhang

Near-infrared-II (NIR-II) imaging has shown great potential in medical diagnosis and surgical navigation, but developing safe, photostable, high-brightness molecular probes remains a great challenge. Due to their ultrasmall size resulting in highly efficient renal clearance, gold nanoclusters have shown great clinical potential. In this work, we systematically explored the effect of different ligands on the luminescence and bioactivity of gold nanoclusters. Our results show that gold nanoclusters protected by thioglycolic acid (TGA) and 6-mercaptohexanoic acid (MHA) exhibit the strongest fluorescence, while 3-mercaptopropionic acid (MPA)- and sodium sulfide (Na2S)-protected gold nanoclusters show the weakest NIR-II signal. Further doping showed that the Cd-doped MPA and MHA-protected gold nanoclusters exhibited enhanced fluorescence, but the cysteine (Cys)-, glutathione (GSH)-, and Na2S-protected gold nanoclusters showed fluorescence quenching after doping, indicating significant ligand selectivity. Because of the unique multi-energy structure and the large number of electronic states at the highest occupied molecular orbital energy level, MPA- and MHA-protected gold nanoclusters exhibit high stability and photostability. In addition, gold nanoclusters with different ligands exhibited different selective enzyme-mimicking activities of peroxidase (POD), superoxide dismutase (SOD), and catalase (CAT). Imaging in vivo showed that gold nanoclusters could accomplish reliable imaging of cerebral vasculature, hindlimb vessels, and spine as well as monitor renal clearance. The stable nanoclusters allow the time window of imaging to reach 125 min for hindlimb imaging and 270 min for spinal imaging. The gold nanoclusters exhibit a high signal-to-noise ratio of up to 11 for whole-body imaging and show efficient renal clearance and low toxicity at an injected dose of 50 mg/kg.

Trimerization-Enhanced NIR-II/Photoacoustic Nanoprobes for Precision Osteosarcoma Imaging

Siyu Lu,Jing Wu,Shuxin Lyu,Yifeng Yu,Jong Seung Kim,Ruiping Zhang,Yuling Xiao,Xuechuan Hong,Xiaodong Zeng

The integration of photoacoustic imaging (PAI) and near-infrared II (NIR-II) fluorescence imaging holds transformative potential for cancer diagnostics but faces challenges in developing high-performance dual-modal contrast agents. We report tH7@CT8, an organic nanoprobe engineered through fluorophore trimerization to address the photothermal conversion efficiency limitations of small-molecule agents. Trimerization of the [1,2,5]thiadiazolo[3,4-g]quinoxaline fluorophore derivative H7 induced aggregation, suppressing fluorescence quantum yield (0.11% vs 0.42% monomer) while amplifying PTCE to 43.1%. Encapsulation within an osteosarcoma-targeting amphiphilic matrix yielded nanoparticles (72 ± 19 nm) with 4-fold enhanced tumor uptake versus nontargeted controls. In orthotopic osteosarcoma models, tH7@CT8 enabled PAI detection and real-time NIR-II guidance with signal (tumor)-to-background ratios >12:1. This work establishes a molecular design strategy to reconcile depth-resolution trade-offs in oncology while advancing organic contrast agents toward clinical translation.

NIR-II Luminescent Nanothermometer for Precise Assessment of Ischemic Stroke

Xiaochen Qiu,Wei Yuan,Chen Jiang,Wei Feng,Fuyou Li

Ischemic stroke, an acute cerebral local blood circulation disorder caused by neurological deficit syndrome, has emerged as a prominent ailment that can affect the well-being and quality of life of the elderly. The difficulty in early assessment and narrow therapeutic time window can increase the risks of mortality and disability. Monitoring the temperature of brain microregions is a promising strategy for assessing the onset of an early stroke, which is positively correlated with the intricate immune response. In this study, an innovative hybrid nanothermometer combining lanthanides and quantum dots was established for contactless monitoring of cerebral lesion temperatures in mice with ischemic stroke. The output of ratio optical signals in the near-infrared-II window (1000–1700 nm) guaranteed deep penetration, high accuracy, and reduced tissue damage. An obvious temperature increase was detected in the minor middle cerebral artery occlusion model mice (4 h after reperfusion), indicating the occurrence of a minor stroke even in the absence of abnormal behaviors. Moreover, a further increase in the brain temperature of the ischemic region resulted in a more severe cerebral lesion, suggesting the potential advancement of the treatment window for early ischemic stroke. The proposed approach can enable precise assessment of ischemic stroke, with significant implications for the incidence and progression of the illness, ultimately enhancing the quality of life of affected patients.

Glass Composite Fiber for Broadband NIR-II Mini Light Source

Yupeng Huang,Shichao Lv,Xueliang Li,Jianrong Qiu,Shifeng Zhou

Near-infrared second window (NIR-II, 1000–1700 nm) light sources present remarkable scientific and technological promise, with wide-ranging applications spanning biomedical imaging, biosensing, and related fields. However, most of the available candidates are in bulk form and it is urgently required to develop mini light sources. Herein, a broadband NIR-II mini fiber-type light source equipped by Ni 2+ activated glass composite fibers is proposed and investigated. It is shown that the transparent glass composite exhibits broadband NIR-II emission with a full width half-maximum approaching 240 nm and an internal quantum efficiency (IQE) larger than 50% by rational control of nanocrystallization. Additionally, the corresponding glass composite fibers are prepared, demonstrating efficient broadband response and distinct on-off gain in NIR-II region. Benefited from above features, an all-fiber mini light source by fusing glass composite fibers with commercial passive fibers is successfully constructed and demonstrates its promising applications for NIR-II imaging. These findings demonstrate significant progress in the development of NIR-responsive photonic materials, enabling advanced applications in photonic technology.

Designing Ultrasensitive NIR-II SERS Nanoprobes: Achieved via Plasmonic Engineering

Wu Yifan,Zhou Wenhao,An Baihong

The second near-infrared (NIR-II, 1000–1700 nm) window is advantageous for bioimaging due to its deep tissue penetration and low autofluorescence. Despite these benefits, creating highly sensitive surface-enhanced Raman scattering (SERS) nanoprobes in this range is challenging. This study introduces an ultra-sensitive SERS nanoprobe developed using plasmonic engineering. We selected the commercial Raman dye IR1061, with an absorption peak at 1061 nm, and synthesized gold nanotriangles (AuNTs) to absorb visible light and transfer energy to NIR-II phonons. The combination of IR1061 and AuNTs significantly improved the nanoprobe's sensitivity to 116 fM in the NIR-II window, due to enhanced electromagnetic fields and SERS effects. Theoretical analysis shows AuNTs have stronger electromagnetic hotspots than gold nanorods, enhancing SERS sensitivity. The nanoprobe penetrates pig tissue over 4 mm and agarose gels up to 5 cm. This work presents a novel approach to ultra-sensitive NIR-II SERS nanoprobes for bioimaging and diagnostics. It can not only overcome the bottleneck of traditional imaging technology, but also provide strong support for high-resolution imaging of deep tissues and provide more accurate information for clinical diagnosis and treatment. Au–Cu Janus Nanostructures as NIR-II Photothermal Antibacterial Agents

Qiuping Yang,Haixia Kong,Liwei Tang,Yanyun Ma,Feng Liu,Maochang Liu,Yi Wang,Pu Zhang,Yiqun Zheng

Metal nanoparticles have been found to exhibit a broad spectrum of antibacterial activities, yet not all metals possess the captivating plasmonic properties necessary for effective interaction with incident light. In this study, we demonstrate the feasibility of synergistically enhancing the Cu-induced antibacterial activity through the incorporation of plasmonic photothermal therapy by hybridlike anchoring the Cu component on a gold (Au) nanoplate that can effectively absorb near-infrared light. To fabricate the Janus structure, plate-like Au seeds were mixed with copper(ii) nitrate, cetyltrimethylammonium bromide, ascorbic acid, and hexamethylenetetramine, resulting in the formation of Au–Cu Janus nanostructures (JNSs). Notably, the plasmonic absorption of Au was preserved without being hindered by the Cu coating. Photothermal measurements revealed that the temperature rise of Au–Cu JNSs aqueous suspensions could reach up to 28.3 °C in just 10 min when exposed to 1064 nm laser light at a power density of 1 W·cm–2, with a photothermal conversion efficiency of 42.14%. Additionally, they show broad-spectrum antibacterial properties, making them potential contenders for addressing a wide range of bacterial infections. Our findings demonstrate that the antibacterial performance of metal nanoparticles composed of less plasmon-active components but with exceptional antibacterial properties can be significantly enhanced by coupling them with a plasmon-active nano-antenna through a Janus architecture. The Au–Cu JNSs possess potential application prospects in photothermal therapy, broad-spectrum antibacterial treatments, biomedical imaging, targeted drug delivery, sensitive diagnostics, and the development of advanced materials.

NIR-II Fluorescent Thermophoretic Nanomotors for Superficial Tumor Photothermal Therapy

Jiwei Jiang,Jing Hu,Mingtong Li,Mingzhi Luo,Bin Dong,Metin Sitti,Xiaohui Yan

Peritumoral subcutaneous injection has been highly envisioned as an efficient yet low-risk administration of photothermal agents for superficial tumor photothermal therapy. However, obstructed by complex subcutaneous tissue, the delivery of injected photothermal agents to the specific tumor remains a critical issue. Herein, the study reports a polydopamine (PDA)-encapsulated spherical core/shell nanomotor with fluorescent indocyanine green (ICG) immobilized on its PDA shell. Upon the first near-infrared (NIR-I) irradiation, this motor can generate favorable photothermal heat, and meantime, emit a robust ICG fluorescence in the second near-infrared window (NIR-II). The heat turns the motor into an active photothermal agent able to perform thermophoretic propulsion along the irradiation direction in subcutaneous tissue, while the ICG fluorescence can direct the subcutaneous propulsion of motors toward specific tumor through real-time NIR-II imaging. These functions endow the motor with the ability of moving to tumor after being injected at peritumoral site, enabling an enhanced photothermal therapy (PTT). The results demonstrated herein suggest an integrated nanorobotic tool for the superficial PTT using peritumoral administration, highlighting an NIR-II imaging-directed subcutaneous propulsion.

NIR-II-activated biocompatible hollow nanocarbons for cancer photothermal therapy

Xu Zhourui,Zhang Yinling,Zhou Weixiao,Wang Lijian,Xu Gaixia,Ma Mingze,Liu Fenghua,Wang Zan,Wang Yucheng,Kong Tiantian,Zhao Binyuan,Wu Weiping,Yang Chengbin

Photothermal therapy has attracted extensive attentions in cancer treatment due to its precise spatial-temporal controllability, minimal invasiveness, and negligible side effects. However, two major deficiencies, unsatisfactory heat conversion efficiency and limited tissue penetration depth, hugely impeded its clinical application. In this work, hollow carbon nanosphere modified with polyethylene glycol-graft-polyethylenimine (HPP) was elaborately synthesized. The synthesized HPP owns outstanding physical properties as a photothermal agent, such as uniform core-shell structure, good biocompatibility and excellent heat conversion efficiency. Upon NIR-II laser irradiation, the intracellular HPP shows excellent photothermal activity towards cancer cell killing. In addition, depending on the large internal cavity of HPP, the extended biomedical application as drug carrier was also demonstrated. In general, the synthesized HPP holds a great potential in NIR-II laser-activated cancer photothermal therapy.

Accurate In Vivo Nanothermometry through NIR-II Lanthanide Luminescence Lifetime

Meiling Tan,Feng Li,Ning Cao,Hui Li,Xin Wang,Chenyang Zhang,Daniel Jaque,Guanying Chen

Luminescence nanothermometry is promising for noninvasive probing of temperature in biological microenvironment at nanometric spatial resolution. Yet, wavelength- and temperature-dependent absorption and scattering of tissues distort measured spectral profile, rendering conventional luminescence nanothermometers (ratiometric, intensity, band shape, or spectral shift) problematic for in vivo temperature determination. Here, a class of lanthanide-based nanothermometers, which are able to provide precise and reliable temperature readouts at varied tissue depths through NIR-II luminescence lifetime, are described. To achieve this, an inert core/active shell/inert shell structure of tiny nanoparticles (size, 13.5 nm) is devised, in which thermosensitive lanthanide pairs (ytterbium and neodymium) are spatially confined in the thin middle shell (sodium yttrium fluoride, 1 nm), ensuring being homogenously close to the surrounding environment while protected by the outmost calcium fluoride shell (CaF 2 , ≈2.5 nm) that shields out bioactive milieu interferences. This ternary structure enables the nanothermometers to consistently resolve temperature changes at depths of up to 4 mm in biological tissues, having a high relative temperature sensitivity of 1.4–1.1% °C −1 in the physiological temperature range of 10–64 °C. These lifetime-based thermosensitive nanoprobes allow for in vivo diagnosis of murine inflammation, mapping out the precise temperature distribution profile of nanoprobes-interrogated area.
