Herein, a novel ratiometric molecularly imprinted electrochemical sensor (MIP-RECS) based on Ag nanoparticles (AgNPs) and ionic liquid (IL) co-modified Ti 2 NTx (AgNPs@Ti 2 NT x @IL) was developed for the determination of patulin (PAT). Firstly, the AgNPs@Ti 2 NT x @IL nanocomposite, was prepared by an in situ reduction and self-assembled method, and then, was cast on the glassy carbon electrode (GCE) to build an electrochemical interface with excellent conductivity and high surface area. After that, the MIP was deposited on the nanocomposite modified GCE by the electro-polymerization technique, with thionine (Thi) and PAT as functional monomer and template molecules. In the sensing platform, the oxidation peaks generated by Thi and AgNPs were used as reference and probe signal, respectively. The introduction of reference effectively eliminates the influence of the matrix effect. At the same time, the oxidation signal of the Ag probe is greatly improved by the high conductivity and large surface area of the AgNPs@Ti 2 NT x @IL nanocomposite. Therefore, the MIP-RECS presented a wide linear range, a very low low limit of detection (LOD), together with high selectivity and excellent repeatability. Additionally, the linearity ranges and LODs of the sensor can be controlled by the amount of Ag, highlighting its robustness in different types of real samples. Furthermore, this sensing model can be used to sensitively detect other residues with poor redox activity, such as imidacloprid and catechol, highlighting its versatility and universality.

Patulin (PAT) is a kind of mycotoxin which must be monitored for the sake of quality and safety in traditional Chinese medicine (TCM) owing to its harm to human health. On this account, a rationally designed ratiometric fluorescent aptasensor was developed based on the study of the interaction mechanism between PAT and its aptamer (PAT-APT). Firstly, CD spectroscopy, molecular docking, and molecular dynamic simulation were applied to investigate the details on how PAT-APT bind with its target molecule. The results indicated that the structure of PAT-APT changed to a certain extent and was stabilized after binding with PAT. C-11, C-37 and C-38 were the key sites for the recognition and interaction between PAT-APT and its target. Secondly, based on these results, a ratiometric aptasensor made use of fluorescence resonance energy transfer (FRET) and synchronous fluorescence spectroscopy was designed. The complementary sequence (cDNA) to the aptamer with appropriate length and hybridization position was obtained through rationally design and optimization. Both PAT-APT and cDNA were labeled by a pair of fluorophores, which could generate FRET when the two single-stranded oligonucleotides hybridized. The accurate detection of PAT could be realized according to the change ratio of fluorescence intensity at the corresponding wavelength of the two fluorophores before and after assay. The aptasensor achieved an ultralow limit of detection 0.16 nM, perfect selectivity, and satisfactory practibility in complex TCM sample. To our knowledge, this is the first aptasensor for PAT designed through the interaction mechanism between aptamer and target molecule. Moreover, the assay for PAT is cost-effective, does not need complicated pretreatment and only takes less than an hour. Summarily, this study makes contribution to safety control of TCM, and provides a thinking mode from mechanism to rational design to conquer the problem of sensitive aptasensing of one component in a complex system.

Tools to detect low molecular weight mycotoxin patulin (PAT) are urgently needed because of its harm to human health. Here, we report a simple, effective, and sensitive fluorescent aptasensor based on sulfur quantum dots encapsulated in MOF-5-NH 2 ( [email protected] 2 ) and self-cycling catalyzed hairpin assembly (scCHA) for the detection of PAT in apple juice. The [email protected] 2 synthesized by a “bottle-around-ship” solvothermal method works as a fluorescent probe and offers enhanced fluorescence intensity and good water-solubility versus MOF-5-NH 2 due to the encapsulation of water-soluble SQDs . Meanwhile, the scCHA designed by the two DNA hairpins is initiated after the formation of PAT/aptamer, which further recycles PAT to effectively capture multiple fluorescence probes ( [email protected] 2 ) and amplify the detection signal. Attributed to the synergistic effect between the strong fluorescence intensity of [email protected] 2 and the efficient PAT utilization of scCHA, the fluorescent aptasensor exhibits high sensitivity with detection limits as low as 0.753 pg/mL. The system has satisfactory specificity, anti-interference, and reproducibility (RSDs of intra-assay and inter-assay were 3.24% and 8.60%, respectively). The recoveries in apple juice were between 89.03% and 107.67%, which were with good consistency versus high-performance liquid chromatography (HPLC). These data collectively show that the developed fluorescent aptasensor has a good practical application value.

Here, a novel molecularly imprinted photopolymer was prepared using CsPbBr 3 quantum dots as the fluorescence source, TpPa-2 as substrate for selective solid fluorescence detection of patulin (PAT). TpPa-2 can promote efficient recognition of PAT due to its unique structure and significantly improve the fluorescence stability and sensitivity. The test results showed that the photopolymer exhibited large adsorption capacity (131.75 mg/g), fast adsorption ability (12 mins), superior reusability and high selectivity. The sensor proposed had good linearity for PAT in the range of 0.2–20 ng/mL and was applied to the analysis of PAT in apple juice and apple jam with a limit of detection as low as 0.027 ng/mL. Therefore it maybe a promising method for solid fluorescence detection of trace PAT in food analysis.

Patulin (PAT) is a mycotoxin that grows on various fruits and grains, and its toxicity research mainly focuses on organ toxicity. However, the research on its impact on nutritional metabolism is still unclear. Vitamin D is an important nutrient and has been reported to have multiple physiological functions. Vitamin D deficiency has been reported to be associated with the risk of developing diseases such as fatal cancers, autoimmune disorders, cardiovascular disease, neurocognitive function impairment, and type 2 diabetes. However, vitamin D need to be converted into biologically active forms through two steps of hydroxylation in the liver or kidney by 25-hydroxylase (CYP2R1) and 25-hydroxyvitamin D 1-alpha-hydroxylase (CYP27B1). In this study, we found that PAT can cause liver and kidney injury, and affect the levels of activated vitamin D products 25(OH)D and 1,25(OH) 2 D. Specifically, we demonstrated that PAT affects the hydroxylation of vitamin D by downregulating 25-hydroxylase (CYP2R1) and 25-hydroxyvitamin D 1-alpha-hydroxylase (CYP27B1) expression and interfering their substrate binding sites. This study can provide a basis for a more comprehensive assessment of the impact of environmental pollution, food safety, and other issues on human health.

There is an urgent need to develop an economical and convenient method for the ultrasensitive detection of patulin (PAT), a mycotoxin that can potentially harm human health when it is found in fruits and their derivatives. In this study, we have developed a novel fluorescent aptasensor that utilizes nitrogen-doped carbon dots (N-CDs) as the fluorescent donor and hexagonal β-Co(OH) 2 nanoplates as the fluorescent acceptor. N-CDs were synthesized through the hydrothermal method, resulting in spherical particles with a diameter of 7.6 nm. These nanoparticles exhibited excellent water solubility and displayed a vibrant blue emission at 448 nm when excited at 360 nm. Cobalt hydroxide nanoplates with a beta crystal structure [β-Co(OH) 2 ] were synthesized using a simple co-precipitation method, exhibiting hexagonal plate-like shapes with uniform lateral sizes of 4–5 μm. The fluorescence of N-CDs can be efficiently quenched by hexagonal β-Co(OH) 2 nanoplates through Förster resonance energy transfer mechanism. The maximum quenching-recovery capability can be achieved when the concentrations of N-CDs-Apt and β-Co(OH) 2 nanoplates are 150 nmol/L and 100 μg/mL, respectively. The pH of the TE buffer should be 8.0, and the incubation time should be 10 min at 25 °C. The developed fluorescent aptasensor displayed an excellent selectivity for PAT determination with a detection limit of 0.57 pg/mL in the linear range of 1.25 pg/mL-100 ng/mL. The rapid PAT determination in fruit juice samples was realized with good recoveries (96.9–105.8%). The developed fluorescent aptasensor based on the interaction between N-CDs and hexagonal β-Co(OH) 2 nanoplates can be a promising method for the rapid and ultrasensitive detection of PAT in agricultural products.
