Ru@UiO-66-NH2 MOFs-Based Dual Emission Ratiometric Fluorescence for Sensitive Sensing of Arginine

Jiawen Fan,Junjie Qi,Jingkun Li,Fuwei Pi

Arginine has been widely applied in the food industry as coloring agents, flavoring agents, and nutritional fortifiers. It is also one of the major components of feed additives. Currently, methods for the highly selective detection of arginine remain absent. For accurate and sensitive detection of L−arginine, a novel ratiometric fluorescence assay based on Ru@UiO-66-NH2was developed and demonstrated in this study. Under optimized detection conditions, the limit of detection (LOD) of this assay for L-arginine was 2.32 μM, which is superior to most assays reported to date. Meanwhile, Ru@UiO-66-NH2showed good stability within 30 days, demonstrating the wide applicability of the proposed assay. The spike-and-recovery rates of the proposed assay for L-arginine in real samples (e.g., tea, grape juice, and serum) were 84.27–113.09%. Overall, the proposed assay showed high sensitivity, good reproducibility, and excellent stability in the detection of L-arginine in both buffer and real samples.

IL-4 and arginine synergy accelerates early bone regeneration via enhanced efferocytosis and ameliorated microenvironment

Lingxi Meng,Rongpu Liu,Siyi Wang,Junyi Wang,Ilya A. Vinnikov,Rong Yang,Xinyu Song,Hongming Zhang,Jiajia Wang,Wenjie Zhang

Apoptosis at bone defects drives an anti-osteogenic oxidative stress environment • IL-4 drives a pro-reparative macrophage phenotype and upregulates the Arg1 pathway • Exogenous Arg fuels efferocytosis and secretion of pro-regenerative factors • Co-delivery of IL-4 and Arg via injectable hydrogel accelerates bone regeneration Summary Host bone microenvironment is a critical factor for successful bone regeneration during implantation. In this work, we identified a spatiotemporal mismatch at drilling-induced injury sites where initial apoptosis outpaces macrophage recruitment, leading to apoptotic cell accumulation and oxidative stress that impairs osteogenic differentiation. To mitigate this phenomenon, we developed a synergistic strategy co-delivering interleukin 4 (IL-4) and arginine (Arg) to reprogram the microenvironment. Mechanistically, IL-4 recruits and polarizes macrophages toward a pro-reparative M2 phenotype, while Arg fuels the arginase-1 metabolic pathway to sustain continual efferocytosis and secretion of pro-regenerative cytokines. In a mouse bone injury model, transcriptomic and histological analyses confirmed that this strategy accelerates apoptotic clearance and establishes an M2-dominant environment, significantly enhancing early bone regeneration. Altogether, this work proposes a novel therapeutic paradigm targeting macrophage efferocytosis to resolve early microenvironmental imbalances, providing a promising therapeutic approach to promote bone defect healing and offering novel insights into optimizing host microenvironments for osseointegration following implantation. Manganese-arginine nanozymes as oxidase mimics for smartphone-based intelligent detection of 4′-O-methylpyridoxine

Zhu Guancheng,Zou Jiahui,Lin Xueer,Zhong Huimin,Jiang Cong,Huang Yanyan

By self-assembly of MnCl 2 and arginine under alkaline conditions, ultra-small MnArg nanoparticles were successfully constructed as oxidase (OXD) mimics for intelligent detection of the Ginkgo toxin 4′- O -methylpyridoxal (MPN). The obtained MnArg nanozymes possessed excellent OXD-like activity and thermal stability. Based on the inhibitory effect of MPN for the catalytic activity of MnArg, this system was utilized for the colorimetric sensing of MPN with a low limit of detection (LOD) of 2.16 µg mL −1 . The detection  system exhibited good selectivity against other potential interferents. FTIR data showed that the presence of MPN binds with MnArg and shields the active sites, thereby interfering with the oxidase-like activity. Combined with a smartphone and the ColorMax software, this nanozyme-based intelligent detection platform could effectively detect MPN with a LOD of 2.1 µg mL −1 . Our MnArg nanozyme–based system was applied to detect real ginkgo nut samples with recoveries of 92.4–108.7%, and the relative standard deviations were less than 0.7%. This work may promote the development of novel nanozymes and expand their applications in the field of food safety detection. Graphical Abstract

Exogenous arginine induced metabolic modulation and biomass enhancement in Nannochloropsis oceanica under high CO2 stress

Ruixue Ma,Jun Cheng,Ying Liu,Dongwei Jia,Jiahao Wang

Given that excessive CO 2 induces reactive oxygen species (ROS) which inhibit photosynthesis and microalgal survival, this study explores how exogenous arginine enhances stress resilience and biomass yield of Nannochloropsis oceanica under high CO 2 stress. Arginine promoted NO synthesis, enhancing antioxidant enzyme activities to combat ROS damage. Transcriptomic analysis revealed that key enzymes in the Calvin cycle (PRK, GAPDH, FBPase and SBPase) and TCA cycle (CS, IDH, SCS and MDH) were significantly up-regulated, supporting carbon fixation and energy metabolism. Arginine also alleviated cell necrosis by extending the G1 phase, promoting DNA replication and protein synthesis, which increased the proportion of viable cells from 47 % to 84 %. Furthermore, the synthesis of chlorophyll a and carotenoid were enhanced, leading to 46 % increase in biomass yield. The results highlighted arginine as a promising metabolic regulator for enhancing stress tolerance and carbon fixation efficiency of N. oceanica under high CO 2 conditions.

Green synthesis of Schiff base based on thiadiazolyl and recognition of arginine and ascorbic acid

Yuting Liu,Yi Zhou,Dawei Yin,Zixu Zhang,Ruilin Zheng,Jiabao Chen,Chi Dang,Yawen Yang,Ziqi Gao

Arginine (Arg) and ascorbic acid (AA) are biologically essential compounds that play a key role in human development and disease resistance. Consequently, it is imperative to detect these molecules expeditiously and efficiently. In the present study, 5-phenyl-1,3,4-thiadizole-2-amine and 2-((5-phenyl-1,3,4-thiadizole-2-yl)imino) methyl) phenol (henceforth designated Probe L) were synthesised catalytically by low eutectic solvent. Subsequently, probe L was utilised as a fluorescence sensor for the detection of Arg and AA. Specifically, the solution of probe L undergoes a colour change from yellow to orange-red in the presence of Arg, and fades when interacting with AA. However, under the fluorescence spectrum, the fluorescence intensity of the probe L solution significantly increases after the addition of Arg and AA. The experiments on the influence of time on the recognition of probe L demonstrated that Arg and AA could be recognised at different times. In the experiments on the influence of pH on the recognition of probe L, it was shown that Arg and AA could be effectively detected within the pH range of 4–12. The curve analysis of Job indicates that 2:3 (probe L: Arg) and 3:2 (probe L: AA) are combined with the stoichiometric ratio. The binding constants of Arg and AA were determined to be 7.43 × 10 3  M −1 and 14.11 × 10 4  M −1 , respectively. The detection limits (LOD) were calculated to be 1.05 × 10 −5  M and 9.37 × 10 −6  M, respectively. In conclusion, this work presents the synthesis of probe L, promoted by DES, and establishes its practicality as a highly selective and stable optical sensor for Arg and AA, which is of great significance for biomedical and environmental monitoring.

Sensitive determination of arginine based on hydrogen bonding by a surface plasmon resonance (SPR) sensor

X.-C. Fan,Q. Wang

A highly sensitive arginine sensor based on optical fiber surface plasmon resonance (SPR) and 11-mercaptoundecanoic acid (MUA) was studied and implemented in this work. The method of modifying the mercaptoundecanoic acid molecule on the surface of the sensing area was proposed initially by using molecular self-assembly technology in order to complete the detection of arginine. The detection principle is a hydrogen bond-based aggregation between mercaptoundecanoic acid and arginine. The close alignment of the mercaptoundecanoic acid molecules allows the aggregation of arginine to affect changes within the surrounding local refractive index. This aggregation was selective for the arginine molecules. The sensor probe has a detection limit of 0.005 μM and a sensitivity of up to 1474 nm/μM. The entire detection system, including the probe, has the advantages of being low cost, small in size, easy to operate, and uses low power. This approach has the potential to be developed as a mobile and fast-detection method.

A novel ratiometric fluorescent sensing strategy of arginine based on lanthanide-based metal-organic framework

Kaitao Xu,Wenqian Cao,Hongxu Chen,Xueqian Xia,Wenxin Lin,Yong Guo,Quan Hu

Arginine (Arg) has been recognized as a crucial biomarker for cystinuria, with its concentration levels markedly influencing human health. In this study, Eu-TATB were synthesized via a solvothermal method using 4,4′,4″- s -triazine-2,4,6-triyl-tribenzoic acid (H₃TATB) as the organic ligand and Eu ions. Eu-TATB, benefiting from its intrinsic photosensitive characteristics and the characteristic Eu 3+ emission at 618 nm, was utilized to construct a novel ratiometric fluorescent sensor. By harnessing the photosensitization of Eu-TATB under ultraviolet light irradiation, significant oxidation of arginine was catalyzed, leading to the emergence of a new blue fluorescence peak at 430 nm. The intensity of this blue fluorescence increased proportionally with the arginine concentration, while the red emission at 618 nm gradually decreased. Utilizing the cross response of this dual-emission intensity, Eu-TATB exhibits ratiometric fluorescence sensing property for arginine, with a broad linear range of 0.1–2 mM and a low limit of detection (LOD) of 3.22 μM, and validated the practicality of the proposed sensor through the quantitative detection of arginine in human serum. Notably, the variable luminescence colors that are visible to naked eye endow the feasibility of Eu-TATB for in situ visualization analysis of arginine.

Bio-based arginine surface-modified ammonium polyphosphate: an efficient intumescent flame retardant for epoxy resin

Chen Cheng,Yi Wang,Yanling Lu,Shaojie Li,Hua Li,Jun Yan,Shiguo Du

In this work, ammonium polyphosphate (APP) was surface-modified by bio-based arginine (Arg) for the first time to enhance its flame retardance for fire-safety epoxy resin (EP). The structure of Arg modified APP (Arg-APP) was characterized by Fourier transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS), 1H nuclear magnetic resonance (1H-NMR), and scanning electron microscopy (SEM). The results illustrated that Arg was attached on the surface of APP through a cation exchange reaction. With Arg acting as the efficient carbon source, the char-forming ability of Arg-APP was significantly improved as illustrated by thermogravimetric analysis (TGA). The flame retardance of EP/APP and EP/Arg-APP composites was evaluated using the limit oxygen index (LOI), vertical burning tests (UL-94), and cone calorimeter tests (CCT). The results showed that at the same weight loading (15 wt%), Arg-APP had better flame retardance and smoke suppression performance compared with pristine APP, which can be attributed to Arg-APP constituting an integrated intumescent flame retardant (IFR) and facilitating formation of char residues with significantly expanded structures and higher carbonization degrees. When the weight loading of Arg-APP reached 25 wt%, the EP/Arg-APP composite could achieve an LOI value as high as 34.7%, pass V-0 requirements in UL-94 tests, and decrease the peak heat release rate and total smoke production by 83.5% and 61.1% compared with neat EP in CCT, respectively, indicating the superior flame retardance performance of Arg-APP. Finally, the effects of the flame retardant additives on the mechanical properties of EP were evaluated by the differential scanning calorimetry (DSC) tests and tensile-strain tests. At the same additive weight loading (15 wt%), the EP/Arg-APP composite showed higher glass-transition temperature and better tensile-strain properties compared with EP/APP composite, which can be attributed to the Arg shell structure improving the compatibility between APP and the organic substrate. In conclusion, this work presents a convenient and environmentally friendly method to improve the practical performance of APP.

Biotin and arginine modified hydroxypropyl-β-cyclodextrin nanoparticles as novel drug delivery systems for paclitaxel

Caixia Yan,Na Liang,Qiang Li,Pengfei Yan,Shaoping Sun

A novel biotin and arginine modified hydroxypropyl-β-cyclodextrin (biotin-Arg(pbf)-HP-β-CD) was successfully synthesized. The hydroxyl groups of HP-β-CD on the primary faces were coupled with carboxyl groups of biotin using arginine as the functional spacer. Using biotin-Arg(pbf)-HP-β-CD as the carrier, paclitaxel (PTX)-loaded nanoparticles were developed by modified emulsion solvent evaporation method. The optimized PTX-loaded biotin-Arg(pbf)-HP-β-CD nanoparticles had a mean diameter of 121.9 nm and zeta potential of -57.7 mV. Transmission electron microscopy (TEM) observation revealed that the nanoparticles were spherical in shape. XRD spectra confirmed the successful encapsulation of PTX. Moreover, in vitro and in vivo evaluations were performed to demonstrate the superior antitumor activity of the PTX-loaded nanoparticles. The cellular uptake study demonstrated the biotin receptor-mediated endocytosis of biotin-Arg(pbf)-HP-β-CD nanoparticles and the increase of cellular uptake by introduction of biotin and arginine. It can be concluded that the biotin-Arg(pbf)-HP-β-CD nanoparticles are efficient tumor-targeting drug delivery systems for PTX.

Self-assembly behaviors of C18 fatty acids in arginine aqueous solution affected by external conditions

Panpan Fan,Yuxian Wang,Jian Shen,Ling Jiang,Wei Zhuang,Yuwang Han,Hongman Zhang

The self-assembly properties of long-chain C 18 fatty acids salts have good application in the field of environmental protection, drug control relieve, renewable energy and so on. Their degree of unsaturation is varied from saturated, monounsaturated to polyunsaturated fatty acids , which influence their self-assembly behaviors. Moreover, different external stimuli can also affect different self-assembly behaviors. In this work, three different fatty acids (stearic acid, C 18:0 ; oleic acid , C 18:1 ; linoleic acid , C 18:2 ) were chosen as the building blocks to self-assemble with arginine, a basic amino acid . The phase behaviors of ion-pairing systems affected by pH, temperature and solvent were investigated based on diagrams in more detail. The rich phase transition was observed through the characterization by cryogenic-transmission electron microscopy (cryo-TEM), polarizing microscopy , particle size analysis and rheological properties. We hope that the phase transition (self-assembly behaviors) controlled by these external conditions may provide a good guide for many related fields.

L-arginine modified mesoporous bioactive glass with ROS scavenging and NO release for periodontitis treatment

Haiyan Yao,Emine Sumeyra Turali Emre,Yuan Fan,Jiaolong Wang,Feng Liu,Junchao Wei

Periodontitis is a chronic inflammatory disease characterized by progressive alveolar bone resorption, and excessive reactive oxygen species (ROS) is a key factor to disease progression. Therefore, scavenging ROS to alleviate inflammation and promote bone regeneration are promising strategies to treat periodontitis. In this study, L-arginine (L-Arg) was used to modify mesoporous bioactive glass (MBG), forming L-Arg modified MBG (MBG@L-Arg), which showed effective ROS-scavenging and NO release properties in cells, and realize the protection and restoration of cell's activity in ROS-rich microenvironment. Furthermore, MBG@L-Arg can induce macrophage polarization from M1 to M2 phenotype, and promote the osteogenic differentiation of MC3T3-E1 cells and human periodontal ligament stem cells (hPDLSCs). MBG@L-Arg also regulated anti-inflammatory and antioxidant systems by inhibiting the NF- κ B signaling pathway and activating the Nrf2 signaling pathway. Besides, NO-PKG signaling pathway was also activated, further promoting bone regeneration. The in vivo results demonstrated that MBG@L-Arg can efficiently inhibit inflammation-induced tissue destruction and promote osteogenesis regeneration. The quantitative bone loss in MBG@L-Arg group was 1.03 ± 0.05 mm, significantly lower than that of the periodontitis group (1.47 ± 0.13 mm), implying that MBG@L-Arg can work as multifunctional materials for periodontal tissue regeneration.

Bioenzyme-responsive L-arginine-based carbon dots: the replenishment of nitric oxide for nonpharmaceutical therapy

Chuyi Han,Qinghua Yu,Junhao Jiang,Xianming Zhang,Fan Wang,Mingyue Jiang,Ruihong Yu,Tao Deng,Chao Yu

Nitric oxide (NO) is a short-lived, bioactive gas that has been found to have affinitive effects on cardiovascular diseases as well as cancer biology, while NO deficiency may cause serious pathological responses. The existing chemically-synthesized NO donors have inevitable systemic toxicity and cannot be released adaptively. Hence, L-arginine, an endogenous NO precursor, merits investigation as a natural efficient NO donor. Herein, we designed amino acid-doped L-arginine CDs-based bioenzyme-responsive NO donors, which could adaptively replenish NO/ONOO− in response to different microenvironments. Our results indicated the mechanism of the NO/ONOO− supplementation of L-arginine-based CDs and their potential for nonpharmaceutical gas-involving theranostics for the first time.
