An Immunoregulatory and Metabolism-Improving Injectable Hydrogel for Cardiac Repair After Myocardial Infarction

Sun Yage,Zhao Xinrui,Zhang Qian,Yang Rong,Liu Wenguang

The hypoxia microenvironment post myocardial infarction (MI) critically disturbs cellular metabolism and inflammation response, leading to scarce bioenergy supplying, prolonged inflammatory phase and high risk of cardiac fibrosis during cardiac restoration. Herein, an injectable hydrogel is prepared by Schiff-base reaction between fructose-1,6-bisphosphate (FBP)-grafted carboxymethyl chitosan (CF) and oxidized dextran (OD), following by loading fucoidan-coated baicalin (BA)-encapsulated zein nanoparticles (BFZ NPs), in which immunoregulatory and metabolism improving functions are integrally included. The grafted FBP serves to enhance glycolysis and provide more bioenergy for cardiomyocytes survival under hypoxia microenvironment, and elevating cellular antioxidant capacity via pentose phosphate pathway. OD with intrinsic anti-inflammatory effect can induce M2 polarization of macrophages to accelerate inflammatory elimination. While facing the possibility of endothelial-to-mesenchymal transition (EndoMT) caused by excessive expressed TGF-β1 secreted from M2 macrophages, BFZ NPs can target endothelia cells and intracellularly release BA to regulate the level of fatty acid oxidation, resulting in retained endothelial features and decreased risk of cardiac fibrosis. After being injecting the hydrogel into rats’ infarcted cardiac, the 28 day-post surgical outcomes demonstrate its benign effects on restoring cardiac functions and attenuating adverse left ventricular remodeling. This study shows a promising measure for MI treatment with immunoregulating and metabolism regulation comprehensively.

An intelligent thymol/alizarin-loaded polycaprolactone/gelatin/zein nanofibrous film with pH-responsive and antibacterial properties for shrimp freshness monitoring and preservation

JinTao He,Qiong Peng,JingJing Wang,Jing Deng,Wen Li,QinLu Lin,FeiFei Zhong,Xu Xia

Improper storage methods cause food resources to be wasted, and the development of multifunctional intelligent packaging can realize freshness monitoring and extend the shelf life. In this study, an intelligent alizarin/thymol-loaded polycaprolactone/gelatin/zein nanofibrous film was prepared and achieved the dual functions of pH-responsive and antibacterial properties. The film was fabricated using electrospun technology and characterized by SEM, FT-IR, WCA, TGA, DSC, and mechanical property tests, which had good antioxidant properties (81.40 ± 0.74 % free radical scavenging). The film could effectively inhibit the growth of Escherichia coli , Staphylococcus aureus , and Shewanella putrefaciens within 12 h and exhibited light yellow-light purple-dark purple color change at pH 2–12, with good color stability and color responsiveness to ammonia in the environment. In the shrimp application, shelf life was extended by 2 days and freshness could be monitored by the color change. This scientific research opens up new possibilities for realizing multifunctional food packaging alternatives.

Antioxidant activities potential of tea polysaccharide fractions obtained by ultra filtration

Yuanfeng Wang,Zhiwei Yang,Xinlin Wei

Three polysaccharide fractions (TPS1, TPS2 and TPS3) with different molecular weights were obtained using ultra filtration membranes from crude tea polysaccharide (CTPS) extracted from abandoned lower grade tea leaves. Each fraction contained different contents of neutral sugar, uronic acid, protein, and total polyphenols. These differences provided basis for the antioxidant and free radical scavenging activity of these polysaccharide fractions. The molecular weights of TPS1, TPS2, and TPS3 were around 2.40   ×   10 5   Da, 2.14   ×   10 4   Da, and 2.46   ×   10 3   Da, respectively. In general, TPS1 and CTPS had stronger antioxidant activity, TPS2 and TPS3 had lower antioxidant activity. TPS1 had higher activity for DPPH and lipid per oxidation inhibition. But it had lower capacity for reducing power and metal chelating. This might be due to its higher content of hexuronic acid and larger molecular weight. The order of inhibition activity of lipid per oxidation of various polysaccharide fractions was the same as DPPH radical scavenging activity, as well as the order of metal chelating activity of various polysaccharide fractions similar to hydroxyl radical scavenging activity, which demonstrated that hydroxyl radical scavenging activity of polysaccharide relied heavily on the Fe 2+ metal chelating to decrease the generation of hydroxyl radical.

A rapid preparation strategy of konjac glucomannan-based fiber film incorporated with elderberry anthocyanins via microfluidic blow spinning for fresh-cut apple preservation

Lihong Lin,Xianrui Chen,Wanxin Hong,Di Zhang,Xinyan Wen,Nitong Bu,Chengrong Wen,Ruojun Mu,Lin Wang,Jie Pang

It is still a challenge to use a fast and efficient method for preserving fresh-cut fruits from browning. To address this problem, we developed konjac glucomannan (KGM) incorporated with elderberry anthocyanins (EA) to form film-forming solution (KEA) combined with polyvinylpyrrolidone (PVP) solution to produce KEA/PVP fiber films by microfluidic blow spinning (MBS). The introduction of PVP and EA improved the spinnability and function properties of KGM-based fiber film, respectively. The fiber diameter of KEA/PVP films was increased with the addition of KEA. The FT-IR and XRD results verified that hydrogen bonds formed among KGM, EA, and PVP, contributing to the enhanced thermal stability, water barrier (WCA: 65°), and mechanical properties (TS: 2.73 MPa) of KEA/PVP films. The KEA/PVP films exhibited excellent antioxidant properties with DPPH and ABTS free radical scavenging rates of 74.69 % and 96.18 %, respectively. Besides, the antibacterial ability of KEA/PVP films against Escherichia coli and Staphylococcus aureus was also enhanced. The fresh-cut apple was effectively preserved that packaged in the KEA/PVP films and exhibited the best preservation effect compared with other groups. This work expanded the application of MBS and natural active compounds incorporated into KGM-based fiber film for fresh-cut apple preservation.

Characterization of flavonoids from Dryopteris erythrosora and evaluation of their antioxidant, anticancer and acetylcholinesterase inhibition activities

Jianguo Cao,Xian Xia,Xuefei Chen,Jianbo Xiao,Quanxi Wang

The profiles and bioactivities of flavonoids extracted from Dryopteris erythrosora were investigated. The total flavonoid content in full plant of D. Erythrosora is about 14.33%. The main flavonoids in D. Erythrosora were identified as gliricidin 7-O-hexoside, apigenin7-O-glucoside, quercetin 7-O-rutinoside, quercetin 7-O-galactoside, keampferol 7-O-gentiobioside, keampferol-3-O-rutinoside, myricetin 3-O-rhamnoside and quercitrin by means of HPLC-DAD–ESI-MS. Flavonoids (0.36   mg/ml) extract from D. erythrosora showed similar 2,2-diphenyl-1-picrylhydrazyl free radical (DPPH ), 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) radical (ABTS ), superoxide anion scavenging potential and ferric reducing antioxidant potential (FRAP) with that of rutin (0.80   mg/ml). However, the antioxidant power by FRAP assay of 0.36   mg/ml flavonoids extract from D. erythrosora was much weaker than that of 0.80   mg/ml rutin. Moreover, the flavonoids extract from D. erythrosora showed obvious cytotoxic effects on A549 cells. The antioxidant activities of flavonoids extracts from 69 ferns showed a significant reciprocal proportion to the total flavonoids contents. The flavonoids extract from D. erythrosora exhibited a dose-dependent inhibition against acetylcholinesterase. Moreover, the anticancer activity slightly increased with improving antioxidant potential of fern flavonoids. Fern flavonoids are excellent function foods.

A carbon dot nanozyme hydrogel enhances pulp regeneration activity by regulating oxidative stress in dental pulpitis

Zhang Yingjuan,Huang Xianxian,Luo Yicai,Ma Xiangyu,Luo Ling,Liang Ling,Deng Tingting,Qiao Yang,Ye Fanggui,Liao Hongbing

Preserving pulp viability and promoting pulp regeneration in pulpitis have attracted widespread attention. Restricted by the oxidative stress microenvironment of dental pulpitis, excessive reactive oxygen and nitrogen species (RONS) trigger uncontrolled inflammation and exacerbate pulp tissue destruction. However, modulating redox homeostasis in inflamed pulp tissue to promote pulp regeneration remains a great challenge. Herein, this work proposes an effective antioxidative system (C-NZ/GelMA) consisting of carbon dot nanozymes (C-NZ) with gelatin methacryloyl (GelMA) to modulate the pulpitis microenvironment for dental pulp regeneration by utilizing the antioxidant properties of C-NZ and the mechanical support of an injectable GelMA hydrogel. This system effectively scavenges RONS to normalize intracellular redox homeostasis, relieving oxidative stress damage. Impressively, it can dramatically enhance the polarization of regenerative M2 macrophages. This study revealed that the C-NZ/GelMA hydrogel promoted pulp regeneration and dentin repair through its outstanding antioxidant, antiapoptotic, and anti-inflammatory effects, suggesting that the C-NZ/GelMA hydrogel is highly valuable for pulpitis treatment.

A strategy to reduce the bitterness of green tea extract: Preparation of cyclodextrin inclusion and assessment of its antioxidant activity and stability

Xuemei Tong,Xin Wang,Yamei Chen,Mengjia Ma,Yongzhen Li,Xiao Yang,Xianggui Chen,Qin Wang,Yukun Huang

Green tea extract (GTE) is rich in polyphenols that are the main body of functional activity, but the natural bitterness greatly restricts its applications in food areas. In this study, the effect of β-cyclodextrin (β-CD) on the bitterness and properties of GTE were investigated. The bitterness intensity was evaluated by determination of catechin content and the electronic tongue analysis. It was found that the bitterness of GTE decreased significantly by complexing with 20 % β-CD. Subsequently, the physicochemical properties were determined, including the determination of functional substances by high performance liquid chromatography, as well as the assessment of antioxidant activity and stability under different conditions (heat, acid/base, time). It was found that β-CD inclusion of active substances in GTE protected its functional components and delayed the deactivation rate. Additionally, the formation of a complex between β-CD and components of GTE was confirmed by UV–vis, FT-IR, thermogravimetric analysis, and scanning electron microscope. Therefore, the findings of the present work suggest that the masking of bitter substances and the protection of functional components of GTE through preparing the β-CD inclusion is a feasible way to expand its applications.

Antioxidant activity and mechanism exploration for microwave-assisted extraction of flavonoids from Scutellariae Radix using natural deep eutectic solvent

Xiaoli Guo,Tingze Ren,Hongbo Li,Qingxin Lu,Xin Di

In this work, a natural deep eutectic solvent combined with microwave-assisted extraction (NADES-MAE) was studied and effectively utilized for the simultaneous extraction of the flavonoids from Scutellaria baicalensis. NADES composed of L-proline and urea were selected to achieve the efficient extraction of baicalin, baicalein, and wogonin. Box Behnken design (BBD) was applied to optimize the extraction conditions. The optimum conditions were as follows: L-proline and urea with a molar ratio of 2:1, water content of 40 %, extraction temperature of 110 °C, extraction time of 20 min, and liquid–solid ratio of 40 mL/g. Compared with the traditional extraction method, NADES-MAE displays the merits of higher yield, shorter extraction time, and less organic solvent consumption. In addition, the as-prepared NADES was characterized by FTIR and 1 H NMR, and the extraction mechanism for the flavonoids was investigated by density functional theory. The outcomes of biological activities in vitro showed that the extracts revealed desirable antioxidant activity. After assessing the ecological impact, the cytotoxicity of NADES was investigated and the results displayed that toxicity of the NADES was very low or even negligible with EC 50  > 2000 mg/L. The results indicated that the suggested approach was sensitive, green, and simple, showing good potential for analysis of active components from natural sources.

Applying Huaimi (Flos Sophorae Immaturus) as a brewing adjunct and its impact on sensory properties of beer

Yongxin Zhang,Xiaochuan Wang,Hua Liu,Zhengcong Peng,Jian Lu,Dianhui Wu

Huaimi is a precious Chinese herbal medicine mainly containing rutin. In this study, Cyan Huaimi (CH) and Golden Huaimi (GH) were introduced at various stages of beer production. A total of 8 beer groups and 1 control group were set up to compare their physicochemical, bioactivity, volatile compounds and sensory characteristics. The results indicated that compared with the control group, adding Huaimi significantly reduced the pH and real extract of beer, and improved the other Physicochemical parameters. Furthermore, the addition of Huaimi brought higher levels of total polyphenols, flavonoids, and phenols compounds in 8 beer groups, particularly rutin content, reaching as high as 212.69 mg/L. Moreover, through the evaluation of DPPH, ABTS, and reducing power methods revealed that adding Huaimi at the boiling stage benefited most in improving antioxidant capacity. However, the results of sensory evaluation demonstrated that panelists preferred the beer with Huaimi added at the saccharification stage because of its smoother foam. Volatiles analysis revealed that adding Huaimi introduced twelve new volatiles to the beer, enhancing its flavor and taste complexity.

Antioxidant properties and changes in vitro digestion of the fermented kiwifruit extract prepared by lactic acid bacteria and yeasts

Yanhua Li,Weijun Wang,Yangyong Deng,Junlong Gao,Jiayun Shi,Liqin Cai

The health benefits of fermented fruits have attracted consumers’ attention. High levels of antioxidant ability in the fermented kiwifruit extract were found at the early stage of fermentation. The co-fermention with Lactobacillus paracasei LG0260 and Kluyveromyces marxianus J2853 showed the highest ABTS radical scavenging ability (ABTS ⋅+ -SA) and superoxide dismutase (SOD) activity. Also, the typical antioxidant components of SOD activity, vitamin C concentration and total phenol content were highly correlated with ABTS ⋅+ -SA. Obviously, polyphenols in the fermented kiwifruit extract evolved into monophenols during fermentation. Compared to undigested samples, the activity of ABTS ⋅+ -SA and reducing power capacity (RP-CA) after the final intestinal digestion decreased and ranged 387.44–531.89 VCμg/mL, 650.95–981.63 VCμg/mL, respectively ( P  < 0.05). Meanwhile, SOD activity on the 10th day of fermentation were still remained 222.82 U/mL, 206.98 U/mL and 217.23 U/mL, respectively. These results suggested that the fermented kiwifruit extract could exhibit antioxidant activity through tolerance to the digestive environment.

Antioxidant capacities and non-volatile metabolites changes after solid-state fermentation of soybean using oyster mushroom (Pleurotus ostreatus) mycelium

Mengxin He,Qing Peng,Xiaoqing Xu,Bo Shi,Yu Qiao

Given the abundance of beneficial properties and enzymes secreted by edible oyster mushrooms, their mycelium could serve as a starter for fermented foods to enhance their nutritional and bioactive quality. This study aimed to investigate the effects on the nutritional ingredients, antioxidant activity, and non-volatile metabolites during solid-state fermentation (SSF) of soybeans by Pleurotus ostreatus mycelium. The results indicated that the contents of dietary fiber and starch in fermented soybeans decreased, while the amounts of protein and lipid increased after SSF (P < 0.05). Analysis of the total phenolic content (TPC) and antioxidant activities of the fermented soybeans revealed that the methanolic extracts significantly increased TPC and antioxidant activities against intracellular reactive oxygen species (ROS) in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages, as well as against DPPH and ABTS radicals in vitro. A total 154 differential metabolites were identified after SSF, and a Spearman correlation study revealed a direct relationship between antioxidant activities and certain metabolites including phenolic compounds, oligopeptides, and free fatty acids etc. Among these metabolites, phenolic compounds produced by the shikimic acid pathway were diverse in variety and had the greatest multiple differences. The study discovered that a potential mechanism involving SSF with P. ostreatus mycelium increased the antioxidant activity of soybeans.

Antioxidant Activity and Volatile Oil Analysis of Ethanol Extract of Phoebe zhennan S. Lee et F. N. Wei Leaves

Liping Yu,Wang Cheng,Meifen Tian,Zhigang Wu,Xiaoli Wei,Xing Cheng,Mingwei Yang,Xuan Ma

The medicinal value ofP. zhennanhas been documented in traditional Chinese medicine books. The aim of this paper was to study the antioxidant activity of alcoholic extracts ofP. zhennanleavesusing 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2-phenyl-4,4,5,5-tetramethylimidazolineoxyl-1-oxyl-3-oxide (PTIO) radical scavenging and ferric ion reducing antioxidant power (FRAP) assays. The active components of the leaves were identified via headspace solid-phase microextraction and gas chromatography–mass spectrometry (HS-SPME-GC-MS). The results showed that the scavenging rate of DPPH was 94.67%with an EC50 value of 0.674 mg/mL at a concentration of 2 mg/mL. The maximum scavenging rate was 47.40% at a Trolox equivalent of 0.33 mg TE/mL for PTIO radicals. The FRAP reached 84.80% at 0.20 mg/mL concentration. The results confirmed the strong antioxidant activity of the extracts. Furthermore, 44 compounds, mostly terpenoids, obtained from the alcoholic extracts ofP. zhennanleaves were analyzed using HS-SPME-GC-MS and 15 of these compounds had a relative content exceeding 1%. The strong antioxidant activity of the alcoholic extracts ofP. zhennanleaves could be attributed to the presence of copaene (33.97%), β-caryophyllene (4.42%), δ-cadinene (11.04%), γ-muurolene (4.78%), cis-calamenene (2.02%), linalool (1.04%), α-pinene (1.46%), borneol acetate (1.5%), and γ-terpinene (0.66%). This study demonstrates the potential medicinal value of alcoholic extracts ofP. zhennanleaves.
