Bacterial outer membrane vesicles-cloaked modified zein nanoparticles for oral delivery of paclitaxel

Zeyu Wang,Yuqi Chu,Xu Tao,Jianchao Li,Lihong Wang,Yuli Sang,Xiuli Lu,Lijiang Chen

To improve the aqueous solubility and oral bioavailability of paclitaxel (PTX), a biomimetic system for oral administration of PTX was efficiently developed as an outer membrane vesicle (OMVs) of sodium caseinate (CAS) modified zein nanoparticles (OMVs-Zein-CAS-PTX-NPs) by Escherichia coli. To verify their structure and properties, the designed nanostructures were thoroughly characterized using various characterization techniques. The results indicated that hydrogen bonds and van der Waals forces mainly drove the interaction between PTX and Zein, but the complex is unstable. The physicochemical stability of PTX-loaded zein nanoparticles was improved by the addition of CAS. The biological characteristics of biofilms are reproduced by nanoparticles cloaked with outer membrane vesicles. OMVs-Zein-CAS-PTX-NPs delayed the release of PTX under simulated gastric and intestinal fluids due to OMVs protection. OMVs-Zein-CAS-PTX-NPs exhibited remarkable antitumor ability in vitro and improved the bioavailability of oral administration of PTX in vivo. Therefore, OMVs cloaked in nanoparticles may be a suitable delivery vehicle to provide an efficient application prospect for the oral administration of PTX.

Pectin-Zein-IPA nanoparticles promote functional recovery and alleviate neuroinflammation after spinal cord injury

Chen Xianghang,Wang Beini,Al Mamun Abdullah,Du Kaiyi,Wang Shengfu,Hu Qianqian,Chen Xinyuan,Lu Yang,Du Anyu,Wu Yueqi,Shao Jiaqin,Wang Shuangshuang,Jiang Chang,Zhou Kailiang,Hu Siwang,Xiao Jian

Introduction Spinal cord injury (SCI) impairs the balance of gut microbiomes, which further aggravates inflammation in the injured areas and inhibits axonal regeneration. The intestinal microbiome plays an important role in SCI and regulating intestinal microbiome promotes SCI repair. However, current studies have shown that indole-3 propionate (IPA), a metabolite of gut bacteria, can promote axonal regeneration. However, the short half-life of IPA limits its effectiveness. Gut microbiota plays a role in the progression of SCI, but the studies about diet regulates intestinal flora metabolites to improve SCI are still limited and lack guiding significance. Results The results showed that Pectin-Zein-IPA NPs treatment improves motor function recovery, inhibits the activation of oxidative stress, enhances axonal regeneration and activates AKT/Nrf-2 signaling pathway following SCI. Further analysis showed that Pec-Zein-IPA NPs treatment reduced the intestinal flora metabolite accumulation of L-methionine, and alleviated neuroinflammation by improving autophagy and inhibiting pyroptosis. Pec-Zein-IPA may reduced neuroinflammation after SCI by decreasing the abundance of Clostridia − UCG-014 , Clostridia − vadinBB60 − group , Shewanella (positively correlated with L-Methionine) and increasing the abundance of Parasutterella (negatively correlated with L-Methionine). Conclusions Our findings provide a strategy for oral drug research in SCI. The results suggest that Pectin-Zein-IPA NPs have potential advantages for treatment and management of SCI. Reducing L-methionine intake may help reduce neuroinflammation after SCI. Graphical

Fabrication and characterization of polydopamine-mediated zein-based nanoparticle for delivery of bioactive molecules

Zhiheng Zhang,Han Jiang,Guo Chen,Wenbo Miao,Qianzhu Lin,Shangyuan Sang,David Julian McClements,Aiquan Jiao,Zhengyu Jin,Jinpeng Wang,Chao Qiu

In this study, a combination of whey protein (hydrophilic coating) and polydopamine (crosslinking agent) was used to improve the stability and functionality of quercetin-loaded zein nanoparticles . There are two key benefits of the core-shell nanoparticles formed. First, the ability of the polydopamine to bind to both zein and whey protein facilitates the formation of a stable core-shell structure, thereby protecting quercetin from any pro-oxidants in the aqueous surroundings. Second, neutral and hydrophilic whey proteins were used for the surface coating of the nanoparticles to further enhance the sustained and slow release of quercetin , facilitating its sustained release into the body at a slow and steady rate. The results of this study will promote the innovative development of precise nutritional delivery systems for zein and provide a theoretical basis for the design and development of dietary supplements based on hydrophobic food nutrient molecules.

Edible electrospun zein nanofibrous scaffolds close the gaps in biofilm formation ability between microorganisms

Meng-Xin Hu,Fei He,Cheng-Kai Tu,Zhe-Xin Chen,Hui Teng,Xin Shao,Ge-Rui Ren,Ya-Xin Guo

Biofilm is an important survival mode of microorganisms in nature. Biofilm culture is supposed to be a way of learning from nature to endow microorganisms with a strong tolerance. However, microorganisms possess different biofilm formation abilities, and universal scaffolds, especially edible scaffolds, suitable for fast biofilm formation are not found yet. Zein is a hydrophobic and biocompatible prolamine-rich protein from corn and is one of the most used biomacromolecules now. In this work, natural zein was utilized to produce electrospun zein nanofibrous biomaterial. The results demonstrate that edible electrospun zein nanofibrous scaffolds significantly enhance biofilm formation, where bacterial probiotics and yeasts are 0.9–1.4 Log cfu/cm 2 (8–25 × ) and 1.7–3.3 Log cfu/cm 2 (50–2000 × ) higher than the values on polystyrene plates and zein films, respectively. Gaps in biofilm formation abilities between bacteria and yeasts are narrowed from 1.7–1.8 Log cfu/cm 2 (50–63 × ) to 0.5–0.6 Log cfu/cm 2 (3–4 × ), respectively. Biofilms quickly formed on electrospun zein nanofibrous scaffolds before the occurrence of serious swelling deformation of the scaffolds. This is the first attempt to use edible electrospun fibrous scaffolds based on natural biomacromolecules for enhancing biofilm formation, crucially important for the application of probiotics and yeast in food and medical industries. Electrospun zein nanofibrous scaffolds can serve as powerful tools to enhance the biofilm formation of valuable food microorganisms and close the huge gaps in biofilm formation abilities between microorganisms, providing a convenient and universal strategy to produce biofilm-phenotyped probiotics and yeasts.

Zein/Shellac Composite Films with Limonin and Resveratrol: Characterization and Application for Strawberry Preservation

Limin Wang,Qianfei Jia,Yuxi Qin,Shufen Li,Zijian Wu

This study aimed to fabricate an active film incorporating limonin (LM) and resveratrol (RES) within a zein/shellac (ZS) matrix for use in strawberry preservation. Zein/shellac composite films embedded with varying concentrations of LM (0–4%w/w) and RES were successfully fabricated using coaxial electrospinning. The prepared films were comprehensively characterized for their mechanical properties, water vapor permeability (WVP), antioxidant capacity (DPPH, ABTS, FRAP), and antibacterial efficacy againstE. coliandS. aureus. Mechanical properties and WVP results revealed that the ZSLM4R film exhibited an elongation at break (EBA) of 28.91%, tensile strength (TS) of 0.93 MPa, elastic modulus (EM) of 40.76 MPa, and a WVP of 1.55 g mm/m2. h. kPa. Furthermore, LM and RES increased the antioxidant properties of the composite film. ZSLM4R’s free radical scavenging activities against DPPH and ABTS were 68.14% and 89.69%, respectively. The composite film also demonstrated strong antibacterial efficacy againstE. coliandS. aureus. When applied to strawberries, ZSLM4R packaging effectively extended the fruit’s shelf life compared to the unwrapped and commercial polyethylene (PE) controls. These obtained results indicate that LM/RES-incorporated zein/shellac composites are a promising eco-friendly packaging alternative for preserving perishable fresh produce and extending its shelf life.

Preparation and characterization of zein-based active packaging films integrated with TiO2 nanotube arrays

Kai Cheng,Jin Du,Fengfeng Xu,Zengkai Wang,Liwei Zhang,Mengqi Bai,Xueqin Wang,Jun Liu

Environmentally-friendly active packaging films provide a novel approach for food preservation, addressing the major challenge of plastic pollution. In this study, we developed a zein-based flexible film incorporating TiO 2 nanotube arrays (TNTAs). We conducted a comprehensive investigation into the microstructure, spectroscopic properties, water resistance, mechanical strength, and antimicrobial activity of the film. The incorporation of TNTAs with varying tube lengths (ranging from 700 nm to 5.2 µm) resulted in their uniform dispersion within the zein matrix and significantly enhanced the mechanical robustness and water resistance of the composite films. XRD, FT-IR, and XPS analyses revealed intermolecular hydrogen bonding between zein matrix and TNTAs. Moreover, under low-power density (1600 mW/cm 2 ) UV irradiation, Zein/TNTAs films exhibited remarkable antimicrobial efficacy against E.coli . These findings highlight the potential application of Zein/TNTAs composite films in active food packaging.

Konjac glucomannan/zein active film loaded with tea polyphenol–ferric nanoparticles for strawberry preservation

Zhenzhen Wu,Lin Wang,Zhiwen Hu,Xiuqing Guan,Yibin Chen,Mingming Xu,Xianrui Chen,Nitong Bu,Jie Duan,Wei Liu,Chen Ma,Jie Pang

With increasing global environmental awareness and concerns about food safety, biodegradable active packaging has garnered widespread attention. In this study, the stability and bioactivity of tea polyphenol (TP) were enhanced through the preparation of TP–ferric nanoparticles (TP–Fe NPs) using metal–polyphenol ion coordination. Moreover, the introduction of Fe ions can further enhance the antibacterial effects of TP–Fe NPs. Using the hydrogen bonding between konjac glucomannan (KGM) and zein to enhance the hydrophobicity and mechanical properties of the film. By employing KGM and zein as the matrix, we incorporated TP–Fe NPs as active fillers to create multifunctional active packaging films. This study aimed to meet the needs of food safety and sustainable development goals. The resulting film exhibited excellent water resistance (water contact angle: 117.73°), mechanical strength (tensile strength: 21.82 MPa, elongation at break: 94.30 %), ultraviolet-shielding ability (>99 %), biodegradability (5 days in soil), and antioxidant (>85 %) and antibacterial (>99 %) properties. Moreover, the film significantly reduced strawberry decay and extended its shelf life by 10 days. These findings provide new insights into the application of nanomaterials in active packaging, highlighting their potential and advantages in food preservation.

Preparation and Characterization of Dihydromyricetin-Loaded Poly(vinyl alcohol)/Gelatin/Zein Composite Electroblowing Nanofibers

Hui Xiang,Qin Li,Longchen Shang,Xiujuan Chen,Lingli Deng,Yexing Tao

In this study, composite nanofibrous membranes composed of poly(vinyl alcohol) (PVA), gelatin, and zein loaded with different contents of dihydromyricetin (DMY) were fabricated via electroblowing spinning (EBS). The effects of DMY content on the microstructure, physicochemical properties, mechanical strength, and functional performance of the membranes were evaluated. Scanning electron microscopy (SEM) analysis showed that the average fiber diameter increased from 174 ± 29 nm to 221 ± 35 nm with increasing DMY content, followed by a slight decrease at higher loading levels, indicating that DMY incorporation influences fiber morphology. Fourier transform infrared spectroscopy (FTIR) results suggested the presence of hydrogen bonding interactions between DMY and the polymer matrix. X-ray diffraction (XRD) and differential scanning calorimetry (DSC) results indicated changes in the physical state of DMY within the nanofibrous system as the loading content increased. All samples exhibited a typical two-stage release behavior, and the highest cumulative release (nearly 55%) was observed at a DMY loading of 22.5%, while further increasing the loading reduced the release efficiency to approximately 45%. The release profiles were well described by a first-order kinetic model. The composite membranes exhibited improved surface hydrophilicity, appropriate water vapor permeability, antioxidant activity, and antibacterial activity againstEscherichia coli(E. coli) andStaphylococcus aureus(S. aureus). This study demonstrates the successful fabrication of DMY-loaded PVA/gelatin/zein nanofibrous membranes and provides preliminary insights into their structure–property–function relationships, release behavior, antioxidant activity, and antibacterial activity against representative bacteria, although further application-oriented validation is still required.

From poor solubility to precision delivery: Zein–κ-carrageenan nanovehicles unlock dihydromyricetin's bioactive potential

Gerui Ren,Ruiqi Hu,Jiajun Mao,Xinpei Cai,Tianrong Wang,Xin Song,Wentao Wu,Caixin Zhuge,Min Huang,Hujun Xie,Kejun Cheng

Dihydromyricetin (DMY) possesses potent antioxidant, anti-inflammatory and neuroprotective activities, yet its potential applications are curtailed by extremely low aqueous solubility and limited bioaccessibility. To overcome these constraints, the Zein-DMY/κ-carrageenan (κ-CGN) nanoparticles were engineered via anti-solvent precipitation at pH 5 (zein:κ-CGN mass ratio of 6:4) that reached 83.1% encapsulation efficiency. Multispectral analysis (FTIR, fluorescence, XRD) combined with molecular docking showed that hydrogen bonding, electrostatic and hydrophobic interactions were the main driving forces for the formation of nanoparticles, and DMY was anchored in the hydrophobic region of Zein. The dense sulfate groups of κ-CGN electrostatically stabilized the surface of nanoparticles, preventing their aggregation. Compared with free DMY, the nanocomposite retained strong antioxidant activity (95.25% for DPPH and 97.20% for ABTS), increased water solubility 4.88-fold (3.12 ± 0.07 mg/mL) and enhanced bioaccessibility 1.65-fold (37.31%). In vitro digestion simulations, coupled with SEM and release kinetics modeling, demonstrated the controlled release characteristics of nanoparticles in intestinal fluid. The release kinetics closely adhered to the Ritger-Peppas model, indicating Fickian diffusion behavior. This scalable, food-grade nanocarrier offers a robust strategy to enhance the solubility, bioaccessibility and targeted intestinal delivery of DMY for functional foods and nutraceuticals.

Astaxanthin Encapsulation in Zein/Chito-Oligosaccharide Composite Nanoparticles: Characterization, Stability, Antioxidant Efficacy and Bioaccessibility

Jian Ma,Feihong Jia,Jiangfeng Song,Dajing Li,Wuyang Huang,Ning Jiang,Haiyan Liu,Yanhong Ma

Astaxanthin (AST) has been widely studied for its potential antioxidant and anti-inflammatory effects; however, its application is limited by poor water solubility and low bioavailability. To address this, zein and chito-oligosaccharides composite nanoparticles (ZCNps) were constructed using antisolvent precipitation for efficient AST delivery. At a zein to COS mass ratio of 10:1, AST-loaded ZCNps (ZCANps) achieved an encapsulation efficiency of 81.23% and drug loading of 4.18 mg/g. Structural characterization confirmed that AST was successfully encapsulated in an amorphous state, stabilized by hydrophobic interactions and hydrogen bonding. ZCANps demonstrated stability under a wide range of conditions, including pH levels of 2–8, NaCl concentrations of up to 50 mmol/L, temperatures of up to 80°C, and storage for 8 weeks at 4°C. Additionally, ZCANps provided 62% UV-light protection for AST. Compared to free AST, the encapsulated form showed significantly enhanced antioxidant activity and bioaccessibility. These results highlight ZCNps as a delivery system for improving the stability, bioavailability, and functional properties of AST in food applications.

Leaf-inspired multifunctional zein films with CD-MOFs and Pickering emulsions for fruit preservation

Jin Yan,Zhengxin Ma,Haoran Ma,Yanling Chen,Chunmei Ding,Jianshu Li

Multifunctional zein films were developed using Pickering emulsions and γ-CD-MOFs. • Pickering emulsions improved mechanical performance while providing active preservation. • γ-CD-MOFs create an effective gas barrier, minimizing O 2 and CO 2 permeability. • The non-toxic, biodegradable film extended shelf-life of cherry tomatoes (15 d) and grapes (10 d). The development of eco-friendly food packaging from biomacromolecules offers a sustainable alternative to conventional petrochemical-based materials. However, effectively addressing both intelligent control and biodegradability remains a significant challenge. Herein, inspired by the nature leaf defense and respiration, for the first time, γ-cyclodextrin-metal-organic frameworks (CD-MOFs) and cinnamaldehyde-loaded Pickering emulsions (PE) are combined within a zein matrix to simultaneously target both external microbial spoilage and internal respiratory physiology of fresh produce. The resulting composite film with an optimal 4 wt% CD-MOFs loading exhibited exceptional gas barrier properties because of its CO 2 adsorption ability, achieving a minimal O 2 permeability and CO 2 permeability, which can efficiently inhibit the respiration of fruit. Furthermore, the PE conferred exceptional UV resistance (~100% at 280–400 nm), potent antioxidant activity (92.5% DPPH and 91.2% ABTS scavenging), and broad-spectrum antimicrobial efficacy (about 100% inhibition of E. coli and S. aureus ). The resulting composite films also exhibited improved mechanical properties and flexibility while maintaining biodegradability and biocompatibility. Application of these films effectively preserved the freshness and visual appeal of various fruits, demonstrating their potential as a safe and effective food preservation material. This work offers a promising strategy for fabricating advanced, multifunctional packaging materials from natural polymers. Enhanced antioxidant peptide extraction from zein using optimized ultrasound-assisted dual-enzyme methods hydrolysis

Xiaoqin Ding,Yabin Zhao,Renlin Zheng,Jimeng Cai,Xinyu Peng,Liangchun Li

In this study, we developed an ultrasound-assisted dual-enzyme hydrolysis method for extracting low-molecular-weight peptides from maize alcohol-soluble proteins. Using a Box-Behnken design within response surface methodology (RSM), we optimized the hydrolysis process, achieving a maximum protein hydrolysis degree of 26.5 % under optimal conditions: substrate concentration 4 g/L, pH 8.3, enzyme addition 6 %, ultrasonic power 480 W/L, ultrasonic duration 2.3 h, and hydrolysis temperature 60 °C. Structural analysis revealed that ultrasound-treated samples exhibited looser structures and more wrinkled surfaces, indicative of increased exposure of hydrophobic and aromatic amino acids. In vitro antioxidant assays demonstrated significantly higher DPPH, hydroxyl radical, and superoxide anion radical scavenging activities in ultrasound-treated samples ( p  < 0.05), with the <3 kDa peptide fraction showing the highest activity. This study highlights the potential of ultrasound-assisted dual-enzyme hydrolysis for enhancing zein utilization and advancing sustainable food additive development.
