Solidago canadensisis one of the most destructive invasive plants in China, and current physical and chemical control methods are often costly, inefficient, and ecologically risky, highlighting the urgent demand for green and efficient ecological alternatives. The native vineHumulus scandensexhibits strong allelopathic potential. In this study, allelochemicals from its leaves and stems were extracted using petroleum ether, ethyl acetate, n-butanol, and n-hexane. Their allelopathic effects onS. canadensiswere systematically evaluated through seed germination, seedling growth, and rhizome propagation assays, combined with widely targeted metabolomics. Leaf extracts showed markedly stronger allelopathic inhibition than stem extracts. Among all fractions, the ethyl acetate extract exerted the most potent suppression on seed germination, significantly reducing germination rate and root elongation in a concentration-dependent manner. At 100 g·L−1, leaf extracts—especially the ethyl acetate phase—significantly decreased belowground fresh weight, rhizome number, and new seedling emergence. The ethyl acetate extract also significantly elevated root antioxidant enzyme activities at 25–100 g·L−1, indicating induced oxidative stress. UPLC-MS/MS-based metabolomics identified 186 differential metabolites among extracts. Ten flavonoids were significantly upregulated in the ethyl acetate extract and were predominantly enriched in the kaempferol aglycone biosynthesis I pathway (MetMap 113) and the flavonoid biosynthesis pathway (ko00941), suggesting that these compounds may play a key role in the observed allelopathic inhibition. Collectively, the ethyl acetate extract ofH. scandensleaves effectively suppressesS. canadensisgrowth by inhibiting seed germination and rhizome tillering and by inducing oxidative stress, probably with the 10 candidate flavonoid metabolites, highlighting its strong potential as a biological control agent against this invasive weed.

Diabetic chronic wounds, as a prevalent and serious complication of diabetes, pose serious threats to patient health and survival, underscoring the urgent need for effective therapeutic strategies. Stem cell therapy has emerged as a promising approach, but its efficacy is often compromised by the hostile wound microenvironment, characterized by excessive reactive oxygen species and persistent inflammation. To address this challenge, we developed a nanozyme-based engineering strategy to enhance the therapeutic potential of periodontal ligament stem cells (PDLSCs) based on a bimetallic CuNi-MOF nanozyme exhibiting dual superoxide dismutase- and catalase-like activities. In vitro experiments demonstrated that, the CuNi-MOF nanozyme effectively alleviated oxidative stress and reduced cellular senescence in PDLSCs. Mechanistic investigation revealed the involvement of the JNK/ERK signaling axis in this protective effect. In a diabetic wound model, nanozyme-engineered PDLSCs (E-PDLSCs), exhibited superior healing compared with untreated PDLSCs. Specifically, E-PDLSCs significantly accelerated re-epithelialization, enhanced collagen deposition, and promoted angiogenesis, while also modulating inflammatory cytokine levels and macrophage polarization. Overall, this study presents a cell engineering strategy that integrates antioxidant nanozymes with stem cells for chronic wound treatment, extending the potential applications of nanozyme in regenerative medicine.

During two-dimensional (2D) culture, stem cells gradually lose their proliferative activity and multipotency due to various physicochemical conditions, which significantly hinder the large-scale clinical application of stem cell therapy. In recent years, three-dimensional (3D) cell culture has been increasingly utilized in the field of stem cell expansion owing to its unique advantages. The superhydrophilicity of zwitterionic hydrogels ensures the maintenance of stem cell stemness during their expansion. This study aims to address a key challenge in the large-scale culture of stem cells in vitro: how to sustain their proliferative capacity and multipotency while achieving efficient cell recovery. To this end, we have designed a novel zwitterionic degradable hydrogel based on host-guest interactions as a 3D carrier for the in vitro culture of adipose-derived stem cells (ADSCs). We synthesized the copolymer poly(sulfobetaine-co-cyclodextrin) (P(SBMA-co-CD)) and adamantane-grafted hyaluronic acid (HA-Ada), and a stable hydrogel was rapidly formed by simply mixing solutions of these two polymers. Leveraging the antifouling properties of zwitterionic groups, this hydrogel effectively maintained the long-term stemness expression of ADSCs during culture. More importantly, we utilized the reversibility of host-guest interactions to disrupt the cross-linked structure of the hydrogel by adding competitive monomers, enabling efficient recovery of stem cells under gentle conditions. This process not only achieved a high recovery rate of stem cells but also avoided the damage to cells caused by traditional cell recovery methods. In summary, this study creatively introduced host-guest interactions into zwitterionic hydrogels and successfully applied it to the 3D culture and recovery of stem cells in vitro. This hydrogel demonstrates functional plasticity in stem cell proliferation, culture, and harvest, holding promise for providing more reliable and efficient solutions in the fields of stem cell therapy and tissue engineering.

Background Cellular senescence refers to a condition where cells permanently cease division while maintaining metabolic activity. Doxorubicin (Dox) is known as an agent of induction of cellular senescence. This study aimed to explore the potential role of human umbilical cord mesenchymal stem cell-derived exosomes (hucMSC-Exo) in mitigating Dox induced senescent. Method NIH3T3 cells were treated by various concentrations of Dox with or without hucMSC-Exo, cell morphology, viability, migration, senescence-associated SA-β-Gal staining were monitored. Cellular senescence was induced in C57BL/6J mice via administration of 5 mg/kg Doxorubicin, followed by treatment with hucMSC-Exo or metformin. Assessments included body weight, liver and kidney weight, colon length, SA-β-Gal staining of kidney and skin, molecular biomarkers of aging such as p16 INK4A , p53, and p21 Waf1/Clip1 to evaluate senescence status. Result We found that after the treatment of exosomes or metformin improved several aging-related phenotypes in both mouse and cellular models, including increases in body weight, liver and kidney weights, and the reduction of SA-β-Gal positive cells in kidney and skin tissues as well as cell models. At the molecular level, hucMSC-Exo resulted in the downregulation of inflammatory factors and senescence markers in liver and kidney tissues as well as cell models. Conclusion Our study demonstrates hucMSC-Exo may ameliorate Dox induced senescence either in NIH3T3 cells or in mice.

Mesenchymal stem cells (MSCs) have become an important choice for regenerative medicine because of their specific differentiation ability. However, MSCs are rapidly cleared by direct injection, and living cells need to be embedded in a 3D semipermeable hydrogel matrix to prolong cell retention time to enhance therapeutic effects. Herein, MnO 2 doped alginate hydrogel was prepared for single stem cell encapsulation (SA@MSC-MnO 2 ) with maintaining biological activity. This hydrogel system could accelerate the differentiation potential of MSCs and promoted 60 % repair of bone defects within 8 weeks. Besides, the doping of MnO 2 could react with H 2 O 2 to generate H 2 O and O 2 , thus achieving the goal of reactive oxygen species (ROS) clearing and improving the bone regeneration effects. We believe that this single stem cell hydrogel encapsulation system will further promote the clinical application of stem cell therapy.

Hydrogels are extensively utilized in stem cell-based tissue regeneration, providing a supportive environment that facilitates cell survival, differentiation, and integration with surrounding tissues. However, designing hydrogels for regenerating hard tissues like bone presents significant challenges. Here, we introduce macroporous hydrogels with spatiotemporally programmed mechanical properties for stem cell-driven bone regeneration. Using liquid-liquid phase separation and interfacial supramolecular self-assembly of protein fibres, the macroporous structure of hydrogels provide ample space to prevent contact inhibition during proliferation. The rigid protein fibre-coated pore shell provides sustained mechanical cues for guiding osteodifferentiation and protecting against mechanical loads. Temporally, the hydrogel exhibits tunable degradation rates that can synchronize with new tissue deposition to some extent. By integrating localized mechanical heterogeneity, macroporous structures, surface chemistry, and regenerative degradability, we demonstrate the efficacy of these stem cell-encapsulated hydrogels in rabbit and porcine models. This marks a substantial advancement in tailoring the mechanical properties of hydrogels for stem cell-assisted tissue regeneration.

Extracellular matrix (ECM) derived from mesenchymal stem cells regulates antioxidant properties and bone metabolism by providing a favorable extracellular microenvironment. However, its functional role and molecular mechanism in mitochondrial function regulation and aged bone regeneration remain insufficiently elucidated. This proteomic analysis has revealed a greater abundance of proteins supporting mitochondrial function in the young ECM (Y-ECM) secreted by young bone marrow-derived mesenchymal stem cells (BMMSCs) compared to the aged ECM (A-ECM). Further studies demonstrate that Y-ECM significantly rejuvenates mitochondrial energy metabolism in adult BMMSCs (A-BMMSCs) through the promotion of mitochondrial respiratory functions and amelioration of oxidative stress. A-BMMSCs cultured on Y-ECM exhibited enhanced multi-lineage differentiation potentials in vitro and ectopic bone formation in vivo. Mechanistically, silencing of silent information regulator type 3 (SIRT3) gene abolished the protective impact of Y-ECM on A-BMMSCs. Notably, a novel composite biomaterial combining hyaluronic acid methacrylate hydrogel microspheres with Y-ECM is developed, which yielded substantial improvements in the healing of bone defects in an aged rat model. Collectively, these findings underscore the pivotal role of Y-ECM in maintaining mitochondrial redox homeostasis and present a promising therapeutic strategy for the repair of aged bone defects.

Remarkable exertions are directed to reveal and understand topographic cues that induce cell mechanical sensitive responses including lineage determination. Extracellular matrix (ECM) is the sophisticated ensemble of diverse factors offering the complicated cellular microenvironment to regulate cell behaviors. However, the functions of only a few of these factors are revealed; most of them are still poorly understood. Herein, the focus is on understanding the curved structure in ECM network for regulating stem cell mechanotransduction. A curved nanofiber network mimicking the curved structure in ECM is fabricated by an improved electrospinning technology. Compared with the straight fibers, the curved fibers promote cell bridge formation because of the cytoskeleton tension. The actomyosin filaments are condensed near the curved edge of the non-adhesive bridge in the bridging cells, which generates higher myosin-II-based intracellular force. This force drives cell lineage commitment toward osteogenic differentiation. This study enriches and perfects the knowledge of the effects of topographic cues on cell behaviors and guides the development of novel biomaterials.

Critical-sized bone defects often lead to non-union and full-thickness defects of the calvarium specifically still present reconstructive challenges. In this study, we show that neurotrophic supplements induce robust in vitro expansion of mesenchymal stromal cells, and in situ transplantation of neurotrophic supplements-incorporated 3D-printed hydrogel grafts promote full-thickness regeneration of critical-sized bone defects. Single-cell RNA sequencing analysis reveals that a unique atlas of in situ stem/progenitor cells is generated during the calvarial bone healing in vivo. Notably, we find a local expansion of resident Msx1+ skeletal stem cells after transplantation of the in situ cell culture system. Moreover, the enhanced calvarial bone regeneration is accompanied by an increased endochondral ossification that closely correlates to the Msx1+ skeletal stem cells. Our findings illustrate the time-saving and regenerative efficacy of in situ cell culture systems targeting major cell subpopulations in vivo for rapid bone tissue regeneration.

Mesenchymal stem cells (MSCs) have demonstrated significant therapeutic potential in heart failure (HF) treatment. However, their clinical application is impeded by low retention rate and low cellular activity of MSCs caused by high inflammatory and reactive oxygen species (ROS) microenvironment. In this study, monascus pigment (MP) nanoparticle (PPM) was proposed for improving adverse microenvironment and assisting in transplantation of bone marrow-derived MSCs (BMSCs). Meanwhile, in order to load PPM and reduce the mechanical damage of BMSCs, injectable hydrogels based on Schiff base cross-linking were prepared. The PPM displays ROS-scavenging and macrophage phenotype-regulating capabilities, significantly enhancing BMSCs survival and activity in HF microenvironment. This hydrogel demonstrates superior biocompatibility, injectability, and tissue adhesion. With the synergistic effects of injectable, adhesive hydrogel and the microenvironment-modulating properties of MP, cardiac function was effectively improved in the pericardial sac of rats. Our results offer insights into advancing BMSCs-based HF therapies and their clinical applications.

Stem cell therapies have made great progress in the treatment of diabetic wounds during recent decades, while their short in vivo residence, alloimmune reactions, undesired behaviors, and dramatic losses of cell functions still hinder the translation of them into clinic. Here, inspired by the natural components of stem cell niches, we presented novel microfluidic hydrogel microcarriers with extracellular matrix (ECM)-like composition and adipose-derived stem cells (ADSCs) encapsulation for diabetic wound healing. As the hydrogel was synthesized by conjugating hyaluronic acid methacryloyl (HAMA) onto the Fibronectin (FN) molecule chain (FN-HAMA), the laden ADSCs in the microcarriers showed improved bioactivities and pro-regenerative capabilities. Based on these features, we have demonstrated that these ADSCs microcarriers exhibited significant promotion of neovascularization, follicular rejuvenation, and collagen deposition in a mouse diabetic wound model. These results indicated that the stem cell niche-inspired FN-HAMA microcarriers with ADSCs encapsulation have great clinical potential for diabetic wound treatment.
