In Situ Forming Injectable Hydrogel For Encapsulation Of Nanoiguratimod And Sustained Release Of Therapeutics

Zhenzhen Ma,Cheng Tao,Lin Sun,Shengbei Qi,Yuan Le,Jiexin Wang,Changhong Li,Xiangyuan Liu,Jianjun Zhang,Jinxia Zhao

Background Iguratimod (IGUR) is a novel disease-modifying antirheumatic drug used for treating rheumatoid arthritis (RA). To improve its bioavailability and to alleviate gastrointestinal side effects, we changed the formulation into nanoiguratimod-loaded hydrogel (NanoIGUR-loaded hydrogel) composites for sustained release of therapeutics. Methods IGUR was first encapsulated in biodegradable polyvinyl alcohol micelle by liquid antisolvent precipitation (LAP) technology, and then loaded into an in situ injectable hyaluronic acid hydrogel, which was cross-linked by PEG (Thiol)2 (HS-PEG-SH) through Michael addition reaction. In vitro, the biological effects (proliferation, migration, and invasion) of NanoIGUR-loaded hydrogel on fibroblast-like synoviocytes (RA-FLS) from RA patients were evaluated. In vivo, the pharmacokinetics of NanoIGUR-loaded hydrogel was assessed and the efficacy of NanoIGUR-loaded hydrogel in treating collagen-induced arthritis (CIA) rats was evaluated. Results By the LAP technique, we acquired the amorphous form nanoiguratimod, with an average size of 458 nm, which had higher dissolution rates and higher stability. The release of IGUR from hydrogel composite in PBS was gradual and sustained for up to 72 hrs compared with nanoiguratimod. Different concentrations of NanoIGUR-loaded hydrogel inhibited the proliferation, migration, and invasion of RA-FLS. The pharmacokinetic parameters showed better bioavailability and longer half-life time with NanoIGUR-loaded hydrogel by subcutaneous administration than oral raw iguratimod. Animal experiments confirmed that subcutaneous injection of NanoIGUR-loaded hydrogel (10 mg/kg every 3 days) and oral raw iguratimod (10mg/kg daily) showed similar efficacy in decreasing arthritis index score, pathological score, and expression of inflammatory cytokines. Conclusion Overall, we demonstrate that NanoIGUR-loaded hydrogel provides a new route of administration and extends the administration interval. It could be a promising drug-delivery approach in the management of RA.

The Enhanced and Tunable Sustained Release of Pesticides Using Activated Carbon as a Carrier

Jun Yang,Wanyu Zang,Zheng Zhang,Peng Wang,Qing Yang

The sustained release of pesticides improves drug utilization efficiency and reduces their adverse effects. Activated carbon (AC) is an excellent adsorbent and promising soil conditioner. It has a rich, porous structure and thus can store and gradually release drugs. In this study, three AC materials with surface areas ranging from 800–2000 m2/g were used and two types of modified activated carbons were prepared, and their capacity as drug carriers was evaluated by using 2,4-Dichlorophenoxyacetic acid sodium (2,4-D sodium) as the model pesticide. The preparations were characterized by scanning electron microscopy, nitrogen physical analysis, and zeta potential. The five preparations showed an enhanced and tunable sustained release of drugs. AC1, with the highest specific surface area, possesses the best drug-loading capacity, reaching 679.18 mg/g, but the lowest drug release rate of 32.31% in 96 h. PDA-AC3 has the lowest specific surface area, showing limited drug-loading ability, 82.94 mg/g, but 100% drug release within 72 h. This study suggests that activated carbon has potent applications in agricultural pest control as an inexpensive, effective, controllable, and safe pesticide carrier. Keywords:activated carbon;sustained release;dopamine;nitrogen-doped activated carbon;pest control

Sustained release of Eugenol from MCM-41 mesoporous silica for preservation of Agaricus bisporus

Kangqing Yuan,Fangxuan Yi,Jiawei Wang,Xiangyou Wang

The postharvest quality of Agaricus bisporus declines rapidly during refrigerated storage. A controlled-release system (Eugenol@MCM-41) was prepared by loading eugenol into MCM-41, with a loading capacity of 51%. Physicochemical characterization and all-atom molecular dynamics (MD) simulations were combined to clarify the sustained-release mechanism. MD simulations provided supportive evidence for favorable eugenol–silica interfacial interactions and restricted surface-associated mobility in the mesoporous model. In vitro release exhibited clear medium dependence and followed the Weibull model, consistent with diffusion-dominated release under confinement. In storage trials at 4 ± 1 °C, Eugenol@MCM-41 extended acceptable mushroom quality to 15 days. The treatment reduced weight loss, maintained firmness, delayed browning, and lowered malondialdehyde accumulation versus the control and pristine MCM-41. These results connect nanoscale confinement with preservation performance and support Eugenol@MCM-41 as an effective eugenol delivery system for preserving highly perishable foods.

Preparation and Properties of PVA/ZIF-8 Sustained-Release Fiber Loaded With Minoxidil Sulfate

Haowei Bai,Yingbo Chen,DengChang Zhu,Yongxiang Shi

To address the limitation of traditional minoxidil sulfate (MXS) nano-drug delivery systems—namely, short dosing cycles, frequent replacement requirements, and inability to fulfill cosmetic coverage needs simultaneously—this study aims to develop a composite fiber with dual functions: sustained release and cosmetic coverage. Employing a green and simple wet spinning technique, MXS-loaded ZIF-8 nanoparticles were functionally composite with a polyvinyl alcohol (PVA) polymer matrix, successfully yielding PVA/ZIF-8 composite fibers. The resulting composite fiber exhibits excellent mechanical properties, with a tensile strength reaching 2.5 cN/dtex and good aging resistance. The fiber inherits the high drug-loading capacity of ZIF-8 nanoparticles, achieving sustained release for up to 220 h. Additionally, the composite fiber possesses outstanding processability, enabling its further weaving into wigs. This study successfully developed a novel PVA/ZIF-8 composite fiber integrating sustained drug release with physical hair coverage. It expands new application directions for functional polymer composites in the biomedical field.

Electrospun cellulose acetate/poly(lactic acid) hybrid fibers for tunable sustained release of ibuprofen

Liu Yaqing,Ye Xiang-Yu,Gou Faliang,Xing Guiying,Yang Peng,Chen Chengguang,Shao Linjun

Precise modulation of drug release profiles from fibrous carriers remains a significant challengein sustained-release formulations. Herein, we present a novel and tunable drug delivery system based on electrospun cellulose acetate (CA) and poly(lactic acid) (PLA) hybrid fibers. Herein, CA/PLA composite fibers were designed and fabricated via electrospinning, and their potential as a drug delivery platform was evaluated. By varying the electrospinning solution concentration from 6% to 14%, smooth and uniform fibers with controlled mean diameters (0.59–1.92 μm) were successfully prepared. Meanwhile, the swelling properties and surface wettability of the fibers were effectively modulated by adjusting the CA/PLA ratio (30–70% PLA). Ibuprofen, used as a model drug, was uniformly encapsulated within the fiber matrices with drug encapsulation efficiency (> 93%). In vitro release studies demonstrated that the ibuprofen release rate decreased with increasing fiber diameter and PLA content, and exhibited pH-dependent behavior. The drug release profiles exhibited excellent fit to the Ritger-Peppas model, indicating a Fickian diffusion mechanism. Furthermore, strong correlations were established between drug release half-lives and key fiber parameters (diameter and PLA content). This work presents a simple yet effective strategy for tailoring drug release kinetics, highlighting the significant potential of CA/PLA composite fibers for advanced fibrous sustained-release drug delivery systems.

Fighting periodontitis with a pH-triggered nanocoating: A sustained-release strategy for simvastatin delivery

Siyu Du,Yi Zheng,Yabing Chen,Yijing Guo,Xinyue Kang,Lei Wang

The growing incidence of periodontitis demands innovative therapies, as current drug treatments are limited by short retention times, rapid clearance, and the risk of antimicrobial resistance. This study introduces a novel pH-responsive nano-miclle system for simvastatin (SIM) delivery. Its core innovation lies in being the first system ((SIM NP)n) of its kind to be stabilized by a cross-linked "outer shell," which significantly enhances its stability and controlled release capabilities. This "smart" system remains stable in a neutral environment (pH 7.4), preventing premature drug release. However, upon encountering the acidic inflammatory microenvironment of periodontitis, the shell degrades, triggering the sustained release of SIM directly at the site of inflammation. In vitro studies demonstrated that the SIM-loaded micelles possess potent immunomodulatory effects: they effectively suppressed pro-inflammatory M1 macrophage polarization (decreasing IL-1β, iNOS) while promoting anti-inflammatory M2 macrophage polarization (increasing Arg-1, IL-10). Mechanistic investigation confirmed this therapeutic effect is mediated by the PI3K/AKT/mTOR signaling pathway. More importantly, in a mouse model of periodontitis, the nano-micelles significantly reduced alveolar bone resorption, demonstrating potent anti-inflammatory and bone-protective efficacy in vivo. In conclusion, this "smart" pH-triggered delivery system for SIM offers a highly promising and effective strategy to overcome the limitations of traditional therapies, providing a new targeted path for the treatment of periodontitis.

Highly stable, temperature- and humidity-responsive entactogens for long-term sustained release of ClO₂

Qiao yuan Luo,Tianrui Zhou,Zhen Wang,Yuntong Liang,Lihua Fu,Chuanhui Xu,Bai Huang,Baofeng Lin

Eutectogels have emerged as promising materials for disinfection applications under complex environmental conditions, owing to their remarkable stability and adaptive properties. In this study, a deep eutectic solvent (DES)-activated eutectogel (PVA/CMC-ZE@NaClO 2 ) was developed through physical cross-linking of polyvinyl alcohol (PVA) and carboxymethyl cellulose (CMC) within a DES medium. The resulting gel exhibits dual-responsive behavior to both temperature (−20 to 50 °C) and humidity (55–90 % RH), enabling intelligent and sustained release of ClO₂ even under extreme environmental conditions. A key innovation lies in the role of DES as an activator, which reacts with NaClO₂ under moisture stimulation to continuously generate gaseous ClO₂. The eutectogel demonstrates high structural integrity across challenging environments—including cold, humid, and dry settings—maintaining stable ClO₂ release for over 20 days. This prolonged disinfectant delivery confers strong antibacterial efficacy, making the material suitable for broad public health applications. Additionally, the gel is readily reshapeable and reusable, while also being fully biodegradable within 20 days under ambient conditions. This work presents the first example of a eutectogel-based slow-release system for ClO₂, offering a new paradigm for designing intelligent, environmentally adaptive materials with extended release performance for disinfection in extreme settings.

Development of γ-CD-MOF/Chitosan platform with sustained release capacity for food preservation

Xuan Wang,Xu Meng,Tao Liang,Jiachun Song,Kamal Eldin Afaf,Hong Duan,Lingyan Zheng,Gang Chen

γ-Cyclodextrin (γ-CD) is one of the most widely used materials for encapsulating hydrophobic functional molecules due to its distinctive structural features, especially an internal hydrophobic cavity surrounded by external hydrophilic surface. However, its limited loading capacity, adsorption efficiency, and poor stability hinder its broader applications. To address this issue, γ-cyclodextrin-based metal-organic frameworks (γ-CD-MOFs) was formed by anchoring potassium ions with γ-CD through hydrothermal self-assembly. After loading different hydrophobic small molecule active substances, a multifunctional composite coating was prepared by embedding them into the chitosan (CS) matrix. The release kinetic model analysis confirmed that the release behavior of Cinnamaldehyde (Cia), 2-methoxycinamaldehyde (2-MCA), and Curcumin (Cur) from γ-CD-MOFs conformed to the Ritger-Peppas equation. Compared with CS, CS/γ-CD-MOF@Cia has a strong inhibitory effect on E. coli O157: H7 and S. aureus . Sprayed on green grapes, the shelf life is extended to 21 days. CS/γ-CD-MOF@2-MCA can significantly reduce the spoilage of milk caused by photooxidation. Furthermore, compared with CS, the oxidation and browning of banana peels were effectively delayed by CS/γ-CD-MOF@Cur, and complete blackening is not observed even after 11 days of storage. By combining the advantages of chitosan and γ-CD-MOF, the active molecules with different functions were encapsulated on the platform of γ-CD-MOF to achieve antibacterial, light-blocking, and antioxidant effects on food. The novel functional coating materials presented in this work offer a promising approach to effectively prolonging the shelf life of fresh foods products.

Glycyrrhizic acid encapsulated in zein/Astragalus polysaccharides nanoparticle: enhanced stability, bioactivity and sustained release

Xin Guo,Yong He,Lei Wang,Qian-Lan Wu,Jing Li,Tian-Le Mao,Kiran Thakur,Yi-Long Ma,Zhao-Jun Wei

Glycyrrhizic acid (GA) exhibits potent antiviral, anti-inflammatory, and hepatoprotective activities. However, its bioactivity is constrained by low stability under varying conditions, including pH and temperature. To improve its stability, GA was loaded into zein-Astragalus polysaccharides (APS) nanoparticles (zein-APS-GA NPs). The optimized nanoparticles had a diameter of 101.10 nm, a polydispersity index of 0.092, and an encapsulation rate of 91.70 %. Simulated digestion experiments revealed a bioaccessibility of 72.84 % for nanoparticles, compared to 30.70 % for free GA. The radical scavenging rates of the nanoparticles ranged from 66.23 % to 73.34 %, and retained over 40 % after 48 h at room temperature. Stability assays confirmed their robustness across a range of ionic strength, pH values, temperatures, and storage durations. Furthermore, zein-APS-GA NPs increased the viability of alcohol-injured AML-12 cells to 95.50 % at 100 μg/mL (containing 91.7 μg/mL GA), significantly surpassing that of free GA (63.29 %, 100 μg/mL), highlighting their enhanced stability and cytoprotective efficacy.

Sustained-Release Photothermal Microneedles for Postoperative Incisional Analgesia and Wound Healing via Hydrogen Therapy

Aining Zhang,Xue Jiang,Bingrui Xiong,Jiayi Chen,Xin Liu,Siyuan Wang,Bofu Li,Mian Peng,Wei Li

Effective management of postoperative pain and wound healing presents significant challenges in clinical settings, driving the need for innovative therapeutic approaches. The analgesic and wound healing effects of hydrogen (H 2 ) have gradually been recognized; however, the lack of efficient hydrogen delivery systems remains a major limitation. This study introduces a novel transdermal drug delivery system, which utilizes sustained-release photothermal microneedles (MNs) to ameliorate incisional pain and accelerate wound healing. Polydopamine (PDA)-modified ZIF-8@ammonia borane (AB) nanoparticles with photothermal conversion properties are designed, along with temperature-responsive QX-314-loaded polycaprolactone (PCL) microspheres for controlled release, which are delivered in vivo by dissolvable MNs. In vitro results showed that PDA@ZIF-8@AB nanoparticles can release H 2 continuously for up to 5 days in an acidic microenvironment, while the photothermal properties of PDA facilitated controlled release of QX-314 through 6 cycles of near-infrared (NIR) exposure. In vivo experiments demonstrated that the MN system provided sustained analgesia for up to 5 days and promoted wound healing in the acidic microenvironment of postoperative incisions. Upon NIR exposure, the photothermal conversion of PDA activated membrane ion channels and induced thermally triggered deformation of PCL@QX-314 microspheres, allowing for on-demand release of QX-314 and targeted neuronal uptake, thus offering personalized analgesia. In vitro cell experiments and in vivo studies confirmed the biocompatibility of the system. This innovative approach not only highlights the dual role of H 2 in pain relief and wound healing but also provides a new personalized treatment strategy for postoperative pain management with promising clinical applications.

Temperature-responsive lauric acid-coated ferrate-SiO2 composite for sustained release and norfloxacin degradation

Xiaoke Ma,Xiao Luo,Dongmei Zhou,Yuan Zhang,Lin Zhao,Yongkui Yang,Peizhe Sun

Chemical oxidation, a cornerstone for groundwater and soil remediation, continues to face two major challenges: the prolonged delivery time required for oxidant diffusion and competitive consumption of oxidants by coexisting reductants. Potassium ferrate (K 2 FeO 4 ) is a highly efficient and eco-friendly water treatment agent owing to its strong oxidizing properties. However, its practical application is hindered by self-decay and substrate interference. Therefore, this study presented a novel slow-release ferrate material (K 2 FeO 4 @SiO 2 ) that integrated the high oxidizing capabilities of ferrates with the structural stability and functional advantages of nanosized SiO 2 . Given the low heating (30–100 °C) process is currently used for the thermal activation of oxidant in in-situ groundwater and soil remediation, a temperature-responsive system (K 2 FeO 4 @SiO 2 -LA) was further developed using lauric acid as a phase-change material to achieve controlled release of ferrate active components. Characterization confirmed the effective encapsulation of K 2 FeO 4 in the temperature-controlled material. The effectiveness of this material for removing norfloxacin (NOR) from water and its underlying degradation mechanism were comprehensively investigated. The material exhibited a removal efficiency of 9.4 % at 30 °C, which increased to 76.4 % at 50 °C. It exhibited excellent degradation performance and precise temperature responsiveness across a broad pH range and maintained high efficiency. High-valent iron (Fe(VI), Fe(V), Fe(IV)) served as the active oxidizing agent in the K 2 FeO 4 @SiO 2 -LA system. The sustained-release mechanism of the SiO 2 core-shell structure effectively suppressed K 2 FeO 4 self-decay, enhancing the production of high-valent iron and the NOR removal efficiency. Moreover, temperature-controlled materials were applied for efficient antibiotic removal from practical waters, offering a novel strategy for managing antibiotic pollution.

Ultrasound Enhancing ZnS for Arsenic Removal from Acid Wastewater by Sustained Release In Situ

Li Rui,Zhang Mengmeng,Li Jing,Zhang Chaobo,Fu Guang

Acid wastewater (AWW) from the non-ferrous metal smelting industry is characterized by a high level of acidity and a high concentration of As. Sulfiding agents are commonly used for arsenic removal from acid wastewater because of their high efficiency. To address the issues associated with traditional sulfiding agents, such as the release of harmful hydrogen sulfide gas and the introduction of large amounts of sodium ions, this paper proposes a new process of ultrasound-enhanced ZnS for arsenic removal from acid wastewater by sustained release in situ. The low solubility of ZnS provides the advantages of matching the reaction rate to arsenic removal efficiency, inhibiting the escape of H 2 S gas, and preventing the introduction of other cations. Ultrasound can open the ZnS surface encapsulation and accelerate the reaction rate. Experimental research has shown that the arsenic removal rate reached 99.96% and the arsenic concentration was only 0.42 mg/L at an acidity of 120 g/L, S/As molar ratio of 3:1, reaction temperature of 60°C, and ultrasonic power of 100 W for 120 min. The development of this process is of great significance as an application prospect for the arsenic removal from acid wastewater in zinc smelting.
