Dual application of Shewanella oneidensis MR-1 in green biosynthesis of Pd nanoparticles supported on TiO2 nanotubes and assisted photocatalytic degradation of methylene blue

Xiaojie Song,Xianyang Shi,Mi Yang

Biosynthesised nanocomposites have attracted growing interests attributed to their ‘green’ synthesis nature in recent years. Shewanella oneidensis MR-1, a dissimilatory metal-reducing bacterium, was used to reduce palladium (II) nitrate to palladium (0) nanoparticles (Pd NPs) under anaerobic conditions, resulting in the in situ formation of Pd NPs immobilised on TiO 2 nanotubes (TNTs) (Pd/TNTs nanocomposites). The Pd/TNTs nanocomposites were characterised by transmission electron microscopy, X-ray diffraction, X-ray photoelectron spectroscopy, energy dispersive X-ray, and electron spin resonance, respectively. The results indicated that Pd NPs are successfully grown on the TNTs without aggregation. Photocatalytic degradation of methylene blue (MB) by Pd/TNTs nanocomposites under simulated sunlight was also investigated. Pd/TNTs nanocomposites had photocatalytic efficiency superior to that of single TiO 2 nanomaterials. The photocatalytic activity of Pd/TNTs nanocomposites can be enhanced by S. oneidensis MR-1. The results showed that after only 10 min, the degradation ratio of MB reached 98.7% by Pd/TNTs nanocomposites when simultaneously assisted with S. oneidensis MR-1.

Quorum sensing-driven biofilm cathode of Shewanella oneidensis for enhanced uranium adsorption: Elucidating the role of exogenous AHLs and validation in actual uranium-contaminated groundwater

Jing Sun,Hong Shu,Xiaoyan Wu,Tingting Liu,Guangyue Li,Zhao Cui,Ting Li,Xiaowei Zhou,Zhongran Dai,Qian Li

Large quantities of low-concentration uranium-contaminated water from the nuclear industry poses a critical environmental challenge due to the limitations of current methods on removal efficiency and adsorption selectivity. This study developed a novel biofilm cathode for enhanced uranium adsorption by leveraging the quorum sensing (QS) system of Shewanella oneidensis MR-1. Biofilm formation was induced using exogenous acyl-homoserine lactones (AHLs), with C4-HSL (10 μmol/L) identified as the most efficient signal molecule. It significantly improved biofilm properties, increasing thickness by 107.7 % versus the control without AHLs, boosting extracellular protein content, and increasing the proportion of living cells. Microstructure analysis (CLSM) revealed a “base-polysaccharide-gel” three-dimensional structure, where proteins, lipids, and β-polysaccharides form a functional gel layer that provides the primary functional matrix for uranium binding. The C4-HSL-induced biofilm exhibited a 43.4 % higher reduction peak current and a 33.3 % decrease in electron transfer resistance, confirming improved electron transfer efficiency. Furthermore, coupling CLSM, SEM-EDS, FT-IR and XPS analysis indicated uranium capture was primarily dominated by complexation/coordination with functional groups on extracellular polymeric substances (EPS), supplemented by electrochemical reduction of 25.91 % soluble U(VI) to insoluble U(IV). Accordingly, a possible mechanism model of QS-driven biofilm cathode is proposed for optimizing biofilm structure and enhancing uranium capture. Finally, verification tests in actual uranium-contaminated groundwater demonstrated the biofilm cathode exhibited exceptional performance with a 99.4 % uranium recovery rate and high selectivity (K d,U at 34.57 L/g). These findings highlight the significant potential of AHLs-triggered QS as a powerful strategy to optimize bioelectrochemical properties for highly-efficient uranium remediation and resource recovery.

Synergistic antibacterial effects and mechanisms of different phenolic acids against Shewanella putrefaciens from fish, with emphasis on the combination of gallic acid and caffeic acid

Qiuying Li,Chenyang Jia,Jiaxing Sun,Tong Sun,Jianrong Li,Xuepeng Li

The strong spoilage capability and cold adaptability of Shewanella putrefaciens present a serious threat to the quality and safety of fish. This study aimed to investigate the synergic antibacterial effects of six phenolic acids (Gallic acid, GA; Caffeic acid, CA; Protocatechuic acid; Ferulic acid; Chlorogenic acid; p-Coumaric acid) in pairs against S . putrefaciens , and to illustrate the antibacterial mechanisms. The fractional inhibitory concentration index (FICI) of 15 combinations of phenolic acids against S. putrefaciens ranged from 0.5-0.75, suggesting synergistic or partial synergistic antibacterial effects of these phenolic acids. Time-kill curves and morphologic observation demonstrated that GA and CA (GC) exhibited the strongest synergistic effect against S. putrefaciens . GC synergistically damaged the structure integrity of S. putrefaciens , leading to the leakage of intracellular substances and inhibiting the activity of Na + K + -ATPase and respiratory chain dehydrogenase. Both GA and CA could bind to DNA and cause changes in DNA content. Proteomics analysis revealed that 399 proteins were affected in the GC groups, a number significantly higher than that in the single GA and CA groups. GC inhibited dissimilatory nitrate and sulfate reduction, and enhanced fatty acid degradation and assimilatory sulfate reduction, which might cause the imbalance of energy supply and ROS accumulation. Meanwhile, the antioxidant-related enzymes, efflux systems and some two-components systems were upregulated, while chemotaxis were inhibited. GC synergistically inhibited S. putrefaciens in turbot, and retarded the spoilage process. The results revealed multi-targeted synergistic antibacterial mechanisms of GC against S. putrefaciens , suggesting GC might be a good strategy for controlling S. putrefaciens and fish spoilage.

Carbon nanotubes mediate electron transfer between Shewanella oneidenis MR-1 and nano zero-valent iron to enhance the elimination of Cr(VI) from aqueous media

Haoran Tian,Qingyu Wu,Qiuju Wu,Chengyou Sun,Ruimei Zhang,Jie Wei,Haiyin Xu,Zhiming Liu,Chao Huang,Ping Wang

Nano zero-valent iron (nZVI) has been widely employed for the elimination of heavy metals from wastewater. However, the practical application of nZVI is greatly limited due to its fast aggregation and surface passivation. In this study, a carbon nanotube-supported nZVI composite (nZVI@CNTs) was synthesized and coupled with Shewanella oneidensis MR-1 to enhance the removal of Cr(VI) from aqueous media. The results demonstrated that the coupling of nZVI@CNTs and MR-1 (nZVI@CNTs/MR-1) exhibited superior performance in eliminating Cr(VI) compared to using nZVI@CNTs or MR-1 alone. Electrochemical characterization demonstrated that CNTs acting as electron shuttles improve the electron transfer from MR-1 to nZVI, thus facilitating the passivation layer dissolution. XRD analysis proved that MR-1 can effectively promote the dissolution of the Fe(III) passivation layer with the existence of CNTs. FTIR and XPS analyses confirmed that CNTs contain many functional groups (such as C − C and C = O) related to extracellular electron transfer, thus promoting the electron transfer of MR-1 to nZVI. Three-dimensional excitation-emission matrix (3D-EEM) analysis showed that the extracellular polymeric substances (EPS) generated from MR-1 participated in the Cr(VI) elimination. The mechanism analysis demonstrated the Fe(II) dissolved from the nZVI passivation layer reduces Cr(VI) to Cr(III) and then removes it by co-precipitation, and the EPS secreted by MR-1 also participates in the Cr(VI) adsorption and reduction. Generally, the coupling of nZVI@CNTs and MR-1 successfully addresses the drawbacks of nZVI and demonstrates a remarkable ability to remove Cr(VI).

Plasmonic Probing Single-Cell Bio-Current Waves with a Shrinking Magnetite Nanoprobe

Zhuodong Tang,Rui Liu,Xueqin Chen,Di Gao,Jian-Rong Zhang,Jun-Jie Zhu,Zixuan Chen

Probing of the single-cell level extracellular electron transfer highlights the maximum output current for microbial fuel cells (MFCs) at hundreds of femtoampere per cell, which is difficult to achieve by existing devices. Past studies focus on the external factors for boosting charge-extraction efficiency from bacteria. Here, we elucidate the intracellular factors that determine this output limit by monitoring the respiratory-driven shrinking kinetics of a single magnetite nanoprobe immobilized on a single Shewanella oneidensis MR-1 cell with plasmonic imaging. Quantified dissolving of nanoprobes unveils a previously undescribed bio-current fluctuation between 0 and 2.7 fA on a ∼40 min cycle. Simultaneously tracing of endogenous oscillations indicates that the bio-current waves are correlated with the periodic cellular electrokinesis. The unsynchronized electron transfer capability in the cell population results in the mean current of 0.24 fA per cell, significantly smaller than in single cells. It explains why the averaged output current of MFCs cannot reach the measured single-cell currents. This work offers a different perspective to improve the power output by extending the active episodes of the bio-current waves.

A Novel Ag@AgCl Nanoparticle Synthesized by Arctic Marine Bacterium: Characterization, Activity and Mechanism

Shuang Li,Hui Zhang,Bailin Cong,Peiqing He,Wenqi Liu,Shenghao Liu

An additive- and pollution-free method for the preparation of biogenic silver and silver chloride nanoparticles ([email protected]NPs) was developed from the bacteriaShewanellasp. Arc9-LZ, which was isolated from the deep sea of the Arctic Ocean. The optimal synthesizing conditions were explored, including light, pH, Ag+concentration and time. The nanoparticles were studied by means of ultraviolet-visible (UV-Vis) spectrophotometry, energy dispersive spectrometry (EDS), X-ray diffraction (XRD) and inductively coupled plasma optical emission spectrometers (ICP-OES). The transmission electron microscope (TEM) showed that the nanoparticles were spherical and well dispersed, with particle sizes less than 20.00 nm. With[email protected]nanoparticles, the kinetic rate constants for congo red (CR) and rhodamine B (RhB) dye degradation were 2.74 × 10−1min−1and 7.78 × 10−1min−1, respectively. The maximum decolourization efficiencies of CR and RhB were 93.36% and 99.52%, respectively.[email protected]nanoparticles also showed high antibacterial activities against the Gram-positive and Gram-negative bacteria. The Fourier transform infrared spectroscopy (FTIR) spectrum indicated that the O-H, N-H and -COO- groups in the supernatant of Arc9-LZ might participate in the reduction, stabilization and capping of nanoparticles. We mapped the schematic diagram on possible mechanisms for synthesizing[email protected]NPs.Keywords:[email protected]nanoparticles;synthesis mechanisms;Shewanellasp.;dye reduction;antibacterial activity

The Combined Antibacterial Mechanism of Ferulic Acid and ε-Polylysine Hydrochloride in Shewanella putrefaciens and the Effect of Their Application on the Storage Quality of Refrigerated Crayfish (Procambarus clarkii) with Plasma-Activated Water

Yue Cui,Tengteng Zhang,Dandan Zhao,Sai Gao,Yinchu Liu,Xinyu Yang,Han Lu,Xiaoguang Gao

This study aimed to investigate the mechanism underlying the synergistic antimicrobial effect of ferulic acid (FA) and ε-polylysine hydrochloride (PL) on Shewanella putrefaciens (S. putrefaciens) and their application on crayfish (Procambarus clarkii). The treatment with FA and PL exhibited a strong synergistic inhibitory effect against S. putrefaciens. The combination of 1/4 Minimum Inhibitory Concentration (MIC) FA and 1/4 MIC PL was the most effective, damaging the cell structure and inhibiting the growth ofS. putrefaciens. Plasma-activated water (PAW) can induce microbial inactivation through physical action. In addition, treatments with FA, PL, PAW, PAW-FA, PAW-PL, and PAW + PL-FA substantially decreased total viable counts (TVCs), total volatile base nitrogen (TVB-N), the thiobarbituric acid value (TBA), and the juice loss rate of crayfish, with FA-PL showing the best effect. This study confirmed the antimicrobial efficacy of PL, FA, and PAW, indicating their potential as effective preservatives for controlling spoilage in freshwater crustaceans.
