Shape-complementary microspheres pack into irregular postoperative cavities and act as a local curcumin reservoir. • Hydrated microsphere beds retain integrity during compression, repeated loading and fluid flushing. • Nanosheet content and laser power tune mild heat-assisted curcumin release for local tumor-cell suppression. Postoperative recurrence of triple-negative breast cancer often starts from residual cells left in an irregular lumpectomy cavity, where a short-lived drug depot is easily washed away and a weak filler gives little structural support. We designed injectable SFMA/MXene/curcumin-loaded mesoporous silica nanoparticle (Cur@MSN) microspheres as a cavity-filling chemophotothermal depot rather than a membrane-like wound dressing. XPS confirmed silica-, curcumin- and MXene-associated signals, including Ti-O contributions in the composite. Mechanical tests showed that MXene/MSN reinforcement increased single-particle stiffness and improved hydrated packed-bed compression resistance, cyclic modulus retention and anti-flushing stability. Curcumin release continued for 14 days, and near-infrared irradiation increased the 32 h release from 68.97 ± 0.55% to 82.93 ± 0.45%. Normal 10A and NIH3T3 cells retained high viability, whereas 4 T1 cells showed concentration-dependent inhibition. In vivo thermal screening identified 2.0 mg/mL MXene with 0.50 W/cm 2 irradiation as a mild heating condition (∼43 °C). In a postoperative 4 T1 model, Cur/MXene microspheres with irradiation reduced bioluminescent tumor signals, supporting a locally retained, mechanically reinforced depot for recurrence-oriented therapy. The widespread use of 4-nitrophenol (4-NP) and curcumin, although beneficial across numerous industries, poses potential risks to human health and the environment. Thus, creating a sensitive and straightforward detection strategy is crucial for the rapid monitoring of these pollutants. Herein, a nitrogen-doped carbon dots (N-CDs)-based dual-excitation ratiometric fluorescent probe is developed, capable of detecting 4-NP and curcumin. The N-CDs are prepared from tris (hydroxymethyl) aminomethane (Tris) and phloroglucinol via a facile hydrothermal method, exhibiting a strong cyan fluorescence centered at 497 nm under excitation at 399 nm and 469 nm. In the presence of 4-NP, the fluorescence excited at 399 nm is significantly quenched compared to that excited at 469 nm due to the inner filter effect (IFE) and static quenching, achieving a limit of detection (LOD) of 76 nM within a linear range of 0.076–110 μM. In contrast, curcumin induces nearly complete fluorescence quenching at both excitation wavelengths, resulting from the quenching mechanism of dominant IFE and partial fluorescence resonance energy transfer. This response allows for curcumin detection over a concentration range of 0.081–90 μM with a low LOD of 81 nM. Furthermore, a smartphone-assisted sensing platform is constructed for the visual detection of 4-NP and curcumin in actual samples, achieving LODs as low as 117 nM and 123 nM, respectively. Therefore, this work enhances the capability for multi-analyte monitoring in complex systems and provides a practical and instructive strategy for detecting 4-NP and curcumin.

Curcumin (CUR) is among the most appropriate and natural-based anticancer drugs that can be applied effectively treat different classes of cancers. However, CUR suffers from a low half-life and stability in the body, which has restricted the efficacy of its delivery applications. This study is dedicated to introducing the pH-sensitive nanocomposite of chitosan (CS)/gelatin (GE)/carbon quantum dots (CQDs) as an applicable nanocarrier for enhancing CUR half-life and its delivery restrictions. The CS/GE hydrogel was synthesized by the physical crosslinking method, which improves the biocompatibility of this hydrogel. Moreover, the water-in-oil-in-water (W/O/W) double emulsion approach is involved in fabricating the drug-loaded CS/GE/CQDs@CUR nanocomposite. Afterward, drug encapsulation (EE) and loading efficiencies (LE) have been determined. Furthermore, FTIR and XRD assessments were performed to confirm the CUR incorporation into the prepared nanocarrier and crystalline features of the nanoparticles. Then, by employing Zeta potential and dynamic light scattering (DLS) analysis, the size distribution and stability of the drug-loaded nanocomposites have been assessed, which indicated monodisperse and stable nanoparticles. Furthermore, field emission scanning electron microscopy (FE-SEM) was utilized that confirmed the homogeneous distribution of the nanoparticles with smooth and quite spherical structures. In vitro drug release pattern was studied and the kinetic analysis was performed using a curve fitting technique to determine the governing release mechanism at both acidic pH and physiological conditions. The obtained outcomes from release data revealed a controlled release behavior with a 22-hour half-life, while the EE% and EL% were acquired at 46.75 % and 87.5 %, respectively. In addition, the MTT assay has been carried out on U-87 MG cell lines to evaluate the cytotoxicity of the nanocomposite. The findings showed that the fabricated nanocomposite of CS/GE/CQDs can be assumed as a biocompatible CUR nanocarrier, while the drug-loaded nanocomposite of CS/GE/CQDs@CUR showed enhanced cytotoxicity compared to the pure CUR. Based on the obtained results, this study suggests the CS/GE/CQDs nanocomposite as a biocompatible and potential nanocarrier for ameliorating CUR delivery restrictions to treat brain cancers.

In this work, double-layered emulsions stabilized by bacterial cellulose nanofibers/soy protein isolate/chitosan complex were prepared using layer-by-layer self-assembly technology and used for the delivery of curcumin, and the physical and delivery properties of the prepared double-layered emulsions were investigated. The zeta-(ζ) potential of colloidal particles confirmed that the chitosan was adsorbed on surface of the single-layered emulsion droplets by attractive electrostatic interactions. Meanwhile, the physical properties showed that the double-layered emulsions displayed high pH-sensitivity and showed relatively higher stability under acid environment. Furthermore, the in vitro digestion result showed that the curcumin loaded by double-layered emulsions were more stable during simulated gastric digestion as compared with that loaded by single-layered emulsions. Especially, the bioaccessibility of curcumin delivered by the double-layered emulsions (77.4 ± 3.25%) was significantly higher than that of curcumin delivered by single-layered emulsions (32.8 ± 2.56%). Therefore, it can be concluded that the prepared double-layered emulsions showed a great potential application in the delivery of fat-soluble bioactive compounds.

Cancer-associated fibroblasts (CAFs) are a major component of the tumor microenvironment (TME). In hepatocellular carcinoma (HCC), quiescent hepatic stellate cells (HSCs) could be activated to become CAFs, which play a critical role in tumor progression and drug resistance. Therefore, recent efforts have been focused on combining anti-HSC and pro-apoptotic activities to improve anti-tumor efficacy of drugs. In this study, glycyrrhetinic acid and hyaluronic acid–modified liposomes (GA-HA-Lip) were prepared for co-delivery of curcumin (CUR) and berberine (BBR) for the treatment of HCC. Furthermore, we established the LX-2+BEL-7402 co-cultured cell model and implanted the m-HSCs+H22 cells into a mouse to evaluate the anti-tumor effect of CUR&BBR/GA-HA-Lip both in vitro and in vivo . The results showed that CUR&BBR/GA-HA-Lip could accumulate in tumor tissues and be taken up by HSCs and BEL-7402 cells simultaneously. Compared with free CUR, the combination therapy based on GA-HA-Lip exhibits stronger pro-apoptotic and anti-proliferation effect both in vitro and in vivo . The anti-tumor mechanistic study revealed that CUR&BBR/GA-HA-Lip could inhibit the activation of HSCs and restrain drug resistance of tumor cells. In summary, CUR&BBR/GA-HA-Lip could be a promising nano-sized formulation for anti-tumor therapy.

In order to study the effects of the compound preservatives (curcumin and piperine (CP)) and vacuum packaging (VP) on the quality of salmon during cold chain logistics suffered from temperature abuse, the physiochemical indexes (texture, water holding capacity (WHC), total volatile basic nitrogen (TVB-N), thiobarbituric acid reactive substances (TBARS), free amino acids (FAA) contents), microbial indicators (total mesophilic bacteria count (MBC), total psychrotrophic bacteria count (PBC), H2S-producing bacteria count (HSBC)) were determined, and the moisture changes were explored by near-infrared (NIR) spectroscopy and low-field nuclear magnetic resonance (LF-NMR). The results showed that the treatment of curcumin and piperine in combination with vacuum packaging could maintain the quality of salmon suffered from temperature abuse most effectively. At the end of storage, the MBC of VP+CP was only 4.95 log CFU/g, which was about 1 log CFU/g lower than the control sample stored at the same condition. The combined treatment also retarded the increase of TVB-N, TBARS, and the decrease of hardness, springiness, and a* value, as well as water migration in salmon, contributing to higher water holding capacity and better appearance. Besides, VP+CP retarded the decrease of free glutamate, which contributed to umami taste. Due to the biological activity and safety of the preserves, the combined treatment could be a promising method for preservation of seafood.

In this study, ovalbumin (OVA) interacted with pectin (PE) to form soluble electrostatic complexes to improve the functional properties of high internal phase Pickering emulsions (HIPEs) under extreme conditions. The results showed that the stability of the OVA-PE soluble complexes-stabilized HIPEs was significantly better than that of the free OVA-stabilized HIPEs and was modulated by the biopolymer ratio. In particular, the complexes at an OVA:PE ratio of 1:1 (C-1:1) may form particulates with a core-shell structure by a flocculation mechanism. The C-1:1-stabilized HIPEs had the smallest oil droplet size (11.34 ± 1.14 μm) and the best resistance to extreme environmental stresses due to their strong, rigid structure and dense interfacial architecture. The in vitro digestion results showed that the bioaccessibility (from 18.3% ± 0.5% to 38.8% ± 4.8%) of curcumin improved with increasing PE content. Our work is helpful in understanding OVA-PE complexes as stabilizers for HIPEs and their potential applications in food delivery systems.

The therapy of tumor recurrence resulting from surgical resection of osteosarcoma is still a significant challenge for clinicians. In this study, we designed curcumin (CM) and indocyanine green (ICG) co-loaded poly(L-lactic acid) (PLLA) (PLLA-CM-ICG) composite microparticles , combining photothermal therapy with chemotherapy to enhance osteosarcoma therapy synergistically. PLLA-CM-ICG microparticles of sizes of (838.3 ± 5.4) nm were prepared by solution-enhanced dispersion by a supercritical CO 2 process. ICG in the PLLA-CM-ICG microparticles was more stable than free ICG. The PLLA-CM-ICG microparticles displayed pH- and near-infrared (NIR)-responsive CM release behavior. Moreover, NIR irradiation improved the anticancer activity of the PLLA-CM-ICG microparticles at a concentration of 250 μg/mL on MG-63 cells. Therefore, PLLA-CM-ICG microparticles could be employed as promising drug delivery systems for synergic photothermo-chemotherapy in osteosarcoma treatment.

Curcumin (CUR) is a promising edible phytochemical compound with ideal ulcerative colitis (UC) treatment activity; however, it is characteristically instable in the digestive tract and has a short retention time in colon. Therefore, we designed and fabricated an oral food-grade nanocarrier composed of tannic acid (TA)-coated, Genipin (Gnp)-crosslinked human serum albumin (HSA) to encapsulate CUR (TA/CUR-NPs). The resulting CUR nanoparticles (NPs) were about 220 nm and −28.8 mV. With the assistance of TA layer and Gnp-crosslinking, the entire nano-scaled system effectively delayed CUR release in simulated gastric fluid, prolonged its colon adhesion and increased its uptake in Caco-2 cells. As expected, TA/CUR-NPs oral administration significantly alleviated colitis symptoms in DSS-treated mice when compared with controls by inhibiting the TLR4-linked NF-κB signaling pathway. Collectively, this study indicates that we have developed a convenient, eco-friendly, nano-scaled vehicle for oral delivery of CUR with anti-UC benefit.

Aim: To design novel emulsifiers with the ability to improve the storage and digestion stability of curcumin emulsions, besides to investigate the influence of phenolic acids types on the emulsify ability of soy protein-pectin-phenolic acids complexes obtained by ultrasonication. Methods: The ternary complexes were characterised by particle size, morphology, zeta potential, X-ray diffraction, Fourier transform infra-red and fluorescence spectroscopy. Additionally, changes in droplet size, charge, and microstructure were monitored as quantitative stability index of curcumin emulsions. Results: Phenolic acid types significantly affected the formation of ternary complexes. Soy protein-pectin-ferulic acid complex (S-P-F) stabilised curcumin emulsion had the best emulsifying property, followed by soy protein-pectin- ellagic acid (S-P-E), and soy protein-pectin-tannic acid complexes (S-P-T). Moreover, S-P-F emulsion was found to retain efficiently cucumin within 30 days storage (77.35%) and simulated gastrointestinal tract (64.09%).Conclusion: Protein-polysaccharide-phenolic acids emulsions are effective oral delivery systems for hydrophobic bioactives.

It is still a tremendous challenge to combat multidrug resistant (MDR) cancer up to now, due to cancer resistance to chemotherapeutic agents. Herein, we put a new strategy for effective treatment of MDR cancer by improving chemotherapeutic efficacy via chemosensitizer in combination with phototherapy. Our previous work has demonstrated poly (ethylene glycol) - poly (curcumin-dithiodipropionic acid) (PEG-PCDA) is a good pre-chemosensitizer and enhances the sensitivity of MDR cancer cells to doxorubicin (DOX). In this study, we firstly synthesized mesoporous carbon nanoparticles (MCNs) having excellent photothermal and photodynamic effects, which could load a high dose of hydrophobic drug and presented pH -responsive drug release characteristics. On this basis, we developed a multifunctional MCN based drug delivery system to take chemo/photo- co-therapy against MDR human breast cancer MCF-7/ADR cells. The MCN based drug delivery system loaded 36.4% DOX and was coated with 23.2% PEG-PCDA, which could take photothermal and photodynamic effects under near-infrared (NIR) irradiation and trigger drug release by pH, glutathione (GSH), and/or NIR irradiation, termed as PEG-PCDA/DOX@MCNs. Once the PEG-PCDA/DOX@MCNs had been delivered into MCF-7/ADR cells, DOX release was triggered by degradation of PEG-PCDA via GSH and weak acidic pH, and in combination with MCN yielded photothermal and photodynamic effects under NIR irradiation, PEG-PCDA/DOX@MCNs could reduce premature drug release and then enhance anti-MDR cancer efficacy.

Curcumin (CUR) exhibits potential inhibitory effects on tumor growth; however, its hydrophobicity and instability limit its clinical applications. In the present study, we developed CUR nanoparticles (CUR-NPs) and evaluated their biochemical characteristics. Cell uptake and proliferation were assessed using scratch and Transwell assays, respectively. Western blotting was performed to investigate the expression levels of proteins related to the NF-κB/PRL-3 signaling pathway, inflammatory response, cell proliferation, and cell migration in SKOV3 cells. Our findings showed that the blank vector was not cytotoxic to cells, allowing us to disregard any effects caused by the vector itself. CUR-NPs exhibited concentration- and time-dependent inhibitory effects on cell proliferation, surpassing those of CUR alone. Increasing the concentration of CUR-NPs resulted in a reduced cell scratch-healing ability and lower chamber migration capacity. Compared to the control group, expression levels of proteins associated with NF-κB/PRL-3 signaling pathway, inflammatory response (TNF-α and IL-6), cell proliferation (cyclin E1 and cyclin A1), as well as cell migration (N-cadherin and vimentin) were significantly elevated in the lipopolysaccharide (LPS) stimulation and NF-κB p65 overexpression groups. Conversely, E-cadherin expression was significantly decreased under these conditions. However, treatment with high concentrations of CUR-NPs effectively reversed these changes. These results highlight the significant ability of CUR-NPs to inhibit human ovarian cancer cell proliferation and migration, while suppressing inflammatory responses through the regulation of the NF-κB/PRL-3 signaling pathway.
