A novel fungus Penicillium crocicola LF-2022 for high-efficiency biodegradation of brominated flame retardants: Performance, pathways, soil remediation, and transcriptomic mechanisms

Yan Li,Ruiying Wu,Zhenglin Xia,Zhinan Jiang,Haitao Wu,Chen Lin,Jiaqing Chen,Fei Li

Tetrabromobisphenol A (TBBPA) and decabromodiphenyl ether (BDE-209) are typical brominated flame retardants (BFRs) with environmental persistence, bioaccumulation potential, and high toxicity, posing serious threats to ecosystems and human health. In this study, a novel fungal strain, Penicillium crocicola LF-2022 ( P. crocicola LF-2022), was identified, exhibiting excellent biodegradation capabilities toward both BFRs. Within six days, the strain achieved maximum degradation rates of 81.1% for TBBPA and 97.7% for BDE-209. The strain degraded both pollutants via intracellular and extracellular pathways, with cytochrome P450 enzymes playing a key role. Degradation pathway analysis revealed that TBBPA was degraded through reductive debromination and isopropylidene bridge cleavage, whereas BDE-209 underwent sequential debromination, ether bond cleavage, and ring-opening reactions, ultimately leading toward mineralization. In soil remediation experiments, P. crocicola LF-2022 effectively degraded TBBPA and BDE-209 even under non-sterile conditions, achieving approximately 55.3% and 82.9% degradation, respectively, demonstrating strong contaminant detoxification capability. Seed toxicity evaluation confirmed the detoxification efficacy of the fungal treatment. After remediation, wheat seedling root and shoot lengths increased markedly compared with untreated controls. Moreover, malondialdehyde content in plant tissues decreased significantly, while superoxide dismutase, peroxidase, and catalase activities recovered to near-control levels, indicating substantial BFR detoxification. Transcriptomic analysis revealed distinct degradation strategies: TBBPA stress upregulated genes related to membrane integrity, ABC transporters, CYP450 monooxygenases, and antioxidant enzymes, whereas BDE-209 stress induced upregulation of secreted hydrolases, lipoxygenases, and cAMP/Ras signaling components. Collectively, this study provides a robust, stress-tolerant fungal strain for efficient BFR degradation and establishes a foundation for the bioremediation of BFR-contaminated sites under non-sterile conditions.

Antifungal potential of the novel antimicrobial peptide N1-6L7A against Penicillium digitatum in Citrus sinensis (L.) Osbeck and its mechanism of action

Jiayi Song,Maomao Ma,Yijian Wu,Yan Hu,Xinyi Wu,Peipei Guan,Bin Zeng

Green mold, caused by Penicillium digitatum , is one of the most destructive postharvest diseases of citrus fruits, leading to significant rot and economic losses. In this study, the antifungal activity and underlying mechanism of the novel antimicrobial peptide (AMP) N1–6L7A against P. digitatum were investigated. Additionally, the potential application of N1–6L7A for controlling postharvest green mold in Citrus sinensis (L.) Osbeck was examined. N1–6L7A exhibited potential antifungal efficacy by inhibiting both mycelial growth and spore germination of P. digitatum . Furthermore, it demonstrated excellent thermal stability, pH tolerance, and biosafety. The results of fluorescence microscopy assay, cell component leakage, and microscopic observations revealed that N1–6L7A severely damaged the fungal membrane of P. digitatum , leading to the release of intracellular nucleic acids and proteins. N1–6L7A treatment also induced excessive intracellular reactive oxygen species (ROS) production, collapse of mitochondrial membrane potential, and decreased activities of malate dehydrogenase (MDH) and ATPase. Gel retardation assay indicated that P. digitatum -DNA was a target of N1–6L7A. Furthermore, transcriptomic analysis suggested that N1–6L7A might inhibit the growth of P. digitatum by blocking the expression of genes related to the ribosome, amino acid metabolism, and pyrimidine metabolism. Meanwhile, the peptide potentially promoted the expression of genes involved in steroid biosynthesis. Notably, in vivo assays confirmed that N1–6L7A effectively controlled P. digitatum infection in citrus fruits and enhanced host resistance by activating defense-related enzymes. Collectively, our results suggested that N1–6L7A could be used as an alternative fungicide to prevent green mold in citrus fruits.

Effects of cellulose-degrading fungus Penicillium griseofulvum on the structure characteristics and adsorption properties of soluble dietary fiber from Citrus aurantium L

Jie Tang,Chaoran Yang,Xiaoting Qin,Jingyu Si,Xuewen Dong,Xiaobo Hu,Qiang Yu

Citrus aurantium L. (CAL) was a potential low-cost source of soluble dietary fiber (SDF). To make full use of this resource, a considerable efficiency cellulose degrading strain ( Penicillium griseofulvum ) was screened from soil humus and used in the modification of CAL-SDF. After fermentation by Penicillium griseofulvum , the structure characteristics and adsorption properties of CAL-SDF were improved. Under the optimum fermentation conditions, the yield of SDF enlarged from 5.47% to 20.0%. Scanning electron microscope (SEM) and Fourier infrared spectroscopy (FTIR) analysis showed that the microstructure became loose and porous after fermentation, unfermented SDF (U-SDF) and fermented SDF (F-SDF) had characteristic intake peaks of polysaccharide . The results of X-ray diffraction (XRD) and thermogravimetric analysis (TGA) showed that the crystallization degree (from 16.50% to 23.15%) of SDF was improved by fermentation, which increased the thermal stability of SDF. The monosaccharide composition and average molecular weight (Mw) results showed that fermentation altered the monosaccharide percentage of SDF as well as significantly reduced the molecular weight (from 576,914 to 112,977 Da). Together, these alterations reduced the water holding capacity (from 11.31 to 1.31 g/g) and oil holding capacity (from 14.24 to 12.86 g/g) of SDF, but significant ( P  < 0.05) increased water solubility (from 89.35% to 92.09%), the glucose dialysis retardation index (GDRI), cholesterol adsorption capacity (CAC) and nitrite adsorption capacity (NIAC). In summary, Penicillium griseofulvum could be used as a potential strain for the modification of the CAL-SDF. Moreover, F-SDF may be used as a functional food component, which will increase CAL's value proposition.

Fermentation of grapefruit peel by an efficient cellulose-degrading strain, (Penicillium YZ-1): Modification, structure and functional properties of soluble dietary fiber

Wenjie Ma,Yuting Liang,Huasi Lin,Yi Chen,Jianhua Xie,Fengling Ai,Ziwen Yan,Xiaobo Hu,Qiang Yu

In the study, a highly efficient cellulose-degrading strain was screened, which was identified as a fungus in the genus Penicillium sp., named YZ-1. The content of soluble dietary fiber was greatly increased by the treatment of this strain. In addition, the effects of soluble dietary fiber from high-pressure cooking group (HG-SDF), strain fermentation group (FG-SDF) and control group (CK-SDF) on the physicochemical structure, and in vitro hypolipidemic activity were investigated. The results showed that the physicochemical structure of the raw materials was improved after fermentation, and FG-SDF exhibited the loosest structure, the highest viscosity and thermal stability. Furthermore, compared to CK-SDF and HG-SDF, FG-SDF showed the most significant improvement in functional properties, including cholesterol adsorption capacity (CAC), inhibition of pancreatic lipase activity (LI) and mixed bile acid adsorption capacity (BBC). Overall, these findings will provide new insights into dietary fiber modification and improve the comprehensive use value of grapefruit by-products.

G protein-cAMP signaling pathway mediated by PGA3 plays different roles in regulating the expressions of amylases and cellulases in Penicillium decumbens

Yibo Hu,Guodong Liu,Zhonghai Li,Yuqi Qin,Yinbo Qu,Xin Song

Heterotrimeric G proteins (G proteins) have been extensively investigated for their regulatory functions in morphogenesis and development in filamentous fungi. In addition, G proteins were also shown to be involved in the regulation of cellulase expression in some fungi. Here, we report the different regulatory effects of PGA3, a group III G protein α subunit, on the expressions of amylases and cellulases in Penicillium decumbens . Deletion of pga3 resulted in impaired amylase production and significantly decreased transcription of the major amylase gene amy15A . Supplementation of exogenous cAMP or its analog dibutyryl-cAMP restored amylase production in Δ pga3 strain, suggesting an essential role of PGA3 in amylase synthesis via controlling cAMP level. On the other hand, the transcription of major cellulase gene cel7A-2 increased, nevertheless cellulase activity in the medium was not affected, in Δ pga3 . The above regulatory effects of PGA3 are carbon source-independent, and are achieved, at least, by cAMP-mediated regulation of the expression level of transcription factor AmyR. The functions of PGA3 revealed by gene deletion were partially supported by the analysis of the mutant carrying dominantly-activated PGA3. The results provided new insights into the understanding of the physiological functions of G protein-cAMP pathway in filamentous fungi.

Metabolomics analysis based on UHPLC-QTOF-MS/MS to explore the synthesis mechanism and culture conditions optimization of Penicillium EF-2 exopolysaccharide

Gang Wang,Jia Kong,Tuo Leng,Weidong Zhang,Ting Chen,Xizhe Xu,Zhibing Huang,Jianhua Xie

Penicillium exopolysaccharide (EPS) inhibits galactose lectins and enhances immunity. However, EPS production is low and its synthesis mechanism remains unclear. Penicillium EF-2 strains with high EPS production were selected for this study, and Penicillium fermentation conditions were subsequently improved. The optimal culture conditions were 30 g/L lactose, 6 g/L yeast extract powder, 4 d seed age, 10 % inoculation amount, 3 d of secondary fermentation time, and the final EPS yield was 3.97 g/L. UHPLC-Q-TOF-MS/MS was used to explore the mechanism of EPS synthesis at the metabolic level. Optimal carbon source: lactose and optimal nitrogen source: yeast extract can provide precursors for EPS synthesis through related metabolic pathways. Moreover, regulating the energy, vitamin, and lipid metabolic pathways created favourable conditions for EPS synthesis and secretion. These findings explain the mechanism of EPS synthesis at the metabolic level and provide a theoretical basis for optimising and industrialising EPS production.

Sodium trans-2-hexenylate damages the mitochondrial function of Penicillium digitaum and plays a significant role in citrus fruit postharvest disease control

Qiuli OuYang,MingChen Yang,Yonghua Zhang,Lu Li,Xiaoli Tan,Nengguo Tao

Huge postharvest citrus infections are produced by Penicillium digitatum . The antifungal activity of sodium trans -2-hexenylate (STH) against the P. digitatum growth and possible mechanism were investigated. The minimal inhibitory and fungicidal concentrations of STH against P. digitatum were 0.4 and 0.8 g L –1 , respectively. STH decreased the green mold incidence of Ponkan fruit without negatively impacting fruit quality. STH altered and distorted the external appearance of P. digitatum mycelia rather than the cell wall. Correspondingly, STH decreased the lipid and ergosterol levels while degrading the cell membrane integrity. Meanwhile, STH affected mitochondrial function by lowering the mitochondria membrane potential (MMP) and ATP levels, as well as malate dehydrogenase (MDH) and succinate dehydrogenase (SDH) activities. Furthermore, MMP damage occurred earlier than that of cell membrane damage. Our current study showed that STH may be an acceptable substitute to chemical fungicide in postharvest disease control in citrus fruit and its underlying inhibitory mechanism is attributed to its ability to impair the mitochondrial function of P. digitatum .

New Drimane Sesquiterpenes and Polyketides from Marine-Derived Fungus Penicillium sp. TW58-16 and Their Anti-Inflammatory and α-Glucosidase Inhibitory Effects

Xiaoshuang Gou,Danmei Tian,Jihua Wei,Yihan Ma,Yixue Zhang,Mei Chen,Wenjuan Ding,Bin Wu,Jinshan Tang

Marine fungi-derived natural products represent an excellent reservoir for the discovery of novel lead compounds with biological activities. Here, we report the identification of two new drimane sesquiterpenes (1and2) and six new polyketides (3–8), together with 10 known compounds (9–18), from a marine-derived fungusPenicilliumsp. TW58-16. The planar structures of these compounds were elucidated by extensive 1D and 2D NMR, which was supported by HR-ESI-MS data. The absolute configurations of these compounds were determined by experimental and calculated electronic circular dichroism (ECD), and their optical rotations compared with those reported. Evaluation of the anti-inflammatory activity of compounds1–18revealed that compound5significantly inhibited the release of nitric oxide (NO) induced by lipopolysaccharide (LPS) in RAW264.7 cells, correlating with the inhibition of expression of inducible nitric oxide synthase (iNOS). In addition, we revealed that compounds1,3–6,14,16,and18showed strongα-glucosidase inhibitory effects with inhibition rates of 35.4%, 73.2%, 55.6%, 74.4%, 32.0%, 36.9%, 88.0%, and 91.1%, respectively, which were comparable with or even better than that of the positive control, acarbose. Together, our results illustrate the potential of discovering new marine-based therapeutic agents against inflammation and diabetes mellitus.Keywords:marine-derived fungus;drimane sesquiterpene;polyketide;anti-inflammatory effect;α-glucosidase inhibitory effect

The effect of a consortium of Penicillium sp. and Bacillus spp. in suppressing banana fungal diseases caused by Fusarium sp. and Alternaria sp.

Win T.T.,Bo B.,Malec P.,Fu P.

AimsThis study sought to utilize indigenous soil micro‐organisms to suppress wilt‐causing fungal pathogens of the banana. Methods and ResultsFungal pathogens were isolated from wilt‐affected rhizospheric soil, and potential antagonistic bacterial strains were isolated from healthy rhizospheric soil in the same area from which fungal pathogens were isolated. The antifungal activity of isolated micro‐organisms against fungal pathogens was studied both in vitro and in vivo against fungal pathogens. It was found that Fusarium oxysporum and Alternaria sp. were pathogenic, while Penicillium sp., Bacillus velezensis and Bacillus subtilis were antagonistic. Moreover, it was seen that B. velezensis, B. subtilis and Penicillium sp. inhibited the growth of the two fungal pathogens in both in vitro and in vivo experiments. Further investigation indicated that B. velezensis, B. subtilis and Penicillium sp. were able to produce enzymatic antifungal compounds (chitinase and β‐1,3‐glucanase). The spray application around rhizome revealed that a combination of Bacillus spp. and Penicillium sp. in greenhouse conditions gave the highest reduction in disease severity by up to 60% to both fungal pathogens among the treatments.ConclusionsBanana disease is seen to be induced not only by F. oxysporum but also by Alternaria sp. The isolated indigenous micro‐organisms can effectively control both the pathogens. The combination of isolated antagonistic micro‐organisms has thus demonstrated substantial potential for suppressing banana disease.Significance and Impact of the StudyAn antagonistic consortium isolated in this study has demonstrated remarkable potential for controlling fungal diseases caused by Fusarium sp. and Alternaria sp. Therefore, the use of indigenous microflora to improve disease suppression of banana plants against soil‐borne pathogens is a preferable approach.

The cellulose binding region in Trichoderma reesei cellobiohydrolase I has a higher capacity in improving crystalline cellulose degradation than that of Penicillium oxalicum

Jian Du,Xiu Zhang,Xuezhi Li,Jian Zhao,Guodong Liu,Baoyu Gao,Yinbo Qu

Commercial cellulase preparations for lignocellulose bioconversion are mainly produced by the fungus Trichoderma reesei . The maximum cellulose conversion of T. reesei cellulase mixture was 15%–20% higher than that of Penicillium oxalicum in the hydrolysis of corncob residue and Avicel. Nevertheless, both preparations hydrolyzed more than 92% of cellulose in NaOH-mercerized Avicel. When added to Avicel hydrolysis residue that was less reactive to P. oxalicum cellulases, cellobiohydrolase I (CBH I) from T. reesei resulted in a higher cellulose conversion than its homologous proteins from P. oxalicum and Aspergillus niger at the same protein loadings. Further domain exchange experiment attributed the high hydrolytic efficiency of T. reesei CBH I to its inter-domain linker and cellulose-binding domain. The results in part explained the superior performance of T. reesei cellulases on the degradation of native crystalline cellulose, and highlighted the important role of cellulose-binding region in determining the degree of hydrolysis by cellulases.

Penicillium expansum lipase-coated magnetic Fe3O4–polymer hybrid hollow nanoparticles: a highly recoverable and magnetically separable catalyst for the synthesis of 1,3-dibutylurea

Jun Liu,Wenjing Wang,Huiwen Liu,Yaoliang Zhou,Haibo Zhang,Xiaohai Zhou

Herein, amino-epoxy supports were innovatively imported onto magnetic nanoparticles (Fe3O4–polymer hybrid nanospheres) for immobilizing enzymes. This new support has a coating layer with dual functional groups (epoxy and amino-epoxy). Consequently, this support has great anionic exchange power and a high number of epoxy groups. The acquired immobilized Penicillium expansum lipase in combination with this heterofunctional support represents a novel class of heterogeneous catalyst towards the synthesis of 1,3-dibutylurea from ethylene carbonate and butylamine, which has not been very commonly catalyzed by enzymes. After optimization of the reaction conditions, the yield of 1,3-dibutylurea was 77% under solvent free conditions at 60 °C. Moreover, after completion of reaction, the catalyst was simply recovered by an external conventional magnet and recycled without significant loss in the catalytic activity (up to ten cycles).

Prochloraz-Based Hydrogen-Bonded Formulations with Salicylic Acid, Ascorbic Acid, or Sorbic Acid for Postharvest Control of Penicillium expansum in Korla Fragrant Pears

Yong Li,Muhammad Ahmad,Xin Gao,Shuren Liu,Rongshan Xu,Haiting Hao,Lan Wang,Hongqiang Dong,Jiamei Zhu

Postharvest diseases caused by fungal pathogens remain a major constraint on fruit storage and quality. Prochloraz (PRO) is widely used for disease control, but its practical application is limited by poor water solubility, relatively high toxicity, and the absence of antioxidant activity. In this study, three PRO-based hydrogen-bonded formulations were prepared using salicylic acid (SA), ascorbic acid (AA), or sorbic acid (SOR) as co-formers at a 1:1 molar ratio. The formulations were characterized by1H NMR, and their physicochemical properties, antifungal activity, disease control efficacy, and acute toxicity to zebrafish larvae were evaluated. All three formulations formed through hydrogen bonding and showed improved water solubility compared with pure PRO. PRO-SA exhibited the strongest antifungal activity againstPenicillium expansum(60.57% reduction in EC50) and the best disease control on Korla fragrant pears at 400 mg/L. PRO-AA showed the greatest reduction in acute toxicity to zebrafish larvae (8.04-fold) and a 54-fold increase in water solubility. PRO-SOR provided a balanced improvement in antifungal activity (41.33% reduction in EC50) and moderate toxicity reduction (1.48-fold). These results indicate that the choice of co-former determines the functional profile of PRO-based formulations, allowing tailored optimization for antifungal efficacy, reduced acute toxicity, or a balanced combination. This work offers a practical strategy for improving the performance of existing fungicides.
