Ochratoxin A(OTA) is positively correlated with an increased risk of developing cancer in nephrotoxic and hepatotoxic patients. Therefore, it is of great significance for the highly sensitive, highly selective, and timely detection of OTA. We described here an electrochemical aptasensor for OTA analysis, which took advantage of the favorable properties of gold nanoparticles (AuNPs) functionalized zinc oxide (ZnO) composites and the intercalative binding between methylene blue (MB) and nucleic acid. There were two label-free aptamers: one to capture OTA and another serving as complementary DNA (cDNA), enabling connection to the ZnO-Au composite’s immobilized electrode. Once OTA was present, the aptamer could capture OTA and detach from the electrode interface, thus, preventing MB from accessing electrode surface for efficient electron transfer; a decreased peak current was monitored by differential pulse voltammetry. The aptasensor presented nice analytical performance for OTA detection in the range of 0.1–30,000 pg·mL−1, with a detection limit of 0.05 pg·mL−1. Moreover, the developed biosensor could be applied to actual sample (wine and beer) analysis. Keywords:food safety;electrochemical aptasensor;intercalative binding;zinc oxide-gold composites;Ochratoxin A

Ochratoxin A (OTA) exposure in food poses great threat to human health. Magnetic solid phase extraction (MSPE) is an effective sample pre-treatment method based on liquid chromatographic analysis to monitor OTA in food. Herein, magnetic covalent organic frameworks (Fe 3 O 4 @COFs) nanospheres were prepared and well characterized. The efficient and selective extraction of OTA relied on large specific surface and inherent porosity of COFs and the hydrogen bonding, π-π and hydrophobic interactions between COF shell and OTA. The MSPE parameters were investigated in detail. Under the optimal MSPE conditions, the developed Fe 3 O 4 @DhaTab-based-MSPE-HPLC-FLD method for OTA displayed good linearity (0.1-800 μg L −1 ) with R 2 (0.996) and low limit of detection (0.03 μg L −1 ). The proposed method was applied to determine OTA in beer, Chinese Baijiu and vinegar with satisfactory recoveries (72–111%) and relative standard deviations (0.2 to 5.5%).

Ochratoxin A (OTA) is the most toxic class of ochratoxins and has become a major threat to the environment, humans and animals. Therefore, research on the methods for its detection is also more urgent. Herein, we propose a low-background electrochemical biosensor based on a DNA tetrahedron-besieged primer and a DNAzyme-activated programmatic rolling circle amplification (RCA) that can be ultimately utilized for OTA detection in wine samples. Low-background detection can be achieved using the besieged primer via sequenced assembly of DNA tetrahedral nanostructures so that non-specific extensions of primer can be avoided. The target OTA-mediated DNAzyme activation initiates the programmatic RCA. Additionally, the catalytic property of silver nanoclusters (AgNCs) is integrated with the electrochemical assay to achieve high sensitivity for OTA detection. Benefiting from the aforementioned processes, a low-background, and highly sensitive electrochemical biosensor has been successfully constructed. This design is capable of detecting OTA at concentrations from 1 pg/mL to 10 ng/mL, and its lowest concentration limit is 0.773 pg/mL. Simultaneously, its validation in the detection of actual samples reveals that the proposed electrochemical biosensor has a lot of potential in food safety and environmental detection.

A facile and efficient strategy is developed to modify aptamers on the surface of the magnetic metal-organic framework MIL-101 for the rapid magnetic solid-phase extraction of ochratoxin A. To the best of our knowledge, this is the first attempt to create a robust aptamer-modified magnetic MIL-101 with covalent bonding for the magnetic separation and enrichment of ochratoxin A. The saturated adsorption of ochratoxin A by aptamer-modified magnetic MIL-101 was 7.9 times greater than that by magnetic metal-organic framework MIL-101 due to the former's high selective recognition as well as good stability. It could be used for extraction more than 12 times with no significant changes in the extraction efficiency. An aptamer-modified magnetic MIL-101-based method of magnetic solid-phase extraction combined with ultra high performance liquid chromatography with tandem mass spectrometry was developed for the determination of trace ochratoxin A with limit of detection of 0.067 ng/L. Ochratoxin A of 4.53–13.7 ng/kg was determined in corn and peanut samples. The recoveries were in the range 82.8–108% with a relative standard deviation ( n = 5) of 4.5–6.5%. These results show that aptamer-modified magnetic MIL-101 exhibits selective and effective enrichment performance and have excellent potential for the analysis of ultra-trace targets from complex matrices.

In this study, a novel aptasensor based on Aptamer/NH 2 Janus particles is developed for the detection of Ochratoxin A(OTA). By coating gold on the hemispherical surface of the aminated polystyrene particles, Ochratoxin A aptamer is immobilized on the surface of the gold layer for selective identification and the other hemispherical able to bind to Glassy carbon electrode via peptide bond. Under optimum conditions, the sensor exhibited a wide dynamic range of OTA concentration from 1 × 10 −5 nM to 10 nM, and the detection limit is 3.3 × 10 −3 pM on condition of acceptable stability and reproducibility. The sensors were showed excellent performance in the detection of OTA in red wine sample with recoveries between 95.7% and 100.18%, which studied by the standard addition spiking technique. This work provides a new idea and method for preparing immune electrochemical sensors and is expected to be used for the OTA detection in red wine sample analysis.

This work proposed a novel and facile strategy for in-situ deposition of silver nanoparticles (AgNPs) on polydopamine nanospheres (PDANSs), and used the as-synthesized nanocomposite (AgNPs/PDANSs) as electrochemical tracer for an ultrasensitive aptasensor of ochratoxin A (OTA). The morphology and composition of PDANSs and AgNPs/PDANSs were characterized by electron microscopies and energy dispersive X-ray spectroscopy, and the results showed that AgNPs uniformly on PDANSs surface. AgNPs/PDANSs were combined with streptavidin (SA) to obtain signal probe for constructing of OTA aptasensor. Sequentially, the thiolated capture DNA (cDNA) was firstly immobilized on the gold electrode (GE) to capture the OTA aptamer, and then SA/AgNPs/PDANSs nanoprobes could bind with the end biotin of the aptamer through biotin-streptavidin biorecognition. The monitoring of the proposed OTA aptasensor was realized by the electrochemical stripping signal of AgNPs on PDANSs, and PDANSs was used for signal amplification. In the presence of OTA, the aptamer was preferentially bonded with OTA because of their high affinity, and resulted in the decrease of signal tags on the aptasensor surface, which reduced the electrochemical response signal of AgNPs. Under optimal experimental conditions, the proposed aptasensor offered a wide linear range from 1.0 × 10−6 μM to 1.0 × 10−2 μM and a low detection limit of 0.57 pM (S/N = 3). Meanwhile, This AgNPs/PDANSs-based OTA aptasensor had excellent specificity, stability and reproducibility, and was successfully employed for OTA detection in red wine samples, which showed promising application in electrochemical analysis of other toxic molecules.

A fluorometric aptamer-based assay for ochratoxin A (OTA) is described. It is making use of magnetic separation and a cationic conjugated fluorescent polymer. Amino-tagged aptamer (Apt) against OTA is immobilized on magnetic beads (MBs) to form a conjugate of type Apt-MBs. The immobilized aptamer is partially complementary to carboxyfluorescein-labeled DNA which binds to the Apt-MBs via hybridization if OTA is absent. Only few FAM-DNA will remain in the supernatant after magnetic separation, and only weak fluorescence resonance energy transfer (FRET) occurs on addition of the fluorescent polymer. If, however, OTA is present, it will bind to the aptamer and prevent the hybridization between Apt-DNA and FAM-DNA. This results in the presence of large amounts of FAM-DNA in the supernatant after magnetic separation. On addition of fluorescent polymer, efficient FRET occurs from the polymer to FAM-DNA. Fluorescence, best measured at excitation/emission peaks of 370/530 nm, increases with increasing concentrations of OTA. This assay is highly sensitive and selective. The detection limit is as low as 0.11 ng mL −1 . This is 6 times lower than the aptamer assay without using the fluorescent polymer. Conceivably, this method has a wider scope in that it may be extended to other mycotoxins by simply changing the aptamer.

A fluorescent biosensor for ochratoxin A was fabricated on the basis of a new nanocomposite (Fe 3 O 4 /g-C 3 N 4 /HKUST-1 composites). Fe 3 O 4 /g-C 3 N 4 /HKUST-1 was synthesized in this work for the first time, which combined HKUST-1 with g-C 3 N 4 to improve its chemical stability. Fe 3 O 4 /g-C 3 N 4 /HKUST-1 composites have strong adsorption capacity for dye-labeled aptamer and are able to completely quench the fluorescence of the dye through the photoinduced electron transfer (PET) mechanism. In the presence of ochratoxin A (OTA), it can bind with the aptamer with high affinity, causing the releasing of the dye-labeled aptamer from the Fe 3 O 4 /g-C 3 N 4 /HKUST-1 and therefore results in the recovery of fluorescence. The fluorescence intensity of the biosensor has a linear relationship with the OTA concentration in the range of 5.0–160.0 ng/mL. The LOD of sensor is 2.57 ng/mL (S/N=3). This fluorescence sensor based on the Fe 3 O 4 /g-C 3 N 4 /HKUST-1 composites has been applied to detect OTA in corn with satisfying results.

A multiplexed, sensitive and specific detection method is highly desirable for the simultaneous detection of several pathogenic bacteria and bio-toxins. In our previous work, multicolor upconversion nanoparticles (UCNPs) via doping with various rare-earth ions to obtain well-separated emission peaks by means of a solvothermal method were synthesized and were successfully applied as luminescence labels in the detection of three pathogenic bacteria. One of the basic achievements of our group has been to establish that the key to increasing the number of simultaneous detection components is the preparation of more UCNPs, the emission peaks of which can be distinguished from each other. According to this vision, NaYF 4 :Yb 0.2 , Tm 0.02 UCNPs were obtained via a thermal-decomposition protocol, which has a main near-infrared (NIR) UC emission at 804 nm under 980 nm excitation. The emission peak at 804 nm was well-separated from the emission peaks of UCNPs we have reported at 477 nm, 542 nm, and 660 nm. It means both the excitation and the emission of NaYF 4 :Yb 0.2 , Tm 0.02 UCNPs are located in the NIR spectral range (NIR-to-NIR UC emission), the so-called biological window. This result establishes the basis of achieving simultaneous detection of four components. To confirm the analytical performance of this NaYF 4 :Yb 0.2 , Tm 0.02 UCNPs, a novel near-infrared magnetic aptasensor for the detection of Ochratoxin A (OTA) was developed using the OTA aptamer-conjugated near-infrared upconversion nanoparticles (apt-UCNPs) and the complementary oligonucleotide-modified magnetic nanoparticles (cDNA-MNPs). The apt-UCNPs and cDNA-MNPs were hybridized to form a poly-network structure of MNP-UCNP nanocomposites. When the target OTA was introduced, the aptamer combined with the priority target and the cDNA-MNPs were replaced. The proposed method achieved a linear range between 0.01 and 100 ng mL −1 , with a detection limit as low as 0.005 ng mL −1 . Then, we successfully applied this method to measure Ochratoxin A (OTA) in beer samples and the results demonstrated that the method possessed a high sensitivity and good selectivity for the determination of OTA and thus is applicable to the determination of OTA in beer samples. This satisfying result shows that the NaYF 4 :Yb 0.2 , Tm 0.02 UCNPs we synthesized has a great prospect in multiplexed simultaneous detection.

Ochratoxin A (OTA), the most harmful and abundant ochratoxin, is chemically stable and commonly existed in foodstuffs. In this work, upconversion luminescent-magnetic microbeads (UCLMMs) -based cytometric bead array for OTA detection with a less reagent consumption and high sensitivity has been established and optimized. In UCLMMs, upconversion nanocrystals (UCNs) for optical code present a weak background noise and no spectral cross talk between the encoding signals and target labels under two excitation conditions to improve detection sensitivity. While the superparamagnetic Fe 3 O 4 nanoparticles (Fe 3 O 4 NPs) aim for rapid analysis. The results show that the developed method has a sensitivity of 9.553 ppt below HPLC with a 50-μL sample and can be completed in <2 h with good accuracy and high reproducibility. Therefore, different colors of UCLMMs will become a promising assay platform for multiple mycotoxins after further improvement.

OTA can induce hepatotoxicity. Our previous research has shown that miRNAs play important roles in the OTA-induced hepatotoxicity. And miR-122 is the most abundant miRNA in the liver and is involved in diverse biological processes. This study was performed to clarify the role of miR-122 in OTA-induced hepatotoxicity. The expression levels of miR-122 and the target genes were quantified by real-time PCR. The OTA-induced apoptosis of hepatocyte and HepG2 cells was evaluated using a TUNEL kit, a CCK-8 kit, a flow cytometer and Hoechst 33342. miR-122 was inhibited in HepG2 cells. The results revealed that OTA affected rat hepatocyte apoptosis. miR-122 decreased at 4 weeks but increased at 13 weeks in the OTA-treated livers, and increased in the OTA-treated HepG2 cells; and the mRNA levels of CCNG1 and Bcl-w increased at 4 weeks and decreased at 13 weeks in the high-dose OTA-treatment groups and decreased in HepG2 cells. The apoptosis of HepG2 cells displayed a dose-related increase with OTA. However, the inhibition of miR-122 greatly reduced OTA-induced apoptosis. p53 decreased in vivo and in vitro. miR-122 is a primary effector of OTA-induced hepatocyte apoptosis through the CCNG1/p53 pathway and Bcl-w/caspase-3 pathway in vivo and in vitro. And miR-122 plays an important role in OTA-induced hepatotoxicity.

Ochratoxin A (OTA), a fungal toxin, induces various toxic effects in animals and humans through the enrichment of toxin residues. In this work, a dual-modal biosensor based on gold nanoparticles (AuNPs) and branched hybridization chain reaction (bHCR) was proposed for the detection of OTA. The strategy is contingent on OTA aptamer-bridged occurrence of bHCR and the salt-induced aggregation of AuNPs. OTA-apt/cDNA could be used to specifically identify the OTA and trigger bHCR reactions, producing a long-branched dsDNA polymer. The electroactive molecule-methylene blue (MB) can be inserted into the superstructure of branched DNA due to the formation of DNA polymers, leading to dynamic changes in MB redox signaling. The residual DNA hairpins were added and adhered to the surface of AuNPs, but they were inadequate to prevent the AuNPs from salt-induced aggregation. The dual-modal yields limits of detection of 4.8 pM (electrochemical assay) and 0.25 nM (colorimetric assay), respectively. It exhibited excellent specificity against common mycotoxins (AFB1, DON, FB1, ZEN), with satisfactory recoveries in corn flour (92.9–108.3 %). This aptasensor, which adopts a dual-modal strategy, features self-calibration to reduce false-positive results and improve accuracy. It demonstrates significant advantages in mycotoxin detection.
