Copper formate–lysine nanoparticles with polyphenol oxidase-like activity for the detection of epinephrine

Sun Ping,Zhou Yue,Qiu Tong,Peng Jian

Laccase is an enzyme known for its eco-friendly uses in environmental cleanup and biotechnology. However, it has limitations such as low stability, high cost, and complex recycling. So, there is a need for laccase mimics that can effectively imitate its properties. Herein, we created copper formate–lysine nanoparticles (Cuf-Lys) that mimic laccase’s activity. The developed Cuf-Lys demonstrated remarkable polyphenol oxidase-like activity, stability, and recyclability, making them suitable for the fabrication of efficient colorimetric sensors for the detection of epinephrine. These sensors had a specific response and could accurately measure epinephrine concentrations ranging from 2.5 to 50 μM, with a detection limit as low as 1 μM. Furthermore, the biosensor demonstrated high sensitivity and selectivity when applied to the detection of rutin. The limit of detection for rutin was determined to be 0.16 μM while in the linear concentration range of 0.25 to 150.0 μM. We believe that Cuf-Lys provide a new route for the design of laccase mimics, showing potential applications for biomedical diagnosis and environmental monitoring.

Preparation and antibacterial study of waterborne polyurethane modified with lysine and quaternary ammonium salt

Mengting Xu,Yu Wang,Jichao Shi,Dandan Wu,Lin Lin,Runping Jia,Yinghao Zhai,Hongxiang Qian

Bacterial surface contamination can lead to the formation of biofilms, infection by pathogens and damage the surface of materials, which can seriously endanger health and even life. However, single-system antibacterial waterborne polyurethane (WPU) cannot meet its application in the field of high antibacterial activity. Therefore, in this study, WPU was synergistically modified by lysine (Lys) and quaternary ammonium salts (QAS), and they were introduced into the WPU molecular framework by chemical bonding to form a composite system, named WKPU. The results showed that WKPU had excellent antibacterial activity against both E. coli and S. aureus compared to a single system (Lys or QAS). When the contents of both Lys and QAS were 7%, the antibacterial activity of WKPU7 was optimal, from the sum (E. coli: 8.4 mm + 8.3 mm = 16.7 mm and S. aureus: 9.7 mm + 9.5 mm = 19.2 mm) of the inhibition circles of Lys and QAS to 19.2 mm and 20.0 mm, respectively, demonstrating the net efficiency increase of 15% and 4.2%, which fully reflects the effect of “1 + 1 > 2”. This highlights their great potential as antibacterial materials that could be used in highly antibacterial fields, such as medical apparatus and food packaging.

Lactate-mediated Ran lactylation at lysine 123 promotes astrocytes polarization after oxygen-glucose deprivation/reoxygenation

Zhiqiang Wang,Man Li,Shuming Guo,Chen Deng,Beiyang Wang,Fang Gao,Junqiao Lv,Lin Sun

Ischemia and hypoxia near the injury site following spinal cord injury (SCI) lead to early lactate accumulation. Reactive astrocytes polarize and migrate to the lesion site, sealing it off and restricting immune cell infiltration. However, the specific mechanisms through which lactate influences astrocyte polarization remain poorly understood. In this study, we found that lactate can promote astrocyte proliferation, migration and differentiation into A2 after oxygen-glucose deprivation/reoxygenation (OGD/R). Inhibiting signal transducer and activator of transcription 3 (STAT3) nuclear transport reversed lactate-driven astrocyte polarization both in vivo and in vitro. Moreover, lactylome analysis identified nonhistone protein Ran as a target of lactylation at lysine 123 (K123). Ran silencing or mutation at K123 reversed the effects of lactate on STAT3 nuclear transport and astrocyte polarization. We also found that Ran lactylation is regulated by SIRT1. Taken together, our results show that lactate promotes astrocyte polarization via Ran lactylation-dependent regulation of STAT3 nuclear transport.

An innovative method for immobilizing sucrose isomerase on ε-poly-l-lysine modified mesoporous TiO2

Lingtian Wu,Yi Liu,Bo Chi,Zheng Xu,Xiaohai Feng,Sha Li,Hong Xu

Sucrose isomerase (SIase) is the key enzyme in the enzymatic synthesis of isomaltulose. Mesoporous titanium dioxide (M-TiO 2 ) and ε-poly- l -lysine-functionalized M-TiO 2 (EPL-M-TiO 2 ) were prepared as carriers for immobilizing SIase. SIase was effectively immobilized on EPL-M-TiO 2 (SI-EPL-M-TiO 2 ) with an enzyme activity of 39.41   U/g, and the enzymatic activity recovery rate up to 93.26%. The optimal pH and temperature of immobilized SIase were 6.0 and 30   °C, respectively. SI-EPL-M-TiO 2 was more stable in pH and thermal tests than SIase immobilized on M-TiO 2 and free SIase. K m of SI-EPL-M-TiO 2 was 204.92   mmol/L, and v max was 45.7   μmol/L/s. Batch catalysis reaction of sucrose by SI-EPL-M-TiO 2 was performed under the optimal conditions. The half-life period of SI-EPL-M-TiO 2 under continuous reaction was 114   h, and the conversion rate of sucrose after 16 batches consistently remained at around 95%, which indicates that SI-EPL-M-TiO 2 has good operational stability. Thus, SI-EPL-M-TiO 2 can be used as a biocatalyst in food industries.

Core-predominant gut fungus Kazachstania slooffiae promotes intestinal epithelial glycolysis via lysine desuccinylation in pigs

Hu Jun,Chen Jianwei,Hou Qiliang,Xu Xiaojian,Ren Jing,Ma Libao,Yan Xianghua

Background Gut fungi are increasingly recognized as important contributors to host physiology, although most studies have focused on gut bacteria. Post-translational modifications (PTMs) of proteins play vital roles in cell metabolism. However, the contribution of gut fungi to host protein PTMs remains unclear. Mining gut fungi that mediate host protein PTMs and dissecting their mechanism are urgently needed. Results We studied the gut fungal communities of 56 weaned piglets and 56 finishing pigs from seven pig breeds using internal transcribed spacer (ITS) gene amplicon sequencing and metagenomics. The results showed that Kazachstania slooffiae was the most abundant gut fungal species in the seven breeds of weaned piglets. K. slooffiae decreased intestinal epithelial lysine succinylation levels, and these proteins were especially enriched in the glycolysis pathway. We demonstrated that K. slooffiae promoted intestinal epithelial glycolysis by decreasing lysine succinylation by activating sirtuin 5 (SIRT5). Furthermore, K. slooffiae -derived 5′-methylthioadenosine metabolite promoted the SIRT5 activity. Conclusions These findings provide a landscape of gut fungal communities of pigs and suggest that K. slooffiae plays a crucial role in intestinal glycolysis metabolism through lysine desuccinylation. Our data also suggest a potential protective strategy for pigs with an insufficient intestinal energy supply. Video Abstract

Polymethyl methacrylate resin containing ε-poly-L-lysine and 2-methacryloyloxyethyl phosphorylcholine with antimicrobial effects

Hao Yang,Xianju Xie,Xiaowei Li,Yuxing Bai

Statement of problem Polymethyl methacrylate (PMMA) is commonly used in dentistry, including as a denture base material. However, the colonization of a PMMA surface by microbial microorganisms could increase the risk of oral diseases such as denture stomatitis and gingivitis. The development of PMMA with antibacterial properties should improve its clinical application, but whether adding ε-poly-L-lysine (ε-PL) and 2-methacryloyloxyethyl phosphorylcholine (MPC) provides antimicrobial effects is unclear. Purpose This in vitro study aimed to develop a novel antibacterial PMMA resin containing the natural nontoxic antibacterial agent ε-PL and the protein repellent agent MPC. The mechanical properties, protein repellency, and antimicrobial activities of the resin were then evaluated. Material and methods Different mass fractions of ε-PL and MPC were mixed into PMMA as the experimental groups, with unaltered PMMA as the control group. The flexural strength (n=10) and surface roughness (n=6) of the resulting mixtures were measured to determine their mechanical properties. The antiprotein properties were measured by using the micro bicinchoninic acid method (n=6). The antimicrobial effect of the resin was assessed using live/dead staining (n=6) and methyltransferase (MTT) assays (n=10). According to the variance homogeneity and normal distribution results, 1-way analysis of variance followed by the Tukey honestly significant difference test or the Welch test and the Games-Howell test were used (α=.05 for all tests). Results No significant differences were found in the flexural strength values and surface roughness of the specimens containing 1.5% MPC and 1.5% ε-PL compared with those of the control ( P >.05). The addition of ε-PL to the PMMA resin alone significantly increased its bactericidal properties ( P <.05). Adding both ε-PL and MPC further increased the antibacterial activity of the PMMA resin without increasing protein adhesion more than in the control group. Conclusions The incorporation of both ε-PL and MPC into PMMA improved its antibacterial capacity without affecting its mechanical properties and did not increase protein adhesion. Therefore, the novel PMMA fabricated in this study shows promise for dental applications.

Integration of Bile Acid-Based Facial Amphiphilicity and Lysine Chirality for Eradicating Drug-Resistant Bacteria

Yunpeng Gao,Ran Chen,Kaojin Wang,Caihong Lin,Weimin Xiao,Qian Sun,Yukang Liu,Meng Zhang,Lin Wang,Yong-Guang Jia,Xiaoxia Zhu

In response to the increasing threat of drug-resistant bacteria, numerous antibacterial peptide (AMP) mimics have been developed and shown substantial efficacy in killing bacteria. However, controlling the secondary structure of these mimics and clarifying the relationship between their conformation and antibacterial activity remain significant challenges. This study integrates facially amphiphilic bile acid scaffolds with chiral lysine centers to construct a series of bile acid–lysine polymers (BALP-n) as AMP mimics. Incorporating chiral centers into the polymer backbone allowed the induction and modulation of a stable antiparallel β-sheet secondary structure, reinforced by hydrogen bonding and hydrophobic interactions. For example, BALP-1 exhibited 51.9% antiparallel β-sheet content at pH 7.4 and demonstrated rapid bactericidal activity against Gram-positive bacteria, Gram-negative bacteria, and drug-resistant strains. In a weakly acidic environment, the antiparallel β-sheet content increased to 56.2%, accompanied by enhanced antibacterial performance. This chirality-driven strategy enables pH-responsive “on-demand” activation and improved membrane selectivity, distinguishing it from conventional cationic antibacterial polymers. BALP-1 also showed high stability under elevated temperatures and in the presence of proteinase K. Moreover, these AMP mimics displayed excellent biocompatibility, potent in vivo antibacterial activity, and the ability to promote wound healing, offering a promising approach to combat drug-resistant bacteria and biofilms.

Kinetics of CO2 Absorption into Aqueous Basic Amino Acid Salt: Potassium Salt of Lysine Solution

Shufeng Shen,Ya-nan Yang,Yangyang Bian,Yue Zhao

Aqueous amino acid salts are considered as an attractive alternative to alkanolamine solvents (e.g., MEA) for carbon dioxide (CO2) absorption. The kinetics of CO2 into unloaded aqueous solutions of potassium lysinate (LysK) was studied using a wetted wall column at concentrations ranging from 0.25 to 2.0 M and temperatures from 298 to 333 K. Physicochemical properties of aqueous LysK solutions such as density, viscosity, and physical solubility of CO2 were measured to evaluate the reaction rate constants. The reaction pathway is described using zwitterion mechanism taking into account the effect of ionic strength on the reaction rate. Under the fast pseudo-first-order regime, the reaction rate parameters were obtained and correlated in a power-law reaction rate expression. LysK shows higher chemical reactivity toward CO2 than the industrial standard MEA and most of amino acid salts. Its reaction rate constants increase considerably with concentration and temperature. The reaction order is found to be an average value of 1.58 with respect to LysK. The forward second-order kinetic rate constant, k20, are obtained as 31615 and 84822 m3 kmol–1 s–1 at 298 and 313 K, respectively with activation energy of 51.0 kJ mol–1. The contribution of water to the zwitterion deprotonation seems to be more significant than that of LysK for the above-mentioned kinetic conditions.

Non-covalent Lysine–docosahexaenoic acid amphiphilic complexes as interfacial stabilizers for emulsions and lutein delivery

Jiaqi Shang,Zhenyuan Wang,Yuanyuan Li,Xiaohui Zheng,Dan Cheng,Mi Wang,Jiaheng Zhang

Non-covalent amphiphilic complexes offer potential as interfacial stabilizers in emulsion-based delivery systems. However, the relationship between their molecular composition, interfacial assembly, and emulsion performance remains inadequately understood. Lysine–docosahexaenoic acid (Lys-DHA) complexes with varying mass ratios were synthesized to investigate how the Lys/DHA ratio influences oil–water interfacial behavior and lutein delivery. Analytical techniques and computational simulations revealed that Lys and DHA interacted through electrostatic forces, hydrogen bonding, and van der Waals interactions. Higher DHA content increased hydrophobic exposure and facilitated interfacial adsorption. The Lys-DHA (1:3) complex exhibited optimal interfacial properties, characterized by the lowest interfacial tension, highest surface pressure, fastest diffusion rate, and a predominantly elastic interfacial film. Emulsions stabilized by Lys-DHA (1:3) displayed smaller droplet sizes, lower ζ-potential, and more excellent physical stability, with an average droplet size of 317.97 nm and ζ-potential of −51.37 mV. Regarding lutein delivery, these emulsions achieved encapsulation efficiencies exceeding 80%, retained over 70% of lutein after 14 d, and enhanced lutein bioaccessibility to 44.9%. These results highlight the potential of Lys-DHA complexes as functional lipid-containing amphiphilic interfacial stabilizers, offering insights into the correlation between non-covalent molecular interactions and interfacial/emulsion characteristics.

Lysine restriction attenuates obesity-related anxiety-like behaviors via increasing Oscillibacter ruminantium-mediated tryptophol regulation

Zhao Feng,Wang Jiao,Liang Weijuan,Sun Fanwei,Liu Zhaoyi,Wu Yuehua,Khan Ahmad,Wang Chunsong,Zhou Ruiqi,Feng Huaili,You Ting,Duan Xinhao,Zhang Hongyang,Qiu Jingfu,Chen Chengzhi

Background The contribution of dietary interventions, particularly lysine-restricted diets (LRD), to ameliorating obesity-associated anxiety-like behaviors remains elusive, with limited evidence clarifying microbiota-metabolite-brain axis pathways involved. Results Herein, our results demonstrated that LRD attenuated high-fat diet (HFD)-induced anxiety-like behaviors while concomitantly reducing body weight and improving glucose metabolism. Furthermore, LRD significantly remodeled gut microbiota composition, most notably enriching the abundance of Oscillibacter. We then screened and identified that both viable and encapsulated Oscillibacter ruminantium alleviated obesity- and chronic restraint stress-induced anxiety-like behaviors, with microencapsulated formulations conferring superior anxiolytic efficacy. Notably, the obtained data revealed that LRD reversed HFD-induced the depletion of tryptophol, which was established as the metabolite of Oscillibacter ruminantium, and LRD could also alleviate the intestinal barrier impairment caused by HFD. Mechanistic results further demonstrated that tryptophol supplementation ameliorated HFD-driven anxiety-like phenotypes by inhibiting FTO-mediated reader protein IGF2BP1, and thereby reducing m⁶A modification of Lgr6 mRNA. Conclusion LRD attenuates obesity-associated anxiety-like behaviors by enriching Oscillibacter ruminantium and modulating tryptophol production, thereby activating the FTO-IGF2BP1-LGR6 signaling axis. These findings would indicate LRD as a microbiota-directed therapeutic strategy for neurobehavioral comorbidities of metabolic stress. Graphical Video

Effects of pH, linoleic acid, and reheating on volatile compounds in glucose-lysine model system

Feng Du,Yiqun Huang,Zhijie Liu,Junjian Miao,Keqiang Lai

This study aimed to investigate the effects of pH (5.8, 7.0, and 10.0), linoleic acid , and reheating (heating: 180 °C, 30 min; reheating: 100 °C, 10 min) on the reactants, α-dicarbonyl compounds, and volatile compounds in the glucose-lysine model system, with a specific focus on exploring the influences of volatile compounds. The results showed that under three different pH glucose-lysine model system yielded the detection of 9, 15, and 29 volatile compounds, respectively. Notably, both the number and content of volatile compounds exhibited an upward trend as the pH increased. Reheating treatment led to changes in both the number and content of volatile compounds. The addition of linoleic acid affected the generation of heterocyclic compounds associated with the Maillard reaction . Volatile compounds such as aldehydes and alcohols were generated in systems containing linoleic acid, resulting in a higher number and content of volatile compounds compared to systems without linoleic acid.

Konjac glucomannan/sodium alginate/ε-poly-l-lysine hydrogel promotes esophageal and colonic wound healing

Tianyu Zhou,Zhaoxue Liu,Lei Xu,Xinli Mao,Haifeng Jin,Yangyang Xiong,Guangwu Chen,Yong Lv,Li Cen,Chunren Wang,Yu Zhang,Kexin Ye,Qien Shen,Jiaming Zhou,Bin Lv,Jianying Dai,Chaohui Yu,Zhe Shen

Endoscopic submucosal dissection (ESD) is widely used to treat gastrointestinal mucosal and submucosal lesions. However, it may cause bleeding, perforation, and stricture. Although these complications can be avoided by introducing materials such as polyglycolic acid and carboxymethyl cellulose sheets, such approaches are expensive and time-consuming. Herein, we report a hydrogel prepared by combining a colloidal solution composed of konjac glucomannan (KGM) and sodium alginate (SA) and a fixative solution containing ε-poly- l -lysine (ε-PLL) and calcium chloride. The two solutions were mixed on the wound surface to form the KGM/SA/ε-PLL hydrogel through hydrogen bonds, coordination bonds, and electrostatic attraction. The effectiveness and convenience of applying the KGM/SA/ε-PLL hydrogel to promote wound healing in the esophagus and colon were assessed in vitro and in vivo . We found that the hydrogel stimulated epithelial proliferation, reduced inflammation, promoted recapillarization, and inhibited fibrosis in the esophagus and colon. Therefore, the KGM/SA/ε-PLL hydrogel is an effective and convenient agent that can promote post-ESD wound healing and is recommended for ulcer bed protection in daily clinical practice.
