Superior aqueous dispersibility mitigates nanoparticle toxicity in biological systems while preventing agglomeration. Herein, a hydrogen-bond-mediated aqueous synthesis strategy is proposed, utilizing β-cyclodextrin (β-CD) and sodium citrate (SC) to directly fabricate metal oxide nanoparticles in a single step. This approach not only suppresses particle aggregation but also preserves the molecular integrity of surface modifiers under high-temperature reaction conditions. Taking the preparation of gadolinium oxide as an example. Through modifier molecules on Gd 2 O 3 surfaces, the hydrophilicity is markedly enhanced, enabling stable dispersion in aqueous media, which is a critical prerequisite for biomedical applications. This green synthesis method, regulated by hydrogen-bonding interactions, overcomes safety concerns associated with organic solvents in traditional solvothermal techniques and gadolinium ion leakage in co-precipitation methods, while eliminating complex post-modification steps. The longitudinal relaxation rate (r1) of Gd 2 O 3 UPs-220, prepared by this method, achieves 8.43 m m −1 s −1 at a magnetic field strength of 7.0 T, demonstrating excellent magnetic resonance imaging (MRI) enhancement performance. This synthetic strategy provides an approach for preparing water-dispersible metal oxides.

The development of highly efficient magnetic resonance imaging (MRI) contrast agent contributes to the early and accurate diagnosis of cancer. Herein, we developed a novel iron oxide and gadolinium oxide (IO/GdO) nanocomposite as T 1 -T 2 dual modal MRI contrast agent through hydrothermal synthesis method. Subsequently, tumor-targeting ligand (PDGFB-PEG) was further conjugated to the surface of IO/GdO nanocomposite (PDGFB-pIO/GdO) to fabricate tumor-targeting MRI probe for accurate diagnosis of orthotopic prostatic cancer. IO/GdO nanocomposite exhibited obvious mesoporous structure, which would promote the contact between water molecules and Gd or Fe centers and enhance the T 1 relaxation rate (r 1 ) and T 2 relaxation rate (r 2 ) of IO/GdO composite. In addition, the r 1 and r 2 values of IO/GdO composite were up to be 11.6 mM −1 s −1 and 102.7 mM −1 s −1 , respectively, and the r 2 /r 1 ratio of IO/GdO nanocomposite was calculated to be 8.85, demonstrating a great potential as dual-mode MRI contrast agent. Further study indicated that PDGFB-pIO/GdO exhibited an excellent biocompatibility. Besides, PDGFB-pIO/GdO presented a higher accumulation in tumor than untargeted pIO/GdO, confirming its excellent in vivo tumor-targeting ability. Lastly, in orthotopic prostatic cancer model, the systemic delivery of PDGFB-pIO/GdO significantly enhanced T 1 - and T 2 -weighted signal of orthotopic prostatic cancer, achieving high quality of MR images and realizing accurate diagnosis of orthotopic prostatic cancer. Collectively, we proposed a rational designed nanoplatform, PDGF-B conjugating mesoporous IO/GdO nanocomposites, suggesting potential for diagnostic identification of prostate cancer via T 1 -T 2 dual-modal MRI contrast enhancement.

Peritoneal dialysis-associated peritonitis (PDAP), primarily caused by bacterial infections, is a severe complication and a major contributor to mortality in peritoneal dialysis patients. Early detection of bacterial infections and prompt therapeutic interventions are critical for effective PDAP management. Herein, we report the development of a novel apoptotic body (AB)-hitchhiked manganese-based T1-weighted magnetic resonance imaging (MRI) probe (PMA) for noninvasive and in situ bacterial detection in the abdominal cavity, offering a potential early warning system for PDAP. When administered to PDAP model mice, the PMA probes selectively accumulated in bacteria-infected regions via AB-mediated macrophage targeting. Subsequently, the PMA probes disintegrated in the acidic microenvironment of bacterial infection, which resulted in the MRI signal ON, therefore imaging the bacterial infection in vivo accurately. Remarkably, due to the protein-assisted T1-weighted signal amplification, the PMA probe-based MRI strategy detected bacterial loads in the abdominal cavity approximately 10 times lower than those identified by biochemical analysis for PDAP diagnosis. This approach achieves sensitive bacterial detection, holding great potential for the early diagnosis of PDAP in clinical practice in the future.

Developing a feasible way to prepare highly dispersed heterometallic nanoparticles incorporated in porous carbon composites is of significant importance for multifunctional materials. In this work, heterometallic γ-Fe 2 O 3 and GdPO 4 nanoparticles that are incorporated in ordered mesoporous carbon composites are facilely prepared by a one-pot in situ method using a Wells–Dawson-like cluster of [Fe 6 Gd 6 (μ 3 -O) 2 (CO 3 )(O 3 PPh) 6 (O 2 C t Bu) 18 ] ({Fe 6 Gd 6 P 6 } for short) as the precursor. It is verified that the γ-Fe 2 O 3 and GdPO 4 nanoparticles are highly dispersed and embedded into the carbon matrix with a particle size smaller than 5 nm, even when the carbon matrix is changed from 2D hexagonal P6mm to 3D body-centered cubic Im-3m symmetry. Additionally, a formation mechanism is proposed. Furthermore, dual-mode magnetic resonance (MR) imaging and drug carrier properties are evaluated by in vitro experiments, which show a satisfactory T 1 - and T 2 -weighted MR imaging effect with r 1 and r 2 relaxivity values of 2.7 and 183.7 mM −1 s −1 , respectively, and doxorubicin hydrochloride carrier amount of 102 mg g −1 , identifying a combined function for potential diagnostic and therapy.

A pH-responsive nanoplatform, hydroxylated mesoporous nanosilica (HMNS) coated by polyethylenimine (PEI) coupled with gadolinium and folic acid (FA) (Gd-FA-Si), was designed to deliver anticancer drug targeting and to promote contrast effect for tumor cells using magnetic resonance (MR) spectrometer. Doxorubicin (DOX) was chosen as the anticancer drug and loaded into nanopores of HMNS, then its release in simulated body fluid could be controlled through adjusting the pH. This nanoplatform could significantly enhance the MR contrast effect, and the highest theoretical relaxivity per nanoplatform could even be approximately 1.28 × 106 mm–1s–1 because of the high Gd payload (2.61 × 105 per nanoplatform). The entire system possessed a high targeting performance to Hela and MDA-MB-231 cells because the FA located in the system could specifically bind to the folate–receptor sites on the surface of cell. Compared with free DOX, the nanoplatform presented a higher cell inhibition effect on the basis of cell assay. Therefore, this nanoplatform could be potentially applied as a tumor-targeted T1 MR contrast agent and pH-sensitive drug carrier system.

Construction of stimuli-responsive theranostic nanoagents that can increase the accuracy of imaging diagnosis and boost the therapeutic efficacy has been demonstrated for a promising approach for diagnosis and treatment of cancer. Herein, we constructed a novel theranostic agent with Co–P nanocomposites as core, mesoporous silica as shell, and manganese dioxide (MnO2) nanosheets as gatekeeper, which have been employed for pH-activatable T1/T2 dual-modality magnetic resonance imaging (MRI)-guided chemotherapeutical and photothermal combination anticancer therapy in vitro and in vivo. Co–P core-enabled theranostic platform could be applied for both photothermal therapy and T2-weighted MRI in the normal circulation owing to its strong near-infrared absorbance and intrinsic magnetic properties. In the acidic environment of tumors, MnO2 cap could be dissolved into Mn2+ ions to not only realize pH-responsive on-demand drug release but also activate T1-weighted MRI contrast enhancement. Such T1/T2 dual-mode MR imaging provides further comprehensive details and accurate information for tumor diagnosis, and the on-demand chemo-photothermal synergetic therapy greatly improved the therapeutic effectiveness and effectively mitigated side effects. These findings demonstrate that Co–P@mSiO2@DOX–MnO2 are promising as pH-responsive theranostic agents for tumor diagnosis and treatment, and stimulate interest in exploration of novel stimuli-responsive theranostic nanoagents which posssess good potential for clinical application in the future.

Development of imaging methods that can detect target molecules will be of great significance for non-invasive molecular diagnosis. Vascular endothelial growth factor receptor-2 (VEGFR-2) is a broadly expressed target, and VEGFR-2-specific agents have been used in biological therapy.
In this study, amine-reactive coupling was used to label the polypeptide VEGF125–136 with a Gd(DOTA) complex to create a magnetic resonance contrast agent, VEGF125–136-Gd, which binds to VEGFR-2. Using T1-weighted magnetic resonance imaging, we explored the tumor cell-targeting
ability of VEGF125–136-Gd and its enhancement of T1-weighted image intensity using human hepatoma (HepG2) cells and found that the signal-to-noise ratio (SNR) increased commensurately with the concentration of VEGF125–136-Gd. At a concentration of 2 mM, the SNR produced by VEGF125–136-Gd
was 5.4-fold higher than that produced by a Gd-labelled non-targeting polypeptide (NTPP-Gd) control. We also evaluated the tumor-targeting efficiency of VEGF125–136-Gd in nude mice injected with human hepatoma (HepG2) cells. Preliminary in vivo imaging of VEGFR-2 in tumor tissue
revealed a targeting peak 60 min post administration of VEGF125–136-Gd, the intensity of which was 23% higher than the SNR of NTPP-Gd at the same time point. Our findings may help lay the foundation for clinical applications of VEGF125–136-Gd in targeted imaging.

Here, a novel poly(lactic-co-glycolic acid) (PLGA)-based nanoparticles (NPs) for magnetic resonance (MR) and fluorescence imaging was developed for cell imaging. PLGA NPs loaded with fluorescent dye Nile red (NR) and surface-coated with poly(ethyleneimine) (PEI) were produced in a single
step nanoprecipitation process. Diethylenetriamine pentaacetic dianhydride (DTPA) was conjugated to PLGA/NR@PEI NPs through amidation reaction between –COOH of DTPA and –NH2 of PEI, which can chelate gadolinium (Gd3+) as an MR imaging contrast agent. The PLGA/NR@PEI-DTPA-Gd
NPs exhibited a uniform particle size of ∼200 nm and were stable in culture medium. These NPs had a high T
1 relaxivity (R
1) of 28.36 mM−1S−1. They did not introduce serious cytotoxicity against A549 lung cancer cells.
Furthermore, fluorescence and MR imaging studies on A549 lung cancer cells in vitro revealed that PLGA/NR@PEI-DTPA-Gd NPs can serve as an efficient fluorescence/MR dual-modality imaging nanoprobe.

Introduction: Magnetic Resonance Imaging (MRI)-guided photothermal therapy (PTT) holds significant promise for the treatment of solid tumors, however, its diagnostic and therapeutic efficacy in malignant pleural mesothelioma (MPM) remains underexplored. To address the limitations of traditional gadolinium (Gd)-based MRI contrast agents, such as intolerance in patients with renal insufficiency and T1 signal attenuation at high field strengths, as well as oxidative stress damage caused by manganese ion leakage from manganese-based materials, this study aims to develop highly chelated and stable manganese polydopamine (Mn-PDA) multifunctional nanoprobes for MRI monitoring and PTT treatment of MPM.Methods: Mn2+ and dopamine were chelated into Mn-PDA NP nanospheres in an ethanol-water system. The physicochemical properties were characterized using Transmission Electron Microscopy (TEM), Dynamic Light Scattering (DLS), zeta potential, Fourier Transform Infrared Spectroscopy (FT-IR), X-ray Photoelectron Spectroscopy (XPS), and Electron Paramagnetic Resonance (EPR). The photothermal performance under 808 nm laser irradiation was also evaluated. The MRI imaging performance (longitudinal/transverse relaxation rates, r1/r2) and manganese ion deposition quantification of Mn-PDA NPs were evaluated in vitro and in vivo at 3.0 T MR scanner. Additionally, the PTT efficacy of Mn-PDA NPs was evaluated in MSTO-211H cells and xenograft tumor models. Biocompatibility was assessed through H&E staining of major organs and blood biochemical analyses. Results: Mn-PDA NPs exhibited a spherical morphology with uniform size (∼200 nm), along with high photothermal conversion efficiency, favorable T1-weighted MRI contrast enhancement and low cytotoxicity. At 8 hours post-injection in vivo, Mn-PDA NPs resulted in a 42.9% enhancement in T1-weighted MRI signals within MPM tumors, accompanied by specific tumor accumulation. Under 808 nm laser irradiation, Mn-PDA NPs inhibited MPM tumor growth, as evidenced by reduced tumor volume, expanded areas of tumor necrosis (H&E staining), and downregulated Ki-67 expression. Moreover, stable body weight, normal histological features of major organs, and unaltered blood biochemical parameters were observed in the animals.Discussion: These findings indicate that Mn-PDA NPs are effective dual-functional agents, serving as both MRI contrast agents and photothermal therapeutics. They exhibit good tumor targeting ability, excellent imaging performance, potent therapeutic efficacy, and high biosafety, thereby offering a promising therapeutic strategy for MPM management.

Multimodal imaging is a potent technique that integrates complementary information from various imaging modalities, offering comprehensive insights into disease diagnosis. However, the development of biocompatible multimodal probes remains a challenge. In this study, we present the synthesis, characterization, and application of gadolinium and ytterbium co-doped carbon dots (Yb/Gd-CDs) for triple-modal fluorescence (FL)/magnetic resonance (MR)/computed tomography (CT) imaging. Notably, Yb and Gd were successfully incorporated into the lattice of CDs using a facile one‐pot hydrothermal approach to obtain Yb/Gd-CDs. The Yb/Gd-CDs demonstrated an ultra-small size, excellent colloidal stability, and low toxicity, making them suitable for further in vivo applications. Moreover, the Yb/Gd-CDs exhibited excellent fluorescence, high X-ray attenuation (40.17 HU/g/L), and high T 1 relaxivity (11.16 mM -1 s −1 ) with an ultra-low r 2 /r 1 ratio of 1.08, showcasing remarkable performance in in vivo fluorescence, T 1 -weighted MR, and CT imaging of mice. Yb/Gd-CDs show great promise for in vivo multimodal imaging, and this work provides a highly biocompatible CDs-based theranostic platform with multiple functions for disease diagnosis.

Personalized radiotherapy strategies enabled by the construction of hypoxia-guided biological target volumes (BTVs) can overcome hypoxia-induced radioresistance by delivering high-dose radiotherapy to targeted hypoxic areas of the tumor. However, the construction of hypoxia-guided BTVs is difficult owing to lack of precise visualization of hypoxic areas. This study synthesizes a hypoxia-responsive T 1 , T 2 , T 2 mapping tri-modal MRI molecular nanoprobe (SPION@ND) and provides precise imaging of hypoxic tumor areas by utilizing the advantageous features of tri-modal magnetic resonance imaging (MRI). SPION@ND exhibits hypoxia-triggered dispersion-aggregation structural transformation. Dispersed SPION@ND can be used for routine clinical BTV construction using T 1 -contrast MRI. Conversely, aggregated SPION@ND can be used for tumor hypoxia imaging assessment using T 2 -contrast MRI. Moreover, by introducing T 2 mapping, this work designs a novel method (adjustable threshold-based hypoxia assessment) for the precise assessment of tumor hypoxia confidence area and hypoxia level. Eventually this work successfully obtains hypoxia tumor target and accurates hypoxia tumor target, and achieves a one-stop hypoxia-guided BTV construction. Compared to the positron emission tomography-based hypoxia assessment, SPION@ND provides a new method that allows safe and convenient imaging of hypoxic tumor areas in clinical settings.

Mn(IV)-Based nanoparticles (NPs) are effective in improving tumor oxygenation (hypoxia) and reducing endogenous hydrogen peroxide and acidity in the tumor region. However, the optimized reduction conditions of conventional Mn(IV)-based NPs are generally reported at pH ≤ 6.5, while the usual pH range of the tumor microenvironment (TME) is 6.5–7.0. The dissatisfactory imaging performance in the weakly acidic environment may limit their further application in tumor diagnosis. In this study, Mn(III) was introduced in a nanoplatform, because it is reduced into Mn(II) in weakly acidic environments. Arg-Gly-Asp (RGD) peptide-decorated bovine serum albumin (BSA) was employed as the stabilizer and scaffold to fabricate Mn(III)- and Mn(IV)-integrated nanocomposites (RGD-BMnNPs) with suitable size, good stability, and excellent biocompatibility. The as-prepared NPs showed clear contrast enhancement at pH 6.5–6.9 in vitro as well as sensitive and rapid T1-weighted imaging performance within the tumor region in a glioblastoma (U87MG) orthotopic model, owing to the intrinsic disproportionation reaction of Mn(III) in the weakly acidic environment. In addition, these NPs could be used for efficient siRNA delivery. They showed superior advantages in this process, including increased tumour uptake, improved tumor accumulation and enhanced therapeutic effects with the modulation of the TME. These novel albumin-stabilized manganese-based NPs combined with efficient drug delivery capacity hold great potential to serve as intelligent theranostic agents for further clinical translation.
