Efficient direct electrocatalysis of nano-dodecahedron for the highly sensitive and selective detection of rutin

Guangquan Mo,Qiuxia Li,Liu Zhu,Zhixiong Ruan,Songqing Mei

Herein, hollow and porous nano-dodecahedron embedded with cobalt oxide nanoparticles (HPND-CN) was prepared for the detection of rutin (Rut). HPND-CN was synthesized by the pyrolysis of zeolitic imidazolate frameworks (ZIFs). HPND-CN displayed a uniform rhombic dodecahedral morphology. For the main architecture, the original framework was carbonized after pyrolysis leading to a porous graphitized polyhedron with nitrogen doping. Notably, in the polyhedron, spherical cobalt oxide nanoparticles with more CoO and less Co 2 O 3 were evenly implanted. With embedded oxide nanoparticles, active doping species, polyhedral morphology, and porous graphitized structure, HPND-CN displayed a strong electrocatalysis towards the redox of Rut. A pair of well-defined symmetric peaks for Rut redox was clearly observed. Based on the electrocatalysis of HPND-CN, an efficient Rut sensor was developed with a low detection limit of 1.9 nM and broad detection range from 7.0 nM to 1.0 μM. HPND-CN prepared sensor also exhibited a superb selectivity showing no interference from common inorganic ions, carbohydrates, and amino acids. Besides, excellent repeatability, reproducibility and reliability were observed. Moreover, HPND-CN sensor was successfully applied to the measurement of Rut in health-care tablets with convinced results, indicating a substantial electrochemical sensing platform for the electrocatalysis and detection of macromolecules.

GQDs/PEDOT Bilayer Films Modified Electrode as a Novel Electrochemical Sensing Platform for Rutin Detection

Rongqian Meng,Qiaoling Li,Shengjian Zhang,Jianke Tang,Chunlei Ma,Riya Jin

A glassy carbon electrode (GCE) modified with graphene quantum dots (GQDs) and poly (3, 4- ethylenedioxythiophene) (PEDOT) bilayer films (GQDs/PEDOT/GCE) was constructed and used as a novel electrochemical sensor for rutin detection. The GQDs/PEDOT/GCE sensor was fabricated by two- step electrodeposition and was characterized by transmission electron microscopy (TEM), scanning electron microscopy (SEM), electrochemical impedance spectroscopy (EIS), and cyclic voltammetry (CV). Compared with individual GQDs/GCE and PEDOT/GCE, the current response of the composite electrode to rutin increased by about 10 times and 3 times respectively. The composite electrode exhibited a strong electrical response toward rutin redox due to the cooperative action of the large surface area of the GQDs which allowed strong analyte adsorption and the high electroconductivity of PEDOT. The current response of the GQDs/PEDOT/GCE electrochemical sensor was more linear at rutin concentrations between 0.05 μM and 10 μM and had a detection limit of 11 nM. The sensor also presented a high selectivity, high stability, and excellent recovery rate and could be used for the effective and reliable analysis of rutin in real samples.

Preparation of Nb2O5 decorated Reduced Graphene Oxide nanocomposites and their electrochemical behaviors for efficient rutin detection

Zhao Liang,Zhang Wen,Han Yu,Hu Chenxi

Nb 2 O 5 decorated Reduced Graphene Oxide (Nb 2 O 5 -RGO) nanocomposites have been successfully prepared through a facile hydrothermal process, offering a synergistic hybrid structure that enhances electron-transfer efficiency and exposes abundant interfacial active sites. While Nb 2 O 5 -based materials have shown promise in electrocatalysis, their application in flavonoid sensing, particularly rutin detection, has rarely been reported, representing a key novelty of this work. Electrochemical investigation using cyclic voltammetry (CV) and differential pulse voltammetry (DPV) demonstrates that the Nb 2 O 5 -RGO-modified glassy carbon electrode (Nb 2 O 5 -RGO /GCE) exhibits excellent electrocatalytic activity toward rutin oxidation, with an ultralow detection limit of 8.7 × 10 −9 M (S/N = 3), which is superior to most electrochemical platforms reported to date, further highlighting the high sensitivity. In addition, the sensor shows strong repeatability, long-term stability, and satisfactory recovery (93.6–97.2 %) in real pharmaceutical tablet samples. Benefiting from its efficient charge-transfer pathways and robust structural stability, the Nb 2 O 5 -RGO nanocomposite represents a promising platform for practical electrochemical sensing. This work not only expands the application scope of Nb 2 O 5 -based materials, but also provides new guidance for designing high-performance metal-oxide/graphene hybrid sensors.

Nitrogen and sulfur co-doped reduced graphene oxide-gold nanoparticle composites for electrochemical sensing of rutin

Fen-Ying Kong,Rong-Fang Li,Sheng-Feng Zhang,Zhong-Xia Wang,Heng-Ye Li,Hai-Lin Fang,Wei Wang

The extraordinary biological and physiological properties of rutin have been used in the preparation of therapeutical drugs. So it is very significant to develop a simple and sensitive method for rutin determaination in pharmaceutical samples. Herein, nitrogen and sulfur co-doped reduced graphene oxide attached with gold nanoparticles (NS-rGO/AuNPs) was developed for electrochemical determination of rutin. The NS-rGO was first prepared by a simple one-step thermal annealing method, and AuNPs were embedded onto NS-rGO through the interaction force of Au with N and S atoms. Detailed characterizations showed that the AuNPs are well dispersed and the presence of nitrogen and sulfur are significant. The NS-rGO/AuNPs modified electrode exhibited a significantly enhanced electrocatalytic activity towards rutin compared with NS-rGO, N-rGO and S-rGO modified electrodes. The excellent performance was achieved by the synergistic effect of AuNPs and heteroatoms co-doping. As a consequence, the NS-rGO/AuNPs based sensor showed a wide linear range of 0.2–1400 nM with a nanomolar detection limit of 0.067 nM (S/N = 3). Also the sensor exhibited satisfying selectivity, reproducibility, and stability. Finally, the sensor was successfully used to determine rutin in pharmaceutical tablets.

Fabrication of rutin-protein complexes to form and stabilize bilayer emulsions: Impact of concentration and pretreatment

Ao Huang,David Julian McClements,Shunjing Luo,Tingting Chen,Jiangping Ye,Chengmei Liu

In present study, the impact of rutin concentration and pretreatment (direct mixing versus pH-driven) of rutin-protein complexes to form and stabilize bilayer emulsions was examined. Bilayer emulsions prepared using the pH-driven method contained smaller oil droplets and had a higher resistance to aggregation and creaming than those prepared using the direct method. The concentration of whey protein isolate at the oil droplet surfaces decreased as the rutin concentration was increased. The oil-water interfacial tension decreased as the rutin concentration in the rutin-whey protein isolate complexes increased, reaching a value of 10.4 mN/m at 0.6 mg/mL rutin. Fluorescence spectroscopy suggested that rutin interacted with the whey protein isolate surfaces mainly through hydrophobic interactions. Moreover, there appeared to be more binding sites and stronger binding interactions when the pH-driven method was used to form the complexes, rather than the direct mixing method. This study suggests that rutin may be used to improve the functional performance of whey protein isolate as emulsifiers in bilayer emulsions.

Multi-omics Analysis of Klebsiella pneumoniae Revealed Opposing Effects of Rutin and Luteolin on Strain Growth

Wang Zhibin,Shen Wanxia,Li Yuejiao,Wang Xiaoyun,Zhong Xiaolin,Wang Xing

The emergence of pathogenic bacteria resistant to conventional antibiotics is becoming increasingly challenging. Plant-derived flavonoids are potential alternatives to antibiotics, owing to their antimicrobial properties. However, the molecular mechanisms through which they inhibit the growth of pathogenic microorganisms remain unclear. Therefore, Klebsiella pneumoniae ATCC700603 was separately incubated in two flavonoids to elucidate their inhibitory mechanism. Metabolomic and transcriptomic analyses were performed after 4-h incubation. In total, 5483 genes and 882 metabolites were identified. Compared to the untreated control, rutin and luteolin activated 507 and 374 differentially expressed genes (DEGs), respectively. However, the number of differential abundant metabolites (DAMs) remained the same. The top 10 correlated DEGs and DAMs were identified within each comparative group after a correlation analysis. Rutin induced the accumulation of unique metabolites and suppressed gene expression whereas luteolin did not. Our results explain the disparate effects of these two flavonoids and demonstrate the inhibitory mechanism of rutin on strain growth.

The protective effect of rutin on sciatic nerve injury in acrylamide-exposed rats and its mechanisms

Suqiu Dong,Chunmei Zhang,Yunjue Wang,Shuping Liu,Junhua Yang,Lixia Li,Yuxin Ma,Jing Liu

Rutin (Rut) is a flavonoid with pharmacological activities such as anti-inflammatory and antioxidant. Acrylamide (ACR) is a toxic substance widely found in human life that can induce neurotoxicity. Some studies have confirmed that neurotoxicity caused by ACR induces myelin damage, which in turn causes neurological dysfunction. Therefore, we established a rutin intervention model to investigate the protective effect of Rut on ACR-induced sciatic nerve injury in rats and its mechanism. The results showed that superoxide dismutase (SOD) activity and glutathione (GSH) content increased and lactate dehydrogenase (LDH) activity decreased in the middle and high dose groups of Rut compared with the ACR group, and the expression of Myelin basic protein (MBP), Extracellular-regulated kinase 1/2(ERK1/2), Phosphorylated extracellular regulated kinase 1/2 (P-ERK1/2), and Nuclear factor E−2-associated factor (Nrf2) was promoted in the Rut-protected group, which suggests that Rutin has a protective effect on ACR-induced sciatic nerve injury and that the mechanism of Rutin's protective effect is related to activation of the ERK1/2 pathway and alleviation of oxidative stress injury.

Water solubility-based Rutin encapsulation by HPCD improves its bioavailability, antioxidant capacity and enzyme inhibitory functions

Shengpeng Zhang,Menglin Jiang,Yao Chen,Genyi Zhang

The water solubility of food bioactives is one of the crucial factors influencing their bioavailability and biological functions. The encapsulation and cellular bioavailability of Rutin, as a representative of bioactive compounds with poor water solubility, was studied. The results showed that Rutin encapsulation by hydroxypropyl-β-cyclodextrin (HPCD) formed an inclusion complex in a molar ratio of 1:1. The encapsulation of Rutin by HPCD exhibited a semi-edged form in which the pyran glucose unit and part of the aromatic ring of Rutin are in the cavity of HPCD through hydrogen bonding and hydrophobic interactions. Rutin encapsulation by HPCD substantially increased the water solubility of Rutin by 51 times. The formed nanoscale inclusion complex (IC) also significantly increased the thermal stability of Rutin with a release kinetics fitting the Korsmeyer-Peppas model. The Rutin encapsulation increased the antioxidant property of Rutin (∼50%) and the inhibitory function on glycolytic enzymes of α-amylase. IC also significantly reduced the postprandial glycemic response to cooked starch (peak glucose level of starch, starch + Rutin, starch + IC are 10.3, 7.2 and 4.7 mmol/L) in mice. Further study using a cell model revealed a significantly increased bioavailability (∼10%). Thus, Rutin encapsulation by HPCD significantly increased its water solubility, bioavailability and biological functions, and high water solubility-induced increase of contact opportunities between Rutin and other components (free radicals, and enzymes) in the reaction system might be the underlying mechanism.

Preparation of Rutin–Whey Protein Pickering Emulsion and Its Effect on Zebrafish Skeletal Muscle Movement Ability

Yiting Zhang,Wenyun Xiong,Yijing Ren,Jian Huang,Xiaoying Wang,Ou Wang,Shengbao Cai

Nutritional supplementation enriched with protein and antioxidants has been demonstrated to effectively strengthen skeletal muscle function and mitigate the risk of sarcopenia. Dietary protein has also been a common carrier to establish bioactive delivery system. Therefore, in this study, a Pickering emulsion delivery system for rutin was constructed with whey protein, and its structural characteristics, bioaccessibility, and molecular interactions were investigated. In the in vivo study, zebrafish (n= 10 in each group), which have a high genetic homology to humans, were treated with dexamethasone to induce sarcopenia symptoms and were administered with rutin, whey protein and the Pickering emulsion, respectively, for muscle movement ability evaluation, and zebrafish treated with or without dexamethasone was used as the model and the control groups, respectively. Results showed that the Pickering emulsion was homogeneous in particle size with a rutin encapsulation rate of 71.16 ± 0.15% and loading efficiency of 44.48 ± 0.11%. Rutin in the Pickering emulsion exhibited a significantly higher bioaccessibility than the free form. The interaction forces between rutin and the two components of whey proteins (α-LA and β-LG) were mainly van der Waals forces and hydrogen bonds. After treatment for 96 h, the zebrafish in Picking emulsion groups showed a significantly increased high-speed movement time and frequency, an increased level of ATP, prolonged peripheral motor nerve length, and normalized muscular histological structure compared with those of the model group (p< 0.05). The results of this study developed a new strategy for rutin utilization and provide scientific evidence for sarcopenia prevention with a food-derived resource.

Rapid Separation of Representative Monomer Compound Rutin From Natural Product by Molecularly Imprinted Macroporous Adsorption Resin

Yubai Qi,Xueqin Jin,Yuepeng Yang,Hong Du,Defu Li,Xinying Li

Monomer compounds from natural products are the major source of active pharmaceutical molecules, which provide great opportunities for discovering of new drugs. However, natural products contain a large number of rather complex compounds. It is difficult to obtain high-purity monomer compounds from complex natural products. Herein, we developed molecularly imprinted macroporous adsorption resins (MIRs-PDA) for the selective adsorption of target monomer compounds from natural products. The response surface methodology was employed to optimize the fabrication parameters. The separation performance of MIRs-PDA for the representative monomer compound rutin was thoroughly studied. We found that MIRs-PDA presented large adsorption capacity (107.82 mg/g), high selectivity (selectivity coefficient was 2.90), good repeatability, and reusability. MIRs-PDA can reach 78.5% equilibrium adsorption capacity in 20 min, showing quick binding kinetic behavior. Furthermore, MIRs-PDA showed good selective and specific adsorption performance for rutin from a representative real sample of Fagopyrum tataricum extract solution. The study of the adsorption mechanism showed that the adsorption of rutin on MIRs-PDA was spontaneous and thermodynamically feasible. The kinetic parameters adapted well to the pseudo-second-order kinetic model, and there existed equivalent binding sites in MIRs-PDA. The MIRs-PDA show promising application prospects for the separation of monomer compounds in natural products.

Preparation of blue sulfur quantum dots in ammonia for the detection of Rutin and cell imaging

Qi Ma,Yueyue Zu,Sanhu Zhao,Hailong Li,Jinping Song,Yongsheng Qiao

Sulfur quantum dots (SQDs) have garnered increasing attention for their versatile applications in bioimaging, LEDs, and fluorescence sensing. As a new type of heavy-metal-free fluorescent nanomaterial, developing high quantum yield SQDs using small molecules as capping agents poses a significant challenge. Here, we present a novel synthesis approach of SQDs using (NH 4 ) 2 S x and sulfur powder as sulfur sources, and the small molecule N-acetyl-L-cysteine (NAC) as a stabilizer in a weak alkaline system. The synthesized blue SQDs (B-SQDs) were extensively characterized using various techniques. B-SQDs exhibited a fluorescence quantum yield of 15.55 % and particle sizes ranging from 2.7 nm to 6.2 nm, showcasing exceptional optical stability, dispersibility, low cytotoxicity, and efficient cell uptake. The as-acquired B-SQDs were explored for potential applications in sensing and cell bioimaging. The findings revealed that B-SQDs exhibited high sensitivity and selectivity towards the drug molecule Rutin. The detection limit was calculated as 50 nM. This methodology was effectively employed for Rutin determination in actual urine samples, yielding satisfactory recovery rates ranging from 95.68 % to 103.93 % with a relative standard deviation below 4.28 %. The fluorescence quenching of B-SQDs by Rutin is influenced by the concentration levels of Rutin. At high Rutin concentrations, fluorescence quenching mechanisms include dynamic quenching and static quenching. Conversely, at low Rutin concentrations, it mainly involves a dynamic quenching mechanism and the IFE process. Moreover, B-SQDs could also accurately identify intracellular Rutin. This research offers a robust approach for Rutin detection in biological samples with implications for disease diagnosis and pharmacology.

Insight into the binding mechanism of rutin and lysozyme: Based on spectroscopy and molecular simulation technology

Lili Liu,Le Yang,Hui Chen,Weiwei Cheng,Yue Ding,Feng Xiao

Lysozyme (LYZ) is an excellent natural food preservative and can also be used as a bioactive carrier loading small molecules to enhance its stability and antioxidant properties. This research explored the intricate mechanism of interaction between LYZ and rutin. Multiple spectroscopic techniques was used first to confirm that rutin caused a fluorescence burst in LYZ. LYZ amino acid microenvironment was altered. The main driving forces driving the formation of the complex between rutin and LYZ were hydrogen bonding and van der Waals forces. In addition, the incorporation of rutin improved the overall stability and oxidation resistance of the complexes. The results of molecular docking and molecular dynamics simulation further show that rutin and LYZ are stably bound by hydrogen bonds and other interactions. The investigation contributed precious information for the development of novel natural preservatives and the design of advanced small molecular carriers.
