Due to the high-glucose environment of diabetic wounds, a significant proliferation of bacteria can occur, resulting in an inflammatory response that extends the inflammatory phase of the wound, thereby complicating the healing process in diabetic wounds. Eliminating the proliferation of bacteria plays a crucial role in promoting the healing of diabetic wounds. Under near-infrared (NIR) laser irradiation, l -arginine-modified natural product puerarin (Pue)-derived carbon nanoparticles ( l -Arg-CNP) not only exhibited excellent photothermal effects but also produced reactive oxygen species (ROS) to react with l -arginine ( l -Arg) for producing Nitric Oxide (NO), thus contributing to a synergistic antibacterial therapy in diabetic wound. At the same time, l -Arg-CNP retained Pue’s original characteristics to promote cell proliferation and angiogenesis. Following the loading of l -Arg-CNP into microneedle patches ( l -Arg-CNP@MN), it can deliver them into the deeper wound, effectively killing bacteria, reducing inflammatory infiltration, and promoting neovascularization at the wound site. It offers an effective therapeutic strategy for treating diabetic wound healing.

Puerarin (PUE) is a Chinese traditional medicine known to enhance glucose uptake into the insulin cells to downregulate the blood glucose levels in the treatment of type II diabetes. Nevertheless, the bioavailability of pristine PUE is limited due to its poor solubility and low intestinal permeability. In this work, we demonstrate that the solubility of PUE can be significantly enhanced via its co-crystallization with L-Proline (PRO). Two crystalline phases, namely, the solvate-free form [PUE][PRO] (I) and the solvated form [PUE]2[PRO]∙EtOH∙(H2O)2(II) are isolated. These two phases are characterized by single-crystal X-ray diffraction (SCXRD), powder X-ray diffraction (PXRD), Fourier-transformed infrared (FT-IR) spectra, nuclear magnetic resonance (NMR), and thermogravimetric analysis in association with differential scanning calorimetry (TGA-DSC). The solubility and dissolution rate of bothIandIIin water, gastrointestinal tract at pH 1.2, and phosphate buffer at pH 6.8 indicates a nearly doubled increase as compared to the pristine PUE. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay of pristine PUE,IandIIagainst murine colon cancer cell lines CT-26 and human kidney cell lines HEK-293 indicated that neither compound exhibits obvious cytotoxicity after 24 h. This work showcases that the readily available and biocompatible PRO can be a promising adjuvant to enhance the physicochemical properties of PUE toward orally administered drug formulation with improved pharmacokinetics.Keywords:puerarin;L-proline;pharmaceutical co-crystal;crystal structure;solubility and dissolution rate;Chinese traditional medicine;Hirschfeld analysis;hydrogen-bonding

In this study, one advanced composite scaffold was designed. Its characteristics, effect on repairing large-sized bone defects and related mechanisms were explored. Methods In vitro , a bi-layered composite scaffold constructing with one layer of electrospun poly (3-hydroxybutyrateco-3-hydroxyvalerate) (PHBV) nanofiber membrane and one layer of UV-crosslinked methacrylated gelatin (MeGel) hydrogel were fabricated. Moreover, two types of bio-components including nano hydroxyapatite (nHAP) and puerarin (Pur) were encapsulated into the MeGel hydrogel layer to impart the composite scaffolds with multiple biofunctions. In vivo , the calvarial defect models with Sprague Dawley rats were created to ascertain the impact of PHBV/MeGel, PHBV/MeGel-HAP, and PHBV/MeGel-HAP-Pur composite scaffolds. Results The in vitro studies demonstrated that all the composite scaffolds loading with or without nHAP and Pur exhibited great water swelling and retention properties, as well as excellent rheological and mechanical properties. Furthermore, the PHBV/MeGel-nHAP-Pur composite scaffolds could facilitate the osteogenesis and vascularization, but inhibit osteoclast formation. Also, hydrogel layer could provide a benificial micro-environment for the cell infiltration. In vivo study demonstrated that the PHBV/MeGel-nHAP-Pur composite scaffolds obviously promote the healing of rat cranial defect with large injury size, and the healing rate was roughly 99.25% after 8 weeks of treatment. Conclusion Our present study suggests that the bi-layered PHBV/MeGel-nHAP-Pur composite scaffolds with multiple bio-functions show huge potential for the repair and regeneration of large-sized bone defects.

In this work, PtCu bimetallic nanoparticle was deposited on poly (styrene sulfonate) (PSS) functionalized graphene (Gr) to form a nanocomposite PtCu/PSS-Gr and its enzyme-like activity was investigated. Benefiting from the synergistic effect from Pt and Cu monometal as well as the superior properties of PSS-Gr, such as large surface area, good dispersity, strong adsorption of substrate and additional peroxidase-like activity, the PtCu/PSS-Gr nanocomposite was demonstrated as an excellent peroxidase mimic to catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) in the presence of H 2 O 2 . Combined with molecularly imprinted polymer (MIP), a new colorimetric approach for puerarin detection was proposed with the linear range of 2 × 10 −5 -6 × 10 −4 mol L −1 and LOD of 1 × 10 −5 mol L −1 . The combination of MIP with PtCu/PSS-Gr nanocomposite not only endowed the determination of puerarin with high selectivity, but also realized the detection of small molecules which are neither substrate of the nanozyme nor substances with strong oxidizing or reducing activity by using peroxidase-like catalytic activity of nanozyme, expanding the application of nanozyme.

Aim: To synthesize a puerarin nanoparticle based on glycyrrhetinic acid (GA)-PEG-PBLA and evaluate it in vivo. Materials & methods: In this study, drug nanoparticle was synthesized, characterized and assessed as puerarin delivery system. Nanoparticle GA-PEG-PBLA could combine with puerarin via hydrophobic interaction to form the compound. Puerarin could be quickly and efficiently loaded via the nanoparticle GA-PEG-PBLA at pH 7.4. Further, GA-PEG-PBLA-mediated puerarin delivery system could target for the liver that had GA receptor binding. The antiliver ischemia/reperfusion injury role of puerarin/GA-PEG-PBLA was measured in rats using free puerarin and puerarin/PEG-PBLA as the controls. Results: GA-PEG-PBLA displayed efficient loading and sustained release. Puerarin/GA-PEG-PBLA showed strengthened antiliver ischemia/reperfusion injury characteristics. Conclusion: Overall, the results show that GA-PEG-PBLA could be regarded as an underlying puerarin nanoparticle.

Background Alzheimer’s disease (AD) is a neurodegenerative disorder characterized by progressive cognitive decline, the formation of amyloid-β plaques and neurofibrillary tangles, and synaptic dysfunction. Puerarin (PUE), an isoflavone C-glycoside extracted from Pueraria lobata , has demonstrated potential in attenuating AD-related cognitive decline; however, the mechanisms governing the effects of PUE remain incompletely understood. Purpose To investigate the mechanisms underlying PUE’s anti-AD effects. Methods Presenilin 1/2 conditional double knockout mice served as the AD model. Following one month of PUE administration, memory function was evaluated through behavioral testing, and hippocampal synaptic plasticity was assessed using electrophysiological recordings. D‑serine levels were quantified via ELISA. Additionally, Western blotting and qRT-PCR were employed to analyze the expression of glutamate receptor subunits and key enzymes involved in D‑serine metabolism. The mechanistic role of D‑serine was further validated through intrahippocampal D‑serine injections in mice and the application of a serine racemase (SR) inhibitor in N2a cells. Results PUE administration reversed memory impairment and enhanced both hippocampal long-term potentiation and synaptic transmission by increasing the open probability of N-methyl-D-aspartate receptor (NMDAR) channels. Furthermore, PUE elevated D‑serine levels and upregulated the expression of SR. Notably, these therapeutic effects were replicated by exogenous D‑serine and abolished by treatment with an SR inhibitor. Conclusion These findings indicate that PUE alleviates cognitive deficits by promoting SR-mediated D‑serine biosynthesis and subsequently restoring NMDAR activity. Accordingly, PUE represents a viable therapeutic candidate for the prevention and management of AD.

Objective The effect of PUE on enhancing the anti-cancerous efficacy of DDP on drug-resistant A549/DDP cancer and the underlying mechanisms were thoroughly investigated.Materials and Methods The cytotoxicity of PUE, DDP, and PUE + DDP to A549 cells and A549/DDP cells, respectively, is determined by cell apoptosis experiments. Anti-proliferation effect of PUE, DDP, and PUE + DDP on A549 cells and A549/DDP cells is evaluated by the cell cloning assay. Qualitative and quantitative analysis of the levels of PUE, DDP, and PUE + DDP of cell proliferation-related genes and proteins expressions in A549/DDP cells are determined by Western blot assay. The levels of VEGF in A549/DDP cells after different treatment strategies are determined by ELISA assay. Qualitative and quantitative determination of VEGF expression in tumor tissues are done by immunohistochemical staining. Results In vitro cellular experiments revealed that co-incubation of A549/DDP cells with PUE and DDP led to a dramatically decreased cell viability and cell survival rate compared with the cells only treated by DDP. Such a stimulating effect of PUE on DDP was further confirmed in vivo with results shown that the A549/DDP cancer-bearing mice treaded by combination therapy achieved the lowest tumor growth rate and longest survival time. Conclusion Taking these results together, we can draw the conclusion that the PUE enhances the anti-tumor effect of DDP on the drug-resistant A549 cancer in vivo and in vitro through activation of the Wnt signaling pathway.

Puerarin is a bioactive flavonoid isolated from Kudzu roots that possesses numerous health benefits. However, its poor bioavailability and existing complex delivery systems with safety issues are challenging tasks for its incorporation into functional foods. Preparing modified-starch-stabilized Pickering emulsions containing microencapsulated puerarin with improved bioaccessibility was the key objective of the present research work. Acid-hydrolyzed high-amylosePueraria montanastarch (PMS) was modified with octenyl succinic anhydride (OSA) and evaluated as an emulsifier to prepare emulsions. The FTIR, SEM, and XRD results showed that PMS was successfully modified. Furthermore, the emulsification index (EI), mean droplet size, and ζ-potential values showed that modified starch with a higher degree of substitution (DS) enhanced the storage stability of emulsions. Similarly, the retention degree and encapsulation efficiency results of puerarin proved the assumption after storage of 16 d. The Pickering emulsions also helped in the controlled release of microencapsulated puerarin in vitro. The study outcomes proved that Pickering emulsions stabilized with OSA-modified PMS have promising applicability in functional foods as efficient food-grade delivery systems, enhancing oral supplementation and accessibility of puerarin. Keywords:Pueraria montanastarch;OSA modification;microencapsulation;puerarin;Pickering emulsions;in vitro release

Puerarin, the most important flavonoid in Puerariae Lobatae Radix (PLR), has high biological activity but poor oral absorption. This study investigated how to enhance the absorption and pharmacokinetics of puerarin through modulating the intestinal barrier. Analyses of intestinal absorption, pharmacokinetics demonstrated enhanced puerarin absorption with daidzin, daidzein and ligustrazine (the main active ingredient in vinegar and Chuanxiong Rhizoma (CXR)). In particular, PLR combined with CXR enhanced the absorption and pharmacokinetics of puerarin through elevating puerarin solubility, regulating P-glycoprotein efflux and reducing claudin-5 expression. The combination of PLR and CXR reduced the diversity of gut microbiota and the ratio of Firmicutes to Bacteroidetes . In conclusion, combining PLR with CXR enhanced the absorption and pharmacokinetics of puerarin by modulating the intestinal barrier and influenced gut microbiota. The gut microbiota is likely to be a potential target for the combination of PLR and CXR.

In this study, a tip foam named as MIL-101(Cr)-NH₂/chitosan (MIL-101(Cr)-NH₂/CS) was prepared by cross-linking chitosan (CS) with MIL-101(Cr)-NH₂ using glutaraldehyde as the cross-linking agent. After freeze-drying, the internal structure of the MIL-101(Cr)-NH₂/CS foam became porous, thereby exposing more MIL-101(Cr)-NH₂, enriching binding sites, ensuring efficient adsorption, and allowing smooth passage of the sample solution. This also facilitates the inhalation and extrusion of the sample solution. Based on the unique advantages of the MIL-101(Cr)-NH₂/CS foam, a method combining pipette tip solid-phase extraction (PT-SPE) with high-performance liquid chromatography equipped with a fluorescence detector (HPLC-FLD) was established for the detection of puerarin and its metabolites (e.g., daidzein and equol) in rat serum. The method exhibited good linearity in the range of 0.5 to 5000 ng/mL, with limits of detection (LOD), lower limits of quantification (LLOQ), and limits of quantification (LOQ) ranging from 0.2 to 15 ng/mL, 0.5 to 50 ng/mL, and 1 to 100 ng/mL, respectively. The relative standard deviations (RSDs) of intra-day and inter-day precision for puerarin, daidzein, and equol were less than 19.8 %, meeting relevant regulatory requirements. The extraction recoveries of puerarin, daidzein, and equol ranged from 80.17 % to 91.70 %, 80.02 % to 87.41 %, and 80.29 % to 85.39 %, with corresponding RSDs of 3.2 % to 10.4 %, 11.2 % to 13.8 %, and 7.9 % to 11.1 %. These results indicate that the developed method meets the methodological requirements for the quantitative analysis of biological samples. The application of MIL-101(Cr)-NH₂/CS foam in PT-SPE further expands the application scope of metal-organic frameworks (MOFs) in solid-phase extraction (SPE) and provides a reference for PT-SPE technology in handling small-volume samples in complex matrices.

Puerarin has been reported to have anti-inflammatory, antioxidant, immunity enhancement, neuroprotective, cardioprotective, antitumor, and antimicrobial effects. However, due to its poor pharmacokinetic profile (low oral bioavailability, rapid systemic clearance, and short half-life) and physicochemical properties (e.g., low aqueous solubility and poor stability) its therapeutic efficacy is limited. The hydrophobic nature of puerarin makes it difficult to load into hydrogels. Hence, hydroxypropyl-β-cyclodextrin (HP-βCD)-puerarin inclusion complexes (PIC) were first prepared to enhance solubility and stability; then, they were incorporated into sodium alginate-grafted 2-acrylamido-2-methyl-1-propane sulfonic acid (SA-g-AMPS) hydrogels for controlled drug release in order to increase bioavailability. The puerarin inclusion complexes and hydrogels were evaluated via FTIR, TGA, SEM, XRD, and DSC. Swelling ratio and drug release were both highest at pH 1.2 (36.38% swelling ratio and 86.17% drug release) versus pH 7.4 (27.50% swelling ratio and 73.25% drug release) after 48 h. The hydrogels exhibited high porosity (85%) and biodegradability (10% in 1 week in phosphate buffer saline). In addition, the in vitro antioxidative activity (DPPH (71%), ABTS (75%), and antibacterial activity (Staphylococcus aureus,Escherichia coli, and Pseudomonas aeruginosa)indicated the puerarin inclusion complex-loaded hydrogels had antioxidative and antibacterial capabilities. This study provides a basis for the successful encapsulation of hydrophobic drugs inside hydrogels for controlled drug release and other purposes. Keywords:hydrogel;flavonoids;antioxidant;inclusion complexes;oral formulation
